Siamese fighting fish (Betta splendens) are freshwater fish that are commonly found in Thailand and other Southeast Asian countries. In the present study, chrysin-loaded polymeric micelles (CPs) were developed and investigated for the masculinizing effects, survival rate, growth indices, and toxicity on Siamese fighting fish. CPs were prepared using a poloxamer. The micelle system of CPs that were formed at a chrysin-to-polymer ratio of 1:2 was found to be the most suitable monodispersed system and exhibited a nanosized diameter (74.2 ± 1.6 nm) with a narrow size distribution (0.288 ± 0.012). In vivo studies were performed using Siamese fighting fish larvae as animal models. In the in vivo toxicity study, the fish larvae were immersed in aqueous systems containing CPs that had five different chrysin concentrations of 1, 10, 100, 1000, and 10,000 ng/mL for 24, 48, and 72 h. Blank polymeric micelles and water were used as controls. The in vivo masculinization effect of CPs with different chrysin concentrations on the fish larvae was evaluated after 5 weeks of exposure. The results demonstrated that CPs with a chrysin concentration of 1000 ng/mL showed a masculinization effect of 94.59 ± 2.76% with a high fish larvae survival rate of 72.45 ± 5.09% and low toxicity. The present document corresponds to the final report (deliverable No. 7) of the specific agreement (SA 2) "To determine the incidence and epidemiological characteristics of ciguatera cases in Europe" within the Framework Partnership Agreement GP/EFSA/AFSCO/2015/03 "Risk characterization of ciguatera food poisoning in Europe". This report summarises the activities performed, results and conclusions obtained in the specific agreement. The objective of the SA2 was to estimate the incidence and to describe the epidemiological characteristics of ciguatera in the EU/EEA. A protocol for a harmonized ciguatera surveillance in the European Union (EU)/Economic European Area (EEA) was elaborated. The protocol includes a ciguatera case definition and questionnaires for collecting information on ciguatera cases or outbreak. Around half of the countries answered the data call for reporting cases. Four countries reported 34 outbreaks from 2012 to 2019. Spain and Portugal reported outbreaks due to consumption of fish captured in the Canary Islands and Madeira (autochthonous outbreaks), mainly due to consumption of Seriola spp. and Ephinephelus spp. In more than half of these outbreaks, the fish was captured by sport fishing. Germany and France reported outbreaks due to consumption of fish imported from outside the EU (imported outbreaks, mainly Lutjanus spp). Spain, Germany and France reported outbreaks in travellers to tropical endemic territories (travel related outbreaks). All the outbreaks cases presented neurological symptoms, most of them had also gastrointestinal symptoms and few outbreak cases mentioned cardiovascular symptoms. Six countries (Austria, France, Germany, Portugal, Spain and Switzerland) reported 34 single cases. The incidence rate in the EU/EEA was very low (0.0054 cases per 100.000 inhabitants per year). Abstract As yet European strains of Raphidiopsis raciborskii (previously Cylindrospermopsis raciborskii) have not been found to produce known cyanotoxins although their extracts have caused adverse effects in mammals, as shown using in vitro and in vivo experimental models. The present study investigated whether R. raciborskii isolated from Western Poland and Ukraine can affect fish cells using in vitro exposures of hepatocytes and red blood cells (RBC), and brain homogenates obtained from common carp (Cyprinus carpio) to 1.0% and 0.1% extracts of 7 strains. The studied extracts evoked different responses of catalase activity in hepatocytes with both increase and decrease observed under low and high concentrations. The cellular thiol pool was also altered with most extracts inducing a decrease in the activity of glutathione-S-transferase, and Ukrainian strains leading to an increase in glutathione level and a decrease in metallothionein content. All the studied extracts induced comparable reactive oxygen species formation, lipid peroxidation, protein carbonylation and DNA fragmentation in hepatocytes, and all but one increased the activity of caspase-3. Only one extract caused lysosomal membrane destabilization as measured by neutral red retention in RBC. In contrast to extracts of Ukrainian isolates, exposure of brain homogenates to extracts of Polish strains induced an increase in acetylcholinesterase activity suggesting the neurotoxic action of their exudates. The results indicate that both Polish and Ukrainian strains of R. raciborskii may pose a toxicological risk to freshwater fish, and further, that Polish strains may produce compound(s) evoking neurotoxic effects. Pesguard FG161™, a mixture of d-tetramethrin and cyphenothrin (1:3 ratio), is extensively used to achieve rapid control of adult dengue vector, Aedes aegypti, during the disease outbreaks. Both d-tetramethrin and cyphenothrin are synthetic pyrethroids that are known to have adverse effects on non-mammalian organisms such as fish. The present study intended to use zebrafish embryo toxicity model to investigate the toxic effect of the above binary mixture on fish. Particularly, zebrafish embryo toxicity model provides an alternative to acute fish toxicity tests in terms of animal welfare perspective as the embryos are not considered live until 5 days after fertilization. The zebrafish embryos (2 hrs after fertilization) were exposed to a binary mixture of pyrethroids at different concentrations (d-tetramethrin: 0.01 – 1.20 μmolL-1 and cyphenothrin: 0.03 – 3.20 μmolL-1) for 24, 48, and 72 hrs at room temperature (26°C) according to the OECD guideline no. 236. Percentage mortality of embryos were calculated by observing the lethal endpoints and LC50 values were calculated for each time interval employing the probit analysis. This binary mixture was highly toxic to zebrafish embryos and was found to be concentration and time dependent. LC50 values at 24 hrs (d-tet: 0.58 μmolL-1, cyp: 1.74 μmolL-1) were significantly reduced in 48 hrs (d-tet: 0.11 μmolL-1, cyp: 0.33 μmolL-1) and 72 hrs (d-tet: 0.03 μmolL-1, cyp: 0.09 μmolL-1). Coagulation of embryos was the most common lethal effect observed and lack of somite formation and lack of heartbeat were also observed. The present study revealed that the binary mixture is highly toxic to zebrafish embryos even when based on nominal concentrations. Hence, extensive use of these pesticides could be detrimental to fish population and integrated vector control methods which involve the minimum use of insecticides are recommended. Abstract Fish consumption is an important route of exposure to persistent organic pollutants (POPs) in dolphins as well as humans. In order to assess the potential risks associated with these contaminants, 39 whole fish and 37 fillets from fish representing species consumed by dolphins and humans captured from Charleston Harbor and tributaries, South Carolina, USA, were measured for a suite of POPs. Polychlorinated biphenyls (PCBs) were the predominant contaminant with concentrations ranging from 5.02 to 232.20 ng/g in whole fish and 5.42–131.95 ng/g in fillets (weight weight ww) followed by total organochlorine pesticides (OCPs) and polybrominated diphenyl ethers (PBDEs). Total POPs levels varied by location and species with general trends indicating significantly higher levels in fish from the Cooper (93.4 ng/g ww) and Ashley Rivers (56.2 ng/g ww) compared to Charleston Harbor (31.6 ng/g ww). Mullet and spot were found to have significantly higher PCBs, OCPs and total POPs, 2–3 times higher than red drum; mullet were also significantly higher in OCPs compared to seatrout. PCB concentrations in whole fish and fillets exceeded EPA human screening values for cancer risk in all fish sampled. For PCBs in fillets, all samples had values of maximum allowable meals per month that were less than the EPA, FDA guidelines for recommended fish meals per month, suggesting lower (more stringent) allowable fish meals per month. All fish exceeded PBDE wildlife values and all fish except two exceeded the level where 95% of the dolphin population would have tissue levels below the health effect threshold. Considering that POP concentrations in fish potentially consumed by humans exceed human health effect thresholds levels, consumption advisories should be considered as a prudent public health measure. The former lead-zinc mine at Mestersvig, Greenland, continues to contaminate the surrounding environment despite its operations ceasing over 50 years ago. Elevated concentrations of heavy metals are found in water, sediment and biota in the terrestrial, freshwater and marine environments. To shed light on the present contamination and its potential effects on local fish we investigated gill and liver histology of sculpins ( Myoxocephalus spp.) around the former mining area. Two species of sculpins were caught; shorthorn sculpins ( M. scorpius ; n = 16) and fourhorn sculpins ( M. quadricornis ; n = 17) at a contaminated site, Nyhavn, and shorthorn sculpins ( M. scorpius ; n = 25) at the reference site. In a previous study we found histopathological changes in the tissues of the sculpins, and we suspected this to be related to elevated heavy metal tissue concentrations. Concentrations of Fe, Hg, Mn, Pb, Se and Zn were significantly higher in sculpins at Nyhavn compared to the reference site. Reference NOED and LOEC thresholds for biochemistry, tissue lesions, growth, survival and reproduction for hepatic Hg, As, Cd and Pb from the ERED database were exceeded in both sculpin species. Histopathological investigations of the sculpins gills revealed significant increases in the prevalence of hyperplastic epithelium, inflammation, intensity of neutral and total mucus cells and chloride cells along with an increased infection of colonial Peritricha. At the contaminated Nyhavn site, fourhorn sculpins had a significantly higher prevalence of chondroplastic tissue and intensity of neutral, mixed and total mucus cells in the gills compared to the shorthorn sculpins. The data indicate that both sculpin species could be useful indicator species for environmental monitoring of metal pollution in Arctic areas. However, confounding effects of gender and species should be investigated further. The decline in marine and freshwaters catches in recent years in Colombia has led to a change in dietary habits, with an increase in the purchase and consumption of imported fish. This is of particular concern as fish are sometimes caught in mercury-contaminated waters, and are subsequently sold canned or uncanned. In addition, canned tuna has received little attention as it is widely assumed that concentrations are low. In this study, total mercury (THg) and methylmercury (MeHg) concentrations were evaluated in three imported fish species marketed in Colombia, Prochilodus lineatus, Prochilodus reticulatus, and Pangasianodon hypophthalmus, plus four brands of canned tuna and one of sardines. One brand of tuna showed the highest mean concentrations of THg (0.543 ± 0.237 μg/g, wet weight, ww) and MeHg (0.518 ± 0.337 μg/g ww), while concentrations in P. hypophthalmus were approximately 30 times lower (≈0.02 µg/g ww). The estimated weekly intake (EWI) in children was above the provisional tolerable weekly intake (PTWI) of MeHg established by the Joint FAO/World Health Organization (WHO) Expert Committee on Food Additives (JECFA) in 2007, 1.6 μg/kg body weight (bw) per week, for all the canned tuna brands. Values for adults were below PTWI, whereas for women of childbearing age, values were above PTWI only for brand D of canned tuna. The estimate of the potential risk indicated that MeHg levels in canned tuna can generate negative effects in vulnerable groups, while the EWI of fresh fish did not pose a threat to the general population. Therefore, establishing strategies to address the high consumption of canned tuna, and continuous monitoring to control commercial food, are recommended to decrease Hg exposure. To tackle the ever-present global concern regarding human exposure to persistent organic pollutants (POPs) via food products, this study strived to indicate associations between organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) in lake-fish tissue depending on the species and sampling season. Apart from the monitoring initiatives recommended in the Global Monitoring Plan for POPs, the study discussed 7 OCPs and 18 PCB congeners determined in three Cyprinidae species (rudd, carp, and Prussian carp) from Vransko Lake (Croatia), which are widely domesticated and reared as food fish across Europe and Asia. We exploit advanced classification algorithms, the Kohonen self-organizing maps (SOM) and Decision Trees (DT), to search for POP patterns typical for the investigated species. As indicated by SOM, some of the dioxin-like and non-dioxin-like PCBs (PCB-28, PCB-74, PCB-52, PCB-101, PCB-105, PCB-114, PCB-118, PCB-156 and PCB-157), α-HCH and β-HCH caused dissimilarities among fish species, but regardless of their weight and length. To support these suggestions, DT analysis sequenced the fish species and seasons based on the concentration of heavier congeners. The presented assumptions indicated that the supplemental application of SOM and DT offers advantageous features over the usually rough interpretation of POPs pattern and over the single use of the methods. Three aquatic products, tilapia fish, lobster and crab, were collected from the landing site of fishermen in Makoko, a fishing suburb in Lagos State, Nigeria. They were analyzed for their proximate composition, and concentration of heavy metals (Cr, Cd, and Pb) was also measured in them. The nutrient values varied among the three organisms. The lobsters and crabs had higher protein values while tilapia was richer in lipid and moisture content. In most cases, these differences were not significant. The concentration of heavy metals also differed in the three organisms. All organisms had high concentrations of the three heavy metals analyzed. The heavy metal load as observed in this study is consistent with some previously reported values in literature. Plausible reasons for these differences were discussed in light of size, trophic level, choice of food of the organisms, and anthropogenic activities. It may be advisable that heavy metal concentrations are regularly monitored in edible aquatic food products so that human health is not at risk. [ABSTRACT FROM AUTHOR]

To trace the most concerned bioavailable mercury (Hg) in aquatic environment, fish samples were collected from three typical regions in China, including 3 rivers and 1 lake in the Tibetan Plateau (TP, a high altitude background region with strong solar radiation), the Three Gorges Reservoir (TGR, the largest artificial freshwater reservoir in China), and the Chinese Bohai Sea (CBS, a heavily human-impacted semi-enclosed sea). The Hg isotopic compositions in fish muscles were analyzed. The results showed that anthropogenic emissions were the main sources of Hg in fish from TGR and CBS because of the observed negative δ 202 Hg and positive Δ 199 Hg in these two regions (TGR, δ 202 Hg: − 0.72 to − 0.29‰, Δ 199 Hg: 0.15 – 0.52‰; CBS, δ 202 Hg: − 2.09 to − 0.86‰, Δ 199 Hg: 0.07 – 0.52‰). The relatively higher δ 202 Hg and Δ 199 Hg (δ 202 Hg: − 0.37 – 0.08‰, Δ 199 Hg: 0.50 – 1.89‰) in fish from TP suggested the insignificant disturbance from local anthropogenic activities. The larger slopes of Δ 199 Hg/Δ 201 Hg in fish from TGR (1.29 ± 0.14, 1 SD) and TP (1.25 ± 0.06, 1 SD) indicated methylmercury (MeHg) was produced and photo-reduced in the water column before incorporation into the fish. In contrast, the photoreduction of Hg 2+ was the main process in CBS (slope of Δ 199 Hg/Δ 201 Hg: 1.06 ± 0.06, 1 SD). According to the fingerprint data of Hg isotopes, the most important source for aquatic bioavailable Hg in TP should be the long-range transported Hg, contrasting to the anthropogenic originated MeHg from surface sediments and runoffs in TGR and inorganic Hg from continental inputs in CBS. Therefore, the isotopic signatures of Hg in fish can provide novel clues in tracing sources and behaviors of bioavailable Hg in aquatic systems, which are critical for further understanding the biogeochemical cycling of Hg. Selenium pollution from the E.W. Brown Electric Generating Station was investigated in Herrington Lake, KY. Coal ash wastewater is discharged as surface water overflow from ash disposal ponds into the lake via a National Pollutant Discharge Elimination System permit issued by the Kentucky Division of Water, but the permit does not restrict or limit the amount of selenium released. Unpermitted discharges occur from seeps and drainage through leaks in ash pond dams. Together, these discharges have resulted in selenium concentrations in water, sediment, benthic macroinvertebrates, and fish that are 2–9 times the level that is toxic for fish reproduction and survival. A large proportion (12.2%, or 25 times background) of juvenile largemouth bass ( Micropterus salmoides , the only species examined) exhibited spinal and/or craniofacial malformations that are consistent with selenium poisoning. Teratogenic Deformity Index values indicated a 3.05% population-level impact on the bass fishery, with total selenium-induced mortality (including pre-swimup mortality) estimated to be in excess of 25% per year. Carbon Nanotubes are among the most promising materials for the technology industry. Their unique physical and chemical proprieties may reduce the production costs and improve the efficiency of a large range of products. However, the same characteristics that have made nanomaterials interesting for industry may be responsible for inducing toxic effects on the aquatic organisms. Since the carbon nanotubes toxicity is still a controversial issue, we performed tests of acute and subchronic exposure to a commercial sample of multiwalled carbon nanotubes in two fish species, an exotic model ( Danio rerio ) and a native one ( Astyanax altiparanae ). Using the alkaline version of the comet assay on erythrocytes and the piscine micronucleous, also performed on erythrocytes, it was verified that the tested carbon nanotubes sample did not generate apparent genotoxicity by means of single/double DNA strand break or clastogenic/aneugenic effects over any of the species, independently of the exposure period. Although, our findings indicate the possibility of the occurrence of CNTs-DNA crosslinks. Apparently, the sample tested induces oxidative stress after subchronic exposure as shown by activity of superoxide dismutase and catalase. Notothenioid fish and invertebrate samples from Antarctica were collected in the austral summer of 2009, and analyzed for persistent organic pollutants (POPs), including polycyclic aromatic hydrocarbons (PAHs), organochlorine pesticides (OCPs), and polybrominated diphenylethers (PBDEs), as well as δ13C and δ15N stable isotopes for trophic level determination. In this study, the POP levels in the Antarctic biota samples were found to be ranked in the following order: OCPs > PAHs >> PBDEs. The POP levels in notothenioid fish and krill correlate to trophic levels; however, the POP concentrations in intertidal benthic invertebrates are higher than in notothenioid fish implying that specific biogeochemical factors may affect bioaccumulation in the Antarctica ecosystem. Biomagnification of POPs may have a smaller role than bioconcentration in Antarctica environment. In addition to the source, transport, exposure, and absorption for each group of POPs in the short food chain in Antarctica, the biological variation among species, interaction habitats, diet and metabolism are also factors for future studies on contaminant bioaccumulation. Background: We have come across the plant secondary metabolites having pesticidal properties being used in the form of pesticides. In this study, we used one of newly available and plant metabolite used as pesticide in wide agricultural fields - Karanjin, obtained from seeds of plant Pongamia pinnata. Methods: The study was conducted during the month of March 2016 at the Department of Zoology, Osmania University. The fish common carp- Cyprinus carpio was exposed to sub-lethal concentration of karanjin i.e., 1/10th of 96 h LC50 value (0.28 ppm) for a period of 21 d. Moreover, at 24 h, 7 d, 14 d and 21 d, the peripheral blood of both control and exposed group fishes were studied for the presences of micronuclei and other nuclear abnormalities. Results: The micronuclei were completely absent, i.e., were not seen during the sub-lethal exposure period. There were seen nuclear abnormalities such as blebbed nuclei (BN), notched nuclei (NN), differently shaped nuclei (DSN), pear-shaped nuclei (PSN), circular nuclei (CN), lobed nuclei (LN) and Karyolysed nuclei (KN). The control group showed few nuclear abnormalities. Conclusion: During the sub-lethal exposure, as the days of exposure increased, the types of aberrations and their number also increased. Silver nanoparticles (Ag-NPs) and its byproducts can spread pollution in aquatic habitat. Liver and gills are key target for toxicity. Oxidative stress, tissue alterations, and hemotoxicity are assumed to be associated with Ag-NPs in target animals. Cerium oxide nanoparticles (nano-ceria) show antioxidant potential in scavenging the free radicals generated in Ag-NP-induced oxidative stress. We determined ameliorated role of nano-ceria against Ag-NP-induced toxicity in fresh water Labeo rohita (L. rohita). Four groups were used in study including control, nano-ceria, Ag-NPs, and Ag-NPs + nano-ceria. Ag-NPs (30 mg l−1) and nano-ceria (50 µg kg−1) were given through water and prepared feed, respectively. The samples were taken after 28 days. Results demonstrated that pre-treatment of nano-ceria recovered L. rohita from Ag-NP-induced toxicity and oxidative stress. Nano-ceria pre-treatment actively mimics the activity of GST, GSH, CAT, and SOD. Furthermore, Ag-NPs’ treatment caused severe inflammation and necrosis in hepatic parenchyma which leaded to congestion of blood in hepatic tissues. Accumulation of a yellow pigment in hepatic tissue was also seen due to necrosis of affected cells. In nano-ceria pre-treatment, there was no congestion in hepatic tissue. Vacuolization of cells and necrosis in some area was recorded in nano-ceria pre-treated group, but the gill and hepatic tissue showed improvement against Ag-NP-induced damage. Nano-ceria pre-treatment also improved hematological parameters in Ag-NP-treated fish. The levels of 20 metals (aluminum, boron, barium, calcium, cadmium, cobalt, chromium, copper, iron, potassium, lithium, magnesium, manganese, molybdenum, sodium, nickel, lead, strontium, vanadium, and zinc) were analyzed in muscle and liver tissue of Trachurus picturatus marketed in the Canary Islands (Spain) by using inductively coupled plasma optical emission spectrometry. In the liver samples, the mean concentrations in milligrams per kilogram wet weight (wt) of Al (14.7), B (0.99), Ba (1.64), Ca (314), Cd (2.52), Co (0.15), Cu (4.07), Fe (106), Li (3.89), Mn (0.85), Mo (0.16), Na (1510), Ni (0.51), Pb (0.36), Sr (3.54), V (0.78), and Zn (23.13) were higher than those detected in the muscle samples in milligrams per killogram wet wt, which were as follows: Al (8.76), B (0.07), Ba (0.30), Ca (210), Cd (0.01), Co (0.01), Cu (1.51 ), Fe (7.33), Li (1.08), Mn (0.12), Mo (0.01), Na (697), Ni (0.11), Pb (0.04), Sr (1.45), V (0.01), and Zn (4.69). The mean concentrations of Cr, K, and Mg (0.14, 1,904, and 243 mg/kg wet wt, respectively) were higher in muscle than in liver (0.05, 1,333 and 236 mg/kg wet wt, respectively). The mean concentrations of Cd and Pb (0.01 and 0.04 mg/kg wet wt) in muscle did not exceed the maximum limits established by a European Commission regulation (0.1 mg of Cd/kg and 0.3 mg of Pb/kg, respectively). Considering a mean daily consumption of fish muscle for the adult population of 31.9 g/day published in the report on food consumption by the Spanish Ministry of Agriculture and Fisheries, Food and Environment, Mg made the highest contributions to the intake (2.58% for adult women of 60 kg and 2.22% for men of 70 kg), and the estimated intakes of Al (0.35 to 0.46 mg/day), Cd (0.55 to 0.74 mg/day) and Pb (1.66 to 5.53 mg/day) were below the respective established tolerable intakes. In conclusion, the results of this study show that the consumption of muscle from this benthopelagic species can be considered safe in terms of maximum legal limits, while consumption of liver is discouraged as a major source of exposure to toxic metals, such as Al, Cd, and Pb. Dinoflagellate species belonging to the genera Gambierdiscus and Fukuyoa produce ciguatoxins (CTXs), potent neurotoxins that concentrate in fish causing ciguatera fish poisoning (CFP) in humans. While the structures and toxicities of ciguatoxins isolated from fish in the Pacific and Caribbean are known, there are few data on the variation in toxicity between and among species of Gambierdiscus and Fukuyoa. Quantifying the differences in species-specific toxicity is especially important to developing an effective cell-based risk assessment strategy for CFP. This study analyzed the ciguatoxicity of 33 strains representing seven Gambierdiscus and one Fukuyoa species using a cell based Neuro-2a cytotoxicity assay. All strains were isolated from either the Caribbean or Gulf of Mexico. The average toxicity of each species was inversely proportional to growth rate, suggesting an evolutionary trade-off between an investment in growth versus the production of defensive compounds. While there is 2- to 27-fold variation in toxicity within species, there was a 1740-fold difference between the least and most toxic species. Consequently, production of CTX or CTX-like compounds is more dependent on the species present than on the random occurrence of high or low toxicity strains. Seven of the eight species tested (G. belizeanus, G. caribaeus, G. carolinianus, G. carpenteri, Gambierdiscus ribotype 2, G. silvae and F. ruetzleri) exhibited low toxicities, ranging from 0 to 24.5 fg CTX3C equivalents cell-1, relative to G. excentricus, which had a toxicity of 469 fg CTX3C eq. cell-1. Isolates of G. excentricus from other regions have shown similarly high toxicities. If the hypothesis that G. excentricus is the primary source of ciguatoxins in the Atlantic is confirmed, it should be possible to identify areas where CFP risk is greatest by monitoring only G. excentricus abundance using species-specific molecular assays. The present study reports total mercury (THg) and methylmercury (MeHg) concentrations in 32 different lean fish species from the Western Mediterranean Sea, with a special focus on the Balearic Islands. The concentrations of THg ranged between 0.05 mg/kg ww and 3.1 mg/kg ww (mean 0.41 mg/kg ww). A considerable number of the most frequently fish species consumed by the Spanish population exceed the maximum levels proposed by the European legislation when they originate from the Mediterranean Sea, such as dusky grouper (100% of the examined specimens), common dentex (65%), conger (45%), common sole (38%), hake (26%) and angler (15%), among others. The estimated weekly intakes (EWI) in children (7–12 years of age) and adults from the Spanish population (2.7 µg/kg bw and 2.1 µg/kg bw, respectively) for population only consuming Mediterranean fish were below the provisional tolerable weekly intake (PTWI) of THg established by EFSA in 2012, 4 µg/kg bw. However, the equivalent estimations for methylmercury, involving PTWI of 1.3 µg/kg bw, were two times higher in children and above 50% in adults. For hake, sole, angler and dusky grouper, the most frequently consumed fish, the estimated weekly intakes in both children and adults were below the maximum levels accepted. These intakes correspond to maximum potential estimations because fish from non-Mediterranean origin is often consumed by the Spanish population including the one from the Balearic Islands. Exposure to contaminants in fish may be associated with adverse health outcomes even as fish consumption is generally considered beneficial. Risk assessments conducted to support regulatory analyses rely on quantitative fish consumption estimates. Here we report the results of a national survey of high-frequency fish consumers (n = 2099) based on a survey population statistically representative of ~17.6 million U.S. individuals consuming three or more fish meals per week. The survey was conducted during 2013 using an on-line survey instrument. Total fish consumption averaged 111 g/day from market, restaurant and self-caught sources. Depending on the season, the incidence of individuals reporting consumption of self-caught species ranged between 10–12% of our high-frequency fish consuming demographic, averaging approximately 30 g/day and comprising 23% of total fish consumption from all sources of fish. Recreational or self-caught consumption rates vary regionally and are poorly understood, particularly for high-frequency consumers, making it difficult to support national-scale assessments. A divergence between sport-fishing and harvesting of fish as a food-staple is apparent in survey results given differences in consumption patterns with income and education. Highest consumption rates were reported for low income respondents more likely to harvest fish as a food staple. By contrast, the incidence of self-caught fish consumption was higher with income and education although overall consumption rates were lower. Regional differences were evident, with respondents from the East-South Central and New England regions reporting lowest consumption rates from self-caught fish on the order of 12–16 g/day and those from Mountain, Pacific and Mid-Atlantic regions reporting highest rates ranging from 44 to 59 g/day. Respondent-specific consumption rates together with national-level data on fish tissue concentrations of PCBs, MeHg, and PFOS suggest that 10–58% of respondents reporting self-caught fish consumption are exposed to concentrations of these contaminants that exceed threshold levels for health effects based on a target hazard index of one, representing 2.3 M to 19 M individuals. The results of this nationwide survey of high-frequency fish consumers highlights regional and demographic differences in self-caught and total fish consumption useful for policy analysis with implications for distributional differences in potential health impacts in the context of both contaminant exposures as well as protective effects. Thirteen microelements (Al, As, Cd, Co, Cr, Cu, Fe, Li, Mn, Ni, Ti, V, and Zn) and four macroelements (Ca, K, Mg, and Na) were determined in the edible muscle tissue of fish in Cienfuegos Bay. Eight species, Albula vulpes, Diapterus rhombeus, Gerres cinereus, Haemulon carbonarium, Haemulon sciurus, Micropogonias furnieri, Kyphosus sectatrix, and Lutjanus cyanopterus were studied. The total concentrations and extracted concentrations using HCl were also determined in sediments. The quality of the analysis was guaranteed by validating the used analytical methods with certified reference materials of fish and sediments. The high content of macroelements confirmed the importance of fish as a valuable source of food for the local population. New information is provided on bioaccumulation of toxic elements As, Cd, Cu, Zn, and Cr in fish of the bay, with concentrations that overcame the local or international maximum allowable levels for human consumption in most samples analyzed. Moray eels (Gymnothorax species) from tropical waters have long been known to be high-risk species, and the consumption of particularly the viscera or ungutted eels can result in severe ciguatera (known as Gymnothorax or moray eel poisoning), characterized by prominent neurological features. In this review, the main objective was to describe the risk and severity of ciguatera caused by eating moray eels in different parts of the world. Moray eels can accumulate very high ciguatoxin (CTX) levels in the flesh and particularly the liver. Therefore, even the smaller ones can be toxic and the consumption of an average portion (particularly liver) can result in severe or fatal ciguatera. Moray eels (particularly when ungutted) must never be served in gatherings since they can cause mass poisoning because of their large sizes and high CTX levels. Silver nanoparticles (17.78 ± 12.12 nm) were synthesized by the reduction of 0.5 M silver nitrate using formaldehyde as reducing and triethylamine as promoting and stabilizing agent. The particles were grain like agglomerates with spherical, centered-face cubic and crystalline in nature. The sample was highly pure with amine (NH) as associated and capping molecules. Further, the genotoxicity and oxidative stress of these particles were evaluated using Labeo rohita (L. rohita) as genetic model exposed (10-55 mg L-1 dose) through aquatic medium for 28 days. The cells were produced with micronuclei, fragmented, lobed and buds nuclei in dose dependent manner. The highest incidence of comet was recoded (27.34 ± 5.68) at 55 mg L-1 Ag-NPs and 14 days treatment. Then frequency was decreased to 22.65 ± 6.66% after 28 days due to complex repair mechanism. Moreover, the treatment also produces the oxidative stress and disturbs the level of GST in gill and liver tissue. There was a sharp decline in the activities of GST and this decrease of activity increase the MDA content. Further, the elevated level of GSH represents that the liver has started defensive mechanism against oxyraidcals. In Australia, trace organic contaminants (TrOCs) and endocrine active compounds (EACs) have been detected in rivers impacted by sewage effluent, urban stormwater, agricultural and industrial inputs. It is unclear whether these chemicals are at concentrations that can elicit endocrine disruption in Australian fish species. In this study, native rainbowfish ( Melanotaenia fluviatilis ) and introduced invasive (but prevalent) mosquitofish ( Gambusia holbrooki ) were exposed to the individual compounds atrazine, estrone, bisphenol A, propylparaben and pyrimethanil, and mixtures of compounds including hormones and personal care products, industrial compounds, and pesticides at environmentally relevant concentrations. Vitellogenin (Vtg) protein and liver Vtg mRNA induction were used to assess the estrogenic potential of these compounds. Vtg expression was significantly affected in both species exposed to estrone at concentrations that leave little margin for safety (p < 0.001). Propylparaben caused a small but statistically significant 3× increase in Vtg protein levels (p = 0.035) in rainbowfish but at a concentration 40× higher than that measured in the environment, therefore propylparaben poses a low risk of inducing endocrine disruption in fish. Mixtures of pesticides and a mixture of hormones, pharmaceuticals, industrial compounds and pesticides induced a small but statistically significant increase in plasma Vtg in rainbowfish, but did not affect mosquitofish Vtg protein or mRNA expression. These results suggest that estrogenic activity represents a low risk to fish in most Australian rivers monitored to-date except for some species of fish at the most polluted sites. Fish venoms are often poorly studied, in part due to the difficulty in obtaining, extracting, and storing them. In this study, we characterize the cardiovascular and neurotoxic effects of the venoms from the following six species of fish: the cartilaginous stingrays Neotrygon kuhlii and Himantura toshi, and the bony fish Platycephalus fucus, Girella tricuspidata, Mugil cephalus, and Dentex tumifrons. All venoms (10-100 μg/kg, i.v.), except G. tricuspidata and P. fuscus, induced a biphasic response on mean arterial pressure (MAP) in the anesthetised rat. P. fucus venom exhibited a hypotensive response, while venom from G. tricuspidata displayed a single depressor response. All venoms induced cardiovascular collapse at 200 μg/kg, i.v. The in vitro neurotoxic effects of venom were examined using the chick biventer cervicis nerve-muscle (CBCNM) preparation. N. kuhlii, H. toshi, and P. fucus venoms caused concentration-dependent inhibition of indirect twitches in the CBCNM preparation. These three venoms also inhibited responses to exogenous acetylcholine (ACh) and carbachol (CCh), but not potassium chloride (KCl), indicating a post-synaptic mode of action. Venom from G. tricuspidata, M. cephalus, and D. tumifrons had no significant effect on indirect twitches or agonist responses in the CBCNM. Our results demonstrate that envenoming by these species of fish may result in moderate cardiovascular and/or neurotoxic effects. Future studies aimed at identifying the molecules responsible for these effects could uncover potentially novel lead compounds for future pharmaceuticals, in addition to generating new knowledge about the evolutionary relationships between venomous animals. The aim of this study was to investigate and compare total mercury (Hg), selenium (Se), and Se:Hg molar ratios in fish muscles (phytophages n=3; benthophages n=32; predators n=5) and semiaquatic carnivores, including piscivores (the European otter n=8, the feral American mink n=7) and the omnivorous raccoon (n=37) from a riverine European ecosystem in a Se-deficient area. The Hg concentration in fish reached 0.337 μg/g dry weight, dw (0.084 μg/g wet weight, ww). We found significant differences among Hg levels in tested vertebrate groups (predators vs benthophages: 0.893 vs 0.281 μg/g; piscivores vs omnivores: 6.085 vs 0.566 μg/g dw). Fish groups did not differ in Se concentrations, with a mean value of 0.653 μg/g dw. Significant differences were revealed between Se levels in piscivorous and omnivorous carnivores (0.360 vs 0.786 μg/g dw, respectively). Fish Se:Hg molar ratio values were >2.2. Benthophages had higher the ratio than predators but similar to phytophages. Among carnivores, piscivores had much lower the ratio than raccoon (0.14 vs 3.75) but raccoon and fish medians did not significantly differ. We found almost two times higher Se levels in fish and raccoons compared to piscivores, possibly resulting from lower fish Se digestibility by piscivores in contrast to higher absorption of plant Se by many fish and omnivorous raccoons. Considering that a tissue Se:Hg molar ratio <1 may be connected with a Hg toxicity potential increase, we assume that piscivores in Se-deficient area are in worse situation and more exposed to Hg than fish and omnivores. Pufferfish, Lagocephalus sceleratus, L. suezensis and L. spadiceus, are widely distributed Lessepsian species along the Turkish and other eastern Mediterranean coasts. L. sceleratus is considered one of the worst invasive species in the Mediterranean Sea and recently caused human intoxication by accidental consumption. Therefore, methods to assess pufferfish toxicity and accurately identify toxic pufferfish are important for food safety. Firstly, 20 specimens of L. sceleratus were collected from the eastern Mediterranean coasts of Turkey. Tetrodotoxin (TTX) content was determined among different tissues (muscle, liver, gonad, kidney, intestine and skin) by liquid chromatography-tandem mass spectrometry. All of the specimens contained TTX. The highest TTX concentration was detected in the ovary (80.0 µg TTX/g), followed by the intestine (48.8 µg TTX/g), kidney (34.0 µg TTX/g) and liver (25.4 µg TTX/g). The flesh and testis contained detectable amounts of TTX (3.4 µg TTX/g and 2.6 µg TTX/g, respectively). TTX analogs were also detected in the tissues, with 4,9-anhydro-5,6,11-trideoxyTTX and 5,6,11-trideoxyTTX being the major analog. A species-specific PCR method was developed to identify three species of genus Lagocephalus based on the nicotinamide adenine dinucleotide dehydrogenase subunit 2 or cytochrome oxidase subunit I regions of the mitochondrial DNA of L. sceleratus, L. suezensis, and L. spadiceus. The designed primers amplified only DNA of target species and not those of other fish and shellfish species. The species-specific PCR method successfully identified these pufferfish species and could be applied to thermally treated products of the pufferfish with only 1 ng of extracted DNA. In the past few decades, extensive application of azoxystrobin has led to great concern regarding its adverse effects on aquatic organisms. The objective of the present study was to evaluate the reproductive toxicity of azoxystrobin to zebrafish. After adult zebrafish of both sexes were exposed to 2, 20 and 200 μg/L azoxystrobin for 21 days, egg production, the fertilization rate, the gonadosomatic index (GSI) and hepatosomatic index (HSI), 17β-estradiol (E2), testosterone (T) and vitellogenin (Vtg) concentrations, and histological alterations in the gonads and livers were measured. Meanwhile, expression alterations of genes encoding gonadotropins and gonadotropin receptors ( fshb , lhb, fshr and lhr ), steroid hormone receptors ( era , er2b and ar ), steroidogenic enzymes ( cyp11a , cyp11b , cyp17 , cyp19a , cyp19b , hsd3b and hsd17b ) in the hypothalamic-pituitary-gonad (HPG) axis and vitellogenin ( vtg1 and vtg2 ) in the livers were also investigated. The results showed that reduced egg production and fertilization rates were observed at 200 μg/L azoxystrobin. In female zebrafish, reduced E2 and Vtg concentrations, decreased GSI, increased T concentrations, and histological alterations in the ovaries and livers were observed at 200 μg/L azoxystrobin, along with significant down-regulation of lhb , cyp19b , lhr , cyp19a , vtg1 and vtg2 , and up-regulation of cyp17 , hsd3b and hsd17b . In male zebrafish, increased E2 and Vtg concentrations, reduced T concentration and GSI, and histological alterations in the testes and livers were observed after exposure to 20 and 200 μg/L azoxystrobin, along with significant up-regulations of cyp19b , cyp11a , cyp17 , cyp19a , hsd3b and hsd17b , vtg1 and vtg2 . Moreover, cyp11a , hsd3b , cyp19a , vtg1 and vtg2 in male zebrafish were significantly up-regulated after treatment with 2 μg/L azoxystrobin. The results of the present study indicate that azoxystrobin led to reproductive toxicity in zebrafish and male zebrafish were more sensitive to azoxystrobin than female zebrafish. Short-chain chlorinated paraffins (SCCPs) are ubiquitous in the environment and might cause adverse environmental and human health effects. Little is known about the relative toxicity of different SCCP compounds especially during development. The objective of this study was to characterize and compare effects of seven SCCP groups at environmentally relevant levels, using a zebrafish ( Danio rerio ) model. Observations on malformation, survival rates at 96 h post fertilization (hpf), and hatching rates at 72 hpf indicated that the C 10- groups (C 10 H 18 Cl 4 , 1,2,5,6,9,10-C 10 H 16 Cl 6 and C 10 H 15 Cl 7 ) were more toxic than the C 12- groups (C 12 H 22 Cl 4 , C 12 H 19 Cl 7 and 1,1,1,3,10,12,12,12-C 12 H 18 Cl 8 ) and Cereclor 63L. The C 10- groups were also more potent than C 12- groups and Cereclor 63L in decreasing thyroid hormone levels. Among the three compounds within the C 10- group, the compounds with less chlorine content had stronger effects on sub-lethal malformations but less effects on triiodothyronine (T3) and tetraiodothyronine (T4). Only C 10 H 18 Cl 4 significantly decreased the mRNA expression of tyr , ttr , dio2 and dio3 at a dose-dependent manner suggesting that the specific mode of actions differ with different congeners. The mechanisms of disruption of thyroid status by different SCCPs could be different. C 10 H 18 Cl 4 might inhibit T3 production through the inhibition effect on dio2 . These results indicate that SCCP exposure could alter gene expression in the hypothalamic-pituitary-thyroid (HPT) axis and thyroid hormone levels. The mechanisms of disruption of thyroid status by different SCCPs could be different. Our results on the relative developmental toxicities of SCCPs will be useful to reach a better understanding of SCCP toxicity supporting environmental risk evaluation and regulation and used as a guidance for environmental monitoring of SCCPs in the future. Local communities in some Asian, African and American countries, use plant toxins in fish poisoning for fishing activities; however, the effects of this practice on the particular wild fish assemblages is unknown. This study was conducted with the aim to investigate the effects of fish poisoning using Tephrosia candida, on freshwater fish diversity and abundance in streams at the boundary of the World Natural Heritage site, Sinharaja Forest Reserve, Sri Lanka. A total of seven field trips were undertaken on a bimonthly basis, from May 2013 to June 2014. We surveyed five streams with similar environmental and climatological conditions at the boundary of Sinharaja forest. We selected three streams with active fish poisoning practices as treatments, and two streams with no fish poisoning as controls. Physico-chemical parameters and flow rate of water in selected streams were also measured at bimonthly intervals. Fish were sampled by electrofishing and nets in three randomly selected confined locations (6 x 2 m stretch) along every stream. Fish species were identified, their abundances were recorded, and Shannon-Weiner diversity index was calculated for each stream. Streams were clustered based on the Bray-Curtis similarity matrix for fish composition and abundance. Physico-chemical parameters of water were not significantly different among streams (P > 0.05). A total of 15 fish species belonging to four different orders Cypriniformes, Cyprinodontiformes, Perciformes and Siluriformes were collected; nine species (60%) were endemic, and six (40%) were native species. From these, 13 fish species were recorded in streams with no poisoning, while five species were recorded in streams where poisoning was practiced. Four endemic and one native fish species were locally extinct in streams where fish poisoning was active. Fish abundance was significantly higher in control streams (32-39/m2) when compared to treatment streams (5-9/m2) (P < 0.05). We concluded that fish poisoning with T. candida may have resulted in a significant reduction in species richness, total abundance and Shannon-Weiner diversity index of fish species in the studied streams at the boundary of Sinharaja forest. We recommend that this fishing practice should be strictly regulated, and to inform local communities about the legal status of fish poisoning, its consequences and the importance of fish diversity for local livelihoods and conservation purposes. Chemical induced changes in fish biomarkers vitellogenin (VTG), secondary sex characteristics (SSC), and sex ratio indicate modes/mechanisms of action (MOAs) of EAS (estrogen, androgen and steroidogenesis) pathways. These biomarkers could be used for defining MOAs and the causal link between MOAs and adverse effects in fish for the identification of endocrine disrupting chemicals (EDCs). This paper compiled data sets of 150 chemicals for VTG, 57 chemicals for SSC and 38 chemicals for sex ratio in fathead minnow, medaka and zebrafish. It showed 1) changes in fish biomarkers can indicate the MOAs as anticipated; 2) in addition to EAS pathways, chemicals with non-EAS pathways induced changes in fish biomarkers; 3) responses of fish biomarkers did not always follow the anticipated patterns of EAS pathways. These responses may result from the interaction of chemical-induced multiple MOAs and confounding factors like fish diet, infection, culture conditions, general toxicity and stress response. The complex response of fish biomarkers to a chemical of interest requires EDC testing at multiple biological levels. Interpretation of fish biomarker data should be combined with relevant information at different biological levels, which is critical for defining chemical specific MOAs. The utility of fish biomarker data for identification, classification, PBT assessment, risk assessment, and testing of EDCs in the regulatory context was discussed. Metal bioaccumulation and induction of biomarkers such as lipid peroxidation (LPO), superoxide dismutase (SOD), catalase (CAT), glutathione S transferase (GST), reduced glutathione (GSH) and DNA damage are potential indicators of stress in Channa punctatus exposed to effluents. In canal water, receiving thermal power plant discharges, Fe and Ni concentrations exceeded the recommended guidelines set by the United Nations Environment Programme Global Environment Monitoring System (UNEPGEMS). Fe was highly bioavailable and accumulated in all organs (liver, kidney, muscle and integument). The highest metal pollution index (MPI) value of 41.2 was observed in kidney and the lowest 13.5 in muscle tissue. LPO, SOD, CAT and GST levels were significantly higher in liver and kidney, whereas GSH levels declined significantly compared to fish from the reference site. Concomitant damage to DNA was observed with significantly higher mean tail length in the exposed fish gill cells (26.5 µm) and in liver (20.8 µm) compared to reference fish. Fish venom remains a virtually untapped resource. There are so few fish toxin sequences for reference, which increases the difficulty to study toxins from venomous fish and to develop efficient and fast methods to dig out toxin genes or proteins. Here, we utilized Chinese yellow catfish (Pelteobagrus fulvidraco) as our research object, since it is a representative species in Siluriformes with its venom glands embedded in the pectoral and dorsal fins. In this study, we set up an in-house toxin database and a novel toxin-discovering protocol to dig out precise toxin genes by combination of transcriptomic and proteomic sequencing. Finally, we obtained 15 putative toxin proteins distributed in five groups, namely Veficolin, Ink toxin, Adamalysin, Za2G and CRISP toxin. It seems that we have developed a novel bioinformatics method, through which we could identify toxin proteins with high confidence. The results show that although there is a positive interspecies correlation, the relationship is poor. Analysis on the excess toxicity calculated from toxic ratios (TR) shows that many chemicals have close toxicities and share the same MOAs between the two species. Linear relationships between the toxicities and octanol/water partition coefficient (log KOW) for baseline and less inert compounds indicate that the internal critical concentrations (CBRs) approach a constant both to fish and V. fischeri for neutral hydrophobic compounds. These compounds share the same toxic mechanisms and bio-uptake processes between species. On the other hand, some hydrophilic compounds exhibit different toxic effects with greatly different log TR values between V. fischeri and fish species. These hydrophilic compounds were identified as reactive MOAs to V. fischeri, but not to fish. The interspecies correlation is improved by adding a hydrophobic descriptor into the correlation equation. This indicates that the differences in the toxic ratios between fish and V. fischeri for these hydrophilic compounds can be partly attributed to the differences of bioconcentration between the two species, rather than the differences of reactivity with the target macromolecules. These hydrophilic compounds may more easily pass through the cell membrane of V. fischeri than the gill and skin of fish, react with the target macromolecules and exhibit excess toxicity. The compounds with log KOW > 7 exhibiting very low toxicity (log TR < –1) to both species indicate that the bioconcentration potential of a chemical plays a very important role in the identification of excess toxicity and MOAs. Toxicological evaluation of Clarias gariepinus from bitumen-polluted River Oluwa, Nigeria, was carried out in furtherance of studies on the environmental impacts of the bitumen exploration in Ondo State, Nigeria. Samples were taken from three different (two polluted and one as control) sites. The effect of changes in monthly seasonal flow rate was assessed for the sites of study. Blood plasma clinical-chemical parameters (BCCPs) and histological changes/lesions in various organs were evaluated as markers of pollution in the fish blood using standard methods. The result of the physicochemical properties of water from the sampling points revealed some of the values conforming to approved standards while others showed deviation. Significant differences were found in the blood and histological endpoints between the control and the polluted sites as well as between the two seasons evaluated across the sites. Ecotoxicological effects of industrial chemicals (Rig wash, Oil eater, Nalco, Glycol?) and pesticides (Propoxur, Deltamethrin, Atrazine, Furadan) on Tilapia guineensis (fish) and Aporrectodea longa (earthworms) were tested using the Organisation for Economic Cooperation and Development (OECD) # 203 and 207 protocols. The water and soil ratings indicate that the test chemicals were toxic to the organisms. The estimated 96 hour lethal concentration LC50 values for Rig wash, Oil eater, Nalco EC1304A/COT 505, Glycol, Propoxur, and Deltamethrin were 26.34±0.46, 6.02±0.30, 3.07±0.14, 1.31±0.01, 20.91±0 and 0.01±0 mg/l respectively. In the earthworm bioassay, the estimated 14-day LC50 values for Rigwash, Oil eater, Nalco EC1304A/COT 505, Glycol, Atrazine and Furadan were 80.05±3.5, 151.55±10.7, 172.63±14.2, 63.72±2.43, 4.97±0 and 0.29±0 mg/kg respectively. Safety factors are arbitrarily built in around the LC50 values in order to arrive at environmentally tolerable concentrations. The concentration of a chemical in the receiving environment should not exceed 10% of the LC50. The organisms exposed to the test chemicals showed significant difference when compared with the levels measured in the control group. The observed sensitivity of the test organisms to the chemicals indicates that adherence to standard safety limits/measures should be maintained during use and disposal of hazardous chemicals. Toxicity data to fish and algae were used to investigate excess toxicity between species. Results show that chemicals exhibiting excess toxicity to fish also show excess toxicity to algae for most of the compounds. This indicates that they share the same mode of action between species. Similar relationships between log K OW and toxicities to fish and algae for baseline and less inert compounds suggest that they have similar critical body residues in the two species. Differences in excess toxicity for some compounds suggest that there is a difference of physiological structure and metabolism between fish and algae. Some reactive compounds (e.g. polyamines) exhibit greater toxic effects for algae than those for fish because of relatively low bio-uptake potential of these hydrophilic compounds in fish as compared with that in algae. Esters exhibiting greater toxicity in fish than that in algae indicate that metabolism can affect the discrimination of excess toxicity from baseline level. Algae growth inhibition is a very good surrogate for fish lethality. This is not only because overall toxicity sensitivity to algae is greater than that to fish, but also the excess toxicity calculated from algal toxicity can better reflect reactivity of compounds with target molecules than fish toxicity. The purpose of this study was to assess selected potentially toxic elements and persistent organic pollutants concentrations in the Shahid Rajaei Dam reservoir as well as their accumulation in barbel and Leuciscus cephalus fish species. The concentration of arsenic, mercury, nickel, chromium and cadmium in all water samples is less than WHO drinking water standard. Chromium shows a higher concentration than WHO standard in both fish species, while nickel and mercury content in barbel fish is higher than WHO standard. The result of principle component analyses indicates an increased elemental concentration due to application of phosphorus fertilizers in the agricultural lands, especially paddy field, ending up to the Tajan River and Shahid Rajaei Dam reservoir. Ni, Co, Mn, Pb and Cr display a quasi-independent behavior within the groups (PC1, PC2 and PC3) reflecting contribution of both geogenic and anthropogenic sources. The concentrations of persistent organic pollutants in all water samples, except M-5 sample, are less than WHO and EPA drinking water standards, while persistent organic pollutants (detected in both fish species) indicate higher contents than those of EPA standard values. Based on carcinogenic health risk value, maximum allowable fish consumption for arsenic is two meals per month. The result of noncarcinogenic health risk of mercury indicates one and three meals consumption per month for barbel and L. cephalus fishes, respectively. The calculation of maximum allowable fish consumption based on carcinogenic health for dieldrin, ∑HCH, heptachlor epoxide, ∑DDT and ∑PCBs reveals monthly meals limitation of both fish species grown in Shahid Rajaei Dam. The modes of toxic action (MOAs) play an important role in the assessment of the ecotoxicity of organic pollutants. However, few studies have been reported on the MOAs in rat toxicity. In this paper, the toxic contributions of functional groups in 1255 aromatic compounds were calculated from regression and were then compared with the toxic contributions in aliphatic compounds. The results show that some functional groups have same toxic contributions both in aromatic and aliphatic compounds, but some have not. To investigate the MOAs in rat toxicity, the distribution of toxic ratio (TR) was examined for well-known baseline and less inert compounds and thresholds of log TR = 0.3 and 0.5 were used to classify baseline, less inert and reactive compounds. The results showed that some compounds identified as baseline compounds in fish toxicity were also classified as baseline compounds in rat toxicity. Except for phenols and anilines which were identified as less inert compounds in fish toxicity, aromatic compounds with functional groups such as ether, nitrile, nitrophenol, isocyanatoe and chloro were identified as less inert chemicals in rat toxicity. Reactive compounds identified in fish toxicity exhibit greater toxicity to rats. These compounds can undergo nucleophilic substitution, acylation and Schiff base formation with biological macromolecules. The critical body residues (CBRs) calculated from absorption and bioconcentration show that log 1/CBRs in rat toxicity are not equal to that in fish for some compounds. The aim of this study was to elucidate how vitamin E (alpha tocopherol) may ameliorate the toxicity of the pesticide chlorpyrifos in Atlantic salmon. Freshly isolated hepatocytes were exposed to vitamin E, chlorpyrifos or a combination of vitamin E and chlorpyrifos (all 100 μM). Transcriptomics (RNA-seq) and metabolomics were used to screen for effects of vitamin E and chlorpyrifos. By introducing vitamin E, the number of upregulated transcripts induced by chlorpyrifos exposure was reduced from 941 to 626, while the number of downregulated transcripts was reduced from 901 to 742 compared to the control. Adding only vitamin E had no effect on the transcriptome. Jak-STAT signaling was the most significantly affected pathway by chlorpyrifos treatment according to the transcriptomics data. The metabolomics data showed that accumulation of multiple long chain fatty acids and dipeptides and amino acids in chlorpyrifos treated cells was partially alleviated by vitamin E treatment. Significant interaction effects between chlorpyrifos and vitamin E were seen for 15 metabolites, including 12 dipeptides. The antioxidant had relatively modest effects on chlorpyrifos-induced oxidative stress. By combining the two data sets, the study suggests that vitamin E supplementation prevents uptake and accumulation of fatty acids, and counteracts inhibited carbohydrate metabolism. Overall, this study shows that vitamin E only to a moderate degree modifies chlorpyrifos toxicity in Atlantic salmon liver cells. [ABSTRACT FROM AUTHOR]

The toxicity induced by insecticides in aquatic organisms is of utmost relevance because it may give a clue about the degree of health or damage of the involved ecosystem. In the present report, we determined the effect of dieldrin (DD) and chlorpyrifos (CP) on the freshwater crayfish, Cambarellus montezumae. The organisms (4-6cm in diameter) were collected in the Ignacio Ramírez Reservoir, situated at 50km Northeast of Mexico City, in the Rio Lerma Basin. Initially, we determined the LC50 value with the Probit method, then the DNA damage with single cell gel electrophoresis (comet assay applied at 24, 48, and 72h of exposure) to the brain and hepatopancreas of animals exposed (in reconstituted water) to 0.05 and 0.5μg/L of each insecticide. In the hepatopancreas of the same organisms, we determined the lipid peroxidation by applying the TBARS test. DNA damage and lipid peroxidation were also evaluated with the same methods to organisms exposed in water from the reservoir. In regard to the LC50, at 72h of exposure, we found a value of 5.1μg/L and a value of 5.62μg/L for DD and CP, respectively. The comet assay applied at different exposure times showed significant DNA damage to both organs, with respect to the control level. In the case of DD, statistical significance was observed for the two doses in the whole evaluated schedule. CP was genotoxic in the brain with the high dose at 72h, and in the hepatopancreas with the two tested doses at all evaluated exposure times. Also, a significant lipid peroxidation increase was detected with the two doses of insecticides. In the study with water from the reservoir, a more pronounced DNA damage was detected. Our results showed strong DNA damage induced by both insecticides in the crayfish, as well as a correlation with the lipid peroxidation effect, suggesting that oxidative stress is involved in the genotoxic alteration. Our results also showed the usefulness of the studied organism as well as the applied tests for the evaluation of toxicological effects, and suggested the pertinence of applying the comet assay to other freshwater organisms to evaluate the bioaccumulation of insecticides. Zebrafish is increasingly used to assess biological properties of chemical substances and thus is becoming a specific tool for toxicological and pharmacological studies. The effects of chemical substances on embryo survival and development are generally evaluated manually through microscopic observation by an expert and documented by several typical photographs. Here, we present a methodology to automatically classify brightfield images of wildtype zebrafish embryos according to their defects by using an image analysis approach based on supervised machine learning. We show that, compared to manual classification, automatic classification results in 90 to 100% agreement with consensus voting of biological experts in nine out of eleven considered defects in 3 days old zebrafish larvae. Automation of the analysis and classification of zebrafish embryo pictures reduces the workload and time required for the biological expert and increases the reproducibility and objectivity of this classification. The article offers information about ciguatera fish poisoning, a marine toxicity that may occur following ingestion of large fish. An overview of the cases of patients who suffered ciguatera fish poisoning is discussed. Information about the pathogenesis, clinical manifestations, differential diagnosis and management of ciguatera fish poisoning is also offered.

Ciguatera fish poisoning (CFP) is a foodborne illness caused by fish containing ciguatoxin (CTX). The toxin is produced by the microalgae Gambierdiscus spp. which are then eaten by reef fish; humans contract the illness when eating either fish that have eaten the algae, or carnivorous fish that have eaten those fish. CTX is an odorless, tasteless, and colorless neurotoxin that blocks voltage-sensitive Na+ channels and accumulates in many tissues of the fish, especially the viscera. The illness is typically mild to moderate in severity with gastrointestinal (diarrhea, cramping, nausea, vomiting) and neurological (paraesthesias, cold allodynia, fatigue, pruritis) manifestations. Rarely, the disease can be more severe with significant neuropathic or cardiac effects such as bradycardia and hypotension. Endemic to Hawai'i and islands throughout the Caribbean and Pacific, CFP incidence rates range from several to thousands of cases per 100,000 per year. Since fishing is important for local food supply, exportation, and recreation throughout the Pacific, CFP is medically and economically significant in these areas. We present a case of CFP from Hawai'i to illustrate the disease, demonstrating that the diagnosis is primarily clinical, with confirmatory tests from fish samples available in some cases. Treatment is supportive and symptomatic with no disease specific remedy. The prognosis for most cases is good with a short duration of self-limited symptoms, but for some cases neurological sequelae can become chronic. With no effective treatment, education on which species of reef fish and which body parts to avoid eating is essential in the prevention of CFP. [ABSTRACT FROM AUTHOR]

Seventy-seven finfish species (300 composites of three fish) were obtained from commercial vendors in six regions of the United States: Great Lakes, mid-Atlantic, New England, northwest, southeast, and southwest. Total mercury in fish muscle tissue ranged from 1 ppb (channel catfish) to 1,425 ppb (king mackerel). Of the top 10 most commonly consumed seafoods in the United States, all finfish species, including salmon species (13 to 62 ppb), Alaskan pollock (11 ppb), tilapia (16 ppb), channel catfish (1 ppb), Atlantic cod (82 ppb), and pangasius (swai) (2 ppb), had low total mercury concentrations. However, two large predatory species, king mackerel and swordfish (1,107 ppb), contained mercury concentrations above the current U.S. Food and Drug Administration action level of 1,000 ppb, indicating that consumers may be unaware that species that are high in mercury are being sold in the marketplace. Twenty-six species of anemonefish of the genera Amphiprion and monospecific Premnas, use only 10 species of anemones as hosts in the wild (Families: Actiniidae, Stichodactylidae and Thalassianthidae). Of these 10 anemone species some are used by multiple species of anemonefish while others have only a single anemonefish symbiont. Past studies have explored the different patterns of usage between anemonefish species and anemone species; however the evolution of this relationship remains unknown and has been little studied over the past decade. Here we reopen the case, comparing the toxicity of crude venoms obtained from anemones that host anemonefish as a way to investigate why some anemone species are used as a host more than others. Specifically, for each anemone species we investigated acute toxicity using Artemia francisca (LC50), haemolytic toxicity using ovine erythrocytes (EC50) and neurotoxicity using shore crabs (Ozius truncatus). We found that haemolytic and neurotoxic activity varied among host anemone species. Generally anemone species that displayed greater haemolytic activity also displayed high neurotoxic activity and tend to be more toxic on average as indicated by acute lethality analysis. An overall venom toxicity ranking for each anemone species was compared with the number of anemonefish species that are known to associate with each anemone species in the wild. Interestingly, anemones with intermediate toxicity had the highest number of anemonefish associates, whereas anemones with either very low or very high toxicity had the fewest anemonefish associates. Effect concentrations in the toxicity assessment of chemicals with fish and fish cells are generally based on external exposure concentrations. External concentrations as dose metrics, may, however, hamper interpretation and extrapolation of toxicological effects because it is the internal concentration that gives rise to the biological effective dose. Thus, we need to understand the relationship between the external and internal concentrations of chemicals. The objectives of this study were to: (i) elucidate the time-course of the concentration of chemicals with a wide range of physicochemical properties in the compartments of an in vitro test system, (ii) derive a predictive model for toxicokinetics in the in vitro test system, (iii) test the hypothesis that internal effect concentrations in fish (in vivo) and fish cell lines (in vitro) correlate, and (iv) develop a quantitative in vitro to in vivo toxicity extrapolation method for fish acute toxicity. To achieve these goals, time-dependent amounts of organic chemicals were measured in medium, cells (RTgill-W1) and the plastic of exposure wells. Then, the relation between uptake, elimination rate constants, and log KOW was investigated for cells in order to develop a toxicokinetic model. This model was used to predict internal effect concentrations in cells, which were compared with internal effect concentrations in fish gills predicted by a Physiologically Based Toxicokinetic model. Our model could predict concentrations of non-volatile organic chemicals with log KOW between 0.5 and 7 in cells. The correlation of the log ratio of internal effect concentrations in fish gills and the fish gill cell line with the log KOW was significant (r>0.85, p = 0.0008, F-test). This ratio can be predicted from the log KOW of the chemical (77% of variance explained), comprising a promising model to predict lethal effects on fish based on in vitro data. The contamination of aquatic ecosystem with heavy metals and pesticides has increased dramatically over the years, and this has generated a lot of attention by environmental experts. There are reports on impacts of different single toxicants on aquatic organisms but not much has been done to investigate the impacts of mixtures of toxicants on aquatic life. In this study we investigated the effects of multiple stressors of Pb and cypermethrin on ionoregulation in the African catfish (Clarias gariepinus). Adults C. gariepinus were exposed to either borehole water (control), 2 mg/L Pb, 0.5 μg/L cypermetrin or a mixture of 2 mg/L Pb and 0.5 μg/L cypermethrin for 96 h, after which the levels of plasma Na+, Cl- and K+ were determined as a measure of branchial ionoregulatory disturbance. The exposure of C. gariepinus to 0.5 μg/L cypermethrin resulted in a significant reduction in plasma Na+ (43% decrease) and Cl- (31% decrease) but a 2-fold increase in plasma K+ concentrations. Exposure to a mixture of cypermethrin and Pb resulted in 53% decrease in plasma Na+, 40% decrease in plasma Cl- and a 3- fold increase in plasma K+. There was no significant difference between the control and those exposed to just only 2 mg/L Pb for all the parameters measured. The results of this study indicated that exposure of aquatic organisms to combined stressors of heavy metals pollution as well as pesticide contaminantion could have a serious effect on the physiology especially on ionic regulation in the exposed fish.
