Roads impact a disproportionately larger area than the land area that they occupy, often called the road effect zone. This effect extends to wildlife populations. Impacts on federally listed species include habitat loss and fragmentation, road mortality, the barrier effect, and the habituation effect. Per the Endangered Species Act, these impacts must be assessed before roadway impacts occur. This publication provides an overview of the potential impacts to 29 of Florida’s threatened and endangered animal species from roads. Potential mitigation options include road configuration, signage, speed limits, habitat protection, improved technology, wildlife fencing, and wildlife crossing structures. The goal is to educate transportation engineers and planners to allow for sustainable transportation infrastructure growth while protecting, conserving, and ultimately recovering Florida’s threatened and endangered species.

At least nine populations of non-human primates have established in Florida since the 1930s. These include three species: rhesus macaques (Macaca mulatta), vervet monkeys (Chlorocebus sabaeus), and squirrel monkeys (Saimiri sp.). Invasion success has varied among the three species. As of 2021, a single, growing population of rhesus macaques was present in central Florida; a single, stable population of vervet monkeys was present in south Florida; and only two squirrel monkeys remained of a previous population on a private property in south Florida. Demonstrated and potential impacts of the three species also varied. Continued monitoring and research of introduced primate populations in Florida is critical to better understand impacts and management needs.

In this document we introduce a standardized set of seven terms compiled by the members of the UF|IFAS Invasive Species Council that can describe most situations involving invasive species. We do so to promote a common language among Florida’s Extension and outreach professionals. This will limit confusion caused by excessive jargon used to describe invasive species and the fact that much of this jargon is used incorrectly. This confusion hinders stakeholder understanding of environmental and economic impacts caused by invasive species which can prevent stakeholders from adopting behaviors that can both limit the introduction of new invasive species and decrease the impacts of those already established. The seven terms we propose are “native”, “nonnative”, “introduced”, “established”, “invasive”, “nuisance”, and “range change”. We also list six terms to avoid due to their tendency to create confusion and/or be misused and misinterpreted (“native invasive”, “invasive exotic”, “invasive weed”, “alien”, “foreign”, and “nonindigenous”) as well as precise definitions of common legal terms (“noxious weed”, “injurious”, “prohibited”, and “conditional/restricted”). By speaking this common language, Florida’s Extension and outreach professionals can deliver a simplified and unified message about the impacts of invasive species and how to best prevent and control them, potentially increasing the environmental and socioeconomic benefits of invasive species Extension and outreach programs.

Frogs control garden pests such as insects and slugs, and serve as a food source for many larger wildlife species. Research on the substances frogs secrete through their skins has even led to the creation of new painkillers and antibiotics. Most frogs in Florida are reclusive and harmless to people, but two species of frogs that have invaded Florida can be harmful to humans and their pets. This 4-page fact sheet presents some facts about native frogs, describes the problems invasive frogs cause, and provides suggestions on how to cope with problem frogs. Background. Childhood arterial ischemic stroke (AIS) is rare and may be difficult to diagnose. Management of acute stroke in any age group is time sensitive, so awareness of the manifestations and appropriate diagnostic procedures for pediatric AIS is vital to establishing care. We present a pediatric case of arterial ischemic stroke that presented to the emergency department (ED) after two seizures. Case Report. A five-year-old female with an existing seizure disorder presented to a pediatric ED after having two seizures. Postictal upon arrival, she underwent a computed tomography (CT) scan of her head. Family reported that she had complained of a severe headache and vomited; her seizures were described as different from those she had experienced in the past. Loss of grey white matter differentiation on the CT warranted magnetic resonance imaging (MRI), which demonstrated a right-sided stroke. After a complicated course in the hospital, the patient was discharged to a rehabilitation hospital. Why Should an Emergency Physician Be Aware of This? It is important that emergency physicians recognize that a seizure may be the initial symptom of a pediatric stroke regardless of an established seizure history. Pediatric seizures are relatively common; however consideration of the diagnosis of pediatric stroke may prevent unnecessary delays in treatment.

Abstract Background Nucleosome-mediated chromatin compaction has a direct effect on the accessibility of trans-acting activators and repressors to DNA targets and serves as a primary regulatory agent of genetic expression. Understanding the nature and dynamics of chromatin is fundamental to elucidating the mechanisms and factors that epigenetically regulate gene expression. Previous work has shown that there are three types of canonical sequences that strongly regulate nucleosome positioning and thus chromatin accessibility: putative nucleosome-positioning elements, putative nucleosome-repelling sequences, and homopolymeric runs of A/T. It is postulated that these elements can be used to remodel chromatin in C. elegans. Here we show the utility of such elements in vivo, and the extreme efficacy of a newly discovered repelling sequence, PRS-322. Results In this work, we show that it is possible to manipulate nucleosome positioning in C. elegans solely using canonical and putative positioning sequences. We have not only tested previously described sequences such as the Widom 601, but also have tested additional nucleosome-positioning sequences: the Trifonov sequence, putative repelling sequence-322 (PRS-322), and various homopolymeric runs of A and T nucleotides. Conclusions Using each of these types of putative nucleosome-positioning sequences, we demonstrate their ability to alter the nucleosome profile in C. elegans as evidenced by altered nucleosome occupancy and positioning in vivo. Additionally, we show the effect that PRS-322 has on nucleosome-repelling and chromatin remodeling.

Abstract The layered transition-metal dichalcogenide WTe2 is characterized by distinctive transport and topological properties. These properties are largely determined by electronic states close to the Fermi level, specifically to electron and hole pockets in the Fermi sea. In principle, these states can be manipulated by changes to the crystal structure. The precise impact of particular structural changes on the electronic properties is a strong function of the specific nature of the atomic displacements. Here, we report on time-resolved X-ray diffraction and infrared reflectivity measurements of the coherent structural dynamics in WTe2 induced by femtosecond laser pulses excitation (central wavelength 800 nm), with emphasis on a quantitative description of both in-plane and out-of-plane vibrational modes. We estimate the magnitude of these motions, and calculate via density functional theory their effect on the electronic structure. Based on these results, we predict that phonons periodically modulate the effective mass of carriers in the electron and hole pockets up to 20%. This work opens up new opportunities for modulating the peculiar transport properties of WTe2 on short time scales.

The use of intense light to control transient properties of quantum materials is a subject of current interest in ultrafast condensed matter physics. Here, a new up-conversion pathway for the optical control of Raman-active modes is demonstrated via a terahertz field-driven nonlinear process in the strongly-correlated metal, V2O3.

Gas chromatography-electroantennographic detection (GC-EAD), typically combined with a flame ionization detector (FID), is a widely used technique for studying insect semiochemicals. Combining mass spectrometry (MS) with GC-EAD is extremely beneficial, but has seldom been adopted, possibly due to the practical challenges associated with incorporating an MS (under vacuum) into a GC-EAD system. We describe a novel method of incorporating MS into a GC-EAD system, where the FID is maintained in the system rather than being replaced by an MS. With this method, effluent is still split between EAD and FID, with the FID being used to assign components that elicit antennal responses, as in a normal GC-FID/EAD system. The MS is introduced via a second effluent splitter inserted before the split between the EAD and the FID. This method allows for EAD, FID, and MS data to be obtained from a single injection, which is especially useful for analysis of thermally desorbed and solid-phase microextraction (SPME) samples. This configuration is also relatively simple to implement and resolves some of the practical challenges associated with dividing effluent between a detector at atmospheric pressure (a live antenna) and a detector under vacuum (the MS). We present test runs with hawkmoth antennae and floral volatiles to demonstrate the effectiveness of this system, and discuss the challenges and practical solutions to incorporating MS into a GC-EAD system that retains an FID detector.

Marine protected areas (MPAs) are ubiquitous in global ocean conservation and play a pivotal role in achieving local, national, and regional area-based conservation targets. Often, such targets are merely met on “paper” and lack the political or managerial resources to produce positive conservation outcomes. Here, we apply the MPA Guide – a framework for assessing the quantity and quality of marine protected areas – to Guam and the Commonwealth of the Northern Mariana Islands (CNMI), two U.S. territories in the Western Pacific. We reviewed the enabling legislation and applicable management documents for all MPAs (n=18). We found that all but three (3) MPAs in the Mariana Islands are actively managed, and these areas are either fully or highly protected – the highest tier of the MPA Guide. Lightly protected areas are associated with high use/high-density tourism activities. Total area protected varies at the jurisdictional scale: 0.83% of Guam’s territorial waters (out to 12 nm) and 23.73% of EEZ under some spatial management; CNMI has 20.39% of territorial waters and 25.91% of EEZ in MPAs. These results emphasize the importance of quality, quantity, and scale when determining effective conservation, especially in overlapping and contested jurisdictional authority areas.

One of the fundamental problems in continental rift segmentation and propagation is how strain is accommodated along large rift-bounding faults (border faults) since the segmentation of propagating border faults control the expression of rift zones, syn-rift depo-centers, and long-term basin evolution. In the Southern Malawi Rift, where previous studies on the early-stage rifting only assessed border fault structure from surficial and topographic expression, we integrate surface and subsurface data to investigate border fault segmentation, linkage, and growth as proxies for strain accommodation along the Bilila-Mtakataka Fault (BMF) System. We used 30 m-resolution topographic relief maps, electrical resistivity tomography (ERT), and high-resolution aeromagnetic data to characterize the detailed fault geometry and provide a more robust estimate of along-fault displacement distribution. Our results reveal a discrepancy between sub-aerial segmentation of the BMF geometry (six segments), scarp height (five segments) reflecting the most recent episodes of fault offset, and cumulative throw (three composite segments) reflecting the long-term fault offset. We also observe that although the BMF exhibits continuity of sub-aerial scarps along its length, the throw distribution shows a higher estimate at the Northern-to-Central segment relay zone (423 m absolute, 364 m moving median) compared to the Central-to-Southern segment relay zone (371 m absolute, 297 m moving median). The ERT profiles across the relay zones suggest a shallower basement and a possible canyon-mouth alluvial fan stratigraphy at the Central-to-Southern segment relay zone, contrasting the deeper basement and “simpler” electrical stratigraphy at the Northern-to-Central relay. The results suggest a more complex long-term evolution of the BMF than was assumed in previous studies. A comparison of BMF’s maximum displacement-vs-length with those of other Malawi Rift border faults and global normal fault populations suggest that although the BMF has possibly reached its maximum length, it remains largely under-displaced as its 580–837 m maximum displacement is significantly lower than that of faults of equivalent length. We suggest that the BMF may continue to accrue significant strain as tectonic extension progresses in the Southern Malawi Rift, thus posing a major seismic hazard in the region.

von Humboldt’s tree-line concept has dominated mountain ecology for almost two hundred years, and is considered a key indicator for monitoring change in biome boundaries and biodiversity shifts under climate change. Even though the concept of life zones and elevation gradients are a globally observed phenomenon, they have not been thoroughly explored for many contexts. One such example is the tree-line ecotone, a widely used conceptual tool to track climate change in many regions, which has limited application in the widespread tree-sparse, grassy systems that comprise a third of the world’s mountain systems. Among grasses (Poaceae), temperature is linked to variation in photosynthetic performance and community dominance for C3 and C4 metabolic groups, due to its role in limiting photorespiration in the C3 photosynthesis process. Here, we investigate this community shift in grassland-dominated mountains to demonstrate the role of climate in driving this transition and discuss the potential applications of this tool to mountain ecosystem conservation worldwide. For identifying grass-dominated mountains worldwide, we measured the grass-cover using satellite data. We then compiled Poaceae distribution data for ten grass-dominated mountains spanning from 42°S to 41°N and determined the temperature intervals and elevation ranges at which each genus was found, testing for effects of temperature, precipitation, and latitudinal gradients on the dominance of C3-C4 grasses. Temperature was the main driver of C3 dominance, with the richness of C3 genera tending to surpass the taxonomic dominance of C4 plants along mountain temperature gradients where the annual mean temperature was colder than ca. 14.6°C. Similar patterns were observed in eight out of ten mountains, suggesting that this may constitute an isotherm-driven ecotone. Consequently, this C3-C4 transition offers a promising tool for monitoring climate change impacts in grassy mountains. C3-C4 grass community shifts in response to environmental change will likely have major implications for fire frequency and severity, rangeland productivity and livelihoods, food security, and water budgets in mountain systems. Given the severity of the implications of global change on these social-ecological systems, we propose that a “grass-line” monitoring protocol be developed for global application.

Pharmacogenomics (PGx) studies how a person’s genes affect the response to medications and is quickly becoming a significant part of precision medicine. The clinical application of PGx principles has consistently been cited as a major opportunity for improving therapeutic outcomes. Several recent studies have demonstrated that most individuals (> 90%) harbor PGx variants that would be clinically actionable if prescribed a medication relevant to that gene. In multiple well-conducted studies, the results of PGx testing have been shown to guide therapy choice and dosing modifications which improve treatment efficacy and reduce the incidence of adverse drug reactions (ADRs). Although the value of PGx testing is evident, its successful implementation in a clinical setting presents a number of challenges to molecular diagnostic laboratories, healthcare systems, providers and patients. Different molecular methods can be applied to identify PGx variants and the design of the assay is therefore extremely important. Once the genotyping results are available the biggest technical challenge lies in turning this complex genetic information into phenotypes and actionable recommendations that a busy clinician can effectively utilize to provide better medical care, in a cost-effective, efficient and reliable manner. In this paper we describe a successful and highly collaborative implementation of the PGx testing program at the University of Minnesota and MHealth Fairview Molecular Diagnostic Laboratory and selected Pharmacies and Clinics. We offer detailed descriptions of the necessary components of the pharmacogenomic testing implementation, the development and technical validation of the in-house SNP based multiplex PCR based assay targeting 20 genes and 48 SNPs as well as a separate CYP2D6 copy number assay along with the process of PGx report design, results of the provider and pharmacists usability studies, and the development of the software tool for genotype-phenotype translation and gene-phenotype-drug CPIC-based recommendations. Finally, we outline the process of developing the clinical workflow that connects the providers with the PGx experts within the Molecular Diagnostic Laboratory and the Pharmacy.

Topological photonic structures can be understood by solving the eigenvalue problem of Maxwell’s equations in the static case. Here, the authors study Floquet topological phases in nonlinear photonic crystals under external drive and show how non-reciprocal transport can be achieved in a Floquet Chern insulator.

Despite increased focus on elucidating the various reproductive strategies employed by orchids, we still have only a rather limited understanding of deceptive pollination systems that are not bee- or wasp-mediated. In Europe, the orchid Neotinea ustulata has been known to consist of two phenologically divergent varieties, neither of which provide rewards to its pollinators. However, detailed studies of their reproductive biology have been lacking. Our study aimed to characterize and understand the floral traits (i.e., morphology, color, and scent chemistry) and reproductive biology of N. ustulata. We found that the two varieties differ in all their floral traits; furthermore, while Neotinea ustulata var. ustulata appears to be pollinated by both bees (e.g., Anthophora, Bombus) and flies (e.g., Dilophus, Tachina), var. aestivalis is pollinated almost entirely by flies (i.e., Nowickia, Tachina). Tachinids were also found to be much more effective than bees in removing pollinaria, and we show experimentally that they use the characteristic dark inflorescence top as a cue for approaching inflorescences. Our results thus suggest that while both N. ustulata varieties rely on tachinids for pollination, they differ in their degree of specialization. Further studies are, however, needed to fully understand the reproductive strategy of N. ustulata varieties.

Geographical variation in flower color of a plant species may reflect the outcome of selection by pollinators or may reflect abiotic factors such as soil chemistry or neutral processes such as genetic drift. Here we document striking geographical structure in the color of capitula of the endemic South African grassland daisy Gerbera aurantiaca and ask which of these competing explanations best explains this pattern. The color of capitula ranges from predominantly red in the southwest to yellow in the center, with some northern populations showing within-population polymorphism. Hopliine scarab beetles were the most abundant flower visitors in all populations, apart from a yellow-flowered one where honeybees were frequent. In a mixed color population, yellow, orange and red morphs were equally attractive to hopliine beetles and did not differ significantly in terms of fruit set. Beetles were attracted to both red and yellow pan traps, but preferred the latter even at sites dominated by the red morph. We found no strong associations between morph color and abiotic factors, including soil chemistry. Plants in a common garden retained the capitulum color of the source population, even when grown from seed, suggesting that flower color variation is not a result of phenotypic plasticity. These results show that flower color in G. aurantiaca is geographically structured, but the ultimate evolutionary basis of this color variation remains elusive.

The workplace provides an important delivery point for health promotion, yet many programs fail to engage men. A gender-sensitive 8-week team challenge-based intervention targeting increased physical activity was delivered at a petrochemical worksite. The purpose of this study was to examine men’s pre–post physical activity and sleep following the intervention, as well as to explore program acceptability and gather men’s recommendations for health promotion. Pre–post surveys assessed physical activity, sleep, program exposure, acceptability, and suggestions for continued support. Overall, 328 men completed baseline surveys and 186 (57%) completed follow-up surveys. Walking increased by 156.5 min/week, 95% confidence interval (61.2, 251.8), p = .001. Men with higher program exposure increased moderate and vigorous activity 49.4 min more than those with low exposure ( p = .026). Sleep duration and quality were higher postintervention, though changes were modest. Program acceptability was high as was intention to maintain physical activity. Men’s suggestions to enable physical activity involved workplace practices/resources, reducing workload, and leadership support. These findings suggest that a gender-sensitive physical activity workplace intervention showed promise for improving physical activity and sleep among men. The men’s suggestions reflected workplace health promotion strategies, reinforcing the need for employers to support ongoing health promotion efforts.

The GroES/GroEL chaperonin from E. coli

South Africa is a megadiverse country with three globally recognised biodiversity hotspots within its borders. Bees in particular show high diversity and endemism in the western part of the country. Not much is currently known about the floral host preferences of indigenous bees in South Africa, with data only available from observational studies. Pollen metabarcoding provides provenance information by utilising DNA analyses instead of floral visitation and traditional microscopic identification to identify pollinator food plants, which can be time consuming and imprecise. In this study, we sampled pollen from leaf-cutter bees (Megachilidae) specimens maintained in a historic insect collection (National Collection of Insects, South Africa) that were originally collected from two florally important areas in South Africa (Succulent Karoo and Savanna) and used metabarcoding to determine pollen provenance. We also sampled pollen from leafcutter bee species with wider distributions, that extend across many different biomes, to determine if these 'generalist' species show relaxed floral host specificity in some biomes. Metabarcoding involved sequencing of the nuclear internal transcribed spacer 2 (ITS2) region. Amplicons were compared to a sequence reference database to assign taxonomic classifications to family level. Sequence reads were also clustered to OTUs based on 97% sequence similarity to estimate numbers of plant species visited. We found no significant difference in the mean number of plant taxa visited in the Succulent Karoo and Savanna regions, but the widespread group visited significantly more floral hosts. Bees from the widespread group were also characterised by a significantly different composition in pollen assemblage. The time since specimens were collected did not have an effect on the mean number of taxa visited by any of the bee species studied. This study highlights national history collections as valuable sources of temporal and spatial biodiversity data.

The most basic level of eukaryotic gene regulation is the presence or absence of nucleosomes on DNA regulatory elements. In an effort to elucidate in vivo nucleosome patterns, in vitro studies are frequently used. In vitro, short DNA fragments are more favorable for nucleosome formation, increasing the likelihood of nucleosome occupancy. This may in part result from the fact that nucleosomes prefer to form on the terminal ends of linear DNA. This phenomenon has the potential to bias in vitro reconstituted nucleosomes and skew results. If the ends of DNA fragments are known, the reads falling close to the ends are typically discarded. In this study we confirm the phenomenon of end bias of in vitro nucleosomes. We describe a method in which nearly identical libraries, with different known ends, are used to recover nucleosomes which form towards the terminal ends of fragmented DNA. Finally, we illustrate that although nucleosomes prefer to form on DNA ends, it does not appear to skew results or the interpretation thereof.

Past performance is a key consideration when rationalising the allocation of grants and other opportunities to individual researchers. The National Research Foundation of South Africa (NRF) has long used a highly structured system of ‘rating’ the past performance of individual researchers. This system relies heavily on peer review, and has seldom been benchmarked against bibliometric measures of research performance such as Hirsch’s h-index. Here I use data for about 600 rated researchers in the biological sciences to evaluate the extent to which outcomes of peer review correspond to bibliometric measures of research performance. The analysis revealed that values of the h-index based on the Scopus database are typically 5–20 for researchers placed in the NRF’s C rating category (‘established’), 20–40 for those in the B rating category (‘considerable international recognition’) and >40 for those in the A rating category (‘leading international scholars’). Despite concerns that citation patterns differ among disciplines, the mean h-index per rating category was remarkably consistent across five different disciplines in the biological sciences, namely animal sciences, plant sciences, ecology, microbiology and biochemistry/genetics. This observation suggests that the NRF rating system is equitable in the sense that the outcomes of peer review are generally consistent with bibliometric measures of research performance across different disciplines in the biological sciences. However, the study did reveal some notable discrepancies which could reflect either bias in the peer-review process or shortcomings in the bibliometric measures, or both. Significance: • NRF rating categorisations (estimates of standing in a research field based on peer review) are a reasonably good predictor of the h-index of individual researchers in the biological sciences. • The relationships between rating categorisations and the h-index are remarkably consistent across five sub-disciplines in the biological sciences. • Peer review and the h-index were compared in terms of their relative advantages and disadvantages and the combined use of both approaches is advocated for measurement of research performance.

Abstract Western honeybees (Apis mellifera L.), native to Europe and Africa, have been transported worldwide and are now one of the most important global crop pollinator species. Although the relative contribution of honeybees to global crop pollination is increasingly recognized, relatively little is known about their importance as pollinators in wild plant communities. The only remaining wild and unmanaged western honeybee populations are in Africa. We investigated the importance of honeybees as pollinators of diverse wild plant communities in two protected areas within the Maputaland–Pondoland–Albany biodiversity hotspot in South Africa. Sites were far from any known areas of beekeeping, and so all honeybees were most likely from wild colonies. Honeybees visited a large proportion of flowering plant species within these two communities (40% and 35%) and also provided a substantial proportion of visits to the plants they visited (40% and 32%, respectively). However, when pollinator importance indices (based on abundance and the size and purity of pollen loads) were calculated for a small subset of plants, honeybees were only important pollinators of 29% of the plants they visited. Our data provide a first step in determining the importance of honeybees as flower visitors and pollinators in wild plant communities and the potential impacts of honeybee declines on these highly diverse grassland ecosystems. Our work suggests that many plants in the grassland systems studied are visited by non‐Apis flower visitors and therefore that conservation efforts should also focus on these pollinator groups.

An outbreak of bacterial soft rot and blackleg of potato has occurred since 2014 with the epicenter being in the northeastern region of the United States. Multiple species of Pectobacterium and Dickeya are causal agents, resulting in losses to commercial and seed potato production over the past decade in the Northeastern and North Central United States. To clarify the pathogen present at the outset of the epidemic in 2015 and 2016, a phylogenetic study was made of 121 pectolytic soft rot bacteria isolated from symptomatic potato; also included were 27 type strains of Dickeya and Pectobacterium species, and 47 historic reference strains. Phylogenetic trees constructed based on multilocus sequence alignments of concatenated dnaJ, dnaX and gyrB fragments revealed the epidemic isolates to cluster with type strains of D. chrysanthemi, D. dianthicola, D. dadantii, P. atrosepticum, P. brasiliense, P. carotovorum, P. parmentieri, P. polaris, P. punjabense, and P. versatile. Genetic diversity within D. dianthicola strains was low, with one sequence type (ST1) identified in 17 of 19 strains. Pectobacterium parmentieri was more diverse, with ten sequence types detected among 37 of the 2015–2016 strains. This study can aid in monitoring future shifts in potato soft rot pathogens within the U.S. and inform strategies for disease management.

Elaborated kettle trap flowers to temporarily detain pollinators evolved independently in several angiosperm lineages. Intensive research on species of Aristolochia and Ceropegia recently illuminated how these specialized trap flowers attract particular pollinators through chemical deception. Morphologically similar trap flowers evolved in Riocreuxia; however, no data about floral rewards, pollinators, and chemical ecology were available for this plant group. Here we provide data on pollination ecology and floral chemistry of R. torulosa. Specifically, we determined flower visitors and pollinators, assessed pollen transfer efficiency, and analysed floral scent chemistry. R. torulosa flowers are myiophilous and predominantly pollinated by Nematocera. Pollinating Diptera included, in order of decreasing abundance, male and female Sciaridae, Ceratopogonidae, Scatopsidae, Chloropidae, and Phoridae. Approximately 16% of pollen removed from flowers was successfully exported to conspecific stigmas. The flowers emitted mainly ubiquitous terpenoids, most abundantly linalool, furanoid (Z)-linalool oxide, and (E)-β-ocimene—compounds typical of rewarding flowers and fruits. R. torulosa can be considered to use generalized food (and possibly also brood-site) deception to lure small nematocerous Diptera into their flowers. These results suggest that R. torulosa has a less specific pollination system than previously reported for other kettle trap flowers but is nevertheless specialized at the level of Diptera suborder Nematocera.

Dickeya and Pectobacterium spp. both cause blackleg and soft rot of potato, which can be a yield-reducing factor to potato production. The purpose of this study was to examine the interaction between these two bacterial genera causing potato infection, and subsequent disease development and yield responses under field conditions. Analysis of 883 potato samples collected in Northeastern USA using polymerase chain reaction determined that Dickeya dianthicola and P. parmentieri were found in 38.1% and 53.3% of all samples, respectively, and that 20.6% of samples contained both D. dianthicola and P. parmentieri. To further investigate the relationship between the two bacterial species and their interaction, field trials were established. Potato seed pieces of “Russet Burbank”, “Lamoka”, and “Atlantic” were inoculated with bacterial suspension of D. dianthicola at 107 colony-forming unite (CFU)/mL using a vacuum infiltration method, air dried, and then planted in the field. Two-year results showed that there was a high correlation (p < 0.01) between yield loss and percent of inoculated seed pieces. In a secondary field trial conducted in 2018 and 2019, seed pieces of potato “Shepody”, “Lamoka” and “Atlantic” were inoculated with D. dianthicola, P. parmentieri, or mixture of both species, and then planted. In 2019, disease severity index, as measured by the most sensitive variety “Lamoka”, was 16.2 with D. dianthicola inoculation, 10.4 with P. parmentieri, 25.4 with inoculation with both bacteria. Two-year data had a similar trend. Thus, D. dianthicola was more virulent than P. parmentieri, but the co-inoculation of the two species resulted in increased disease severity compared to single-species inoculation with either pathogen.

A critical area of emphasis for science educators is the identification of effective means of teaching and engaging undergraduate students. Personal microbiome analysis is a means of identifying the microbial communities found on or in our body. We hypothesized the use of personal microbiome analysis in the classroom could improve science education by making courses more applied and engaging for undergraduate students. We determined to test this prediction in three Brigham Young University undergraduate courses: Immunology, Advanced Molecular Biology Laboratory, and Genomics. These three courses have a two-week microbiome unit and students during the 2016 semester students could submit their own personal microbiome kit or use the demo data, whereas during the 2017 semester students were given access to microbiome data from an anonymous individual. The students were surveyed before, during, and after the human microbiome unit to determine whether analyzing their own personal microbiome data, compared to analyzing demo microbiome data, impacted student engagement and interest. We found that personal microbiome analysis significantly enhanced the engagement and interest of students while completing microbiome assignments, the self-reported time students spent researching the microbiome during the two week microbiome unit, and the attitudes of students regarding the course overall. Thus, we found that integrating personal microbiome analysis in the classroom was a powerful means of improving student engagement and interest in undergraduate science courses.
