1. Enzymatic conversion of human blood group A kidneys to universal blood group O

Abstract

Abstract ABO blood group compatibility restrictions present the first barrier to donor-recipient matching in kidney transplantation. Here, we present the use of two enzymes, FpGalNAc deacetylase and FpGalactosaminidase, from the bacterium Flavonifractor plautii to enzymatically convert blood group A antigens from the renal vasculature of human kidneys to ‘universal’ O-type. Using normothermic machine perfusion (NMP) and hypothermic machine perfusion (HMP) strategies, we demonstrate blood group A antigen loss of approximately 80% in as little as 2 h NMP and HMP. Furthermore, we show that treated kidneys do not bind circulating anti-A antibodies in an ex vivo model of ABO-incompatible transplantation and do not activate the classical complement pathway. This strategy presents a solution to the donor organ shortage crisis with the potential for direct clinical translation to reduce waiting times for patients with end stage renal disease.

Patient-reported symptoms are critical for diagnosis and evaluation of treatment benefit in DG. The Diabetic Gastroparesis Symptom Severity Diary (DGSSD), a new patient-reported outcome measure, was developed for use in clinical trials to support product approval and labeling claims for DG treatments. Materials and methods: Initial DGSSD development was based on a review of the existing instruments and qualitative research (focus groups and cognitive debriefing interviews) in 41 patients with DG. Psychometric evaluations (individual items and composite scores) were conducted using data from Phase IIa and IIb relamorelin clinical trials. Results: Qualitative research in patients with DG resulted in a six-item DGSSD, included in the Phase IIa trial, addressing symptom severity for nausea, vomiting, abdominal pain, early satiety, and bloating, as well as vomiting frequency. An item addressing severity of postprandial fullness (PPF) was subsequently added based on regulatory advice and included in the Phase IIb trial. Measurement properties were generally strong for weekly averages of daily item and composite scores. Item-level intraclass correlation coefficients ranged from 0.79 to 0.97 and correlations with other measures matched hypothesized patterns; the discriminating ability and responsiveness of the DGSSD were also supported. Multiple methods supported the computation of a composite score based on items addressing nausea, abdominal pain, bloating, and PPF severity. Conclusion: Qualitative and quantitative evidence support use of the DGSSD as a reliable and valid measure from which to derive endpoints to evaluate treatment benefit in future DG interventional trials. 5. Preparation of $^{87}$Rb and $^{133}$Cs in the motional ground state of a single optical tweezer

Abstract

We report simultaneous Raman sideband cooling of a single $^{87}$Rb atom and a single $^{133}$Cs atom held in separate optical tweezers at 814\,nm and 938\,nm, respectively. Starting from outside the Lamb-Dicke regime, after 45\,ms of cooling we measure probabilities to occupy the three-dimensional motional ground state of 0.86$^{+0.03}_{-0.04}$ for Rb and 0.95$^{+0.03}_{-0.04}$ for Cs. Our setup overlaps the Raman laser beams used to cool Rb and Cs, reducing hardware requirements by sharing equipment along the same beam path. The cooling protocol is scalable, and we demonstrate cooling of single Rb atoms in an array of four tweezers. After motional ground-state cooling, a 938\,nm tweezer is translated to overlap with a 814\,nm tweezer so that a single Rb and a single Cs atom can be transferred into a common 1064\,nm trap. By minimising the heating during the merging and transfer, we prepare the atoms in the relative motional ground state with an efficiency of 0.81$^{+0.08}_{-0.08}$. This is a crucial step towards the formation of single RbCs molecules confined in optical tweezer arrays.

6. Feshbach Spectroscopy of Cs Atom Pairs in Optical Tweezers

Abstract

We prepare pairs of $^{133}$Cs atoms in a single optical tweezer and perform Feshbach spectroscopy for collisions of atoms in the states $(f=3, m_f=\pm3)$. We detect enhancements in pair loss using a detection scheme where the optical tweezers are repeatedly subdivided. For atoms in the state $(3,-3)$, we identify resonant features by performing inelastic loss spectroscopy. We carry out coupled-channel scattering calculations and show that at typical experimental temperatures the loss features are mostly centred on zeroes in the scattering length, rather than resonance centres. We measure the number of atoms remaining after a collision, elucidating how the different loss processes are influenced by the tweezer depth. These measurements probe the energy released during an inelastic collision, and thus give information on the states of the collision products. We also identify resonances with atom pairs prepared in the absolute ground state $(f=3, m_f=3)$, where two-body radiative loss is engineered by an excitation laser blue-detuned from the Cs D$_2$ line. These results demonstrate optical tweezers to be a versatile tool to study two-body collisions with number-resolved detection sensitivity., Comment: 18 pages, 6 figures

Abstract

We report the preparation of exactly one $^{87}$Rb atom and one $^{133}$Cs atom in the same optical tweezer as the essential first step towards the construction of a tweezer array of individually trapped $^{87}$Rb$^{133}$Cs molecules. Through careful selection of the tweezer wavelengths, we show how to engineer species-selective trapping potentials suitable for high-fidelity preparation of Rb $+$ Cs atom pairs. Using a wavelength of 814~nm to trap Rb and 938~nm to trap Cs, we achieve loading probabilities of $0.508(6)$ for Rb and $0.547(6)$ for Cs using standard red-detuned molasses cooling. Loading the traps sequentially yields exactly one Rb and one Cs atom in $28.4(6)\,\%$ of experimental runs. Using a combination of an acousto-optic deflector and a piezo-controlled mirror to control the relative position of the tweezers, we merge the two tweezers, retaining the atom pair with a probability of $0.99^{(+0.01)}_{(-0.02)}$. We use this capability to study hyperfine-state-dependent collisions of Rb and Cs in the combined tweezer and compare the measured two-body loss rates with coupled-channel quantum scattering calculations., Comment: 22 pages, 6 figures

Abstract

In quantum optomechanics, finding materials and strategies to limit losses has been crucial to the progress of the field. Recently, superfluid 4He was proposed as a promising mechanical element for quantum optomechanics. This quantum fluid shows highly desirable properties (e.g. extremely low acoustic loss) for a quantum optomechanical system. In current implementations, superfluid optomechanical systems suffer from external sources of loss, which spoils the quality factor of resonators. In this work, we propose a new implementation, exploiting nanofluidic confinement. Our approach, based on acoustic resonators formed within phononic nanostructures, aims at limiting radiation losses to preserve the intrinsic properties of superfluid 4He. In this work, we estimate the optomechanical system parameters. Using recent theory, we derive the expected quality factors for acoustic resonators in different thermodynamic conditions. We calculate the sources of loss induced by the phononic nanostructures with numerical simulations. Our results indicate the feasibility of the proposed approach in a broad range of parameters, which opens new prospects for more complex geometries., Comment: 20 pages, 19 figures

The properties of $\widehat{V}_m$ are discussed and contrasted with those of the standard modular value of a projector. The associated notion of a faux qubit is introduced and its possible utility in quantum computation is noted. 13. Experimental Evidence for a Dynamical Non-Locality Induced Effect in Quantum Interference Using Weak Values

Abstract

The quantum theoretical concepts of modular momentum and dynamical non-locality, which were introduced four decades ago, have recently been used to explain single particle quantum interference phenomena. Although the non-local exchange of modular momentum associated with such phenomena cannot be directly observed, it has been suggested that effects induced by this exchange can be measured experimentally using weak measurements of pre- and post-selected ensembles of particles. This paper reports on such an optical experiment that yielded measured weak values that were consistent with the theoretical prediction of an effect induced by a non-local exchange of modular momentum.

A generalized analytical tripartite loss model is posited for Mach-Zehnder interferometer (MZI) phase sensitivity which is valid for both arbitrary photon input states and arbitrary system environmental states. This model is shown to subsume the phase sensitivity models for the lossless MZI and the ground state MZI. It can be employed to develop specialized models useful for estimating phase sensitivities, as well as for performing associated design trade-off analyses, for MZIs which operate in environmental regimes that are not contained within the ground state MZI's envelope of validity. As a simple illustration of its utility, the model is used to develop phase sensitivity expessions for an MZI with "excited" internal arms and an MZI with "excited" output channels. These expressions yield a conditional relationship between the expected number of photons entering an MZI and its efficiency parameters which-when satisfied-predicts an enhanced phase sensitivity for the MZI with "excited" output channels relative to that for the MZI with "excited" internal arms., Comment: 16 pages no figures This is the model used for the research results reported in Rev. 17. Three-tone coherent microwave electromechanical measurement of a superfluid Helmholtz resonator

Abstract

We demonstrate electromechanical coupling between a superfluid mechanical mode and a microwave mode formed by a patterned microfluidic chip and a 3D cavity. The electric field of the chip-cavity microwave resonator can be used to both drive and detect the motion of a pure superflow Helmholtz mode, which is dictated by geometric confinement. The coupling is characterized using a coherent measurement technique developed for measuring weak couplings deep in the sideband unresolved regime. The technique is based on two-probe optomechanically induced transparency/amplification using amplitude modulation. Instead of measuring two probe tones separately, they are interfered to retain only a signal coherent with the mechanical motion. Abstract

Health disparities in childhood‐onset systemic lupus erythematosus (SLE) disproportionately impact marginalized populations. Socioeconomically patterned missing data can magnify existing health inequities by supporting inferences that may misrepresent populations of interest. Our objective was to assess missing data and subsequent health equity implications among participants with childhood‐onset SLE enrolled in a large pediatric rheumatology registry. We evaluated co‐missingness of 12 variables representing demographics, socioeconomic position, and clinical factors (e.g., disease‐related indices) using Childhood Arthritis and Rheumatology Research Alliance Registry childhood‐onset SLE enrollment data (2015–2022; n = 766). We performed logistic regression to calculate odds ratios (ORs) and 95% confidence intervals (95% CIs) for missing disease‐related indices at enrollment (Systemic Lupus Erythematosus Disease Activity Index 2000 [SLEDAI‐2K] and/or Systemic Lupus International Collaborating Clinics/American College of Rheumatology Damage Index [SDI]) associated with data missingness. We used linear regression to assess the association between socioeconomic factors and SLEDAI‐2K at enrollment using 3 analytic methods for missing data: complete case analysis, multiple imputation, and nonprobabilistic bias analyses, with missing values imputed to represent extreme low or high disadvantage. On average, participants were missing 6.2% of data, with over 50% of participants missing at least 1 variable. Missing data correlated most closely with variables within data categories (i.e., demographic). Government‐assisted health insurance was associated with missing SLEDAI‐2K and/or SDI scores compared to private health insurance (OR 2.04 [95% CI 1.22, 3.41]). The different analytic approaches resulted in varying analytic sample sizes and fundamentally conflicting estimated associations. Our results support intentional evaluation of missing data to inform effect estimate interpretation and critical assessment of causal statements that might otherwise misrepresent health inequities.

20. Small fatigue crack propagation in a near alpha titanium alloy, IMI 829

Abstract

Small crack growth rates are assessed for the near alpha titanium alloy, IMI 829, with beta processed microstructures. Constant load amplitude tests were conducted using three test piece geometries which encompassed plain surfaces and stress concentrations. Two computerised crack length measurement techniques were used: A switched d.c. potential drop method and a photomicroscopic system. The p.d. system together with a clip gauge were used to assess crack closure. Crack growth is structure sensitive, characterised by crack path tortuosity, crack bifurcation and a faceted fracture surface. Colony and prior beta grain boundaries provide barriers to propagation which result in temporary arrest in many instances. Small crack growth rates were compared and contrasted with available long crack data for the same alloy and microstructure. A transition, ΔKT, was observed for the small crack growth rate data. This transition is most marked for through thickness cracks at notches and least marked for part through cracks in plain surfaces. Small crack growth behaviour was observed for values of ΔK less than ΔK_T with growth rates up to a factor of 10 greater than long crack rates. For values of ΔK greater than ΔKT the data merge, although in some instances rates for through thickness small cracks were slower. The transition from small to long crack behaviour occurs as the monotonic crack tip plastic zone approaches the widmanstatten alpha colony size. At low ΔK there was no evidence of a threshold even at 3.6 MNm^-3/2. Load ratio was not observed to affect growth rates. The results suggest that the significant difference between crack voltage signals during a load cycle, for ΔK above and below ΔKT, could be due to crack tip plasticity rather than closure in this study. Crack closure was not evident from crack opening displacements obtained using the clip gauge.

Abstract

In recent years, students in police academies and higher education institutions around the world have worked together to analyse cold cases including long-term missing persons cases in collaboration with investigators and prosecutors. In 2020, three European organisations, the Police Expert Network on Missing Persons (PEN-MP), AMBER Alert Europe and Locate International, succeeded in connecting these educational organisations enabling them to work collectively on cases and conduct cold case analyses (CCA) across international borders. The International Cold Case Analysis Project (ICCAP) learning objectives were to 1) collect the necessary information about the victim, 2) reconstruct the crime, and 3) investigate trace control. In a learning objective-based evaluation using Computer-Assisted Web Interviewing, 76 participating students from the German and International ICCAP teams were asked to complete a pre- and post-review questionnaire to self-assess their personal competence development. Participants reported significant increases in competence in all evaluated areas, thus demonstrating that authentic and relevant collaborations can enrich the learning environment, promote the use of professional skills, and provide significant knowledge exchange opportunities between academia and industry. Drawing on case studies of cold case missing persons' investigations and unidentified found remains, this article shares how university academics, students and community volunteers can work together nationally and internationally to find out what has happened to missing people and how we can more effectively identify the previously unidentified. In so doing, we share the expertise required to progress these cold cases and provide recommendations to support other institutions and organisations in adopting this innovative approach.

Abstract

In the spinel oxide cathode family, LiNi0.5Mn1.5O4 (LNMO) shows a high operating voltage (≈4.7 V vs Li/Li⁺) and excellent Li-ion mobility with stable 3D conducting channels. Ni/Mn cation disordered and ordered phases usually coexist in LNMO materials, and they have distinct structural and electrochemical properties, resulting in different battery performances for LNMO materials with different phase compositions. Identifying the correlation between phase compositions and electrochemical properties is of significance to the improvement of battery performance and understanding of degradation mechanisms. Herein, the disordered/ordered phase compositions in LNMO materials are tailored by post-annealing strategies and their impacts on electrochemical performance and degradation mechanisms from the surface to the bulk are systematically investigated. The ordered phase increases rapidly as Mn³⁺ is oxidized to Mn⁴⁺ through a post-annealing process. LNMO with an intermediate fraction of disordered and ordered phases gives rise to improved cycling stability. This article further reports that a high ordered phase fraction can preferentially protect Ni from dissolution during cycling. However, these results suggest that the transition metal dissolution and surface structural change of LNMO do not exhibit a direct correlation with cycling stability. These results indicate the capacity fading mainly correlates with the bulk structural distortion, leading to decreased Li-ion kinetics.

Abstract

The goal was to characterize short‐term kidney status and describe variation in early care utilization in a multicenter cohort of patients with childhood‐onset systemic lupus erythematosus (cSLE) and nephritis. We analyzed previously collected prospective data from North American patients with cSLE with kidney biopsy‐proven nephritis enrolled in the Childhood Arthritis and Rheumatology Research Alliance (CARRA) Registry from March 2017 through December 2019. We determined the proportion of patients with abnormal kidney status at the most recent registry visit and applied generalized linear mixed models to identify associated factors. We also calculated frequency of medication use, both during induction and ever recorded. We identified 222 patients with kidney biopsy–proven nephritis, with 64% class III/IV nephritis on initial biopsy. At the most recent registry visit at median (interquartile range) of 17 (8–29) months from initial kidney biopsy, 58 of 106 patients (55%) with available data had abnormal kidney status. This finding was associated with male sex (odds ratio [OR] 3.88, 95% confidence interval [95% CI] 1.21–12.46) and age at cSLE diagnosis (OR 1.23, 95% CI 1.01–1.49). Patients with class IV nephritis were more likely than class III to receive cyclophosphamide and rituximab during induction. There was substantial variation in mycophenolate, cyclophosphamide, and rituximab ever use patterns across rheumatology centers. In this cohort with predominately class III/IV nephritis, male sex and older age at cSLE diagnosis were associated with abnormal short‐term kidney status. We also observed substantial variation in contemporary medication use for pediatric lupus nephritis between pediatric rheumatology centers. Additional studies are needed to better understand the impact of this variation on long‐term kidney outcomes.

Abstract

Children with well‐controlled juvenile idiopathic arthritis (JIA) frequently experience flares after medication discontinuation, but the outcomes of these flares have not been well described. The objective of this study was to characterize the rates and predictors of disease recapture among children with JIA who restarted medication to treat disease flare. Children with JIA who discontinued conventional synthetic or biologic disease‐modifying antirheumatic drugs for well‐controlled disease but subsequently experienced a flare and restarted medication were identified from the Childhood Arthritis and Rheumatology Research Alliance (CARRA) registry. The primary outcome was inactive disease (ID) (physician global assessment <1 and active joint count = 0) 6 months after flare. A total of 333 patients had complete data for ID at 6 months after flare. The recapture rate for the cohort was 55%, ranging from 47% (persistent oligoarthritis) to 69% (systemic arthritis) (P= 0.4). Approximately 67% of children achieved ID by 12 months. In the multivariable model, history and reinitiation of biologic drugs were associated with increased odds of successful recapture (odds ratio [OR] 4.79 [95% confidence interval (95% CI) 1.22–18.78] and OR 2.74 [95% CI 1.62–4.63], respectively). Number of joints with limited range of motion was associated with decreased odds (OR 0.83 per 1 joint increase [95% CI 0.72–0.95]). Approximately half of JIA flares post‐discontinuation were recaptured within 6 months, but rates of recapture varied across JIA categories. These findings inform shared decision‐making for patients, families, and clinicians regarding the risks and benefits of medication discontinuation. Better understanding of biologic predictors of successful recapture in JIA are needed.

This is routinely performed with point‐of‐care analysers and doses of medications are adjusted based on the results.\ud \ud Objectives: \ud \ud To investigate the performance of two point‐of‐care analysers (IDEXX Catalyst Dx and IDEXX VetStat) against a reference laboratory method for the measurement of blood sodium, potassium and chloride concentrations, as well as sodium: potassium ratios, in dogs diagnosed with and treated for hypoadrenocorticism.\ud \ud Methods: \ud \ud Forty‐eight dogs were enrolled into a prospective cross‐sectional study. Paired blood samples were taken and tested on two point‐of‐care analysers and at a reference laboratory. Statistical analysis was then performed with Bland‐Altman analysis and Passing‐Bablok regression. The clinical effects of inaccurate electrolyte analysis were investigated.\ud \ud Results: \ud \ud In total, 329 samples were tested on the Catalyst analyser, while another 72 samples were tested on the VetStat. Passing‐Bablok regression identified both proportional and constant bias for some analytes. There was poor agreement between sodium and chloride concentrations on both analysers. Both analysers tended to give higher results than the reference method for all analytes, except for potassium when measured on the VetStat.\ud \ud Clinical Significance: \ud \ud There are inherent differences between the electrolyte concentrations measured by these two point‐of‐care analysers and reference laboratory methods in dogs with hypoadrenocorticism.

39. The outcome of bone graft surgery for nonunion of fractures of the scaphoid

Abstract

Data on 806 patients undergoing bone graft surgery for a scaphoid fracture nonunion were retrospectively collected at 19 centres in the United Kingdom. Each centre contributed at least 30 cases. Sufficient data were available in 462 cases to study factors that influenced the outcome of surgery. Overall union occurred in at least 69%, and nonunion in at least 22%, with 9% of cases having ‘uncertain union status’. Union appeared to be adversely influenced by smoking and the time between acute scaphoid fracture and nonunion surgery, with adjusted odds ratios of 1.8 and 2.4, respectively, but neither achieved the pre-determined significance level of 0.003. The type of bone graft (vascular vs non-vascular; iliac crest vs distal radius) did not appear to influence outcome. Further large multicentre prospective studies with clear definitions of ‘union’ and other factors are needed to clarify whether modification of surgical technique can influence union.

Contains fulltext : 167711.pdf (Publisher’s version ) (Closed access) IMPORTANCE: The 16p11.2 BP4-BP5 duplication is the copy number variant most frequently associated with autism spectrum disorder (ASD), schizophrenia, and comorbidities such as decreased body mass index (BMI). OBJECTIVES: To characterize the effects of the 16p11.2 duplication on cognitive, behavioral, medical, and anthropometric traits and to understand the specificity of these effects by systematically comparing results in duplication carriers and reciprocal deletion carriers, who are also at risk for ASD. DESIGN, SETTING, AND PARTICIPANTS: This international cohort study of 1006 study participants compared 270 duplication carriers with their 102 intrafamilial control individuals, 390 reciprocal deletion carriers, and 244 deletion controls from European and North American cohorts. Data were collected from August 1, 2010, to May 31, 2015 and analyzed from January 1 to August 14, 2015. Linear mixed models were used to estimate the effect of the duplication and deletion on clinical traits by comparison with noncarrier relatives. MAIN OUTCOMES AND MEASURES: Findings on the Full-Scale IQ (FSIQ), Nonverbal IQ, and Verbal IQ; the presence of ASD or other DSM-IV diagnoses; BMI; head circumference; and medical data. RESULTS: Among the 1006 study participants, the duplication was associated with a mean FSIQ score that was lower by 26.3 points between proband carriers and noncarrier relatives and a lower mean FSIQ score (16.2-11.4 points) in nonproband carriers. The mean overall effect of the deletion was similar (-22.1 points; P < .001). However, broad variation in FSIQ was found, with a 19.4- and 2.0-fold increase in the proportion of FSIQ scores that were very low (100) compared with the deletion group (P < .001). Parental FSIQ predicted part of this variation (approximately 36.0% in hereditary probands). Although the frequency of ASD was similar in deletion and duplication proband carriers (16.0% and 20.0%, respectively), the FSIQ was significantly lower (by 26.3 points) in the duplication probands with ASD. There also were lower head circumference and BMI measurements among duplication carriers, which is consistent with the findings of previous studies. CONCLUSIONS AND RELEVANCE: The mean effect of the duplication on cognition is similar to that of the reciprocal deletion, but the variance in the duplication is significantly higher, with severe and mild subgroups not observed with the deletion. These results suggest that additional genetic and familial factors contribute to this variability. Additional studies will be necessary to characterize the predictors of cognitive deficits.

Abstract

We report the preparation of exactly one 87Rb atom and one 133Cs atom in the same optical tweezer as the essential first step towards the construction of a tweezer array of individually trapped 87Rb133Cs molecules. Through careful selection of the tweezer wavelengths, we show how to engineer species-selective trapping potentials suitable for high-fidelity preparation of Rb + Cs atom pairs. Using a wavelength of 814 nm to trap Rb and 938 nm to trap Cs, we achieve loading probabilities of 0.508(6) for Rb and 0.547(6) for Cs using standard red-detuned molasses cooling. Loading the traps sequentially yields exactly one Rb and one Cs atom in 28.4(6)% of experimental runs. Using a combination of an acousto-optic deflector and a piezo-controlled mirror to control the relative position of the tweezers, we merge the two tweezers, retaining the atom pair with a probability of. We use this capability to study hyperfine-state-dependent collisions of Rb and Cs in the combined tweezer and compare the measured two-body loss rates with coupled-channel quantum scattering calculations. Abstract

We have recently been funded by the UK Prevention Research Partnership (UKPRP) to develop a UK school food network. The overarching aim is to build a community working towards a more health-promoting food and nutrition system in UK schools (primary and secondary). Here we describe the current status of school food research, including a review of the literature supporting the health-promoting schools approach and outline the opportunities for intervention and innovation establishment of the network present. Key potential school food research themes are described, and their prioritisation within the network, as well as network activities that have been planned, with the ultimate ambition of reducing socio-economic diet-related inequalities, and, consequently, non-communicable disease risk. Abstract

Initial internet-based cognitive behavioral therapy (iCBT) programs for anxiety disorders in children and young people (CYP) have been developed and evaluated, however these have not yet been widely adopted in routine practice. The lack of guidance and formalized approaches to the development and dissemination of iCBT has arguably contributed to the difficulty in developing iCBT that is scalable and sustainable beyond academic evaluation and that can ultimately be adopted by healthcare providers. 49. The state of play: how commensurate are BTECs and A-levels in sport and physical education

Abstract

The somewhat forever-changing landscape of education in England has recently challenged the post-16 sector in new ways, with funding and a knowledge-based curriculum just some of the new initiatives institutions must acclimatise to. Sport provision, a generic term used in this paper to encompass level three sport and physical education programmes, is not in limbo, but certainly faces challenges. This research focused on exploring the current ‘state of play’ of post-16 sport provision. Thirteen semi-structured interviews were carried out on teachers focusing solely on the Advanced (A)-level and Business and Technology Education Council (BTEC) national routes. Interviews were recorded and transcribed verbatim. The data were analysed using the six phases outlined by Braun and Clarke in the form of a thematic analysis. The main findings were that the content is perceived to be challenging due to the depth and detail of knowledge required on topic areas investigated on the A-level route, in contrast to the breadth, related to the number of units of study on the BTEC national programmes. Practical knowledge also emerged as a key theme, with results indicating a diminished importance of the practical aspects of both programmes, and conflicting views were evident on how practical knowledge should be assessed, with some preferring the grading of practical performance only and others preferring wider aspects of performance to feature in the grade awarded. Finally, post-16 options in sport and PE were perceived to be ‘a positive thing’ but the caveat with this is the concerns regarding programme symmetry and how commensurate respective programmes are.

Abstract

The impact of varying the tip clearance of each rotor on the performance of a counter-rotating axial compressor has been investigated based on numerical simulations. The main purpose was to investigate the sensitivity to the tip clearance of each of the two individual rotors and the corresponding aerodynamic mechanisms associated with the performance variation in this compressor. The results indicated that both the total pressure ratio and the efficiency decreased as the tip clearance was increased, and the sensitivity curve for peak efficiency for both rotors was found to be an approximately linear negative relationship with increasing tip clearance. The variations of peak efficiency and stability margin of Rotor 2 were more sensitive to changing tip clearance than Rotor 1. An optimum combination of tip gaps existed for this compressor, i.e. 0.5t for Rotor 1 and 0.25x fcr Rotor 2 (where r represents the nominal tip clearance value). At this optimum configuration, the peak efficiency and stability margin were improved by 0.63% and 29.4%, respectively. The location of the onset of the tip leakage vortex was found to be shifted downstream when the tip clearance increased. The nature of the lip leakage flow for each rotor was found to be influenced by the variation of tip clearance in the other rotor, fotor 2 showed a more significant impact on Rotor 1.
