Abstract

Within the study of the urban heat island (UHI) in Echirolles and Grenoble (France, the eastern part of the alpine arc), two temperature measurement networks have been deployed. The aim is to measure the temperature gradients associated with the UHI in summer. A total of 62 measurement points has been installed in the various neighborhoods on 3-meter-high streetlights, starting in summer 2019. The preliminary classification of the different neighborhood typologies according to ``Local Climate Zone'' guided the choice of location for the temperature sensors. These urban observations respond to a dual challenge: firstly, to observe temperature located in complex topographical situations with valleys, and secondly, to observe the urban climate in neighborhoods where social considerations are important. Municipalities of Echirolles and Grenoble were involved in the investigation. The ADEME-funded (The French Agency for Ecological Transition) CASSANDRE research program analyzes and processes these observations to study the vulnerability of inhabitants to heat waves and more generally to summer heat stress.

Hurricane Gustav which approaches the Western coast of Louisiana at the end of August 2008 is presented like one of most intense since Katrina, which earlier has considerably affected the city of News Orleans three years ago. However, if the hazard is comparable, the vulnerability is less in particular because of an effective prevention. This article compares various hurricanes of the Atlantic Ocean, Katrina, Gustav, Hanna and Ike, by differentiating the natural hazard from the vulnerability, especially in Haiti and the United States.

Abstract

The climate of the French Alps remains poorly described, and its interannual variability is difficult to study because of the wide variety of local topoclimatic characteristics and the lack of available long-term time-series. Regional consequences by certain climatic threat on various economic activities are nevertheless expected. This article thus proposes a diagnosis of the contemporary climatic variations (1959-2009) on the scale of French northern Prealps (Vercors and Chartreuse) by using the time series of 4 reference stations. Observed changes are then related to known or potential impacts on the regional socioeconomic activities.

4. A Focus on Relapsed Multiple Myeloma

Abstract

Multiple myeloma (MM) is a relapsing disease for many patients with multiple myeloma. At relapse, patients have many options for treatment once disease has progressed. Advanced practitioners are well suited to set expectations for ongoing therapy and underscore the importance of continued disease monitoring. Criteria for relapsed myeloma rely on biomarker and radiologic imaging, as well as physical exam and awareness of new bone pain or changes in physiologic function. The treatment of patients with relapsed MM requires a personalized approach and considers patient desires in regard to aggressiveness of therapy and willingness to participate in a clinical trial. The prognosis of patients with relapsed MM depends upon disease characteristics at baseline or throughout, as patients may acquire adverse cytogenetic abnormalities through various lines of treatment. Empowering patients to understand their diagnosis, interpret labs, and take an active role in treatment selection through shared decision-making can improve patients’ quality of life and enhance adherence.

Abstract

The chemokine CCL28 is highly expressed in mucosal tissues, but its role during infection is not well understood. Here, we show that CCL28 promotes neutrophil accumulation in the gut of mice infected with Salmonella and in the lung of mice infected with Acinetobacter. Neutrophils isolated from the infected mucosa expressed the CCL28 receptors CCR3 and, to a lesser extent, CCR10, on their surface. The functional consequences of CCL28 deficiency varied between the two infections: Ccl28−/− mice were highly susceptible to Salmonella gut infection but highly resistant to otherwise lethal Acinetobacter lung infection. In vitro, unstimulated neutrophils harbored pre-formed intracellular CCR3 that was rapidly mobilized to the cell surface following phagocytosis or inflammatory stimuli. Moreover, CCL28 stimulation enhanced neutrophil antimicrobial activity, production of reactive oxygen species, and formation of extracellular traps, all processes largely dependent on CCR3. Consistent with the different outcomes in the two infection models, neutrophil stimulation with CCL28 boosted the killing of Salmonella but not Acinetobacter. CCL28 thus plays a critical role in the immune response to mucosal pathogens by increasing neutrophil accumulation and activation, which can enhance pathogen clearance but also exacerbate disease depending on the mucosal site and the infectious agent.

Abstract

Searching for positive selection signals across genomes has identified functional genetic variants responding to environmental change. In Native Americans of Mexico, we used the fixation index (Fst) and population branch statistic (PBS) to identify SNPs suggesting positive selection. The 103 most differentiated SNPs were tested for associations with metabolic traits, the most significant association was FADS2/rs174616 with body mass index (BMI). This variant lies within a linkage disequilibrium (LD) block independent of previously reported FADS selection signals and has not been clearly associated with metabolic phenotypes. We tested this variant in two independent cohorts with cardiometabolic data. In the Genetics of Atherosclerotic Disease (GEA) cohort, the derived allele (T) was associated with increased BMI, lower LDL-C levels and a decreased risk of subclinical atherosclerosis in women. Significant gene-diet interactions affected lipid, apolipoprotein and adiponectin levels with differences according to sex, involving mainly total and complex dietary carbohydrate%. In the Genotype-related Effects of PUFA trial, the derived allele was associated with lower Δ-6 desaturase activity and erythrocyte membrane dihomo-gamma-linolenic acid (DGLA) levels, and with increased Δ-5 desaturase activity and eicosapentaenoic acid levels. This variant interacted with dietary carbohydrate% affecting Δ-6 desaturase activity. Notably, the relationship of DGLA and other erythrocyte membrane LC-PUFA indices with HOMA-IR differed according to rs174616 genotype, which has implications regarding how these indices should be interpreted. In conclusion, this observational study identified rs174616 as a signal suggesting selection in an independent linkage disequilibrium block, was associated with cardiometabolic and erythrocyte measurements of LC-PUFA in two independent Mexican cohorts and showed significant gene-diet interactions.

Abstract

International audience; This study details the specificity of the two heat waves that occurred in the city of Grenoble (Isère, France) in June and July 2019. The processed temperature data includes 7 stations, in particular some that have never before been analysed from a climatological analysis perspective. The analyses also cover the daily temperature values projected at the scale of the urban area according to the RCP 8.5 scenario over the period 2006-2100. The results confirm that the city possesses a high degree of spatial variability during an extreme hot event, which the current measurement network is not yet able to discriminate sufficiently, not to mention the vertical contrasts that also exist. The heat waves of 2019 are a priori potential analogues of the conditions expected between now and 2050 (whereas the exceptional heat wave of 2003 would surely be representative of the normal at the end of the century). These feedbacks and quantified scientific elements on the heat wave hazard help the City of Grenoble, which is very involved in the study of climate risk, to improve and anticipate its programming and tactical urban planning to adapt to climate change, by building its own typical diagnosis of urban microclimatic characteristics.; -Cette étude détaille la spécificité des deux vagues de chaleurs qui se sont produites à l'échelle de la ville de Grenoble (Isère, France) en juin et juillet 2019. Les données thermiques traitées regroupent 7 stations, en particulier certaines encore jamais analysées dans une perspective d'analyse climatologique. Les analyses portent aussi sur les valeurs thermiques quotidiennes projetées à l'échelle de l'aire urbaine en fonction du scénario RCP 8.5 sur la période 2006-2100. Les résultats confirment que la ville possède une grande variabilité spatiale lors d'un événement chaud extrême, que le réseau de mesure actuel n'arrive pas encore à discriminer suffisamment, sans compter les contrastes verticaux qui existent également. Les vagues de chaleur de 2019 sont a priori des analogues potentiels des conditions attendues d'ici 2050 (alors que l'aléa exceptionnel de 2003 serait sûrement représentatif de la normale de fin du siècle). Ces retours d'expérience et éléments scientifiques quantifiés sur l'aléa caniculaire aident la Ville de Grenoble, très impliquée dans l'étude du risque climatique, à améliorer et anticiper ses programmations et son urbanisme tactique pour s'adapter au changement climatique, en se construisant son propre diagnostic typique des caractéristiques microclimatiques urbaines. Abstract

Este trabajo analiza cuantitativamente el impacto en la responsabilidad social del alumnado universitario de Magisterio, a partir del desarrollo de una actividad de análisis de un centro educativo y su contexto, así como del diseño de estrategias para mejorar sus necesidades. En cuanto a la metodología utilizada, se ha suministrado un cuestionario ad hoc de 15 ítems a partir de la Escala de Responsabilidad Social (De la Calle, García y Ortega, 2008), en 217 alumnos y alumnas del primer curso del Grado de Magisterio. Se ha realizado un análisis descriptivo de resultados pre-test y post-test, cuantitativo multivariante de la escala mediante ANOVA, de correlaciones mediante Pearson y de comparación de resultados pre-test y post-test mediante T-Student. Los resultados muestran un incremento significativo de valores tras la actividad en todos los análisis realizados. Además, se han detectado valores más elevados y relacionados entre sí en los constructos y en el análisis de comparación de muestras post-test, con respecto al pre-test. En el apartado de discusión, los resultados están en la línea del desarrollo de comportamientos sociales responsables a partir de la realización de proyectos educativos de autores como Fonseca et al., (2019) o Bustamante y Navarro (2007). Se concluye la evidencia del efecto positivo de la realización de la práctica en la responsabilidad social del alumnado.

Abstract

Arachis hypogaea L. is an annual legume that is one of the most consumed plant species. On the other hand, it belongs to one of the most monitored clinically important allergens worldwide. The polymorphism of this species based on allergen coding genes could be useful in its characterisation, but previously, no allergen-based marker techniques have been developed for peanuts. A new type of DNA-based markers of coding regions were used to analyse the variability of 21 peanut accessions - BBAP (Bet v1 based amplicon polymorphism), PBAP (profilin based amplicon polymorphism), and VBAP (vicilin based amplicon polymorphism). All of the used technique provided polymorphic fingerprints and distinguished the analysed peanut accessions. The effectivity of these techniques corresponds to the presence of the allergen homologous sequences that are a part of the A. hypogaea genome. VBAP was the most effective in distinguishing the analysed peanut accessions when compared to the results of BBAP and PBAB. For BBAP, two of the analysed accessions provided the same fingerprinting pattern. The ability of the used markers to detect polymorphisms was comparable, with an average polymorphism information content (PIC) value of 0.47.

Abstract

Sarcocystis aucheniae is a coccidian parasite that produces macroscopic sarcocysts in South American camelid (SAC) muscles and causes a disease known as SAC sarcocystosis. This parasitosis hampers the commercialization of llama and alpaca meat, a vital economic activity in the Andean regions. No control or prevention methods are available, and diagnosis is based on postmortem visual inspection of carcasses. The aim of this study was to identify S. aucheniae B-cell epitopes suitable for the development of diagnostic methods for SAC sarcocystosis. To this end, sarcocyst immunoreactive protein bands were analyzed via mass spectrometry, and proteins in each band were identified in silico by searching in the parasite transcriptome. Five highly antigenic, hydrophilic B-cell epitopes, predicted not to cross-react with antibodies against other coccidia, were selected for future development of peptide-based serological tests. In addition, conserved domains present in the identified proteins allowed us to unravel metabolic pathways and mechanisms active in the parasitic stages present in sarcocysts, including aerobic respiration, antioxidant activity, signal transduction, protein synthesis and processing, and host–pathogen interactions. This study provides novel information on the biology of S. aucheniae, as well as new protein sequences that can be used for the development of diagnostic tests and chemotherapeutic approaches for SAC sarcocystosis.

Abstract

Thermal extremes generate health risks that are increasingly taken into account, including in tropical Africa. This work compares the duration and the frequency of spring heat waves (HWs) in West Africa according to 5 indices: Warm Spell (WS), Heat Index (HI) with and without the relative humidity, Apparent Temperature (AT), and Excess Heat Factor (EHF). The daily relative humidity and temperature data (minimum, maximum, dew point are provided by the GSOD database for the period 1973-2014 and cover 145 stations spread over a large African area (3-24°N; 24°W-36°E). The results show that the duration of the HWs is short on the coast (3 days) and they last longer inland the continent and in the Sahelian belt. HWs’ frequency varies greatly according to the heat indices used, from 3% of spring days in Cape Verde to 30 % in Central Sahel for the EHF., Les extrêmes thermiques génèrent des risques sanitaires de plus en plus étudiés, y compris en Afrique tropicale. Ce travail compare les durée et fréquence des vagues de chaleur (HW) printanières (avril à juin) en Afrique de l’Ouest selon 5 indices : Warm Spell (WS), Heat Index (HI) avec et sans prise en compte de l’humidité relative, Température Apparente (AT) et Facteur de chaleur excessive ou Excess Heat Factor (EHF). Les données quotidiennes de température (minimale, maximale, point de rosée) et d’humidité relative proviennent de la base GSOD sur la période 1973-2014 et couvrent 145 stations réparties sur un large espace africain tropical (3°N-24°N ; 24°W-36°E). Les résultats montrent que la durée des HW est courte sur le littoral (3 jours, où elles sont rares) et plus longue au coeur du continent en zone sahélienne. La fréquence des HW varie selon les indices de chaleur utilisés : de 3% des jours de printemps au Cap Vert à 30% au Sahel Central selon l’EHF.

Abstract

Después de 30 años de Presupuestos Participativos (PP), ha habido muchos experimentos y pocos diagnósticos. Pero, ¿cómo evaluar una experiencia de presupuesto participativo sin un modelo de evaluación propio? El objetivo de este trabajo es evaluar el PP de la ciudad española de Zaragoza utilizando un modelo desarrollado por los propios investigadores. El método implicó un modelo sintético para evaluar el caso práctico con información empírica obtenida mediante observación directa, entrevista estructurada y análisis de fuentes secundarias. Los resultados aportan una importante innovación: un área e indicadores relacionados con el diseño y la planificación. Los resultados aportan la medición de 105 indicadores agrupados en 35 alcances, 15 criterios y cuatro esferas. El resultado fue una valoración positiva, media-alta, de la experiencia.

Abstract

Trends in daily maximum (TX) and minimum (TN) temperatures and indices of warm extremes are studied in tropical North Africa, west of the eastern African highlands, from 1961 to 2014. The analysis is based on the concatenation and cross-checking of two observed databases. Due to the large number of missing entries (~25%), a statistical infilling using probabilistic principal component analysis was applied. Averaged over 90 stations, the linear trends of annual mean TX and TN equal respectively +0.021 °C/yr and +0.028 °C/yr. The frequency of very hot days (TX > 35°C) and tropical nights (TN > 20°C), as well as the frequency of daily TX and TN above the 90th percentile (p90) (“warm days” and “warm nights”), roughly follows the variations of mean TX and TN, respectively. Background Venous thromboembolism (VTE) and cardiovascular (CV) disease can occur in patients with multiple myeloma. Although VTE and CV disease are separate medical conditions, they can be serious and even life-threatening. . Objectives The objectives of this article are to describe risk factors for cancer-associated VTE, describe the influence of CV disease on patients with multiple myeloma, and review the approaches to VTE and CV disease identification and treatment. . Methods PubMed and CINAHL® databases were used to identify literature to describe VTE and CV in patients diagnosed with multiple myeloma. . Findings When present in patients with multiple myeloma, VTE and CV disease can limit patient tolerance for myeloma treatment and, therefore, decrease therapeutic options.

Background About 85% of patients with multiple myeloma develop bone disease. In these patients, lytic bone lesions can cause fractures, poor circulation, blood clots, pain, poor mobility, and decreased quality of life. . Objectives This article presents consensus statements to guide nurses in the assessment and management of bone disease, pain, and mobility in patients with multiple myeloma at varying points in their disease trajectory. . Methods Members of the International Myeloma Foundation Nurse Leadership Board reviewed previously provided recommendations, current recommendations based on literature review, and evidence-based grading. . Findings Oncology nurses play a key role in maximizing bone health, minimizing skeletal injury, maximizing pain control, and improving quality of life in patients by enhancing patient mobility and safety. Clinician assessment accompanied by effective interventions reduces patient injury and optimizes functioning in patients with multiple myeloma.

Abstract

Cet article fait un état des lieux sur la notion de services climatiques, et sur le déficit actuel de leurs orientations et applications à l’échelle urbaine. Il détaille particulièrement les enjeux que représentent ces nouveaux services dans le cadre de projets liés à l’adaptation aux changements climatiques pour les municipalités. Le constat de l\'inadéquation des mesures climatiques collectées et des réseaux de mesures est également proposé, en les mettant en perspective avec de nouvelles approches de veille climatique, en particulier la mesure citoyenne et les nouvelles sources d’observation associées. L’exemple français de la ville de Grenoble sert ensuite à illustrer le déficit actuel de données climatiques utiles aux diagnostics et services urbains, alors même que les problématiques locales sur les extrêmes thermiques sont stratégiques, en particulier pour la plateforme municipale participative et d’anticipation ‘Grenoble Ville de Demain’., This paper provides an overview of the concept of climate services and the current shortfall in their orientations and applications at the urban scale. It details the issues that these new services represent in projects related to adaptation to climate change for municipalities. The observation of the inadequacy of the collected climate measurements and of the climate networks is also done, putting them in perspective with new approaches to climate monitoring, in particular the citizen observations and the associated new sources of monitoring. The example of the French city of Grenoble is then used to illustrate the current deficit of climate data useful for urban diagnostics and services, even though the local issues on temperature extremes are strategic, especially for the municipal sharing platform ‘Grenoble City of Tomorrow\'.

Abstract

International audience; Great effort is made to address heat waves (HWs) in developed countries because of their devastating impacts on society, economy, and environment. However, HWs are still understudied over developing countries. This is particularly true in West Africa, and especially in the Sahel, where temperatures recurrently reach critical values, such as during the 2010 HW event in the western Sahel. This work aims at characterizing the Sahelian HWs during boreal spring seasons (April-May-June) and understanding the mechanisms associated with such extreme events. Over the last three decades, Sahelian HWs have been becoming more frequent, lasting longer, covering larger areas, and reaching higher intensities. The physical mechanisms associated with HWs are examined to assess the respective roles of atmospheric dynamics and radiative and turbulent fluxes by analyzing the surface energy budget. Results suggest that the greenhouse effect of water vapor is the main driver of HWs in the western Sahel, increasing minimum temperatures by enhanced downward longwave radiation. Atmospheric circulation plays an important role in sustaining these warm anomalies by advecting moisture from the Atlantic Ocean and the Guinean coasts into the Sahel. Maximum temperature anomalies are mostly explained by increased downward shortwave radiation due to a reduction in cloud cover. Interannual variability of HWs is affected by the delayed impact of El Niño-Southern Oscillation (ENSO), with anomalous temperature warming following warm ENSO events, resulting from an amplified water vapor feedback.

Abstract

International audience; This study aims in filling the gap in understanding the relationship between trend and extreme in diurnal and nocturnal temperatures (Tx and Tn) over the Gulf of Guinea area and the Sahel. Time-evolution and trend of Tx and Tn anomalies, extreme temperatures and heat waves are examined using regional and station-based indices over the 1900-2012 and 1950-2012 periods respectively. In investigating extreme temperature anomalies and heat waves, a percentile method is used. At the regional and local scales, rising trends in Tx and Tn anomalies, which appear more pronounced over the past 60 years, are identified over the two regions. The trends are characterized by an intensification of: i) nocturnal/Tn warming over the second half of the 20th century; and ii) diur-nal/Tx warming over the post-1980s. This is the same scheme with extreme warm days and warm nights. Finally annual number of diurnal and nocturnal heat waves has increase over the Gulf of Guinea coastal regions over the second half of the 20th century, and even more substantially over the post-1980s period. Although this trend in extreme warm days and nights is always overestimated in the simulations, from the Coupled Model Intercompar-ison Project Phase 5 (CMIP5), those models display rising trends whatever the scenario, which are likely to be more and more pronounced over the two regions in the next 50 years.

Abstract

As in many other cancers, survivorship of multiple myeloma involves handling treatment, recovery from therapeutic interventions, the effects of the disease, and ongoing therapies. Although mobility challenges vary among survivors of multiple myeloma, these patients have an increased risk of impaired mobility because of side effects of therapy and the pathology of the disease, as well as other factors (e.g., increasing age). Health maintenance increasingly is becoming a part of the cancer control continuum, and nurses have the opportunity to help survivors of multiple myeloma optimize their functional mobility and safety, thereby preserving quality of life. The purpose of these practice recommendations is to provide the healthcare professional with information on mobility, fall risk, and planned activity as an integral part of the plan of care for patients with multiple myeloma. Tools for nurses and physicians for assessing and evaluating the newly diagnosed patient, the patient undergoing treatment, and the long-term survivor of multiple myeloma will be provided.

Abstract

Novel therapies approved over the past decade for the management of multiple myeloma have contributed to improved overall survival in patients with newly diagnosed and relapsed disease. Nurses play a key role in educating, advocating for, and supporting patients throughout the continuum of care. Identifying potential and actual comorbid conditions associated directly with multiple myeloma and its treatment is important, as is confirming those that are patient specific so that prompt intervention can take place; therefore, the International Myeloma Foundation Nurse Leadership Board identified the most significant needs of patients diagnosed with multiple myeloma as bone health, health maintenance, mobility and safety, sexual dysfunction, and renal health. The Nurse Leadership Board then developed a survivorship care plan to assist healthcare providers and patients with multiple myeloma, their partners, and their caregivers to identify these needs.

Abstract

Tourism economy is deeply dependent on climate change. For example, skiing needs snow in quantity and quality, and sun is a factor of attractiveness in summer. In order to diagnose precisely climate change perceptions during the twenty last years, this work focuses on tourism actors in activity (restaurant owner, hotel owner...) in the Drôme department. These actors were questioned about their personal climate change experiences and feelings. This survey allows to understand how local climate evolutions are felt and which impacts on tourism activities are perceived. This work is realized within the context of the DECLIC research project (Drôme : Eau, CLimat et Impacts liés aux Changements), which is itself included in a broader national program named GICC-2 leading by the MEEDDM., Ce travail établit un diagnostic de la perception des évolutions climatiques et de leurs impacts potentiels sur les activités de tourisme que se font les professionnels du tourisme (hébergeurs, restaurateurs, institutionnels...) dans le département de la Drôme (sud-est de la France).Cette étude est effectuée dans le cadre du projet de recherche DECLIC (Drôme : Eau, CLimat, Impacts liés aux Changements) qui s'inscrit dans le programme national GICC-2 du MEEDDM. Les données sont issues du résultat d'une enquête exploratoire réalisée numériquement auprès des professionnels du tourisme du département (à partir de l'annuaire de l'ADT). Les méthodes d'analyse s'appuient sur des traitements statistiques bi- et multi-variés. Les résultats montrent la préoccupation sérieuse des acteurs en ce qui concerne l'évolution du climat et leur volonté de s'adapter.

Abstract

IntroductionImmunoglobulin A (IgA) is the main antibody isotype in body fluids such as tears, intestinal mucous, colostrum, and saliva. There are two subtypes of IgA in humans: IgA1, mainly present in blood and mucosal sites, and IgA2, preferentially expressed in mucosal sites like the colon. In clinical practice, immunoglobulins are typically measured in venous or capillary blood; however, alternative samples, including saliva, are now being considered, given their non-invasive and easy collection nature. Several autoimmune diseases have been related to diverse abnormalities in oral mucosal immunity, such as rheumatoid arthritis, Sjogren’s syndrome, and systemic lupus erythematosus (SLE).MethodsWe decided to evaluate the levels of both IgA subtypes in the saliva of SLE patients. A light chain capture-based ELISA measured specific IgA1 and IgA2 levels in a cohort of SLE patients compared with age and gender-matched healthy volunteers.ResultsSurprisingly, our results indicated that in the saliva of SLE patients, total IgA and IgA1 subtype were significantly elevated; we also found that salivary IgA levels, particularly IgA2, positively correlate with anti-dsDNA IgG antibody titers. Strikingly, we also detected the presence of salivary anti-nucleosome IgA antibodies in SLE patients, a feature not previously reported elsewhere.ConclusionsAccording to our results and upon necessary validation, IgA characterization in saliva could represent a potentially helpful tool in the clinical care of SLE patients with the advantage of being a more straightforward, faster, and safer method than manipulating blood samples.

International audience; Les changements climatiques sont déjà perceptibles en montagne (GIEC, 2007), avec notamment un réchauffement dans les Alpes et les Préalpes double de celui enregistré à l'échelle de l'hémisphère nord. Cette étude, menée dans le cadre du programme GICC-DECLIC, examine l'intensité des extrêmes thermiques estivaux actuels et futurs dans les Préalpes françaises. Les données météorologiques sont issues de sorties des trois modèles numériques de climat (ALADIN, LMDz, MAR) utilisés par le projet ANR-SCAMPEI, calibrées par rapport aux observations à maille fine de 8 km (réanalyses de type SAFRAN de Météo-France). L'étude porte sur l'analyse des températures minimales et maximales d'été, pour la période de référence (1961-1990), les périodes simulées 2021-50 et 2071-2100. Les résultats montrent notamment l'intensification des extrêmes thermiques estivaux chauds (TN et TX) d'ici les années 2080, en particulier en zone collinaire en juillet avec +6,2°C pour les températures minimales et +7,5°C pour les maximales.

Le cyclone Gustav qui aborde les côtes ouest de la Louisiane fin août 2008 est présenté comme l’un des plus intenses depuis Katrina qui a considérablement affecté la ville de la Nouvelle Orléans trois ans plus tôt. Or, si l’aléa cyclonique est comparable, la vulnérabilité l’est moins, notamment parce que la prévention est plus efficace. Cet article compare différents cyclones de l’Atlantique, en particulier Katrina, Gustav, Hanna et Ike, en différenciant l’aléa de la vulnérabilité, spécialement à Haïti et aux Etats-Unis.Hurricane Gustav which approaches the Western coast of Louisiana at the end of August 2008 is presented like one of most intense since Katrina, which earlier has considerably affected the city of News Orleans three years ago. However, if the hazard is comparable, the vulnerability is less in particular because of an effective prevention. This article compares various hurricanes of the Atlantic Ocean, Katrina, Gustav, Hanna and Ike, by differentiating the natural hazard from the vulnerability, especially in Haiti and the United States.

The climate of the French Alps remains poorly described, and its interannual variability is difficult to study because of the wide variety of local topoclimatic characteristics and the lack of available long-term time-series. Regional consequences by certain climatic threat on various economic activities are nevertheless expected. This article thus proposes a diagnosis of the contemporary climatic variations (1959-2009) on the scale of French northern Prealps (Vercors and Chartreuse) by using the time series of 4 reference stations. Observed changes are then related to known or potential impacts on the regional socioeconomic activities., Le climat des Alpes françaises reste encore mal décrit, et sa variabilité interannuelle est difficile à étudier du fait des spécificités topoclimatiques locales et du manque de séries climatiques longues. Les conséquences régionales du changement de certaines contraintes climatiques sur plusieurs secteurs d'activité sont pourtant redoutées. Cet article propose donc un diagnostic des variations climatiques contemporaines (1959-2009) à l'échelle des Préalpes du Nord (Vercors et Chartreuse) en utilisant des chroniques stationnelles pluviothermiques. Les changements observés sont ensuite mis en relation avec des impacts connus ou potentiels sur les activités socio-économiques régionales.

Abstract

Remodeling of the bone marrow microenvironment in chronic inflammation and in aging reduces hematopoietic stem cell (HSC) function. To assess the mechanisms of this functional decline of HSC and find strategies to counteract it, we established a model in which the Sfrp1 gene was deleted in Osterix+ osteolineage cells (OS1Δ/Δ mice). HSC from these mice showed severely diminished repopulating activity with associated DNA damage, enriched expression of the reactive oxygen species pathway and reduced single-cell proliferation. Interestingly, not only was the protein level of Catenin beta-1 (bcatenin) elevated, but so was its association with the phosphorylated co-activator p300 in the nucleus. Since these two proteins play a key role in promotion of differentiation and senescence, we inhibited in vivo phosphorylation of p300 through PP2A-PR72/130 by administration of IQ-1 in OS1Δ/Δ mice. This treatment not only reduced the b-catenin/phosphop300 association, but also decreased nuclear p300. More importantly, in vivo IQ-1 treatment fully restored HSC repopulating activity of the OS1Δ/Δ mice. Our findings show that the osteoprogenitor Sfrp1 is essential for maintaining HSC function. Furthermore, pharmacological downregulation of the nuclear b-catenin/phospho-p300 association is a new strategy to restore poor HSC function.

Abstract

BackgroundUntil now, most of the research addressing long-term humoral responses in coronavirus disease 2019 (COVID-19) had only evaluated the serum titers of anti-severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) IgGs, without the assessment of the baseline antiviral clinical and immune profile, which is the aim of this study and may be the key factor leading to a broad and sustained antibody response.MethodsWe included 103 patients with COVID-19. When the patients sought medical attention (baseline), a blood sample was drawn to perform immunophenotype of lymphocytes by flow cytometry. The patients were assessed 15 days after baseline and then every month until the third month, followed by a last visit 6 months after recruitment. We evaluated the anti-SARS-COV-2 IgG at all time points, and the serum levels of cytokines, chemokines, anti-cellular (AC) antibodies and neutrophil extracellular traps were also assessed during the follow-up. The primary outcome of the study was the presence of a sustained immune humoral response, defined as an anti-SARS-CoV-2 IgG titer >4.99 arbitrary units/mL in at least two consecutive measures. We used generalized lineal models to assess the features associated with this outcome and to assess the effect of the changes in the cytokines and chemokines throughout time on the development of a sustained humoral immune response.ResultsAt baseline the features associated to a sustained immune humoral response were the diagnosis of critical disease, absolute number of lymphocytes, serum IP-10, IL-4, IL-2, regulatory T cells, CD8+ T cells, and positive AC antibodies. Critical illness and the positivity of AC antibodies were associated with a sustained humoral immune response after 3 months, whilst critical illness and serum IL-13 were the explanatory variables after 6 months.ConclusionA sustained immune humoral response is strongly related to critical COVID-19, which is characterized by the presence of AC antibodies, quantitative abnormalities in the T cell compartment, and the serum cytokines and chemokines during acute infection and throughout time.

To examine practice variation in hematopoietic stem cell transplantation (HSCT) nursing and to identify the gap between recommended standards of practice and actual practice across settings. Additional practices relevant to HSCT nursing also were explored.Cross-sectional, descriptive survey.National and international cancer centers.A convenience sample was obtained from the 2006 Oncology Nursing Society Blood and Marrow Stem Cell Transplant Special Interest Group membership list (N = 205). Most participants were women (94%) with a median age of 45 years. The primary role was bedside nurse (46%), with an adult-only population (78%) in an academic (84%), inpatient (68%-88%) center. 39 (94%) U.S. states and 7 (6%) non-U.S. countries were represented.Survey development was guided by Dillman Mail and Internet survey design. Electronic questionnaires were conducted with Zoomerang Market Tools.Infection control practices across bone marrow transplantation settings.Descriptive statistics revealed minimal practice variation regarding infection control across transplantation types or conditioning regimens. Practices regarding implementation of restrictions on patients' hygiene, diet, and social interactions varied by phase of transplantation, with the greatest variations occurring during the post-transplantation phase. Sixty-two percent of respondents reported using published guidelines; 72% reported using organization-specific policies.Although published standards are under consideration, practice variation exists across transplantation centers. Whether the variation is caused by a lack of compliance with published guidelines or by the poor delineation of details for providers to translate the guidelines into practice is not known.Identifying gaps in the literature and inconsistencies in HSCT practices is an important first step in designing evidence-based projects that can be used to standardize practice and link best practices to improved patient outcomes.

International audience; L'objectif de ce travail est d'analyser les types de circulations atmosphériques responsables ou concomitantes d'épisodes thermiques extrêmes, en milieu de moyenne montagne, en prenant l'exemple des hauts plateaux du Vercors (région Rhône-Alpes, France). Les épisodes thermiques extrêmes sont discriminés par approche fréquentielle à l'échelle de la journée sur la période octobre 2004-décembre 2008. Les données sont mesurées au pas de temps de 15 minutes à la station de Gerland (44°52'N - 5°28'E, 1510 mètres d'altitude), située dans le massif préalpin du Vercors, puis moyennées à l'échelle quotidienne. Les extrêmes thermiques sont repérés en sélectionnant les centiles les plus froids et les plus chauds des données quotidiennes de température de l'air. Le type de circulation atmosphérique est analysé grâce aux travaux de James (2007) qui propose une méthode de classification objective des types de temps sur l'Europe, à partir de la classification de Hess et Brezowsky. Les résultats permettent alors de mieux comprendre les types de circulation atmosphérique associés ou responsables des événements thermiques extrêmes dans le massif du Vercors.

Abstract

Abstract In this study, we investigated the influence of the loss of cathepsin K (Ctsk) gene on the hematopoietic system in vitro and in vivo. We found that cultures with lineage− SCA1+ KIT+ (LSK) cells on Ctsk deficient stromal cells display reduced colony formation and proliferation, with increased differentiation, giving rise to repopulating cells with reduced ability to repopulate the donor LSKs and T cell compartments in the bone marrow (BM). Subsequent in vivo experiments showed impairment of lymphocyte numbers, but, gross effects on early hematopoiesis or myelopoiesis were not found. Most consistently in in vivo experimental settings, we found a significant reduction of (donor) T cell numbers in the BM. Lymphocyte deregulation is also found in transplantation experiments, which revealed that Ctsk is required for optimal regeneration of small populations of T cells, particularly in the BM, but also of thymic B cells. Interestingly, cell nonautonomous Ctsk regulates both B and T cell numbers, but T cell numbers in the BM require an additional autonomous Ctsk‐dependent process. Thus, we show that Ctsk is required for the maintenance of hematopoietic stem cells in vitro, but in vivo, Ctsk deficiency most strongly affects lymphocyte homeostasis, particularly of T cells in the BM.

Abstract

Nurses play an essential role in managing the care of patients with multiple myeloma, who require education and support to receive and adhere to optimal therapy. The International Myeloma Foundation created a Nurse Leadership Board comprised of oncology nurses from leading cancer centers and community practices. An assessment survey identified the need for specific recommendations for managing key side effects of novel antimyeloma agents. Myelosuppression, thromboembolic events, peripheral neuropathy, steroid toxicities, and gastrointestinal side effects were selected for the first consensus statements. The board developed recommendations for healthcare providers in any medical setting, including grading of side-effect toxicity and strategies for managing the side effects in general, with specific recommendations pertaining to the novel agents.

Abstract

Patients with myeloma are at risk for serious and life-threatening thromboembolic events because of their disease, individual risk factors, and antimyeloma or other medications. The International Myeloma Foundation's Nurse Leadership Board developed this consensus statement for assessment and prevention of thromboembolic events. Prophylactic measures are categorized as mechanical, regimen related, and antithrombotic drug, based on individual and myeloma-related risk factors. Aspirin is suggested for patients with no or one risk factor, low-molecular-weight heparin or full-dose warfarin for patients with two or more risk factors, and low-molecular-weight heparin or full-dose warfarin for all patients with therapy-related risks, including high-dose dexamethasone, doxorubicin, or multiagent chemotherapy.

Abstract

International audience; L'utilisation de la télédétection satellitale permet d'étudier la variabilité spatiale et temporelle de l'enneigement. Après un rappel des concepts fondamentaux, grâce aux données SPOT-Vegetation (1998-2008) à un pas de temps décadaire, et par le biais d'un indice analytique de type Normalized Difference Snow Ice Index (NDSII), ce travail propose d'analyser la couverture nivale dans la région du Vercors (Préalpes françaises). Les résultats indiquent, d'une part, une forte variabilité interannuelle de l'enneigement et, d'autre part, sa sensibilité importante à des écarts thermiques moyens relativement faibles, souvent proches de 1°C. C'est notamment le cas en début et en fin de saison hivernale. L'évolution régionale des superficies enneigées est aussi influencée à la fois par des écarts thermiques majeurs (exemples de 2000 et 2006), mais aussi par une tendance régionale générale à la hausse des températures atmosphériques observée depuis la fin des années 1980.

Abstract

Parmi les six massifs francais reconnus, celui de Corse est l'un des plus petits, mais la topographie insulaire est accidentee. Pourtant, cet espace est convoite pour l'attrait touristique lie a l'authenticite et la preservation des paysages. L'essentiel des villages de la montagne corse est situe entre 300 et 900 metres d'altitude. Or, des qu'un evenement meteorologique extreme se produit, la population montagnarde est momentanement coupee du reste de l'ile. Nous presentons ici les grands traits du climat dans le domaine de montagne en Corse, c'est-a-dire au-dessus de 800 metres d'altitude, avant de proposer deux phenomenes meteorologiques extremes ayant touche tout ou partie du massif insulaire. Il apparait que le climat de la montagne corse presente des caracteres typiquement mediterraneens comme des jours de forte chaleur et un deficit hydrique en ete, mais aussi des nuances alpines, tel un plus grand nombre de jours de pluie et de gel.

Abstract

Abstract We identified the main changes in serum metabolites associated with severe (n = 46) and mild (n = 19) COVID-19 patients by gas chromatography coupled to mass spectrometry. The modified metabolic profiles were associated to an altered amino acid catabolism in hypoxic conditions. Noteworthy, three α-hydroxyl acids of amino acid origin increased with disease severity and correlated with altered oxygen saturation levels and clinical markers of lung damage. We hypothesize that the enzymatic conversion of α-keto-acids to α- hydroxyl-acids helps to maintain NAD recycling in patients with altered oxygen levels, highlighting the potential relevance of amino acid supplementation during SARS-CoV-2 infection.

Las personas con discapacidad intelectual que sufren algún tipo de abuso y pretenden denunciarlo se enfrentan a obstáculos que impiden y/o desaniman a finalizar el proceso. Las investigaciones refieren que un alto número de víctimas con discapacidad intelectual no llega a los servicios correspondientes por la poca veracidad que se otorga a sus relatos y porque el familiar y/o profesional decide no denunciar. Este trabajo identifica circunstancias de abuso que sufren estas personas y sus implicaciones en el proceso judicial a través de grupos focales con profesionales y entrevistas a familiares y personas con discapacidad intelectual víctimas, señalando, también, recomendaciones para superar los obstáculos.

Abstract

Lupus nephritis (LN) is one of the most common manifestations of systemic lupus erythematosus (SLE), characterized by abnormal B cell activation and differentiation to memory or plasma effector cells. However, the role of these cells in the pathogenesis of LN is not fully understood, as well as the effect of induction therapy on B cell subsets, possibly associated with this manifestation, like aged-associated B cells (ABCs). Consequently, we analyzed the molecules defining the ABCs subpopulation (CD11c, T-bet, and CD21) through flow cytometry of blood samples from patients with lupus presenting or not LN, following up a small sub-cohort after six months of induction therapy. The frequency of ABCs resulted higher in LN patients compared to healthy subjects. Unexpectedly, we identified a robust reduction of a CD21hi subset that was almost specific to LN patients. Moreover, several clinical and laboratory lupus features showed strong and significant correlations with this undefined B cell subpopulation. Finally, it was observed that the induction therapy affected not only the frequencies of ABCs and CD21hi subsets but also the phenotype of the CD21hi subset that expressed a higher density of CXCR5. Collectively, our results suggest that ABCs, and more importantly the CD21hi subset, may work to assess therapeutic response since the reduced frequency of CD21hi cells could be associated with the onset of LN.

Abstract

Abstract Neuromyelitis Optica (NMO) is an autoimmune disease with a higher prevalence in non-European populations. Because the Mexican population resulted from the admixture between mainly Native American and European populations, we used genome-wide microarray, HLA high-resolution typing and AQP4 gene sequencing data to analyze genetic ancestry and to seek genetic variants conferring NMO susceptibility in admixed Mexican patients. A total of 164 Mexican NMO patients and 1,208 controls were included. On average, NMO patients had a higher proportion of Native American ancestry than controls (68.1% vs 58.6%; p = 5 × 10–6). GWAS identified a HLA region associated with NMO, led by rs9272219 (OR = 2.48, P = 8 × 10–10). Class II HLA alleles HLA-DQB1*03:01, -DRB1*08:02, -DRB1*16:02, -DRB1*14:06 and -DQB1*04:02 showed the most significant associations with NMO risk. Local ancestry estimates suggest that all the NMO-associated alleles within the HLA region are of Native American origin. No novel or missense variants in the AQP4 gene were found in Mexican patients with NMO or multiple sclerosis. To our knowledge, this is the first study supporting the notion that Native American ancestry significantly contributes to NMO susceptibility in an admixed population, and is consistent with differences in NMO epidemiology in Mexico and Latin America.

Abstract

Multi-drug resistance in bacteria is a major health problem worldwide. To overcome this issue, new approaches allowing for the identification and development of antibacterial agents are urgently needed. Peptides, due to their binding specificity and low expected side effects, are promising candidates for a new generation of antibiotics. For over two decades, a large diversity of antimicrobial peptides (AMPs) has been discovered and annotated in public databases. The AMP family encompasses nearly 20 biological functions, thus representing a potentially valuable resource for data mining analyses. Nonetheless, despite the availability of machine learning-based approaches focused on AMPs, these tools lack evidence of successful application for AMPs’ discovery, and many are not designed to predict a specific function for putative AMPs, such as antibacterial activity. Consequently, among the apparent variety of data mining methods to screen peptide sequences for antibacterial activity, only few tools can deal with such task consistently, although with limited precision and generally no information about the possible targets. Here, we addressed this gap by introducing a tool specifically designed to identify antibacterial peptides (ABPs) with an estimation of which type of bacteria is susceptible to the action of these peptides, according to their response to the Gram-staining assay. Our tool is freely available via a web server named ABP-Finder. This new method ranks within the top state-of-the-art ABP predictors, particularly in terms of precision. Importantly, we showed the successful application of ABP-Finder for the screening of a large peptide library from the human urine peptidome and the identification of an antibacterial peptide.

Abstract

BackgroundMost of the explanatory and prognostic models of COVID-19 lack of a comprehensive assessment of the wide COVID-19 spectrum of abnormalities. The aim of this study was to unveil novel biological features to explain COVID-19 severity and prognosis (death and disease progression).MethodsA predictive model for COVID-19 severity in 121 patients was constructed by ordinal logistic regression calculating odds ratio (OR) with 95% confidence intervals (95% CI) for a set of clinical, immunological, metabolomic, and other biological traits. The accuracy and calibration of the model was tested with the area under the curve (AUC), Somer’s D, and calibration plot. Hazard ratios with 95% CI for adverse outcomes were calculated with a Cox proportional-hazards model.ResultsThe explanatory variables for COVID-19 severity were the body mass index (BMI), hemoglobin, albumin, 3-Hydroxyisovaleric acid, CD8+ effector memory T cells, Th1 cells, low-density granulocytes, monocyte chemoattractant protein-1, plasma TRIM63, and circulating neutrophil extracellular traps. The model showed an outstanding performance with an optimism-adjusted AUC of 0.999, and Somer’s D of 0.999. The predictive variables for adverse outcomes in COVID-19 were severe and critical disease diagnosis, BMI, lactate dehydrogenase, Troponin I, neutrophil/lymphocyte ratio, serum levels of IP-10, malic acid, 3, 4 di-hydroxybutanoic acid, citric acid, myoinositol, and cystine.ConclusionsHerein, we unveil novel immunological and metabolomic features associated with COVID-19 severity and prognosis. Our models encompass the interplay among innate and adaptive immunity, inflammation-induced muscle atrophy and hypoxia as the main drivers of COVID-19 severity.

Abstract

Neuromyelitis Optica Spectrum Disorder (NMOSD) is a demyelinating autoimmune disease of the central nervous system, more prevalent in individuals of non-European ancestry. Few studies have analyzed genetic risk factors in NMOSD, and HLA class II gene variation has been associated NMOSD risk in various populations including Mexicans. Thymopoietin (TMPO) has not been tested as a candidate gene for NMOSD or other autoimmune disease, however, experimental evidence suggests this gene may be involved in negative selection of autoreactive T cells and autoimmunity. We thus investigated whether the missense TMPO variant rs17028450 (Arg630Cys, frequent in Latin America) is associated with NMOSD, and whether this variant shows an interaction with HLA-class II rs9272219, previously associated with NMOSD risk. A total of 119 Mexican NMOSD patients, 1208 controls and 357 Native Mexican individuals were included. The HLA rs9272219 “T” risk allele frequency ranged from 21 to 68%, while the rs17028450 “T” minor allele frequency was as high as 18% in Native Mexican groups. Both rs9272219 and rs17028450 were significantly associated with NMOSD risk under additive models (OR = 2.48; p = 8 × 10–10 and OR = 1.59; p = 0.0075, respectively), and a significant interaction between both variants was identified with logistic regression models (p = 0.048). Individuals bearing both risk alleles had an estimated 3.9-fold increased risk of NMOSD. To our knowledge, this is the first study reporting an association of TMPO gene variation with an autoimmune disorder and the interaction of specific susceptibility gene variants, that may contribute to the genetic architecture of NMOSD in admixed Latin American populations.
