Apple consumption is highly recomended for a healthy diet and is the most important fruit produced in temperate climate regions. Unfortunately, it is also one of the fruit that most ofthen provoks allergy in atopic patients and the only treatment available up to date for these apple allergic patients is the avoidance. Apple allergy is due to the presence of four major classes of allergens: Mal d 1 (PR-10/Bet v 1-like proteins), Mal d 2 (Thaumatine-like proteins), Mal d 3 (Lipid transfer protein) and Mal d 4 (profilin). In this work new advances in the characterization of apple allergen gene families have been reached using a multidisciplinary approach. First of all, a genomic approach was used for the characterization of the allergen gene families of Mal d 1 (task of Chapter 1), Mal d 2 and Mal d 4 (task of Chapter 5). In particular, in Chapter 1 the study of two large contiguos blocks of DNA sequences containing the Mal d 1 gene cluster on LG16 allowed to acquire many new findings on number and orientation of genes in the cluster, their physical distances, their regulatory sequences and the presence of other genes or pseudogenes in this genomic region. Three new members were discovered co-localizing with the other Mal d 1 genes of LG16 suggesting that the complexity of the genetic base of allergenicity will increase with new advances. Many retrotranspon elements were also retrieved in this cluster. Due to the developement of molecular markers on the two sequences, the anchoring of the physical and the genetic map of the region has been successfully achieved. Moreover, in Chapter 5 the existence of other loci for the Thaumatine-like protein family in apple (Mal d 2.03 on LG4 and Mal d 2.02 on LG17) respect the one reported up to now was demonstred for the first time. Also one new locus for profilins (Mal d 4.04) was mapped on LG2, close to the Mal d 4.02 locus, suggesting a cluster organization for this gene family, as is well reported for Mal d 1 family. Self-incompatibility (SI) systems have evolved in many flowering plants to prevent self-fertilization and thus promote outbreeding. Pear and apple, as many of the species belonging to the Rosaceae, exhibit RNase-mediated gametophytic self-incompatibility, a widespread system carried also by the Solanaceae and Plantaginaceae. Pear orchards must for this reason contain at least two different cultivars that pollenize each other; to guarantee an efficient cross-pollination, they should have overlapping flowering periods and must be genetically compatible. This compatibility is determined by the S-locus, containing at least two genes encoding for a female (pistil) and a male (pollen) determinant. The female determinant in the Rosaceae, Solanaceae and Plantaginaceae system is a stylar glycoprotein with ribonuclease activity (S-RNase), that acts as a specific cytotoxin in incompatible pollen tubes degrading cellular RNAs. Since its identification, the S-RNase gene has been intensively studied and the sequences of a large number of alleles are available in online databases. On the contrary, the male determinant has been only recently identified as a pollen-expressed protein containing a F-box motif, called S-Locus F-box (abbreviated SLF or SFB). Since F-box proteins are best known for their participation to the SCF (Skp1 - Cullin - F-box) E3 ubiquitine ligase enzymatic complex, that is involved in protein degradation through the 26S proteasome pathway, the male determinant is supposed to act mediating the ubiquitination of the S-RNases, targeting them for the degradation in compatible pollen tubes. Attempts to clone SLF/SFB genes in the Pyrinae produced no results until very recently; in apple, the use of genomic libraries allowed the detection of two F-box genes linked to each S haplotype, called SFBB (S-locus F-Box Brothers). Durante il secolo scorso sono state individuate alcune mutazioni per il colore della buccia della varietà William che invece di essere giallo arriva a maturazione con diverse tonalità di colore rosso. L’intensità e la tipologia del fenotipo dovuto a questa mutazione mostra una variabilità all’interno dei diversi cloni rossi di questa cultivar: Max Red Bartlett, Rosired e Sensation. Questa mutazione è ereditabile e usando come genitore uno dei sopra-citati mutanti per il rosso sono state prodotte altre cultivar caratterizzate da buccia rossa come Cascade. Max Red Bartlett presenta una intensa colorazione rossa nelle prime fasi di maturazione per poi striarsi perdendo di lucentezza e non ricoprendo totalmente la superficie del frutto. Max Red Bartlett ha inoltre il problema di regressione del colore. Questa mutazione infatti non è stabile e dopo qualche anno può regredire e presentare il fenotipo di William. Diverso è invece lo sviluppo per esempio di Rosired che durante le prime fasi di accrescimento del frutto è identica a Williams (di colore verde con la parte del frutto rivolta verso il sole leggermente rossastra) per poi virare e mantenere un vivo colore rosso su tutta la superficie del frutto. Questa tesi si è proposta di caratterizzare questa mutazione che coinvolge in qualche modo la via biosintetica per la sintesi del colore. In particolare si è cercato di investigare sui probabili geni della via degli antociani coinvolti e in quale modo vengono espressi durante la maturazione del frutto, inoltre si è cercato di trovare quali specifiche molecole venissero diversamente sintetizzate. Le cultivar utilizzate sono state William e Max Red Bartlett. Di quest’ultima era già disponibile una mappa molecolare, ottenuta sulla popolazione di’incrocio di Abate Fetel (gialla) x MRB (rossa) con AFLP e SSR, quest’ultimi hanno permesso di denominare i diversi linkage group grazie alla sintenia con le altre mappe di pero e di melo.
