Toxoplasma gondii is a protozoan with worldwide prevalence, known to affect a large variety of warm-blooded hosts. However, its ability to induce long-lasting infections in cold-blooded animals remains unclear. The most likely source of infection is through consumption of meat containing tissue cysts or by ingestion of food or water contaminated with oocysts. The current global climate change trend and the progressive degradation of natural habitats are prone to alter the distribution of ectotherm populations over a short period of time, which may favor contact between these animals and the protozoan. In association, alligator meat is considered a delicacy in many regions and its consumption has been previously related to a diversity of foodborne diseases. In that sense, we proposed in this study to search for specific antibodies against T. gondii in serum samples of two common species of alligators from the Brazilian fauna (Melanosuchus niger and Caimam crocodilus). We obtained the serum samples from 84 alligators from the Araguaia region, which were tested by agglutination assays that do not require species-specific secondary antibodies (Modified Agglutination Test – MAT; Indirect Hemagglutination Assay – IHA). From the 84 samples tested, eight (9.5%) were positive by MAT. From those, seven (87.5% of MAT+, 8.3% of the total) were also positive by IHA, reassuring a probable exposure of these animals to the parasite. Direct parasite detection in muscle fragments of one serologically reactive alligator did not yield positive results. Our results provide serological evidence that Brazilian alligators may be exposed to T. gondii and further studies should be performed to elucidate whether alligators are natural hosts of this ubiquitous protozoan parasite., Graphical abstract Image 1, Highlights • Exposure to T. gondii was verified in sera of 84 Brazilian alligators. • The serum samples were analyzed to 2 serological assays: MAT and HAI. • 7 samples were found to be positive in bothserological tests.

Toxoplasma gondii is an obligate intracellular protozoan parasite that causes toxoplasmosis in humans, domestic animals and wild animals. Although there is serological evidence of T. gondii infection in wild animals, little is known about the role of wildlife in the epidemiological chain of this parasite. The South American coati (Nasua nasua) is an omnivorous species capable of adaptation to different environments. It can be found throughout Brazil, and in anthropized environments it can come into close contact with domestic animals. The present study found occurrences of T. gondii antibodies in South American coatis living in the Tietê Ecological Park, in the municipality of São Paulo, Brazil, through the modified agglutination test (MAT). In total, 99 samples were obtained and 70.70% (n = 70) were positive for anti-T. gondii antibodies, with titers that ranged from 50 to 3200. The data from this study indicate that South American coatis are exposed to this parasite. Apesar das evidências sorológicas da infecção por T. gondii em animais selvagens, pouco se sabe sobre o papel da vida selvagem na cadeia epidemiológica deste parasito. Os quatis (Nasua nasua) são uma espécie onívora capaz de se adaptar em diferentes ambientes. Eles são encontrados em todo território brasileiro, e em ambientes antropizados podem apresentar um contato próximo com animais domésticos. O presente estudo verificou a ocorrência de anticorpos anti-T. gondii em quatis habitantes do Parque Ecológico do Tietê, São Paulo, Brasil, pelo Teste de Aglutinação Modificada (MAT). No total, 99 amostras foram obtidas das quais 70,70% (n = 70) foram positivas para anticorpos anti-T. gondii, com títulos de anticorpos que variaram de 50 a 3200. Os dados obtidos neste estudo indicam que quatis sul-americanos são expostos a este parasito.

For this purpose, 226 samples were submitted to mice bioassay and screened by PCR based on 18S rRNA sequences. A total of 15 T. gondii isolates were obtained, including samples from four armadillos (three Dasypus novemcinctus, one Euphractus sexcinctus), three collared anteaters (Tamandua tetradactyla), three whited-lipped peccaries (Tayassu pecari), one spotted paca (Cuniculus paca), one oncilla (Leopardus tigrinus), one hoary fox (Pseudalopex vetulus), one lineated woodpecker (Dryocopus lineatus) and one maned wolf (Chrysocyon brachyurus). DNA from the isolates, originated from mice bioassay, and from the tissues of the wild animal, designated as “primary samples”, were genotyped by PCR–restriction fragment length polymorphism (PCR/RFLP), using 12 genetic markers (SAG1, SAG2, alt.SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L258, PK1, CS3 and Apico). A total of 17 genotypes were identified, with 13 identified for the first time and four already reported in published literature. Results herein obtained corroborate previous studies in Brazil, confirming high diversity and revealing unique genotypes in this region. Given most of genotypes here identified are different from previous studies in domestic animals, future studies on T. gondii from wildlife is of interest to understand population genetics and structure of this parasite.

Sarcocystis neurona and Neospora hughesi are coccidian protozoa that can cause neurological illness in horses in America. In this study we report seroprevalence of Neospora spp. andS. neurona in sera of 333 donkeys from the northeastern region of Brazil. Antibodies to Neospora spp. were detected in 2% (7 donkeys) of 333 sera tested by the indirect fluorescent antibody test (IFAT) with a cut-off dilution of 1:40. Antibodies to S. neurona were found in 3% (10 donkeys) of the samples tested by IFAT (cut-off ≥50) and 21% (69 donkeys) by the direct agglutination test (SAT ≥50). The SAT and IFAT results for S. neurona showed a poor concordance (value of Kappa=0.051). This is the first report ofNeospora spp. antibodies in Brazilian donkeys and the first detection of antibodies against S. neurona in this animal species. Resumo Sarcocystis neurona e Neospora hughesi são protozoários coccídios que infectam equídeos e são responsáveis por doenças neurológicas nessas espécies. Neste estudo, a soroprevalência de infecção porS. neurona e Neospora spp. foi determinada em amostras de 333 soros de jumentos da Região Nordeste do Brasil. Anticorpos contra Neospora spp. foram detectados em 2% (7 jumentos) dos 333 animais examinados pela reação de imunofluorescência indireta (RIFI), com ponto de corte de 40. Anticorpos contra S. neurona foram detectados em 3% (10 jumentos) das amostras pela RIFI (ponto de corte de 50) e em 21% (69 jumentos) pela técnica de aglutinação direta (SAT - ponto de corte de 50). SAT e RIFI, para diagnóstico de S. neurona, apresentaram uma baixa concordância (Kappa = 0,051). Essa é a primeira observação de anticorpos anti-N. caninum em jumentos brasileiros e a primeira detecção de anticorpos contra S. neurona nessa espécie.

In this study, serum samples of 53 wild animals from two different states from the southeast region of Brazil were analyzed for the presence of anti–Toxoplasma gondii antibodies by the modified agglutination test (MAT), with a cut-off of 1 : 5 for birds and of 1 : 25 for mammals. Out of the sampled animals, 27 were birds and 26 were mammals, and from this total, 83% (n = 44) were free-living animals. Antibodies were found in 13 mammals, from which 11 were free-living animals, and in five birds, all of which were free-living. In this study, T. gondii antibodies were detected in four bird species (crested seriema, Cariama cristata; buff-necked ibis, Theristicus caudatus; picazuro pigeon, Patagioenas picazuro; and burrowing owl, Athene cunicularia) and in a giant anteater (Myrmecophaga tridactyla) for the first time.

Toxoplasma gondii affects mainly warm-blooded animals, including birds. Even though previous experimental data indicate that raptors are resistant to clinical infection, there is no information regarding the susceptibility of Brazilian birds of prey to T. gondii. The present study aimed to observe how the crested caracara, a common raptor in Brazil, interacts with T. gondii using an experimental model. Seven crested caracaras, seronegative for T. gondii, were separated into infected (n=5) and control groups (n=2). Birds from the infected group were fed T. gondii-infected Calomys callosus, a rodent present in Brazilian savanna and described as highly susceptible to infection by the parasite, for three consecutive days, while control animals were fed non-infected rodents. All infected birds produced T. gondii-specific IgG antibodies that were firstly detected at day 7 post-infection, with peak production detected between 15 and 30dpi. No significant alterations in clinical and hematological parameters were observed throughout the experimental period, and parasites were sparsely found in muscular tissues after the birds were euthanized. In conclusion, our results demonstrated that crested caracaras are resistant to oral infection with T. gondii, suggesting that the host-parasite relationship between both species has reached a remarkable equilibrium.

Valores basais da PIO foram aferidos em ambos os olhos (M0). Cateterizou-se previamente a artéria braquial para obtenção de parâmetros hemogasométricos e cardiorrespiratórios. Anestesia foi induzida com ISO a 5V% e mantida por 40 minutos com 2,5V%. PIO e amostras de sangue foram avaliadas em diferentes momentos até o final do procedimento. Após recuperação, uma segunda anestesia foi realizada com SEV a 6% e mantida com 3,5%. Os parâmetros foram aferidos nos mesmos momentos estabelecidos previamente. A PIO decresceu significativamente (P=0,012) de M0 em todos os momentos e não houve diferença estatística entre ISO e SEV. Correlações significativas entre PIO e PaCO2 e entre PIO e pH sangüíneo foram observadas apenas para a anestesia com SEV. O pH sangüíneo decresceu paralelamente a PIO, enquanto a PaCO2 aumentou, em carcarás anestesiados com isofluorano e sevofluorano. The intraocular pressure (IOP) and its correlations with arterial carbon dioxide partial pressure (PaCO2) and arterial pH were studied in five crested caracaras (Caracara plancus) anesthetized with isoflurane (ISO) and sevoflurane (SEV). Baseline IOP values were measured in both eyes (M0). Brachial artery was previously catheterized to obtain blood gas and cardiorespiratory analysis. Anesthesia was induced with 5% ISO and maintained with 2.5% for 40 minutes. IOP measurements and blood samples were evaluated in different moments until the end of the procedure. After recovering, a second anesthesia was induced with 6% SEV and maintained with 3.5%. Parameters were evaluated at the same time points of the previous procedure. IOP reduced significantly (P= 0.012) from M0 at all time points and no significative changes were observed between ISO and SEV anesthesias. Correlation between IOP and PaCO2 and between PIO and blood pH were found only for SEV. IOP and blood pH decreased in parallel with IOP, whereas values of PaCO2 increased in caracaras anesthetized with isoflurane and sevoflurane. The occurrence of antibodies against Toxoplasma gondii was determined in donkeys, mules, and horses from different regions of Brazil. Serum samples from 304 donkeys (67.11%), 118 horses (26.05%), and 31 mules (6.84%) were analyzed by means of the indirect fluorescent antibody test (cutoff = 64). Antibodies against T. gondii were detected in 129 equids (28.47%) (82 donkeys, 32 horses, and 15 mules). Tissue samples from 19 seropositive and 50 seronegative animals were obtained in order to isolate the parasite by means of mouse bioassay, and T. gondii was isolated from a donkey. Through genotypic characterization of the isolate, by means of polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP) using 11 genotypic markers, the genotype #163 (TgCkBr220), which has already been described in chickens in Brazil, was identified.

Use of serological tests in the diagnosis of infectious diseases in wild animals has several limitations, primarily the difficulty of obtaining species-specific reagents. Wild canids, such as maned wolves (Chrysocyon brachyurus), are highly predisposed to infection by Toxoplasma gondii and, to a lesser extent, to Neospora caninum. The aim of the present study was to evaluate homologous, heterologous, and affinity conjugates in enzyme-linked immunosorbent assays (ELISAs) and indirect fluorescent antibody tests (IFATs) for detecting immunoglobulin (Ig) G antibodies against T. gondii and N. caninum in maned wolves. Serum samples were obtained from 59 captive animals in Brazil and tested by ELISA for T. gondii serology and IFAT for N. caninum serology using 3 different enzymatic and fluorescent conjugates: homologous (guinea pig anti-maned wolf IgG-peroxidase and -fluorescein isothiocyanate [FITC]), heterologous (rabbit anti-dog IgG-peroxidase and -FITC), and affinity (protein A-peroxidase and -FITC). Seropositivity to T. gondii was comparable among the homologous (69.5%), heterologous (74.6%), and affinity (71.2%) enzymatic conjugates. A significant positive correlation was found between the antibody levels determined by the 3 enzymatic conjugates. The highest mean antibody levels (ELISA index = 4.5) were observed with the protein A-peroxidase conjugate. The same seropositivity to N. caninum (8.5%) was found with the homologous and heterologous fluorescent conjugates, but protein A-FITC was not able to detect or confirm any positive samples with homologous or heterologous conjugates. Our results demonstrate that homologous, heterologous, and affinity conjugates might be used in ELISA for serological assays of T. gondii in wild canids, whereas for N. caninum infection, only the homologous or heterologous fluorescent conjugates have been shown to be useful.

Background and Methods Sera were tested for Brucella spp., Leptospira spp. and Toxoplasma gondii antibodies in 68 free-ranging New World monkeys from a forest fragment of the Brazilian Cerrado. Results and Conclusion All animals were negative for Brucella spp. and Leptospira spp. However, 75% of Alouatta caraya and 16.6% of Callithrix penicillata were positive for T. gondii. The implications for conservation and health management are discussed.

Toxoplasma gondii é um protozoário formador de cistos capaz de infectar diversas espécies de aves e mamíferos domésticos e selvagens, incluindo o homem. Apesar de evidências sorológicas da infecção por T. gondii em animais selvagens, pouco se sabe sobre o papel da vida selvagem na cadeia epidemiológica e tampouco a susceptibilidade das variadas espécies a este parasito. O presente trabalho consistiu no isolamento e caracterização genotípica de T. gondii de tecidos de animais selvagens, de vida livre e de cativeiro, provenientes de diversas localidades do Brasil e na detecção sorológica de anticorpos contra o parasito nas amostras em que foi possível obter o soro. A sorologia foi realizada em 54 amostras de soros de aves e mamíferos de várias espécies por meio do Teste de Aglutinação Modificado (MAT). Deste total, 18 amostras (cinco de aves e 13 de mamíferos) foram positivas para anticorpos anti-T. gondii. Para o isolamento do parasito foi realizado o bioensaio em camundongos. Homogenados de coração e cérebro de cada um dos animais foram submetidos à digestão péptica e inoculados em grupos de cinco camundongos. Tecidos dos camundongos que vinham à óbito eram examinados para constatar a presença de formas de T. gondii. Seis semanas após inoculação, foi colhido sangue dos camundongos para a realização de testes sorológicos (MAT) para detecção de anticorpos anti-T. gondii e dois meses após a inoculação estes animais foram submetidos à eutanásia para a procura por cistos teciduais do parasito. Por meio desta prova biológica foi possível isolar T. gondii em 18 animais selvagens (16 de diversas espécies de mamíferos e duas de aves) provenientes de diferentes localidades. T. gondii foi isolado em uma coruja-buraqueira (Athene cunicularia), um pica-pau-de-banda-branca (Dryocopus lineatus), um gato-do-mato-pequeno (Leopardus tigrinus), um lobo-guará (Chrysocyon brachyurus), uma mucura (Didelphis marsupialis), uma paca (Cuniculus paca), três queixadas (Tayassu pecari), uma raposa-do-campo (Pseudalopex etulus), três tamanduás-mirim (Tamandua tetradactyla), três tatus-galinha (Dasypus novemcinctus) e dois tatuspeba (Eufractus sexcinctus). Dezesseis dos 18 isolados obtidos foram letais para 100% dos camundongos infectados. O isolado de um tatu-peba não causou mortalidade em camundongos e o isolado da coruja-buraqueira causou 50% de mortalidade. A caracterização genotípica dos isolados foi realizada pela técnica de PCR/RFLP utilizando 12 marcadores genotípicos. As amostras primárias dos tecidos provenientes dos animais selvagens que foram positivas na PCR de triagem também foram submetidas à caracterização genotípica. Por meio da PCR/RFLP foi possível obter o genótipo completo de 22 amostras, 15 delas provenientes de isolados e sete de amostras primárias de tecidos. Dos 18 isolados obtidos através do bioensaio, em três (tatu-peba, coruja-buraqueira e mucura) não foi possível obter a caracterização completa de todos os 12 marcadores utilizados. Pela análise das 22 amostras caracterizadas foram observados 17 genótipos diferentes, sendo que 13 deles são inéditos. A mortalidade de camundongos infectados foi comparada com a ocorrência dos diferentes alelos no marcador CS3 nos 15 genótipos provenientes de isolados, dos quais, sete apresentaram o alelo tipo I, seis o alelo tipo II e os alelos u-1 e u-3 foram encontrados em um isolado cada. Todos os isolados apresentaram 100% de mortalidade para os camundongos infectados. Although studies indicate that wild animals are frequently positive for antibodies anti-T. gondii, the role of wild life in the epidemiology of this parasite is not well understood nor the susceptibility of different wild species. The present study aimed to isolate T. gondii from free-living and captive wild birds and mammals from different locations from Brazil, to perform the genotipic characterization of T. gondii found in the analyzed tissue samples and to detect aintibodies anti-T. gondii in samples which were possible to obtain the serum. Serology was performed in 54 serum samples from different species of wild birds and mammals by the Modified Agglutination Test (MAT). From this total, 18 samples (five from birds and 13 from mammals) were seropositive for antibodies anti-T. gondii. For the isolation of the parasite, mice bioassay was performed. Brain and heart homogenates were submitted to peptic digestion and were inoculated in groups of five mice per animal sampled. Tissues of mice that died were examined for the presence of T. gondii. Mice were bled six weeks post-inoculation, and their sera were tested for anti-T. gondii antibodies by MAT. Surviving mice were euthanized two months after the inoculation and their brains were examined for the presence of T.gondii tissue cysts. T. gondii was isolated in 18 wild animals (16 from different species of mammals and two from birds) from different locations. T. gondii was isolated from one burrowing owl (Athene cunicularia), one lineated woodpecker (Dryocopus lienatus), three collared anteater (Tamandua tetradactyla), one hoary fox (Pseudalopex vetulus), one maned wolf (Chrysocyon brachyurus), one oncilla (Leopardus tigrinus), one opossum (Didelphis marsupialis), one paca (Cuniculus paca), three nine-banded armadillo (Dasypus novemcinctus), two six-banded armadillo (Euphractus sexcincticus) and three white-lipped peccary (Tayassu pecari). Sixteen of the 18 isolates were lethal to 100% of inoculated mice. The genotypic characterization was performed using 12 PCR-restriction fragment length polymorphism (RFLP) Primary tissue samples which were positive in a trial PCR were also submitted to genotypic characterization. It was possible, by PCR/RFLP, to obtain the complete genotype of 22 samples, 15 from isolates and seven from primary tissue samples. It was not possible to accomplish the complete genotypic characterization in three isolates; one burrowing owl, one opossum and one sixbanded armadillo. In this 22 samples characterized, a total of 17 different genotypes were found with 13 of them described for the first time. Mortality of infected mice was compared between the different alleles of the marker CS3 in the 15 genotypes originated from isolates, which seven were type I, six were type II and alleles u-1 and u-3 were found in one isolate each. All the isolates presented 100% of mortality in infected mice.

Brazil is one of the regions with the highest prevalences of Toxoplasma gondii in humans and animals. Because free-range chickens become infected by feeding from ground contaminated with oocysts, the prevalence of T. gondii in this host has been widely used as an indicator of the strains prevalent in the environment. The genetic variability among T. gondii isolates from different healthy and sick hosts all over the world has been recently studied. Three clonal genetic lineages (Types I, II and III) were initially recognised as predominant in Western Europe and the United States. T. gondii strains are genetically diverse in South America. In Brazil, recombination plays an important role in strain diversification. The objective of this study was to genetically characterise T. gondii isolates from free-range chickens from Espírito Santo state, Southeast region, Brazil, using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). A total of 44 isolates among 47 previously described isolates (TgCkBr234-281) from free-range chickens were included in this study. Strain typing was performed using 12 PCR-RFLP markers: SAG1, SAG2, alt. SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, Apico and CS3. Eleven genotypes were identified. Ten isolates (23%) were grouped into four novel genotypes. Four isolates, distributed in four counties, corresponded to the Type BrI lineage, the genotype found most frequently in Brazil. No clonal Types I, II or III lineages were found. Two novel genotypes were represented by single isolates. Unique alleles were identified for the markers SAG1, c22-8 and CS3, and for the first time, a unique allele was found for the marker SAG3. Although a large number of T. gondii genotypes have already been identified from a variety of animal hosts in Brazil, new genotypes are continuously identified from different animal species. This study confirmed the diversity of T. gondii in Brazil and demonstrates clonal Type I, II and III lineages are rare in this country.

Toxoplasma gondii isolates are highly diverse in domestic animals from Brazil. However, little is known about the genetics of this parasite from wild mammals in the same region. Reveal genetic similarity or difference of T. gondii among different animal populations is necessary for us to understand transmission of this parasite. Here we reported isolation and genetic characterisation of three T. gondii isolates from wild animals in Brazil. The parasite was isolated by bioassay in mice from tissues of a young male red handed howler monkey (Alouatta belzebul), an adult male jaguarundi (Puma yagouaroundi), and an adult female black-eared opossum (Didelphis aurita). The monkey and the jaguarundi had inhabited the Zoo of Parque Estadual Dois Irmãos, Pernambuco State, Northeastern Brazil, for 1 year and 8 years, respectively. The wild black-eared opossum was captured in São Paulo State, Southeastern Brazil, and euthanised for this study because it was seropositive for T. gondii (titre 1:100 by the modified agglutination test, MAT). Ten PCR-RFLP (Polymerase Chain Reaction-Restriction Fragment Length Polymorphism) markers, SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1 and Apico, were used to genotype the isolates. T. gondii was isolated from the brain and heart homogenate of the monkey, the muscle homogenate of the jaguarundi, and the heart homogenate of the black-eared opossum. This was the first isolation of T. gondii from a neotropical felid from Brazil. The isolate from the monkey (TgRhHmBr1) was not virulent in mice, whereas the isolates from the jaguarundi (TgJagBr1) and the black-eared opossum (TgOpBr1) were virulent in mice. The genotype of the isolate from the monkey has been identified in isolates from a goat and ten chickens in the same region of Brazil, suggesting that it may be a common lineage circulating in this region. The genotypes of the isolates from the jaguarundi and the black-eared opossum have not been previously reported. Although there are already 88 genotypes identified from a variety of animal hosts in Brazil, new genotypes are continuously being identified from different animal species, indicating an extremely high diversity of T. gondii in the population.
