Orestias ascotanensis (Cyprinodontidae) is a teleost pupfish endemic to springs feeding into the Ascotan saltpan in the Chilean Altiplano (3,700 m.a.s.l.) and represents an opportunity to study adaptations to high-altitude aquatic environments. We have de novo assembled the genome of O. ascotanensis at high coverage. Comparative analysis of the O. ascotanensis genome showed an overall process of contraction, including loss of genes related to G-protein signaling, chemotaxis and signal transduction, while there was expansion of gene families associated with microtubule-based movement and protein ubiquitination. We identified 818 genes under positive selection, many of which are involved in DNA repair. Additionally, we identified novel and conserved microRNAs expressed in O. ascotanensis and its closely-related species, Orestias gloriae. Our analysis suggests that positive selection and expansion of genes that preserve genome stability are a potential adaptive mechanism to cope with the increased solar UV radiation to which high-altitude animals are exposed to.

Prenatal stress affects brain functionality and sexual behavior. The medial prefrontal cortex (mPFC) participates in the integration and processing of sexual stimuli. Electroencephalographic (EEG) theta activity has been associated with attention as well as rewarding and sexually motivated states. Considering that the induction of sexual motivation requires attention to, and the adequate processing of, sexual stimuli, this study aimed to evaluate the effects of exposure to stress during the prenatal period on EEG activity in the mPFC during nose pokes in adulthood, actions which are indicators of attention to a receptive female. Eighteen sexually experienced male rats were used, nine stressed prenatally by immobilization during days 14-21 of gestation (stress-exposed group). The other nine formed the control group. All rats were implanted bilaterally in the mPFC (specifically in prelimbic areas) and were allowed one intromission with a receptive female to induce a sexually motivated state before the experimental session. During this session, both nose pokes and non-contact erections in the male rats were evaluated in the presence of an inaccessible receptive female. EEGs were recorded only during nose pokes. The stress-exposed group presented lower nose poke duration, fewer non-contact erections, and lower relative power of the theta band (4-7 Hz) in both prefrontal areas. Considering that the prevalence of this band is associated with attention and motivational processes, these data confirm the deleterious effect of prenatal stress on attention and sexual activation to sexually relevant stimuli in male rats during adulthood.Lay summariesPrenatal stress diminishes attention and activation behaviors in receptive females.Prenatal stress decreases prefrontal activation in the presence of receptive females.Prenatal stress decreases prefrontal theta rhythms in male rats.

El objetivo del presente trabajo es analizar la jurisdicción laboral desde su aparición en la Constitución del 5 de febrero de 1917, sus diversas reglamentaciones desde el 18 de agosto de 1931 cuando surge la primera Ley Federal del Trabajo, hasta la última reforma del 2012. Esto a fin de determinar si se cumplieron con los objetivos y propuestas de los Diálogos por la Justicia Cotidiana, en la reforma Constitucional al artículo 123 apartado A del 24 de febrero del año en curso, y en la iniciativa de reforma a la Ley Federal del Trabajo del 28 de abril de 2016.

Purpose The purpose of this paper is to propose a new typology to leverage the different knowledge gaps existing between universities and industries. This new typology classifies interactions into training-oriented, diffusion-oriented, service-oriented, development-oriented, and research-oriented. Design/methodology/approach The Brazilian 2010 Census data available from the CNPq Research Group Directory were used to test the typology. Findings It was found that, among the five types of interactions proposed, the most frequent were the development-oriented (39 per cent), diffusion-oriented (23 per cent), and research-oriented (16 per cent) ones. These findings indicate that it is likely that university-industry (U-I) interactions have improved in terms of quality in an attempt to attain a joint higher technological development. Originality/value The paper provides the U-I interaction debate with a new perspective about the kinds of interactions that could effectively bridge the knowledge gaps between universities and industries, especially in a developing country context.

Packaging of DNA into chromatin regulates DNA accessibility and consequently all DNA-dependent processes. The nucleosome is the basic packaging unit of DNA forming arrays that are suggested, by biochemical studies, to fold hierarchically into ordered higher-order structures of chromatin. This organization has been recently questioned using microscopy techniques, proposing an irregular structure. To address the principles of chromatin organization, we applied an in situ differential MNase-seq strategy and analyzed in silico the results of complete and partial digestions of human chromatin. We investigated whether different levels of chromatin packaging exist in the cell. We assessed the accessibility of chromatin within distinct domains of kb to Mb genomic regions, performed statistical analyses and computer modelling. We found no difference in MNase accessibility, suggesting no difference in fiber folding between domains of euchromatin and heterochromatin or between other sequence and epigenomic features of chromatin. Thus, our data suggests the absence of differentially organized domains of higher-order structures of chromatin. Moreover, we identified only local structural changes, with individual hyper-accessible nucleosomes surrounding regulatory elements, such as enhancers and transcription start sites. The regulatory sites per se are occupied with structurally altered nucleosomes, exhibiting increased MNase sensitivity. Our findings provide biochemical evidence that supports an irregular model of large-scale chromatin organization.

El envejecimiento es un proceso continuo en el desarrollo del ser humano que es físico, biológico y social que se construye cada día para lograr las capacidades de cada persona. Objetivo: Determinar los factores asociados al envejecimiento activo, en especial las redes sociales, en una población de la sierra ecuatoriana. Metodología: Investigación de enfoque mixto de campo y de corte transversal, en 30 adultos mayores de la Parroquia Antonio José Holguín. Resultados: Se evidencia el bienestar físico el 90% de los participantes son independientes en actividades básicas de la vida diaria, y 63% necesitan cierta ayuda en las actividades instrumentales. Conclusión: Las redes sociales influyen en el envejecimiento activo de las personas de la tercera edad. Existe una relación del bienestar físico, mental y social con el uso de redes sociales como medios de comunicación.

Summary Chromatin-associated non-coding RNAs modulate the epigenetic landscape and its associated gene expression program. The formation of triple helices is one mechanism of sequence-specific targeting of RNA to chromatin. With this study, we show an important role of the nucleosome and its relative positioning to the triplex targeting site (TTS) in stabilizing RNA-DNA triplexes in vitro and in vivo. Triplex stabilization depends on the histone H3 tail and the location of the TTS close to the nucleosomal DNA entry-exit site. Genome-wide analysis of TTS-nucleosome arrangements revealed a defined chromatin organization with an enrichment of arrangements that allow triplex formation at active regulatory sites and accessible chromatin. We further developed a method to monitor nucleosome-RNA triplexes in vivo (TRIP-seq), revealing RNA binding to TTS sites adjacent to nucleosomes. Our data strongly support an activating role for RNA triplex-nucleosome complexes, pinpointing triplex-mediated epigenetic regulation in vivo.

Triplexes are noncanonical DNA structures, which are functionally associated with regulation of gene expression through ncRNA targeting to chromatin. Based on the rules of Hoogsteen base-pairing, polypurine sequences of a duplex can potentially form triplex structures with single-stranded oligonucleotides. Prediction of triplex-forming sequences by bioinformatics analyses have revealed enrichment of potential triplex targeting sites (TTS) at regulatory elements, mainly in promoters and enhancers, suggesting a potential function of RNA–DNA triplexes in transcriptional regulation. Here, we have quantitatively evaluated the potential of different sequences of human and mouse ribosomal RNA genes (rDNA) to form triplexes at different salt and pH conditions. We show by biochemical and biophysical approaches that some of these predicted sequences form triplexes with high affinity, following the canonical rules for triplex formation. We further show that RNA triplex-forming oligos (TFOs) are more stable than their DNA counterpart, and point mutations strongly affect triplex formation. We further show differential sequence requirements of pyrimidine and purine TFO sequences for efficient binding, depending on the G–C content of the TTS. The unexpected sequence specificity, revealing distinct sequence requirements for purine and pyrimidine TFOs, shows that in addition to the Hoogsteen pairing rules, a sequence code and mutations have to be taken into account to predict genomic TTS.

Antecedentes: El infarto agudo del miocardio (IAM) es consecuencia de la disrupción del endotelio vascular que lleva a oclusión del vaso coronario por un trombo. En la coronariografía de algunos de estos pacientes es posible encontrar arterias coronarias sin lesiones significativas. Objetivo: Comparar características basales, presentación clínica, tratamiento y mortalidad intrahospitalaria de pacientes con IAM, con y sin lesiones significativas. Métodos: Se incluyeron 3.194 pacientes registrados en la ficha electrónica GEMI que además del diagnóstico de IAM, hubiesen sido sometidos a estudio coronariográfico. Se consideró "sin lesiones significativas" si ninguna estenosis era al 50%. Resultados: El grupo sin lesiones significativas correspondió a 116 pacientes (3,6%). CHD3 and CHD4 (Chromodomain Helicase DNA binding protein), two highly similar representatives of the Mi-2 subfamily of SF2 helicases, are coexpressed in many cell lines and tissues and have been reported to act as the motor subunit of the NuRD complex (nucleosome remodeling and deacetylase activities). Besides CHD proteins, NuRD contains several repressors like HDAC1/2, MTA2/3 and MBD2/3, arguing for a role as a transcriptional repressor. However, the subunit composition varies among cell- and tissue types and physiological conditions. In particular, it is unclear if CHD3 and CHD4 coexist in the same NuRD complex or whether they form distinct NuRD complexes with specific functions. We mapped the CHD composition of NuRD complexes in mammalian cells and discovered that they are isoform-specific, containing either the monomeric CHD3 or CHD4 ATPase. Both types of complexes exhibit similar intranuclear mobility, interact with HP1 and rapidly accumulate at UV-induced DNA repair sites. But, CHD3 and CHD4 exhibit distinct nuclear localization patterns in unperturbed cells, revealing a subset of specific target genes. Furthermore, CHD3 and CHD4 differ in their nucleosome remodeling and positioning behaviour in vitro. As preocupações, desejos e necessidades dos consumidores à jusante tendem a guiar, cada vez mais, as iniciativas dos produtores à montante e esta é uma realidade também na agricultura, com a seleção de alimentos mais saudáveis, saborosos e com valor agregado por parte dos compradores, gerando um ambiente propício para produtos com estas características. O azeite extra virgem de oliva, cujas características se enquadram nos novos hábitos alimentares, vem assumindo uma posição de destaque entre os consumidores do mundo todo e não é diferente no Brasil. A despeito da pouca tradição na produção olivícola, o Brasil e em especial o Rio Grande do Sul, ao longo da última década tem iniciado uma produção concreta deste alimento, o qual vem chegando lentamente às gôndolas dos supermercados locais e nacionais gozando de uma reputação excelente. Diante, portanto, da possível emergência deste sistema de produção agrícola, da demanda potencial deste produto e da possibilidade de satisfazê-la por meio da produção local em contraposição às importações, entendeu-se que um estudo deste fenômeno recente e por conseguinte pouco analisado pode lançar as bases teóricas para a sua compreensão. Assim, teorias ligadas à governança foram utilizadas para a identificação da organização e coordenação do sistema. Para a análise dos elementos básicos do sistema, o referencial teórico utilizado foi o de sistemas complexos e para a compreensão da relação dos elementos básicos na formação de um sistema complexo, aplicou-se o ferramental da economia institucional evolucionária. Em função deste referencial teórico e da problemática a ser trabalhada, optou-se por um método reconhecidamente institucional com uma abordagem qualitativa, ou seja, um estudo de caso em modo exploratório, realizando entrevistas com participantes considerados chave no sistema e submetendo posteriormente os dados colhidos no campo aos dados secundários e ao referencial teórico. Os resultados encontrados apontam para um gradual florescimento da organização do sistema, com a coordenação não mercadológica do setor, entende-se governança, a cargo dos governos estadual e federal, mas não foram encontrados, até o momento, indícios de uma governança por parte da base, isto é, dos produtores. No que tange ao relacionamento dos elementos para a emergência do sistema, verificou-se que esta se dá por meio das trocas de informações constantes e transações que ocorrem entre os agentes e a economia local, influenciando uns aos outros em uma dinâmica base-topo e no sentido inverso, topo-base, quando o sistema reage às influências e inicia a estimular as estruturas de governança e infra-estrutura institucional. Nesta perspectiva, entende-se que os elementos do sistema que está efetivamente emergindo foram caracterizados, bem como sua relação com a economia local e que é imprescindível, para os participantes, que este sistema complexo seja entendido e tratado como tal, com intervenções adequadas, de modo que possa, desta forma, propiciar ganhos a todos os envolvidos. Consumers concerns, desires and necessities in downstream tend to increasingly guide initiatives of producers in the upstream and this is a reality also in agriculture, with the selection of healthier, tastier, and with higher market value food, which generates a welcoming environment for products with such characteristics. Extra virgin olive oil, whose characteristics fit in the new dietary habits, has been assuming a highlighted position among consumers from the whole world, which is not different in Brazil. Despite having little tradition in olive cultivation, Brazil, especially the state of Rio Grande do Sul, has started since the last decade a solid production of this product, which has slowly reached national and international supermarkets’ shelves while receiving an excellent reputation. Therefore, with the possible emergence of this agricultural production system, and by the high demand of the product as well as the possibility of fulfilling this demand through local production instead of imports, we assume that a study about this recent, and consequently little studied, phenomenon can provide theoretical basis for its comprehension. Hence, governance theories were used to identify the system’s organization and coordination. In order to analyze basic elements of the system, the theoretical background used was that of complex systems; for the comprehension of the basic elements’ relation in the formation of a complex system, the apparatus of institutional-evolutionary economics was applied. Having in mind this theoretical background and the problem studied, we opted for a method acknowledgedly institutional with a qualitative approach i.e. an exploratory case study conducting interviews with participants considered as fundamental in the system, submitting afterwards the data collected in field to the secondary data and the theoretical background. The results obtained point to a gradual blooming of the system’s organization, with coordination not subjected to the market action of the sector, by which is understood governance, of Federal and State responsibility. However, until the moment evidences were not found of a governance of the base i.e. the producers. In reference to the relationship of the elements for the system’s emergence, it was verified that this occurs by means of constant information exchange and transactions that occur between the agents and the local economy. The agents and the local economy influence each other reciprocally in a bottom-up dynamics and in the reverse, topdown, when the system reacts to influences and begins to stimulate governance structures and institutional infrastructure. In this perspective, it is understood that the system’s elements that are effectively emerging were characterized, as well as their relation with local economy. It is crucial for the participants that this complex system can be understood and treated as so, with adequate interventions in a way that it can propitiate gains to all parts involved.

The endoplasmic reticulum (ER) of eukaryotic cells is involved in the synthesis and processing of proteins and lipids in the secretory pathway. These processing events that proteins undergo in the ER may present major limiting steps for recombinant protein production. Increased protein synthesis, accumulation of improperly processed or mis-folded protein can induce ER stress. To cope with ER stress, the ER has quality control mechanisms, such as the unfolded protein response (UPR) and ER-associated degradation to restore homeostasis. ER stress and UPR activation trigger multiple physiological cellular changes. Here we review cellular mechanisms that cope with ER stress and illustrate how this knowledge can be applied to increase the efficiency of recombinant protein expression.

Se realizó una correlación entre World Referente Base (WRB) y Soil Taxonomy (ST), para los suelos de la VII Región del Maule de Chile. Once Grupos de Suelos de Referencia definidos por WRB fueron identificados: Lixisols (499.965 ha), Durisols (62.539 ha). Umbrisols (62.387 ha), Phaeozems (52.823 ha), Cambisols (46.982 ha), Andosols (45.645 ha), Regosols (13.701 ha), Fluvisols (6.908 ha), Luvisols (6.132 ha), Planosols (4.606 ha) y Vertisols (640 ha). Estos suelos fueron localizados principalmente en la Depresión Intermedia (Graben Central) de Chile. Los Andosols se describieron hacia la Cordillera de los Andes y los Luxisols principalmente hacia la Cordillera de la Costa. Las correlaciones más estrechas se observaron entre Lixisols y Alfisols, Umbrisols, Cambisols e Inceptisols, Phaeozems y Mollisols, y finalmente entre Andosols y Andisols. Los Durisols y los restantes Grupos de Suelos de Referencia no mostraron una correlación definida hacia algún Orden de Suelos A soil correlation between World Reference Base (WRB) and Soil Taxonomy (ST), for soils from the 'Region del Maule' (Chile) was carried out. These soils were located mainly in the Central Valley (Graben Central) of Chile. Andosols were described towards the Andean Mountains and Lixisols mainly towards the Coastal Cordillera. Lixisols and Alfisols, Umbrisols, Cambisols and Inceptisols, Phaeozems and Mollisols, and finally Andosols and Andisols were described as the best soil correlated Reference Soil Groups and Soil Order. Durisols and the remaining Reference Soil Groups did not show any defined correlation towards any Soil Order
