Polymeric films containing pomegranate peel extract (PPE) can act as a drug-delivery platform for topical treatment of candidiasis. The composition, mechanical resistance, and in vitro antifungal activity of a polymeric film containing PPE at 1.25 mg.mL−1 were investigated. Films were prepared using a solvent casting technique. The incorporation of PPE in the polymeric matrix gave rise to homogeneous, smooth, shiny, and yellowish-brown films. FTIR spectra of the film containing PPE showed differences without compromising the stability of the extract and the matrix. SEM analysis showed the existence of interruptions in the continuity of the films with extract, which promoted a reduction in the mechanical parameters without significantly changing the tensile strength and elongation at break. Films showed adequate mechanical properties and antifungal activity against Candida albicans, C. glabrata, C. krusei and C. tropicalis.

The present study aimed to evaluate the biological activity of films containing pomegranate peel extract at 1.25% w/w prepared by the electrospinning technique for use as wound dressing. The antimicrobial activity of films loaded with extract was demonstrated by the formation of zones of inhibition of Staphylococcus aureus, Staphylococcus epidermidis, and Candida albicans, suggesting that the release of the extract in the medium occurred at an adequate concentration to inhibit the proliferation of microorganisms. No zone of inhibition was observed for Escherichia coli. These results suggest that the film produced by electrospinning containing 1.25% w/w of the extract might serve as a bioactive wound dressing.

Binary high-density polyethylene (HDPE) composites with natural microtalc (TN), semicrystalline synthetic nanotalc (NTC) and amorphous synthetic (NTA) nanotalc with 1% m/m content were prepared by mixing in the molten state in a twin-screw extruder followed by injection molding. The thermal properties were investigated by thermogravimetric analysis (TGA) and by differential scanning calorimetry (DSC) and showed that the thermal stability of the composites was slightly improved with the incorporation of talcs. The morphology of composites, studied using scanning electron microscopy (SEM), revealed a good dispersion of synthetic talc in composites. The dynamic-mechanical (DMA) properties of the samples were analyzed and showed that the storage modulus was improved with the incorporation of talcs. The tensile strength and impact strength showed a moderate drop for all composites, up to 7.5% and 8.4% respectively, while the elongation at break showed a marked gain of 50% for the HDPE/NTA sample. These results suggest that NTA particles act, with great efficiency, as an internal lubricating agent or plasticizer. This research showed that NTA is a favorable filler when the main objective is to improve the ductility and the rigidity/toughness balance of HDPE and when some loss of tensile strength and impact resistance are tolerated.

In this work, hybrid hydrogels were prepared based on poly(acrylic acid-co-acrylamide) hydrogels (Gel) and self-assembled nanostructured polyaniline (PANI). These hybrid hydrogels were prepared by in situ UV-assisted aniline polymerization during hydrogel network formation in a controlled interfacial reaction. They were then tested as flexible quasi-solid electrolytes with a counter electrode in dye-sensitized solar cells. This electrolyte-counter electrode system was prepared on top of a titanium dioxide layer deposited on a transparent and conductive anode electrode (ITO). Three different nanostructured Gel–PANI hybrid hydrogels were fabricated by employing different PANI concentrations and used for constructing electrolyte–counter electrode systems that displayed conductivities between 0.003 S/cm and 0.02 S/cm. Dye-sensitized solar cells (DSSC) that had the configuration ITO/TiO2/Gel–PANI/ITO were prepared and tested using natural dye extracted from Myrciaria cauliflora. The energy conversion efficiency was evaluated under AM1.5 (100 mW/cm2) light irradiation and was found to reach values above 2.0%. The device’s energy conversion efficiency was found to be higher than those of others with the same natural dye. The process proved to be fast and facile, and it utilized an inexpensive natural dye.

Hybrid delivery systems can release multiple drugs with different profiles and have several applications, including skin dressing. In this work, the co-solvent technique was used for the preparation of nanometric vesicles based on poly(styrene-b-ethylene oxide) block copolymer (BCPVs) containing adapalene (AD). The BCPVs were incorporated into collagen and gelatin matrices together with free AD and silver sulfadiazine (SSD). The AD content of BCPVs and their release capacity were analyzed by using ultraviolet-visible spectroscopy (UV–Vis). The gelatin and collagen matrices were evaluated for their ability to release AD and SSD through an in vitro release study. The obtained results confirmed that the production of empty and AD-loaded BCPVs was viable. The degree of AD encapsulation in BCPVs was 9.0% and the in vitro test revealed a constant, slow, and prolonged release of AD content from AD-loaded BCPVs. The combination of free and encapsulated multiple drugs in hybrid delivery systems based on gelatin and collagen matrices was shown to act as a skin dressing that combined the progressive release of large amounts of drugs within the first hours of use (to restrict infection) with a more prolonged and slow release of AD to enhance skin healing.

In this work, the effect of synthesizing shape memory polyurethanes in aqueous dispersions instead of in organic solvents on the structure and properties of the obtained polymers was investigated. Shape memory polyurethanes based on polycaprolactone diol and isophorone diisocyanate were synthesized by two routes: (1) aqueous dispersion (PU/SMWATER) and (2) dissolution in THF (PU/SMTHF). The samples were analyzed by infrared spectroscopy (FTIR), X-ray diffraction (XRD), static light scattering (SLS), atomic force microscopy (AFM), differential scanning calorimetry (DSC), dynamic mechanical analysis (DMA), and mechanical tests. The aqueous dispersion method led to the production of polyurethanes with a higher degree of phase separation and a higher degree of crystallinity. The morphology of the obtained polyurethanes demonstrated that PU/SMWATER displays a structure with better defined phase separated domains. The polyurethanes exhibited similar average molar masses, soft segment glass transitions (Tgs) and mechanical properties. The lower degrees of phase separation and crystallinity of the PU/SMTHF led to lower values for the shape memory properties (shape recovery ratio (Rr)). The observed changes in the structure of the polyurethanes due to the replacement of organic solvent by an aqueous dispersion during their syntheses confirmed the preparation of shape memory polyurethanes with enhanced shape memory properties. Packaging materials based on natural polymers are becoming popular because these polymers are derived from renewable resources and can display biodegradability. However, natural polymers are typically highly hydrophilic, and packaging films produced with them can absorb water, leading to dimensional changes and reductions in mechanical properties. In this work, films of thermoplastic starch (TPS) and blends of TPS/low-density polyethylene (LDPE) were prepared, and their surfaces were decorated to alter their hydrophilic characteristics. Micro- (polymer microspheres) and nano-components (polycaprolactone electrospun nanofibers) were deposited on the surface of starch and its blends with LDPE. Nanofibers containing silver sulfadiazine were also used to decorate the starch and its blends to change the topography (evaluated by SEM and atomic force microscopy) and to provide anti-bacterial properties. High values of contact angle (CA) and low values of CA hysteresis were measured for the modified surfaces, including a CA higher than 150° and a hysteresis lower than 10°. Therefore, it was demonstrated that the intrinsically hydrophilic surfaces of biodegradable natural polymers could be altered by creating topographical patterns through decoration with nano-/micro-components to generate hydrophobic and even superhydrophobic features that can be useful in minimizing the water absorption of these materials.

Electrospinning technique has been explored to produce nanofibers incorporated with drugs as alternative drug delivery systems for therapeutic purposes in various organs and tissues. Before such systems could potentially be used, their biocompatibility must be evaluated. In this study, dexamethasone acetate-loaded poly(ɛ-caprolactone) nanofibers (DX PCL nanofibers) were developed for targeted delivery in the vitreous cavity in the treatment of retinal diseases. Ocular biocompatibility was tested in vitro and in vivo. DX PCL nanofibers were characterized by scanning electron microscopy (SEM) and Fourier Transform InfraRed spectroscopy (FTIR) and the in vitro drug release from nanofibers was evaluated. The in vitro biocompatibility of DX PCL nanofibers was tested on both ARPE-19 and MIO-M1 cells using the cytotoxicity (MTT) test by morphological studies based on staining of the actin fibers in ARPE-19 cells and GFAP in MIO-M1 cells. The in vivo biocompatibility of DX PCL nanofibers was investigated after intravitreous injection in the rat eye, using spectral domain Optical Coherence Tomography (OCT) imaging of the retina. SEM results indicated that nanometric fibers were interconnected in a complex network, and that they were composed of polymer. FTIR showed that polymer and drug did not chemically interact after the application of the electrospinning technique. PCL nanofibers provided controlled DX release for 10 days. DX PCL nanofibers were not cytotoxic to the ocular cells, allowing for the preservation of actin fibers and GFAP in the cytoplasm of ARPE-19 and MIO-M1 cells, respectively, which are biomarkers of these ocular cell populations. DX PCL nanofibers did not affect the retinal and choroidal structures, and they did not induce abnormalities, hemorrhages, or retinal detachment, suggesting that the nanofibers were well tolerated. In eyes receiving DX PCL nanofibers, SD-OCT images were corroborated with histological analysis of neuroretina and choroid, which are ocular tissues that are extremely sensitive to toxic agents. Finally, the preservation of cone and rod photoreceptors indicated the light sensitivity of the animals. In conclusion, DX PCL nanofibers exhibited ocular biocompatibility and safety in the rodent eye and allow the release of dexamethasone. Further studies are required to appreciate the potential of these new drug delivery systems for the treatment of retinal diseases.

Age-related macular degeneration (AMD) is a degenerative ocular disease that affects the central retina. It is considered the main cause of blindness and loss of vision worldwide. Angiogenic factors are associated with AMD, which has led to the use of antiangiogenic drugs, such as bevacizumab, to treat the disease using frequent intravitreal injections. In the present study, biodegradable core shell nanofibers containing bevacizumab were prepared by the coaxial electrospinning technique. It is thought that the shell could control the release of the drug, while the core would protect and store the drug. Poly(caprolactone) (PCL) and gelatin were used to form the shell of the nanofibers, while poly(vinyl alcohol) (PVA) and bevacizumab comprised the core. The nanofibers were characterized using microscopy techniques, thermal analysis, and FTIR. The results showed that core-shell nanofibers were produced as designed. Bevacizumab activity was evaluated using a chicken embryo chorioallantoic membrane (CAM) assay. An enzyme-linked immunosorbent assay was used to quantify the amount of the drug released from the different nanofibers in vitro. The toxicity of the nanofibers was evaluated in human retinal pigment epithelial (ARPE) cells. The CAM results demonstrated that bevacizumab maintained its antiangiogenic activity when incorporated into the nanofibers. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) tests revealed that the nanofibers showed no cellular toxicity, even in the presence of bevacizumab. The core-shell structure of the nanofibers reduced the release rate of bevacizumab compared with PVA nanofibers. The bevacizumab-loaded biodegradable nanofibers presented interesting properties that would potentially constitute an alternative therapy to intravitreal injections to treat AMD.

In this study a novel approach for improving the photo-stabilization of polymers by incorporation of nanoparticles of a new niobium compound is reported. For the first time, this approach uses the electronic properties of niobium oxyhydroxide to absorb UV light efficiently to avoid the decomposition of the polymers. The innovative results of this work consist of preparing nanoparticles of niobium oxyhydroxide and its modification by treatment with hydrogen peroxide to alter their ability to absorb radiation and act as a photo-stabilizer for preventing polypropylene degradation. Synchrotron radiation was employed: (i) to explore the niobium nanoparticle form and (ii) to investigate the macromolecular structure of neat PP and PP/Nb compounds before and after degradation. The results from this research study using FTIR-ATR, DSC and microscopies, as TEM, SEM and OM, indicate that the incorporation of niobium oxyhydroxide nanoparticles into the PP matrix provides stability against the action of ultraviolet radiation.

Biocompatibility is a requirement for the development of nanofibers for ophthalmic applications. In this study, nanofibers were elaborated using poly(e-caprolactone) via electrospinning. The ocular biocompatibility of this material was investigated. MIO-M1 and ARPE-19 cell cultures were incubated with nanofibers and cellular responses were monitored by viability and morphology. The in vitro biocompatibility revealed that the nanofibers were not cytotoxic to the ocular cells. These cells exposed to the nanofibers proliferated and formed an organized monolayer. ARPE-19 and MIO-M1 cells were capable of expressing GFAP, respectively, demonstrating their functionality. Nanofibers were inserted into the vitreous cavity of the rat’s eye for 10 days and the in vivo biocompatibility was investigated using Optical Coherence Tomography (OCT), histology and measuring the expression of pro-inflammatory genes (IL-1β, TNF-α, VEGF and iNOS) (real-time PCR). The OCT and the histological analyzes exhibited the preserved architecture of the tissues of the eye. The biomaterial did not elicit an inflammatory reaction and pro-inflammatory cytokines were not expressed by the retinal cells, and the other posterior tissues of the eye. Results from the biocompatibility studies indicated that the nanofibers exhibited a high degree of cellular biocompatibility and short-term intraocular tolerance, indicating that they might be applied as drug carrier for ophthalmic use.

Cooperation between researchers in the areas of medical, pharmaceutical and materials science has facilitated the development of pharmaceutical dosage forms that elicit therapeutic effects and protective action with a single product. In addition to optimizing pharmacologic action, such dosage forms provide greater patient comfort and increase success and treatment compliance. In the present work, we prepared semipermeable bioactive electrospun fibers for use as wound dressings containing silver sulfadiazine complexed with β-cyclodextrin in a poly(Ɛ-caprolactone) nanofiber matrix aiming to reduce the direct contact between silver and skin and to modulate the drug release. Wound dressings were prepared by electrospinning, and were subjected to ATR-FT-IR and TG/DTG assays to evaluate drug stability. The hydrophilicity of the fibrous nanostructure in water and PBS buffer was studied by goniometry. Electrospun fibers permeability and swelling capacity were assessed, and a dissolution test was performed. In vitro biological tests were realized to investigate the biological compatibility and antimicrobial activity. We obtained flexible matrices that were each approximately 1.0 g in weight. The electrospun fibers were shown to be semipermeable, with water vapor transmission and swelling indexes compatible with the proposed objective. The hydrophilicity was moderate. Matrices containing pure drug modulated drug release adequately during 24 h but presented a high hemolytic index. Complexation promoted a decrease in the hemolytic index and in the drug release but did not negatively impact antimicrobial activity. The drug was released predominantly by diffusion. These results indicate that electrospun PCL matrices containing β-cyclodextrin/silver sulfadiazine inclusion complexes are a promising pharmaceutical dosage form for wound healing.

The properties of glass fiber-reinforced polymer composites are closely related to the fiber-matrix interface. Interfacial treatments to improve mechanical properties are usually limited to enhance interfacial adhesion. In this work, Layer-by-Layer (LbL) technique was introduced to build a novel interface in polymer composites. Different numbers of bilayers of poly(diallyldimethylammonium chloride) and poly(sodium 4-styrenesulfonate) with carbon nanotubes were deposited through LbL on the surface of woven glass fibers (GFs). Polypropylene composites containing the modified GFs were prepared by compression molding. Thermogravimetric analysis, scanning electron microscopy and Raman spectroscopy proved that multilayers of polymers with carbon nanotubes could be deposited on GFs surface. Mechanical tests on composites with modified GFs revealed an increase in Flexural Modulus and toughness. The overall results attested that the LbL technique can be used to design interfaces with different compositions to perform diverse tasks, such as to improve the stiffness of composites and to encapsulate active nanocomponents.

O polipropileno (PP) tem uma degradação natural lenta e representa um importante constituinte na gestão dos resíduos sólidos, enquanto o poli(hidroxibutirato) (PHB) é um polímero biodegradável, mas tem as desvantagens de dificuldade de processamento e custo. O objetivo deste trabalho foi avaliar o efeito de nanopartículas de TiO2, do método de processamento e do envelhecimento acelerado na estrutura e propriedades da mistura PP/PHB/TiO2. As amostras foram produzidas por etapas de extrusão e injeção e foram caracterizadas por MEV/EDS, FTIR e análise térmica. Os resultados mostraram que a incorporação de 3% de nanopartículas de TiO2 levou a um aumento do tamanho médio da fase dispersa rica em PHB. A mudança na sequência da adição de nanopartículas de TiO2 permitiu guiar a incorporação desses nanocomponentes para a fase rica em PP. As misturas produzidas se mostraram mais estáveis termicamente do que PHB puro e a adição de TiO2 foi eficaz em aumentar a estabilidade térmica dos compósitos. As análises termogravimétricas e por FTIR mostraram que blendas contendo nanopartículas de TiO2 tiveram sua degradação mais afetada nos ensaios de envelhecimento acelerado. Os ensaios de extração com solvente mostraram que um maior conteúdo de produtos de fotodegradação puderam ser extraídos de amostras contendo mais elevadas concentrações de TiO2. Polypropylene (PP) has a slow natural degradation rate. On the other hand, poly(hydroxybutyrate) (PHB) is biodegradable but has the disadvantages of cost and difficulty of processing. The aim of this work was to evaluate the effect of TiO2 nanoparticles, processing conditions and accelerated aging on the structure and properties of PP/PHB/TiO2 mixtures. Samples were prepared by extrusion and injection molding. The incorporation of 3% TiO2 nanoparticles led to an increase in the average size of the dispersed phase rich in PHB. Changes in the sequence of TiO2 nanoparticles incorporation were able to guide the inclusion of nanoparticles to the PP-rich phase. The produced composites were more thermally stable than pure PHB and the incorporation of TiO2 was effective to increase the thermal stability of the composites. Thermal and FTIR analyses showed that blends containing TiO2 nanoparticles had their degradation more affected by accelerated aging treatments. Solvent extraction results revealed that a larger amount of soluble products from the photodegradation could be extracted from samples containing TiO2 submitted to accelerated aging treatments.

Polyurethane waterborne synthesis was performed using a two-step method, commonly referred to as a prepolymer method. Nanocomposites based on waterborne polyurethane and cellulose nanocrystals were prepared by the prepolymer method by altering the mode and step in which the nanofillers were incorporated during the polyurethane formation. The morphology, structural, thermal, and mechanical properties of the resulting nanocomposite films were evaluated by Fourier transform infrared spectroscopy (FTIR), small angle X-ray scattering (SAXS), scanning electron microscopy (SEM), and tensile tests. FTIR results indicated that the degree of interaction between the nanofillers and the WPU through hydrogen bonds could be controlled by the method of cellulose nanocrystal incorporation. Data obtained from SAXS experiments showed that the cellulose nanocrystals as well as the step of the reaction in which they are added influenced the morphology of the polyurethane. The reinforcing effect of CNCs on the nanocomposites depends on their morphology.

Materials based on a mixture of poly(vinylidene fluoride) (PVDF), poly(2-hydroxyethyl methacrylate) (PHEMA) and LiClO4 were produced to evaluate their ionic behavior and morphological structure to determine if they can be used as an electrolyte in electrical devices. FTIR chemical analysis results indicated the presence of interactions between PVDF, PHEMA and LiClO4. MDSC results showed that the transition temperatures of the polymers shifted to higher temperatures for systems containing high concentrations of each polymer. SEM images indicated that there was some miscibility between the polymers especially for the 25 and 75 wt. % compositions. In terms of the electrical performance, the ionic conductivity level of the electrolyte could be controlled by changing the composition of the system.

The subretinal transplantation of retinal pigment epithelial cells (RPE cells) grown on polymeric supports may have interest in retinal diseases affecting RPE cells. In this study, montmorillonite based polyurethane nanocomposite (PU-NC) was investigated as substrate for human RPE cell growth (ARPE-19 cells). The ARPE-19 cells were seeded on the PU-NC, and cell viability, proliferation and differentiation were investigated. The results indicated that ARPE-19 cells attached, proliferated onto the PU-NC, and expressed occludin. The in vivo ocular biocompatibility of the PU-NC was assessed by using the HET-CAM; and through its implantation under the retina. The direct application of the nanocomposite onto the CAM did not compromise the vascular tissue in the CAM surface, suggesting no ocular irritancy of the PU-NC film. The nanocomposite did not elicit any inflammatory response when implanted into the eye of rats. The PU-NC may have potential application as a substrate for RPE cell transplantation.

Targeted drug delivery to the ocular tissues remains a challenge. Biodegradable intraocular implants allow prolonged controlled release of drugs directly into the eye. In this study, we evaluated an anterior suprachoroidal polyurethane implant containing dexamethasone polyurethane dispersions (DX-PUD) as a drug delivery system in the rat model of endotoxin-induced uveitis (EIU). In vitro drug release was studied using PUD implants containing 8%, 20%, and 30% (wt/wt) DX. Cytotoxicity of the degradation products of DX-PUD was assessed on human ARPE-19 cells using 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazolium bromide (MTT) test. Short-term ocular biocompatibility of suprachoroidal DX-PUD implants was evaluated in normal rat eyes. Endotoxin-induced uveitis was then induced in rat eyes preimplanted with DX-PUD. Clinical examination was performed at 24 hours; eyes were used to assess inflammatory cell infiltration and macrophage/microglial activation. Cytokine and chemokine expression in the iris/ciliary body and in the retina was investigated using quantitative PCR. Feasibility of anterior suprachoroidal PUD implantation was also tested using postmortem human eyes. A burst release was followed by a sustained controlled release of DX from PUD implants. By-products of the DX-PUD were not toxic to human ARPE-19 cells or to rat ocular tissues. Dexamethasone-PUD implants prevented EIU in rat eyes, reducing inflammatory cell infiltration and inhibiting macrophage/microglial activation. Dexamethasone-PUD downregulated proinflammatory cytokines/chemokines (IL-1β, IL-6, cytokine-induced neutrophil chemoattractant [CINC]) and inducible nitric oxide synthase (iNOS) and upregulated IL-10 anti-inflammatory cytokine. Polyurethane dispersion was successfully implanted into postmortem human eyes. Dexamethasone-PUD implanted in the anterior suprachoroidal space may be of interest in the treatment of intraocular inflammation.

Bamboo is an alternative sustainable material for use in product design and has been incorporated into the concepts of eco-design. Here, we investigated the mechanical properties and morphologies of low density polyethylene (LDPE)/bamboo flour (BF) composites that were modified with polyethylene-graft-maleic anhydride (PE-g-MA) and glycerol. Scanning electron microscopy (SEM) and tensile tests of the composites demonstrated poor adhesion between the filler and matrix. Contact angle measurement showed that the surface of LDPE was modified by the presence of the load. The thermal stability of the composites was studied by measuring the oxidation induction time (OIT). Preliminary bacterial penetration tests were performed using culture inoculums of E. coli and S. aureus to investigate the natural antibacterial and bacteriostatic properties attributed to bamboo. Furthermore, bamboo may have interesting antioxidant activity with potential for use in food packaging applications.

The purpose of this study was to develop triamcinolone acetonide-loaded polyurethane implants (TA PU implants) for the local treatment of different pathologies including arthritis, ocular and neuroinflammatory disorders. The TA PU implants were characterized by FTIR, SAXS and WAXS. The in vitro and in vivo release of TA from the PU implants was evaluated. The efficacy of TA PU implants in suppressing inflammatory-angiogenesis in a murine sponge model was demonstrated. FTIR results revealed no chemical interactions between polymer and drug. SAXS results indicated that the incorporation of the drug did not disturb the polymer morphology. WAXS showed that the crystalline nature of the TA was preserved after incorporation into the PU. The TA released from the PU implants efficiently inhibited the inflammatory-angiogenesis induced by sponge discs in an experimental animal model. Finally, TA PU implants could be used as local drug delivery systems because of their controlled delivery of TA.

The treatment of posterior segment ocular diseases, such as uveitis, by using eye drops and oral drugs is usually not effective due to the body's natural barriers to drug penetration. In this study, ocular implants to treat uveitis were synthesized by incorporating dexamethasone acetate, an important type of corticoid used in the treatment of some uveitis, into a biodegradable polyurethane containi clay nanoparticles. Biodegradable polyurethane nanocomposites having poly(caprolactone) oligomers as soft segments were obtained by delaminating clay particles within a polyurethane aqueous dispersion. The drug was incorporated into the polymer by dispersing it in the waterborne polyurethane followed by a drying step. Nanoparticles derived from clay were demonstrated to be able to tailor the mechanical properties of polyurethanes to achieve values that can match the properties of ocular soft tissues. Infrared spectra (FTIR) showed that the presence of clay particles was able to change the microphase separation process typical of polyurethanes. X-ray diffraction and small angle x-ray scattering (SAXS) results were explored to show that the incorporation of both dexamethasone acetate and nanocomponents derived from clay led to a less defined two-phase polyurethane. The presence of clay nanoparticles increased the rate of drug release measured in vitro . Human retinal pigment epithelial cells (ARPE-19) were cultured in contact with polyurethanes and polyurethane nanocomposites, and the viability of them (evaluated by using MTT assay after 7 days) showed that no toxic components were released from polyurethanes containing no drugs during the test.

Direct compression is one of the most popular techniques to prepare tablets but only a few commercial excipients are well adapted for this process into controlled release formulations. In the last years, the introduction of new materials for drug delivery matrix tablets has become more important. This paper evaluated the physicochemical and flow properties of new polymeric excipient of ethyl acrylate, methyl methacrylate and butyl metacrylate, synthesized by suspension polymerization using cellulose nanowhiskers as co-stabilizer, to be used as direct compression for modified release tablets. Infrared spectroscopy (FTIR) confirmed the success of the copolymerization reaction. Scanning electron microscopy (SEM) showed that excipient was obtained how spherical beads. Thermal properties of the beads were characterized by thermogravimetric (TG) analysis. Particle size analysis of the beads with cellulose nanowhiskers (CNWB) indicated that the presence of the nanowhiskers led to a reduction of particle size and to a narrower size distribution. In vitro test showed that the nanowhiskers and beads produced are nontoxic. Parameters such as Hausner ratio, Carr's index and cotangent of angle α were employed to characterize the flow properties of CNWB beads. Furthermore, the beads are used to produce tablets by direct compression contained propranolol hydrochloride as model drug. Dissolution tests performed suggested that beads could be used as excipient in matrix tablets with a potential use in drug controlled release.

A porous biodegradable polyurethane nanocomposite based on poly(caprolactone) (PCL) and nanocomponents derived from montmorillonite (Cloisite®30B) was synthesized and tested to produce information regarding its potential use as a scaffold for tissue engineering. Structural and morphological characteristics of this nanocomposite were studied by infrared spectroscopy (FTIR), X-ray diffraction (XRD), small angle X-ray scattering (SAXS) and scanning electron microscopy (SEM). The reaction between polyurethane oligomers with isocyanate endcapped chains and water led to the evolution of CO2, which was responsible for building interconnected pores with sizes ranging from 184 to 387 μm. An in vitro cell-nanocomposite interaction study was carried out using neonatal rat calvarial osteoblasts. The ability of cells to proliferate and produce an extracellular matrix in contact with the synthesized material was assessed by an MTT assay, a collagen synthesis analysis, and the expression of alkaline phosphatase. In vivo experiments were performed by subcutaneously implanting samples in the dorsum of rats. The implants were removed after 14, 21, and 29 days, and were analyzed by SEM and optical microscopy after tissue processing. Histology crosssections and SEM analyses showed that the cells were able to penetrate into the material and to attach to many location throughout the pore structure. In vitro and in vivo tests demonstrated the feasibility for polyurethane nanocomposites to be used as artificial extracellular matrices onto which cells can attach, grow, and form new tissues.

Os polímeros representam uma das classes de materiais mais versáteis disponíveis para aplicações em diversas áreas, inclusive a farmacêutica. Polímeros naturais, naturais modificados e sintéticos são empregados como excipientes farmacêuticos para a formulação de cosméticos e medicamentos de liberação convencional e de liberação modificada. Nos dias atuais, polímeros são desenvolvidos para atuarem como moduladores e direcionadores da liberação de fármacos em sítios específicos no organismo. Polímeros biodegradáveis, bioadesivos, biomiméticos e hidrogéis responsivos têm sido amplamente incluídos em formulações farmacêuticas. Os avanços na idealização de novos Sistemas de Liberação de Fármacos somente são e serão permitidos a partir do desenvolvimento de polímeros projetados especificamente para a área farmacêutica. Neste sentido, o presente artigo visa a revisar e apresentar informações sobre o uso de polímeros em aplicações farmacêuticas que poderão ser úteis no planejamento de novos sistemas com desempenhos superiores.

Three series of shape memory poly(ester–urethane) with varying hard-segment contents were synthesized. The materials were designed to display a three-phase structure consisting of a disperse phase formed by crystallites and hard domains embedded in an amorphous matrix. The initial undeformed morphology was investigated using techniques such as modulated differential scanning calorimetry, Fourier transform infrared spectroscopy, and wide angle X-ray scattering. These techniques were used to determine the phase separation, hydrogen-bonding structure, and crystalline fraction of the specimens prior to thermo-mechanical treatments. The obtained information was correlated with small angle X-ray scattering investigations of morphological changes that occurred during shape memory cycling. The deformation cycle led to the formation of an oriented nanostructure derived from chain alignment. The nanostructure recovered was observed to be triggered by the melting of the crystallites and bulk incompatibility. A relationship between the ability of the studied poly(ester–urethane) specimens to recover their original shape and their original nanostructure was determined.

Reprocessing of single-use intravascular catheters is a common practice in public health services and hospitals. The determination of safe number of reprocessing cycles before the catheter integrity becomes compromised has been a priority issue. The present paper addresses the evaluating molecular and micro-structural integrity of reprocessed cardiac angiographic catheters. The Fourier Transform Infrared Spectroscopy and Scanning Electron Microscopy were carried out to elucidate morphological changes. The tensile test was performed on catheters to examine changes in bulk characteristics. In this work, samples of catheters were reprocessed until nine times and sterilized by hydrogen peroxide plasma. It was observed that the number of hydrogen-bonded carbonyls groups increased in 0.05 u.a. ( p p = 0.0003) at increasing number of reprocessing cycles, also suggestive of crosslinking in this material.

This work aims to investigate the influence of the formation of ion pairing between all-trans retinoic acid (RA) and a lipophilic amine (stearylamine; STE) on the drug encapsulation efficiency (EE) and stability of solid lipid nanoparticles (SLNs). The SLNs were characterized for EE and size. The EE and particle size were significantly improved and reduced, respectively, when the surfactant or co-surfactant concentration increased. However, while the formulation without STE allowed only 13% of RA encapsulation, the EE for RA-STE-loaded SLNs was 94%. The stability studies showed a significant decrease in EE for the SLNs without STE, while, for SLNs loaded with RA and STE, the EE remained constant after 360 days. The interactions among ion pairing components and the lipid matrix were investigated through small-angle X-ray scattering (SAXS). The SAXS analysis revealed the presence of RA in the crystalline form in SLNs without ion pairing, while crystalline RA was not observed in SLNs loaded with RA/amine. Skin irritation studies showed that the SLNs loaded with the ion pairing were significantly less irritating when compared to the marketed RA-cream. This novel SLN formulation represents a promising alternative for topical treatment of acne with RA.

The combination of polymers with macromers derived from inorganic species and oligomers to yield controlled hyper structures can potentially allow the design of materials with tunable mechanical, optoelectronic properties and chemical behavior. In this work, novel hybrid structures were processed based upon the controlled insertion of macromers derived from inorganic groups and oligomers into previously chemically modified polymer frameworks. The chemical procedure involved a series of steps that started with the creation of a functionalized framework to act as host for the construction of the polymer–macromer hyper structures. The designed polymer framework was obtained by incorporating highly polar groups, such as sulfonic acid, into poly(aryl ether sulfones). Following this step, the newly promoted reactive sites were used to insert alkoxysilane groups through reacting modified poly(aryl ether sulfones) with specifically selected silane compounds. The chemical reactions and the obtained novel structures were characterized using NMR, FTIR, AFM and thermal analysis. Results showed that structural parameters such as the concentration of siloxane bonds across the material and the density of cross-links could be controlled by selecting different conditions of reaction. Completely homogeneous and also heterogeneous, but still controlled, structures could be produced using the described procedure. The developed polymer structures containing controlled profiles of concentration, densities and chemical functionalities can have tailorable mechanical properties and chemical activity.

Controlled delivery of drugs is a major issue in the treatment of ocular diseases, such as in the treatment of uveitis. In this study, dexamethasone acetate, an important type of corticoid used in the treatment of some uveitis, was incorporated into biodegradable polyurethanes (PU) having different macromolecular architectures. The biodegradable polyurethanes were obtained by preparing PU aqueous dispersions having poly(caprolactone) and/or poly(ethylene glycol) as soft segments. The drug was incorporated into the polymer by dissolving it in the PU aqueous dispersion. FTIR results showed the presence of the drug in the polymer with its original chemical structure. Small angle X-ray scattering (SAXS) results were explored to show that the incorporation of dexamethasone acetate led to the modification of the nanostructure of the polyurethane having only poly(caprolactone) as the soft segment, while the drug did not change significantly the microphase separated structure of PU having both poly(caprolactone) and poly(ethylene glycol) as soft segments. The evaluation of the release of the drug in vitro demonstrated that the obtained biodegradable polyurethanes were well succeeded in delivering dexamethasone acetate at an almost constant rate for 53 weeks. The presence of poly(ethylene glycol) together with poly(caprolactone) as soft segment in biodegradable PU was able to increase the rate of dexamethasone acetate release when compared to the rate of drug release from PU having only poly(caprolactone).

Bioactive glass nanoparticles-loaded poly(ɛ-caprolactone) nanofibers (BIOG PCL nanofibers) were synthesized and evaluated as substrates for ocular cells (ARPE-19). BIOG PCL nanofibers were characterized using SEM, FTIR, and DSC, and thein vitrodegradation profile was also investigated. Thein vitroocular biocompatibility of nanofibers was exploited in Müller glial cells (MIO-M1 cells) and in chorioallantoic membrane (CAM); and the proliferative capacity, cytotoxicity, and functionality were evaluated. Finally, ARPE-19 cells were seeded onto BIOG PCL nanofibers and they were investigated as supports forin vitrocell adhesion and proliferation. SEM images revealed the incorporation of BIOG nanoparticles into PCL nanofibers. Nanoparticles did not induce modifications in the chemical structure and semicrystalline nature of PCL in the nanofiber, as shown by FTIR and DSC. MIO-M1 cells exposed to BIOG PCL nanofibers showed viability, and they were able to proliferate and to express GFAP, indicating cellular functionality. Moreover, nanofibers were well tolerated by CAM. These findings suggested thein vitroocular biocompatibility and absence of toxicity of these nanofibers. Finally, the BIOG nanoparticles modulated the protein adsorption, and, subsequently, ARPE-19 cells adhered and proliferated onto the nanostructured supports, establishing cell-substrate interactions. In conclusion, the biodegradable and biocompatible BIOG PCL nanofibers supported the ARPE-19 cells.

Investigation of novel biomaterials for bone engineering is based on the development of porous scaffolds, which should match the properties of the tissue that is to be replaced. These materials need to be biocompatible, ideally osteoinductive, osteoconductive, and mechanically well-matched. In the present paper, we report the preparation and characterization of hybrid macroporous scaffold of polyvinyl alcohol (PVA)/bioactive glass through the sol–gel route. Hybrids containing PVA (80, 70 and 60 wt%) and bioactive glass with composition 58SiO2–33CaO–9P2O5 were synthesized by foaming a mixture of polymer solution and bioactive glass via sol–gel precursor solution. PVA with two different degree of hydrolysis (DH), 98.5% (high degree) and 80% (low degree) were also investigated, in order to evaluate the influence of residual acetate group present in polymer chain on the final structure and properties of 3D porous composite produced. The microstructure, morphology and crystallinity of the hybrid porous scaffolds were characterized by X-ray diffraction (XRD), Infrared Fourier Transform spectrometry (FTIR) and Scanning electron microscopy (SEM/EDX) analysis. In addition, specific surface area was assessed by B.E.T. nitrogen adsorption method and mechanical behavior was evaluated by compression tests. Preliminary cytotoxicity and cell viability were also performed by the MTT assay. VERO cell monolayers were grown in 96-well microtiter plates. The results have clearly showed that hybrid foams of polyvinyl alcohol/bioactive glass (PVA/BG) with interconnected macroporous 3D structure were successfully produced. All the tested hybrids of PVA/BG have showed adequate cell viability properties for potential biological applications.

Glass fiber reinforced plastics have been used in many structural applications for the last 40 years. Although extensively used, glass fibers are still expensive for many applications that require low cost raw materials. Glass fibers also have high density and leachable components that can reduce specific mechanical properties (property to weight ratios) and performance of composites. In this work, a novel inorganic short fiber (Silexil) obtained from mineral resources was characterized and tested as a reinforcing agent in plastics. This type of fiber was characterized by electron microscopy, water picnometry, Knoop microhardness and infrared spectroscopy. Fibers were also incorporated into polyurethane and the composites obtained were mechanically tested. Results showed that the novel fiber has a composition close to 100% pure SiO2 and bulk density (1.73 g/cm3) lower than the density of commercial e-glass fibers (2.65 g/cm3). The novel short fibers are 200–600 μm long and have diameters measuring 10 μm. They have also microhardness values comparable to commercial glass fibers and adjustable by changing the temperature of heat treatment. Mechanical properties of the composites showed that the novel fiber is able to reinforce polyurethane. The advantages of this type of fiber when compared to commercial glass fibers would then be: production of low cost composites with lower densities and comparable mechanical properties.

OBJETIVO: Comparar dois novos biomateriais: compósito a 10% (Escola de Engenharia da Universidade Federal de Minas Gerais - UFMG - Brasil) e compósito a 20% (Escola de Engenharia da Universidade Federal de Minas Gerais - UFMG - Brasil), a dois materiais controles: Medpor® (Porex Surgical Inc. - EUA) e Polipore® (Homus Com. Ind. Ltda - Brasil). MÉTODOS: Estudo experimental randomizado duplo cego: em 28 coelhos, 56 implantes foram colocados por via transconjuntival após a remoção de fragmento de cartilagem da reborda orbitária superior de ambas as órbitas, formando-se 4 grupos: Grupo I: Compósito a 10%/Medpor®; Grupo II: Compósito a 10%/Polipore®; Grupo III: Compósito a 20%/Medpor®; Grupo IV: Compósito a 20%/Polipore®. Após permanência média de 19,7 semanas, o material explantado foi avaliado histomorfometricamente em microscopia óptica, e o estudo estatístico foi feito pelo teste t de Student. RESULTADOS: Nenhum implante sofreu extrusão, ocorrendo uma única exposição (Compósito a 20%). Comprovado similar biocompatibilidade dos materiais nos quatro grupos. A histomorfometria no entanto revelou nos compósitos a presença estatisticamente significativa de pseudocápsula mais espessa, com aumento do número de fibroblastos, maior quantidade de colágeno e neovasos, e menor número de células inflamatórias. Foi possível detectar na interface implante-tecido nos compósitos, a presença de camada de hidroxiapatita carbonatada, funcionando como cola biológica. A integração tecidual dos implantes através dos poros foi maior nos controles que nos compósitos. CONCLUSÃO: Os compósitos foram aprovados quanto a biocompatibilidade, e apresentaram adicionalmente algumas vantagens sobre o Medpor® e o Polipore®.PURPOSE: To compare two new biomaterials: Composite 10% (Engineering School - Universidade Federal de Minas Gerais - UFMG - Brazil) and Composite 20% (Engineering School - Universidade Federal de Minas Gerais - UFMG - Brazil), with two similar materials as control: MedporTM (Porex Surgical Inc. - USA) e PoliporeTM (Homus Com. Ind. Ltda - BRASIL). METHODS: In vivo randomized experimental double blind study: a transconjunctival approach was performed in both cartilaginous superior orbital walls of 28 rabbits for 56 implants divided into 4 groups: GI: Composite 10%/MedporTM; GII: Composite 10%/PoliporeTM; GIII: Composite 20%/MedporTM; GIV: Composite 20%/PoliporeTM. The mean rate for implant time of permanence was 19.7 weeks. The Student's t test was used for statistical analysis. RESULTS: Revealed similar biocompatibility of all materials and none of the implants was extruded; one Composite 20% had partial exposure. Histomorphometry showed a greater bioactivity of the composites with significantly thicker fibrous encapsulation, with more collagen and fibroblasts, exuberant angiogenesis, and a lower inflammatory cell mean compared to controls. The presence of precipitation of a hydroxycarbonate apatite layer, which acts as an implant-tissue bond was confirmed. Composites presented a smaller tissular invasion index of the pores compared to controls. CONCLUSIONS: The composites showed a good biocompatibility and some additional advantageover the MedporTM and PoliporeTM controls.

Biossensores têm a função de detectar e analisar tanto quantitativa como qualitativamente a presença de uma certa molécula em um determinado meio, isto com resposta em tempo real e sem etapas intermediárias de processo. Para isso, o biossensor possui sempre em seu conjunto um sistema de reconhecimento biológico que o diferencia de qualquer outro tipo de sensor. O objetivo deste trabalho foi desenvolver hidrogéis condutores a partir da combinação de polianilina com hidrogel poli(metacrilato de hidroxietila) e avaliar alguns aspectos associados à possível utilização deste como base de um biossensor capaz de detectar a presença de frações tóxicas do antígeno do veneno do escorpião Tytius Serrulatus. Hidrogéis condutores foram sintetizados a partir da mistura, em diferentes proporções, de polianilina e poli(metacrilato de hidroxietila) em um solvente comum. A condutividade elétrica (método de quatro pontas), capacidade de inchamento e morfologia (microscopia eletrônica de varredura) foram avaliadas para blendas com diferentes composições. Antígenos relativos ao veneno do escorpião Tytius Serrulatus foram incorporados às blendas condutoras a partir do inchamento dos hidrogéis por soluções aquosas contendo as moléculas de interesse. A imobilização e bioatividade das biomacromoléculas nos hidrogéis foram constatadas através de ensaio imunoabsorvente ELISA cujos resultados foram avaliados por espectroscopia na região do ultravioleta-vísivel. Os resultados mostraram que a incorporação de baixos conteúdos de Pani (20% em massa) em hidrogéis de poli(metacrilato de hidroxietila) foi suficiente para a produção de materiais com elevada condutividade elétrica e grande capacidade de inchamento. Moléculas associadas ao veneno do escorpião Tytius Serrulatus foram imobilizadas com sucesso nos hidrogéis condutores e mantiveram suas capacidades funcionais.
