Abstract

We conducted a systematic review and meta-analysis of the associations between prenatal exposure to persistent organic pollutants (POPs) and childhood obesity. We focused on organochlorines (dichlorodiphenyltrichloroethane [DDT], dichlorodiphenyldichloroethylene [DDE], hexachlorobenzene [HCB], and polychlorinated biphenyls [PCBs]), perfluoroalkyl and polyfluoroalkyl substances (PFAS), and polybrominated diphenyl ethers (PBDEs) that are the POPs more widely studied in environmental birth cohorts so far. We search two databases (PubMed and Embase) through July/09/2021 and identified 33 studies reporting associations with prenatal organochlorine exposure, 21 studies reporting associations with prenatal PFAS, and five studies reporting associations with prenatal PBDEs. We conducted a qualitative review. Additionally, we performed random-effects meta-analyses of POP exposures, with data estimates from at least three prospective studies, and BMI-z. Prenatal DDE and HCB levels were associated with higher BMI z-score in childhood (beta: 0.12, 95% CI: 0.03, 0.21; I2 : 28.1% per study-specific log increase of DDE and beta: 0.31, 95% CI: 0.09, 0.53; I2 : 31.9% per study-specific log increase of HCB). No significant associations between PCB-153, PFOA, PFOS, or pentaPBDEs with childhood BMI were found in meta-analyses. In individual studies, there was inconclusive evidence that POP levels were positively associated with other obesity indicators (e.g., waist circumference).

Abstract

BackgroundBiological aging estimators derived from DNA methylation data are heritable and correlate with morbidity and mortality. Consequently, identification of genetic and environmental contributors to the variation in these measures in populations has become a major goal in the field.ResultsLeveraging DNA methylation and SNP data from more than 40,000 individuals, we identify 137 genome-wide significant loci, of which 113 are novel, from genome-wide association study (GWAS) meta-analyses of four epigenetic clocks and epigenetic surrogate markers for granulocyte proportions and plasminogen activator inhibitor 1 levels, respectively. We find evidence for shared genetic loci associated with the Horvath clock and expression of transcripts encoding genes linked to lipid metabolism and immune function. Notably, these loci are independent of those reported to regulate DNA methylation levels at constituent clock CpGs. A polygenic score for GrimAge acceleration showed strong associations with adiposity-related traits, educational attainment, parental longevity, and C-reactive protein levels.ConclusionThis study illuminates the genetic architecture underlying epigenetic aging and its shared genetic contributions with lifestyle factors and longevity.

Abstract

Hierarchical model fitting has become commonplace for case-control studies of cognition and behaviour in mental health. However, these techniques require us to formalise assumptions about the data-generating process at the group level, which may not be known. Specifically, researchers typically must choose whether to assume all subjects are drawn from a common population, or to model them as deriving from separate populations. These assumptions have profound implications for computational psychiatry, as they affect the resulting inference (latent parameter recovery) and may conflate or mask true group-level differences. To test these assumptions we ran systematic simulations on synthetic multi-group behavioural data from a commonly used multi-armed bandit task (reinforcement learning task). We then examined recovery of group differences in latent parameter space under the two commonly used generative modelling assumptions: (1) modelling groups under a common shared group-level prior (assuming all participants are generated from a common distribution, and are likely to share common characteristics); (2) modelling separate groups based on symptomatology or diagnostic labels, resulting in separate group-level priors. We evaluated the robustness of these approaches to variations in data quality and prior specifications on a variety of metrics. We found that fitting groups separately (assumptions 2), provided the most accurate and robust inference across all conditions. 20. A universal method for accounting greenhouse gas emissions from the activities of higher education institutions using a hybrid life-cycle approach

Abstract

Organisations of all types are significant contributors to international greenhouse gas emissions and the business case for supporting low-carbon practices is gathering pace. Three noteworthy barriers to reporting greenhouse gas emissions in a Higher Education context, namely, time resources, financial resources, and data quality, were found to hinder the production of full greenhouse gas emission assessments encompassing all Scope 1, 2 and, 3 value chain sources. Higher Education Institutions are chosen for study due to their positioning as key components of education systems across the globe. Transcendent of international borders, socio-political regimes, and economic systems, the sector is a significant actor on climate issues. Overseeing education and research activities, institutions are often likened to towns in their size and operational scope. The carbon reduction targets of institutions in the United Kingdom are assessed for pragmatism and were deemed overambitious. Furthermore, they do not account for Scope 3 sources, often the largest proportion of the carbon footprint. A critical assessment of common organisational greenhouse gas assessment methodologies was undertaken and a gap in knowledge highlighted. Whilst theoretical environmental standards are designed to be universally applicable, their practical application to higher education is little explored and in practice, debatable. A theoretical methodology, sympathetic to university environmental practitioners’ requirements was proposed to bridge this gap. Clear and unambiguous guidance that avoided assumptions was developed. Among the numerous benefits, the use of external data sources was reduced and the potential for double counting was eradicated through the use of cut-off criteria, which excluded all paid-for Scope 3 services. The methodology performed favourably against three baseline test parameters: a baseline of 150 hours to complete the emissions assessment, corresponding to financial costs of £24,200 and to satisfy the requirements of the verification standard, ISO14064-part 3. A series of potential strategies, incorporating the use of the proposed methodology (verifiable using industry verification standards) were outlined: whilst the sector should make ambitious pledges to decarbonise along the trajectory set by the Paris agreement, it should not be at the expense of quality research and teaching.

Abstract

DNA methylation has become a biomarker of great interest in the forensic and clinical fields. In criminal investigations, the study of this epigenetic marker has allowed the development of DNA intelligence tools providing information that can be useful for investigators, such as age prediction. Following a similar trend, when the origin of a sample in a criminal scenario is unknown, the inference of an individual’s lifestyle such as tobacco use and alcohol consumption could provide relevant information to help in the identification of DNA donors at the crime scene. At the same time, in the clinical domain, prediction of these trends of consumption could allow the identification of people at risk or better identification of the causes of different pathologies. In the present study, DNA methylation data from the UK AIRWAVE study was used to build two binomial logistic models for the inference of smoking and drinking status. A total of 348 individuals (116 non-smokers, 116 former smokers and 116 smokers) plus a total of 237 individuals (79 non-drinkers, 79 moderate drinkers and 79 drinkers) were used for development of tobacco and alcohol consumption prediction models, respectively. The tobacco prediction model was composed of two CpGs (cg05575921 in AHRR and cg01940273) and the alcohol prediction model three CpGs (cg06690548 in SLC7A11, cg0886875 and cg21294714 in MIR4435–2HG), providing correct classifications of 86.49% and 74.26%, respectively. Validation of the models was performed using leave-one-out cross-validation. Additionally, two independent testing sets were also assessed for tobacco and alcohol consumption. Considering that the consumption of these substances could underlie accelerated epigenetic ageing patterns, the effect of these lifestyles on the prediction of age was evaluated. 35. Aetiology of an unknown liver disease in northern Ethiopia

Abstract

Cases of a novel form of liver disease, localised to a cluster of villages in the Tigray region of Northern Ethiopia, were first reported in 2001. Up to January 2010, 591 cases were recorded, including 228 deaths. Symptoms include epigastric and abdominal swelling. Children are particularly susceptible, with some children having died within three months of the onset of symptoms, whereas some adults report living with the disease for many years. The pattern of spread suggested an environmental toxin was the causative agent, with investigations implicating the plant toxins, pyrrolizidine alkaloids. The ultimate aim of the research presented in this thesis was to determine the aetiology of the disease and a multi-disciplinary approach, involving clinical research, ethnography, biochemical and metabonomic analysis and toxicology, has been employed to achieve this. The disease was first characterised clinically: The disease presents with epigastric pain, initial bloody diarrhoea and progressive ascites with either hepatomegaly or splenomegaly or both. Histology of liver biopsy specimens revealed centrilobular necrosis in acute cases and bile ductular proliferation and fibrosis in chronic cases. Biochemical analysis identified γ-glutamyl transferase as a sensitive indicator of the disease. Ethnographic investigation of the lifestyle of the residents of the affected villages revealed a monotonous diet, the presence of a similar disease among livestock and suggested millet and animal products as potential sources of exposure to the aetiological agent(s). The pesticide DDT was identified as a further potential risk factor for the disease. A global metabonomic analysis of patient urine samples using 1H NMR spectroscopy was conducted and several metabolites associated with the disease were identified. Targeted LC-MS assays were then developed to detect pyrrolizidine alkaloid exposure among patients, either through detection of the parent alkaloid, acetyllycopsamine (AL), in urine samples or extraction and detection of pyrrolic metabolite adducts from whole blood. Relative AL urinary concentrations were found to be significantly higher among cases. AL was shown to be hepatotoxic in the mouse model, and at high doses induced centrilobular hepatic necrosis and metabolic changes similar to that observed in the human disease. Finally, DDT was detected at high levels in patient urine and serum samples. DDT was shown to substantially enhance the toxic effects of AL in the mouse model, through induction of the CYP3A11 enzyme, and in combination with AL induced liver pathology closely resembling the human disease. In conclusion, the available evidence suggests that the disease arises from co-exposure to DDT and pyrrolizidine alkaloids, including AL.

39. The Impact of Threat of Shock-Induced Anxiety on Memory Encoding and Retrieval

Abstract

Anxiety disorders are the most common mental health disorders, and daily transient feelings of anxiety (or "stress") are ubiquitous. However, the precise impact of both transient and pathological anxiety on higher-order cognitive functions, including short- and long-term memory, is poorly understood. A clearer understanding of the anxiety-memory relationship is important as one of the core symptoms of anxiety, most prominently in post-traumatic stress disorder (PTSD), is intrusive reexperiencing of traumatic events in the form of vivid memories. This study therefore aimed to examine the impact of induced anxiety (threat of shock) on memory encoding and retrieval. Eighty-six healthy participants completed tasks assessing: visuospatial working memory, verbal recognition, face recognition, and associative memory. Critically, anxiety was manipulated within-subjects: information was both encoded and retrieved under threat of shock and safe (no shock) conditions. Results revealed that visuospatial working memory was enhanced when information was encoded and subsequently retrieved under threat, and that threat impaired the encoding of faces regardless of the condition in which it was retrieved. Episodic memory and verbal short-term recognition were, however, unimpaired. These findings indicate that transient anxiety in healthy individuals has domain-specific, rather than domain-general, impacts on memory. Future studies would benefit from expanding these findings into anxiety disorder patients to delineate the differences between adaptive and maladaptive responding.
