Abstract: The ASY-EOS experiment at GSI laboratory measured the direct and elliptic flow of neutrons and light charged particles in the reaction 197Au+197 Au at 400 A MeV incident energy. The ratio of elliptic flow of neutrons with respect to that of the light charged particles was used as main experimental observable to probe the density dependence of the symmetry energy term of the nuclear equation of state. Results, obtained by comparison of the experimental data with the UrQMD model predictions, strongly support a moderately soft to linear density dependence of the symmetry energy at suprasaturation densities below 2ρ0.

Flow results obtained for the Au+Au system, in comparison with predictions of the UrQMD transport model, confirm the moderately soft to linear density dependence of the symmetry energy deduced from the earlier FOPI-LAND data.

These results have been confirmed by an independent analysis based on the Tübingen QMD simulations. In order to obtain an improved data set for Au+Au collisions and to extend the study to other systems, a new experiment was carried out at the GSI laboratory by the ASY-EOS collaboration. Abstract: The construction of a new array to study femtoscopy and multi-particle correlations in heavy ion collisions at intermediate energies (E=20-1000 AMeV) has been started at the INFN. The project, named FARCOS (Femtoscope ARray for COrrelations and Spectroscopy) is aimed at developing of a detection system with high pixelation capabilities in order to perform precision measurements of particle correlations for nuclear dynamics and spectroscopy. We present first detection simulations for FARCOS and first results related to the commissioning of CsI(T1) crystals, an important detection stage of each telescope.

Abstract: Directed and elliptic flows of neutrons and light charged particles were measured for the reaction 197Au+197Au at 400 MeV/nucleon incident energy within the ASY-EOS experimental campaign at the GSI laboratory. The detection system consisted of the Large Area Neutron Detector LAND, combined with parts of the CHIMERA multidetector, of the ALADIN Time-of-flight Wall, and of the Washington-University Microball detector. The latter three arrays were used for the event characterization and reaction-plane reconstruction. In addition, an array of triple telescopes, KRATTA, was used for complementary measurements of the isotopic composition and flows of light charged particles. From the comparison of the elliptic flow ratio of neutrons with respect to charged particles with UrQMD predictions, a value \gamma = 0.72 \pm 0.19 is obtained for the power-law coefficient describing the density dependence of the potential part in the parametrization of the symmetry energy. It represents a new and more stringent constraint for the regime of supra-saturation density and confirms, with a considerably smaller uncertainty, the moderately soft to linear density dependence deduced from the earlier FOPI-LAND data. Abstract: Objectives: To describe clinical characteristics of patients with Still's disease treated with methotrexate (MTX) and to assess drug effectiveness evaluating change in disease activity, reduction of inflammatory markers, and glucocorticoid (GC)-sparing effect. Methods: Patients with Still's disease treated with MTX were assessed among those included in AIDA Network Still Disease Registry. Results: In this registry, 171 patients with Still's disease were treated with MTX (males 43.3%, age 37.1 ± 16.0 years). They were mainly characterised by joint features and fever without a prominent multiorgan involvement. MTX was administered with GCs in 68.4% of patients, with other conventional synthetic DMARDs in 6.4%, and with biologic DMARDs in 25.1%. A significant reduction of the modified systemic score was observed, and 38.6% patients were codified as being in clinical remission at the end of follow-up. The concomitant administration of a biologic DMARD resulted a predictor of the clinical remission. Furthermore, a reduction of inflammatory markers and ferritin levels was observed following the administration of MTX. Additionally, a marked reduction of the dosage of concomitant GCs was identified, while 36.7% discontinued such drugs. Male gender appeared as a predictor of GC discontinuation. MTX was discontinued in 12.3% of patients because of adverse effects, and in 12.3% for lack of efficacy. Conclusions: Clinical characteristics of patients with Still's disease treated with MTX were described, mainly joint features and fever without a prominent multiorgan involvement. The clinical usefulness of MTX was reported in reducing the disease activity, decreasing the inflammatory markers, and as GC-sparing agent.

Abstract: The elliptic-flow ratio of neutrons with respect to protons in reactions of neutron rich heavy-ions systems at intermediate energies has been proposed as an observable sensitive to the strength of the symmetry term in the nuclear Equation Of State (EOS) at supra-saturation densities. The recent results obtained from the existing FOPI/LAND data for $^{197}$Au+$^{197}$Au collisions at 400 MeV/nucleon in comparison with the UrQMD model allowed a first estimate of the symmetry term of the EOS but suffer from a considerable statistical uncertainty. In order to obtain an improved data set for Au+Au collisions and to extend the study to other systems, a new experiment was carried out at the GSI laboratory by the ASY-EOS collaboration in May 2011., Comment: Talk given by P. 14. Obesity and Psychiatric Disorders in a Sample of Obese Candidates for Bariatric Surgery in Campania Region

Abstract: Introduction: Obesity is now considered as pandemic. 40% of Italian population is overweight or obese. Many studies emphasize the association of obesity with mental disorders specifically depressive and anxiety disorders, substance use disorder, personality disorder. It has to be distinguished from the mental dimensions: impulsivity, mood, anxiety and body image connected to the emotional regulation system producing eating behaviors. Obesity subjects differ in eating behaviors: gorging, snacking, sweeteating, grazing and binge that are characterised by different level of psychopathology. Mental disorders are also associated to eating behaviors. Bariatric surgery is considered gold standard therapy for obesity. However, follow-up studies underline that the association obesity-mental disorders determines weight loss failure. Methods: 2205 obese subjects underwent psychiatric assessment before bariatric surgery. Patients were divided into two groups as result of psychiatric assessment: 1392 obese subjects without association with mental disorders and 813 with mental disorders. These last (mean age 37,63 SD ± 12,07; 181 M, 632 W; mean body mass index (BMI), 45,16 SD ± 12,14) were enrolled in this study. Every patient underwent psychiatric evaluation. The absence of mental disorders was considered an exclusion criteria. Results: In our sample, Binge Eating Disorder (BED) and Night Eating Syndrome (NES) have the major prevalence. General Anxiety Disorder (GAD) and Major Depressive Disorder (MDD) are also related to obesity. Educational level, in our sample, does not influence in a significant way the mental disorders. Age does not change the psychopathological frame. Conclusion: This study emphasizes that the association between obesity and mental disorders is high among bariatric surgery candidates of our region. The relationship with eating behaviors is connected to the general features of the mental disorders but is not yet clear the reciprocal influences nor why some obese subjects suffer from both and some others does not.

Abstract: The elliptic-flow ratio of neutrons with respect to protons in reactions of neutron rich heavy-ions systems at intermediate energies has been proposed as an observable sensitive to the strength of the symmetry term in the nuclear Equation Of State (EOS) at supra-saturation densities. The recent results obtained from the existing FOPI/LAND data for $^{197}$Au+$^{197}$Au collisions at 400 MeV/nucleon in comparison with the UrQMD model allowed a first estimate of the symmetry term of the EOS but suffer from a considerable statistical uncertainty. In order to obtain an improved data set for Au+Au collisions and to extend the study to other systems, a new experiment was carried out at the GSI laboratory by the ASY-EOS collaboration in May 2011., Comment: Talk given by P. Abstract: The design of modern multi-detector arrays for Intermediate Energy Nuclear Physics experiments has to face the problem of the improvement of the Charge and Mass Identification of Heavy and Light Charged Products. Most of the detector arrays developed for the identification of mass and atomic number of the emitted fragments, use detection units composed by thin silicon detectors and by thick scintillator crystals read-out by photodiodes. The light output of the CsI(TI) crystal can be modeled - at least in a given energy range - with a combination of two exponential functions with different time constants. The possibility of storing the digitized waveforms allows performing further off-line analyses. In the present work we determine - by direct least-square fitting of the base-line subtracted output waveforms - the four-vector defining the light output (τ f, τ s, h f, h s). In this way it is possible to provide an experimental determination of the scintillator parameters for a wide energy range and for different particle types. The proposed method has been applied to the waveforms acquired with the CHIMERA 4π detector array during runs at the INFN Laboratori Nazionali del Sud Superconducting Cyclotron in Catania, Italy with different beam/target pairs. In this paper we present the results obtained with a 21.5 MeV/u 20Ne beam bombarding a 12C target. © 2011 IEEE.

Abstract: The elliptic-flow ratio of neutrons with respect to protons or light complex particles in reactions of heavy ions at pre-relativistic energies has been proposed as an observable sensitive to the strength of the symmetry term of the nuclear equation of state at supra-saturation densities. In the ASY-EOS experiment at the GSI laboratory, flows of neutrons and light charged particles were measured for 197Au+197Au collisions at 400 MeV/nucleon. Flow results obtained for the Au+Au system, in comparison with predictions of the UrQMD transport model, confirm the moderately soft to linear density dependence of the symmetry energy deduced from the earlier FOPI-LAND data.

Abstract: The design of modern multi-detector arrays for Intermediate Energy Nuclear Physics experiments has to face the problem of the improvement of the Charge and Mass Identification of Heavy and Light Charged Products. We are fixing our attention to the products stopping in the silicon detector that can be identified in mass by measuring Energy and Time of Flight - (E, ToF) technique - and in charge by measuring the Energy and the Rise Time of the signal - (E, RT) technique With this last technique, one of the points of largest physical interest is the minimum energy of the reaction products that allows charge identification. To this purpose we have done some tests, using the ion beam of the Superconducting Cyclotron of Laboratori Nazionali del Sud in Catania, aimed to evaluate the performance of the CHIMERA Silicon detectors in reverse mounting. In this paper we will discuss the experimental results obtained in reverse mounting in a true beam experiment and we will critically analyze the advantages and drawbacks of reverse mounting w.r.t. front mounting, correlating the measured detector properties in terms of physical properties and biasing conditions of the detector itself.

Abstract: ObjectiveIn the light of the current COVID-19 epidemic and the availability of effective vaccines, this study aims to identify factors associated with non-response to anti-SARS-CoV-2 vaccines as immunological alteration associated with immune rheumatic diseases (IRD) and immunosuppressive medications may impair the response to vaccination.MethodsVolunteers in the health profession community with IRD, age, and sex-matched controls (CTRL) who underwent vaccination with two doses of BNT162b2 were recruited for this study. Anti-Trimeric Spike protein antibodies were assayed eight ± one weeks after the second vaccine dose. Univariate and logistic regression analyses were performed to identify factors independently associated with non-response and low antibody titers.ResultsSamples were obtained from 237 IRD patients (m/f 73/164, mean age 57, CI 95% [56-59]): 4 autoinflammatory diseases (AI), 62 connective tissue diseases (CTD), 86 rheumatoid arthritis (RA), 71 spondylarthritis (SpA) and 14 vasculitis (Vsc). 232 CTRL were recruited (m/f 71/161, mean age 57, CI 95% [56-58]). Globally, IRD had a lower seroconversion rate (88.6% vs 99.6%, CI 95% OR [1.61-5.73], pConclusionThe response to anti-SARS-CoV-2 vaccines is often impaired in IRD patients under treatment and may pose them at higher risk of severe COVID-19. Specific vaccination protocols are desirable for these patients.

Abstract: International audience; In the ASY-EOS experiment flows of neutrons and light charged particles were measured for (197)Au+(197)Au collisions at 400 MeV/nucleon, in order to investigate the strength of the symmetry term of the nuclear equation of state at supra-saturation densities. By comparing the experimental data with the UrQMD transport model predictions, we have extracted a new constraint in agreement with the moderately soft to linear density dependence obtained in the former analysis on FOPI-LAND data, but reducing the associated uncertainty by a factor ∼ 2.

Abstract: Directed and elliptic flows of neutrons and light charged particles were measured for the reaction 197Au+197Au at 400 MeV/nucleon incident energy within the ASY-EOS experimental campaign at the GSI laboratory. The detection system consisted of the Large Area Neutron Detector LAND, combined with parts of the CHIMERA multidetector, of the ALADIN Time-of-flight Wall, and of the Washington-University Microball detector. The latter three arrays were used for the event characterization and reaction-plane reconstruction. In addition, an array of triple telescopes, KRATTA, was used for complementary measurements of the isotopic composition and flows of light charged particles. From the comparison of the elliptic flow ratio of neutrons with respect to charged particles with UrQMD predictions, a value \gamma = 0.72 \pm 0.19 is obtained for the power-law coefficient describing the density dependence of the potential part in the parametrization of the symmetry energy. It represents a new and more stringent constraint for the regime of supra-saturation density and confirms, with a considerably smaller uncertainty, the moderately soft to linear density dependence deduced from the earlier FOPI-LAND data. Abstract: International audience; The ASY-EOS experiment at GSI laboratory measured the direct and elliptic flow of neutrons and light charged particles in the reaction 197Au+197 Au at 400 A MeV incident energy. The ratio of elliptic flow of neutrons with respect to that of the light charged particles was used as main experimental observable to probe the density dependence of the symmetry energy term of the nuclear equation of state. Results, obtained by comparison of the experimental data with the UrQMD model predictions, strongly support a moderately soft to linear density dependence of the symmetry energy at suprasaturation densities below 2ρ0.

Abstract: International audience; The elliptic-flow ratio of neutrons with respect to protons or light complex particles in reactions of heavy ions at pre-relativistic energies has been proposed as an observable sensitive to the strength of the symmetry term of the nuclear equation of state at supra-saturation densities. In the ASY-EOS experiment at the GSI laboratory, flows of neutrons and light charged particles were measured for ^197Au+^197Au collisions at 400 MeV/nucleon. Flow results obtained for the Au+Au system, in comparison with predictions of the UrQMD transport model, confirm the moderately soft to linear density dependence of the symmetry energy deduced from the earlier FOPI-LAND data.

Abstract: We investigated the merits of different event parameters in the identification of Light Charged Particles (LCPs) with CsI(Tl) scintillators read out by photodiodes at high incident energy (400 MeV/u). This investigation is made possible by digital signal processing the output signals. As in the conventional analogue case, the digitized signals allow the discrimination of light charged particles by computing the fast and slow components. In addition other identification parameters as the rise time of the output pulses of the CsI(Tl) come out nearly for free. Aim of this paper is the investigation of novel identification plots and the probe of their merits, in particular at relativistic energies. When the energies of the light charged particles exceed those corresponding to their ranges in CsI(Tl), the points related to the punching-through particles gather in the corresponding scatter plots giving rise to a cusp. The punch-through energies provide a complete set of data for the energy calibration of the reaction products stopped in the CsI(Tl). The obtained results suggest that at relativistic beam energies the energy vs. rise time computation provide an efficient figure for the discrimination in charge and mass of LCPs.

Abstract: The elliptic-flow ratio of neutrons with respect to protons in reactions of neutron rich Heavy-Ion at intermediate energies has been recently proposed as an observable sensitive to the strength of the symmetry term in the nuclear equation of state (EOS) at supra-saturation densities. The recent results obtained from the existing FOPI/LAND data for 197Au+197Au collisions at 400 MeV/nucleon in comparison with the UrQMD model allowed a first estimate of the symmetry term of the EOS but suffer from a considerable statistical uncertainty. In order to obtain an improved data set for Au+Au collisions and to extend the study to other systems, a new experiment was carried out at the GSI laboratory by the ASY-EOS collaboration in May 2011. © Owned by the authors, 2012.

Abstract: In recent years, immune checkpoint inhibitors have significantly changed the field of oncology, emerging as first-line treatment, either alone or in combination with other regimens, for numerous malignancies, improving overall survival and progression-free survival in these patients. However, immune checkpoint inhibitors might also cause severe or fatal immune-related adverse events, including adverse cardiovascular events. Initially, myocarditis was recognized as the main immune checkpoint inhibitor-related cardiac event, but our knowledge of other potential immune-related cardiovascular adverse events continues to broaden. Recently, preclinical and clinical data seem to support an association between immune checkpoint inhibitors and accelerated atherosclerosis as well as atherosclerotic cardiovascular events such as cardiac ischemic disease, stroke, and peripheral artery disease. In this review, by offering a comprehensive overview of the pivotal role of inflammation in atherosclerosis, we focus on the potential molecular pathways underlying the effects of immune checkpoint inhibitors on cardiovascular diseases. Moreover, we provide an overview of therapeutic strategies for cancer patients undergoing immunotherapy to prevent the development of cardiovascular diseases.

Abstract: To coordinate cellular physiology, eukaryotic cells rely on the rapid exchange of molecules at specialized organelle-organelle contact sites 1,2 . Endoplasmic reticulum-mitochondrial contact sites (ERMCSs) are particularly vital communication hubs, playing key roles in the exchange of signalling molecules, lipids and metabolites 3,4 . ERMCSs are maintained by interactions between complementary tethering molecules on the surface of each organelle 5,6 . However, due to the extreme sensitivity of these membrane interfaces to experimental perturbation 7,8 , a clear understanding of their nanoscale organization and regulation is still lacking. Here we combine three-dimensional electron microscopy with high-speed molecular tracking of a model organelle tether, Vesicle-associated membrane protein (VAMP)-associated protein B (VAPB), to map the structure and diffusion landscape of ERMCSs. We uncovered dynamic subdomains within VAPB contact sites that correlate with ER membrane curvature and undergo rapid remodelling. We show that VAPB molecules enter and leave ERMCSs within seconds, despite the contact site itself remaining stable over much longer time scales. This metastability allows ERMCSs to remodel with changes in the physiological environment to accommodate metabolic needs of the cell. An amyotrophic lateral sclerosis-associated mutation in VAPB perturbs these subdomains, likely impairing their remodelling capacity and resulting in impaired interorganelle communication. These results establish high-speed single-molecule imaging as a new tool for mapping the structure of contact site interfaces and reveal that the diffusion landscape of VAPB at contact sites is a crucial component of ERMCS homeostasis., (© 2024. Abstract: Dysregulated neuronal excitability is a hallmark of amyotrophic lateral sclerosis (ALS). We sought to investigate how functional changes to the axon initial segment (AIS), the site of action potential generation, could impact neuronal excitability in ALS human induced pluripotent stem cell (hiPSC) motor neurons. We find that early TDP-43 and C9orf72 hiPSC motor neurons show an increase in the length of the AIS and impaired activity-dependent AIS plasticity that is linked to abnormal homeostatic regulation of neuronal activity and intrinsic hyperexcitability. In turn, these hyperactive neurons drive increased spontaneous myofiber contractions of in vitro hiPSC motor units. In contrast, late hiPSC and postmortem ALS motor neurons show AIS shortening, and hiPSC motor neurons progress to hypoexcitability. At a molecular level, aberrant expression of the AIS master scaffolding protein ankyrin-G and AIS-specific voltage-gated sodium channels mirror these dynamic changes in AIS function and excitability. 45. Surgical oncological emergencies in octogenarian patients.

Abstract: Background: Surgical oncological emergencies represent a frequent challenge in acute settings, with postoperative courses characterized by high morbidity and mortality. An accurate selection of patients who could benefit from surgery is essential to avoid unnecessary invasive treatment. In this study, we tried to determine if advanced age (>80 years) represents a risk factor for negative short-term outcome in patients undergoing emergency surgery for acute abdominal oncological illness., Methods: We retrospectively analyzed the records of patients who underwent emergency oncological surgery at the Department of Acute Care Surgery of Padua General Hospital from January 2018 to December 2022. One hundred two cancer patients were included in the study. Among them, 42 were aged ≥80 years (41%). Multiple preoperative and postoperative parameters were recorded, and the follow-up period was at least 90 days. Multivariate logistic regression analyses were used to identify factors associated with short-term postoperative outcomes., Results: In the octogenarian group, 30-day mortality was 11% vs. 9.5% in the younger group [p = not significant ( ns )] and 90-day mortality was 17.6% in the octogenarian group vs. 20.5% in the younger group (p = ns ). Postoperative morbidity and hospital length of stay were not significantly different in the two groups. Low albumin levels [odds ratio (OR) 30.6, 9.51-87.07] and elevated lactate dehydrogenase (LDH) levels (OR 26.4, 9.18-75.83) were predictive for short-term mortality in surgical oncological emergencies., Conclusion: Advanced age is not a risk factor for negative outcomes in surgical oncological emergencies. Therefore, surgical options should be considered in octogenarians with oncological emergencies and acceptable clinical conditions. Abstract: Diabetes mellitus, a group of metabolic disorders characterized by high levels of blood glucose caused by insulin defect or impairment, is a major risk factor for cardiovascular diseases and related mortality. Patients with diabetes experience a state of chronic or intermittent hyperglycemia resulting in damage to the vasculature, leading to micro- and macro-vascular diseases. These conditions are associated with low-grade chronic inflammation and accelerated atherosclerosis. Several classes of leukocytes have been implicated in diabetic cardiovascular impairment. Although the molecular pathways through which diabetes elicits an inflammatory response have attracted significant attention, how they contribute to altering cardiovascular homeostasis is still incompletely understood. In this respect, non-coding RNAs (ncRNAs) are a still largely under-investigated class of transcripts that may play a fundamental role. This review article gathers the current knowledge on the function of ncRNAs in the crosstalk between immune and cardiovascular cells in the context of diabetic complications, highlighting the influence of biological sex in such mechanisms and exploring the potential role of ncRNAs as biomarkers and targets for treatments. 47. Enrichment of human embryonic stem cell-derived V3 interneurons using an Nkx2-2 gene-specific reporter.

Abstract: V3 spinal interneurons are a key element of the spinal circuits, which control motor function. However, to date, there are no effective ways of deriving a pure V3 population from human pluripotent stem cells. Here, we report a method for differentiation and isolation of spinal V3 interneurons, combining extrinsic factor-mediated differentiation and magnetic activated cell sorting. We found that differentiation of V3 progenitors can be enhanced with a higher concentration of Sonic Hedgehog agonist, as well as culturing cells in 3D format. To enable V3 progenitor purification from mixed differentiation cultures, we developed a transgene reporter, with a part of the regulatory region of V3-specific gene Nkx2-2 driving the expression of a membrane marker CD14. We found that in human cells, NKX2-2 initially exhibited co-labelling with motor neuron progenitor marker, but V3 specificity emerged as the differentiation culture progressed. At these later differentiation timepoints, we were able to enrich V3 progenitors labelled with CD14 to ~ 95% purity, and mature them to postmitotic V3 interneurons. This purification tool for V3 interneurons will be useful for in vitro disease modeling, studies of normal human neural development and potential cell therapies for disorders of the spinal cord., (© 2023. Abstract: Human pluripotent stem cell-derived hepatocytes (hPSC-Heps) may be suitable for treating liver diseases, but differentiation protocols often fail to yield adult-like cells. We hypothesised that replicating healthy liver niche biochemical and biophysical cues would produce hepatocytes with desired metabolic functionality. Using 2D synthetic hydrogels which independently control mechanical properties and biochemical cues, we found that culturing hPSC-Heps on surfaces matching the stiffness of fibrotic liver tissue upregulated expression of genes for RGD-binding integrins, and increased expression of YAP/TAZ and their transcriptional targets. Alternatively, culture on soft, healthy liver-like substrates drove increases in cytochrome p450 activity and ureagenesis. Knockdown of ITGB1 or reducing RGD-motif-containing peptide concentration in stiff hydrogels reduced YAP activity and improved metabolic functionality; however, on soft substrates, reducing RGD concentration had the opposite effect. Furthermore, targeting YAP activity with verteporfin or forskolin increased cytochrome p450 activity, with forskolin dramatically enhancing urea synthesis. hPSC-Heps could also be successfully encapsulated within RGD peptide-containing hydrogels without negatively impacting hepatic functionality, and compared to 2D cultures, 3D cultured hPSC-Heps secreted significantly less fetal liver-associated alpha-fetoprotein, suggesting furthered differentiation. Abstract: Background: Cell therapy with autologous peripheral blood mononuclear cells (PB-MNCs) may help restore limb perfusion in patients with diabetes mellitus and critical limb-threatening ischemia (CLTI) deemed not eligible for revascularization procedures and consequently at risk for major amputation (no-option). Fundamental is to establish its clinical value and to identify candidates with a greater benefit over time. Assessing the frequency of PB circulating angiogenic cells and extracellular vesicles (EVs) may help in guiding candidate selection., Methods: We conducted a prospective, non-controlled, observational study on no-option CLTI diabetic patients that underwent intramuscular PB-MNCs therapy, which consisted of more cell treatments repeated a maximum of three times. The primary endpoint was amputation rate at 1 year following the first treatment with PB-MNCs. We evaluated ulcer healing, walking capability, and mortality during the follow-up period. We assessed angiogenic cells and EVs at baseline and after each cell treatment, according to primary outcome and tissue perfusion at the last treatment [measured as transcutaneous oxygen pressure (TcPO 2 )]., Results: 50 patients were consecutively enrolled and the primary endpoint was 16%. TcPO 2 increased after PB-MNCs therapy (17.2 ± 11.6 vs 39.1 ± 21.8 mmHg, p < .0001), and ulcers healed with back-to-walk were observed in 60% of the study population (88% of survivors) during follow-up (median 1.5 years). Patients with a high level of TcPO 2 (≥ 40 mmHg) after the last treatment showed a high frequency of small EVs at enrollment., Conclusions: In no-option CLTI diabetic patients, PB-MNCs therapy led to an improvement in tissue perfusion, a high rate of healing, and back-to-walk. Coupling circulating cellular markers of angiogenesis could help in the identification of patients with a better clinical benefit over time., (© 2022. The Author(s).)

50. Serological Response to BNT162b2 Anti-SARS-CoV-2 Vaccination in Patients with Inflammatory Rheumatic Diseases: Results From the RHEUVAX Cohort.

Abstract: Objective: In the light of the current COVID-19 epidemic and the availability of effective vaccines, this study aims to identify factors associated with non-response to anti-SARS-CoV-2 vaccines as immunological alteration associated with immune rheumatic diseases (IRD) and immunosuppressive medications may impair the response to vaccination., Methods: Volunteers in the health profession community with IRD, age, and sex-matched controls (CTRL) who underwent vaccination with two doses of BNT162b2 were recruited for this study. Anti-Trimeric Spike protein antibodies were assayed eight ± one weeks after the second vaccine dose. Univariate and logistic regression analyses were performed to identify factors independently associated with non-response and low antibody titers., Results: Samples were obtained from 237 IRD patients (m/f 73/164, mean age 57, CI 95% [56-59]): 4 autoinflammatory diseases (AI), 62 connective tissue diseases (CTD), 86 rheumatoid arthritis (RA), 71 spondylarthritis (SpA) and 14 vasculitis (Vsc). 232 CTRL were recruited (m/f 71/161, mean age 57, CI 95% [56-58]). Globally, IRD had a lower seroconversion rate (88.6% vs 99.6%, CI 95% OR [1.61-5.73], p<0.001) and lower antibody titer compared to controls (median (IQR) 403 (131.5-1012) versus 1160 (702.5-1675), p<0.001). After logistic regression, age, corticosteroid (CCS), Abatacept and Mycophenolate Mofetil (MMF) use were associated with non-response. Lower antibody titer was associated with the use of MMF, ABA, CCS, Rituximab, tumor necrosis factor inhibitor, JAK inhibitors, and higher age., Conclusion: The response to anti-SARS-CoV-2 vaccines is often impaired in IRD patients under treatment and may pose them at higher risk of severe COVID-19.
