Ternstroemia es un género con ca. 141 especies de los trópicos húmedos del mundo, con una especie que alcanza Corea y Japón. Una conjunción de ecosistemas higrófilos y sistemas de montaña con alta diversidad e historias geológicas complementarias, convierte a Venezuela en el país con el mayor número de especies a nivel global, incluyendo una alta densidad de taxones (ca. 2,8 × 10-5spp./km2) y un elevado grado de endemismo (32%). Se reconocen 28 especies, una de ellas descrita aquí (Ternstroemia igapicola sp. nov.), y se proponen seis sinónimos nuevos (Ternstroemia brevipes var. blanchetii, T. camelliifolia var. minor, T. grandiosa, T. laevigata, [previamente sinonimizada, pero bajo un concepto diferente], T T. steyermarkii var. submontana y T. unilocularis), cinco lectotipificaciones (T. andina, T. brevipes, T. camelliifolia, T. meridionalis, T. meridionalis var. nigricans y T. schomburgkiana) y siete registros nuevos por país (T. klugiana, T. prancei y T. schomburgkiana para Venezuela, T. discoidea y T. klugiana para Brasil, T. crassifolia para Colombia y T. distyla para Perú). Ternstroemia gleasoniana es confirmada para Venezuela, mientras T . brasiliensis y T. punctata, previamente reportadas para ese país, son excluidas. Una clave, cuatro ilustraciones, tres mapas y una tabla permiten diferenciar las especies aceptadas.

Enquadramento: O modelo de cuidados prestados durante o parto na água é interpessoal, integral e respeitado. Objetivo: Conhecer a perceção das mulheres sobre os cuidados recebidos pelas enfermeiras especialistas de saúde materna e obstétrica durante o parto na água na perspetiva da teoria de enfermagem de Jean Watson. Metodologia: Investigação empírica, qualitativa, interpretativa com 24 mulheres portuguesas que vivenciaram o parto na água no hospital ou no domicílio, com tratamento dos dados baseados no método de análise de conteúdo de Bardin. Resultados: A partir das narrativas, emergiram cinco categorias - ambiente acolhedor; a mulher no controlo do seu próprio parto; relação de confiança; expectativas correspondidas e o companheiro como parte do processo. Conclusão: O Processo Clinical Caritas foi percebido nas narrativas através do respeito e do carinho durante os cuidados prestados, proporcionando uma relação e cooperação, que promoveu posteriormente uma cura interior através desse processo de cuidados. A perceção das participantes do estudo quanto aos cuidados recebidos revelou um ambiente seguro, íntimo, respeitado, acolhedor, sem intervenções desnecessárias, favorável e amparado pelo companheiro.

As the SARS-CoV-2 pandemic continues to rage worldwide, the emergence of numerous variants of concern (VOC) represents a challenge for the vaccinal protective efficacy and the reliability of commercially available high-throughput immunoassays. Our study demonstrates the administration of two doses of the BNT162b2 vaccine that elicited a robust SARS-CoV-2-specific immune response which was assessed up to 3 months after full vaccination in a cohort of 37 health care workers (HCWs). SARS-CoV-2-specific antibody response, evaluated by four commercially available chemiluminescence immunoassays (CLIA), was qualitatively consistent with the results provided by the gold-standard in vitro neutralization assay (NTA). However, we could not observe a correlation between the quantity of the antibody detected by CLIA assays and their neutralizing activity tested by NTA. Almost all subjects developed a SARS-CoV-2-specific T-cell response. Moreover, vaccinated HCWs developed a similar protective neutralizing antibodies response against the EU (B.1), Alpha (B.1.1.7), Gamma (P.1), and Eta (B.1.525) SARS-CoV-2 variants, while Beta (B.1.351) and Delta (B.1.617.2) strains displayed a consistent partial immune evasion. These results underline the importance of a solid vaccine-elicited immune response and a robust antibody titre. We believe that these relevant results should be taken into consideration in the definition of future vaccinal strategies.

Objective: Gastroenteritis caused by a single pathogen or multiple pathogens remains a major diagnostic challenge for the laboratory. The treatment of diarrhoea is based on microbiological results. Diagnosis is achieved using different laboratory techniques that have variable sensitivity and specificity. xTAG GPP is a new multiplex PCR assay that simultaneously detects 15 different pathogens responsible for diarrhoea. The results of the first multicentre study in Italy to evaluate the potential clinical application of the GPP assay in the laboratory diagnosis of diarrhoea are reported here. Methods: Faeces specimens (N = 664) from hospitalized patients were tested with the GPP assay using a Luminex 200 instrument. All specimens were run using comparator methods following a routine algorithm: culture for bacteria, enzyme immunoassay and PCR for viruses, and microscopy for parasites. Results: Of the samples tested with the GPP, 53.61% (356/664) gave positive results, as compared to 45.33% by routine testing. Of the positive specimens, 34.55% showed the presence of genomic DNA from multiple pathogens. The Luminex method showed an increase in the percentage of positivity of 8.28%. Conclusions: The GPP assay can be considered a helpful tool for the detection of gastrointestinal pathogens, with a hands-on time of 5 h; it provides accurate data for the clinical management of hospitalized patients and for epidemiological surveillance.

Four new species, Siphocampylus acanthidiotrichus J.R. Grande (Campanulaceae), Phyllanthus huberi Riina & P.E. Berry (Phyllanthaceae), Sohnreyia maigualidensis J.R. Grande & Kallunki (Rutaceae), and Turnera maigualidensis J.R. Grande & Arbo (Turneraceae) are described and illustrated from the summit areas of Sierra de Maigualida in the Gua - yana Highlands of southern Venezuela. To date, one genus, 32 spe cies, one subspecies, and one variety of angiosperms are known only from the Maigualida meadows and shrublands, between 1800 m and the highest elevation areas at 2400 m. Here we describe the first iteroparous species for the genus Sohnreyia K. Krause, a Turnera L. species with apically villose, apiculate petals, and a Siphocampylus Pohl species with aculeiform hairs and pinnatisect leaves. The new species, Siphocampylus acanthidiotrichus, together with S. Tillettii Steyerm., represents a morphologically distinct subgroup within the genus.

The almost unexplored Sierra de Maigualida is the largest and highest non sedimentary, granitic mountain system in the Guayana Shield. Because of its particular geological constitution, a surprisingly large number of new plant taxa has been found in its high mountain ecosystems. In this first installment, two new species, Sauvagesia cryptothallis (Ochnaceae) and Aegiphila uasadiana (Lamiaceae) are described and illustrated. Sauvagesia cryptothallis belongs to Sauvagesia sect. Imthurnianae and is closely related to S. imthurniana, differing mainly in the axis of the inflorescence being densely covered by 5-8 tightly overlapping bracts, hiding the entire axis, and with the principal and secondary leaf veins almost flat, not salient on the lower surface, and poorly differentiated from tertiary veins. Aegiphila uasadiana is a very distinctive species somewhat similar to A. luschnathii, from Brazil. It can be distinguished by its leaves that are coriaceous, conspicuously revolute, entire, shiny, gla brescent, conspicuously bullate, and foveate adaxially; the inflorescences that are relatively compact and crowned at the apex of the twigs; the calices that are adpressed-pubescent, densely so at the base, progressively glabrescent towards the apex, truncate apex, 4-apiculate, each apiculi of ca. 0.4 mm long, and stamens that are largely exserted from the corolla.

Clinical infection and death caused by bluetongue virus infection has been reported in the Eurasian lynx. Bluetongue virus surveillance in the Iberian lynx revealed widespread and repeated exposure to serotypes 1 and 4 in wild and captive populations of this species. This exposure is possibly from a spillover event from sympatric ruminants.

Currently, in dairy farming, there is growing concern for the welfare of calves during the critical period between the separation from their dams and weaning. During weaning, rationed feeding is a practice used to improve feed efficiency and control the calves' growth, but it could also have negative consequences associated with hunger and feed restriction behavior. One such consequence could be the performance by calves of stereotyped behaviors indicative of poor welfare, such as non-nutritive oral behaviors. We hypothesized that making a Total Mixed Ration (TMR) available to calves, in addition to the standard pelleted starter diet, thanks to its structural and nutritional characteristics, could help to focus the oral behavior of the subjects towards nutritional activities and therefore limit the development of stereotyped behaviors, even when the amount of starter is restricted. To test this hypothesis, 30 female Holstein calves (equipped with an accelerometer based on an ear tag), were randomly assigned to one of three treatments: a control diet (CTR) with an ad libitum calf starter but without TMR; Treatment 1 (TRT1) with both ad libitum calf starter and ad libitum TMR; Treatment 2 (TRT2) with ad libitum TMR and a restricted amount of a calf starter (50% of the intake starter of the control group day by day). All animals were kept in individual cages equipped with a slow-flow teat bucket apparatus for milk feeding and with access to separated buckets (one for drinking water, one for the starter, and one for the TMR) placed on the outside of the front gate of each cage. Sucking behavior, as well as resting, ruminating and activity behavior, was recorded individually from 7 days of life to weaning (63 d of age) by an automated monitoring system based on ear-tag accelerometers (SCR eSense, Allflex, Irving, TX, USA). The results showed that in the CTR group, there was a greater sucking activity compared to the TRT1 group (26.25 min/head/day vs. 16.83 min/head/day, p = 0.0181), while the TRT2 group showed intermediate values (20.41 min/head/day). We hypothesized that this increased sucking activity may be explained by the oral activity directed at sucking the Milk Bar tube observed only in CTR calves after complete consumption of available milk and could indicate a higher stress level than in the TRT1 group. The time spent resting was significantly lower for the CTR group compared to the TRT1 group (9.20 h/day vs. 11.15 h/day, p = 0.0049) while the TRT2 group was in an intermediate situation (10.65 h/day). Furthermore, the increase in time dedicated to rest observed in TRT1, in light of the good vitality of the calves shown by the daily patterns of resting behavior and daily activity, also seems to suggest an improvement in welfare conditions compared to calves receiving pelleted starter alone. Based on these results, we can deduce that providing TMR ad libitum in the diet of weaning calves could be an effective strategy to improve calf welfare due to the reduction in stereotyped oral activity. However, further studies are needed to establish threshold values useful for distinguishing physiological from pathological resting times in weaning calves raised in individual cages.

Babesiosis, caused by protozoan parasites of the genus Babesia , is an emerging tick-borne disease of significance for both human and animal health. Babesia parasites infect erythrocytes of vertebrate hosts where they develop and multiply rapidly to cause the pathological symptoms associated with the disease. The identification of new Babesia species underscores the ongoing risk of zoonotic pathogens capable of infecting humans, a concern amplified by anthropogenic activities and environmental changes. One such pathogen, Babesia MO1 , previously implicated in severe cases of human babesiosis in the United States, was initially considered a subspecies of B. divergens , the predominant agent of human babesiosis in Europe. Here we report comparative multiomics analyses of B. divergens and B. MO1 that offer insight into their biology and evolution. Our analysis shows that despite their highly similar genomic sequences, substantial genetic and genomic divergence occurred throughout their evolution resulting in major differences in gene functions, expression and regulation, replication rates and susceptibility to antiparasitic drugs. Furthermore, both pathogens have evolved distinct classes of multigene families, crucial for their pathogenicity and adaptation to specific mammalian hosts. Leveraging genomic information for B. MO1 , B. divergens , and other members of the Babesiidae family within Apicomplexa provides valuable insights into the evolution, diversity, and virulence of these parasites. This knowledge serves as a critical tool in preemptively addressing the emergence and rapid transmission of more virulent strains.

Research into novel anti- Helicobacter pylori agents represents an important approach for the identification of new treatments for chronic gastritis and peptic ulcers, which are associated with a high risk of developing gastric carcinoma. In this respect, two series of azobenzenesulfonamides were designed, synthesized, and tested against a large panel of human and bacterial CAs to evaluate their inhibitory activity. In addition, computational studies of the novel primary benzenesulfonamides ( 4a - j ) were performed to predict the putative binding mode to both HpCAs. Then, the antimicrobial activity versus H. pylori of the two series was also studied. The best-in-class compounds were found to be 4c and 4e among the primary azobenzenesulfonamides and 5c and 5f belonging to the secondary azobenzenesulfonamides series, showing themselves to exert a promising anti- H. pylori activity, with MIC values of 4-8 μg/mL and MBCs between 4 and 16 μg/mL. Moreover, the evaluation of their toxicity on a G. mellonella larva in vivo model indicated a safe profile for 4c , e and 5c , f . The collected results warrant considering these azobenzenesulfonamides as an interesting starting point for the development of a new class of anti- H. pylori agents.

Introduction: In Portugal, evidence of clinical outcomes within home-based hospitalization programs remains limited. Despite the adoption of homebased hospitalization services, it is still unclear whether these services represent an effective way to manage patients compared with inpatient hospital care. Therefore, the aim of this study was to evaluate the outcomes of home-based hospitalization compared with conventional hospitalization in a group of patients with a primary diagnosis of infectious, cardiovascular, oncological, or 'other' diseases., Methods: An observational retrospective study using anonymized administrative data to investigate the outcomes of home-based hospitalization (n = 209) and conventional hospitalization (n = 192) for 401 Portuguese patients admitted to CUF hospitals (Tejo, Cascais, Sintra, Descobertas, and the Unidade de Hospitalização Domiciliária CUF Lisboa). Data on demographics and clinical outcomes, including Barthel index, Braden scale, Morse scale, mortality, and length of hospital stay, were collected. The statistical analysis included comparison tests and logistic regression., Results: The study found no statistically significant differences between patients' admission and discharge for the Barthel index, Braden scale, and Morse scale scores, for both conventional and home-based hospitalizations. In addition, no statistically significant differences were found in the length of stay between conventional and home-based hospitalization, although patients diagnosed with infectious diseases had a longer stay than patients with other conditions. Although the mortality rate was higher in home-based hospitalization compared to conventional hospitalization, the mortality risk index (higher in home-based hospitalization) assessed at admission was a more important predictor of death than the type of hospitalization., Conclusion: The study found that there were no significant differences in outcomes between conventional and home-based hospitalization. Home-based hospitalization was found to be a valuable aspect of patient- and family-centered care. However, it is noteworthy that patients with infectious diseases experienced longer hospital stays.

The aim of this study was to evaluate the antimicrobial efficacy of an air gas soft jet CAP for its potential use in removing oral biofilms, given that plasma-based technologies have emerged as promising methods in periodontology. Two types of biofilms were developed, one by Streptococcus mutans UA 159 bacterial strain and the other by a complex mixture of saliva microorganisms isolated from a patient with periodontitis. This latter biofilm was characterized via Next Generation Sequencing to determine the main bacterial phyla. The CAP source was applied at a distance of 6 mm for different time points. A statistically significant reduction of both CFU count and XTT was already detected after 60 s of CAP treatment. CLSM analysis supported CAP effectiveness in killing the microorganisms inside the biofilm and in reducing the thickness of the biofilm matrix. Cytotoxicity tests demonstrated the possible use of CAP without important side effects towards human gingival fibroblasts cell line. The current study showed that CAP treatment was able to significantly reduce preformed biofilms developed by both S. mutans and microorganisms isolated by a saliva sample. Further studies should be conducted on biofilms developed by additional saliva donors to support the potential of this innovative strategy to counteract oral pathogens responsible for periodontal diseases., (© 2024. Severe musculoskeletal diseases, such as those associated with congenital or traumatic events, that result in missing limbs may compromise the fitness and survival of free-living felids. Here we report the space use of four amputee individuals from three felid species captured from 2017 to 2022 in Missouri (USA), Toledo and Badajoz (Spain), and Suitai Khairkhan Mountain (Mongolia). We describe home ranges and daily travel distances post-release of free-living felids that had either suffered a traumatic amputation or following a surgical amputation. We compared these data with those reported in the literature for felids without amputations. Forelimb or hindlimb amputation did not affect the hunting, mating, or territory patrolling behavior of any of the individuals. However, we recorded significant differences in the daily movement before and after the traumatic event of the Iberian lynx forelimb amputee. We attribute this difference to the physical impairment, although we consider other variables that may have played a role. Nevertheless, all animals appeared to cope well with their limb loss, showing home ranges and daily distances within those recorded for their sex and species. Unless amputee felids represent a threat to domestic livestock or humans, our data suggest these individuals may remain free-living as they contribute to local population persistence and appear to maintain good general health and welfare., Competing Interests: The authors declare that they do not have competing interests., (© 2024 The Authors. In recent years, there has been a considerable increasing interest in the use of the greater wax moth Galleria mellonella as an animal model. In vivo pharmacological tests, concerning the efficacy and the toxicity of novel compounds are typically performed in mammalian models. However, the use of the latter is costly, laborious and requires ethical approval. In this context, G. mellonella larvae can be considered a valid option due to their greater ease of use and the absence of ethical rules. Furthermore, it has been demonstrated that the immune system of these invertebrates has similarity with the one of mammals, thus guaranteeing the reliability of this in vivo model, mainly in the microbiological field. To better develop the full potential of this model, we present a novel approach to characterize the hemocyte population from G. mellonella larvae and to highlight the immuno modulation upon infection and treatments. Our approach is based on the detection in isolated hemocytes from G. mellonella hemolymph of cell membrane markers typically expressed by human immune cells upon inflammation and infection, for instance CD14, CD44, CD80, CD163 and CD200. This method highlights the analogies between G. mellonella larvae and humans. Furthermore, we provide an innovative tool to perform pre-clinical evaluations of the efficacy of antimicrobial compounds in vivo to further proceed with clinical trials and support drug discovery campaigns., (© 2024. Unlike most RNA and DNA viruses that broadly stimulate mRNA decay and interfere with host gene expression, human cytomegalovirus (HCMV) extensively remodels the host translatome without producing an mRNA decay enzyme. By performing a targeted loss-of-function screen in primary human fibroblasts, we here identify the host CCR4-NOT deadenylase complex members CNOT1 and CNOT3 as unexpected pro-viral host factors that selectively regulate HCMV reproduction. We find that the scaffold subunit CNOT1 is specifically required for late viral gene expression and genome-wide host responses in CCR4-NOT-disrupted cells. By profiling poly(A)-tail lengths of individual HCMV and host mRNAs using nanopore direct RNA sequencing, we reveal poly(A)-tails of viral messages to be markedly longer than those of cellular mRNAs and significantly less sensitive to CCR4-NOT disruption. Our data establish that mRNA deadenylation by host CCR4-NOT is critical for productive HCMV replication and define a new mechanism whereby herpesvirus infection subverts cellular mRNA metabolism to remodel the gene expression landscape of the infected cell. Outer membrane vesicles (OMVs) are spherical, lipid-based nano-structures, which are released by Gram-negative bacteria in both in vitro and in vivo conditions. The size and composition of OMVs depend on not only the producer bacterial species but also cells belonging to the same strain. The mechanism of vesicles' biogenesis has a key role in determining their cargo and the pattern of macromolecules exposed on their surface. Thus, the content of proteins, lipids, nucleic acids, and other biomolecules defines the properties of OMVs and their beneficial or harmful effects on human health. Many studies have provided evidence that OMVs can be involved in a plethora of biological processes, including cell-to-cell communication and bacteria-host interactions. Moreover, there is a growing body of literature supporting their role in horizontal gene transfer (HGT). During this process, OMVs can facilitate the spreading of genes involved in metabolic pathways, virulence, and antibiotic resistance, guaranteeing bacterial proliferation and survival. For this reason, a deeper understanding of this new mechanism of genetic transfer could improve the development of more efficient strategies to counteract infections sustained by Gram-negative bacteria. In line with this, the main aim of this mini-review is to summarize the latest evidence concerning the involvement of OMVs in HGT.

Background: The healthy immigrant paradox has found wide support in the literature. To evaluate this hypothesis that immigrants have better health outcomes than the native population, this study aimed to compare the premature cancer mortality between the native and immigrant populations in Spain., Methods: We obtained the 2012-15 cause-specific mortality estimates from administrative records and participant characteristics data from the 2011 Spanish census. Using Cox proportional hazards regression models, we calculated the risks of mortality of the native and immigrant populations, and the latter populations' risk based on their regions of origin, and determined the effects of covariates of interest on the calculated risk., Results: Our results show that the risk of premature cancer mortality is lower among immigrants than among natives, and this gap is higher among men than among women. There is a lower mortality rate among Latin American immigrants (Latino men are 81% less likely to die prematurely from cancer than native-born men, and Latino women are 54% less). Moreover, despite social class disparities, immigrants' advantage in cancer mortality remained constant and decreased with increasing length of residence in the host country., Conclusions: This study provided novel evidence on the 'healthy immigrant paradox', associated with the fact that migrants are favorably selected at origin, cultural patterns of the societies of origin and, in the case of men, there is some convergence or an 'unhealthy' integration that explains the fact that this advantage over natives is lost with more years of residence in Spain., (© The Author(s) 2023.
