Abstract

This communication will provide the latest information about the progress of the “Research Project for the Study of Georgian Grapes and Wine Culture”, managed by the National Wine Agency of Georgia since 2014. Local and foreign institutions continue to work together with the aim of stimulating multidisciplinary scientific research activity on Georgian viticulture and viniculture and to reconstruct their development from Neolithic civilizations to the present. The project is multidisciplinary in nature, merging contributions from archaeology, history, ethnography, molecular genetics, biomolecular archaeology, palaeobotany, ampelography, enology, climatology and other scientific fields.

Abstract

Major surgical societies advised using non-operative management of appendicitis and suggested against laparoscopy during the COVID-19 pandemic. The hypothesis is that a significant reduction in the number of emergent appendectomies was observed during the pandemic, restricted to complex cases. The study aimed to analyse emergent surgical appendectomies during pandemic on a national basis and compare it to the same period of the previous year. This is a multicentre, retrospective, observational study investigating the outcomes of patients undergoing emergent appendectomy in March–April 2019 vs March–April 2020. The primary outcome was the number of appendectomies performed, classified according to the American Association for the Surgery of Trauma (AAST) score. Secondary outcomes were the type of surgical technique employed (laparoscopic vs open) and the complication rates. One thousand five hundred forty one patients with acute appendicitis underwent surgery during the two study periods. 1337 (86.8%) patients met the inclusion criteria: 546 (40.8%) patients underwent surgery for acute appendicitis in 2020 and 791 (59.2%) in 2019. According to AAST, patients with complicated appendicitis operated in 2019 were 30.3% vs 39.9% in 2020 (p = 0.001). We observed an increase in the number of post-operative complications in 2020 (15.9%) compared to 2019 (9.6%) (p < 0.001). The following determinants increased the likelihood of complication occurrence: undergoing surgery during 2020 (+ 67%), the increase of a unit in the AAST score (+ 26%), surgery performed > 24 h after admission (+ 58%), open surgery (+ 112%) and conversion to open surgery (+ 166%). In Italian hospitals, in March and April 2020, the number of appendectomies has drastically dropped. During the first pandemic wave, patients undergoing surgery were more frequently affected by more severe appendicitis than the previous year's timeframe and experienced a higher number of complications. Abstract

Major surgical societies advised using non-operative management of appendicitis and suggested against laparoscopy during the COVID-19 pandemic. The hypothesis is that a significant reduction in the number of emergent appendectomies was observed during the pandemic, restricted to complex cases. The study aimed to analyse emergent surgical appendectomies during pandemic on a national basis and compare it to the same period of the previous year. This is a multicentre, retrospective, observational study investigating the outcomes of patients undergoing emergent appendectomy in March–April 2019 vs March–April 2020. The primary outcome was the number of appendectomies performed, classified according to the American Association for the Surgery of Trauma (AAST) score. Secondary outcomes were the type of surgical technique employed (laparoscopic vs open) and the complication rates. One thousand five hundred forty one patients with acute appendicitis underwent surgery during the two study periods. 1337 (86.8%) patients met the inclusion criteria: 546 (40.8%) patients underwent surgery for acute appendicitis in 2020 and 791 (59.2%) in 2019. According to AAST, patients with complicated appendicitis operated in 2019 were 30.3% vs 39.9% in 2020 (p = 0.001). We observed an increase in the number of post-operative complications in 2020 (15.9%) compared to 2019 (9.6%) (p 24 h after admission (+ 58%), open surgery (+ 112%) and conversion to open surgery (+ 166%). In Italian hospitals, in March and April 2020, the number of appendectomies has drastically dropped. During the first pandemic wave, patients undergoing surgery were more frequently affected by more severe appendicitis than the previous year's timeframe and experienced a higher number of complications. Abstract

Plants develop in a microbe-rich environment and must interact with a plethora of microorganisms, both pathogenic and beneficial. Indeed, such is the case of Pseudomonas, and its model organisms P. fluorescens and P. syringae, a bacterial genus that has received particular attention because of its beneficial effect on plants and its pathogenic strains. The present study aims to compare plant-beneficial and pathogenic strains belonging to the P. syringae species to get new insights into the distinction between the two types of plant-microbe interactions. In assays carried out under greenhouse conditions, P. syringae pv. syringae strain 260-02 was shown to promote plant-growth and to exert biocontrol of P. syringae pv. tomato strain DC3000, against the Botrytis cinerea fungus and the Cymbidium Ringspot Virus. This P. syringae strain also had a distinct volatile emission profile, as well as a different plant-colonization pattern, visualized by confocal microscopy and gfp labeled strains, compared to strain DC3000. Despite the different behavior, the P. syringae strain 260-02 showed great similarity to pathogenic strains at a genomic level. However, genome analyses highlighted a few differences that form the basis for the following hypotheses regarding strain 260-02. P. syringae strain 260-02: (i) possesses nonfunctional virulence genes, like the mangotoxin-producing operon Mbo; (ii) has different regulation pathways, suggested by the difference in the autoinducer system and the lack of a virulence activator gene; (iii) has genes encoding DNA methylases different from those found in other P. syringae strains, suggested by the presence of horizontal-gene-transfer-obtained methylases that could affect gene expression.

Abstract

Mgaloblishvili, a Vitis vinifera cultivar, exhibits unique resistance traits against Plasmopara viticola, the downy mildew agent. This offers the unique opportunity of exploring the molecular responses in compatible and incompatible plant-pathogen interaction. In this study, whole transcriptomes of Mgaloblishvili, Pinot noir (a V. vinifera susceptible cultivar), and Bianca (a resistant hybrid) leaves, inoculated and non-inoculated with the pathogen, were used to identify P. viticola effector-encoding genes and plant susceptibility/resistance genes. Multiple effector-encoding genes were identified in P. viticola transcriptome, with remarkable expression differences in relation to the inoculated grapevine cultivar. Intriguingly, five apoplastic effectors specifically associated with resistance in V. vinifera. Gene coexpression network analysis identified specific modules and metabolic changes occurring during infection in the three grapevine cultivars. Analysis of these data allowed, for the first time, the detection in V. vinifera of a putative P. viticola susceptibility gene, encoding a LOB domain-containing protein. Finally, the de novo assembly of Mgaloblishvili, Pinot noir, and Bianca transcriptomes and their comparison highlighted novel candidate genes that might be at the basis of the resistant phenotype. These results open the way to functional analysis studies and to new perspectives in molecular breeding of grapevine for resistance to P. viticola.

Between August 2011 and June 2013, wood samples from diseased vines showing dead spur and cordons, shoot dieback associated with sunken necrotic bark lesions and brown to black vascular streaking were collected from numerous diseased vineyards. Several fungal species were isolated from the margin between healthy and symptomatic tissue. Three species of Botryosphaeriaceae, namely, Diplodia seriata, Neofusicoccum australe, and N. vitifusiforme and one species of Diaporthaceae, namely, Diaporthe neotheicola, were observed to be associated with the decline of old vines. Other fungal species were recovered from diseased wood, namely, Alternaria alternata, Botryotinia fuckeliana (anamorph of Botrytis cinerea), Acremonium spp., Aspergillus spp., and Fusarium spp. In addition, Penicillium spp. inter- and intra-species diversity were assessed based on virtual RFLP gel analyses and identification of restriction enzymes able to distinguish fungi strains within species based on determination of single nucleotide polymorphism (SNP) lineages within cluster members based on the sequencing of the internal transcribed spacer (ITS1-5.8S-ITS2) region of the rDNA.

Entre agosto de 2011 y junio de 2013, se recolectaron muestras de madera de vid enfermas que mostraban espolones y cordones muertos, desvirones de tallos asociados con lesiones de corteza necrótica hundidas y rayas vasculares de color café a negro de numerosos viñedos enfermos. Se aislaron varias especies de hongos del margen entre tejido sano y sintomático. Tres especies de Botryosphaeriaceae, a saber, Diplodia seriata, Neofusicoccum australe, y N. vitifusiforme y una especie de Diaporthaceae, a saber, Diaporthe neotheicola, se observaron a ser asociado con la disminución de viñas viejas. Otras especies fúngicas se recuperaron de la madera enferma, a saber, Alternaria alternata, Botryotinia fuckeliana (anamorfo de Botrytis cinerea), Acremonium spp., Aspergillus spp. Y Fusarium spp. Además, Penicillium spp. inter e intra-diversidad especies fueron evaluados en base del gel RFLP virtual análisis y la identificación de las enzimas de restricción capaz de distinguir cepas de hongos dentro de las especies basado en la determinación del polimorfismo de un solo nucleótido (SNP) linajes dentro de los miembros de clúster en base a la secuenciación del espaciador transcrito interno (ITS1-5.8S-ITS2) región del rDNA.

Abstract

Apple proliferation (AP), caused by 'Candidatus Phytoplasma mali', is one of the most important phytoplasma diseases in Europe. Because of its economic importance, efforts are focused on the study of 'Ca. Phytoplasma mali' characterization, molecular mechanisms of disease induction, transmission by insect vectors, and plant defense responses. However, very few studies have investigated the effect of 'Ca. Phytoplasma mali' on microbial diversity. In this work, we describe for the first time the endophytic bacterial community associated with healthy and AP phytoplasma-infected apple roots, suggesting the hypothesis that 'Ca. Phytoplasma mali' influences microbial diversity in apple plants. En- dophytic bacterial diversity associated with healthy and 'Candidatus Phytoplasma mali'-infected apple (Malus domestica, Borkh.) trees was analyzed by cultivation-dependent and -independent methods. Sequence analysis of 16S rRNA gene showed the presence of Proteobacteria, Acidobacteria, Bacteroidetes, Actinobacteria, Chlamydiae, and Firmicutes. Bacterial group distribution differed in healthy and diseased plants. Library analyses underscored 24 and 17 operational taxonomic units (OTUs) in healthy and infected roots, respectively, with a dominance of the Betaproteobacteria group. Pseudomonas fluorescens was prevalent in healthy roots, while Burkholderia sp. in the infected ones. The combination of culturing methods and cloning analysis allowed an accurate description of the endophytic community in apple trees, revealing that bacterial richness in healthy plants is higher than that observed in those infected by 'Ca. Phytoplasma mali'. Evidences from the present study reinforce the idea that uncultivable bacterial pathogens can modify the composition of endophytic bacterial communities associated with infected plants. Abstract

Phytoplasmas are cell-wall less, uncultivable bacteria, belonging to the class Mollicutes. They parasitize both plant hosts and insect vectors, but knowledge of their biological and molecular properties is limited. Analyses of 16S rDNA nucleotide sequences revealed that 16SrV group phytoplasma strains identified in grapevine plants are closely related, but associated with distinct ecologies. In the present study, we investigated the evolutionary relationships of eight representative 16SrV phytoplasma strains (EY1, ULW, ALY, HD1, PGY-A, FD-C, FD92 and FD70) by sequencing 15 ribosomal protein (rp) genes positioned within the S10-spc operon. Phylogenetic analyses of concatenated nucleotide and amino acid sequences underscored that 16SrV phytoplasma strains infecting grapevine cluster together in a distinct clade, supporting the hypothesis that they have a probable common origin. Furthermore, sequence typing revealed the presence of single nucleotide polymorphisms (SNPs) distinguishing the analyzed 16SrV phytoplasma strains. In detail, in silico restriction digest assays on rplV-rplO nucleotide sequences showed that the DNA segment including the genes rplR, rpsE, rpmD, and rplO contained numerous SNPs positioned within restriction sites for AluI and HhaI endonucleases, revealing the presence of distinct strain-specific virtual RFLP patterns. Based on SNPs identified in this study, candidate rp-genes could be proposed for finer analyses. For example, in the case of Flavescence dorée (FD) disease, SNPs located within the gene rpsE could be used for distinguishing FD phytoplasma strains. In-depth investigation will be performed for confirming and describing more accurately the complex population structure of FD phytoplasmas of 16SrV-C and -D subgroups. [ABSTRACT FROM AUTHOR]

Abstract

Grapevine “bois noir”, related to the presence of ‘Candidatus Phytoplasma solani’ (‘Ca. P. solani’), represents a serious threat in several vine-growing areas worldwide. In surveys conducted over two years, mild and/or moderate symptoms and lower pathogen titer were mainly associated with ‘Ca. P. solani’ strains harboring a secY gene sequence variant (secY52), whereas severe symptoms and higher titer were mainly observed in grapevines infected by phytoplasma strains carrying any one of another four variants. A comparison of amino acid sequences of the protein SecY of ‘Ca. P. solani’ strains revealed the presence of conservative and semi-conservative substitutions. The deduced three-dimensional (3D) protein analysis unveiled that one semi-conservative substitution identified in the sequence variant secY52 is responsible for a structural disordered region that probably confers a flexibility for binding to distinct molecular complexes. In fact, the other analyzed variants show an organized structure and the 3D in silico prediction allowed the identification of β-sheets. Thus, differences in symptom severity and pathogen concentration observed in grapevines infected by ‘Ca. P. solani’ strains carrying distinct secY gene sequence variants suggest a possible relationship between SecY protein structure and phytoplasma strain virulence.

Abstract

Bois noir (BN), associated with &lsquo, Candidatus Phytoplasma solani&rsquo, (CaPsol), is the most widespread disease of the grapevine yellows complex worldwide. In this work, BN epidemiology was investigated in a case study vineyard where an unusual CaPsol strain, previously detected only in other host plants, was found to be prevalent in grapevine. Experimental activities included: symptom observation, sampling of symptomatic vines, Auchenorrhyncha specimens, and weeds, molecular detection and typing of CaPsol strains, statistical analyses for determining possible relationships between CaPsol relative concentration, strain type, and symptom severity. Among insects, Reptalus quinquecostatus was the most abundant and was found to be highly infected by CaPsol, while Hyalesthes obsoletus, the main CaPsol vector, was not caught. Moreover, R. quinquecostatus harbored CaPsol strains carrying uniquely the stamp sequence variant St10, also identified as prevalent in vines and in the majority of weeds, and all the secY variants identified in the vineyard. Statistical analyses revealed that CaPsol strains carrying the St10 variant are not associated with severe symptoms, suggesting their possible moderate virulence. Based on such evidence, a new BN epidemiological pattern related to these CaPsol strains and involving grapevine, R. quinquecostatus, and/or weeds is proposed. Furthermore, the possible presence of other players (vectors and weeds) involved in CaPsol transmission to grapevines was highlighted.
