Tetranychus urticae Koch is a worldwide agricultural pest mite that feeds on more than 1100 kinds of crops. The mite has developed a high level of tolerance to high temperatures, but the physiological mechanism underlying the outstanding adaptability of this pest to high temperatures remains unclear. To clarify the physiological mechanisms of T. urticae in response to short-term heat stress, four temperatures (36, 39, 42, and 45 °C) and three short-term heat durations (2, 4, and 6 h) were conducted to test the effects on protein content, the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), and the total antioxidant capacity (T-AOC). The results showed that protein content, antioxidant enzyme activity, and T-AOC in T. urticae were significantly induced by heat stress. These results suggest that heat stress induces oxidative stress and that antioxidant enzymes play an important role in reducing oxidative damage in T. urticae. The data of this study will provide a basis for further research on the molecular mechanisms of thermostability and ecological adaptability of T. urticae.

Plant fiber/plastic composites (PPCs) have been widely used in food contact materials (FCMs) for many benefits, such as their claimed better environmental footprint compared to conventional plastics. However, their safety is still not fully understood and must be comprehensively evaluated. Non-volatiles extracted from six PPCs with different plant fibers and polymer matrices were characterized by employing ultra-high-performance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry in combination with various spectral libraries and manual elucidation, taking into account spectral similarity and characteristic product ions. A total of 115 compounds were tentatively identified, 50 of which were oligomers or their derivatives from the sample with polylactic acid (PLA) and polybutylene adipate terephthalate (PBAT) as the polymer matrix, and some of them were Cramer rules class III substances based on the threshold of toxicological concern (TTC). Seven reaction products between PLA and PBAT monomers, as well as four derivatives of melamine, were elucidated and well detailed for the first time. In addition, bisphenol S was detected in all samples even though its origin remains to be further explored. Isoprothiolane, as an insecticide and fungicide used to control a range of rice pests, was identified in the sample with rice husk as fillers, experimentally confirming the presence of agrochemicals in samples containing plant fibers.

In order to clarify the physiological mechanisms of T. urticae in response to short-term heat stress, four high temperatures (36, 39, 42, and 45°C), three short-term heat durations (2, 4, and 6 h) were conducted to test the effects on the protein content of T. urticae and the activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and total antioxidant capacity (T-AOC), respectively. The results showed that protein content, antioxidant enzymes activity, and T-AOC in T. urticae were significantly affected by both the high temperature and duration after treatment, meanwhile, they showed same trends of increased at 25–39℃, then decreased at 39–42℃ except for CAT activity. The highest values of SOD activity, POD activity and T-AOC were all obtained at 39℃-4h, and that of CAT activity was obtained at 39℃-2h. The higher levels of T-AOC serve an important signal demonstrating that T. urticae has other antioxidant mechanisms except for antioxidant enzymes. Those results suggested that heat stress induces oxidative stress, and antioxidant enzymes played important role in reducing oxidative damage in T. urticae. which provided a basis for further research on the molecular mechanisms of thermostability and ecological adaptability of T. urticae.

Background The lack of effective prognostic models for colorectal cancer (CRC) makes it crucial to establish a reliable prognostic model for colorectal cancer. The aim of this study was to develop a reliable prognostic model in CRC. Using CRC samples from the TCGA database, a multi-gene combination model was screened for prognosis in CRC patients and the prognostic power of the multi-gene combination model was validated. We screened a multi-gene combination model for risk stratification and prognosis of CRC patients, which is important for individualised clinical treatment and long-term management of colorectal cancer patients. Results To solve these questions, we used data from colorectal cancer patients in the TCGA database to uncover differential genes, screened variables using one-way COX analysis and LASSO regression analysis, and then screened for the polygenic combination most associated with colorectal cancer and assessed its significance on survival events using multi-factor COX analysis and ROC curves to assess polygenic combinations. Finally, using gene enrichment analysis, the polygenic combinations were found to be associated with tRNA biosynthesis, homologous recombination, nucleotide excision repair, ribosome biogenesis in eukaryotes and DNA replication. Conclusions The multi-gene combination (FCGBP, GSTM1, IGLV4-69, IGSF9 and PCOLCE2) model screened in this study is capable of prognosticating colorectal cancer, and the findings of this study could shed new light on the prognosis of colorectal cancer in the clinical setting.

To study the physiological mechanisms of Neoseiulus barkeri in response to short-term heat stress, the eggs and the emerged adults were exposed to 38, 40, and 42°C, 85% ± 5%RH,16 h:8 h (L:D) for 2, 4, and 6 h. The activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) as well as the protein content of N. barkeri were examined. All treatments caused significant different changes compared to the untreated control. The protein content increased as the temperature increased, while it showed different changing trends with the prolongation of exposure duration. The enzymatic activity of SOD, CAT, and POD was significantly affected by the temperature treatment. Both the maximum and minimum level of the three enzymes after a short-term heat stress differed significantly to the control group (p < 0.05). The highest values of three enzymatic activities were all obtained at 40°C-4 h. Person correlation analysis indicates that the high temperature was the primary factor affecting the enzymatic activity, while the exposure duration of the heat stress was the secondary factor. In general, the short-term heat stress increased the protein content of Neoseiulus barkeri and up-regulated the expression of SOD, CAT, and POD activities as well.

The spider mite Tetranychus urticae is one of the most economically key mite pests of fields and greenhouse crops over the world. In order to clarify the control efficacy of Neoseiulus barkeri on T. urticae in the presence of entomopathogenic fungus Acremonium hansfodii. This study was conducted to evaluate the control of combining N. barkeri with A. hansfodii against T. urticae. The results showed that the functional response of N. barkeri treated with Ahy1 strain to T. urticae was Holling type II equation. Among these, the highest predation number and the shortest attack time were obtained for the predator feeding on eggs, then followed by feeding on nymphs, female adults ranked as the lowest. That of N. barkeri to T. urticae treated with Ahy1 strain was also Holling type II equation. The predation capacity of N. barkeri on nymphs and female adults of T. urticae treated by Ahy1 strain increased with the increase of the concentration of Ahy1 strain, but that when feeding eggs had no significant difference. Therefore, the combination of EPF with N. barkeri on control of TSSM had synergistic effects, which will provide the guidance to carry out combined utilization of entomogenous fungi with predatory mites.

Chemotherapy resistance is the dominant challenge in the treatment of acute myeloid leukemia (AML). Nuclear factor E2-related factor 2 (Nrf2) exerts a vital function in drug resistance of many tumors. Nevertheless, the potential molecular mechanism of Nrf2 regulating the base excision repair pathway that mediates AML chemotherapy resistance remains unclear. Here, in clinical samples, we found that the high expression of Nrf2 and base excision repair pathway gene encoding 8-hydroxyguanine DNA glycosidase (OGG1) was associated with AML disease progression. In vitro, Nrf2 and OGG1 were highly expressed in drug-resistant leukemia cells. Upregulation of Nrf2 in leukemia cells by lentivirus transfection could decrease the sensitivity of leukemia cells to cytarabine, whereas downregulation of Nrf2 in drug-resistant cells could enhance leukemia cell chemosensitivity. Meanwhile, we found that Nrf2 could positively regulate OGG1 expression in leukemia cells. Our chromatin immunoprecipitation assay revealed that Nrf2 could bind to the promoter of OGG1. Furthermore, the use of OGG1 inhibitor TH5487 could partially reverse the inhibitory effect of upregulated Nrf2 on leukemia cell apoptosis. In vivo, downregulation of Nrf2 could increase the sensitivity of leukemia cell to cytarabine and decrease OGG1 expression. Mechanistically, Nrf2-OGG1 axis-mediated AML resistance might be achieved by activating the AKT signaling pathway to regulate downstream apoptotic proteins. Thus, this study reveals a novel mechanism of Nrf2-promoting drug resistance in leukemia, which may provide a potential therapeutic target for the treatment of drug-resistant/refractory leukemia.

Flies of the Colocasiomyia toshiokai species group depend exclusively on inflorescences/infructescences of the aroid tribe Homalomeneae. The taxonomy and reproductive biology of this group is reviewed on the basis of data and samples collected from Southeast Asia. The species boundaries are determined by combining morphological analyses and molecular species delimitation based on sequences of the mitochondrial COI (cytochrome c oxidase subunit I) gene. For the phylogenetic classification within this species group, a cladistic analysis of all the member species is conducted based on 29 parsimony-informative, morphological characters. As a result, six species are recognised within the toshiokai group, including one new species, viz. C. toshiokai, C. xanthogaster, C. nigricauda, C. erythrocephala, C. heterodonta and C. rostrata sp. n. Various host plants are utilised by these species in different combinations at different localities: Some host plants are monopolized by a single species, while others are shared by two or three species. C. xanthogaster and C. heterodonta cohabit on the same host plant in West Java, breeding on spatially different parts of the spadix. There is a close synchrony between flower-visiting behaviour of flies and flowering events of host plants, which indicate an intimate pollination mutualism.

Background/Aims: Parkinson’s disease (PD) is a prevalent disease that leads to motor and cognitive disabilities, and oxidative stress (OS) injury was found to be related to the etiology of PD. Increasing evidence has shown that SHC3 is aberrantly expressed in neurons. The current study examines the involvement of SHC3 silencing in OS injury in the nigral dopamine neurons in rats with PD via the PI3K-AKT-FoxO signaling pathway. Methods: To study the mechanisms and functions of SHC3 silencing in PD at the tissue level, 170 rats were selected, and a lentivirus-based packaging system was designed to silence SHC3 expression in rats. Furthermore, PC12 cells were selected for in vitro experimentation. To evaluate the effect of SHC3 silencing in nigral dopamine neuronal growth, an MTT assay, propidium iodide (PI) single staining and Annexin V-PI double staining were performed to detect cell viability, cell cycle progression and cell apoptosis, respectively. Results: SHC3 shRNA led to decreased SOD and MDA levels and enhanced GSH activity, indicating that SHC3 silencing leads to motor retardation. SHC3 silencing repressed the extent of Akt and FoxO phosphorylation, thereby inhibiting the PI3K-AKT-FoxO signaling pathway. Furthermore, in cell experiments, SHC3 silencing suppressed PC12 cell proliferation and cell cycle progression, whereas it enhanced cell apoptosis. Conclusion: The current study provides evidence suggesting that SHC3 silencing may aggravate OS injury in nigral dopamine neurons via downregulation of the PI3K-AKT-FoxO signaling pathway in PD rats.

Physical examination including systolic blood pressure (SBP) and heart rate (HR) was performed before, 5 min after, and 20 min after intra-rectal use. Toxicities were graded using the Radiation Therapy Oncology Group standard. A two-sided Fisher’s exact test was used to compare proportions between groups. A mixed-effects model was used to analyze multiple measurements of SBP and HR. Survival curves were calculated using the Kaplan–Meier method and compared between groups using the log-rank test.Results: All patients completed the protocol treatment and reported no cardiovascular symptoms after intra-rectal administration. There were no differences in SBP and HR between these two groups at any time point (before, 5 min after, and 20 min after epinephrine). At 5 weeks after the start of radiotherapy, the incidence of rectal toxicity≥grade 2 was 27.8% (5/18) for the control group versus 12.5% (2/16) for the epinephrine group, but was not statistically significant (p=0.4). There was no rectal toxicity≥grade 2 in these two groups beyond 2-year follow-up. The 5-year biochemical relapse-free survival was 75.0% and 72.2% for the epinephrine and control group, respectively.Conclusion: Results of this pilot randomized trial have demonstrated that intra-rectal administration of epinephrine is feasible and safe in prostatic radiotherapy. A newly developed wall function for the velocity-profile of steady turbulent flow in various hydraulic regimes near rough walls was created. The wall function was then applied into two approaches to obtain the total flow energy loss (mechanical energy loss) of turbulent pipe flow. The energy loss values calculated through the above two approaches both predicted the experimental results well. The energy loss coefficient calculated through the second approach gave a more precise definition of the total flow characteristics than the Manning roughness coefficient as it was proved to be a function of the field statistics. It was furtherly found that this energy loss can be decomposed into the energy loss due to dissipation caused by molecular viscosity and the dissipation of turbulent kinetic energy, and that its density distribution falls mainly into the near-wall region.

Purpose Retinoblastoma is a malignant tumor of the developing retina that mostly occurs in children. Our study aimed to investigate the effect of tripartite motif-containing protein 59 (TRIM59) on retinoblastoma growth and the underlying mechanisms. Methods We performed bioinformatic analysis of three datasets (GSE24673, GSE97508, and GSE110811) from the Gene Expression Omnibus database. Quantitative reverse-transcription PCR and immunoblotting of three retinoblastoma cell lines were conducted to verify TRIM59 as a differentially expressed gene. Specific siRNAs were used to inhibit TRIM59 expression in the HXO-Rb44 cell line. A lentiviral vector was transfected into the Y79 cell line to overexpress TRIM59. The effects of TRIM59 on retinoblastoma cell proliferation, cell cycling, and apoptosis were explored in vitro using the abovementioned cell lines. The effect of TRIM59 expression on retinoblastoma cell proliferation was evaluated in a mouse xenograft tumor model. Results TRIM59 expression in three retinoblastoma cell lines was remarkably elevated compared with normal control. Knocking down TRIM59 expression remarkably suppressed cell proliferation and growth and promoted cell apoptosis in HXO-Rb44 cells, whereas TRIM59 overexpression promoted tumor progression in Y79 cells. Silencing TRIM59 also markedly inhibited in vivo tumor growth in the xenograft model. Mechanistic studies revealed that TRIM59 upregulated phosphorylated p38, p-JNK1/2, p-ERK1/2, and p-c-JUN expression in retinoblastoma cells. Notably, the p38 inhibitor SB203580 attenuated the effects of TRIM59 on cell proliferation, apoptosis, and the G1/S phase transition. Conclusions TRIM59 plays an oncogenic role in retinoblastoma and exerts its tumor-promotive function by activating the p38-mitogen-activated protein kinase pathway.

In the present experiment, effects of sodium hydrosulfide (NaHS), a H2S donor, on the oxidative damage, antioxidant capacity and the growth of cucumber hypocotyls and radicles were studied under 100 mmol L−1 NaCl stress. NaCl treatment significantly induced accumulation of H2O2 and thiobarbituric acid-reactive substances (TBARS) in cucumber hypocotyls and radicles, and application of NaHS dramatically reduced the accumulation of H2O2 and lipid peroxidation. However, the alleviating effects greatly depended on the concentrations of NaHS, and 400 μmol L−1 NaHS treatment showed the most significant effects. Corresponding to the change of lipid peroxidation, higher activities of antioxidant enzymes as well as the antioxidant capacity indicated as DPPH scavenging activity, chelating activity of ferrous ions and hydroxyl radical (·OH) scavenging activity were induced by NaHS treatment under NaCl stress, especially by 400 μmol L−1 NaHS treatment. With the alleviating lipid peroxidation, the amylase activities in cotyledons were increased, and the length of cucumber hypocotyls and radicles were significantly promoted by NaHS treatment under NaCl stress. Unlike the effects of NaHS, pretreatment with other sodium salts including Na2S, Na2SO4, NaHSO4, Na2SO3, NaHSO3 and NaAc did not show significant effects on the growth of cucumber hypocotyls and radicles. These salts do not release H2S. Based on above results, it can be concluded that the effects of NaHS in the experiment depended on the H2S rather than other compounds derived from NaHS, and the alleviating effects might related with its function in modulating antioxidant capacity and amylase activities.

Elm (Ulmus) has a long history of use as a high-quality heavy hardwood famous for its resistance to drought, cold, and salt. It grows in temperate, warm temperate, and subtropical regions. This is the first report of Ulmaceae chloroplast genomes by de novo sequencing. The Ulmus chloroplast genomes exhibited a typical quadripartite structure with two single-copy regions (long single copy [LSC] and short single copy [SSC] sections) separated by a pair of inverted repeats (IRs). The lengths of the chloroplast genomes from five Ulmus ranged from 158,953 to 159,453 bp, with the largest observed in Ulmus davidiana and the smallest in Ulmus laciniata. The genomes contained 137-145 protein-coding genes, of which Ulmus davidiana var. japonica and U. davidiana had the most and U. pumila had the fewest. The five Ulmus species exhibited different evolutionary routes, as some genes had been lost. In total, 18 genes contained introns, 13 of which (trnL-TAA+, trnL-TAA-, rpoC1-, rpl2-, ndhA-, ycf1, rps12-, rps12+, trnA-TGC+, trnA-TGC-, trnV-TAC-, trnI-GAT+, and trnI-GAT) were shared among all five species. The intron of ycf1 was the longest (5,675bp) while that of trnF-AAA was the smallest (53bp). All Ulmus species except U. davidiana exhibited the same degree of amplification in the IR region. To determine the phylogenetic positions of the Ulmus species, we performed phylogenetic analyses using common protein-coding genes in chloroplast sequences of 42 other species published in NCBI. The cluster results showed the closest plants to Ulmaceae were Moraceae and Cannabaceae, followed by Rosaceae. Ulmaceae and Moraceae both belonged to Urticales, and the chloroplast genome clustering results were consistent with their traditional taxonomy. The results strongly supported the position of Ulmaceae as a member of the order Urticales. In addition, we found a potential error in the traditional taxonomies of U. davidiana and U. davidiana var. japonica, which should be confirmed with a further analysis of their nuclear genomes. This study is the first report on Ulmus chloroplast genomes, which has significance for understanding photosynthesis, evolution, and chloroplast transgenic engineering.

The thyroid transcription factor 1 (TTF-1) gene is associated with the differentiation of lung epithelial cells and has been reported to be an independent prognostic factor for lung adenocarcinoma patients. The aim of the present study was to detect the expression of TTF-1 in human lung cancer cell lines and to evaluate the association of overexpressed TTF-1 with Ki-67 and apoptosis in the A549 cell line. We also investigated the expression of TTF-1 and Ki-67 in Xuanwei lung adenocarcinoma. TTF-1 mRNA expression was evaluated in 10 non-small cell lung cancer (NSCLC) cell lines by quantitative real-time RT-PCR (qRT-PCR). Overexpression of TTF-1 in A549 cells was achieved by transient transfection. The TTF-1 and Ki-67 proteins were detected by immunohistochemistry and apoptosis was detected by flow cytometry. We also investigated immunohistochemically the expression of TTF-1 and Ki-67 in 62 resected cases of Xuanwei lung adenocarcinoma. Overall the expression of TTF-1 mRNA in the 10 cell lines was low. Overexpression of TTF-1 mRNA was found only in 3 (30%) of 10 NSCLC cell lines, including 1 (25%) of 4 adenocarcinoma cell lines. A549 cells overexpressing TTF-1 were found to have repressed expression of Ki-67 (P=0.012) and increased apoptosis (P=0.000). Immunohistochemical analysis of resected cases of Xuanwei lung adenocarcinoma (n=62) showed the expression of TTF-1 in 58 (93%) of 62 and Ki-67 in 22 (35%) of 62. Patients with strong immunohistochemical expression TTF-1 were statistically associated with well-differentiated phenotype (P=0.006) and inverse correlation with Ki-67 expression (P=0.016). These data suggest that TTF-1 may serve as a tumor suppressor gene based on its inverse correlation with Ki-67 proliferative activity and increase of cellular apoptosis.

To increase oil accumulation in the high-biomass vegetative organs of tobacco ( Nicotiana tabacum L.) plants for renewable production of biofuels, VgDGAT1a isolated from developing seeds of Vernonia galamensis L. was ectopically overexpressed in tobacco leaves using a constitutive promoter. The transgenic tobacco leaves showed a 3.5–5.0-fold increase in oil content compared to the control, with a maximum increase of 9.2% (DW). The transgenic leaves also showed a substantial change in fatty acid composition, with significant enhancement of linoleic acid (18 : 2) and notable reduction of -linolenic acid (18 : 3). The overexpression of VgDGAT1a exhibited no deleterious effect on other phenotypes in the tobacco plant. These results will facilitate development of a novel system for lipid metabolic engineering in vegetative organs of plants, as well as provide a platform for the production of biofuels using the vegetative organs of commercial non-food crops.

AIM: To compare the gene expression between La (NO3) 3-exposed and control rats in vivo. METHODS: Rats were fed La (NO3) 3 once daily at a dose of 20 mg/kg for one month by gavage. Gene expression of hepatocytes was detected using mRNA differential display (DD) technique and cDNA microarray and compared between treated and control groups. RESULTS: Six differentially expressed sequence tags were cloned by DD, of which five were up regulated and one was down regulated in treated rats. Two sequences were determined. One band was novel. The other shared 100% sequence homology with AU080263 Sugano mouse brain mncb Mus musculus cDNA clone MNCb-5435 5’. With DNA microarray, 136 differentially expressed genes were identified including 131 over-expressed genes and 5 under-expressed genes. Most of these differentially expressed genes were cell signal and transmission genes, genes associated with metabolism, protein translation and synthesis. CONCLUSION: La (NO3) 3 could change the expression levels of some kinds of genes. Further analysis of the differentially expressed genes would be helpful for understanding the wide biological effect spectrum of rare earth elements.

The title compound, C37H36N4O2, has been obtained by the reaction of 2-[3,6-bis­(diethyl­amino)-9H-xanthen-9-yl]benzoyl chloride with 8-amino­quinoline. The mol­ecular packing in the crystal structure is stabilized by weak C—H⋯O hydrogen bonding.
