The study of hydrothermal travertines contributes to the understanding of the interaction between physico-chemical processes and the role played by microbial mats and biofilms in influencing carbonate precipitation. Three active travertine sites were investigated in Central Italy to identify the types of carbonate precipitates and the associated microbial mats at varying physico-chemical parameters. Carbonate precipitated fabrics at the decimetre- to millimetre-scale and microbial mats vary with decreasing water temperature: a) at high temperature (55-44°C) calcite or aragonite crystals precipitate on microbial mats of sulphide oxidizing, sulphate reducing and anoxygenic phototrophic bacteria forming filamentous streamer fabrics, b) at intermediate temperature (44-40°C), rafts, coated gas bubbles and dendrites are associated with Spirulina cyanobacteria and other filamentous and rod-shaped cyanobacteria, c) low temperature (34-33°C) laminated crusts and oncoids in a terraced slope system are associated with diverse Oscillatoriales and Nostocales filamentous cyanobacteria, sparse Spirulina and diatoms. At the microscale, carbonate precipitates are similar in the three sites consisting of prismatic calcite (40-100 µm long, 20-40 µm wide) or acicular aragonite crystals organized in radial spherulites, overlying or embedded within biofilm EPS (Extracellular Polymeric Substances). Microsparite and sparite crystal size decreases with decreasing temperature and clotted peloidal micrite dominates at temperatures < 40°C, also encrusting filamentous microbes. Carbonates are associated with gypsum and Ca-phosphate crystals; EPS elemental composition is enriched in Si, Al, Mg, Ca, P, S and authigenic aluminium-silicates form aggregates on EPS. This study confirms that microbial communities in hydrothermal travertine settings vary as a function of temperature. Carbonate precipitate types at the microscale do not vary considerably, despite different microbial communities suggesting that travertine precipitation, driven by CO2 degassing, is influenced by biofilm EPS acting as template for crystal nucleation (EPS-mediated mineralization) and affecting the fabric types, independently from specific microbial metabolism.

Holocene fringing reef development around Bora Bora is controlled by variations in accommodation space (as a function of sea-level and antecedent topography) and exposure to waves and currents. Subsidence ranged from 0 to 0·11 m kyr−1, and did not create significant accommodation space. A windward fringing reef started to grow 8·7 kyr bp, retrograded towards the coast over a Pleistocene fringing reef until ca 6·0 kyr bp, and then prograded towards the lagoon after sea-level had reached its present level. The retrograding portion of the reef is dominated by corals, calcareous algae and microbialite frameworks; the prograding portion is largely detrital. The reef is up to 13·5 m thick and accreted vertically with an average rate of 3·12 m kyr−1. Lateral growth amounts to 13·3 m kyr−1. Reef corals are dominated by an inner Pocillopora assemblage and an outer Acropora assemblage. Both assemblages comprise thick crusts of coralline algae. Palaeobathymetry suggests deposition in 0 to 10 m depth. An underlying Pleistocene fringing reef formed during the sea-level highstand of Marine Isotope Stage 5e, and is also characterized by the occurrence of corals, coralline algal crusts and microbialites. A previously investigated, leeward fringing reef started to form contemporaneously (8·78 kyr bp), but is thicker (up to 20 m) and solely prograded throughout the Holocene. A shallow Pocillopora assemblage and a deeper water Montipora assemblage were identified, but detrital facies dominate. At the Holocene reef base, only basalt was recovered. The Holocene windward–leeward differences are a consequence of less accommodation space on the eastern island side that eventually led to a more complex reef architecture. As a result of higher rates of exposure and flushing, the reef framework on the windward island side is more abundant and experienced stronger cementation. In the Pleistocene, the environmental conditions on the leeward island side were presumably unfavourable for fringing reef growth.

The earliest diagenetic post mortem exposure of biogenic carbonates at the sea floor and in the uppermost sediment column results in the colonization of hard-part surfaces by bacterial communities. Some of the metabolic redox processes related to these communities have the potential to alter carbonate shell properties, and hence affect earliest diagenetic pathways with significant consequences for archive data. During a three-month in vitro study, shell subsamples of the ocean quahog Arctica islandica (Linnaeus, 1767) were incubated in natural anoxic sediment slurries and bacterial culture medium of the heterotrophic Shewanella sediminis HAW-EB3. Bulk analyses of the liquid media from the Shewanella sediminis incubation revealed an over ten-fold increase in total alkalinity, dissolved inorganic carbon and ΩAragonite, and the alteration of the Mg/Ca, Mg/Sr and Sr/Ca ratios relative to control incubations without cultures. Ion ratios were most affected in the incubation with anoxic sediment, depicting a 25% decrease in Mg/Ca relative to the control. Shell-sample surfaces that were exposed to both incubations displayed visible surface dissolution features, and an 8 wt% loss in calcium content. No such alteration features were detected in control shells. Apparently, alteration of shell-carbonate properties was induced by microbially-driven decomposition of shell intercrystalline organic constituents and subsequent opening of pathways for pore-fluid – crystal exchange. This study illustrates the potential influence of benthic bacterial metabolism on biogenic carbonate archives during the initial stages of diagenetic alteration within a relatively short experimental duration of only three months. These results suggest that foremost the biological effect of bacterial cation adsorption on divalent-cation ratios has the potential to complicate proxy interpretation. Results shown here highlight the necessity to consider bacterial metabolic activities in marine sediments for the interpretation of palaeo-environmental proxies from shell carbonate archives. Deregulated dynamics of the extracellular matrix (ECM) are one of the hallmarks of cancer. Studies on tumor mechanobiology are thus expected to provide an insight into the disease pathogenesis as well as potentially useful biomarkers. Type I collagen is among the major determinants of breast ECM structural and tensile properties, and collagen modifications during tumor evolution drive a number of disease-related processes favoring cancer progression and invasion. We investigated the use of 3D collagen-based scaffolds to identify the modifications induced by cancer cells on the mechanical and structural properties of the matrix, comparing cell lines from two breast tumor subtypes with different clinical aggressiveness. Orthotopic implantation was used to investigate the collagen content and architecture of in vivo breast tumors generated by the two cell lines. MDA-MB-231, which belongs to the aggressive basal-like subtype, increased scaffold stiffness and overexpressed the matrix-modifying enzyme, lysyl oxidase (LOX), whereas luminal A MCF-7 cells did not significantly alter the mechanical characteristics of extracellular collagen. This replicates the behavior of in vivo tumors generated by MDA-MB-231, characterized by a higher collagen content and higher LOX levels than MCF-7. When LOX activity was blocked, the ability of MDA-MB-231 to alter scaffold stiffness was impaired. Our model could constitute a relevant in vitro tool to reproduce and investigate the biomechanical interplay subsisting between cancer cells and the surrounding ECM and its impact on tumor phenotype and behavior.
