To reach their target site, systemic pesticides must enter the plant from a spray droplet applied in the field. The uptake of an active ingredient (AI) takes place via the barrier-forming cuticular membrane, which is the outermost layer of the plant, separating it from the surrounding environment. Formulations are usually used which, in addition to the AI, also contain stabilizers and adjuvants. Adjuvants can either have surface-active properties or they act directly as barrier-modifying agents. The latter are grouped in the class of accelerating adjuvants, whereby individual variants may also have surface-active properties. The uptake of a pesticide from a spray droplet depends essentially on its permeability through the cuticular barrier. Permeability defines a combined parameter, which is the product of AI mobility and AI solubility within the cuticle. In recent decades, several tools have been developed that allowed the determination of individual parameters of organic compound penetration across the cuticular membrane. Nevertheless, earlier studies showed that mainly cuticular waxes are the barrier-determining component of the cuticular membrane and additionally, it was shown that mainly the very-long-chain aliphatic compounds (VLCAs) are responsible for establishing an effective barrier. However, the barrier-determining role of the individual VLCAs, being classified according to their respective functional groups, is still unknown. Therefore, the following objectives were pursued and achieved in this work: (1) A new ATR-FTIR-based approach was developed to measure the temperature-dependent real-time diffusion kinetics of organic models for active ingredients (AIs) in paraffin wax, exclusively consisting of very-long chain alkanes. (2) The developed ATR-FTIR approach was applied to determine the diffusion kinetics of self-accelerating adjuvants in cuticular model waxes of different VLCA composition. At the same time, wax-specific changes were recorded in the respective IR spectra, which provided information about the respective wax modification. (3) The ATR-FTIR method was used to characterize the diffusion kinetics, as well as to determine the wax-specific sorption capacities for an AI-modeling organic compound and water in cuticular model waxes after adjuvant treatment. Regarding the individual chemical compositions and structures, conclusions were drawn about the adjuvant-specific modes of action (MoA). In the first chapter, the ATR-FTIR based approach to determine organic compound diffusion kinetics in paraffin wax was successfully established. The diffusion kinetics of the AI modelling organic compounds heptyl parabene (HPB) and 4-cyanophenol (CNP) were recorded, comprising different lipophilicities and molecular volumes typical for AIs used in pesticide formulations. Derived diffusion coefficients ranged within 10-15 m2 s-1, thus being thoroughly higher than those obtained from previous experiments using an approach solely investigating desorption kinetics in reconstituted cuticular waxes. An ln-linear dependence between the diffusion coefficients and the applied diffusion temperature was demonstrated for the first time in cuticular model wax, from which activation energies were derived. The determined activation energies were 66.2 ± 7.4 kJ mol-1 and 56.4 ± 9.8 kJ mol-1, being in the expected range of already well-founded activation energies required for organic compound diffusion across cuticular membranes, which again confirmed the significant contribution of waxes to the cuticular barrier. Deviations from the assumed Fickian diffusion were attributed to co-occurring water diffusion and apparatus-specific properties. In the second and third chapter, mainly the diffusion kinetics of accelerating adjuvants in the cuticular model waxes candelilla wax and carnauba wax were investigated, and simultaneously recorded changes in the wax-specific portion of the IR spectrum were interpreted as indications of plasticization. For this purpose, the oil derivative methyl oleate, as well as the organophosphate ester TEHP and three non-ionic monodisperse alcohol ethoxylates (AEs) C12E2, C12E4 and C12E6 were selected. Strong dependence of diffusion on the respective principal components of the mainly aliphatic waxes was demonstrated. The diffusion kinetics of the investigated adjuvants were faster in the n-alkane dominated candelilla wax than in the alkyl ester dominated carnauba wax. Furthermore, the equilibrium absorptions, indicating equilibrium concentrations, were also higher in candelilla wax than in carnauba wax. It was concluded that alkyl ester dominated waxes feature higher resistance to diffusion of accelerating adjuvants than alkane dominated waxes with shorter average chain lengths due to their structural integrity. This was also found either concerning candelilla/policosanol (n-alcohol) or candelilla/rice bran wax (alkyl-esters) blends: with increasing alcohol concentration, the barrier function was decreased, whereas it was increased with increasing alkyl ester concentration. However, due to the high variability of the individual diffusion curves, only a trend could be assumed here, but significant differences were not shown. The variability itself was described in terms of fluctuating crystalline arrangements and partial phase separation of the respective wax mixtures, which had inevitable effects on the adjuvant diffusion. However, diffusion kinetics also strongly depended on the studied adjuvants. Significantly slower methyl oleate diffusion accompanied by a less pronounced reduction in orthorhombic crystallinity was found in carnauba wax than in candelilla wax, whereas TEHP diffusion was significantly less dependent on the respective wax structure and therefore induced considerable plasticization in both waxes. Of particular interest was the AE diffusion into both waxes. Differences in diffusion kinetics were also found here between candelilla blends and carnauba wax. However, these depended equally on the degree of ethoxylation of the respective AEs. The lipophilic C12E2 showed approximately Fickian diffusion kinetics in both waxes, accompanied by a drastic reduction in orthorhombic crystallinity, especially in candelilla wax, whereas the more hydrophilic C12E6 showed significantly retarded diffusion kinetics associated with a smaller effect on orthorhombic crystallinity. The individual diffusion kinetics of the investigated adjuvants sometimes showed drastic deviations from the Fickian diffusion model, indicating a self-accelerating effect. Hence, adjuvant diffusion kinetics were accompanied by a distinct initial lag phase, indicating a critical concentration in the wax necessary for effective penetration, leading to sigmoidal rather than to exponential diffusion kinetics. The last chapter dealt with the adjuvant-affected diffusion of the AI modelling CNP in candelilla and carnauba wax. Using ATR-FTIR, diffusion kinetics were recorded after adjuvant treatment, all of which were fully explicable based on the Fickian model, with high diffusion coefficients ranging from 10-14 to 10-13 m2 s-1. It is obvious that the diffusion coefficients presented in this work consistently demonstrated plasticization induced accelerated CNP mobilities. Furthermore, CNP equilibrium concentrations were derived, from which partition- and permeability coefficients could be determined. Significant differences between diffusion coefficients (mobility) and partition coefficients (solubility) were found on the one hand depending on the respective waxes, and on the other hand depending on treatment with respective adjuvants. Mobility was higher in candelilla wax than in carnauba wax only after methyl oleate treatment. Treatment with TEHP and AEs resulted in higher CNP mobility in the more polar alkyl ester dominated carnauba wax. The partition coefficients, on the other hand, were significantly lower after methyl oleate treatment in both candelilla and carnauba wax as followed by TEHP or AE treatment. Models were designed for the CNP penetration mode considering the respective adjuvants in both investigated waxes. Co-penetrating water, which is the main ingredient of spray formulations applied in the field, was likely the reason for the drastic differences in adjuvant efficacy. Especially the investigated AEs favored an enormous water uptake in both waxes with increasing ethoxylation level. Surprisingly, this effect was also found for the lipophilic TEHP in both waxes. This led to the assumption that the AI permeability is not exclusively determined by adjuvant induced plasticization, but also depends on a “secondary plasticization”, induced by adjuvant-attracted co-penetrating water, consequently leading to swelling and drastic destabilization of the crystalline wax structure. The successful establishment of the presented ATR-FTIR method represents a milestone for the study of adjuvant and AI diffusion kinetics in cuticular waxes. In particular, the simultaneously detectable wax modification and, moreover, the determinable water uptake form a perfect basis to establish the ATR-FTIR system as a universal screening tool for wax-adjuvants-AI-water interaction in crop protection science., Um ihren Zielort zu erreichen, müssen systemische Pestizide aus einem auf dem Feld ausgebrachten Sprühtropfen in die Pflanze gelangen. Letztere werden in der Klasse der beschleunigenden Adjuvantien zusammengefasst, wobei einzelne Varianten auch oberflächenaktive Eigenschaften haben können. Die Aufnahme eines Pestizids aus einem Sprühtropfen hängt im Wesentlichen von seiner Durchlässigkeit durch die kutikuläre Barriere ab. Die Permeabilität ist ein kombinierter Parameter, der sich aus der Mobilität und der Löslichkeit des Wirkstoffs in der Kutikula ergibt. In den letzten Jahrzehnten wurden mehrere Methoden entwickelt, die die Bestimmung einzelner Parameter der Permeation organischer Verbindungen durch die Kutikularmembran ermöglichen. Frühere Studien zeigten jedoch, dass hauptsächlich kutikuläre Wachse die barrierebestimmende Komponente der Kutikula darstellen, und darüber hinaus wurde gezeigt, dass hauptsächlich die sehr langkettigen aliphatischen Verbindungen (VLCAs) für die Errichtung einer wirksamen Barriere verantwortlich sind. Die Rolle der einzelnen VLCAs, die nach ihren jeweiligen funktionellen Gruppen klassifiziert werden, ist jedoch in Bezug auf die Bestimmung der Barriereeigenschaften noch unbekannt. Daher wurde in dieser Arbeit folgende Ziele verfolgt und erreicht: (1) Ein neuer ATR-FTIR-basierter Ansatz wurde entwickelt, um die temperaturabhängige Echtzeit-Diffusionskinetik von organischen Modellen für Wirkstoffe (AI) in ausschließlich aus Alkanen bestehendem Paraffinwachs zu messen. (2) Der entwickelte ATR-FTIR-Ansatz wurde zur Bestimmung der Diffusionskinetik von selbstbeschleunigenden Adjuvantien in kutikulären Modellwachsen unterschiedlicher VLCA-Zusammensetzung angewendet. Gleichzeitig wurden wachsspezifische Veränderungen in den jeweiligen IR-Spektren aufgezeichnet, welche Informationen über die jeweilige Wachsmodifikation lieferten. (3) Die ATR-FTIR-Methode wurde zur Charakterisierung der Diffusionskinetik, sowie zur Bestimmung der wachsspezifischen Sorptionskapazitäten für eine AI-modellierende organische Verbindung und von Wasser in kutikulären Modellwachsen nach Adjuvans-Behandlung verwendet. Im Hinblick auf die einzelnen chemischen Zusammensetzungen und Strukturen wurden Rückschlüsse auf die adjuvansspezifischen Wirkweisen (MoA) gezogen. Im ersten Kapitel wurde der ATR-FTIR-basierte Ansatz zur Bestimmung der Diffusionskinetik organischer Verbindungen in Paraffinwachs erfolgreich etabliert. Es wurde die Diffusionskinetik der organischen AI-Modellverbindungen Heptylparaben (HPB) und 4-Cyanophenol (CNP) aufgezeichnet, die unterschiedliche Lipophilitäten und Molekülvolumina aufweisen, wie sie für AIs in Pestizidformulierungen typisch sind. Die abgeleiteten Diffusionskoeffizienten lagen im Bereich von 10-15 m2 s-1 und waren damit höher als die zuvor in rekonstituierten kutikulären Wachsen beobachteten Diffusionskoeffizienten. Zum ersten Mal wurde eine ln-lineare Abhängigkeit zwischen den Diffusionskoeffizienten und der angewandten Diffusionstemperatur in kutikulärem Modellwachs nachgewiesen, aus der schließlich Aktivierungsenergien abgeleitet wurden. Die ermittelten Aktivierungsenergien betrugen 66.2 ± 7.4 kJ mol-1 und 56.4 ± 9,8 kJ mol-1 und lagen damit im erwarteten Bereich der bereits gut begründeten Aktivierungsenergien, die für die Diffusion organischer Verbindungen durch kutikuläre Membranen erforderlich sind. Dies bestätigte abermals den signifikanten Beitrag der Wachse zur kutikulären Barriere. Abweichungen von der angenommenen Fick'schen Diffusion wurden auf die gleichzeitig stattfindende Wasserdiffusion und gerätespezifische Artefakte zurückgeführt. Im zweiten und dritten Kapitel wurde vor allem die Diffusionskinetik von beschleunigenden Adjuvantien in den kutikulären Modellwachsen Candelillawachs und Carnaubawachs untersucht und gleichzeitig aufgezeichnete Veränderungen im wachspezifischen Teil des IR-Spektrums als Hinweise auf eine Plastifizierung interpretiert. Zu diesem Zweck wurden das Ölderivat Methyloleat, sowie der Organophosphatester TEHP und drei nichtionische monodisperse Alkoholethoxylate (AEs) C12E2, C12E4 und C12E6 ausgewählt. Es wurde eine starke Abhängigkeit der Adjuvansdiffusion von den jeweiligen Hauptkomponenten der hauptsächlich aliphatisch strukturierten Wachse nachgewiesen. So war die Diffusionskinetik der untersuchten Adjuvantien in dem hauptsächlich aus n-Alkanen bestehenden Candelillawachs schneller als in dem von Alkylestern dominierten Carnaubawachs. Darüber hinaus waren die Gleichgewichtsabsorptionen, die auf Gleichgewichtskonzentrationen hinweisen, in Candelillawachs ebenfalls höher als in Carnaubawachs. Daraus wurde gefolgert, dass Wachse mit hohen Alkylesteranteilen aufgrund ihrer strukturellen Integrität einen höheren Widerstand gegen die Diffusion von beschleunigenden Adjuvantien aufweisen als Wachse mit kürzeren durchschnittlichen Kettenlängen. Dies wurde auch bei Candelilla/Policosanol- (n-Alkohol) oder Candelilla/Reiskleiewachs-Mischungen (Alkylester) festgestellt: Mit steigender Alkoholkonzentration nahm die Barrierefunktion ab, während sie mit steigender Alkylesterkonzentration zunahm. Aufgrund der hohen Variabilität der einzelnen Diffusionskurven konnte hier jedoch nur ein Trend vermutet werden, signifikante Unterschiede zeigten sich jedoch nicht. Die Variabilität selbst wurde mit schwankenden kristallinen Anordnungen und teilweiser Phasentrennung der jeweiligen Wachsmischungen erklärt, die sich zwangsläufig auf die Diffusion der Adjuvantien auswirkten. Die Diffusionskinetik hing jedoch auch stark von den untersuchten Adjuvantien ab. In Carnaubawachs wurde eine deutlich langsamere Methyloleat-Diffusion festgestellt, die mit einer weniger ausgeprägten Verringerung der orthorhombischen Kristallinität einherging als in Candelillawachs, während die TEHP-Diffusion deutlich weniger von der jeweiligen Wachsstruktur abhängig war und in beiden Wachsen eine erhebliche Plastifizierung bewirkte. Von besonderem Interesse war die AE-Diffusion in den untersuchten Wachsen. Auch hier wurden Unterschiede in der Diffusionskinetik zwischen Candelillamischungen und Carnaubawachs festgestellt. Diese hingen jedoch gleichermaßen vom Ethoxylierungsgrad der jeweiligen AEs ab. Das lipophile C12E2 zeigte in beiden Wachsen eine annähernd Fick‘sche Diffusionskinetik, die mit einer drastischen Verringerung der orthorhombischen Kristallinität einherging, insbesondere im Candelillawachs, während das hydrophilere C12E6 eine deutlich verzögerte Diffusionskinetik zeigte, die mit einer geringeren Auswirkung auf die orthorhombische Kristallinität einherging. Die individuellen Diffusionskinetiken der untersuchten Adjuvantien zeigten teilweise drastische Abweichungen vom Fick‘schen Diffusionsmodell, was auf einen selbstbeschleunigenden Effekt hindeutet. Die Diffusionskinetik der Adjuvantien wurde von einer ausgeprägten anfänglichen Verzögerungsphase begleitet, die auf das Erreichen einer kritischen Konzentration im Wachs hindeutet. Es wird angenommen, dass aufgrund der initialen Verzögerungsphase letztlich sigmoidale, statt Fick’sche Diffusionskinetiken vorlagen. Das letzte Kapitel befasste sich mit der adjuvansbeeinflussten Diffusion der für Wirkstoffe modellhaften organischen Substanz CNP in Candelilla- und Carnaubawachs. Mittels ATR-FTIR wurden Diffusionskinetiken nach Adjuvans-Behandlung aufgezeichnet, die alle auf der Grundlage des Fick‘schen Modells vollständig erklärbar waren, einhergehend mit hohen Diffusionskoeffizienten von 10-14 bis 10-13 m2 s-1. Es ist offensichtlich, dass die in dieser Arbeit vorgestellten Diffusionskoeffizienten durchweg eine durch die Plastifizierung bedingte erhöhte CNP-Mobilität belegen. Darüber hinaus wurden CNP-Gleichgewichtskonzentrationen abgeleitet, aus denen Verteilungs- und Permeabilitätskoeffizienten bestimmt werden konnten. Signifikante Unterschiede zwischen Diffusionskoeffizienten (Mobilität) und Verteilungskoeffizienten (Löslichkeit) wurden zum einen in Abhängigkeit von den jeweiligen Wachsen und zum anderen in Abhängigkeit von den jeweiligen Adjuvantien festgestellt. Die CNP-Mobilität war in Candelillawachs nur nach Behandlung mit Methyloleat höher als in Carnaubawachs. Die Behandlung mit TEHP und AEs führte zu einer höheren CNP-Mobilität in dem polaren, von Alkylestern dominierten Carnaubawachs. Die Verteilungskoeffizienten hingegen waren nach der Behandlung mit Methyloleat sowohl in Candelilla- als auch in Carnaubawachs deutlich niedriger als nach der Behandlung mit TEHP oder AE. Es wurden Modelle für den CNP-Penetrationsmodus unter Berücksichtigung der jeweiligen Adjuvantien in den beiden untersuchten Wachsen entwickelt. Der Grund für die drastischen Unterschiede in der Wirksamkeit der Adjuvantien liegt wahrscheinlich im Ko-Penetrieren von Wasser, dem Hauptbestandteil der auf dem Feld angewandten Spritzformulierungen. Insbesondere die untersuchten AEs begünstigten eine enorme Wasseraufnahme in beiden Wachsen mit zunehmendem Ethoxylierungsgrad. Überraschenderweise wurde dieser Effekt auch für das lipophile TEHP in beiden Wachsen gefunden. Dies führte zu der Vermutung, dass die AI-Permeabilität nicht ausschließlich durch die adjuvansinduzierte Plastifizierung bestimmt wird, sondern auch von einer "sekundären Plastifizierung" abhängt, die durch die Ko-Penetration von Wasser induziert wird und so zur Quellung und drastischen Destabilisierung der kristallinen Wachsstruktur führt. Die erfolgreiche Etablierung der vorgestellten ATR-FTIR-Methode stellt einen Meilenstein für die Untersuchung der Diffusionskinetik von Adjuvantien und AIs in kutikulären Wachsen dar. Insbesondere die gleichzeitig nachweisbare Wachsmodifikation und darüber hinaus die bestimmbare Wasseraufnahme bilden eine perfekte Grundlage, um das ATR-FTIR-System als universelles Screening-Tool für Wachs-Adjuvans-AI-Wasser-Interaktionen in der Pflanzenschutzwissenschaft zu etablieren.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Landscaping is essential for ecological balance, as green spaces are being increasingly occupied with urbanization. It is incongruous that in a country with one of the richest, most diverse and important floras in the world, most of the plants used in landscaping are still exotic, some of which are invasive. With the little appreciation of native species, the urgency of modifying this dynamic is understandable, and analyzing the high rate of urbanization in a global context, the water crisis and the threat of extinction of so many species of fauna and flora, it appears that the Cities must now be seen as ecological entities, where human action must try to reestablish the local environmental balance. Since the State of São Paulo is the most urbanized place in Brazil, concentrating the largest contingent of population residing in urban areas, in addition to the presence of two threatened biomes, Atlantic Forest and Cerrado, we limit one of its most economically significant Metropolitan Regions as an object of study, the Metropolitan Region of Sorocaba (RMS), in order to carry out a survey of local landscape demand and which are the main species offered in regional commerce in the scale of production, sale and management, identifying the groups with the highest output and if there is a market for herbs, shrubs and local native trees for ornamentation, and what material is currently available to promote this sector. The methodology focused on bibliographic research to understand the importance of biodiversity in urban gardens, and quantitative and qualitative research in the floristic market of this regional cut, as well as data collection of working professionals and population profile on the subject, with the purpose of understanding if there is a demand for these vegetations and how their supply and availability in nurseries and floras works. Thus, the standardization of landscapes in the region was verified, where the most offered plants are exotic and little diverse, and the professionals responsible for the projects have an aesthetic vision in disagreement with the population sample studied. L'aménagement paysager est essentiel pour l'équilibre écologique, car les espaces verts sont de plus en plus occupé par l'urbanisation. Es incongruente que en un país con uno de los más rico, diverso e importante en el mundo, siendo la mayoría de las plantas utilizadas en el paisajismo exóticos, algunos de los cuales son invasivos. Con poca apreciación de las especies nativas, comprender la urgencia de cambiar esta dinámica, y analizar el alto índice de urbanización en un contexto global, la crisis del agua y la amenaza de extinción de tantas especies de fauna y flora, parece que las ciudades ahora deben ser vistas como entidades ecológicas, donde la acción la actividad antrópica debe tratar de restablecer el equilibrio ambiental local. La metodología se centró en la investigación. literatura para comprender la importancia de la biodiversidad en los jardines urbanos, y investigación cuantitativa y cualitativa en el mercado florístico de este corte regional, así como recopilación de datos de los profesionales en activo y el perfil de la población sobre el tema, con el objetivo de con el propósito de entender si hay demanda de estas vegetaciones y cómo la oferta y su disponibilidad en viveros y flora. É incongruente que num país com uma das floras mais ricas, diversas e importantes do mundo, ainda a maior parte das plantas utilizadas no paisagismo seja exótica, algumas das quais, invasoras. Com a pouca valorização das espécies nativas, compreende-se a urgência em modificar essa dinâmica, e analisando a alta taxa de urbanização num contexto mundial, a crise hídrica e a ameaça de extinção de tantas espécies de fauna e flora, constata-se que as cidades agora devem ser vistas como entidades ecológicas, onde a ação antrópica deve tentar reestabelecer o equilibro ambiental local. Sendo o Estado de São Paulo o local mais urbanizado do Brasil, concentrando o maior contingente populacional residindo em áreas urbanas, além de presente dois biomas ameaçados, mata atlântica e cerrado, limitamos uma de suas Regiões Metropolitanas mais significativas economicamente como objeto de estudo, a Região Metropolitana de Sorocaba (RMS), afim de realizar um levantamento da demanda paisagística local e quais são as principais espécies ofertadas no comércio regional na escala de produção, venda e manejo, identificando os grupos de maior saída e se há mercado para herbáceas, arbustivas e arbóreas nativas locais para ornamentação, e qual material é disponibilizado atualmente para fomentar esse setor. A metodologia se concentrou na pesquisa bibliográfica para compreensão da importância da biodiversidade nos jardins urbanos, e investigação quantitativa e qualitativa no mercado florístico desse recorte regional, bem como levantamento de dados dos profissionais atuantes e perfil da população perante o tema, com o propósito de compreender se existe demanda dessas vegetações e como funciona a oferta e disponibilidade das mesmas em viveiros e floras. Verificou-se assim, a padronização de paisagens na região, onde as plantas mais ofertadas são exóticas e pouco diversas, e os profissionais responsáveis pelos projetos possuem visão estética em discordância com a amostra populacional estudada. Pharmaceuticals as pollutants have garnered increased scientific interest over recent years. Their continuous release results in a pseudo persistence, which can lead to a constant exposure of organisms to active pharmaceutical ingredients (APIs). APIs are commonly found in wastewater. Most wastewater treatment plants (WWTPs) are not equipped to sufficiently remove APIs from their effluent streams. Sewage sludge used as fertilizer presents another route of contamination. This thesis focuses on the uptake and metabolization of antidepressant medication (ADM) by edible plants. Citalopram and venlafaxine were chosen as two common antidepressants. Cress, peas, and maize were grown hydroponically in deliberately contaminated tap water solutions. The plants were separated into different parts, homogenized, and extracted with a 2:1 solution of hydrochloric acid (0.1 M) and acetonitrile (ACN). The samples were separated with reversed phase high performance liquid chromatography (RP HPLC) using a water/methanol gradient. The detector of choice was a quadrupole time of flight (QToF) instrument equipped with a drift tube (DT) for ion mobility (IM) measurements and collision cell for MS/MS. The detector was operated as MS, MS/MS, IM MS, and IM MS/MS. The main focus of this thesis is the untargeted screening for potential API metabolites using a statistical analysis of IM MS data, comparing extracts of treated and untreated plants; and subsequently validating the identity of these metabolite candidates using targeted MS/MS and IM MS/MS. A set of targeted screening methods were also performed in MS mode, by searching the generated HPLC MS data using a m/z database of hypothetical phase I and phase II metabolites of venlafaxine and citalopram. In addition to metabolite screening, this thesis includes a quasi quantitative uptake study, monitoring the uptake of both APIs in cress. Es ist bekannt, dass Pflanzen einen spezifischen Lebensraum für mikrobielle Gemeinschaften darstellen, die mit ihrem Wirt in wesentlichen funktionellen Zusammenhängen interagieren. Es wird eine mögliche Verbindung zwischen den Samen und der Kontinuität des Pflanzenmikrobioms über Generationen hinweg angenommen. Kürzlich wurde gezeigt, dass Samen während der Keimung unterschiedliche mikrobielle Gemeinschaften an die Phyllosphäre und die Wurzeln während der Entwicklung der Pflanze weitergeben. Derzeit ist jedoch noch nur wenig bekannt über die metabolischen Fähigkeiten des vertikal vererbten Mikrobioms. Das Ziel dieser Studie war es, Erkenntnisse über das vertikal übertragene Mikrobiom zu gewinnen. Insbesondere die Unterschiede in der funktionellen Vielfalt zwischen Wurzeln und Phyllosphäre und ihre Fähigkeit, verschiedene Kohlenstoffquellen zu verstoffwechseln. Weiters wurde ein endophytischer Rhodanobacter isoliert und sequenziert. Die Studie zeigte, dass der Artenreichtum, die Shannon-Diversität sowie die Anzahl der Bakterien in den Wurzeln höher waren als in der Phyllosphäre. Außerdem war die Stoffwechselkapazität zwischen Wurzeln und Phyllosphäre unterschiedlich. Die größten Unterschiede bei der Verstoffwechselung von Kohlenstoff gab es bei D-Xylose, Phenylalanin, 4-Hydroxybenzoesäure, α-Cyclodextrin und Glykogen. Im Allgemeinen war die Zusammensetzung des Mikrobioms der Wurzeln vielfältiger als die der Phyllosphäre, aber beide wurden von Pseudomonas (Phyllosphäre: 88,2 %, Wurzeln: 84,4 %) und Yersinia (Phyllosphäre: 11,7 %, Wurzeln: 15,5 %) dominiert. Das sequenzierte Genom des Rhodanobacters wurde zu einem 4 Mb großen Genom zusammengesetzt und ca. 1800 verschiedene Gene wurden annotiert. Drei Gene mit möglichen wachstums- und gesundheitsfördernden Funktionen wurden entdeckt: pqqC2, budB und nirK. Außerdem wurden drei verschiedene Sekretionssysteme (Typ-II-, Typ-IV- und Typ-VI-Sekretionssystem) und mehrere Resistenzgene gefunden. Plant endosphere is a specific habitat for endophytic microbial communities where essential functional interactions occurs. Recently, seeds were shown to transmit distinct microbial communities to phyllosphere and roots of new plants during germination, ensuring the continuity of the plant microbiome across generations. However, information regarding the metabolic capability of these vertically transmitted communities is lacking. The aim of this study was to identify the differences in microbial functional diversity transmitted from seed to roots and phyllosphere and their ability to metabolize different carbon sources. Among the isolated seed endophytes, Rhodanobacter was selected for whole genome sequencing to gain further insights into the function of the seed microbiome. The study showed that species richness, Shannon diversity as well as the abundance of bacteria were higher in roots than phyllosphere. Further, the metabolic capacity was different between roots and phyllosphere. Main differences of carbon metabolization occurred in D-xylose, phenylalanine, 4-hydroxy benzoic acid, α-cyclodextrin and glycogen. Carbon source had a signficant impact on species richness and Shannon diversity in roots, but not in phyllosphere. In general, the composition of the rhizosphere microbiome was more diverse than the microbiome of the phyllosphere, but both were dominated by Pseudomonas (phyllosphere: 88,2%, roots: 84,4%) and Yersinia (phyllosphere: 11,7%, roots: 15,5%). The sequenced Rhodanobacter genome was assembled to an approximately 4 Mb genome and about 1800 different genes were annotated. Three genes in possible growth and health promoting functions were detected: pqqC2 (function in phosphate solubilization), budB (function in acetoine/2,3-butanediol synthesis) and nirK (function in nitric oxide synthesis). Further, three different secretion systems (type-II-, type-IV- and type-VI-secretion system) and several resistance genes were found. The increasing demand for pharmaceuticals and personal care products (PPCPs) in human and veterinary medicine has led to multiple contamination sites of these products in the ecosystem. These substances enter the environment via direct excretion from human or animal consumption and improper disposal of unused products. The PPCPs enter the municipal wastewater system and are subsequently filtered by wastewater-treatment plants (WWTPs). However, studies investigating the removal efficiency of WWTPs show incomplete elimination of PPCPs. Inevitably, these compounds contaminate surface waters and ground waters. As the effects of climate change causes droughts and aridity in many regions of the world, the demand for reclaimed, treated wastewater is high. Consequently, the irrigation with treated wastewater leads to contamination in agriculture. Furthermore, the application of manure as fertilizer is a direct source for contamination. Thus, PPCPs enter crops and plants cultivated to produce food. Compared to human metabolism, plants have a different strategy to deal with xenobiotic substances. They first transform these substances into compounds of reduced plant toxicity and store. The biotransformation step involves three phases. In phase I the polarity of the xenobiotic is increased and sites for conjugation in phase II are introduced to the chemical structure. Subsequently, the phase I metabolite is conjugated with glycosides or amino acids. Finally, the conjugated substance is stored in the cell wall or the vacuoles. As the plant cannot simply excrete the potentially harmful compounds, it is reasonable to assume that they accumulate in the plant matrix. This study aims to identify phase I and phase II metabolites of two commonly used active pharmaceutical ingredients (API) in the treatment of hypertension, azilsartan and olmesartan and their prodrugs azilsartan medoxomil and olmesartan medoxomil. To investigate metabolization and uptake of these compounds, plants were cultivated in a solution, containing the active pharmaceutical ingredient (API). Garden cress, maize, pea, onion, carrot, barley, sorghum, rye, triticale was used as model plants. The plant matrix was separated into roots, stem, and leaves, extracted, and analyzed with reverse phase high performance liquid chromatography coupled to drift-tube-ion-mobility quadrupole time-of-flight mass spectrometer (RP-HPLC-DTIM-QTOF-MS). Targeted MS/MS data was compared to a library containing the m/z ratios of possible phase I and phase II metabolites. Furthermore, group comparison of API-solution treated plants and plants cultivated in tap water was conducted via statistical analysis. Up-regulated substances in the API-treated group were further investigated with tandem MS/MS experiments. The fragmentation pattern of possible phase II metabolites was compared to the pattern of azilsartan or olmesartan and their phase I metabolites. Additionally, an alternating frame-all ions method was developed, which yielded valuable precursor and associated product ion information. Direct correlation of product ions and associated precursor ions was possible with the generated collision cross section (DTCCSN2). With this workflow 11 metabolites of azilsartan and 7 metabolites of olmesartan were tentatively identified by comparison of the fragmentation pattern of API and metabolite in tandem MS/MS experiments. For quantification of olmesartan and azilsartan via QqQ-MS, an optimized MRM method was developed. Olmesartan was identified and quantified via external calibration in a water sample from local river in Reichenthal with 0.9 µg L-1. Azilsartan was detected in this sample below the LOQ. The same water sample was then used to cultivate garden cress hydroponically. Subsequently, olmesartan was identified in the root matrix of this garden cress sample and quantified via matrix matched calibration. During the course of evolution, land plants evolved a peculiar haplodiplontic life cycle in which both the haploid gametophyte and the diploid sporophyte are multicellular. Within the life cycle, the phase transition between these two stages is called the alternation of generations. However, within land plants the dominant generation varies. Following the alternation of generations on molecular level is difficult to analyse in flowering plants due to their diploid-dominant generation and their highly reduced gametophytic generation. Bryophytes, in contrast, are haploid-dominant plants, have more easily tractable (morphologically larger) generations and share with flowering plants the fundamental regulatory networks for switching between vegetative and reproductive growth. This switch is most often regulated by transcription associated proteins (TAPs). Combining an orthology detection tool (proteinortho), protein-family categorization (TAPscan), literature search and phylogenetic inference a set of candidate genes, representing single copy TAPs potentially involved in embryo development, was identified. In bryophytes the knowledge of sexual reproduction and evolution of plant sexual reproduction is limited as compared to seed plants. Even though, bryophyte plants defective in gametangiogenesis or embryo/sporophyte development often can be grown as gametophytic tissue and vegetatively propagated contrary to flowering plants. Two master regulators (swi3a/b, hag1) were identified. It could be shown that they are indeed crucial for sexual reproduction and thus the knowledge about the regulation of sexual reproduction in bryophytes could be amended. Deletion of the respective genes led to a loss of sporophyte development due to an impairment in the male germ line in the latest steps of spermatozoid ripening on the part of swi3a/b, and an impairment in the male and female germ lines (disturbed ripening) on the part of hag1. Hag1 is a highly conserved gene throughout all eukaryotic kingdoms, which was proven by phylogenetic analyses and underlined by mutants impaired in the male germ line in moss as well as flowering plants. In contrast, the SWI3 family diversified during land plant evolution (one SWI3 gene in algae, four SWI3 genes in Arabidopsis thaliana). That was underlined by the phenotypical analyses in Physcomitrium patens and A. thaliana, which showed a shift from a male exclusive role of swi3a/b in P. patens towards an involvement in male and female gametogenesis as well as embryogenesis in A. thaliana. These findings underline the hypothesis that on one hand TAPs can be highly conserved and can serve as single copy phylogenetic markers, whereas on the other hand TAP families expanded with plant complexity as drivers for morphogenic evolution. All in all, TAPs are important developmental regulators, that can be used for analysing the plants’ potential to adapt to new conditions., Im Laufe der Evolution entwickelten Landpflanzen einen haplodiplontischen Lebenszyklus, in dem sowohl der haploide Gametophyt als auch der diploide Sporophyt mehrzellig sind. Bryophyten hingegen sind haploid dominante Pflanzen, haben leichter untersuchbare (morphologisch größere) Generationen und haben mit den Blütenpflanzen die grundlegenden regulatorischen Netzwerke für den Wechsel zwischen vegetativem und reproduktivem Wachstum gemeinsam. Dieser Wechsel wird häufig durch transkriptionsassoziierte Proteine (TAPs) reguliert. Durch die Kombination aus einem orthologermittlungs Programm (Proteinortho), Proteinfamilien Kategorisierung (TAPscan), Literatursuche und phylogenetischer Inferenz konnte eine Reihe von Kandidatengenen identifiziert warden. Diese repräsentieren TAPs mit nur einer Kopie, welche möglicherweise in die Embryonalentwicklung involviert sind. Bei Bryophyten ist das Wissen über sexuelle Reproduktion und die Evolution derselbigen im Vergleich zu Samenpflanzen begrenzt. Bryophyten, welche Defekte in der Gametangiogenese oder Embryo-/Sporophytenentwicklung zeigen, können als gametophytisches Gewebe angezogen werden und im Gegensatz zu Blütenpflanzen vegetativ vermehrt werden. Es wurden zwei Masterregulatoren (swi3a/b, hag1) identifiziert und für diese wurde gezeigt, dass sie essentiell für die sexuelle Reproduktion sind. Folglich wurde das Wissen über die Regulation eben jener bei Bryophyten erweitert. Loss-of-function Mutationen führten zu einem Verlust der Sporophytenentwicklung bedingt durch eine Beeinträchtigung der männlichen Keimbahn in den finalen Schritten der Spermatozoidreifung seitens swi3a/b und eine Beeinträchtigung der männlichen und weiblichen Keimbahn (gestörte Reifung) seitens hag1. Hag1 ist ein hochkonserviertes Gen in allen eukaryotischen Reichen, was durch phylogenetische Analysen impliziert und durch Mutanten, die in der männlichen Keimbahn sowohl in Moosen als auch in Blütenpflanzen beeinträchtigt sind, unterstrichen wurde. Im Gegensatz dazu diversifizierte die SWI3-Familie innerhalb der Evolution von Landpflanzen (ein SWI3-Gen in Algen, vier SWI3-Gene in A. thaliana). Dies wurde durch phänotypische Untersuchungen bei P. patens und A. thaliana verdeutlicht, die eine Verschiebung von einer ausschließlichen Kontrolle der männlichen Keimbahn von swi3a/b bei P. patens zu einer Beteiligung an der männlichen und weiblichen Gametogenese sowie der Embryogenese bei A. thaliana zeigten. Diese Analysen unterstreichen die Hypothese, dass einerseits TAPs hoch konserviert sind und als taxonomische Marker herangezogen werden können, andererseits TAP Familien im Zuge der Evolution morphologischer Komplexität expandierten. Zusammengefasst sind TAPs wichtige Entwicklungsregulatoren, die für das Verständnis des Anpassungspotentials von Pflanzen an neue Bedingungen von besonderem Interesse sind.

Plant cell walls represent the first line of defence against biotic stresses, including phytopathogens and herbivores. The mustard leaf beetle Phaedon cochleariae feeds on brassicaceous plants and possesses various plant cell wall degrading enzymes for the digestion of plant cell wall polysaccharides. Amongst those, polygalacturonases (PGs) of the glycoside hydrolase family 28 (GH28) facilitate the breakdown of pectin. Several PG-inhibiting proteins (PGIPs) – plant-derived, cell wall-associated proteins containing multiple leucine-rich repeat (LRR) motifs – have been shown to inhibit microbial PGs and thus contribute to the defence against phytopathogens. This thesis takes a biochemical approach to elucidate the interaction between the P. cochleariae pectinolytic system and plant-derived inhibitors. It provides the first direct evidence that beetle PGs are inhibited by PGIPs and suggests that PGIPs play a role in the plant defence against herbivorous beetles. Beyond the “classical” PG-PGIP interaction, my studies link new players – PG pseudoenzymes and PGIP-like proteins – to the complex interplay between herbivore and plant. These discoveries form the basis for future in-depth studies on how pectin digestion in herbivorous insects and the PGIP-mediated plant defence system co-evolved.

The right timing of phenological events is crucial for species fitness. Species should be highly synchronized with mutualists, but desynchronized with antagonists. With climate warming phenological events advance in many species. However, often species do not respond uniformly to warming temperatures. Species-specific responses to climate warming can lead to asynchrony or even temporal mismatch of interacting species. A temporal mismatch between mutualists, which benefit from each other, can have negative consequences for both interaction partners. For host-parasitoid interactions temporal asynchrony can benefit the host species, if it can temporally escape its parasitoid, with negative consequences for the parasitoid species, but benefit the parasitoid species if it increases synchrony with its host, which can negatively affect the host species. Knowledge about the drivers of phenology and the species-specific responses to these drivers are important to predict future effects of climate change on trophic interactions. In this dissertation I investigated how different drivers act on early flowering phenology and how climate warming affects the tritrophic relationship of two spring bees (Osmia cornuta & Osmia bicornis), an early spring plant (Pulsatilla vulgaris), which is one of the major food plants of the spring bees, and three main parasitoids of the spring bees (Cacoxenus indagator, Anthrax anthrax, Monodontomerus). In Chapter II I present a study in which I investigated how different drivers and their change over the season affect the reproductive success of an early spring plant. For that I recorded on eight calcareous grasslands around Würzburg, Germany the intra-seasonal changes in pollinator availability, number of co-flowering plants and weather conditions and studied how they affect flower visitation rates, floral longevity and seed set of the early spring plant P. vulgaris. I show that bee abundances and the number of hours, which allowed pollinator foraging, were low at the beginning of the season, but increased over time. However, flower visitation rates and estimated total number of bee visits were higher on early flowers of P. vulgaris than later flowers. Flower visitation rates were also positively related to seed set. Over time and with increasing competition for pollinators by increasing numbers of co-flowering plants flower visitation rates decreased. My data shows that a major driver for early flowering dates seems to be low interspecific competition for pollinators, but not low pollinator abundances and unfavourable weather conditions. Chapter III presents a study in which I investigated the effects of temperature on solitary bee emergence and on the flowering of their food plant and of co-flowering plants in the field. Therefore I placed bee cocoons of two spring bees (O. cornuta & O. bicornis) on eleven calcareous grasslands which differed in mean site temperature. On seven of these grasslands the early spring plant P. vulgaris occurred. I show that warmer temperatures advanced mean emergence in O. cornuta males. However, O. bicornis males and females of both species did not shift their emergence. Compared to the bees P. vulgaris advanced its flowering phenology more strongly with warmer temperatures. Co-flowering plants did not shift flowering onset. I suggest that with climate warming the first flowers of P. vulgaris face an increased risk of pollinator limitation whereas for bees a shift in floral resources may occur. In Chapter IV I present a study in which I investigated the effects of climate warming on host-parasitoid relationships. I studied how temperature and photoperiod affect emergence phenology in two spring bees (O. cornuta & O. bicornis) and three of their main parasitoids (C. indagator, A. anthrax, Monodontomerus). In a climate chamber experiment with a crossed design I exposed cocoons within nest cavities and cocoons outside of nest cavities to two different temperature regimes (long-term mean of Würzburg, Germany and long-term mean of Würzburg + 4 °C) and three photoperiods (Würzburg vs. Snåsa, Norway vs. constant darkness) and recorded the time of bee and parasitoid emergence. I show that warmer temperatures advanced emergence in all studied species, but bees advanced less strongly than parasitoids. Consequently, the time period between female bee emergence and parasitoid emergence decreased in the warm temperature treatment compared to the cold one. Photoperiod influenced the time of emergence only in cocoons outside of nest cavities (except O. bicornis male emergence). The data also shows that the effect of photoperiod compared to the effect of temperature on emergence phenology was much weaker. I suggest that with climate warming the synchrony of emergence phenologies of bees and their parasitoids will amplify. Therefore, parasitism rates in solitary bees might increase which can negatively affect reproductive success and population size. In this dissertation I show that for early flowering spring plants low interspecific competition for pollinators with co-flowering plants is a major driver of flowering phenology, whereas other drivers, like low pollinator abundances and unfavourable weather conditions are only of minor importance. With climate warming the strength of different drivers, which act on the timing of phenological events, can change, like temperature. I show that warmer temperatures advance early spring plant flowering more strongly than bee emergence and flowering phenology of later co-flowering plants. Furthermore, I show that warmer temperatures advance parasitoid emergence more strongly than bee emergence. Whereas temperature changes can lead to non-uniform temporal shifts, I demonstrate that geographic range shifts and with that altered photoperiods will not change emergence phenology in bees and their parasitoids. In the tritrophic system I investigated in this dissertation climate warming may negatively affect the reproductive success of the early spring plant and the spring bees but not of the parasitoids, which may even benefit from warming temperatures., Der richtige Zeitpunkt von phänologischen Ereignissen ist maßgeblich für das Überleben und die Fortpflanzung einer Art. Arten sollten eine möglichst hohe Synchronisation mit Mutualisten aufweisen, aber eine möglichst geringe mit Antagonisten. Die Klimaerwärmung führt dazu, dass sich bei vielen Arten phänologische Ereignisse verfrühen. Allerdings reagieren Arten unterschiedlich auf wärmere Temperaturen. Artspezifische Reaktionen auf die Klimaerwärmung können zu Asynchronität oder sogar zu zeitlicher Diskrepanz bei interagierenden Arten führen. Eine zeitliche Diskrepanz zwischen Mutualisten, die voneinander profitieren, kann sich negativ auf beide Interaktionspartner auswirken. Bei Wirt-Parasitoid Beziehungen kann der Wirt von einer zeitlichen Diskrepanz profitieren, wenn er seinem Parasitoid zeitlich entfliehen kann, was wiederum negative Folgen für den Parasitoid haben kann. Jedoch kann der Parasitoid profitieren, wenn er die Synchronisation mit seinem Wirt erhöhen kann, was wiederum den Wirt negativ beeinflussen kann. Das Wissen über die Treiber von phänologischen Ereignissen und die artspezifischen Reaktionen auf diese Treiber sind von Bedeutung um die Auswirkungen des Klimawandels auf trophische Beziehungen vorherzusagen. In meiner Doktorarbeit habe ich untersucht, wie verschiedene Treiber mit einer frühen Blüte zusammenhängen und wie der Klimawandel die tritrophische Beziehung von zwei Frühlingsbienen (Osmia cornuta & Osmia bicornis), einer Frühlingspflanzenart (Pulsatilla vulgaris), die eine der wichtigen Futterpflanzen der Bienen ist, und der drei Hauptparasitoiden der Frühlingsbienen (Cacoxenus indagator, Anthrax anthrax, Monodontomerus) beeinflusst. In Kapitel II präsentiere ich eine Studie, in der ich den Einfluss verschiedener Treiber und ihre saisonale Veränderung auf den Fortpflanzungserfolg einer Frühlingspflanzenart untersucht habe. Dazu habe ich auf acht Kalkmagerrasen bei Würzburg (Deutschland) die innersaisonalen Veränderungen der Bestäuberverfügbarkeit, der Anzahl an gleichzeitig blühenden Pflanzenarten und die Wetterbedingungen aufgezeichnet. Des Weiteren habe ich erforscht wie diese Faktoren die Blütenbesuchsrate, die Blütenlanglebigkeit und den Samenansatz der Frühlingspflanze P. vulgaris beeinflussen. Ich konnte zeigen, dass die Anzahl an Bienen und die Anzahl an Stunden, die ein Furagieren von Bestäubern ermöglicht hätten, am Anfang der Saison niedrig waren und mit der Zeit zunahmen. Jedoch war die Blütenbesuchsrate und die geschätzte Anzahl an Bienenbesuchen höher bei frühen als bei späten P. vulgaris Blüten. Die Blütenbesuchsrate wirkte sich auch positiv auf den Samenansatz aus. Die Blütenbesuchsrate nahm mit der Zeit und mit zunehmender Konkurrenz um Bestäuber durch eine zunehmende Anzahl an gleichzeitig blühenden Pflanzenarten ab. Meine Daten zeigen, dass ein Hauptreiber von frühen Blühzeitpunkten die geringe zwischenartliche Konkurrenz um Bestäuber ist, aber nicht die niedrige Bestäuberanzahl und ungünstige Wetterbedingungen. Kapitel III präsentiert eine Studie, in welcher ich die Auswirkungen der Temperatur auf den Schlupf von Solitärbienen und die Blüte ihrer Futterpflanzen und gleichzeitig blühenden Pflanzen im Freiland untersucht habe. Dafür habe ich Bienenkokons von zwei Frühlingsbienen (O. cornuta & O. bicornis) auf elf Kalkmagerrasen, die sich in der mittleren Flächentemperatur unterschieden, platziert. Auf sieben dieser Kalkmagerrasen kam die Frühlingspflanzenart P. vulgaris vor. Ich konnte zeigen, dass wärmere Temperaturen den mittleren Schlupf von O. cornuta Männchen verfrühen. Die Männchen von O. bicornis und die Weibchen beider Arten haben ihren Schlupfzeitpunkt jedoch nicht verschoben. Im Vergleich zu den Bienen verfrühte P. vulgaris seine Blühphänologie bei warmen Temperaturen stärker. Die gleichzeitig blühenden Pflanzenarten verschoben ihren Blühbeginn nicht. Die Daten zeigen, dass wärmere Temperaturen den Bienenschlupf weniger stark verfrühen als die Blüte ihrer Futterpflanze. Das lässt darauf schließen, dass mit dem Klimawandel die ersten Blüten von P. vulgaris ein erhöhtes Risiko haben nicht bestäubt zu werden, während die Bienen möglicherweise auf andere Blühressourcen ausweichen müssen. Kapitel IV beschreibt eine Studie, in welcher ich die Auswirkungen der Klimaerwärmung auf Wirt-Parasitoid Beziehungen untersucht habe. Dabei habe ich die Auswirkungen von Temperatur und Photoperiode auf die Schlupfphänologie zweier Frühlingsbienen (O. cornuta & O. bicornis) und drei ihrer Hauptparasitoide (C. indagator, A. anthrax, Monodontomerus) erforscht. In einem Klimakammerexperiment mit gekreuztem Design habe ich Kokons in Nesthöhlen und Kokons außerhalb von Nesthöhlen, zwei verschiedenen Temperaturregimen (Langzeitmittel von Würzburg, Deutschland und Langzeitmittel von Würzburg + 4 °C) und drei Photoperioden (Würzburg, Deutschland contra Snåsa, Norwegen contra Dauerdunkel) ausgesetzt und die Zeitpunkte des Bienen- und Parasitoidenschlupfes aufgezeichnet. Ich konnte zeigen, dass warme Temperaturen in allen untersuchten Arten den Schlupfzeitpunkt verfrühten, jedoch bei den Bienen weniger stark als bei den Parasitoiden. Eine Folge daraus ist, dass sich die Zeitspanne zwischen dem Schlupf der Bienenweibchen und dem Schlupf der Parasitoide im warmen Temperaturregime im Vergleich zum kalten verkürzte. Die Photoperiode hatte auf den Zeitpunkt des Schlupfes nur in Kokons außerhalb von Nisthöhlen einen Effekt (außer beim Schlupf von O. bicornis Männchen). Die Daten zeigen auch, dass der Effekt der Photoperiode auf die Schlupfphänologie im Vergleich zu dem Effekt der Temperatur viel schwächer war. Daraus schließe ich, dass sich im Zuge der Klimaerwärmung die Synchronisation der Schlupfphänologien von Bienen und ihren Parasitoiden verstärken wird. Eine Folge davon könnten erhöhte Parasitierungsraten bei Solitärbienen sein, welche den Reproduktionserfolg und die Populationsgröße negativ beeinflussen können. In dieser Doktorarbeit habe ich gezeigt, dass einer der Haupttreiber einer frühen Blüte bei Frühlingspflanzen geringe zwischenartliche Konkurrenz um Bestäuber mit später gleichzeitig blühenden Pflanzenarten ist, während andere Treiber, wie geringe Bestäuberabundanzen und ungünstige Wetterbedingungen nur von geringer Bedeutung sind. Im Zuge des Klimawandels könnte sich die Stärke verschiedener Treiber, die den Zeitpunkt von phänologischen Ereignissen beeinflussen, verändern. Ich konnte außerdem zeigen, dass wärmere Temperaturen die Blüte von frühen Frühlingspflanzen stärker verfrühen, als den Schlupf von Bienen und die Blüte von später gleichzeitig blühenden Pflanzenarten. Des Weiteren zeigte ich, dass wärmere Temperaturen den Schlupf von Parasitoiden stärker verfrühen als den Schlupf der Bienen. Ich konnte zeigen, dass während Temperaturveränderungen zu verschieden starken zeitlichen Verschiebungen führen können, Verschiebungen von geografischen Verbreitungsgebieten und damit geänderten Photoperioden die Schlupfphänologie von Bienen und ihren Parasitoiden wahrscheinlich nicht ändern werden. In dem tritrophischen System, das ich in dieser Doktorarbeit untersucht habe, könnte die Erwärmung des Klimas den Fortpflanzungserfolg der frühen Frühlingspflanze und der Frühlingsbienen negativ beeinflussen, aber wahrscheinlich nicht die der Parasitoide, die vielleicht sogar davon profitieren können.

Eisen (Fe) liegt an vierter Stelle in der Fülle von Elementen in der Erdkruste, aber Fe-Mangel ist ein weit verbreitetes Problem bei Pflanzen und Tieren, da die Fe-Oxide unlöslich sind. Fe-Mangel führt zu einer Verringerung der Pflanzenproduktivität und am deutlichsten zu einer erhöhten Fe-induzierten Anämie beim Menschen. Es wurde angenommen, dass die Biofortifizierung von Fe ein praktischer Ansatz zur Verbesserung der Nährstoffqualität in Pflanzen und damit auch in Lebensmitteln oder Tierfutter ist. In dieser Arbeit wurden neue Strategien zur Erhöhung des Fe-Gehalts in der Modellpflanze Arabidopsis getestet. Vakuoläre Fe-Transportgene der VTL-Familie wurden in Kombination mit dem neu entdeckten Fe-Regulierungsprotein IMA1oder dem Fe-Bindungspeptid NAS3 überexprimiert. Zusammenfassend wurde gezeigt, dass die Überexpression von VTL1, 2, 3, 4 oder 5, IMA1 oder NAS3 mit erhöhtem Eisengehalt im Samen korreliert. Die Expression der Gene für die Aufnahme von Fe und die Homöostase bestätigten den erhöhten Fe-Gehalt in diesen überexprimierenden Pflanzen. Die doppelte Überexpression der VTL-Gene in Kombination mit IMA1 oder NAS3 führte zu keinem weiteren Anstieg des Fe-Gehaltes, der wahrscheinlich durch die Regulation der VTL-Gene durch IMA1-Expression und den Mangel an erhöhtem Nicotianamin im Fall von VTL5 / NAS3-Überexpressionpflanzen verursacht würde. Zukünftige Forschung sollte der Übertragbarkeit dieser Ergebnisse auf Kulturpflanzen gewidmet sein., Iron (Fe) ranks fourth in an abundance of elements in the Earth’s crust, but Fe deficiency is a widespread problem in plants and animals because of the insolubility of Fe oxides. Fe deficiency leads to reduce plant productivity and most significantly to enhanced Fe-induced anemia in humans. Fe biofortification has been suggested to be a practical approach for improving the nutritional quality of plants for food or feed. In this work, we have tested new strategies for increasing Fe content in the model plant Arabidopsis. Vacuolar Fe transport genes of the VTL family (VIT1-like) were over-expressed in combination with the newly discovered Fe regulatory protein IMA1 (IronMan1) or the Fe-binding peptide NAS3. Over-expression of each of the five VTL genes (VTL1 – 5) led to an increased Fe content by 2- to 3-fold in Arabidopsis seeds. IMA1 was greatly induced under Fe deficiency. Over-expression of IMA1 resulted in an Fe deficiency response also in Fe-sufficient plants. Fe content was an increase by 3-fold in seed, leaves, roots, and seedlings of Arabidopsis. The expression of Fe uptake and homeostasis genes was greatly induced in over-expressing plants independent of the Fe supply compared to the wild type. Analyses of NAS3 OE plants showed that Fe content in seeds was increased approximately 2-fold compared to WT. In conclusion, we have demonstrated that single over-expression of VTL1, 2, 3, 4 or 5, IMA1 or NAS3 correlated with increased seed Fe. Expression of Fe uptake and homeostasis genes confirmed the increased Fe content in these over-expressing plants. Double over-expression of the VTL genes in combination with IMA1 or NAS3 resulted in no further increase in Fe likely caused by the regulation of the VTL genes by IMA1 expression and the lack of increased nicotianamine in the case of VTL5/NAS3 over-expressing plants. Future research should be dedicated to extending these findings to crop plants.

Fest verwurzelte Pflanzen können nur über ihre Nachkommen wandern, z.B. in Form von Samen, und nutzen als Vehikel den Wind, das Wasser oder Tiere, oder sie breiten sich nach dem "do-it-yourself-Verfahren" aus. Deshalb findet man in der Pflanzenwelt vielfältige, oft von hoher technischer Raffinesse geprägte Anpassungen von Samen, Früchten und Fruchtständen an ihre transportierenden Medien. Bei enterohämorrhagischen Escherichia coli (EHEC) handelt es sich um humanpathogene Bakterien, welche beim Menschen schwerwiegende gastrointestinale Erkrankungen auslösen können. Der Gastrointestinaltrakt von Rindern wird als Hauptreservoir von EHEC angesehen und die primäre Infektionsquelle des Menschen stellt kontaminiertes, rohes Fleisch dar. Jedoch kam es in den letzten Jahrzehnten vermehrt zu EHEC-Infektionen, welche mit dem Verzehr von rohem Gemüse und Salatpflanzen assoziiert wurden. Heute ist es anerkannt, dass diese Infektionen dadurch begünstigt werden, dass EHEC-Bakterien in der Lage sind, Pflanzen als Sekundärwirte zu nutzen und damit eine Übertragung auf den Menschen erleichtern. Um diese Interaktion zwischen Pathogen und Pflanze besser zu verstehen, sind grundlegende Kenntnisse in der molekularen Anpassung an pflanzliche Bestandteile nötig. Im Rahmen dieser Arbeit wurde die Anpassung verschiedener EHEC Stämme an pflanzliche Bestandteile untersucht, dazu wurden O157:H7 Stamm Sakai, O104:H4 Stamm C227-11phicu und O157:H- Stamm 3072/96 als Prototypen ausgewählt. Zunächst konnte in Wachstumsexperimenten in einem artifiziellen Salatmedium gezeigt werden, dass Inhaltsstoffe des Salatmediums für ein erfolgreiches Wachstum aller drei Stämme ausreichend waren. In der RNA-Sequenzierung wurde die differentielle Genexpression der drei Stämme nach Wachstum in Salatmedium gegenüber dem Wachstum in M9-Minimalmedium bestimmt. Um Gene zwischen den Stämmen anhand einer einheitlichen Genbezeichnung miteinander zu vergleichen, wurde die differentielle Genexpression mit der Grundlage eines geteilten Genoms der drei pathogenen Stämme inklusive Referenzstamm Escherichia coli Stamm K-12 Substamm MG1655 durchgeführt. Analog zum erfolgreichen Wachstum in Anwesenheit pflanzlicher Bestandteile zeichnete sich eine erhöhte Transkription von zahlreichen Genen des Kohlenhydrat- und Peptidmetabolismus in allen drei Stämmen ab. Besonders die Gene des Lactose- (lacZ), Ribose- (rbsAC) und Xylosemetabolismus (xylF) waren in allen Stämmen deutlich hochreguliert. Die deutlichsten transkriptionellen Unterschiede zwischen den Stämmen zeigten sich in der Regulation von Motilitäts- und Chemotaxis-Genen. O104:H4 Stamm C227-11phicu zeigte in Anwesenheit pflanzlicher Bestandteile eine starke Expression aller Gene der drei Flagellenklassen (Klasse I, II und III). Dazu gehören die Gene, welche für den Aufbau der Flagellengrundstruktur (fli, flg), der Ausbildung des Filaments (fliC) sowie des Chemotaxis-Systems (che, tar, tap) verantwortlich sind. Dem entgegen konnte bei O157:H7 Stamm Sakai ausschließlich eine erhöhte Expression der Klasse I und II Flagellengene beobachtet werden. In Übereinstimmung mit den Transkriptomdaten zeigten diese beiden Stämme ein gesteigertes Schwimm- und Schwärmverhalten auf Motilitätsplatten in Anwesenheit von Salatextrakt. Ausschließlich bei dem, ausgelöst durch eine Deletion im Gen flhC, unbeweglichen O157:H- Stamm 3072/96 konnte eine gesteigerte Expression von Virulenzfaktoren der LEE-Pathogenitätsinsel beobachtet werden. Dazu gehörten Gene für den Aufbau des T3SS (esc) sowie T3SS-vermittelten Effektoren (esp). Interessanterweise äußerte sich dieser Stamm als leistungsfähiger Biofilmbildner in M9-Minimalmedium. Des Weiteren konnte gezeigt werden, dass die Komplementierung des intakten flhC-Gens die Beweglichkeit von O157:H- Stamm 3072/96 wieder herstellen konnte. In diesem Zusammenhang konnte ebenfalls festgestellt werden, dass die Deletion im Gen flhC nicht der alleinige Grund für die erhöhte Biofilmbildung dieses Stammes war. Zusätzlich zur Bestimmung der Biofilmbildung wurden die Stämme auf ihre Adhärenz an HT-29 Zellen untersucht. Hierbei konnte für O157:H- Stamm 3072/96 gegenüber den motilen Stämmen eine signifikant erhöhte Adhärenz beobachtet werden, das geringste Adhärenzpotential konnte bei O157:H7 Stamm Sakai festgestellt werden. Die Ergebnisse dieser Studie liefern klare Hinweise, dass die verschiedenen EHEC-Stämme fähig sind, sich an die Nährstoffverfügbarkeiten in einem pflanzlichen Wirt anzupassen. Es kann davon ausgegangen werden, dass Flagellen und das Chemotaxis-System eine entscheidende Rolle in der Auffindung von Pflanzen und deren Erschließung spielen. Außerdem könnten Curli-Strukturen eine elementare Funktion bei der initialen Anheftung und der anschließenden Biofilmbildung auf pflanzlichem Gewebe spielen. Vermutlich sind, neben dem typischen Pflanzen-assoziierten Ausbruchstamm O157:H7 Stamm Sakai, auch weitere EHEC-Stämme in der Lage ihr genetisches Repertoire auszunutzen, um eine Antwort auf die atypische Bedingungen dieser Nische zu gewährleisten. Die in dieser Arbeit gewonnenen Ergebnisse legen nahe, dass die Stämme aufgrund ihrer unterschiedlichen genetischen Ausstattung, neben zahlreichen übereinstimmenden Mechanismen, zusätzlich auch stammspezifische Strategien bei einer Interaktion mit Pflanzen als Sekundärwirte zeigen. Enterohemorrhagic Escherichia coli (EHEC) are human pathogens which are able to cause severe gastrointestinal diseases in humans. The gastrointestinal tract of cattle is considered as the main reservoir for EHEC and contaminated raw meat represents the primary source of infection. Yet there have been increasing reports over the last few decades of EHEC infections that were linked to the consumption of raw vegetables. Today it is generally accepted that EHEC bacteria are able to use plants as their secondary hosts, thus favouring the transmission to humans. To improve the understanding of this pathogen-plant interaction fundamental knowledge about the pathogens’ molecular adaptions towards plant material is urgently required. In the cope of this study the adaption of different EHEC strains towards components of the plant was examined. Therefore O157:H7 strain Sakai, O104:H4 strain C227-11phicu and O157:H strain 3072/96 were chosen as surrogates. In growth experiments performed with an artificial lettuce medium it could be shown that components of the lettuce were sufficient for the proliferation of the three strains. RNA-sequencing was performed to study the differential gene expression of the three strains after the growth in lettuce medium compared to the growth in M9 minimal medium. In order to compare genes according to standardized gene denotations, the differential gene expression analysis was performed on the basis of a shared genome including the genomes of the three pathogenic strains as well as the genome of Escherichia coli strain K-12 substrain MG1655. Analogous to the successful growth in presence of components of the plant an upregulation of genes involved in carbohydrate and peptide metabolism throughout all three strains was observed. Especially genes involved in the catabolism of lactose (lacZ), ribose (rbsAC) and xylose (xylF) were found to be uniformly upregulated. The greatest differences among the strains accounted for the regulation of motility and chemotaxis genes. O104:H4 strain C227-11phicu showed a strong upregulation of all three classes of the flagellar hierarchy (class I, II and III) in presence of plant derived compounds. These included genes involved in the establishment of the basal body hook structure (fli, flg), the synthesis of the flagellar filament (fliC), and the chemotaxis-system (che, tap, tar). In contrast, O157:H7 strain Sakai only featured upregulation of class I and class II genes. According to the transcriptional data both of these strains also showed increased swimming and swarming behaviour on motility plates in presence of lettuce extract. Solely O157:H- strain 3072/96, which is non-motile due to a deletion in the flhC gene, showed an upregulation of virulence factors encoded on the LEE pathogenicity island, including genes involved in the establishment of the T3SS (esc) and T3SS secreted effectors (esp). Interestingly, it was shown for O157:H- strain 3072/96 to have a powerful capacity to form biofilms in M9 minimal medium. Furthermore it was proven that the complementation of an intact flhC gene restored motility in O157:H- strain 3072/96. In this regard it could be shown that the deletion in flhC was not the mere reason for the augmented biofilm formation capacity. In addition to the biofilm formation, the strains’ potential to adhere to HT-29 cells was examined. Here a significantly increased adherence potential for O157:H- strain 3072/96 with respect to the motile strains could be observed, the lowest adherence potential was determined for O157:H7 strain Sakai. The results presented in this study clearly indicate that the different EHEC strains are capable to adapt towards the nutrient availability provided by their plantal host. It can be assumed that flagella and the chemotaxis system play a fundamental role in the finding and exploitation of the plant. Furthermore curli structures might play a crucial role in the initial adherence and the subsequent establishment of a biofilm on plant tissues. Presumably, besides the typical plant associated outbreak strain O157:H7 strain Sakai, there are further strains capable of utilizing their genetic repertoire in order to adapt towards the atypical environmental conditions within this niche. The findings of this study suggest that the strains, besides sharing multiple coinciding mechanisms, are able to adapt in a strain specific manner and use different strategies in coping with plants as their secondary hosts.

Die Rote Liste der Pflanzengesellschaften ist ein Bindeglied zwischen den Roten Listen der Arten und der Biotoptypen. Sie dient sowohl der Bioindikation als auch der Fachplanung des Naturschutzes. Mit Hilfe der Roten Liste können Arten- und Biotopschutzprogramme ergänzt und Aktivitäten zum Schutz und zur Entwicklung bedrohter Lebensräume angeregt werden. Dabei sind sie gerade in unseren dicht bebauten Siedlungen für viele Tiere, vor allem für Insekten, von besonderer Bedeutung. Drimia indica (Roxb.) Jessop (Asparagaceae) is a reputed Ayurvedic medicine for a number of therapeutic benefits, including for cardiac diseases, indigestion, asthma, dropsy, rheumatism, leprosy, and skin ailments. The present work aimed to critically and extensively review its traditional uses, phytochemistry, pharmacology, toxicology, and taxonomy together with the mechanisms of action of selected extracts of D. indica. A systematic literature survey from scientific databases such as PubMed, Scopus, and Web of Science as well as from some textbooks and classical texts was conducted. The plant, mainly its bulb, contains various bioactive constituents, such as alkylresorcinols, bufadienolides, phytosterols, and flavonoids. Various scientific studies have proven that the plant has anthelmintic, anticancer, antidiabetic, antimicrobial, antioxidant, and wound healing activities. The present work concludes that D. indica has the potential to treat various diseases, mainly microbial infections. This review also suggests that bufadienolides, flavonoids, and steroids might be responsible for its bioactive potential.
