Apolipoprotein L1 (APOL1) nephropathy results from several podocyte dysfunctions involving morphological and motility changes, mitochondrial perturbations, inflammatory stress, and alterations in cation channel activity. I propose that this phenotype results from increased hydrophobicity of the APOL1 risk variants, which induces two distinct types of podocyte dysfunctions. On one hand, increased hydrophobic interactions with APOL3 cause intracellular variant isoforms to impair both APOL3 control of Golgi PI(4)P kinase-B (PI4KB) activity and APOL3 control of mitochondrial membrane fusion, triggering actomyosin reorganisation together with mitophagy and apoptosis inhibition (hit 1). On the other hand, increased hydrophobic interactions with the podocyte plasma membrane may cause the extracellular variant isoforms to activate toxic Ca2+ influx and K+ efflux by the TRPC6 and BK channels, respectively (hit 2), presumably due to APOL1-mediated cholesterol clustering in microdomains. I propose that hit 2 depends on low HDL-C/high extracellular APOL1 ratio, such as occurs in cell culture in vitro, or during type I-interferon (IFN-I)-mediated inflammation.

7. Establishing large mammal population trends from heterogeneous count data

Abstract Monitoring population trends is pivotal to effective wildlife conservation and management. However, wildlife managers often face many challenges when analyzing time series of census data due to heterogeneities in sampling methodology, strategy, or frequency. We present a three‐step method for modeling trends from time series of count data obtained through multiple census methods (aerial or ground census and expert estimates). First, we design a heuristic for constructing credible intervals for all types of animal counts including those which come with no precision measure. Then, we define conversion factors for rendering aerial and ground counts comparable and provide values for broad classes of animals from an extant series of parallel aerial and ground censuses. Lastly, we construct a Bayesian model that takes the reconciled counts as input and estimates the relative growth rates between successive dates while accounting for their precisions. Importantly, we bound the rate of increase to account for the demographic potential of a species. We propose a flow chart for constructing credible intervals for various types of animal counts. We provide estimates of conversion factors for 5 broad classes of species. We describe the Bayesian model for calculating trends, annual rates of population increase, and the associated credible intervals. We develop a bespoke R CRAN package, popbayes, for implementing all the calculations that take the raw counts as input. It produces consistent and reliable estimates of population trends and annual rates of increase. Several examples from real populations of large African mammals illustrate the different features of our method. The approach is well‐suited for analyzing population trends for heterogeneous time series and allows a principled use of all the available historical census data. The method is general and flexible and applicable to various other animal species besides African large mammals. It can readily be adapted to test predictions of various hypotheses about drivers of rates of population increase.

12. Towards the Development of a Cream with Antiviral Properties Targeting Both the Influenza A Virus and SARS-CoV-2

Objective: Many severe acute respiratory infections are caused by viral pathogens, and viruses are responsible for a large number of deaths worldwide. Among the most common respiratory viruses are the influenza A virus (IAV) and, more recently, the SARS-CoV-2 that emerged in 2019 and caused the most significant human pandemic of the beginning of the 21st century. Both IAV and SARS-CoV-2 share clinical features and a common transmission route through the emission of viral particles via aerosols and droplets. These penetrate the host after entry from the nose and mouth or an indirect mode of transmission via contact contamination of different media. These facts prompted us to investigate the possibility of designing a soft cream with a virucidal activity targeted against IAV and SARS-CoV-2. Methods: We first investigated the action of chemical compounds known to have antiviral properties such as cyclodextrin, or algae extracts containing sulfated polysaccharides, on cultured cells infected with lentiviral viral particles pseudotyped (VP) with either proteins HA (hemagglutinin) and NA (neuraminidase) from IAV or the G protein from the vesicular stomatitis virus or spike-bearing particles in order to select molecules with antiviral activities in human embryonic kidney (HEK293T) cells. Results: Our results show that some cyclodextrin-containing creams can significantly reduce the stability of HANA- and spike-bearing particles when they are applied prior to challenge with a viral inoculum on skin. Conclusions: We observed some specificities of these creams towards either IAV or SARS-CoV-2, indicating that the neutralization of viral activity is correlated with the mechanism of receptor interaction and entry of these two pathogens.

Summary: Apolipoproteins L1 and L3 (APOLs) are associated at the Golgi with the membrane fission factors phosphatidylinositol 4-kinase-IIIB (PI4KB) and non-muscular myosin 2A. Either APOL1 C-terminal truncation (APOL1Δ) or APOL3 deletion (APOL3-KO [knockout]) reduces PI4KB activity and triggers actomyosin reorganization. We report that APOL3, but not APOL1, controls PI4KB activity through interaction with PI4KB and neuronal calcium sensor-1 or calneuron-1. Both APOLs are present in Golgi-derived autophagy-related protein 9A vesicles, which are involved in PI4KB trafficking. Like APOL3-KO, APOL1Δ induces PI4KB dissociation from APOL3, linked to reduction of mitophagy flux and production of mitochondrial reactive oxygen species. APOL1 and APOL3, respectively, can interact with the mitophagy receptor prohibitin-2 and the mitophagosome membrane fusion factor vesicle-associated membrane protein-8 (VAMP8). While APOL1 conditions PI4KB and APOL3 involvement in mitochondrion fission and mitophagy, APOL3-VAMP8 interaction promotes fusion between mitophagosomal and endolysosomal membranes. We propose that APOL3 controls mitochondrial membrane dynamics through interactions with the fission factor PI4KB and the fusion factor VAMP8.

Background:Aframomum sp. is a genus of plants in the Zingiberaceae family. It includes several species, some of which are used in cosmetics for their various properties, making them useful in skincare products, particularly for anti-aging, moisturizing, and brightening the skin. However, to date, there is no experimental evidence on its natural extracts obtained or modified using microorganisms (bio-fermentation) as an anti-aging agent.Objective: The present study aimed to evaluate the antiaging effect of a Bio-fermented Aframomum angustifolium (BAA) extract on 3D bioprinted skin equivalent.Methods: The consortium of microorganisms contained Komagataeibacter, Gluconobacter, Acetobacter, Saccharomyces, Torulaspora, Brettanomyces, Hanseniaspora, Leuconostoc, Lactobacillus, Schizosaccharomyces. It was developed on a media containing water, sugar, and infused black tea leaves. The seeds of Aframomum angustifolium previously grounded were mixed with the culture medium, and the ferments in growth; this fermentation step lasted 10 days. Then, the medium was collected and filtered (0.22 µm) to obtain the BAA extract. To enhance our comprehension of the impact of BAA extract on skin aging, we developed skin equivalents using bio-printing methods with the presence or absence of keratinocyte stem cells (KSC). These skin equivalents were derived from keratinocytes obtained from both a middle-aged donor, with and without KSC. Moreover, we examined the effects of treating the KSC-depleted skin equivalents with Bio-fermented Aframomum angustifolium (BAA) extract for 5 days. Skin equivalents containing KSC-depleted keratinocytes exhibited histological characteristics typical of aged skin and were compared to skin equivalents derived from young donors.Results: The BAA extract contained specific organic acids such as lactic, gluconic, succinic acid and polyphenols. KSC-depleted skin equivalents that were treated with BAA extract exhibited higher specular reflection, indicating better hydration of the stratum corneum, higher mitotic activity in the epidermis basal layer, improved dermal-epidermal connectivity, and increased rigidity of the dermal-epidermal junction compared to non-treated KSC-depleted equivalents. BAA extract treatments also resulted in changes at the dermis level, with an increase in total collagen and a decrease in global laxity, suggesting that this extract could help maintain youthful-looking skin.Conclusion: In summary, our findings indicated that BAA extract treatments have pleiotropic beneficial effects on skin equivalents and that the bio-fermentation provides new biological activities to this plant.

The need to reconcile food production, the safeguarding of nature, and the protection of public health is imperative in a world of continuing global change, particularly in the context of risks of emerging zoonotic disease (EZD). In this paper, we explored potential land use strategies to reduce EZD risks using a landscape approach. We focused on strategies for cases where the dynamics of pathogen transmission among species were poorly known and the ideas of “land-use induced spillover” and “landscape immunity” could be used very broadly. We first modeled three different land-use change scenarios in a region of transition between the Cerrado and the Atlantic Forest biodiversity hotspots. The land-use strategies used to build our scenarios reflected different proportions of native vegetation cover, as a proxy of habitat availability. We then evaluated the effects of the proportion of native vegetation cover on the occupancy probability of a group of mammal species and analyzed how the different land-use scenarios might affect the distribution of species in the landscape and thus the risk of EZD. We demonstrate that these approaches can help identify potential future EZD risks, and can thus be used as decision-making tools by stakeholders, with direct implications for improving both environmental and socio-economic outcomes.

The clinical severity of multiple sclerosis (MS), an autoimmune disorder of the central nervous system, is thought to be determined by environmental and genetic factors that have not yet been identified. In a recent genome-wide association study (GWAS), a single nucleotide polymorphism (SNP), rs10191329, has been associated with MS severity in two large independent cohorts of patients. Different approaches were followed by the authors to prioritize the genes that are transcriptionally regulated by such an SNP. It was concluded that the identified SNP regulates a group of proximal genes involved in brain resilience and cognitive abilities rather than immunity. Here, by conducting an alternative strategy for gene prioritization, we reached the opposite conclusion. According to our re-analysis, the main target of rs10191329 is N-Acetylglucosamine Kinase (NAGK), a metabolic gene recently shown to exert major immune functions via the regulation of the nucleotide-binding oligomerization domain-containing protein 2 (NOD2) pathway. To gain more insights into the immunometabolic functions of NAGK, we analyzed the currently known list of NAGK protein partners. We observed that NAGK integrates a dense network of human proteins that are involved in glucose metabolism and are highly expressed by classical monocytes. Our findings hold potentially major implications for the understanding of MS pathophysiology.

Abstract While harboring the bulk of the planet's biodiversity, tropical ecosystems have experienced intense land conversion for agriculture. Studies examining the impacts of land‐use change on tropical biodiversity have primarily focused on forest cover loss but have overlooked the ecological potential of habitats surrounding forest fragments to modulate biodiversity loss. We examined whether small‐mammal communities changed with the land cover surrounding forest fragments, and how functional traits affected responses to land cover. Small mammals were sampled in the Brazilian Cerrado using live‐trap transects. Abstract Reconstructed human epidermis equivalents (RHE) have been developed as a clinical skin substitute and as the replacement for animal testing in both research and industry. KiPS, or keratinocytes derived from induced pluripotent stem cells (iPSCs) are frequently used to generate RHE. In this study, we focus on the mitochondrial performance of the KiPS derived from iPSCs obtained from two donors. We found that the KiPS derived from the older donor have more defective mitochondria. Treatment of these KiPS with a plant extract enriched in compounds known to protect mitochondria improved mitochondrial respiration and rendered them fully competent to derive high-quality RHE. Overall, our results suggest that improving mitochondrial function in KiPS is one of the key aspects to obtain a functional RHE and that our plant extracts can improve in this process.

IntroductionThe impact of multi-domain preventive interventions on older adults, in particular on those with higher risk to develop Alzheimer's disease (AD), could be beneficial, as it may delay cognitive decline. However, the precise mechanism of such positive impact is not fully understood and may involve brain reserve and adaptability of brain functional connectivity (FC).MethodsTo determine the effect of multidomain interventions (involving physical activity, cognitive training, nutritional counseling alone or in combination with omega-3 fatty acid supplementation and vs. a placebo) on the brain, longitudinal FC changes were assessed after 36 months of intervention on 100 older adults (above 70 year-old) with subjective cognitive complaints.ResultsNo global change in FC was detected after uni or multidomain preventive interventions. However, an effect of omega-3 fatty acid supplementation dependent on cognitive decline status was underlined for frontoparietal, salience, visual and sensorimotor networks FC. These findings were independent of the cortical thickness and vascular burden.DiscussionThese results emphasize the importance of patient stratification, based on risk factors, for preventive interventions.

Monitoring population trends is pivotal to effective wildlife conservation and management. However, wildlife managers often face many challenges when analyzing time series of census data due to heterogeneities in sampling methodology, strategy, or frequency. We present a three‐step method for modeling trends from time series of count data obtained through multiple census methods (aerial or ground census and expert estimates). First, we design a heuristic for constructing credible intervals for all types of animal counts including those which come with no precision measure. Then, we define conversion factors for rendering aerial and ground counts comparable and provide values for broad classes of animals from an extant series of parallel aerial and ground censuses. Lastly, we construct a Bayesian model that takes the reconciled counts as input and estimates the relative growth rates between successive dates while accounting for their precisions. Importantly, we bound the rate of increase to account for the demographic potential of a species. We propose a flow chart for constructing credible intervals for various types of animal counts. We provide estimates of conversion factors for 5 broad classes of species. We describe the Bayesian model for calculating trends, annual rates of population increase, and the associated credible intervals. We develop a bespoke R CRAN package, popbayes, for implementing all the calculations that take the raw counts as input. It produces consistent and reliable estimates of population trends and annual rates of increase. Several examples from real populations of large African mammals illustrate the different features of our method. The approach is well‐suited for analyzing population trends for heterogeneous time series and allows a principled use of all the available historical census data. The method is general and flexible and applicable to various other animal species besides African large mammals. In adult testis, the cell mobility is essential for spermatogonia differentiation and is suspected to regulate spermatogonial stem cell fate. Netrin-1 controls cell migration and/or survival according to the cellular context. Its involvement in some self-renewing lineages raises the possibility that Netrin-1 could have a role in spermatogenesis.We show that in addition to Sertoli cells, a fraction of murine undifferentiated spermatogonia express the Netrin-1 receptor UNC5c and that UNC5c contributes to spermatogonia differentiation. Receptor loss in Unc5crcm males leads to the concomitant accumulation of transit-amplifying progenitors and short syncytia of spermatogonia. Without altering cell death rates, the consequences of Unc5c loss worsen with age: the increase in quiescent undifferentiated progenitors associated with a higher spermatogonial stem cell enriched subset leads to the spermatocyte I decline. We demonstrate in vitro that Netrin-1 promotes a guidance effect as it repulses both undifferentiated and differentiating spermatogonia.Finally, we propose that UNC5c triggers undifferentiated spermatogonia adhesion/ migration and that the repulsive activity of Netrin-1 receptors could regulate spermatogonia differentiation, and maintain germ cell homeostasis.

In a substantial share of patients suffering from multiple sclerosis (MS), neurological functions slowly deteriorate despite a lack of radiological activity. Such a silent progression, observed in either relapsing-remitting or progressive forms of MS, is driven by mechanisms that appear to be independent from plaque activity. In this context, we previously reported that, in the spinal cord of MS patients, periplaques cover large surfaces of partial demyelination characterized notably by a transforming growth factor beta (TGF-beta) molecular signature and a decreased expression of the oligodendrocyte gene NDRG1 (N-Myc downstream regulated 1). In the present work, we re-assessed a previously published RNA expression dataset in which brain periplaques were originally used as internal controls. When comparing the mRNA profiles obtained from brain periplaques with those derived from control normal white matter samples, we found that, irrespective of plaque activity, brain periplaques exhibited a TGF-beta molecular signature, an increased expression of TGFB2 (transforming growth factor beta 2) and a decreased expression of the oligodendrocyte genes NDRG1 (N-Myc downstream regulated 1) and MAG (myelin-associated glycoprotein). From these data obtained at the mRNA level, a survey of the human proteome allowed predicting a protein–protein interaction network linking TGFB2 to the down-regulation of both NDRG1 and MAG in brain periplaques. To further elucidate the role of NDRG1 in periplaque-associated partial demyelination, we then extracted the interaction network linking NDRG1 to proteins detected in human central myelin sheaths. We observed that such a network was highly significantly enriched in RNA-binding proteins that notably included several HNRNPs (heterogeneous nuclear ribonucleoproteins) involved in the post-transcriptional regulation of MAG. We conclude that both brain and spinal cord periplaques host a chronic process of tissue remodeling, during which oligodendrocyte myelinating functions are altered. Our findings further suggest that TGFB2 may fuel such a process. Overall, the present work provides additional evidence that periplaque-associated partial demyelination may drive the silent progression observed in a subset of MS patients.

Background: Aframomum sp. is a genus of plants in the Zingiberaceae family. It includes several species, some of which are used in cosmetics for their various properties, making them useful in skincare products, particularly for anti-aging, moisturizing, and brightening the skin. However, to date, there is no experimental evidence on its natural extracts obtained or modified using microorganisms (biofermentation) as an anti-aging agent. Objective: The present study aimed to evaluate the antiaging effect of a Biofermented Aframomum angustifolium (BAA) extract on 3D bioprinted skin equivalent. Methods: The consortium of microorganisms contained Komagataeibacter, Gluconobacter, Acetobacter, Saccharomyces, Torulaspora, Brettanomyces, Hanseniaspora, Leuconostoc, Lactobacillus, Schizosaccharomyces. It was developed on a media containing water, sugar, and infused black tea leaves. The seeds of Aframomum angustifolium previously grounded were mixed with the culture medium, and the ferments in growth; this fermentation step lasted 10 days. Then, the medium was collected and filtered (0.22 µm) to obtain the BAA extract. To enhance our comprehension of the impact of BAA extract on skin aging, we developed skin equivalents using bio-printing methods with the presence or absence of keratinocyte stem cells (KSC). These skin equivalents were derived from keratinocytes obtained from both a middle-aged donor, with and without KSC. Moreover, we examined the effects of treating the KSC-depleted skin equivalents with Bio-fermented Aframomum angustifolium (BAA) extract for 5 days. Skin equivalents containing KSC-depleted keratinocytes exhibited histological characteristics typical of aged skin and were compared to skin equivalents derived from young donors. Results: The BAA extract contained specific organic acids such as lactic, gluconic, succinic acid and polyphenols. KSC-depleted skin equivalents that were treated with BAA extract exhibited higher specular reflection, indicating better hydration of the stratum corneum, higher mitotic activity in the epidermis basal layer, improved dermal-epidermal connectivity, and increased rigidity of the dermalepidermal junction compared to non-treated KSC-depleted equivalents. BAA extract treatments also resulted in changes at the dermis level, with an increase in total collagen and a decrease in global laxity, suggesting that this extract could help maintain youthful-looking skin. Conclusion: In summary, our findings indicated that BAA extract treatments have pleiotropic beneficial effects on skin equivalents and that the bio-fermentation provides new biological activities to this plant. The necessity to create national to global-scale biodiversity monitoring systems as part of assessing progress toward biodiversity agendas presents a challenge for signatory countries. This is a brief review of ongoing Brazilian national initiatives that would allow the construction of a general biomonitoring network scheme in protected areas; with additional focus on linking independent monitoring schemes. We discuss some key aspects needed to include monitoring schemes under a single framework that will lead to better evaluation of pressure–state–response indicators for managing biodiversity at several scales; and we point out the potential of embracing citizen science and participatory monitoring to quantify some aspects within those schemes.

50. Aquatic invasive alien rodents in Western France: Where do we stand today after decades of control?

Two aquatic invasive alien rodents, the coypu (Myocastor coypus) and muskrat (Ondatra zibethicus), have taken over a significant amount of wetlands in France. Pays de la Loire is an administrative region of about 32 000 km2 in the Western France with 6.3% of its area in wetlands (excluding the Loire River). Populations of coypus and muskrats are established and a permanent control programme has been set to reduce their impacts. The control plan is based on few professional trappers and many volunteers which makes this programme unique compared to other programme relying on professionals only. The aim of this study is to analyse the temporal and spatial dynamics of coypu and muskrat captures during the last 10 years to evaluate their effectiveness. The number of rodents removed per year increased by 50% in 10 years and reached about 288 000 individuals in 2016 with about 80% of them being coypus. During the same time length, the number of trappers involved in the programme also increased by 50% to reach 3 000 people in 2016. Although the raise of coypus and muskrats trapped can possibly be explained by an increase of the number of trappers, the number of coypus removed per trapper per year increased by 22%. Despite the outstanding number of individuals removed per year, our results suggest that the programme does not limit the population dynamics of coypus. Finally, since 2017, the number of data gathered from municipalities decreased, as did the total number of individuals trapped. Indeed, although rewards are crucial to recruit new volunteers, subsidies from local and regional authorities are declining. Decision makers and financers should be encouraged to fund this programme from the perspectives of the direct or indirect costs related to the presence of aquatic invasive alien rodents in wetlands.
