Nicht nur die mögliche Verwendung von Variolavirus als bioterroristischer Kampfstoff sondern auch das zoonotische Potential pathogener Pockenviren wird als eine erhebliche Gefahr für das öffentliche Gesundheitswesen diskutiert. Zurzeit liegt in der Bevölkerung kein ausreichender Impfschutz vor. Mit Ausnahme der Expositionsprophylaxe steht die Vakzination als einzige Prävention zur Verfügung, von der bekannt ist, dass sie mit erheblichen Impfkomplikationen assoziiert ist. Therapeutische Ansätze sind deshalb von großem Interesse. In dieser Arbeit wurden HEp-2-Zellen mit den Orthopockenviren (OPV) VACV-LE, VACV-MVA-BN, CMLV-CP-19 und Calpoxvirus sowie die Zelllinien 293, A-549, Huh-7 und HSB-2 mit Calpoxvirus infiziert und der Replikationszyklus analysiert. Die Genexpression von Faktoren der Transkription (D7R), der DNA-Replikation (K4L) oder für Strukturproteine (D8L, A56R, A27L) wurde unabhängig vom untersuchten Virus und der Zelllinie gleich reguliert, was auf eine wichtige Rolle während der Virusreplikation hinweist. Ferner wurde ein siRNA-System zur Hemmung der OPV-Gene D7R, K4L, D8L, A56R und A27L etabliert. Die Transkription von D7R, K4L, D8L und A27L wurde in infizierten Zellen effektiv durch spezifisch eingesetzte shRNAs inhibiert und dadurch die Virusreplikation von Calpoxvirus im Vergleich zu den Kontrollzellen 24 und 48 Stunden nach der Infektion gehemmt. Die Inhibition der A27L-Genexpression wurde zusätzlich im Western Blot für Calpoxvirus- und VACV-LE-infizierte 293-Zellen nachgewiesen. A56R wurde weder für die Replikation von Calpoxvirus noch von VACV-LE in der Zellkultur benötigt. Da die Gene D7R, K4L, D8L und A27L essentiell für die Replikation von OPV in der Zellkultur waren, könnten sie potentielle Zielstrukturen für die Entwicklung von antiviralen Substanzen für die Behandlung einer OPV-Infektion darstellen., Not only the potential use of smallpox in a bioterrorist attack but also the zoonotic potential of other poxviruses infecting animals are discussed to be a public health threat. At present, there is no sufficient protection in the human population provided by vaccination. Prophylactic vaccination was effective but has been associated with serious adverse effects. No proven drug treatment is available. Therefore, therapies and new treatment strategies are greatly needed. In this study HEp-2 cells were infected with the orthopoxviruses (OPV) VACV-LE, VACV-MVA-BN, CMLV-CP-19 and Calpoxvirus. In addition, the cell lines 293, A-549, Huh-7 and HSB-2 were infected with Calpoxvirus alone to determine the OPV replication cycle. Irrespective of the analyzed virus and cell line gene expression was regulated similarly for factors involved in transcription (D7R), DNA replication (K4L) or for structural proteins (D8R, A56R, A27L), indicating an important role in virus replication. Furthermore, a siRNA-system for the inhibition of the OPV genes D7R, K4L, D8L, A56R and A27L was established. It could be shown, that specific shRNAs effectively inhibited transcription of D7R, K4L, D8L and A27L in infected cells and repressed Calpoxvirus replication 24 and 48 hours post infection in comparison to control cells. In addition, the inhibition of A27L gene expression was shown in Western Blot analyses for Calpoxvirus- and VACV-LE-infected 293 cells. A56R is not required for virus replication of Calpoxvirus and VACV-LE in cell culture. Since D7R, K4L, D8L and A27L seem to be essential for OPV replication in cell culture, these OPV genes might serve as potential targets for the development of antiviral substances for the treatment of OPV infections.

In dieser Arbeit wurden zwei serologische Teste zur Unterscheidung inzidenter und prävalenter Proben etabliert und validiert, welche auf der Reifung des Immunsystems in der frühen HIV-Infektion basieren. Im BED-ELISA werden anhand von anti-HIV-gp41-spezifischen IgG-Antikörperspiegeln die Proben klassifiziert. Die Aviditäts-Methode (AI) unterscheidet die Bindungsfähigkeit der Antikörper an spezifische Antigene. Das Probenpanel wurde in einem weiteren Inzidenztest, dem IDE-V3-ELISA (Kooperation F. Barin), gemessen welcher den Anstieg der Antikörperreaktivität gegen zwei verschiedene immundominante Epitope nutzt. Wirtsspezifische Marker und virale Eigenschaften wurden untersucht, um Merkmale zu identifizieren, welche die Sensitivität und Spezifität der Teste verbessern könnten. Mit dem BED-ELISA wurde eine Pilotstudie unter Berliner HIV-Patienten durchgeführt. Zur Vereinfachung des Probentransports in Studien wurde der Einfluss einer Filtertrocknung der Plasmaproben auf die Infektiosität von HIV, Stabilität der HIV-RNA und die Antikörperreaktivität im BED-ELISA untersucht. In den Testen wurden inzidente Plasmaproben mit folgenden Sensitivitäten und Spezifitäten richtig klassifiziert: 80% und 86% (BED-ELISA); 74% und 82% (AI); 73% und 84% (IDE-V3). Von allen untersuchten wirtsspezifischen Faktoren korrelierte nur der Gehalt an Antikörpern der IgG3-Subklasse mit der Fehlklassifikation der Proben. Für die Pilotstudie wurden zwischen Feb. 2005 und Nov. 2007 von 132 erstmalig HIV-1 positiv diagnostizierten Patienten Proben genommen und im BED-ELISA analysiert (51% Anteil inzidente Infektionen). Die Antikörperreaktivitäten blieben nach der Filtertrocknung erhalten, so das auf der Grundlage der Ergebnisse eine deutschlandweite Inzidenzstudie mit Filter-getrockneten Plasmaproben geplant wurde, die seit Januar 2008 im Auftrag des BMG zur Verbesserung der Datenlage zur HIV-Inzidenz in Deutschland durchgeführt wird., In this PhD thesis two methods that can differentiate between incident and prevalent infections were established and validated. Both tests are based on the maturation of the immune system during early HIV-infection. The BED-ELISA uses anti-HIV-gp41 specific IgG-antibody levels for differentiation. The avidity method (AI) is based on the binding-capacity of the antibodies to specific antigens in the presence of a chaotropic agent. The sample panel was also evaluated using an additional incidence ELISA, the IDE-V3-ELISA (cooperation with F. Barin). This test is also based on the antibody''s reactivity to two different immune dominant epitopes, allowing incident and prevalent infections to be differentiated. Host specific factors and viral determinants were analysed to provide information that could lead to improvements in the sensitivity and specificity of the incidence tests. With the BED-ELISA a pilot study with HIV-infected patients in Berlin was carried out. The inactivation of HIV-1 after filter-drying of samples, the stability of viral RNA and reactivity of antibodies in the BED-ELISA were analysed to simplify the transport of samples in future studies. Incident plasma samples were identified correctly with following sensitivities and specificity’s: 80% and 86% (BED-ELISA); 74% and 82% (AI); 73% and 84% (IDE-V3). Of all host factors analysed, only the titre of IgG3-antibodies correlated with the incorrect classification of samples. In the pilot study samples from 132 newly diagnosed HIV-positive patients were obtained between February 2005 and November 2007 and analysed in the BED-ELISA (51% proportion of incident infections). It could be shown that filter-drying of plasma samples rendered HIV non-infectious but did not influence antibody reactivity. Based on these results, a German HIV incidence study using filter-dried plasma samples, designed to improve knowledge of HIV-incidence in Germany, was sponsored by the BMG and has been ongoing since January 2008.

Die vorsätzliche Freisetzung von Variola Virus (VARV) und schwere Erkrankungen des Menschen durch zoonotische Affen- (MPXV) und Kuh- (CPXV) pocken Viren stellen nach wie vor eine Bedrohung für die Bevölkerung dar. Klassische Pockenimpfstoffe bergen die Gefahr einer schweren Erkrankung. Deshalb ist die Entwicklung neuer Impfstoffe und Therapeutika von entscheidender Bedeutung. Deren Wirksamkeit und Sicherheit muss zunächst in verschiedenen Tiermodellen bewiesen werden. Existierende Makakken-Primatenmodelle leiden unter sehr artifiziellen Bedingungen der letalen Krankheitsinduktion durch VARV oder MPXV. Aus diesem Grund wurde das Calpox Virus/Krallenaffen-modell etabliert, welches auf einem CPXV aus natürlich infizierten Neuweltaffen (Marmosets) basiert. Das neue Modell hat drei wesentliche Vorteile: Die Arbeit mit Calpox Virus kann unter Sicherheitsstufe 2 durchgeführt werden und ist folglich einfacher in der Handhabung. 2. Die intranasale (i.n.) Infektion von Marmosets (Krallenaffen; Callithrix jacchus) spiegelt den natürlichen Infektionsweg von VARV wieder. Infizierte Affen entwickelten Pocken ähnliche Symptome und verstarben innerhalb von 2-3 Tagen nach Auftreten erster Symptome. Hohe Viruslasten wurden im Blut, Speichel und allen untersuchten Organen nachgewiesen. 3. Die i.n. Titration des Calpox Virus ergab eine 50 % Affen-Infektions-Dosis (MID50) von 8.3x102 pfu. Diese ist um den Faktor 10000 niedriger als in anderen Pocken-Primatenmodellen. Neun bis zehn Wochen nach einer Immunisierung mit dem Lister-Elstree Impfstoff waren alle Krallenaffen gegen eine letale Dosis des Calpox Virus (10 MID50) geschützt. Damit konnte der Nutzen des Calpox Virus/Krallenaffen-modells für die Erforschung neuer Impfstoffe gezeigt werden. Das Calpox Virus/Krallenaffen-modell überwindet wesentliche Nachteile bestehender Primatenmodelle und ist somit ein geeignetes Model für die Evaluierung von neuen Impfstoffen, Impfstrategien und antiviralen Therapien., The intentional re-introduction of Variola virus (VARV), the agents of smallpox, into the human population remains of concern today. Moreover, zoonotic infections with Cowpox (CPXV) and Monkeypox virus (MPXV) cause severe diseases in humans. Smallpox vaccines presently available can have severe adverse effects that are no longer acceptable. The efficacy and safety of new vaccines and antivirals have to be demonstrated by different animal models. The existing primate models, using VARV and MPXV, need very high viral doses that have to be applied intravenously to induce a lethal infection in macaque monkeys. To overcome these drawbacks, the main objective of this study was to develop a primate model in which a smallpox-like disease could be induced by a CPXV virus designated calpox virus which was isolated from a lethal orthopox virus (OPV) outbreak in New World monkeys (marmosets). The new non-human primate model has three major advantages: 1. Working with calpox virus is less challenging and can be done under bio-safety-level two. 2. Mimicking the natural route of VARV infection, intranasally infected marmosets (Callithrix jacchus) reproducibly developed clinical symptoms of an OPV infection and died within two to three days after onset of the first symptoms. High viral loads of calpox virus were detected in blood, saliva and all analyzed organs. 3. Intranasal titration of the virus resulted in a 50 % monkey infectious dose (MID50) of 8.3x102 pfu, a lethal infectious dose 10,000 lower than those used in any other primate model. Moreover, we showed the aptitude of the primate model for the testing of new vaccines since nine to ten weeks after immunization with Vaccinia virus Lister-Elstree marmosets were completely protected against intranasal challenge with 10 MID50 of calpox virus. As the calpox virus/marmoset model overcomes major limitations of current primate models it is suitable to evaluate new vaccines, new vaccination strategies and antiviral therapies.

Als ein Grund für die vertikale Transmission von HIV von der Mutter auf das Kind während der Schwangerschaft bzw. der Geburt wird der Transport von HIV durch die Eihaut diskutiert. Hierbei handelt es sich wahrscheinlich um einen rezeptorvermittelten Transport, der auf einer Interaktion zwischen einer Lektin-bindenden Domäne auf dem viralen Oberflächenglykoprotein gp120 und einem Rezeptor auf der epithelialen Oberfläche beruht. In der vorliegenden Arbeit konnte die in den Transport von zellfreien HIV-1 durch epitheliale Zellen beteiligte Domäne auf gp120 erstmals näher charakterisiert werden. Überlappende Oligopeptide –basierend auf der Aminosäurensequenz von gp120– wurden zur Hemmung der Transzytose von HIV-1 durch humane Amnionzellen verwendet. Vier dieser Oligopeptide inhibierten die Transzytose von HIV-1 signifikant. Ein synthetisches Peptid (Env362-420) mit einer Länge von 59 Aminosäuren, welches die Sequenz der inhibierenden Oligopeptide darstellt, reduzierte die Menge an transportierten Viren ebenfalls, unabhängig vom HIV-1 Subtyp. Im Weiteren konnte der Transport von HIV-1 durch polyklonale Antikörper in Seren HIV-Infizierter, die mit Env362-420 reagierten, und durch Seren, die durch eine Immunisierung von Kaninchen mit Env362-420 gewonnen wurden, inhibiert werden. Antikörper gegen die in den Transport involvierte Domäne konnte in Seren HIV-Infizierter zu jedem Stadium der Infektion nachgewiesen werden. Bei einer Expression der Antikörper in der frühen Infektionsphase wäre ein positiver Einfluss auf die Prognose der Krankheit vorstellbar. Ob ein Zusammenhang zwischen einer Antikörperexpression gegen Env362-420 in HIV-infizierten Schwangeren und der Wahrscheinlichkeit einer HIV-Transmission auf das Kind besteht, muss noch geklärt werden. Env362-420 kann zur Identifizierung des Rezeptors auf der epithelialen Oberfläche, welcher in die Transzytose von HIV involviert ist, und zur Entwicklung von Inhibitoren der Mutter–Kind-Übertragung von HIV herangezogen werden., The transport of HIV through the fetal membranes is discussed as one possible reason for the vertical transmission of HIV from mother to child during pregnancy or labor. HIV can penetrate epithelial barriers by a receptor-mediated transport mechanism involving interaction of a lectin-like domain on the viral glycoprotein gp120 and a receptor on the epithelial surface. In this study the domain on gp120 involved in transcytosis of cell-free HIV-1 through epithelial cells was characterized in more detail. Overlapping oligopeptides of gp120 were used to inhibit transcytosis of HIV 1 through an amnion cell monolayer. Four oligopeptides significantly inhibited transcytosis of HIV 1. A synthetic oligopeptide (Env362-420) with a length of 59 amino acids representing the sequence of the four inhibiting oligopeptides significantly reduced the transport of HIV, independent of the HIV 1 subtype. Furthermore, human HIV-positive sera with antibodies reacting with the domain Env362-420 and rabbit sera raised against the oligopeptide Env362-420 also inhibited the transport of HIV-1. Antibodies directed against the transcytosis domain could be detected in sera from every stage of infection. The development of these antibodies in the early stage of infection might play a role in the outcome of the HIV disease.It has to be investigated whether HIV 1-infected women who developed these antibodies show a lower rate of HIV transmission to their offspring than those without such antibodies. Env362–420 can also be used as a tool to identify the receptor involved in transcytosis on the epithelial cell surface and to develop inhibitors that could help prevent mother-to-child transmission of HIV during pregnancy or labor.

Hepatitis B Virus (HBV) Infektionen verursachen entzündliche Erkrankungen der Leber. Insbesondere die frühen Phasen des HBV Lebenszyklus sind noch nicht geklärt, so ist z.B. der Rezeptorkomplex an den HBV bindet unbekannt. Mittlerweile stehen neue Infektionsmodelle zur Verfügung um den HBV Lebenszyklus zu untersuchen. Dies erfordert eine effiziente Zellkultur basierende Methode um große Mengen infektiöser Partikel zu generieren. Ein Ziel der Arbeit war durch Kultivierung auf Mikrocarrier die HBV Produktion der Zelllinie HepG2.2.15 zu steigern. Die Analyse von Protein und HBV Sekretion, Infektiösität und MAP Signalübertragung ergab eine 18x höhere HBV Produktion bei einer reduzierten Sekretion von subviralen Partikeln durch HepG2.2.15 die auf Mikrocarrier kultiviert wurden. Der Anstieg der Virusproduktion korreliert mit einer verstärkten Aktivierung der MAP Kinase ERK-2, die mit HBV Replikation in Verbindung steht. Ein weiterer wenig verstandener Teil des HBV Lebenszyklus ist der Kernimport des HBV Genoms. Spuren der viralen Polymerase finden sich im Zellkern von HBV infizierten Zellen. Ziel der Arbeit war, Motive in der HBV Polymerase zu finden, die in der Lage sind Zelllokalisation zu beeinflussen. Durch Sequenzvergleich wurde eine konservierte zweiseitige Kernlokalisationssequenz im Terminalen Protein der HBV Polymerase identifiziert, die eine Proteinkinase CKII Erkennungsstelle enthält. Inhibition der CKII Aktivität in HBV infizierten primären Hepatozyten sowie die Zerstörung der CKII Erkennungsstelle im Terminalen Protein inhibieren die HBV Replikation. Die Funktionalität der Kernlokalisationssequenz wurde durch Fusion an GFP bestätigt und war Abhängig von CKII Aktivität in der Zelle. Dies wurde in vitro durch Bindung des Adapterproteins Karyopherin-alpha an CKII-phosphoryliertes Terminales Protein bestätigt. Die HBV Polymerase enthält eine konservierte zweiseitige Kernlokalisationssequenz deren Funktionalität durch CKII Phosphorilierung vermittelt wird., Hepatitis B virus (HBV) infection causes acute and chronic liver inflammation. Especially the early phase of the HBV life cycle is not clearly understood. For example the receptor complex that mediates viral entry is not known. Novel infection models to study the HBV lifecycle are described that demand for a large amount of cell culture generated infectious HBV particles. One aim was to enhance HBV production of the cell line HepG2.2.15 by cultivation on microcarrier substrate. Analysis of protein and viral particle secretion, infectivity, and cellular MAP kinase signaling revealed an up to 18x increased HBV production and a decreased subviral particle secretion by HepG2.2.15 when cultivated on microcarrier. The observed effect was due to an enhanced phospho-activation of MAP kinase ERK-2 that is tightly associated with HBV replication. Another poorly understood part of the HBV lifecycle is the mechanism that delivers the HBV genome into the nucleus. Traces of HBV polymerase can be found in HBV infected cells. The second objective was to identify motifs on the HBV polymerase that determine its subcellular localization. By sequence alignment a conserved bipartite nuclear localization signal was found in the terminal protein of the HBV polymerase encompassing a protein kinase CKII recognition site. Inhibition of CKII kinase in infected primary hepatocytes and destruction of the identified CKII recognition site in the viral polymerase impaired virus production. The functionality of the putative nuclear localization signal was confirmed by fusion to GFP. Moreover, its functionality was depended on CKII activity that was verified by in vitro binding experiments of terminal protein to the import adaptor karyopherin-alpha. This data identified a nuclear localization signal in the HBV polymerase, which functionality is mediated by CKII phosphorylation.

Die vorliegende Arbeit beschäftigt sich mit einem neuartigen proteinbasierten, suizidgentherapeutischen Ansatz zur sicheren und effektiven Behandlung von soliden Tumoren. Verwendet wurden zellpermeable Fusionsproteine auf der Grundlage des bakteriellen Enzyms Cytosin Desaminase, welches spezifisch die Umsetzung der inaktive, nichttoxische Substanz (Prodroge) 5-Fluorcytosin in den hochwirksamen, stark toxischen Wirkstoff 5-Fluoruracil katalysiert. Dieser bewirkt die selektive Zerstörung von Tumorzellen. Durch die Fusion der bakteriellen Cytosin Desaminase (bCD) mit der Sequenz des Zellpermeabilität vermittelnden Peptides HBV-Translokationsmotiv (TLM) des Hepatits B-Virus (HBV) wurden zunächst zellpermeable E.coli Cytosin Desaminase Suizidfusionskonstrukte generiert. Für die bakteriell synthetisierten HBV-TLM-Fusionsproteine konnten eine Hexamerisierung sowie eine spezifische enzymatische Aktivität bei der Umsetzung von Cytosin zu Uracil als strukturelle und funktionelle Voraussetzungen für einen Einsatz in der Suizidgentherapie nachgewiesen werden, die vergleichbar mit dem wt-Protein waren. Bei Versuchen zur Internalisierung der zellpermeablen Fusionsproteine wurde für die Fusionsproteine mit C-terminal fusioniertem HBV-TLM (bCD-HBV-TLM) eine Aufnahme in das Zytoplasma von Hepatomzellen mittels konfokaler Laserscanmikroskopie und differentieller Zellfraktionierung nachgewiesen, nicht jedoch für Fusionsproteine mit N-terminalem HBV-TLM (HBV-TLM-bCD). Die gezeigte Internalisierung des Proteins HBV-TLM-bCD erfolgte effizient und schnell und war unabhängig vom endosomalen Aufnahmeweg. Bei der nachgewiesenen Translokalisation blieb die enzymatische, suizidgentherapeutische Aktivität des zellpermeablen Suizidproteins (HBV-TLM-bCD), d.h. die katalytische Wirkung bei der Umsetzung der Prodroge 5-Fluorcytosin vollständig erhalten, so dass sich dieses Fusionsprotein für einen therapeutischen Einsatz in der Suizidgentherapie eignet. Zusätzlich zur antitumoralen Wirkung können d, This work investigates the application of protein based therapeutic suicide enzyme/prodrug approaches providing novel means for both safe and effective local therapeutic regimes in solid tumors. The concept of the used suicide gene therapy system is based mainly on the transfer of the cell permeable bacterial suicide enzyme cytosine deaminase which specifically convert the inactive, non-toxic prodrug 5-fluorocytosine into the toxic metabolite 5-fluorouracil finally executing the efficient destruction of tumor cells. Employing a novel cell permeable peptide, known as the translocation motif (TLM) of hepatitis B virus (HBV), E.coli cytosine deaminase (bCD) suicide fusion proteins were generated. HBV-TLM fusion proteins formed hexamers (as do parental wt bCD) and retained the specific enzymatic activity of cytosine conversion to uracil also being comparable to parental wtbCD protein. However, only bCD-HBV-TLM fusion proteins, but not HBV-TLM-bCD fusion proteins were found to be taken up to the cytoplasm of target hepatoma cells as demonstrated both by confocal laser scanning microscopy and cell fractionation. Uptake of bCD-HBV-TLM worked both efficiently and rapidly and was found to be independent from the endosomal pathway. Since bCD-HBV-TLM fusion proteins completely retained their suicide enzymatic activity in the course of translocation across the plasma membrane their usage as profound inducers of chemo-sensitivity to 5-fluorocytosine strongly is suggested. Future therapeutic local application of cell permeable bCD-HBV-TLM fusion proteins together with a systemic 5-fluorocytosine prodrug application could result in profound antitumor activities without apparent side effects.

BACKGROUND: Up to 20 million persons are infected with the human retroviruses human T-lymphotropic virus (HTLV)-I and HTLV-II globally. Most data on the seroprevalence of HTLV-I and HTLV-II in Europe are from studies of low-risk blood donors or high-risk injection drug users (IDUs). Little is known about the general population. METHODS: A prospective anonymous study of HTLV-I and HTLV-II seroprevalence among 234,078 pregnant women in Belgium, France, Germany, Italy, Portugal, Spain, and the United Kingdom was conducted. Maternal antibody status was determined by standard methods using sera obtained for routine antenatal infection screens or eluted from infant heel prick dried blood spots obtained for routine neonatal metabolic screens. RESULTS: Anti-HTLV-I/II antibodies were detected and confirmed in 96 pregnant women (4.4 per 10,000, 95% confidence interval [CI]: 3.5-5.2). Of these, 73 were anti-HTLV-I, 17 were anti-HTLV-II, and 6 were specifically anti-HTLV but untyped. The seroprevalence ranged from 0.7 per 10,000 in Germany to 11.5 per 10,000 in France. CONCLUSIONS: Pregnant women better reflect the general population than blood donors or IDUs. The seroprevalence of HTLV-I and HTLV-II in Western Europe is 6-fold higher among pregnant women (4.4 per 10,000) than among blood donors (0.07 per 10,000). These data provide a robust baseline against which changes in HTLV-I and HTLV-II seroprevalence in Europe can be measured.
