The growing use of herbal medicines worldwide requires ensuring their quality, safety, and efficiency to consumers and patients. Quality controls of vegetal extracts are usually undertaken according to pharmacopeial monographs. Analyses may range from simple chemical experiments to more sophisticated but more accurate methods. Nowadays, metabolomic analyses allow a fast characterization of complex mixtures. In the field, besides mass spectrometry (MS), nuclear magnetic resonance spectroscopy (NMR) has gained importance in the direct identification of natural products in complex herbal extracts. For a decade, automated dereplication processes based on 13C-NMR have been emerging to efficiently identify known major compounds in mixtures. Though less sensitive than MS, 13C-NMR has the advantage of being appropriate to discriminate stereoisomers. Since NMR spectrometers nowadays provide useful datasets in a reasonable time frame, we have recently made available MixONat, a software that processes 13C as well as distortionless enhancement by polarization transfer (DEPT)-135 and -90 data, allowing carbon multiplicity (i.e., CH3, CH2, CH, and C) filtering as a critical step. MixONat requires experimental or predicted chemical shifts (δ C) databases and displays interactive results that can be refined based on the userʼs phytochemical knowledge. The present article provides step-by-step instructions to use MixONat starting from database creation with freely available and/or marketed δ C datasets. Then, for training purposes, the reader is led through a 30 – 60 min procedure consisting of the 13C-NMR based dereplication of a peppermint essential oil.

Thirty-three natural products were isolated from the aerial parts of Antidesma bunius, Euphorbiaceae, a plant used in Vietnamese traditional medicine against rheumatoid arthritis. All compounds were reported the first time for this species, and nine constituents resembled undescribed natural products, noticeably three coumarinolignans with 2,2-dimethyl-1,3-dioxolane moiety, two cyclopeptides, and two furofuran-type lignans connected with a phenylpropanoid moiety. The individual structures were elucidated by combining NMR and MS data, and their configuration was established by NOESY and ECD experiments and NMR calculations. Presently it is estimated that many of the approximately 4000 new natural products isolated every year following complicated, long, and expensive isolation processes are already known; because of this, developing new strategies for locating secondary metabolites of interest in complex extracts or fractions is important. Currently, chromatographic and spectroscopic techniques are being used to optimize the isolation and identification of natural products. In this investigation we have used 13C NMR dereplication analyses for the quick identification of a number of triterpenes (friedelin, lupeol, betulinic acid), sterols (euphol, β-sitosterol) and fatty acids (palmitic acid) present in semipurified fractions obtained from the stem bark extract of Clusia flava and to assist in the isolation of the bioactive metabolites trapezifolixanthone and paralycolin A. The complete and correct assignment of the 1H and 13C NMR spectroscopic data for paralycolin A is reported for the first time and the antioxidant and antiAGEs activity of both metabolites is described.

Chalcone-vitamin E hybrids 6’-O-tosyl-3,4,5-trimethoxy-δ-tocopherol-chalcone (1), 3,4,5-trimethoxy-δ-tocopherol-chalcone (2), 6’-O-tosyl-3,4,5-trimethoxy-δ-tocopherol-retrochalcone (3) and 3,4,5-trimethoxy-δ-tocopherol-retrochalcone (4) were synthesized as part of a search for new biological activities in these types of derivatives. We report herein on the photoisomerization products of hybrids 1-4, and the effects of the solvent and substitution patterns in producing secondary products such as flavanone 6, 3-deoxyanthocyanidin 8, and hemiketal 10. Photochemically-induced changes are considered important since structural modifications and/or the presence of additional products can affect the biological activity of this type of semisynthetic hybrids. Los híbridos de chalcona-vitamina E, 6’-O-tosil-3,4,5-trimetoxi-δ-tocoferol-chalcona (1), 3,4,5-trimetoxi-δ-tocoferol-chalcona (2), 6’-O-tosil-3,4,5-trimetoxi-δ-tocoferol-retrochalcona (3) y 3,4,5-trimetoxi-δ-tocoferol-retrochalcona (4), fueron sintetizados como parte de la búsqueda de nuevos perfiles de actividad biológica para este tipo de derivados. En este trabajo reportamos los productos de fotoisomerización de los híbridos 1-4, y los efectos del disolvente, así como de distintos patrones de sustitución en la generación de productos secundarios como la flavanona 6, la 3-deoxiantocianidina 8, y el hemicetal 10. Los cambios fotoinducidos son considerados de gran importancia debido a que la modificación en la estructura y/o la presencia de productos adicionales puede afectar la actividad biológica de este tipo de híbridos semisintéticos.

Endogenous long-chain metabolites of vitamin E (LCMs) mediate immune functions by targeting 5-lipoxygenase (5-LOX) and increasing the systemic concentrations of resolvin E3, a specialized proresolving lipid mediator. SAR studies on semisynthesized analogues highlight α-amplexichromanol (27a), which allosterically inhibits 5-LOX, being considerably more potent than endogenous LCMs in human primary immune cells and blood. Other enzymes within lipid mediator biosynthesis were not substantially inhibited, except for microsomal prostaglandin E2 synthase-1. Compound 27a is metabolized by sulfation and β-oxidation in human liver-on-chips and exhibits superior metabolic stability in mice over LCMs. Pharmacokinetic studies show distribution of 27a from plasma to the inflamed peritoneal cavity and lung. In parallel, 5-LOX-derived leukotriene levels decrease, and the inflammatory reaction is suppressed in reconstructed human epidermis, murine peritonitis, and experimental asthma in mice. Our study highlights 27a as an orally active, LCM-inspired drug candidate that limits inflammation with superior potency and metabolic stability to the endogenous lead.

Alkaloids represent a major group of natural products (NPs), derived from highly diverse organisms. These structurally varied specialized metabolites are widely used for medicinal purposes and also known as toxic contaminants in agriculture and dietary supplements. While the detection of alkaloids is generally facilitated by GC- or LC-MS, these techniques do require considerable efforts in sample preparation and method optimization. Bypassing these limitations and also reducing experimental time, matrix-free laser desorption ionization (LDI) and related methods may provide an interesting alternative. As many alkaloids show close structural similarities to matrices used in matrix-assisted laser desorption ionization (MALDI), they should ionize upon simple laser irradiation without matrix support. With this in mind, the current work presents a systematic evaluation of LDI properties of a wide range of structurally diverse alkaloids. Facilitating a direct comparison between LDI and ESI-MS fragmentation, all tested compounds were further studied by electrospray ionization (ESI). Moreover, crude plant extracts of Atropa belladonna, Cinchona succirubra, and Colchicum autumnale were analyzed by LDI in order to evaluate direct alkaloid detection and dereplication from complex mixtures. Finally, dose-dependent evaluation of MALDI and LDI detection using an extract of Rosmarinus officinalis spiked with atropine, colchicine, or quinine was conducted. Overall, present results suggest that LDI provides a versatile analytical tool for analyzing structurally diverse alkaloids as single compounds and from complex mixtures. It may further serve various potential applications ranging from quality control to the screening for toxic compounds as well as the build up of MS databases. Graphical abstract.

Our aim was to investigate the molecular determinants of liver colonization and potential common biomarkers of sarcomas and sarcomatoid tumors, using the most invasive (M5-T1) of our four experimental models of peritoneal sarcomatoid malignant mesothelioma in the F344 rat. Using an advanced and robust technique of quantitative proteomics and a bank of paraffin-embedded tumor and tissue samples, we analyzed changes in the proteotype patterns of the liver from normal rats, adjacent non-tumorous liver from untreated tumor-bearing rats, and liver from tumor-bearing rats positively responding to repeated administrations of curcumin given intraperitoneally. The identification of proteome alterations accounting for the antitumor effects of curcumin and changes in the liver microenvironment, which favored the induction of an immune response, could be useful to the research community.AbstractInvestigations of liver metastatic colonization suggest that the microenvironment is preordained to be intrinsically hospitable to the invasive cancer cells. To identify molecular determinants of that organotropism and potential therapeutic targets, we conducted proteomic analyses of the liver in an aggressive model of sarcomatoid peritoneal mesothelioma (M5-T1). The quantitative changes between SWATH-MS (sequential window acquisition of all theoretical fragmentation spectra) proteotype patterns of the liver from normal rats (G1), adjacent non-tumorous liver from untreated tumor-bearing rats (G2), and liver from curcumin-treated rats without hepatic metastases (G3) were compared. The results identified 12 biomarkers of raised immune response against M5-T1 cells in G3 and 179 liver biomarker changes in (G2 vs. G1) and (G3 vs. G2) but not in (G3 vs. G1). Cross-comparing these 179 candidates with proteins showing abundance changes related to increasing invasiveness in four different rat mesothelioma tumor models identified seven biomarkers specific to the M5-T1 tumor. Finally, analysis of correlations between these seven biomarkers, purine nucleoside phosphorylase being the main biomarker of immune response, and the 179 previously identified proteins revealed a network orchestrating liver colonization and treatment efficacy. These results highlight the links between potential targets, raising interesting prospects for optimizing therapies against highly invasive cancer cells exhibiting a sarcomatoid phenotype and sarcoma cells.

Alternaria dauci is the causal agent of Alternaria leaf blight (ALB), a foliar disease of carrot crops (Daucus carota) around the world. In terms of phytotoxic metabolites production, A. dauci has received limited attention. Previous studies carried out on the secondary metabolites involved in the pathogenicity of this fungus have only reported the isolation of a ubiquitous non-selective phytotoxin known as zinniol. Because of this, the aim of this research is directed towards the isolation and identification of secondary metabolites involved in the plant-pathogen interaction process. A. dauci was cultured in the Czapek-Dox medium, and the culture filtrate was extracted with ethyl acetate. The leaf-spot assay of fractions resulting from the partition process showed a phytotoxic effect in the ethyl acetate fraction. The chromatographic separation of ethyl acetate fraction allowed the isolation of seven diketopiperazines, identified as cyclo-(pro-val) (1), cyclo-(pro-leu) (2), cyclo-(pro-phe) (3), cyclo-(val-leu) (4), cyclo-(val-phe) (5), cyclo-(leu-phe) (6) and cyclo-(leu-tyr) (7). The structures of the different metabolites were established by comparing their spectroscopic (1H NMR) and spectrometric (GC-MS) data with those reported in the literature. Resumen. Alternaria dauci es el agente causal del tizón de la hoja (ALB), una enfermedad foliar que afecta los cultivos de zanahoria (Daucus carota) alrededor del mundo. En términos de producción de metabolitos fitotóxicos, A. dauci ha recibido una atención muy limitada. Estudios previos llevados a cabo sobre los metabolitos secundarios involucrados en la patogenicidad de este hongo, solo han reportado el aislamiento de una fitotoxina no selectiva y ubicua conocida como zinniol. Debido a lo anterior, el objetivo de esta investigación se dirige al aislamiento e identificación de metabolitos secundarios implicados en la interacción planta-patógeno. Para esto el fitopatógeno se cultivó en medio Czapek-Dox y el filtrado del cultivo se extrajo con acetato de etilo. La evaluación de las fracciones resultantes de la partición, en el ensayo de manchas foliares en hojas, mostró un efecto fitotóxico en la fracción de acetato de etilo. La separación cromatográfica de la fracción de acetato de etilo permitió el aislamiento de siete dicetopiperazinas identificadas como ciclo-(pro-val) (1), ciclo-(pro-leu) (2), ciclo-(pro-phe) (3), ciclo-(val-leu) (4), ciclo-(val-phe) (5), ciclo-(leu-phe) (6) y ciclo-(leu-tyr) (7). Las estructuras de los diferentes metabolitos se establecieron comparando sus datos espectroscópicos (1H RMN) y espectrométricos (CG-EM) con los reportados en la literatura.

Alternaria dauci is the causal agent of Alternaria leaf blight (ALB) in carrot (Daucus carota) crops around the world. In this investigation, the bioassay-guided isolation of the extract from liquid cultures of A. dauci resulted in the isolation of two metabolites identified as &alpha, acetylorcinol (1) and p-hydroxybenzoic acid (2), based on their spectroscopic data and results from chemical correlation reactions. Testing of both metabolites in different assays showed an important phytotoxic activity for p-hydroxybenzoic acid (2) when tested in the leaf-spot assay on parsley (Petroselinum crispum), in the leaf infiltration assay on tobacco (Nicotiana alata) and marigold (Tagetes erecta), and in the immersion assay on parsley and parsnip (Pastinaca sativa) leaves. Quantification of the two metabolites in the crude extract of A. dauci kept at different times showed that p-hydroxybenzoic acid (2) is one of the first metabolites to be synthesized by the pathogen, suggesting that this salicylic acid derivative could play an important role in the pathogenicity of the fungus.

Propolis is a material produced by bees from a combination of plant exudates and wax, used to fill out cracks in the beehive and to defend against intruders and pathogenic microorganisms; it is recognized for its many biological activities and its chemical composition depends on the botanical sources close to the beehive. The objective of this investigation was to isolate and identify metabolites with antioxidant activity present in a propolis sample collected in Yucatan, Mexico. Purification of the bioactive metabolites was carried out using argentation chromatography, while the combination of 1H nuclear magnetic resonance (NMR), laser desorption ionization (LDI), gas chromatography-mass spectrometry (GC-MS) and biosynthetic origin data allowed their identification as resorcinolic lipids. Finally, the resin of Mangifera indica was identified as the botanical source of these metabolites.

Systemic vitamin E metabolites have been proposed as signaling molecules, but their physiological role is unknown. Here we show, by library screening of potential human vitamin E metabolites, that long-chain ω-carboxylates are potent allosteric inhibitors of 5-lipoxygenase, a key enzyme in the biosynthesis of chemoattractant and vasoactive leukotrienes. 13-((2R)-6-hydroxy-2,5,7,8-tetramethylchroman-2-yl)-2,6,10-trimethyltridecanoic acid (α-T-13′-COOH) can be synthesized from α-tocopherol in a human liver-on-chip, and is detected in human and mouse plasma at concentrations (8–49 nM) that inhibit 5-lipoxygenase in human leukocytes. α-T-13′-COOH accumulates in immune cells and inflamed murine exudates, selectively inhibits the biosynthesis of 5-lipoxygenase-derived lipid mediators in vitro and in vivo, and efficiently suppresses inflammation and bronchial hyper-reactivity in mouse models of peritonitis and asthma. Together, our data suggest that the immune regulatory and anti-inflammatory functions of α-tocopherol depend on its endogenous metabolite α-T-13′-COOH, potentially through inhibiting 5-lipoxygenase in immune cells.

International audience; Matrix-free laser desorption ionization (LDI) is a rapid and versatile technique for the ionization of small, UV-light-absorbing molecules. Indeed, many natural products such as polyphenols exhibit inherent LDI properties, potentially facilitating their detection from highly complex samples such as crude extracts. With this in mind, the present work thoroughly evaluated the potential of LDI as an analytical tool for the chemical profiling and differentiation of propolis samples obtained from different global regions. Propolis is a complex bee product containing, among others, significant amounts of phenolic constituents that may show LDI effects. The present work will demonstrate that LDI not only provides reproducible and highly specific fingerprint spectra for each of the tested samples, it further allows their clear differentiation by principal compound analysis (PCA). Contrary to classical analytical approaches such as LC- or GC-MS, LDI does not require time-consuming sample preparation and method optimization procedures. Thus, the technique represents a most interesting analytical tool and potent supplement to classic LC-MS for quality control of herbal pharmaceuticals and dietary supplements. Present results clearly support this approach and further suggest the use of LDI as a versatile tool for the automated analysis of large sample batches on an industrial scale.

M. ulcerans infection is a type of panniculitis beginning mostly with a nodule or an oedema, which can progress to large ulcerative lesions. The lesions are caused by mycolactone, the polyketide toxin of M. ulcerans. Mycolactone plays a central role for host colonization as it has immunomodulatory and analgesic effects. On one hand, mycolactone induces analgesia by targeting type-2 angiotensin II receptors (AT 2 R), causing cellular hyperpolarization and neuron desensitization. Indeed, a single subcutaneous injection of mycolactone into the mouse footpad induces a long-lasting hypoesthesia up to 48 h. It was suggested that the long-lasting hypoesthesia may result from the persistence of a significant amount of mycolactone locally following its injection, which could be probably due to its slow elimination from tissues. To verify this hypothesis, we investigated the correlation between hypoesthesia and mycolactone bioavailability directly at the tissue level. Various quantities of mycolactone were then injected in mouse tissue and hypoesthesia was recorded with nociception assays over a period of 48 h. The hypoesthesia was maximal 6 h after the injection of 4 µg mycolactone. The basal state was reached 48 h after injection, which demonstrated the absence of nerve damage. Surprisingly, mycolactone levels decreased strongly during the first hours with a reduction of 70 and 90% after 4 and 10 h, respectively. Also, mycolactone did not diffuse in neighboring skin tissue and only poorly into the bloodstream upon direct injection. Nevertheless, the remaining amount was sufficient to induce hypoesthesia during 24 h. Our results thus demonstrate that intact mycolactone is rapidly eliminated and that very small amounts of mycolactone are sufficient to induce hypoesthesia. Taken together, our study points out that mycolactone ought to be considered as a promising analgesic.

This Data Descriptor announces the submission to public repositories of the monoterpene indole alkaloid database (MIADB), a cumulative collection of 172 tandem mass spectrometry (MS/MS) spectra from multiple research projects conducted in eight natural product chemistry laboratories since the 1960s. All data have been annotated and organized to promote reuse by the community. Being a unique collection of these complex natural products, these data can be used to guide the dereplication and targeting of new related monoterpene indole alkaloids within complex mixtures when applying computer-based approaches, such as molecular networking. International audience; The genus Mesua L. (Calophyllaceae) comprises approximately 50 species that grow in the restrictive area of South East Asia. Investigations of Mesua species reported the chemodiversity and pharmacological relevance of their phytoconstituents, in particular polyphenolic compounds. To date, about 170 secondary metabolites have been identified from 13 Mesua species, predominantly xanthones and coumarins. Most of them hold a prenylated skeleton and display activities such as antitumor and antimicrobial, antioxidant, anti-inflammatory or immunomodulating properties, which were evaluated using either in vitro assay or, in rare cases, in vivo animal studies. Prenylation of aromatic compounds appears as a key element to enhance their biological activities, including their cytotoxicity and, could be involved in their inhibitory potential towards enzymes playing crucial roles in inflammatory and immune cascades. The aim of this review is to provide a systematic and comprehensive overview of Mesua polyphenolic secondary metabolites and their pharmacological properties. Among them, the structure of coumarin beccamarin T is herein revised and attributed to lepidotol A based on a careful examination of the 1D and 2D NMR spectra of the respective compounds. A considerable attention is also given to the place of prenylated polyphenols as anti-inflammatory and immunomodulatory molecules, to highlight the interest of Mesua metabolites as chemical probes for target discovery.

Modulation of major histocompatibility complex (MHC) expression using drugs has been proposed to control immunity. Phytochemical investigations on Garcinia species have allowed the isolation of bioactive compounds such as polycyclic polyprenylated acylphloroglucinols (PPAPs). PPAPs such as guttiferone J (1), display anti-inflammatory and immunoregulatory activities while garcinol (4) is a histone acetyltransferases (HAT) p300 inhibitor. This study reports on the isolation, identification and biological characterization of two other PPAPs, i.e., xanthochymol (2) and guttiferone F (3) from Garcinia bancana, sharing structural analogy with guttiferone J (1) and garcinol (4). We show that PPAPs 1&ndash, 4 efficiently downregulated the expression of several MHC molecules (HLA-class I, -class II, MICA/B and HLA-E) at the surface of human primary endothelial cells upon inflammation. Mechanistically, PPAPs 1&ndash, 4 reduce MHC proteins by decreasing the expression and phosphorylation of the transcription factor STAT1 involved in MHC upregulation mediated by IFN-&gamma, Loss of STAT1 activity results from inhibition of HAT CBP/p300 activity reflected by a hypoacetylation state. The binding interactions to p300 were confirmed through molecular docking. Loss of STAT1 impairs the expression of CIITA and GATA2 but also TAP1 and Tapasin required for peptide loading and transport of MHC. Overall, we identified new PPAPs issued from Garcinia bancana with potential immunoregulatory properties.

Inflammation contributes to the development of various pathologies, e.g. asthma, cardiovascular diseases, some types of cancer, and metabolic disorders. Leukotrienes (LT), biosynthesized from arachidonic acid by 5-lipoxygenase (5-LO), constitute a potent family of pro-inflammatory lipid mediators. δ-Garcinoic acid (δ-GA) (1), a natural vitamin E analogue, was chosen for further structural optimization as it selectively inhibited 5-LO activity in cell-free and cell-based assays without impairing the production of specialized pro-resolving mediators by 15-LO. A model of semi-quantitative prediction of 5-LO inhibitory potential developed during the current study allowed the design of 24 garcinamides that were semi-synthesized. Secondary metabolites from lichens are known for exhibiting various biological effects such as anti-inflammatory, antioxidant and antibacterial activities. Despite this wide range of reported biological effects, their impact on the formation of advanced glycation end products (AGEs) remains vastly unexplored. The latter are known contributors to lifestyle and age-related diseases such as Alzheimer and Parkinson. Moreover, the development of atherosclerosis and arterial stiffness is causally linked to the formation of AGEs. With this in mind, the present work evaluated the inhibitory effects of secondary lichen metabolites on the formation of pentosidine-like AGEs' by using an in vitro, Maillard reaction based, fluorescence assay. Overall, thirty-seven natural and five synthetically modified compounds were tested, eighteen of which exhibiting IC50 values in the range of 0.05 to 0.70 mM. This corresponds to 2 to 32 fold of the inhibitory activity of aminoguanidine. Targeting one major inhibiting mechanism of AGEs formation, all compounds were additionally evaluated on their radical scavenging capacities in an DPPH assay. Furthermore, as both AGEs' formation and hypertension are major risk factors for atherosclerosis, compounds that were available in sufficient amounts were also tested for their vasodilative effects. Overall, and though some of the active compounds were previously reported cytotoxic, present results highlight the interesting potential of secondary lichen metabolites as anti-AGEs and vasodilative agents.

International audience; Through dereplication analysis, seven known Mammea coumarins were identified in a fraction obtained from Mammea neurophylla dichloromethane bark extract selected for its ability to prevent advanced glycation end-product (AGE) formation. Among them, a careful examination of the NMR dataset of pedilanthocoumarin B led to a structural revision. Inspection of LC-DAD-MSn chromatograms allowed us to predict the presence of four new compounds, which were further isolated. Using spectroscopic methods (1H-, 13C- and 2D-NMR, HRMS, UV), these compounds were identified as new benzoyl substituted 4-phenylcoumarins (iso-pedilanthocoumarin B and neurophyllol C) and 4-(1-acetoxypropyl)coumarins cyclo F (ochrocarpins H and I).

International audience; Propolis is a resinous natural substance collected by honeybees from buds and exudates of various trees and plants; it is widely accepted that the composition of propolis depends on the phytogeographic characteristics of the site of collection. In this study we have analyzed the chemical composition of a propolis collected in Quintana Roo, Mexico, and evaluated its antioxidant, antifungal and antibacterial activities. Unexpectedly, the chemical analysis showed that the main components of the ethanolic extract of a Mexican propolis appeared to be pentacyclic triterpenoids, such as α and β-amyrin derivatives, and sterols. The crude extract did not show antioxidant activity when tested using the DPPH-reduction assay, and it also proved inactive when tested for antifungal and antibacterial activities using microdilution and agar diffusion assays, respectively. The fact that the presence of both α and β-amyrins and their derivatives have been reported from the resin of Bursera simaruba, one of the plants used by the bees for propolis production in Quintana Roo, Mexico, confirms the relationship that exists between the flora available to bees in a given region and the chemical composition of the propolis that they produce.; El propóleo es un producto natural resinoso colectado por abejas a partir de brotes y exudados de diferentes plantas y árboles; es ampliamente aceptado que la composición del propóleo depende de las características fitogeográficas del sitio de colecta. En este estudio analizamos la composición química de un propóleo colectado en Quintana Roo, México, y evaluamos sus actividades antioxidante, antifúngica y antibacteriana. Inesperadamente, el análisis químico mostró que los principales componentes del extracto etanólico del propóleo mexicano parecían ser triterpenoides pentacíclicos, tales como derivados de α y β-amyrina y esteroles. El extracto crudo no mostró actividad antioxidante al ser evaluado en el ensayo de reducción de DPPH, y también se mostró inactivo al ser evaluado en cuanto a su actividad antifúngica y antibacteriana utilizando los ensayos de microdilución y de difusión en agar, respectivamente. El hecho de que la presencia de α y β-amyrina y sus derivados ya se ha reportado en la resina de Bursera simaruba, una de las plantas utilizadas por las abejas para su producción de propóleo en Quintana Roo, México, confirma la relación que existe entre la flora disponible para las abejas en una región en particular y la composición química del propóleo que producen.

Alkaloids represent a group of biologically most interesting compounds commonly used in modern medicines but also known for exhibiting severe toxic effects. Therefore, the detection of alkaloids is an important issue in quality control of plants, dietary supplements, and herbal pharmaceutical and mostly facilitated by methods such as GC or LC-MS. However, benefitting from the development of selective matrices as well as requiring very little sample preparation, MALDI-MS may also provide a valuable supplement to these standard analytical methods. With this in mind, the present study highlights recent advances in the development of bithiophenic matrix molecules designed for the selective detection of alkaloids. Overall four new bithiophenic matrix molecules (BMs) were tested on different analytes belonging to various chemical families such as alkaloids, curcuminoids, benzopyrones, flavonoids, steroids, and peptides (I). All BMs were further compared to the commercial matrices α-cyano-4-hydroxycinnamic acid (CHCA) and 2,5-dihydroxybenzoic acid (DHB) in terms of their signal response as well as their matrix noise formation (II). Based on these results the most promising candidate, 3-(5'-pentafluorophenylmethylsulfanyl-[2,2']bithiophenyl-5-ylsulfanyl)propionitrile (PFPT3P), was tested on highly complex samples such as the crude extracts of Colchicum autumnale, RYTMOPASC ® solution (a herbal pharmaceutical containing sparteine and rubijervine), as well as strychnine-spiked human plasma (III). For the latter, an evaluation of the limit of detection was performed. Eventually, a simplified protocol for the direct MALDI detection of major alkaloids from pulverized plant material of Atropa belladonna and Senecio vulgaris is presented (IV). International audience; RATIONALE: Light-absorbing secondary metabolites from lichens were recently reported to exhibit promising Laser Desorption Ionization (LDI) properties, enabling their direct detection from crude lichen extracts. In addition, many of them display close structural homologies to commercial Matrix-Assisted Laser Desorption Ionization (MALDI) matrices, which is incentive for the evaluation of their matrical properties. The current study systematically evaluated the matrix effects of several structural classes of lichen metabolites: monoaromatic compounds, quinone derivatives, dibenzofuran-related molecules and the shikimate-derived vulpinic acid. Their matrical properties were tested against a wide range of structurally diverse analytes including alkaloids, coumarins, flavonoids and peptides.METHODS: Triplicate automatic positive-ion mode MALDI analyses were carried out and ionization efficiencies were compared with those of structurally related reference matrices (i.e. DHB, HCCA, dithranol and usnic acid) in terms of (i) analyte absolute intensities and (ii) Matrix Suppressing Effect (MSE) scores.RESULTS: Monoaromatic lichen metabolites revealed matrical properties similar to those of DHB when obtained under comparable experimental conditions. Likewise, anthraquinone metabolites triggered ionization of tested analytes in a similar way to the structurally related dithranol. Finally, dibenzofuran derivatives displayed a broad ionization profile, reminiscent of that of (+)-usnic acid.CONCLUSIONS: Lichen metabolites exhibit interesting MALDI matrix properties, especially for medium and low molecular weight analytes. For many of the tested molecules, matrix ion formation was very limited. This proof-of-concept study paves the way for follow-up investigations to assess the matrix properties of lichen metabolites against a wider array of analytes as well as adapting experimental settings to individually optimize the performance of successfully tested candidates.

International audience; Saponins have the potential to favorably modulate rumen fermentation, but there is generally a lack of the chemical structures associated with the described effects. The activity of extracts fromCalendula officinalisandSaponaria officinalisin the rumen was evaluatedin vitro. TheS. officinalisroot extract, reduced CH4production by 8.5% and increased total VFA concentration by 25.2%.C. officinalisandS. officinalisroot extracts and theS. officinalisaerial part extract decreased the acetate to propionate ratio from 8.6 to 17.4%, according to the extract. An HPLC-ELSD analysis indicated that the saponin content ranged from 43.6 to 57.6 mg/g of dry matter (DM) in theC. officinalisextracts and from 224.0 to 693.8 mg/g of DM in theS. officinalisextracts, expressed as the hederacoside C equivalent. Identification of the saponin compounds present in the extracts by HPLC–MSnsuggested that the saponin profile modulated the biological activities, showing the importance of determining the structure of saponins when evaluating extracts.

International audience; Voltage-gated sodium channels (Nav) are molecular targets of clinically used drugs for treatments of various diseases (epilepsy, chronic pain, cardiac arrhythmia...) and also of numerous animal and plant neuro- toxins. The development of easy-to-use screening assays for the search of new ligands from chemicals libraries, animal venoms or plant extracts represents a challenge of a great interest to generate therapeutic hits. Here, we used the mammalian GH3B6 pituitary cell line, which consti- tutively expresses three different neuronal Nav channels isoforms (Nav1.2, Nav1.3 and Nav1.6), to identify novel compounds of pharmaco- logical interest from a library of in-house natural alkaloids. For this screening, we developed a method based on the use of Voltage-Sensor Probes (VSPs) that we adapted to detect both activators and blockers of Nav channels. Among the 62 alkaloids tested, 11 appeared as potent Nav channels inhibitors. Five other compounds were characterized as specific gating modifiers. While most of these alkaloids are already described in the literature, their ability to modulate Nav channels was unknown. In conclusion, we report here the suitability of this novel VSPs-based screening method i) to challenge the discovery and ii) to assess the ac- tivity of novel ligands on Nav channels.

International audience; The study describes bioactive compounds as inhibitors of acetylcholinesterase (AChE), from the stem bark extract of Montrouziera cauliflora, selected among 19 dichloromethane extracts from Clusiaceae species. Our work focused on the development of an original normal phase HPLC microfractionation strategy to rapidly assess highly active zones from this crude active non-polar plant extract. Two different microfraction collection methods were evaluated for the assessment of the AChE inhibition. Two guttiferones and a tocotrienol were directly isolated among five compounds identified off-line by NMR after upscaling the fractionation and their AChE inhibition was evaluated. The strengths and weaknesses of the two microfractionation collection methods for HPLC-AChE activity-based profiling are discussed.

International audience; The synthesis of five light absorbing triazolobithiophenic thiols, which were utilized for producing self-assembled monolayers (SAMs) on gold surfaces, is presented. The monolayer formation was monitored by cyclic voltammetry, indicating excellent surface coverage. The new triazolobithiophenic compounds exhibited an absorption maximum around 340 nm, which is close to the emission wavelength of a standard nitrogen laser. Consequently these compounds could be used to aid ionization in laser desorption mass spectrometry (MS).
