Abstract Background Limited information is available regarding the application of lung-protective ventilation strategies during one-lung ventilation (OLV) across mainland China. A nationwide questionnaire survey was conducted to investigate this issue in current clinical practice. Methods The survey covered various aspects, including respondent demographics, the establishment and maintenance of OLV, intraoperative monitoring standards, and complications associated with OLV. Results Five hundred forty-three valid responses were collected from all provinces in mainland China. Volume control ventilation mode, 4 to 6 mL per kilogram of predictive body weight, pure oxygen inspiration, and a low-level positive end-expiratory pressure ≤ 5 cm H2O were the most popular ventilation parameters. The most common thresholds of intraoperative respiration monitoring were peripheral oxygen saturation (SpO2) of 90–94%, end-tidal CO2 of 45 to 55 mm Hg, and an airway pressure of 30 to 34 cm H2O. Recruitment maneuvers were traditionally performed by 94% of the respondents. Intraoperative hypoxemia and laryngeal injury were experienced by 75% and 51% of the respondents, respectively. The proportions of anesthesiologists who frequently experienced hypoxemia during OLV were 19%, 24%, and 7% for lung, cardiovascular, and esophageal surgeries, respectively. Up to 32% of respondents were reluctant to perform lung-protective ventilation strategies during OLV. Abstract Background Castor is an important industrial raw material. Drought-induced oxidative stress leads to slow growth and decreased yields in castor. However, the mechanisms of drought-induced oxidative stress in castor remain unclear. Therefore, in this study, physiological, biochemical, and RNA-seq analyses were conducted on the roots of castor plants under PEG-6000 stress for 3 d and 7 d followed by 4 d of hydration. Results The photosynthetic rate of castor leaves was inhibited under PEG-6000 stress for 3 and 7 d. Biochemical analysis of castor roots stressed for 3 d and 7 d, and rehydrated for 4 d revealed that the activities of APX and CAT were highest after only 3 d of stress, whereas the activities of POD, GR, and SOD peaked after 7 d of stress. RNA-seq analysis revealed 2926, 1507, and 111 differentially expressed genes (DEGs) in the roots of castor plants under PEG-6000 stress for 3 d and 7 d and after 4 d of rehydration, respectively. GO analysis of the DEGs indicated significant enrichment in antioxidant activity. Furthermore, KEGG enrichment analysis of the DEGs revealed significantly enriched metabolic pathways, including glutathione metabolism, fatty acid metabolism, and plant hormone signal transduction. WGCNA identified the core genes PP2C39 and GA2ox4 in the navajowhite1 module, which was upregulated under PEG-6000 stress. On the basis of these results, we propose a model for the response to drought-induced oxidative stress in castor. Conclusions This study provides valuable antioxidant gene resources, deepening our understanding of antioxidant regulation and paving the way for further molecular breeding of castor plants.

Objective The current study aims to elucidate the critical function of hepatocyte nuclear factor 1-beta (HNF1-β) in lupus nephritis (LN) by investigating its modulation of the Derlin-1/valosin-containing protein (VCP)/VCP-interacting membrane selenoprotein (VIMP) complex, endoplasmic reticulum (ER) stress and podocyte apoptosis.Methods In vitro and in vivo models of LN were established using glomerular podocytes treated with LN serum and MRL/lpr mice, respectively. The expression levels of HNF1-β were analysed in kidney tissues from patients with LN and MRL/lpr mice. To assess the effects of HNF1-β inhibition, an adeno-associated virus vector carrying HNF1-β short hairpin was administered to MRL/lpr mice. In vitro, glomerular podocytes were transfected with HNF1-β small interfering RNA (siRNA) or HNF1-β overexpression plasmids to explore their regulatory effects on the Derlin-1/VCP/VIMP complex and podocyte apoptosis. Dual-luciferase reporter assays and chromatin immunoprecipitation (ChIP) assays were performed to investigate the transcriptional activation of Derlin-1 and VCP promoters by HNF1-β.Results A significant decrease in HNF1-β levels was observed in kidney tissues from patients with LN while MRL/lpr mice exhibited an initial compensatory increase followed by a subsequent decrease in renal HNF1-β expression. Overexpression of HNF1-β transcriptionally upregulated Derlin-1 and VCP mitigating LN serum-induced ER stress and podocyte apoptosis. In contrast, HNF1-β inhibition exacerbated renal dysfunction and structural damage in MRL/lpr mice. Interestingly, HNF1-β inhibition transcriptionally repressed ERP44, leading to calcium ions (Ca²+) release-mediated disruption and inactivation of the Derlin-1/VCP/VIMP complex. This finding suggests that HNF1-β not only regulates the expression of key proteins in the Derlin-1/VCP/VIMP complex but also influences their assembly through Ca²+ release regulation.Conclusion This study provides novel insights into the regulatory mechanisms of HNF1-β in LN emphasising its impact on the Derlin-1/VCP/VIMP complex, ER stress and podocyte apoptosis. These findings have the potential to inform the development of new diagnostic tools and therapeutic strategies for LN.

[Purpose/Significance] Research on the governance system and policy of data elements circulation is an important issue to be solved in the field of data governance in China at present, and research on the policy formulation and governance system of its circulation plays an important role in grasping the security of data circulation in China and promoting the market-oriented allocation of data elements. [Method/Process] First, this study is based on the reality of China's data factor market security and trustworthy, autonomous and controllable requirements. Based on the analysis of the security risk of data circulation, we put forward the data factor market risk governance countermeasures of the "security-fairness-efficiency" triangular structure. Then, based on the three-level system and five-dimensional standards of data factor market governance, we put forward the method of docking the security governance with the trusted ecosystem and the international data governance rule system for cross-border data flow, and constructed a governance system with Chinese characteristics for the national unified data factor market. [Results/Conclusions] Facing the security risks in data sovereignty, data market and data circulation, we should identify and monitor data sovereignty disputes and the operation situation of the circulation market, and establish a multi-party cooperative and joint governance model led by the government, operated by the platform owner, the main body of the enterprise and the participation of users. When assessing the market for data elements, a mixed assessment approach should be adopted, combining qualitative and quantitative aspects, combining expert opinion with objective data, and comparing objectives with results. For different types of data, the control boundaries and scope of use should be clarified in a hierarchical manner, and data ownership, use and income should be clarified; at the same time, a confirmation platform of data rights should be established to audit and register and certify the data service subject, data circulation process, and data circulation rules so as to ensure that the normative nature of data circulation is maintained.

Abstract Bleeding is as particularly a serious phenomenon in Actinidia arguta and has important effects on this plant’s growth and development. Here we used A. arguta to study the effects of bleeding on the growth and development of leaves and fruits after a bleeding episode. We detect and analyze physiological indices of leaves and fruit after bleeding. The result revealed that the relative electrical conductivity and malondialdehyde (MDA) of leaves increased in treatment. Nitro blue tetrazolium chloride (NBT) and 3,3-diaminobenzidine (DAB) staining revealed the accumulation of reactive oxygen species (ROS) in leaves after bleeding. The chlorophyll content and photosynthetic parameter of plants were also decreased. In fruits, pulp and seed water content decreased after the damage, as did fruit vitamin C (Vc), soluble sugar content, and soluble solids content (SSC); the titratable acid content did not change significantly. We therefore conclude that bleeding affects the physiological indices of A. arguta. Our study provides a theoretical basis for understanding the physiological changes of A. arguta after bleeding episodes and laying a timely foundation for advancing research on A. arguta bleeding and long-term field studies should be executed in order to gain insights into underlying mechanisms.

Cupressus gigantea W. C. Cheng & L. K. Fu is an endemic conifer tree species that is distributed widely along the northern portion of the deep gorge of the Yarlung Tsangbo River on the Tibetan Plateau. However, as a key plant species growing on the Tibetan plateau, C. gigantea has since become an endangered species due to habitat loss and degradation, overexploitation, and other factors. It has been listed as a first-grade national protected wild plant species in China. Accordingly, to conserve this plant species, we should obtain more information on its genetic structure. In this study, the genetic diversity and structure among 67 samples were evaluated by the inter-simple sequence repeat (ISSR) technique. Overall, 78 bands were produced with a molecular length of 200 bp to 3100 bp using 10 ISSR primers. The mean values for the average number of alleles (Na), effective number of alleles (Ne), Nei’s gene diversity (H), and Shannon’s information index (I) were 1.529, 1.348, 0.199, and 0.293, respectively. Additionally, the number of polymorphic loci (NPLs) and percentage of polymorphic loci (PPLs) averaged 41.25 and 52.90, respectively. Further, total variation among populations was 14.2%, while that within populations was 85.8%; accordingly, the within-population genetic differentiation was found to be significant (p < 0.001). These results demonstrated that a genetic structure model with K = 3 fitted the data best, which agreed with the unweighted pair group method with arithmetic average (UPGMA) cluster and the principal coordinate analysis (PCoA). These findings are beneficial for ensuring the development and genetic protection of C. gigantea populations in the future.

: As the only woody resurrection plant, Myrothamnus flabellifolia has a strong tolerance to drought and can survive long-term in a desiccated environment. However, the molecular mechanisms related to the stress tolerance of M. flabellifolia are largely unknown, and few tolerance-related genes previously identified had been functionally characterized. WRKYs are a group of unique and complex plant transcription factors, and have reported functions in diverse biological processes, especially in the regulation of abiotic stress tolerances, in various species. However, little is known about their roles in response to abiotic stresses in M. flabellifolia. In this study, we characterized a dehydration-inducible WRKY transcription factor gene, MfWRKY17, from M. flabellifolia. MfWRKY17 shows high degree of homology with genes from Vitis vinifera and Vitis pseudoreticulata, belonging to group II of the WRKY family. Unlike known WRKY17s in other organisms acting as negative regulators in biotic or abiotic stress responses, overexpression of MfWRKY17 in Arabidopsis significantly increased drought and salt tolerance. Further investigations indicated that MfWRKY17 participated in increasing water retention, maintaining chlorophyll content, and regulating ABA biosynthesis and stress-related gene expression. These results suggest that MfWRKY17 possibly acts as a positive regulator of stress tolerance in the resurrection plant M. flabellifolia.

Architectural aesthetics is often studied with a focus on the visual properties of buildings, yet the role of individual aesthetic appreciation abilities in shaping aesthetic pleasure remains overlooked. This study challenges the traditional object-centric perspective by investigating how aesthetic appreciation ability influences architectural aesthetic pleasure through aesthetic judgment. Using Hui-style architecture, a distinctive architectural style originating from the Huizhou region in southern China, as the representative case, a questionnaire survey was conducted with 453 participants. Regression analysis was applied to examine the moderating effects of aesthetic judgment (variety, unity, novelty, and typicality) on the relationship between aesthetic appreciation ability and aesthetic pleasure. The results reveal that individuals with high aesthetic appreciation ability experience greater aesthetic pleasure when exposed to diverse and novel architectural elements. In contrast, those with low aesthetic appreciation ability respond primarily to novelty, showing limited sensitivity to other aesthetic judgments. These findings highlight the nuanced interplay between personal aesthetic capacities and architectural experience, offering valuable insights for artists, architects, and educators seeking to cater to diverse aesthetic preferences and enhance public engagement with architectural beauty. The Cambrian period marks a crucial phase in the initial subduction of the Proto-Tethys Ocean beneath the East Kunlun Orogen. Studying the I-type granites and mafic–ultramafic rocks formed during this period can provide valuable insights into the early Paleozoic tectonic evolution of the region. This paper incorporates petrology, LA-ICP-MS zircon U-Pb geochronology, and whole-rock major and trace element data obtained from the Kekesha intrusion in the eastern section of the East Kunlun Orogen. The formation age, petrogenesis, and magmatic source region of the intrusion are revealed, and the early tectonic evolution process of the subduction of the Proto-Tethys Ocean is discussed. The Kekesha intrusion includes four main rock types: gabbro, gabbro diorite, quartz diorite, and granodiorite. The zircon U-Pb ages are 515.7 ± 7.4 Ma for gabbro, 508.9 ± 9.8 Ma for gabbro diorite, 499.6 ± 4.0 Ma for quartz diorite, and 502.3 ± 9.3 Ma and 501.6 ± 6.2 Ma for granodiorite, respectively, indicating that they were formed in the Middle Cambrian. The geochemical results indicate that the gabbro belongs to the high-Al calc-alkaline basalt series, the gabbro diorite belongs to the medium-high-K calc-alkaline basalt series, the quartz diorite belongs to the quasi-aluminous medium-high-K calc-alkaline I-type granite series, and the granodiorite belongs to the weakly peraluminous calc-alkaline I-type granite series, all of which belong to the medium-high-K calc-alkaline series that have undergone varying degrees of differentiation and contamination. Gabbro and gabbro diorite exhibit significant enrichment in light rare earth elements (LREEs), depletion in heavy rare earth elements (HREEs), and an enhanced negative anomaly in Eu (Europium). Compared to gabbro and gabbro diorite, quartz diorite and granodiorite exhibit more pronounced enrichment in LREEs, more significant depletion in HREEs, and an enhanced negative anomaly in Eu. All four rock types are enriched in large-ion lithophile elements (LILEs) such as Cs, Rb, Th, Ba, and U, and are depleted in high-field-strength elements (HFSEs) such as Nb, Ta, and Ti. This indicates that these rocks originated from the same or similar mixed mantle source regions, and that they are formed in the island-arc tectonic environment. This paper suggests that the gabbro and gabbro diorite are mainly derived from the basic magma formed by partial melting of the lithospheric mantle metasomatized by subducted slab melt in the oceanic crust subduction zone and mixed with a small amount of asthenosphere mantle material. Quartz diorite results from the crystal fractionation of basic magma and experiences crustal contamination during magmatic evolution. Granodiorite forms through the crystal fractionation of basic magma, mixed with partial melting products from quartz diorite. While the lithology of the intrusions differs, their geochemical characteristics suggest they share the same tectonic environment. Together, they record the geological processes associated with island-arc formation in the East Kunlun region, driven by the northward subduction of the Proto-Tethys Ocean during the Early Paleozoic. Based on regional tectonic evolution, it is proposed that the Proto-Tethys Ocean began subducting northward beneath the East Kunlun block from the Middle Cambrian. The Kekesha intrusion formed between 516 and 500 Ma, marking the early stages of Proto-Tethys Ocean crust subduction. [ABSTRACT FROM AUTHOR]

The petrology, geochemistry, and zircon U-Pb chronology of the Huoshaodian pluton in the Liuba area of the western part of the South Qinling tectonic belt are investigated in this study. The Huoshaodian pluton consists of gabbro, quartz diorite, and granodiorite, and the dominated rock type is quartz diorite. The results indicate that the Huoshaodian pluton belongs to the calc-alkaline series. In the chondrite-normalized REE, all of the samples showed similar patterns, with an enrichment of light REEs and depletion of heavy REEs, but they showed slight differences in the degrees of Eu anomalies. The primitive mantle-normalized trace element diagram reveals an enrichment of large-ion lithophile elements (LILEs) and light rare earth elements (LREEs), as well as depleted high field strength elements (HFSEs). The zircon U-Pb dating results reveal that the gabbro, quartz diorite, and granodiorite have crystallization ages of 214.9 ± 0.58 Ma, 215.0 ± 1.2 Ma, and 215.4 ± 1.9 Ma, respectively, indicating that the Huoshaodian pluton was emplaced during the late Triassic period (214.9–215.4 Ma). In terms of petrogenesis, the gabbro of the Huoshaodian pluton originates from a transitional lithospheric mantle that has undergone fluid metasomatism and partial melting. Specifically, it originated through 1%–2% garnet spinel peridotite undergoing partial melting. In addition, the gabbro underwent a slight degree of contamination by crustal materials during its ascent and intrusion, with some continental crust material being incorporated. The quartz diorite and granodiorite of the Huoshaodian pluton are formed through partial melting processes occurring within the normal lower crust. Combined with the previous studies on the early Mesozoic tectonic evolution of the South Qinling, this study proposes that the formation mechanism of the Huoshaodian pluton may be as follows: in the early Triassic, the Mianlue Ocean subducted northward beneath the Qinling microblock, resulting in a large-scale continental-continental collision between the North China Block and the Yangtze Block; when the oceanic crust subducted to a certain depth, the detachment of the subducting slab triggered the upwelling of mantle material. The heat from mantle-derived magma caused the partial melting of the mafic lower crust, while the mafic magma entered into the upper granitic magma chamber and began to mix. Due to the high viscosity contrast and temperature difference between the two end-member magmas, incomplete mixing led to the formation of a melt with distinct adakitic characteristics and a mafic melt representing mantle-derived material. Subgroup analysis revealed no significant difference in intraoperative hypoxemia during OLV between respondents who performed lung-protective ventilation strategies and those who did not. Conclusions: Lung-protective ventilation strategies during OLV have been widely accepted in mainland China and are strongly recommended for esophageal surgery, particularly in tier 2 hospitals. The formation of early Mesozoic granitoid plutons in the Qinling Orogen is widely regarded as a result of the collision and accretion between the Yangtze Block and the South Qinling Block during the early Mesozoic, but the specific magmatic process, source composition, tectonic environment and deep dynamic background remain controversial. This study reports the petrology, zircon U–Pb geochronology, and whole-rock geochemistry of diorites from the Liuba and Qingyangyi plutons in the South Qinling, to provide new evidence for understanding the final collision tectonic evolution process of Qinling Orogenic belt. The Liuba and Qingyangyi plutons, located in the central part of the South Qinling region, are primarily composed of quartz diorite and quartz monzodiorite, respectively. The results indicate that the weighted mean crystallization ages of the quartz diorite in the Liuba pluton range from 216.1 ± 0.8 Ma to 217.1 ± 1.3 Ma, with the weighted mean crystallization ages of its MMEs being 215.4 ± 1.0 Ma. The crystallization ages of the quartz monzodiorite in the Qingyangyi pluton range from 214.6 ± 0.9 Ma to 215.4 ± 0.9 Ma, suggesting that both plutons were formed in the late Triassic. The investigated plutons are characterized as right-leaning and have weak negative Eu anomalies on the chondrite-normalized REE patterns diagram. The large ion lithophile elements (LILE) Rb, Ba, Th and K are relatively enriched, while high-field strength elements (HFSE) Nb, Ta, Ti and P are strongly depleted. The formation of numerous MMEs in the Liuba pluton is the product of magmatic mixing. The Liuba and Qingyangyi plutons are the results of crust thickening and partial melting of lower crust caused by the comprehensive late Triassic collision between the Yangtze Block and the North China Block (NCB), and are the manifestation of magmatic intrusion along the South Qinling tectonic belt in the late Triassic period. The connection between the Ordovician bentonites on the southern margin of the Ordos Basin and the Early Palaeozoic volcanic rocks of the North Qinling Orogenic Belt is crucial for understanding the subduction and collisional closure of the Shangdan Ocean during the Early Palaeozoic. This paper investigates zircon U–Pb ages, geochemistry and Lu–Hf isotopic compositions of zircons in the Upper Ordovician Zhaolaoyu Formation bentonites located on the southern margin of the Ordos Basin. U–Pb dating of zircon indicates a coeval age of 453.3 ± 1.4 Ma (MSWD = 0.99), which represents the crystallisation age during the Late Ordovician Katian stage. The bentonites exhibit higher SiO2 (57.94–77.95 wt.%) and Al2O3 (9.21–14.33 wt.%), classifying them within the low‐potassium alkali basalt to medium‐potassium calc‐alkaline series. The parent rock of the bentonites is likely intermediate to felsic volcanic rocks. The rare earth element partitioning curves of the bentonites are right‐dipping, with a more pronounced negative Eu anomaly (δEu = 0.48–0.67). The zircons in the bentonites yield two‐stage model ages ranging from 546 to 956 Ma, along with εHf(t) values between 5.56 and 13.55. These results indicate that the bentonites are products of volcanic arc magma formed in a subduction–collision environment. The interbedded bentonites in the Upper Ordovician limestones of the southern margin of the Ordos Basin may be associated with the northward subduction of the Shangdan Oceanic crust, reflecting the subduction and consumption of the Proto‐Tethys Ocean along the southern margin of the North China Block. Fatigue failure is one of the most important reasons for the failure of engineering application components. However, due to the high cost of fatigue experiments, it is necessary to use mechanical properties to predict fatigue strength. Based on the true stress-strain curves, a novel model for fatigue strength prediction is established and is used to calculate the fatigue strength. The strength predicted is compared with that calculated by the staircase method and the Basquin equation. The results show that the model could obtain the fatigue strength of the materials only by using tensile strength and work-hardening strength, and it is suitable for other steels, which greatly saves costs and increases accuracy.

miR-30b, which is encoded by the gene located on chromosome 8q24.22, plays an important role in a variety of diseases. In most types of tumors, miR-30b significantly inhibits the proliferation, migration, and invasion of cancer cells through the regulation of target genes. Moreover, miR-30b can inhibit the PI3K/AKT signaling pathway through the regulation of EGFR, AKT, Derlin-1, GNA13, SIX1, and other target genes, thus inhibiting the EMT process of tumor cells and promoting apoptosis. In addition, miR-30 plays a significant role in alleviating drug resistance in tumor cells. Although the use of miR-30b as a clinical diagnostic indicator or anticancer drug is still facing great difficulties in the short term, with the deepening of research, the potential application of miR-30b is emerging.

BackgroundProfessional certification of community health workers (CHWs) is a debated topic. Although intended to promote CHWs, certification may have unintended impacts given the grassroots nature of the workforce. As such, both intended effects and unintended adverse effects should be carefully evaluated. However, there is a lack of published literature describing such effective evaluations with a robust methodology. In this methods paper, we describe a key component of evaluating CHW certification in Massachusetts-the Massachusetts CHW Workforce Survey.MethodsDesign of the surveys was informed by a program theory framework that delineated both positive and negative potential impacts of Massachusetts CHW certification on CHWs and CHW employers. Using this framework, we developed measures of interest and preliminary CHW and CHW employer surveys. To validate and refine the surveys, we conducted cognitive interviews with CHWs and CHW employers. We then finalized survey tools with input from state and national stakeholders, CHWs, and CHW employers. Our sample consisted of three frames based on where CHWs are most likely to be employed in Massachusetts: acute care hospitals, community-based organizations, and ambulatory care health centers, primarily community health centers and federally qualified health centers. We then undertook extensive outreach efforts to determine whether each organization employed CHWs and to obtain CHW and CHW employer contact information. Our statistical analysis of the data utilized inverse probability score weighting accounting for organizational, site, and individual response.Anticipated resultsWave one of the survey was administered in 2016 prior to launch of Massachusetts CHW certification and wave two in 2021. We report descriptive statistics of the three sample frames and response rates of each survey for each wave. Further, we describe select anticipated results related to certification, including outcomes of the program theory framework.ConclusionsThe Massachusetts CHW Workforce Survey is the culmination of 5 years of effort to evaluate the impact of CHW certification in Massachusetts. Our comprehensive description of our methodology addresses an important gap in CHW research literature. The rigorous design, administration, and analysis of our surveys ensure our findings are robust, valid, and replicable, which can be leveraged by others evaluating the CHW workforce.

Integrating biosensors with smartphones is becoming an increasingly popular method for detecting various biomolecules and could replace expensive laboratory-based instruments. In this work, we demonstrate a novel smartphone-based biosensor system with a gradient grating period guided-mode resonance (GGP-GMR) sensor. The sensor comprises numerous gratings which each correspond to and block the light of a specific resonant wavelength. This results in a dark band, which is observed using a CCD underneath the GGP-GMR sensor. By monitoring the shift in the dark band, the concentration of a molecule in a sample can be determined. The sensor is illuminated by a light-emitting diode, and the light transmitted through the GGP-GMR sensor is directly captured by a smartphone, which then displays the results. Experiments were performed to validate the proposed smartphone biosensor and a limit of detection (LOD) of 1.50 × 10−3 RIU was achieved for sucrose solutions. Additionally, multiplexed detection was demonstrated for albumin and creatinine solutions at concentrations of 0–500 and 0–1 mg/mL, respectively; the corresponding LODs were 1.18 and 20.56 μg/mL.

A hydraulic engine mount (HEM) is an advanced tuned mass damper (TMD) system used to isolate the noise and vibration from the engine to the chassis. This paper aims to design a semiactive HEM based on the TMD model, which only tunes damping without affecting the other lumped parameters of the TMD system. Firstly, the dynamic equations of an HEM modeled as a TMD system are derived based on the linear lumped parameter model (LPM). Each lumped parameter of the HEM is analyzed to identify the relevant parameters that affect damping. Secondly, a newly designed semiactive HEM design is proposed that utilizes a helical moving plate to simultaneously control both the inertia track area and length, resulting in precise damping tuning. To illustrate the dynamic performance of the newly designed HEM, calculations are presented based on various parameters for its tunable damping range and dynamic stiffness spectrum. Additionally, to demonstrate the performance of vibration isolation, this paper determines the optimal length and dynamic stiffness of the inertia track to minimize the transmissibility. Thirdly, to reveal the superiority of the newly designed HEM over the MR fluid mount, an example is presented where the MR fluid medium is used in place of conventional hydraulic fluid in the HEM while keeping all other parameters constant. Specifically, the novel semiactive HEM employs conventional hydraulic fluid and is spiral-driven by a moving plate while the MR fluid-based HEM is controlled by an additional tunable magnetic intensity controller. The nonlinear LPM of the newly designed HEM is verified by comparing the dynamic stiffness spectrum with the experimental results in the published literature. Then, the nonlinear LPM of the MR fluid mount is established, and its dynamic stiffness spectrum is calculated and compared with that of the newly designed HEM. The results indicate that the newly designed HEM and MR fluid mount have similar ranges of dynamic stiffness control, but the newly designed HEM does not require expensive MR/ER fluid or additional continuous external energy input to regulate the dynamic stiffness. Moreover, when using inexpensive low-viscosity hydraulic fluid, the newly designed HEM can provide a wider range of dynamic stiffness control.

Introduction: This study investigated the potential link between serum klotho levels and cognitive function in patients with non-alcoholic fatty liver disease (NAFLD). Materials and Methods: Utilizing NHANES data from 2011 to 2014, the research included 356 eligible participants. NAFLD was identified with the United States Fatty Liver Index (US-FLI), and cognition was measured by various tests including the Animal Fluency Test (AFT), Digit Symbol Substitution Test (DSST), Immediate Recall Test (IRT), and Delayed Recall Test (DRT). Weighted logistic regression and restricted cubic splines were employed to analyze the relationship between klotho levels and cognitive scores. Results: A significant nonlinear association was observed between klotho levels and the performance in DSST and Delayed Recall Test (DRT). After controlling for confounding factors, the study found a positive association between higher serum klotho levels and improved cognitive performance in both AFT and DSST. However, there was no significant relationship between klotho levels and the IRT or DRT, regardless of whether the natural logarithm or quartile was considered. Discussion: The findings suggest that a higher serum klotho level may be positively correlated with better cognitive performance in NAFLD patients. [ABSTRACT FROM AUTHOR]

For selectively sensing Tryptophan(Trp), the fluorescent Tb3+@UiO-66-(COOH)2 was prepared through loading Tb3+ into UiO-66-(COOH)2 via post-synthesis method. Transmission electron microscope (TEM), X-ray powder diffractometer (XRD), specific surface area analyzer, and X-ray photoelectron spectroscope (XPS) were used to analysize the morphology, composition and pore characteristics of the porous materials. Meanwhile, ultraviolet-visible spectrophotometer (UV-Vis) and fluorescence spectrophotometer were used to study the fluorescence characteristics. The results show that the energy transfer from organic ligand to Tb3+ leads to a bright green fluorescence. Among the 13 amino acids, only Tryptophan has obvious fluorescence quenching effect. This is because there appears a large overlap between UV-Vis absorption spectrum of Trp and excitation spectrum of the MOF, which indicates a strong competition effect for UV light. The absorption to UV light of Tb3+@UiO-66-(COOH)2 is decreased by Trp,leading to the weak fluorescence. Further investigation indicates the material can sensitively sense Trp (LOD, 5.53 μmol/L) and owns excellent anti-interference ability from other co-existing amino acids. Thus, this work not only demonstrates Tb3+@UiO-66-(COOH)2 can serve as a selective and sensitive sensor for Trp, but also provides a guideline for designing novel sensors for amino acids in the future.

Autism spectrum disorder (ASD) is a group of heterogeneous neurodevelopmental disorders characterized by impairments activities without efficient pharmacological therapies in social interaction, speech and stereotypic patterns. Clinical studies have shown the efficacy of acupuncture as an alternative therapy for autism. The effectiveness of acupuncture as an alternative treatment for autism has been demonstrated through clinical trials. However, the molecular mechanisms that underlie these effects remain unclear. Due to its profound pro-social, anxiolytic, stress management effects, and its potential use for the treatment of psychiatric disorders associated with altered socioemotional competence, oxytocin (OT) released from the hypothalamus has attracted considerable interest. In the past decade, a number of clinical and animal studies have shown that OT administration effectively reduces core symptoms of ASD, especially social behavior deficits. Recently, the endocannabinoid system has emerged as a promising target for the treatment of autism. OT was found to facilitate the endocannabinoid-mediated social reward processes in the nucleus accumbens of the mouse brain. Furthermore, serotonin and dopamine are involved in the reward response mediated by OT. In view of these findings, we conclude that acupuncture may produce therapeutic effects on autism by triggering the hypothalamic oxytocin system, which in turn activates the release of neurotransmitters such as endocannabinoids, dopamine and serotonin. This would be a valuable guide for further research on the mechanism of treatment of autism with acupuncture.

Introduction Kidney stone disease is common, with a lifetime risk in the United States of about 19% in men and 9% in women (1). Studies of families (2), twin concordance [...], The major risk factor for kidney stone disease is idiopathic hypercalciuria. Recent evidence implicates a role for defective calcium reabsorption in the renal proximal tubule. We hypothesized that claudin-2, a paracellular cation channel protein, mediates proximal tubule calcium reabsorption. We found that claudin-2-null mice have hypercalciuria due to a primary defect in renal tubule calcium transport and papillary nephrocalcinosis that resembles the intratubular plugs in kidney stone formers. Our findings suggest that a proximal tubule defect in calcium reabsorption predisposes to papillary calcification, providing support for the vas washdown hypothesis. Claudin-2-null mice were also found to have increased net intestinal calcium absorption, but reduced paracellular calcium permeability in the colon, suggesting that this was due to reduced intestinal calcium secretion. Common genetic variants in the claudin-2 gene were associated with decreased tissue expression of claudin-2 and increased risk of kidney stones in 2 large population-based studies. Finally, we describe a family in which males with a rare missense variant in claudin-2 have marked hypercalciuria and kidney stone disease. Our findings indicate that claudin-2 is a key regulator of calcium excretion and a potential target for therapies to prevent kidney stones.
