Introduction: Dural arteriovenous fistulae (dAVF) are rare acquired intracranial vascular malformations with clinical manifestations varying from asymptomatic lesions, non-hemorrhagic neurological disorders, and intracranial hemorrhage which may be fatal. In addition, patients with dAVF often have cognitive impairments. This study assessed whether endovascular embolization improves cognitive performance in patients with dAVF. Methods: NAIF was a prospective study including all the patients with a clinical diagnosis of dAVF (excluding those with acute intracranial hemorrhage secondary to venous hypertension) who underwent successful embolization at 5 hospitals in Spain between January 4, 2018 and January 7, 2020. Patients were categorized by risk of hemorrhage (Cognard classification 1-2A and 2B-4) via angiography. A neuropsychological evaluation covering 5 domains of cognitive function (memory, attention and executive functions, language, praxis, gnosis) was completed at baseline and a 3-month follow-up. Paired sample t-tests were used to compare mean Z-scores for cognitive tests pre- and post-treatment, where higher Z-scores signify better cognitive status. Results: A preliminary group of 15 patients (mean age 60, [range 23-83 years], 4 [26.7%] female) were included. Six patients presented with low hemorrhagic risk (Cognard1-2A) and 9 with high hemorrhagic risk (Cognard 2B-4). In the post-treatment neuropsychological evaluation, patients exhibited improved performance in executive function in the Semantic and Phonetic Fluency test (-0.57 vs. 0.21; p=0.01) and increased performance in Verbal Memory Learning (-1.45 vs. -0.82; p=0.014) relative to baseline. The remaining cognitive evaluations did not show significant differences, although there was a trend of language improvement in the Boston naming test (0.95 vs. 1.12; p=0.093). Improvements were observed in both high- and low-risk patients, and no patients experienced cognitive worsening after the intervention. Conclusion: This study suggests that endovascular embolization imparts cognitive benefits for dAVF patients undergoing endovascular embolization, and may be beneficial even for patients with a low risk of hemorrhage.

Actualmente, el impacto de la industria de la construcción está siendo una de las causas originarias del agotamiento de recursos no renovables además de la generación de grandes cantidades de residuos y de las emisiones de dióxido de carbono en la atmósfera. En interés de una estrategia de construcción más sostenible, en los últimos años se ha investigado exhaustivamente el uso de fibras vegetales, residuos y materiales reciclados. Como una buena opción, las fibras de residuos textiles junto con fibras vegetales se pueden utilizar como refuerzo interno en materiales con una matriz de cemento con el fin de mejorar su ductilidad y resistencia a las fisuras. Sin embargo, todos los estudios llevados a cabo hasta ahora han sido para placas planas, así pues, también es necesario analizar cuáles son las características en piezas curvadas. El objetivo de este trabajo es definir las características mecánicas de piezas curvadas de cemento reforzadas con no tejidos de residuo textil y fibras vegetales. Además de determinar la influencia del número de capas de no tejido y el grado de curvatura en sus propiedades. También se ha analizado el comportamiento del material en simulaciones numéricas. Para lograr el objetivo se elaboraron piezas con cuatro diámetros diferentes: 9, 16, 20 y 32 cm; y un reforzamiento con cuatro o seis capas de no tejido. Posteriormente, se analizaron sus propiedades mecánicas con un ensayo de flexión. Por otra parte, se compararon las tensiones calculadas con las ecuaciones descritas para placas planas y los resultados obtenidos con simulaciones numéricas atendiendo a la geometría de las piezas. Los resultados indicaron que las piezas de cemento reforzadas con seis capas de no tejido presentan mejores propiedades mecánicas que las reforzadas con cuatro capas. Respecto al grado de curvatura, se han observado diferencias significativas; las piezas de diámetro de 20 cm con seis capas han dado las tensiones más altas de entre todo el conjunto. The aim of this work is to define the mechanical characteristics of curved concrete pieces reinforced with textile waste nonwovens and vegetal fibres. Moreover, it also aims to determine the influence of the number of nonwoven layers as well as the degree of curvature on their properties. The behaviour of the material has also been analysed in numerical simulations. In order to reach the objective, there were prepared composites with four different diameters: 9, 16, 20 and 32 cm; and reinforced with four or six layers of non-woven. Subsequently, their mechanical properties were analysed with a bending test. On the other hand, stresses values were compared depending if they were calculated with the equations described for flat plates or with numerical simulations according to their geometry. The results indicated that concrete pieces reinforced with six nonwoven layers have better mechanical properties than those reinforced with four layers. Significant differences were observed in relation to the degree of curvature; the pieces with a diameter of 20 cm with six layers gave the highest stresses among all the pieces. - Abstract
Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE epsilon 4 allele. Meta-analysis of genome-wide association studies on Alzheimer's disease and related dementias identifies new loci and enables generation of a new genetic risk score associated with the risk of future Alzheimer's disease and dementia.

Actualmente, el impacto de la industria de la construcción está siendo una de las causas originarias del agotamiento de recursos no renovables además de la generación de grandes cantidades de residuos y de las emisiones de dióxido de carbono en la atmósfera. En interés de una estrategia de construcción más sostenible, en los últimos años se ha investigado exhaustivamente el uso de fibras vegetales, residuos y materiales reciclados. Como una buena opción, las fibras de residuos textiles junto con fibras vegetales se pueden utilizar como refuerzo interno en materiales con una matriz de cemento con el fin de mejorar su ductilidad y resistencia a las fisuras. Sin embargo, todos los estudios llevados a cabo hasta ahora han sido para placas planas, así pues, también es necesario analizar cuáles son las características en piezas curvadas. El objetivo de este trabajo es definir las características mecánicas de piezas curvadas de cemento reforzadas con no tejidos de residuo textil y fibras vegetales. Además de determinar la influencia del número de capas de no tejido y el grado de curvatura en sus propiedades. También se ha analizado el comportamiento del material en simulaciones numéricas. Para lograr el objetivo se elaboraron piezas con cuatro diámetros diferentes: 9, 16, 20 y 32 cm; y un reforzamiento con cuatro o seis capas de no tejido. Posteriormente, se analizaron sus propiedades mecánicas con un ensayo de flexión. Por otra parte, se compararon las tensiones calculadas con las ecuaciones descritas para placas planas y los resultados obtenidos con simulaciones numéricas atendiendo a la geometría de las piezas. Los resultados indicaron que las piezas de cemento reforzadas con seis capas de no tejido presentan mejores propiedades mecánicas que las reforzadas con cuatro capas. Respecto al grado de curvatura, se han observado diferencias significativas; las piezas de diámetro de 20 cm con seis capas han dado las tensiones más altas de entre todo el conjunto. Por lo que respecta a la metodología de cálculo de las tensiones, los resultados en las simulaciones son superiores a los de las ecuaciones. Currently, the construction industry's impact is one of the causes of the exhaustion of non-renewable resources as well as the generation of large amounts of waste and carbon dioxide emissions into the atmosphere. For the sake of a more sustainable construction strategy, the usage of vegetal fibres, waste and recycled materials has been extensively researched in the recent years. As a good option, textile waste fibres along with vegetal fibres can be used as internal reinforcement in materials with a cementitious matrix to improve their ductility and crack resistance. However, all studies carried out so far have been for flat plates, so it is necessary to analyse the characteristics of curved pieces. The aim of this work is to define the mechanical characteristics of curved concrete pieces reinforced with textile waste nonwovens and vegetal fibres. Moreover, it also aims to determine the influence of the number of nonwoven layers as well as the degree of curvature on their properties. The behaviour of the material has also been analysed in numerical simulations. In order to reach the objective, there were prepared composites with four different diameters: 9, 16, 20 and 32 cm; and reinforced with four or six layers of non-woven. Subsequently, their mechanical properties were analysed with a bending test. On the other hand, stresses values were compared depending if they were calculated with the equations described for flat plates or with numerical simulations according to their geometry. The results indicated that concrete pieces reinforced with six nonwoven layers have better mechanical properties than those reinforced with four layers. Significant differences were observed in relation to the degree of curvature; the pieces with a diameter of 20 cm with six layers gave the highest stresses among all the pieces. Background: The objective of the present study is to describe the characteristics of interstitial pneumonia with autoimmune features (IPAF) patients, to assess the incidence rate of functional respiratory impairment over time and to evaluate the influence of therapeutic alternatives on the prognosis of these patients. Methods: A longitudinal observational multicenter study was performed (NEREA registry). It was carried out by a multidisciplinary team in seven Hospitals of Madrid. Patients were included from IPAF diagnosis. Main outcome: poor prognosis as functional respiratory impairment (relative decline in FVC % defined as ≥ 5% every 6 months). Covariates: therapy, sociodemographic, clinical, radiological patterns, laboratory and functional tests. Statistics: Survival techniques were used to estimate IR per 100 patients-semester with their 95% confidence interval [CI]. The influence of covariates in prognosis were analyzed through cox multivariate regression models (hazard ratio (HR) and [CI]). Results: 79 IPAF were included, with a mean and a maximum follow-up of 3.17 and 12 years respectively. Along the study, 77.2% received treatment (52 glucocorticoids, 25 mycophenolate, 21 azathioprine, 15 rituximab and 11 antifibrotics). IR was 23.9 [19.9–28.8], and 50% of IPAF developed functional respiratory impairment after 16 months from its diagnosis. Multivariate analysis: usual interstitial pneumonia (UIP) had poorer prognosis compared to non-specific interstitial pneumonia (NSIP) (p = 0.001). In NSIP, positive ANA, increased the risk of poor prognosis. In UIP, glucocorticoids (HR: 0.53 [0.34–0.83]), age (HR: 1.04 [1.01–1.07]), and Ro-antibodies (HR: 0.36 [0.19–0.65]) influenced the prognosis. Conclusions: IPAF have functional impairment during the first years of disease. Factors predicting deterioration differ between radiographic patterns. Our real-life study suggests the potential benefit of particular therapies in IPAF. [ABSTRACT FROM AUTHOR]

- Abstract
Background: Today, a growing number of individuals with mild cognitive impairment (MCI) wish to assess their risk of developing Alzheimer's disease (AD) dementia. The expectations as well as the effects on quality of life (QoL) in MCI patients and their close others through biomarker-based dementia risk estimation are not well studied. Objective: The PreDADQoL project aims at providing empirical data on effects of such prediction on QoL and at developing an ethical and legal framework of biomarker-based dementia risk estimation in MCI. Methods: In the empirical study, 100 MCI-patients and their close others will be recruited from two sites (Germany and Spain). They receive standardized counselling on cerebrospinal fluid (CSF) biomarker-based prediction of AD dementia and a risk disclosure based on their AD biomarker status. A mixed methods approach will be applied to assess outcomes. Results: The pilot-study yielded a specification of the research topics and newly developed questionnaires for the main assessment. Within this binational quantitative and qualitative study, data on attitudes and expectations toward AD risk prediction, QoL, risk communication, coping strategies, mental health, lifestyle changes, and healthcare resource utilization will be obtained. Together with the normative part of the project, an empirically informed ethical and legal framework for biomarker-based dementia risk estimation will be developed. Conclusion: The empirical research of the PreDADQoL study together with the ethical and legal considerations and implications will help to improve the process of counselling and risk disclosure and thereby positively affect QoL and health of MCI-patients and their close others in the context of biomarker-based dementia risk estimation.

- Abstract
Genetic discoveries of Alzheimer’s disease are the drivers of our understanding, and together with polygenetic risk stratification can contribute towards planning of feasible and efficient preventive and curative clinical trials. We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery n = 409,435 and validation size n = 58,190). Here, we add six variants associated with Alzheimer’s disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer’s disease patients in APOE ɛ4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer’s disease.

Objective Methods Results Significance Pathological amyloid‐β (Aβ) accumulation and hyperphosphorylated tau proteins have been described in resected temporal lobe specimens of epilepsy patients. We aimed to determine cerebrospinal fluid (CSF) Aβ1‐42 and p181‐tau levels and cerebral Aβ deposits on positron emission tomography (Aβ PET) and correlate these findings with cognitive performance in adults with drug‐resistant temporal lobe epilepsy (TLE).In this cross‐sectional study, we enrolled individuals with drug‐resistant TLE who were 25–55 years old. Each participant underwent 18F‐flutemetamol PET, determination of CSF Aβ1‐42, p181‐tau, and total tau, and a comprehensive neuropsychological assessment. We evaluated normalized standard uptake value ratios (SUVRs) for different brain regions on Aβ PET.Thirty patients (mean age = 41.9 ± SD 8.1 years, 57% men) were included. The median disease duration was 9.5 (interquartile range = 4–24) years. Twenty‐six patients (87%) had a clinically significant cognitive impairment on neuropsychological evaluation, 18 (69%) of the amnesic type. On Aβ PET, high uptake was observed in both mesial temporal regions (ipsilateral: SUVR z‐score = .90, 95% confidence interval [CI] = .60–1.20; contralateral: SUVR z‐score = .92, 95% CI = .57–1.27; p < .001), which was higher when compared to SUVR z‐scores in all the remaining regions (p < .001) and in the ipsilateral anterior cingulate (SUVR z‐score = .27, 95% CI = .04–.49, p = .020). No significant deposition was observed in other regions. Seven patients (23%) had low Aβ1‐42 levels, and two (7%) had elevated p181‐tau levels in CSF. Higher p181‐tau levels correlated with poorer verbal fluency (R = −.427, p = .044).Our findings reveal a considerable Aβ deposition in mesial temporal regions and ipsilateral anterior cingulate among adults with drug‐resistant TLE. Additionally, abnormal CSF Aβ1‐42 levels were observed in a significant proportion of patients, and p181‐tau levels were associated with verbal fluency. These results suggest that markers of neuronal damage can be observed in adults with TLE, warranting further investigation. [ABSTRACT FROM AUTHOR]

- Abstract
The role of genetic risk markers for Alzheimer's disease (AD) in mediating the neurocognitive endophenotypes (NEs) of subjects with mild cognitive impairment (MCI) has rarely been studied. The aim of the present study was to investigate the relationship between well-known AD-associated single-nucleotide polymorphisms (SNPs) and individual NEs routinely evaluated during diagnosis of MCI, AD, and other dementias. The Fundacio ACE (ACE) dataset, comprising information from 1245 patients with MCI, was analyzed, including the total sample, amnestic MCI (aMCI) (n = 811), and non-amnestic MCI (naMCI) (n = 434). As probable-MCI (Pr-MCI) patients with memory impairment have a higher risk of AD, which could influence the statistical power to detect genetic associations, the MCI phenotype was also stratified into four related conditions: Pr-aMCI (n = 262), Pr-naMCI (n = 76), possible (Pss)-aMCI (n = 549), and Pss-naMCI (n = 358). Validation analyses were performed using data from the German study on Aging, Cognition, and Dementia in primary care patients (AgeCoDe), and the German Dementia Competence Network (DCN). SNP associations with NEs were calculated in PLINK using multivariate linear regression analysis adjusted for age, gender, and education. In the total MCI sample, APOE-epsilon 4 was significantly associated with the memory function NEs delayed recall (DR) (beta = -0.76, p = 4.1 x 10(-10)), learning (beta = -1.35, p = 2.91 x 10(-6)), and recognition memory (beta = -0.58, p = 9.67 x 10(-5)); and with DR in the aMCI group (beta = -0.36, p = 2.96 x 10(-5)). These results were confirmed by validation in the AgeCoDe (n = 503) and DCN (n = 583) datasets. APOE-epsilon 4 was also significantly associated with the NE learning in individuals classified as having Pss-aMCI (beta = -1.37, p = 5.82 x 10(-5)). Moreover, there was a near study-wide significant association between the HS3ST1 locus (rs6448799) and the backward digits working memory NE (beta = -0.52, p = 7.57 x 10(-5))

To determine whether lower performance on executive function tests in subjective cognitive decline (SCD) individuals are associated with higher levels of brain amyloid beta (Aβ) deposition and regional volumetric reduction in areas of interest for Alzheimer's disease (AD). 195 individuals with SCD from the FACEHBI study were assessed with a neuropsychological battery that included the following nine executive function tests: Trail Making Test A and B (TMTA, TMTB), the Rule Shift Cards subtest of BADS, the Automatic Inhibition subtest of the Syndrom Kurz Test (AI-SKT), Digit Span Backwards and Similarities from WAIS-III, and the letter, semantic, and verb fluency tests. All subjects underwent an 18F-Florbetaben positron emission tomography (FBB-PET) scan to measure global standard uptake value ratio (SUVR), and a magnetic resonance imaging (MRI). A multiple regression analysis, adjusted for age, was carried out to explore the association between global SUVR and performance on executive tests. Then, on those tests significantly associated with amyloid burden, a voxel-based morphometry (VBM) analysis was carried out to explore their correlates with grey matter volume. Multiple regression analysis revealed a statistically significant association between Aβ deposition and performance on one of the executive tests (the AI-SKT). Moreover, VBM analysis showed worse AI-SKT scores were related to lower volume in bilateral hippocampus and left inferior frontal regions. In conclusion, in SCD individuals, worse automatic inhibition ability has been found related to higher cerebral Aβ deposition and lower volume in the hippocampus and frontal regions. The MAPT H1 haplotype has been linked to several disorders, but its relationship with Alzheimer's disease (AD) remains controversial. A rare variant in MAPT (p.A152T) has been linked with frontotemporal dementia (FTD) and AD. We genotyped H1/H2 and p.A152T MAPT in 11,572 subjects from Spain (4,327 AD, 563 FTD, 648 Parkinson's disease (PD), 84 progressive supranuclear palsy (PSP), and 5,950 healthy controls). Additionally, we included 101 individuals from 21 families with genetic FTD. MAPT p.A152T was borderline significantly associated with FTD [odds ratio (OR)=2.03; p=0.063], but not with AD. MAPT H1 haplotype was associated with AD risk (OR=1.12; p=0.0005). Stratification analysis showed that this association was mainly driven by APOE ε4 noncarriers (OR=1.14; p=0.0025). MAPT H1 was also associated with risk for PD (OR=1.30; p=0.0003) and PSP (OR=3.18; p=8.59 × 10-8) but not FTD. Alzheimer’s disease (AD) is a severe and incurable neurodegenerative disease, and the failure to find effective treatments suggests that the underlying pathology remains poorly understood. Due to its strong heritability, deciphering the genetic landscape of AD and related dementia (ADD) is a unique opportunity to advance our knowledge. We completed a meta-analysis of genome-wide association studies (39,106 clinically AD-diagnosed cases, 46,828 proxy-ADD cases and 401,577 controls) with the most promising signals followed-up in 25,392 independent AD cases and 276,086 controls. We report 75 risk loci for ADD, including 42 novel ones. Pathway-enrichment analyses confirm the involvement of amyloid/Tau pathways, highlight the role of microglia and its potential interaction with APP metabolism. Numerous genes exhibited differential expression or splicing in AD-related conditions and gene prioritization implies EGFR signaling and TNF-α pathway through LUBAC complex. We also generated a novel polygenic risk score strongly associated with the risk of future dementia or progression from mild cognitive impairment to dementia. In conclusion, by more than doubling the number of loci associated with ADD risk, our study offers new insights into the pathophysiological processes underlying AD and offers additional therapeutic entry-points and tools for translational genomics.

Introduction: Understanding the impact of biomarker-based dementia risk estimation in people with mild cognitive impairment (MCI) and their care partners is critical for patient care., Methods: MCI patients and study partners were counseled on Alzheimer's disease (AD) biomarker and dementia risk was disclosed. Data on mood, quality of life (QoL), and satisfaction with life (SwL) were obtained 1 week and 3 months after disclosure., Results: Seventy-six dyads were enrolled, and two-thirds of the patients opted for biomarker testing. None of the participants experienced clinically relevant depression or anxiety after disclosure. All dyads reported moderate to high QoL and SwL throughout the study. Patients reported more subthreshold depressive symptoms 1 week and lower QoL and SwL 3 months after disclosure. In patients, depression (odds ratio [OR]: 0.76) and anxiety (OR: 0.81) were significant predictors for the decision against biomarker testing., Discussion: No major psychological harm is to be expected in MCI patients and care partners after dementia risk disclosure., Trial Registration: This study is registered in the German clinical trials register (Deutsches Register Klinischer Studien, DRKS): http://www.drks.de/DRKS00011155, DRKS registration number: DRKS00011155, date of registration: 18.08.2017., Highlights: Patients with mild cognitive impairment (MCI) and study partners were counseled on Alzheimer's disease (AD) biomarker-based dementia risk estimation. - Abstract
Background: Optical coherence tomography (OCT) of the retina is a fast and easily accessible tool for the quantification of retinal structural measurements. Multiple studies show that patients with Alzheimer's disease (AD) exhibit thinning in several retinal layers compared to age-matched controls. Subjective cognitive decline (SCD) has been proposed as a risk factor for progression to AD. There is little data about retinal changes in preclinical AD and their correlation with amyloid-β (Aβ) uptake., Aims: We investigated the association of retinal thickness quantified by OCT with Aβ accumulation and conversion to mild cognitive impairment (MCI) over 24 months in individuals with SCD., Methods: One hundred twenty-nine individuals with SCD enrolled in Fundació ACE Healthy Brain Initiative underwent comprehensive neuropsychological testing, OCT scan of the retina and florbetaben (FBB) positron emission tomography (PET) at baseline (v0) and after 24 months (v2). We assessed the association of sixteen retinal thickness measurements at baseline with FBB-PET status (+/-) and global standardize uptake value ratio (SUVR) as a continuous measure at v0 and v2 and their predictive value on clinical status change (conversion to mild cognitive impairment (MCI)) at v2., Results: Mean age of the sample was 64.72 ± 7.27 years; 62.8% were females. Fifteen participants were classified as FBB-PET+ at baseline and 22 at v2. Every 1 μm of increased thickness in the inner nasal macular region conferred 8% and 6% higher probability of presenting a FBB-PET+ status at v0 (OR = 1.08, 95% CI = 1.02-1.14, p = 0.007) and v2 (OR = 1.06, 95% CI = 1.02-1.11, p = 0.004), respectively. Inner nasal macular thickness also positively correlated with global SUVR (at v0: β = 0.23, p = 0.004; at v2: β = 0.26, p = 0.001). No retinal measurements were associated to conversion to MCI over 24 months., Conclusions: Subtle retinal thickness changes in the macular region are already present in SCD and correlate with Aβ uptake.

- Abstract
The apolipoprotein E ( APOE ) gene on chromosome 19q13.32, was the first, and remains the strongest, genetic risk factor for Alzheimer's disease (AD). Additional signals associated with AD have been located in chromosome 19, including ABCA7 (19p13.3) and CD33 ( 19q13.41). The ABCA7 gene has been replicated in most populations. However, the contribution to AD of other signals close to APOE gene remains controversial. Possible explanations for inconsistency between reports include long range linkage disequilibrium (LRLD). We analysed the contribution of ABCA7 and CD33 loci to AD risk and explore LRLD patterns across APOE region. To evaluate AD risk conferred by ABCA7 rs4147929:G>A and CD33 rs3865444:C>A, we used a large Spanish population (1796 AD cases, 2642 controls). The ABCA7 rs4147929:G>A SNP effect was nominally replicated in the Spanish cohort and reached genome-wide significance after meta-analysis (odds ratio (OR)=1.15, 95% confidence interval (95% CI)=1.12-1.19; P = 1.60 x 10
-19 ). CD33 rs3865444:C>A was not associated with AD in the dataset. The meta-analysis was also negative (OR=0.98, 95% CI=0.93-1.04; P =0.48). After exploring LRLD patterns between APOE and CD33 in several datasets, we found significant LD (D' >0.20; P <0.030) between APOE -Ɛ2 and CD33 rs3865444C>A in two of five datasets, suggesting the presence of a non-universal long range interaction between these loci affecting to some populations. In conclusion, we provide here evidence of genetic association of the ABCA7 locus in the Spanish population and also propose a plausible explanation for the controversy on the contribution of CD33 to AD susceptibility., Competing Interests: CONFLICTS OF INTEREST None. The Face-Name Associative Memory Exam (FNAME) is a paired associative memory test created to detect memory deficits in individuals with preclinical Alzheimer's disease (AD). Worse performance on FNAME in cognitively healthy individuals were found related to higher amyloid beta (Aβ) burden measured with Positron-Emission-Tomography using
11 C-PiB (PiB-PET). We previously reported normative data of a Spanish version of FNAME (S-FNAME) in cognitively healthy Spanish-speaking subjects. The aim of the present study was to determine whether performance on S-FNAME was associated with Aβ burden in subjective cognitive decline (SCD) individuals. 200 SCD subjects received neurological and neuropsychological assessments, including the S-FNAME and the Word List task from the Wechsler-Memory-Scale-III (WMS-III). Moreover, they received an MRI and (18)F-Florbetaben Positron-Emission-Tomography (FBB-PET) to measure Aβ burden. Three cognitive factor composites were derived for the episodic memory measures (face-name [SFN-N], face-occupation [SFN-O] and WMS-III) to determine whether episodic memory performance was related to Aβ deposition. Higher global Aβ deposition was significantly related to worse performance on SFN-N but not with SFN-O or WMS-III Composite. Moreover, worse SFN-N performance was significantly related to higher Aβ deposition in bilateral Posterior Cingulate Cortex. The S-FNAME may be a promising neuropsychological tool for detecting SCD individuals with preclinical AD.- Published
