Ecosystem services (ES) are a fundamental component of well-being and sustainable urban development with tremendous potential to enhance urban planning. Recently, several studies have evaluated the environmental performance of urban plans using the ES approach. To strengthen this science-policy integration, it is still necessary to perform ES assessments within the urban planning practice as well as to collect empirical evidence on the impacts of envisioned planning measures on the supply of ES in urban environments across the world. In this research, we analyzed the state-of-the-art of China’s new environmental governance, which aims to change China’s land use policy and particularly the role of Green Infrastructure (GI) regarding urban planning and ES. We focused on the Shanghai Baoshan district Master Plan as a case study, and analyzed it under the lenses of the supply of ES using the matrix approach. We ascertained the supply of ES as delineated in the ecological network plan for 2035, and developed an evaluation framework based on CICES v5.1 and two expert workshops. Our approach used an integrated preliminary ES-assessment, and evaluated the consequences for the supply of ES in Baoshan district, which is adaptable to varying urban geographies. The results of our assessment show that, if realized as planned, the district will increase the overall supply of ES, especially regulating and cultural services, that play an important role within GI on the urban level. In general, the land use plans should include fine-grained information within building blocks to allow for even better assessing of the spatial structure of the supply of ES

Pyruvate kinase (PK) is one of the three rate-limiting enzymes of glycolysis, and it plays a pivotal role in energy metabolism. In this study, we have identified 10 PK genes from the rice genome. Initially, these genes were divided into two categories: cytoplasmic pyruvate kinase (PKc) and plastid pyruvate kinase (PKp). Then, an expression analysis revealed that OsPK1, OsPK3, OsPK4, OsPK6, and OsPK9 were highly expressed in grains. Moreover, PKs can form heteropolymers. In addition, it was found that ABA significantly regulates the expression of PK genes (OsPK1, OsPK4, OsPK9, and OsPK10) in rice. Intriguingly, all the genes were found to be substantially involved in the regulation of rice grain quality and yield. For example, the disruption of OsPK3, OsPK5, OsPK7, OsPK8, and OsPK10 and OsPK4, OsPK5, OsPK6, and OsPK10 decreased the 1000-grain weight and the seed setting rate, respectively. Further, the disruption of OsPK4, OsPK6, OsPK8, and OsPK10 through the CRISPR/Cas9 system showed an increase in the content of total starch and a decrease in protein content compared to the WT. Similarly, manipulations of the OsPK4, OsPK8, and OsPK10 genes increased the amylose content. Meanwhile, the grains of all CRISPR mutants and RNAi lines, except ospk6, showed a significant increase in the chalkiness rate compared to the wild type. Overall, this study characterizes the functions of all the genes of the PK gene family and shows their untapped potential to improve rice yield and quality traits.

Recreational green spaces are associated with human thriving and well-being. During the ongoing COVID-19 pandemic a spotlight has been shed on the interconnection between access to these spaces, human well-being and social equity. Containment measures enacted in many cities effectively precluded people from reaching distant recreational areas during the pandemic and consequently, recreational areas close to home became particularly important. Urban density is often associated with building or population density with the assumption that if either parameter has a high value, the availability of open (green) space is low. Certain densities are associated with specific spatial qualities. Addressing challenges of sustainable development, a detailed evaluation of density is necessary to allow evidence-based arguments, planning and decision-making. In this study we develop a multi-scale analysis method for quantifying and assessing green infrastructures from settlement unit to building level to reach a differentiated view on density, arguing that density can be organized in different ways achieving very different qualities. For this purpose, we use geospatial-data analysis and in-depth neighborhood studies to compare two cities in Asia and Europe, revealing different ways of organizing density in the built environment and identifying a derivation of approaches for sustainable development in dense urban regions.

Starch and storage proteins are the main components of rice (Oryza sativa L.) grains. Despite their importance, the molecular regulatory mechanisms of storage protein and starch biosynthesis remain largely elusive. Here, we identified a rice opaque endosperm mutant, opaque3 (o3), that overaccumulates 57-kDa proglutelins and has significantly lower protein and starch contents than the wild type. The o3 mutant also has abnormal protein body structures and compound starch grains in its endosperm cells. OPAQUE3 (O3) encodes a transmembrane basic leucine zipper (bZIP) transcription factor (OsbZIP60) and is localized in the endoplasmic reticulum (ER) and the nucleus, but it is localized mostly in the nucleus under ER stress. We demonstrated that O3 could activate the expression of several starch synthesis-related genes (GBSSI, AGPL2, SBEI, and ISA2) and storage protein synthesis-related genes (OsGluA2, Prol14, and Glb1). O3 also plays an important role in protein processing and export in the ER by directly binding to the promoters and activating the expression of OsBIP1 and PDIL1-1, two major chaperones that assist with folding of immature secretory proteins in the ER of rice endosperm cells. High-temperature conditions aggravate ER stress and result in more abnormal grain development in o3 mutants. We also revealed that OsbZIP50 can assist O3 in response to ER stress, especially under high-temperature conditions. We thus demonstrate that O3 plays a central role in rice grain development by participating simultaneously in the regulation of storage protein and starch biosynthesis and the maintenance of ER homeostasis in endosperm cells.

BACKGROUND In recent years, the incidence of gastrointestinal (GI) cancer in China has increased annually. Early detection and appropriate therapy are considered to be the key to treat GI cancer. DNMT1 takes an active part in the advancement of GI cancer, which will change as the disease progresses. But its expression characteristics in the dynamic variations of GI carcinogenesis are still unclear. AIM To investigate the expression characteristics of DNMT1 in different GI diseases. METHODS We detected the expression of DNMT1 in 650 cases of different GI diseases by immunohistochemistry, including 90 cases of chronic superficial gastritis (CSG), 72 cases of atrophic gastritis with intestinal metaplasia (AG/GIM), 54 cases of low-grade intraepithelial neoplasia (GLIN), 66 cases of high-grade intraepithelial neoplasia (GHIN), 71 cases of early gastric cancer (EGC), 90 cases of normal intestinal mucosa (NIM), 54 cases of intestinal low-grade intraepithelial neoplasia (ILIN), 71 cases of intestinal high-grade intraepithelial neoplasia (IHIN), and 82 cases of early colorectal cancer (ECRC). RESULTS In the CSG group, all cases showed weakly positive or negative expression of DNMT1. However, in other four groups (AG/GIM, GLIN, GHIN, and EGC), the positive expression rate gradually increased with the severity of the diseases; the negative or weakly positive cases accounted for 55.56% (40/72), 38.89% (21/54), 1.52% (1/66), and 1.41% (1/71), respectively. Besides, the moderately positive cases were 44.44% (32/72), 57.41% (31/54), 80.30% (53/66), and 43.66% (31/71), respectively. The strongly positive cases only existed in the GLIN (3.70%, 2/54), GHIN (18.18%, 12/66), and EGC (54.93%, 39/71) groups. The differences between any two groups were statistically significant (P < 0.05). Similarly, in the NIM group, cases with weakly positive expression of DNMT1 were predominant (91.11%, 82/90), and the rest were moderately positive cases (8.89%, 8/90). In the ILIN, IHIN, and ECRC groups, the rates of cases with weak or negative expression of DNMT1 were 46.30% (25/54), 12.68% (9/71), and 4.88% (4/82), respectively; with moderately positive expression were 53.70% (29/54), 71.83% (51/71), and 34.15% (28/82), respectively; and with strongly positive expression were 0.00% (0/54), 15.49% (11/71), and 60.98% (50/82), respectively. The differences between any two groups were also statistically significant (P < 0.05). CONCLUSION The overexpression of DNMT1 protein could effectively predict early GI cancers and severe precancerous lesions, which may have potential clinical application value.

Purpose: The study was conducted to investigate the relationship of serum pepsinogens PGI, PGII, gastrin-17, and Hp-IgG with colorectal cancer (CRC), aiming to explore the clinical significance of serum markers reflecting gastric function and H. pylori infection in CRC. Methods: A total of 569 CRC cases and 569 age and sex-matched controls were enrolled in this study between June 2012 and April 2016 from The First Hospital of China Medical University. The serum markers reflecting gastric function and H. pylori infection were detected using ELISA, including PGI, PGII, PGI/II ratio, G-17 and Hp-IgG. Information of clinicopathological parameters and tumor biomarkers was collected from the medical records of inpatients, including CEA, CA199, CA125, CA153 and AFP. Results: Serum PGII, G-17 levels and Hp-IgG were increased in CRC, while PGI and PGI/II ratio appeared no significant difference between CRC and controls. Population ageing and shrinking are demographic phenomena with far-reaching implications for sustainability in the current context of extensive and rapid urbanization. This Perspective rationalizes their interface by (a) identifying the challenges and opportunities that ageing and shrinking urban populations will have for implementing the sustainable development goals (SDGs), and (b) discussing some emerging interventions to capitalise on the opportunities and reduce the challenges to achieving sustainability. We argue that a diverse set of context-specific technological, socioeconomic, institutional and governance interventions would be needed to leverage effectively the opportunities and minimize the risks posed by ageing and shrinking urban populations for long-term sustainability.

Sulforaphane (SFN) extracted from broccoli sprout has previously been investigated for its potential properties in cancers, however, the underlying mechanisms of the anticancer activity of SFN remain not fully understood. In the present study, we investigate the effects of SFN on cell proliferation, cell cycle, cell apoptosis, and also the expression of several cell cycle and apoptosis-related genes by MTT assay, flow cytometry and western blot analysis in gastric cancer (GC) cells. The results showed that SFN could impair the colony-forming ability in BGC-823 and MGC-803 cell lines compared with the control. In addition, SFN significantly suppressed cell proliferation by arresting the cell cycle at the S phase and enhancing cell apoptosis in GC cells in a dose-dependent manner. Western blot results showed that SFN treatment significantly increased the expression levels of p53, p21 and decreased CDK2 expression, which directly regulated the S phase transition. The Bax and cleaved-caspase-3 genes involved in apoptosis executive functions were significantly increased in a dose-dependent manner in BGC-823 and MGC-803 cells. These results suggested that SFN-induced S phase cell cycle arrest and apoptosis through p53-dependent manner in GC cells, which suggested that SFN has a potential therapeutic application in the treatment and prevention of GC.

Objective The aim of this study was to investigate the value of the combined expression of the gastric mucosal differentiation protein pepsinogen C (PGC) and gastric cancer (GC)-associated antigen MG7 for the diagnosis of GC and prediction of the development from precancerous conditions to GC. Methods The gastric mucosal biopsies of 285 subjects enrolled from a region with a high incidence of GC were obtained and histopathologically examined. Subjects testing negative for GC (n=208) were followed up from 1998 to 2015. The levels of PGC and MG7 in the biopsies were determined by immunohistochemistry. Results PGC was positive in 91.4% of the non-atrophic gastritis, 26.5% of the atrophic gastritis, and 0% of the GC. MG7 was positive in 15.0% of the non-atrophic gastritis, 82.4% of the atrophic gastritis, and 94.8% of the GC. The non-atrophic gastritis group was predominantly "PGC+MG7-". The atrophic gastritis and GC groups were predominantly "PGC-MG7+". The rate of GC in subjects with "PGC-MG7+" staining was 113.4-fold higher [95% confidence interval (95% CI): 15.3-869.4, P

Multidimensional interactions of multiple factors are more important in promoting cancer initiation. Gene‐gene interactions between protein‐coding genes have been paid great attention, while rare studies refer to the interactions between encoding and noncoding genes. Our research group previously found encoding gene PGC polymorphisms could affect the susceptibility to atrophic gastritis (AG) and gastric cancer (GC). Interestingly, several SNPs in long noncoding RNA (lncRNA) genes, just adjacent to PGC, were found to be associated with AG risk and GC prognosis afterward. This study aims to explore the SNP interactions between PGC and its neighbor lncRNAs on the risk of AG and GC. Genotyping for seven PGC SNPs and seven lncRNA SNPs was conducted using Sequenom MassARRAY platform in a total of 2228 northern Chinese subjects, including 536 GC cases, 810 AG cases, and 882 controls. We found 15 pairwise PGC‐lncRNAs SNPs had interactions: Five pairs were associated with AG risk, and ten pairs were associated with GC risk. Moreover, two GC‐related interactions PGC rs6939861 with lnc‐C6orf‐132‐1 rs7749023 and rs7747696 survived the Bonferroni correction (P correction = 0.049 and 0.007, respectively). Several combinations showed obvious epistasis and cumulative effects on disease risk. Some three‐way interactions of SNPs with smoking and drinking could also be observed. Besides, a few interacting SNPs showed correlations with the expression levels of PGC protein and related lncRNAs in serum. Our study would provide research clues for further screening combination biomarkers uniting both protein‐coding and noncoding genes with the potential in prediction of the susceptibility to GC and its precursor.

As a building component with long history, green envelope has received much attention for its thermal and energy benefits in the past decades. However, few studies have taken into account the two-way effect of green envelope on indoor and outdoor environment during thermal performance evaluation. In this paper, a coupled hygrothermal transfer model of green envelope is developed and validated against field measurements, and the results demonstrate that the model could reasonably reproduce temporal variations of temperature and moisture in green envelope. Then the green envelope model is implemented into an urban canopy model to predict its thermal performance under urban canopy context in Shanghai area. The simulations for a district of office buildings indicate that green envelope decreases sensible heat getting into the indoor and outdoor space significantly, and the maximum indoor temperature difference between buildings with and without green envelope is up to 0.34 °C under free-floating condition. Meanwhile, for outdoor environment, the maximum differences for air temperature, mean radiant temperature and SET* are up to 0.1,0.9 and 0.2 °C respectively under air conditioning condition. What’s more, sensitivity analysis shows that building density has a significant effect on the relative cooling benefit of green envelope against common envelope.

Besides, protein level analysis were performed to compare ERCC6 and ERCC8 expression in different stages of gastric diseases, and to correlate SNPs jointly with gene expression. Sequenom MassARRAY platform method was used to detect polymorphisms of ERCC6 and ERCC8 in 1916 subjects. In situ ERCC6 and ERCC8 protein expression were detected by immunohistochemistry in 109 chronic superficial gastritis, 109 chronic atrophic gastritis and 109 gastric cancer cases. Our results demonstrated pairwise epistatic interactions between ERCC6 and ERCC8 SNPs that ERCC6 rs1917799- ERCC8 rs158572 combination was associated with decreased risk of chronic atrophic gastritis and increased risk of gastric cancer. ERCC6 rs1917799 also showed a significant interaction with ERCC8 rs158916 to reduce gastric cancer risk. The expressions of ERCC6, ERCC8 and ERCC6-ERCC8 combination have similarities that higher positivity was observed in chronic superficial gastritis compared with chronic atrophic gastritis and gastric cancer. As for the effects of ERCC6 and ERCC8 SNPs on the protein expression, single SNP had no correlation with corresponding gene expression, whereas the ERCC6 rs1917799– ERCC8 rs158572 pair had significant influence on ERCC6 and ERCC6-ERCC8 expression. In conclusion, ERCC6 rs1917799, ERCC8 rs158572 and rs158916 demonstrated pairwise epistatic interactions to associate with chronic atrophic gastritis and gastric cancer risk. The ERCC6 rs1917799– ERCC8 rs158572 pair significantly influence ERCC6 and ERCC6-ERCC8 expression.

This work provides a comparative study of the dielectric and piezoelectric properties of randomly oriented and textured 0.88Na0.5Bi0.5TiO3-0.08K0.5Bi0.5TiO3-0.04BaTiO3 (88NBT) ceramics. Textured ceramics were fabricated by template grain growth (TGG) method using NaNbO3 (NN) for templates. For textured ceramics with 4 wt% NN templates, a Lotgering factor of 96% and piezoelectric coefficient d33 of 185 pC/N were obtained. Compared to the randomly oriented ceramics, textured ceramics show lower strain hysteresis (H = 7.6%), higher unipolar strain of 0.041% with corresponding large signal piezoelectric coefficient d33* of 200 pm/V at applied field of 2 kV/mm. This enhancement can be explained by the grain orientation along &lt, 001&gt, direction by texturing, where an engineered domain configuration is formed after polarization, leading to decreased hysteresis and increased piezoelectric property.

The recreational spaces in urban parks are important to our children for the development of their intellectual, physical and psychological wellbeing. Not only will the failure to meet basic demands for safe and secure recreational spaces causes mental and physical harm to the children, it also leads to the loss in opportunity for children to play in open spaces. Therefore, the research in the safety of recreational spaces for children aims to analyse the potential dangers during recreational activities and to promote safety standards so as to create a safe and healthy outdoor environment for our children. Taking children aged from 3 to 7 as study subjects across 13 regional comprehensive parks in Shanghai downtown area, this article presents an empirical research on accidental injuries in urban parks. Several theories from related subjects such as landscape sociology and child psychology were used. Through interviews and questionnaires, 8 types of accidental injuries were identified using a nonprobability sampling technique. Event probability and distribution characteristics as well as its relationship between children's behaviour and recreation space characteristic were analysed by means of descriptive qualitative statistics. As a result of qualitative research, the framework for potential safety hazard chain model is established and 12 potential safety hazard factors that caused accidental injuries and its spatial features are extracted from the aspects of physical environment and human behaviour after coding 320 interview data based on Grounded Theory Approach.

Green roofs have been recognized as an effective way to increase urban green space and improve building micro-environment. This paper investigated the thermal and energy performance of an extensive green roof under free-floating 1 and air-conditioned conditions in Shanghai, a coastal city with hot and humid summer. A field experiment was conducted on two full-scale rooms of a building from 30/07/2014 to 31/08/2014. One room was covered by a green roof while the other one was covered by a common roof. The results show that solar radiation has the strongest correlation with green roof’s cooling effect, and the correlation between solar radiation and heat flux through green roof is negative in free-floating condition but positive under air-conditioned condition. Outdoor temperature is the most correlated factor of green roof’s temperatures but the least correlated one for green roof’s cooling effect. Energy balance analysis indicates that evapotranspiration and long wave radiation dissipate most of heat gain for green roof, and soil water content has a significant impact on energy balance. This paper presents the basic thermal characteristic of extensive green roof in Shanghai and also make it possible for further validation of hygrothermal transfer model of green roof system.

Green roofs and cool roofs are commonly used to improve indoor thermal environment, reduce air-conditioning load and mitigate the Urban Heat Island (UHI) effect. This study aimed to quantify the differences in thermal and energy performance between the two different roof types under the climate of Shanghai. Firstly, field experiment was conducted in the Shanghai area. Thermal performance of green roof, cool roof and common roof in summer and winter were measured. Results showed that, compared to common roof, the cool roof had an average cooling effect of 3.3 °C on the outer roof deck surface in summer, while the green roof only had a cooling effect of 2.9 °C. In winter, the green roof provided good insulation, and could improve outer surface temperature of the roof deck by an average of 3.3 °C compared to the cool roof. A hygrothermal transfer model for green roof was coupled with a dynamic building thermal performance simulation software (THERB) and validated using measured data. The coupled model was used to predict the effect of both roof types on energy performance of a public building. Simulation results showed that green roof could reduce the cooling and heating loads of the top floor by 3.6% and 6.2%, respectively. The cool roof could reduce cooling load by 3.6% and increase heating load by 10.4%. Finally, a parametric analysis was implemented. The functional mechanism of the main parameters of green and cool roof as well as their impacts on thermal and energy performance of public buildings were analyzed in detail. Conclusions drawn from this paper could provide guidance for the design optimization and application of green roof and cool roof in Shanghai area.

So far, stomach-specific biomarkers, gastric cancer(GC)-related environmental factors, and cancer-associated biomarkers are three major classes of serological biomarkers with GC warning potential, joint detection of which is expected to increase the diagnosis efficiency. We investigated whether the combination of serum pepsinogens(PGs), IgG anti-Helicobacter pylori (HpAb), and osteopontin (OPN) can be used as a panel for GC diagnose. Serum was collected from 365 GC patients and 729 healthy individuals,furtherly 332 cases and 332 age- and sex-matched controls were selected for the matched analysis. Serum levels were measured by ELISA. Logistic regression and receiver operator characteristic curve (ROC) were used to assess the associations of biomarkers with GC and the discriminative performance of biomarkers for GC. Since 2018, the integrated regional development of the Yangtze River Delta has been subjected as a national strategy to intensify the interconnection between its cities. However, the questions of open space conservation and planning have so far remained essentially quantitative and strongly informed by regulatory and top-down principles. Focusing on the vast green heart between Shanghai, Suzhou, and Hangzhou, this design-driven research project hypothesizes that Taipu Canal can be upgraded from its current technical role into a civic spine that frames new developments and articulates the rich diversity of open spaces, ecosystems, historic water towns and villages. The research adopts a crossscale method of “contextual prototypes” that combines sampling, typological classification, and prototypical design explorations in pilot projects. A reflective phase zooms out to critically assess how these prototypical strategies can be systemized as structuring principles at the regional scale. The conclusion of the article discusses how this prototypical approach offers an opportunity to inductively complement the top-down Chinese territorial planning system, which needs to cope with increasingly complex conditions and vaster scales.

Stomach carcinogenesis progresses stepwise from normal mucosa/superficial gastritis, atrophic gastritis (GA) to gastric cancer (GC). Host factors independent of or combined with Helicobacter pylori infection may modulate the carcinogenesis process. In this two-stage study, we selected 24 putative functional tag single-nucleotide polymorphisms (tagSNPs) for six H.pylori-related host genes, MUC1, toll-like receptor 4 (TLR4), protein tyrosine phosphatase, non-receptor type 11 (PTPN11), IL-1B, PGC and PGA3-5, and analyzed their influence and interaction with H.pylori on the GA and GC risks. Using high-throughput genotyping, the 24 tagSNPs were preliminarily assessed in a screening population of 552 controls, 254 GA and 236 GC subjects; subsequently, five candidate tagSNPs for gastric diseases risk in the TLR4, PGC and PTPN11 genes were re-evaluated in a larger population of 1276 controls, 907GA and 714 GC subjects. We observed that PGC rs6458238, PGC rs4711690 and PTPN11 rs12229892 were associated with susceptibilities to GA and/or GC. Moreover, rs4711690 and rs12229892 and H.pylori demonstrated significant interaction effects on GA risk. In gastric cancerous specimens, we observed significantly higher messenger RNA level in the subjects carrying the PGC rs6458238 GA genotype than that in subjects with the common GG genotype. These findings indicated that genetic variations of two crucial H.pylori-related host genes, H.pylori's mucosal effecter PGC gene and H.pylori's cellular messenger PTPN11 gene, either dependent or independent of interaction with H.pylori, were associated with the risks of GC and/or GA that precede carcinoma. Functional studies and further independent large-scale studies especially in other ethnic populations are still needed to confirm our results.

Helicobacter pylori, a microaerophilic Gram-negative bacterium, is known to cause chronic gastritis, peptic ulcer and gastric cancer. Genes that are present in certain isolates may determine strain-specific traits such as disease association and drug resistance. In order to understand the pathogenic mechanisms of gastric diseases, identify molecular markers of the diseases associated with H. pylori strains and provide clues for target treatment of H. pylori-related diseases, a subtracted DNA library was constructed from a gastric cancer-associated H. pylori strain and a superficial gastritis-associated H. pylori strain by suppression subtractive hybridization. The presence of gastric cancer-specific genes was identified by dot blot hybridization, DNA sequencing and PCR-based screening. Twelve gastric cancer-specific high-copy genes and nine low-copy genes were found in gastric cancer compared with the superficial gastritis strain. These genes were confirmed by PCR analysis of H. pylori isolates. This paper presents an ultrahigh sensitive pressure and temperature sensor based on a Fabry-Perot interferometer (FPI) using a graphene diaphragm. The cavity of the FPI is the gap between the end-face of fiber Bragg gratings (FBG) and the graphene diaphragm. To prevent the serious cross-impact of pressure and temperature, a FBG is used to replace the single mode fiber. The sensitivities of pressure are 17.4698nm/kPa and 0.0267pm/kPa for FPI and FBG, respectively. The sensitivities of temperature are 5.075nm/°C and 10.9pm/°C, for FPI and FBG, respectively. The resolutions for pressure and temperature measurement are ±0.0267kPa and ±0.459°C with the wavelength resolution of 5pm. The experimental results show that the sensor is able to measure pressure and temperature simultaneously and accurately by a matrix on sensitivity. The proposed sensor can be applied on wide applications such as ocean exploration, biomedical science, and microfluidic, etc.

To investigate the expressions of E-cadherin, β-catenin, transcription factor 4 (TCF4), which are related with Wnt/β-catenin signaling pathway, and Caudal-type Homeobox Protein 2 (CDX2) originated from intestine, in various gastric diseases and its influence on gastric carcinogenesis and differentiation. A total of 319 various gastric diseases samples from gastroscopy were detected in situ using immunohistochemistry, and data were analyzed with χ 2 test and Kendall’s tau-b rank correlation coefficient analysis. Along with the progression from superficial gastritis (SG) to atrophic gastritis (AG) to gastric cancer (GC), the expressions of E-cadherin and β-catenin appeared a decreasing tendency in total, while β-catenin emerged a decreasing tendency in the cell membrane and an increasing in the nucleus, and the highly positive expression rate of intranuclear transcription factor TCF4 appeared an increasing tendency. The transcription factor CDX2 had a higher expression in AG and GC than that in SG. The expression rates of the four proteins above in intestinal type adenocarcinoma were all higher than those in diffuse type carcinoma. There were positive correlations between E-cadherin and β-catenin expressed in cytoplasm, between β-catenin expressed in cell membrane and β-catenin in cytoplasm, between β-catenin expressed in cytoplasm and β-catenin in nucleus, between β-catenin and TCF4 in nucleus, between TCF4 and CDX2 in nucleus, respectively (P 0.05). We found that the four proteins were involved in gastric carcinogenesis and the differentiation to intestinal type adenocarcinoma.

This study sought to examine interactions among polymorphisms of H. pylori- related genes PGC, PTPN11, TLR4 , and IL1B and assess whether their interaction effects were modified by H. pylori infection. Thirteen polymorphisms of the aforementioned genes were genotyped by the Sequenom MassARRAY platform in 714 gastric cancer patients, 907 atrophic gastritis cases and 1276 healthy control subjects. When we considered the host genetic effects alone, gene–gene interactions consistently decreased the risks of gastric cancer and/or atrophic gastritis, including three two-way interactions: PGC rs6912200- PTPN11 rs12229892, PGC rs4711690- IL1B rs1143623 and PTPN11 rs12229892- IL1B rs1143623 and a three-way interaction: PGC rs4711690- PGC rs6912200- PTPN11 rs12229892. When the effect modification of H. pylori infection was evaluated, the cumulative effects of the aforementioned three-way interaction on atrophic gastritis susceptibility switched from being beneficial to being risky by the status of H. pylori infection. These data showed that SNP interactions among H. pylori -related genes PGC, PTPN11 , and IL1B , are associated with susceptibility to gastric carcinogenesis. Moreover, we provided important hints of an effect modification by H. pylori infection on the cumulative effect of PGC and PTPN11 polymorphisms. Functional experiments and further independent large-scale studies especially in other ethnic populations are still needed to confirm our results.

The government attaches great importance to support the high technology industry and promote the high technology industry products export; and the government should deepen reform of the financial system and construct multi-channel financing system to provide an opportunity for the development of enterprise.

BACKGROUND: Pepsinogen C (PGC) plays an important role in sustaining the cellular differentiation during the process of gastric carcinogenesis. This study aimed to assess the role of PGC tagSNPs and their interactions with Helicobacter pylori (H. pylori) in the development of gastric cancer and its precursor, atrophic gastritis. METHODS: Four PGC tagSNPs (rs6941539, rs6912200, rs3789210 and rs6939861) were genotyped by Sequenom MassARRAY platform in a total of 2311 subjects consisting of 642 gastric cancer, 774 atrophic gastritis, and 895 healthy control subjects. The mRNA and protein expression levels of PGC in gastric tissues and in serum were respectively measured by quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR), immunohistochemistry, and Eenzyme-linked immunoabsorbent assay (ELISA). RESULTS: We found associations between PGC rs3789210 CG/GG genotypes and reduced gastric cancer risk and between PGC rs6939861 A variant allele and increased risks of both gastric cancer and atrophic gastritis. As for the haplotypes of PGC rs6941539-rs6912200-rs3789210-rs6939861 loci, the TTCA and TTGG haplotypes were respectively associated with increased and reduced risks of both gastric cancer and atrophic gastritis; additionally, the CTCA haplotype was associated with increased atrophic gastritis risk. Very interestingly, rs6912200 CT/TT genotypes had a positive interaction with H. pylori, synergistically elevating the gastric cancer risk. Moreover, healthy subjects who carried rs6912200 CT, TT and CT/TT variant genotypes had lower histological and serum expression levels of PGC protein. CONCLUSIONS: Our findings highlight an important role of PGC rs3789210 and rs6939861 in altering susceptibility to atrophic gastritis and/or gastric cancer. Moreover, people who carry rs6912200 variant genotypes exhibit higher gastric cancer risk in case of getting H. pylori infection, which strongly suggest a necessity of preventing and/or eliminating H. pylori infection in those individuals.

Background Xeroderma pigmentosum group G (XPG) plays a critical role in preventing cells from oxidative DNA damage. This study aimed to investigate XPG protein expression in different gastric tissues and in patients with diverse prognoses, thus providing insights into its role in the development, progression and prognosis of gastric cancer (GC). Methods A total of 176 GC, 131 adjacent non-tumour tissues, 53 atrophic gastritis (AG) and 49 superficial gastritis (SG) samples were included. Immunohistochemical staining was used to detect XPG protein expression. Results XPG expression was significantly higher in GC tissues compared with adjacent non-tumour tissues. In the progressive disease sequence SG→AG→GC, XPG expression was significantly higher in AG and GC compared with SG. Analysis of clinicopathological parameters and survival in GC patients demonstrated a significant association between XPG expression level and depth of tumour invasion, macroscopic type, Lauren’s classification, smoking, Helicobacter pylori infection and family history. Cox multivariate survival analysis indicated that patients with positive XPG expression had significantly longer overall survival (P = 0.020, HR = 0.394, 95%CI 0.179–0.866), especially in aged younger than 60 years (P = 0.027, HR = 0.361, 95%CI 0.147–0.888) and male patients (P = 0.002, HR = 0.209, 95%CI 0.077–0.571). Conclusions This study demonstrated that XPG protein expression was related to the development, progression and prognosis of GC, and might thus serve as a potential biomarker for its diagnosis and prognosis.

We propose a full-field optical method for the label-free and quantitative mapping of the velocities of red blood cells (RBCs) in capillaries. It integrates spatiotemporal demodulation and an autocorrelation algorithm, and measures RBC velocity according to the ratio of RBC length to lag time. Conventionally, RBC length is assumed to be a constant and lag time is taken as a variable, while our method treats both of them as variables. We use temporal demodulation and the Butterworth spatial filter to separate RBC signal from background signal, based on which we obtain the RBC length by image segmentation and lag time by autocorrelation analysis. The RBC velocity calculated now is more accurate. The validity of our method is verified by an in vivo experiment on a mouse ear. Owing to its higher image signal-to-noise ratio, our method can be used for mapping RBC velocity in the turbid tissue case.

Identifying the genetic variants that alter MUC I protein expression may further our understanding of the risk for development of gastric cancer (GC). We used PCR-SSPs to identify the genotype of MUC1 A/G polymorphism at its 568 site of exon 2 and immunohistochemistry to detect MUC1 protein expression in GC patients and non-cancer subjects and analyzed the association between this polymorphism and MUC1 protein expression. We found that the frequency of AA genotype was significantly high in the GC patients and the risk for GC in AA genotype carriers increased 1.81-fold. Moreover, we found a significant underexpression of MUC protein in GC as compared to non-cancer subjects, which was negatively correlated to AA genotype of MUCI (r=-0.1790, P=0.004). Furthermore, this study provides a possible mechanistic insight that the MUC1 A/G polymorphism at its 568 site disrupts the physiological functions of MUC1 which is important to the physiological protection of gastric mucosa. Thus we have provided evidence that may identify the MUCI A/G polymorphism at 568 site, as a potential genetic factor which leads to an increase in susceptibility for GC through alteration of MUCI gene and MUC1 expression in the population that carry the A allele.
