Aim: To unveil a putative correlation between phage genome flexibility and virion morphogenesis yield. Materials & methods: A deeper analysis of the mechanical properties of three Pseudomonas aeruginosa lytic phage genomes was undertaken, together with full genome cyclizability calculations. Results & conclusion: A putative correlation was established among phage genome flexibility, eclipse timeframe and virion particle morphogenesis yield, with a more flexible phage genome leading to a higher burst size and a more rigid phage genome leading to lower burst sizes. The results obtained are highly relevant to understand the influence of the phage genome plasticity on the virion morphogenesis yield inside the infected bacterial host cells and assumes particular relevance in the actual context of bacterial resistance to antibiotics.

Background The CQC inspection of the Royal Lancaster Infirmary (RLI) in May 2021 rated the performance of the stroke department unsatisfactory, leading to a number of changes. A retrospective audit was performed to determine the impact of these changes for thrombolysed stroke patients. Aim This retrospective audit assessed the performance of the stroke department at the RLI against the parameters set by the ‘Sentinel Stroke National Audit Program’ (SSNAP), comparing 6-month periods before and after the CQC inspection in May 2021. Method Using electronic medical records and SSNAP data, we reviewed every thrombolysed stroke patient at the RLI between November 2020 until April 2021 and from May 2021 until November 2021, assessing 10 parameters and comparing the results with SSNAP targets. Since May 2021, changes to practice introduced included opening a new, larger stroke unit located directly opposite the Emergency Department, ring-fencing stroke beds, doubling the number of stroke specialists and stroke consultants reviewing all suspected stroke patients face-to-face within working hours. Results 46 patients were thrombolysed with 42 confirmed as having had ischaemic strokes on subsequent MRI imaging. All patients were discussed with a stroke consultant before thrombolysis. Mean time from arrival to CT improved from 51 to 34.5 minutes, admission to stroke unit from 7hr53 to 4hr36 and to thrombolysis from 2hr18 to 1hr22. The number of thrombolysis complications decreased from 5 to 2. Since the changes, the SSNAP grade for stroke unit admission improved from C to A and specialist assessments from E to B. Conclusion The changes implemented following the May 2021 CQC inspection have had a positive impact on the care of thrombolysed stroke patients and overall SSNAP grades at RLI. Improvements are still required and the next steps include improving the efficiency of thrombolysis times and further improving SSNAP grades.

This work addresses the analysis of mechanical metamaterials exhibiting, as a distinguishing feature, snap-through instabilities in the elastic regime. Among the various opportunities offered by this feature is the manipulation of the material deformation energy [1] that allows the conception of reusable energy-trapping or energy-dissipation devices [2]. As a task to reach this objective, we present a novel computational methodology for analysing microarchitectures displaying elastic snap-through instabilities. The strategy is based on the construction of a surrogate model that reduces the computational burden if compared with full high-fidelity models of non-linear volumetric finite elements or with nonlinear beam elements. The surrogate model opens the possibility to analyse a large number of unit cells of periodic metamaterials with instabilities. In addition, it permits an efficient assessment of different geometrical configurations proving a suitable tool for its use in microarchitecture topology optimization processes. Noting that, in general, existing designs of this class of metamaterials only achieve energy manipulation in a specific predefined loading direction, in this work, and based on the proposed computational technique, we aim at the analysis of reusable multidirectional isotropic energy manipulation. This objective agrees with more realistic uses of metamaterials in which general boundary conditions, typically the loading directions, are not established a priori.

Background: Patients with COVID-19 can develop acute respiratory distress syndrome (ARDS), which is associated with high mortality. The aim of this study was to examine the functional and morphological features of COVID-19-associated ARDS and to compare these with the characteristics of ARDS unrelated to COVID-19. Methods: This prospective observational study was done at seven hospitals in Italy. We enrolled consecutive, mechanically ventilated patients with laboratory-confirmed COVID-19 and who met Berlin criteria for ARDS, who were admitted to the intensive care unit (ICU) between March 9 and March 22, 2020. All patients were sedated, paralysed, and ventilated in volume-control mode with standard ICU ventilators. Static respiratory system compliance, the ratio of partial pressure of arterial oxygen to fractional concentration of oxygen in inspired air, ventilatory ratio (a surrogate of dead space), and D-dimer concentrations were measured within 24 h of ICU admission. Lung CT scans and CT angiograms were done when clinically indicated. A dataset for ARDS unrelated to COVID-19 was created from previous ARDS studies. Survival to day 28 was assessed. Findings: Between March 9 and March 22, 2020, 301 patients with COVID-19 met the Berlin criteria for ARDS at participating hospitals. Although regular combined aerobic-resistance exercises can ameliorate the inflammatory status and redox balance in elderly population, it is unclear whether protein or specific amino acid supplementation could improve such benefits. Therefore, we aimed to evaluate the inflammatory status and redox indexes through of the saliva of 34 elderly subject nonpractitioners (NP group, 73.3±6.6 years) and 49 elderly subject practitioners of a combined-exercise training in moderate intensity (CET group, 71.9±5.8 years) before (pre) and after (post) 30 days of supplementation with L-glutamine (Gln) or placebo (PL). Our results showed that, both in pre- and postsupplementation, the salivary levels of nitric oxide (NO⋅) and TNF-α were lower, whereas the levels of uric acid and IL-10 (as well as IL-10/TNF-α ratio) were higher in the CET groups than in the NP groups. In postsupplementation, both groups supplemented with Gln (NP-Gln and CET-Gln) showed higher salivary uric acid levels compared to baseline. In addition, lower NO⋅ levels were found in the CET-Gln group postsupplementation than presupplementation values. Whereas the CET-Gln group showed lower GSH levels postsupplementation, NP-Gln subjects showed lower GSSG levels at the same time point, both compared to baseline. Interestingly, salivary peroxidase activity was lower only in NP groups (NP-PL and NP-Gln) postsupplementation than baseline values. A positive significant correlation between salivary peroxidase activity and GSH levels, and also between salivary peroxidase activity and uric acid levels were observed in the CET-Gln group both pre- and postsupplementation. No differences were found in albumin, total antioxidant activity (TEAC), and reducing power analysis between groups, pre- or postsupplementation. In conclusion, the elderly subjects from the CET group showed a better inflammatory response and redox balance and, for the first time, it was shown that daily supplementation with Gln for 30 days can improve these benefits with putative association with a healthy aging.

We use a scanning superconducting quantum interference device (SQUID) to image the magnetic flux produced by a superconducting device designed for quantum computing. The nanometer-scale SQUID-on-tip probe reveals the flow of superconducting current through the circuit as well as the locations of trapped magnetic flux. In particular, maps of current flowing out of a flux-control line in the vicinity of a qubit show how these elements are coupled, providing insight on how to optimize qubit control., 5 pages, 3 figures

The COVID-19 pandemic has made evident the need to develop effective strategies to cushion the psychological consequences of social catastrophes. Preliminary evidence suggests that the use of hallucinogens is a protective factor that mitigates against such stressors. However, the underlying mechanisms must be further explored. This study specifically focused on the potential role of coping strategies in this regard, analyzing them through an online survey completed by a total of 2971 subjects who were followed up with from the beginning of the COVID-19 pandemic until six months after baseline. The survey was published in three different cultures (English, Spanish, and Portuguese), allowing for the collection of data from three different cultural contexts. The results show that coping strategies were generally more related to psychological well-being and psychopathology than to hallucinogenic drug use. However, regarding the latter, users of hallucinogens had higher scores on problem-focused engagement and disengagement and lower scores on wishful thinking than non-users. Longitudinally, while most baseline coping scores were associated with psychological distress and the severity of psychological symptoms, some coping strategies were related to the use of hallucinogens. These results show an adaptive pattern of coping strategies among hallucinogen users. Further research should take into account that coping strategies are only marginally associated with hallucinogenic drug use. Other underlying mechanisms explaining the better adjustment of users of hallucinogens to pandemics should be explored.

SummaryWhole Genome Assembly (WGA) of bacterial genomes with short reads is a quite common task as DNA sequencing has become cheaper with the advances of its technology. The process of assembling a genome has no absolute golden standard (Del Angel et al. (2018)) and it requires to perform a sequence of steps each of which can involve combinations of many different tools. However, the quality of the final assembly is always strongly related to the quality of the input data. CD8+ T-cell activation has been demonstrated to distinguish patients with primary and infection-associated hemophagocytic lymphohistiocytosis (HLH) from patients with early sepsis. We evaluated the activation profile of CD8+ T cells in patients with various forms of secondary HLH (sHLH), including macrophage activation syndrome (MAS). Peripheral blood mononuclear cells from children with inactive systemic juvenile idiopathic arthritis (sJIA, n = 17), active sJIA (n = 27), MAS in sJIA (n = 14), infection-associated HLH (n = 7), and with other forms of sHLH (n = 9) were analyzed by flow cytometry. Compared with patients with active sJIA, in patients with MAS and sHLH of different origins, beside a significant increase in the frequency of CD38high/HLA-DR+CD8+ T cells, we found a significant increase in the frequency of CD8+ T cells expressing the CD4 antigen (CD4dimCD8+ T cells). These cells expressed high levels of the activation markers CD38 and HLA-DR, suggesting they were a subset of CD38high/HLA-DR+CD8+ T cells, as well as of the activation/exhaustion markers CD25, PD1, CD95, and interferon-γ. The frequency of CD4dimCD8+ T cells strongly correlated with most of the laboratory parameters of MAS severity and with circulating levels of CXCL9 and interleukin-18. These findings were confirmed in a prospective replication cohort in which no expansion of any particular T-cell receptor Vβ family in CD3+ T cells of patients with sHLH was found. Finally, frequency of CD4dimCD8+, but not of CD38high/HLA-DR+CD8+ T cells, significantly correlated with a clinical severity score, further supporting the involvement of these cells in MAS/sHLH pathogenesis.

We present a detailed description of the electromagnetic filter for the PTOLEMY project to directly detect the Cosmic Neutrino Background (CNB). Starting with an initial estimate for the orbital magnetic moment, the higher-order drift process of E×B is configured to balance the gradient-B drift motion of the electron in such a way as to guide the trajectory into the standing voltage potential along the mid-plane of the filter. As a function of drift distance along the length of the filter, the filter zooms in with exponentially increasing precision on the transverse velocity component of the electron kinetic energy. This yields a linear dimension for the total filter length that is exceptionally compact compared to previous techniques for electromagnetic filtering. The parallel velocity component of the electron kinetic energy oscillates in an electrostatic harmonic trap as the electron drifts along the length of the filter. Educational interactive technologies are placed in museums to enhance the visitor experience. Great time and money and hard work hours are spent to design, develop and deploy these systems. But do these systems actually achieve their pedagogical goals? Do they contribute to greater effectiveness and user experience (UX)? In this article we report the use of the technological attraction "Adventure in the Solar System", from the Catavento Cultural Museum, in São Paulo, Brazil. The attraction simulates an adventure in space through a spaceship composed of collaborative games designed to teach about astronomy, physics and mathematics. 129 students participated in this empirical study and were classified into three groups (ESI - Elemental Scholl I; ESII - Elemental Scholl II; HS - High Scholl), all aged between 7 and 16 years. The main objective of the study was to analyze learning performance and user experience (UX). The results showed that there were no statistically significant results betwen groups, but higher proportions of positive scores were observed in the ESII and SH groups. Regarding the user experience, ESI and ESII presented more expressive results than SH. The main results are discussed and ideas for future research are presented.

SABRE (Sodium-iodide with Active Background REjection) is a direct dark matter search experiment based on an array of radio-pure NaI(Tl) crystals surrounded by a liquid scintillator veto. Twin SABRE experiments in the Northern and Southern Hemispheres will differentiate a dark matter signal from seasonal and local effects. The experiment is currently in a Proof-of-Principle (PoP) phase, whose goal is to demonstrate that the background rate is low enough to carry out an independent search for a dark matter signal, with sufficient sensitivity to confirm or refute the DAMA result during the following full-scale experimental phase. The impact of background radiation from the detector materials and the experimental site needs to be carefully investigated, including both intrinsic and cosmogenically activated radioactivity. Based on the best knowledge of the most relevant sources of background, we have performed a detailed Monte Carlo study evaluating the expected background in the dark matter search spectral region. The simulation model described in this paper guides the design of the full-scale experiment and will be fundamental for the interpretation of the measured background and hence for the extraction of a possible dark matter signal., Comment: 13 pages, 5 figures

Bruchine beetles are usually considered seed predators, particularly because these beetles consume the seed embryo and kill the seeds. Previous studies suggest that under certain conditions, these insects do not kill the embryo. In this study, we asked whether the germination speed of seeds of the invasive tree Leucaena leucocephala is enhanced by the seed-feeding beetle Acanthoscelides macrophthalmus (Coleoptera: Chrysomelidae: Bruchinae). We also tested whether differences in germination between attacked and intact seeds could be related to seed size. We finally examined whether the number of larvae per seed affected seed germination and whether seedling emergence patterns were similar to those exhibited by germination. We found that compared to intact seeds, proportionally faster germination occurred in seeds attacked by A. macrophthalmus in 14 of the 26 populations studied. In addition, those populations that presented greater germination speed in the attacked seeds during the evaluation time also had the largest seeds. Similar to the germination experiments, seedling emergence was faster in the attacked compared to the intact seeds. Our findings show that the germination speed of L. leucocephala seeds can be enhanced by A. macrophthalmus. However, this effect is much more intense when a single larva develops inside the seed. In evolutionary terms, it is possible that the selection for larger seeds is favoured, increasing the speed of seed germination and contributing to predator satiation at the seed level. However, this effect could be minimized by the production of many seeds by L. leucocephala plants, causing predator satiation by “masting” effects, which would favour the selection for higher seed numbers.

Global warming is affecting biodiversity; however, the extent to which animal reproductive processes respond to predicted temperature increments remains largely unexplored. The thermal environment has a pronounced impact on metabolic rates of ectotherms; therefore, an interesting question to assess is whether temperature increase might affect specific reproductive mechanisms like sperm performance in ectotherms. Moreover, in many species, oviductal fluid (OF) is known to regulate and maintain sperm quality; however, the role of OF in relation to the effects of high temperature on sperm remains unclear. Our aim was to experimentally test the effect of increased temperature on sperm velocity, swimming path and percentage of motility in neutral conditions at ejaculation (without OF) and in female's reproductive tract fluid (with OF), in a social ectotherm lizard model, Tropidurus spinulosus, which has specific thermal requirements for reproduction. Our results suggest that a rising temperature associated with global warming (+4°C) affects negatively sperm dynamics and survival. However, OF ameliorated the harmful effects of high temperature. This is an important point, as this study is the first to have tested the role of OF in preserving sperm from a warmer pre-fertilization environment. These results contribute to our understanding of how thermal environment changes might affect post-copulatory reproductive mechanisms. Proportional electroluminescence (EL) in noble gases is used in two-phase detectors for dark matter searches to record (in the gas phase) the ionization signal induced by particle scattering in the liquid phase. The "standard" EL mechanism is considered to be due to noble gas excimer emission in the vacuum ultraviolet (VUV). In addition, there are two alternative mechanisms, producing light in the visible and near infrared (NIR) ranges. The first is due to bremsstrahlung of electrons scattered on neutral atoms ("neutral bremsstrahlung", NBrS). The second, responsible for electron avalanche scintillation in the NIR at higher electric fields, is due to transitions between excited atomic states. In this work, we have for the first time demonstrated two alternative techniques of the optical readout of two-phase argon detectors, in the visible and NIR range, using a silicon photomultiplier matrix and electroluminescence due to either neutral bremsstrahlung or avalanche scintillation. The amplitude yield and position resolution were measured for these readout techniques, which allowed to assess the detection threshold for electron and nuclear recoils in two-phase argon detectors for dark matter searches. Background: One of the main public health strategies adopted at the beginning of the COVID-19 pandemic consisted of implementing strict lockdowns to stop the transmission of the virus. Despite being an effective measure, the confinement and the associated social isolation create a stressful, potentially lengthy situations that has been proven to have several psychological consequences. Given the potential benefits that certain psychedelic drugs have shown for the treatment of psychological disorders, this study aimed to assess the impact of lifetime psychedelic drug use on mental health in relation to the first strict lockdown adopted by various countries (April-July 2020).Methods: Subjects completed an online survey that inquired about sociodemographic factors, activities, and lifestyle factors during confinement, as well as health and mental health related factors. Subjects were asked about their lifetime use of psychedelic drugs (MDMA, ayahuasca, psilocybin-containing mushrooms, LSD, peyote, San Pedro, Bufo alvarius or 5-MeO-DMT, and others), being classified as regular users (more than once per 6 months), occasional users, or non-users. The survey included psychometric tests used to assess psychological distress, peritraumatic stress, social support, psychopathological symptoms, and personality. Linear regressions were performed with psychedelic drug users as the independent variable and psychometric factors as the outcomes, while correcting for age, gender, language, religion, spirituality, and use of non-psychedelic drugs.Results: The study included 2,974 English, Portuguese, and Spanish speakers (497 regular users of psychedelic drugs, 606 occasional users, and 1,968 non-users). On average, respondents were 36 years old and 70% were female. Psychedelic drug users, especially regular ones, reported less psychological distress, less peritraumatic stress, and more social support. Regarding personality measures, psychedelic drug users scored higher on the novelty-seeking and self-transcendence scales, and lower on cooperativeness.Conclusion: Our findings showed that regular users of psychedelic drugs had less psychological stress and some personality differences when compared to occasional users and non-users. This suggests that either the use of psychedelics might be a protective factor itself or people with certain previous traits are more prone to frequently using psychedelic drugs. Future prospective longitudinal research should investigate the underlying processes observed in this study to develop consistent hypotheses.

The violation of baryon number, B , is an essential ingredient for the preferential creation of matter over antimatter needed to account for the observed baryon asymmetry in the Universe. However, such a process has yet to be experimentally observed. The HIBEAM/NNBAR program is a proposed two-stage experiment at the European Spallation Source to search for baryon number violation. The program will include high-sensitivity searches for processes that violate baryon number by one or two units: free neutron–antineutron oscillation ( n → n ̄ ) via mixing, neutron–antineutron oscillation via regeneration from a sterile neutron state ( n → [ n ′ , n ̄ ′ ] → n ̄ ), and neutron disappearance (n → n′); the effective Δ B = 0 process of neutron regeneration ( n → [ n ′ , n ̄ ′ ] → n ) is also possible. The program can be used to discover and characterize mixing in the neutron, antineutron and sterile neutron sectors. The experiment addresses topical open questions such as the origins of baryogenesis and the nature of dark matter, and is sensitive to scales of new physics substantially in excess of those available at colliders. A goal of the program is to open a discovery window to neutron conversion probabilities (sensitivities) by up to three orders of magnitude compared with previous searches. The opportunity to make such a leap in sensitivity tests should not be squandered. The experiment pulls together a diverse international team of physicists from the particle (collider and low energy) and nuclear physics communities, while also including specialists in neutronics and magnetics.

In the context of the PTOLEMY project, the need for a site with a rather low cosmogenic induced background led us to measure the differential muon flux inside the bunker of Monte Soratte, located about 50~km north of Rome (Italy). The measurement was performed with the Cosmic Ray Cube (CRC), a portable tracking device. The simple operation of the Cosmic Ray Cube was crucial to finalise the measurements, as they were carried out during the COVID-19 lockdown and in a site devoid of scientific equipment. The muon flux measured at the Soratte hypogeum is above two orders of magnitude lower than the flux observed on the surface, suggesting the possible use of the Mt. Recently, an increasing attention in electric and hybrid vehicles is taking place. In order to raise awareness and technical knowledge on vehicles with reduced carbon footprint, several electric car competitions have been launched. Formula SAE is one of these, where talented students from different universities around the globe compete to design a fully electric racing car. This requires the development of high power density drives, with accurate electromagnetic and thermal analyses.This paper presents the design process and the thermal assessment of a high-performance electric motor intended for a FSAE racing car. The starting requirements for the electromagnetic and thermal designs are based on realistic available data from previous competitions and championship regulations.
