Habrobracon hebetor Say (Hymenoptera: Braconidae) is one of the most important parasitoids of many pyralid moths, including the olive leaf moth, Palpita unionalis Hubner (Lepidoptera: Pyralidae). The widespread use of insecticides threatens natural enemies. Assessing the side effects of insecticides on nontarget organisms supports the rational use of insecticides during field application. The present study evaluates the lethal and sublethal effects of three insect growth regulators (IGRs), novaluron, methoxyfenozide, and pyriproxyfen, on P. unionalis and the demographic toxicology of these IGRs on its parasitoid H. hebetor. The LC 50 values of these IGRs on P. unionalis were 0.97, 0.176, and 0.00009 ppm, respectively, indicating that pyriproxyfen was the most toxic. When H. hebetor adults were exposed to these LC 50 levels under laboratory conditions to determine possible side effects. The IGRs did not affect the paralysis and parasitism rates of the parasitoid nor the sex ratio of its offspring. IGR treatments slightly reduced hatching rates and immature survival by 15-25%, indicating moderate effects on the early developmental stages of H. hebetor. The longevity and fecundity of treated females were each reduced to < 50% of their respective values in untreated females. Additionally, some demographic parameters of the parasitoid were significantly affected by the IGRs. Nevertheless, despite these observed effects, the positive net reproductive rate (R 0  > 0) and intrinsic rate of increase (r m > 1) of H. hebetor indicated an exponential population increase that reflects the compatibility of the IGRs with the parasitoid. Our results demonstrated that the tested IGRs could be categorized as relatively harmless compounds to the parasitoid. Palpita forficifera Munroe, 1959 (Lepidoptera: Crambidae) is the main pest of the olive tree (Olea europaea L., Oleaceae) in Brazil and its management has been difficult, as there are few products recommended for its control. This study aimed to evaluate the population dynamics of P. forficifera and the entomofauna of parasitoid eggs and larvae in three olive orchards under different cultivation systems, in Rio Grande do Sul state, Brazil. The study was carried out from October to May, in the 2017/18 and 2018/19 harvests, in olive orchards of different cultivars in the municipalities of Pelotas and Rio Grande, state of Rio Grande do Sul, Brazil. Population dynamics of P. forficifera varied according to the agricultural season, the months sampled, and the orchards (sites) evaluated. The highest infestation reached around 60% of the shoots in some months, depending on the orchard evaluated. During the pest infestation period, the occurrence of the egg parasitoid Trichogramma foersteri Takahashi, 2021 (Hymenoptera: Trichogrammatidae) was observed along with three larval parasitoids belonging to the genera Dolichogenidea Viereck, 1911, Hymenochaonia Dalla Torre, 1898 (Hymenoptera: Braconidae) and Temelucha Förster, 1869 (Hymenoptera: Ichneumonidae). Temelucha hilux Gauld, 2000 is recorded for the first time in association with a host, and its distribution in the country is extended to the southern region, from Brazil. Due to the scarcity of information on pest management, the natural occurrence of natural enemies in crops is of paramount importance in helping to manage P. forficifera in the field. The population dynamics of P. forficifera is also influenced by the parasitoid community, made up of at least four species of parasitic Hymenoptera. Therefore, strategies aimed at managing P. forficifera must be well developed in order to maintain and increase natural biological control in the field.

Insect gut microbes play important roles in digestion, metabolism, development, and environmental adaptation. Parasitoid wasps are one of the most important biological control agents in pest control, while the gut microbial species compositions and the associated functions have been poorly investigated. Two endoparasitoid wasps, Cotesia vestalis and Diadromus collaris , parasitize the larval stage and pupal stage of the diamondback moth, Plutella xylostella , respectively. Using whole-genome shotgun metagenomic sequencing, we characterized the gut microbial composition, diversity, and potential functional roles associated with the two parasitoid wasp larvae. The results reveal that Proteobacteria and Firmicutes are the dominant phyla in the gut of C. vestalis and D. collaris larvae, with Rhizobium and Enterococcus being the dominant genera. The putative microbial functions associated with the two parasitoid wasps might play a virtual role in assisting in consuming the host's nutritional composition. The enriched CAZymes family genes are primarily involved in the degradation and synthesis of chitin. Despite the richness of microbial species and communities, the microbes species and the microbial community structure exhibit significant similarity between the two parasitoid wasps and between the parasitoid wasp and the host P. xylostella . Notably, the prevalence of the genus Enterococcus shared among them suggests a possible link of gut microbes between the host and their associated parasitoids. Our study offers insights into the gut microbe-based interactions between the host and parasitoid wasps for the first time, potentially paving the way for the development of an ecologically friendly biocontrol strategy against the pest P. xylostella .IMPORTANCEEndoparasitoid wasps spend the majority of their lifespan within their host and heavily rely on the host's nutrition for survival. There is limited understanding regarding the composition and physiological impacts of gut microbial communities in parasitoid wasps, particularly during the larval stage, which is directly linked to the host. Based on a thorough characterization of the gut microbe and comprehensive comparative analysis, we found the microbial species of the larval parasitoid wasp Cotesia vestalis and the pupal parasitoid wasp Diadromus collaris were similar, sharing 159 genera and 277 species, as were the microbial community structure. Certain of the dominant microbial strains of the two parasitoid wasps were similar to that of their host Plutella xylostella larvae, revealing host insect may affect the microbial community of the parasitoid wasps. Pest management on hemp is still in its infancy, and biological control options are limited. Helicoverpa zea (corn earworm) is one of the key pests of hemp cultivated outdoors, especially on cultivars grown for cannabinoids and grain. In a three-year study, we assessed the effect of diet on the performance of H. zea and its tachinid parasitoids. Parasitized (bearing fly eggs) and unparasitized (without eggs) H. zea larvae were fed on hemp flowers or an artificial diet. Five tachinid species parasitized H. zea larvae, but the most abundant species were Winthemia rufopicta (68.8%) and Lespesia aletiae (28.3%). Overall, 55.2% of H. zea larvae bearing tachinid eggs died, while the mortality of unparasitized larvae reached 24.7%. The success of tachinids increased by 2-fold when the host larvae were fed on an artificial diet. Our results demonstrated that high protein food (artificial diet), intensity of parasitism, and caterpillar size play a role in the fitness of both the herbivores (H. zea) and its tachinid parasitoids. These findings have important implications for understanding biological control mechanisms and open new insights into the impact of landscape variation on plant-herbivore-parasitoid interactions. A complete three-dimensional reconstruction of the internal damage (oviposition holes, entry and exit galleries, cavities caused by fungal infection) of three destructive pests of olive fruit was obtained using micro-computed tomography. In the case of the olive fruit fly (Bactrocera oleae), complete reconstruction of the galleries was achieved. The galleries were colour-coded according to the size of the internal lumens produced by larval instars. In the case of the olive moth (Prays oleae), we confirmed that the larvae only consume olive stones, leaving pulp tissue intact. This study revealed the evolutionary defensive adaptation developed by larvae, creating entrance/exit gallery in the form of a zigzag with alternating angles to avoid the action of possible parasitoids. In the case of olive fruit rot, caused by fungal infection transmitted by the midge (Lasioptera berlesiana), microtomography revealed the infection cavity, which was delimited by a protective layer of tissue produced by the plant to isolate the infection zone, which contained fungal hyphae and reproductive organs of the fungus. Two ovoid cavities were observed below a single external orifice in the concave necrotic depression. These results were interpreted as successive ovipositions of B. oleae, followed by the parasitoid L. berlesiana. We examined the possibility of a cross-modal effect in naïve Cotesia vestalis, a parasitoid wasp of diamondback moth larvae, by using artificial flower models of four colours (blue, green, yellow, and red) in the absence or presence of floral scent collected from Brassica rapa inflorescences. In a four-choice test, regardless of the floral scent, non-starved female wasps visited green and yellow models significantly more often than blue and red ones, although no significant difference was observed between visits to the green and yellow models. They seldom visited blue and red models. When starved, the wasps became even more particular, visiting yellow significantly more frequently than green models, irrespective of the presence of the floral scent, indicating that they preferred to use yellow visual cues in their food search. Furthermore, a factorial analysis of variance revealed a significant effect of the interaction between model colour and floral scent on the wasps' visits to flower models. The floral scent induced starved and non-starved wasps to visit yellow and green models about twice as often as without the scent. Ascogaster mimetica Viereck is an egg-larval parasitoid that targets Sparganothis sulfureana Clemens, a major cranberry pest in North America. While previous studies have shown that other Ascogaster species respond to cues from their hosts' eggs, it remains unknown whether A. mimetica utilizes these cues to recognize S. sulfureana. We hypothesized that female A. mimetica recognizes S. sulfureana from another cranberry pest, Choristoneura parallela Robinson, based on the presence of surface cues on eggs. To test this, we observed female A. mimetica behavior when exposed to eggs from its host, S. sulfureana; a nonhost, C. parallela; and eggs of S. sulfureana that were washed clean with hexane. Additionally, we tracked parasitism rates in each group. Our results revealed that A. mimetica spent 9.5 times longer walking when exposed to C. parallela eggs and 6 times longer when exposed to hexane-washed S. sulfureana eggs compared to unwashed S. sulfureana eggs. Also, A. mimetica spent 3 times longer grooming when exposed to hexane-washed than unwashed S. sulfureana eggs. In contrast, females spent 6 and 18 times longer drumming and probing/ovipositing on unwashed S. sulfureana eggs than on C. parallela eggs and 5 times longer probing/ovipositing on unwashed S. sulfureana eggs than on hexane-washed S. sulfureana eggs. Higher parasitism rates were observed from unwashed S. sulfureana eggs compared to those from C. parallela eggs and hexane-washed S. sulfureana eggs. One of the challenges in augmentative biological control programs is the definition of releasing strategy for natural enemies, especially when macro-organisms are involved. Important information about the density of insects to be released and frequency of releases usually requires a great number of experiments, which implies time and space that are not always readily available. In order to provide science-based responses for these questions, computational models offer an in silico option to simulate different biocontrol agent releasing scenarios. This allows decision-makers to focus their efforts to more feasible options. The major insect pest in sugarcane crops is the sugarcane borer Diatraea saccharalis, which can be managed using the egg parasitoid Trichogramma galloi. The current strategy consists in releasing 50,000 insects per hectare for each release, in three weekly releases. Here, we present a simulation model to check whether this releasing strategy is optimal against the sugarcane borer. A sensitive analysis revealed that the population of the pest is more affected by the number of releases rather than by the density of parasitoids released. Only the number of releases demonstrated an ability to drive the population curve of the pest towards a negative growth. For example, releasing a total of 600,000 insects per hectare in three releases led to a lower pest control efficacy that releasing only 250,000 insects per hectare in five releases. A higher number of releases covers a wider range of time, increasing the likelihood of releasing parasitoids at the correct time given that the egg stage is short. Based on these results, it is suggested that, if modifications to the releasing strategy are desired, increasing the number of releases from 3 to 5 at weekly intervals is most likely preferable., (© 2024. The brinjal fruit and shoot borer (BFSB), Leucinodes orbonalis Guenée (Lepidoptera: Crambidae), is a very detrimental pest that causes significant economic losses to brinjal crop worldwide. Infested brinjal fruits were collected from vegetable fields located at the ICAR-Indian Agricultural Research Institute (ICAR-IARI), New Delhi, India, during two consecutive seasons (2021-2022). The larvae of the pest were brought to the laboratory and reared under controlled conditions of 25 ± 0.5 °C and 70 ± 5% relative humidity, for the emergence of parasitoids. In addition, the survey of Hymenoptera parasitoids in brinjal was conducted utilizing a sweep net and yellow pan trap over the course of two seasons. The results reveal that five parasitoid species were emerged from L. orbonalis viz., Apanteles hemara Nixon, 1965, Bracon greeni Ashmead 1896 (Hymenoptera: Braconidae), Goryphus nursei (Cameron, 1907), Trathala flavoorbitalis (Cameron, 1907) (Hymenoptera: Ichneumonidae) and Spalangia gemina Boucek 1963 (Hymenoptera: Spalangiidae). Out of these, A. hemara and S. gemina were documented as new occurrences in Delhi. Additionally, A. hemara was recorded for the first time as a parasite on L. orbonalis . Trathala flavoorbitalis was observed during both seasons and exhibited higher parasitism reaching 15.55% and 18.46% in July and August 2022, respectively. However, the average parasitism (%) recorded by A. hemara , B. greeni , G. nursei , T. flavoorbitalis and S. gemina was 3.10%, 1.76%, 1.10%, 9.28% and 1.20% respectively. Furthermore, the findings showed a significant ( p ≤ 0.01) strongly positive correlation between fruit infestation (%) by L. orbonalis and parasitism (%). The survey indicates the presence of a broad group (19 families and 60 species) of Hymenoptera parasitoids in the brinjal crop ecosystem in Delhi which could be valuable in biological control. This study aims to explore the composition of natural enemy species in the fall webworm, Hyphantria cunea (Drury) population and the dynamics of its natural enemy community in Dandong, Liaoning Province, China, where it was first reported. We collected the natural enemy of eggs, larvae, and pupae of H. cunea on host trees at 12 survey sites from June 2019 to October 2020. The results showed that the community consists of 34 species: 20 predatory species, including 15 spiders and 5 insects, and 14 parasitic species, including 10 parasitic wasps and 4 parasitic flies. The top 3 dominant species based on the importance value index for both parasitic and predatory species were Pediobius pupariae > Chouioia cunea > Cotesia gregalis in the natural enemy community of H. cunea. Analysis of all 3 principal components by principal component analysis showed that Clubionidae sp. 1, Parena cavipennis, or other predators were the main factors affecting the natural enemy community. Analysis of the community structure parameters of the H. cunea natural enemy community in different developmental stages across generations revealed the following: (i) Compared with the degree of complexity of the egg and pupal stages, the larval stage was the highest. (ii) The complexity was determined by means of comprehensive evaluation: first-generation larvae in 2020 > first-generation larvae in 2019 > second-generation larvae in 2020 > second-generation larvae in 2019. Parasitic Trichogramma chilonis Ishii, an egg parasitoid of Grapholita molesta, is a critical agent for biological control of insect pests in crop plants. However, the efficiency of T. chilonis is influenced by its resistance to the common pesticide chlorantraniliprole. To elucidate the chlorantraniliprole detoxification mechanism, differentially expressed genes (DEGs) related to chlorantraniliprole resistance were studied at different developmental stages of the wasp. Individuals of T. chilonis were grouped and treated with chlorantraniliprole at different developmental stages. Untreated wasps were used as controls. Transcriptomic analysis identified the DEGs associated with chlorantraniliprole resistance and detoxification in T. chilonis. A total of 1,483 DEGs were associated with chlorantraniliprole resistance at all developmental stages. DEGs that correlated with chlorantraniliprole sensitivity of T. chilonis at different developmental stages were distinct and had various functions. The newly identified DEGs are involved in cytochrome P450- and glutathione metabolism-related pathways, which were predicted to contribute to chlorantraniliprole detoxification. Chlorantraniliprole detoxification by T. chilonis was associated with cytochrome P450- and glutathione-related pathways. Tuta absoluta is one of the most devastating pests of Solanaceae crops in Africa. We previously demonstrated the efficacy of Metarhizium anisopliae isolates ICIPE 18, ICIPE 20 and ICIPE 665 against adult T. absoluta. However, adequate strain selection and accurate spatial prediction are fundamental to optimize their efficacy and formulations before field deployment. This study therefore assessed the thermotolerance, conidial yield and virulence (between 15 and 35 °C) of these potent isolates. Over 90% of conidia germinated at 20, 25 and 30 °C while no germination occurred at 15 °C. Growth of the three isolates occurred at all temperatures, but was slower at 15, 33 and 35 °C as compared to 20, 25 and 30 °C. Optimum temperatures for mycelial growth and spore production were 30 and 25 °C, respectively. Furthermore, ICIPE 18 produced higher amount of spores than ICIPE 20 and ICIPE 665. The highest mortality occurred at 30 °C for all the three isolates, while the LT 50 values of ICIPE 18 and ICIPE 20 were significantly lower at 25 and 30 °C compared to those of ICIPE 665. Subsequently, several nonlinear equations were fitted to the mortality data to model the virulence of ICIPE 18 and ICIPE 20 against adult T. absoluta using the Entomopathogenic Fungi Application (EPFA) software. Spatial prediction revealed suitable locations for ICIPE 18 and ICIPE 20 deployment against T. absoluta in Kenya, Tanzania and Uganda. Some DNA viruses infect host animals usually by integrating their DNAs into the host genome. However, the mechanisms for integration remain largely unknown. Here, we find that Cotesia vestalis bracovirus (CvBV), a polydnavirus of the parasitic wasp C. vestalis (Haliday), integrates its DNA circles into host Plutella xylostella (L.) genome by two distinct strategies, conservatively and randomly, through high-throughput sequencing analysis. We confirmed that the conservatively integrating circles contain an essential "8+5" nucleotides motif which is required for integration. Then we find CvBV circles are integrated into the caterpillar's genome in three temporal patterns, the early, mid and late stage-integration. We further identify that three CvBV-encoded integrases are responsible for some, but not all of the virus circle integrations, indeed they mainly participate in the processes of early stage-integration. Strikingly, we find two P. xylostella retroviral integrases (PxIN1 and PxIN2) are highly induced upon wasp parasitism, and PxIN1 is crucial for integration of some other early-integrated CvBV circles, such as CvBV&#95;04, CvBV&#95;12 and CvBV&#95;24, while PxIN2 is important for integration of a late-integrated CvBV circle, CvBV&#95;21. Our data uncover a novel mechanism in which CvBV integrates into the infected host genome, not only by utilizing its own integrases, but also by recruiting host enzymes. Different factitious hosts were used to mass rear Trichogramma japonicum Ashmead in different parts of the globe because thorough details were lacking in both the laboratory and the field. The objective of this study was to compare, parasitoid, T. japonicum reared in different factitious hosts. Three commonly used factitious host eggs, Corcyra cephalonica (Stainton), Ephestia kuehniella Zeller and Sitotroga cerealella Olivier were tested under laboratory conditions and then in the field over a yellow stem borer, Scirpophaga incertulus (Walker) of rice. The highest parasitism by T. japonicum was observed on E. kuehniella eggs. The parasitoid's highest emergence (88.99%) was observed on S. cerealella eggs at 24 h exposure, whereas at 48 h it was on E. kuehniella eggs (94.66%). Trichogramma japonicum females that emerged from E. kuehniella eggs were significantly long-lived. The days of oviposition by hosts and the host species were significant individually, but not their interaction. Higher proportions of flying T. japonicum were observed when reared on E. kuehniella and C. cephalonica eggs. Field results showed that T. japonicum mass-reared on E. kuehniella showed higher parasitism of its natural host, S. incertulus eggs. Hence, by considering these biological characteristics and field results, E. kuehniella could be leveraged for the mass rearing of quality parasitoids of T. japonicum in India, the Asian continent and beyond., Competing Interests: The authors have declared that no competing interests exist.

Entomopathogenic nematodes (EPNs) have been successfully applied as biological control agents against above ground and soil stages of insect pests. However, for commercial application, it is crucial to mass culture these nematodes using in vitro liquid culture technology, as it is not attainable when using susceptible insects as hosts. Lobesia vanillana (Lepidoptera: Tortricidae) is regarded a sporadic pest of wine grapes in South Africa. The in vivo- and in vitro-cultured South African EPNs, Steinernema yirgalemense and Steinernema jeffreyense (Rhabditida: Steinernematidae), were evaluated against larvae and pupae of L. vanillana in laboratory bioassays. For larvae, high mortality was observed for all treatments: In vitro-cultured S. yirgalemense (98%) performed better than S. jeffreyense (73%), while within in vivo cultures, there was no difference between nematode species (both 83%). No significant difference was detected between in vivo- and in vitro cultures of the same nematode species. The LD50 of the in vitro-cultured S. yirgalemense, was 7.33 nematodes per larva. Mortality by infection was established by dissecting L. vanillana cadavers and confirming the presence of nematodes, which was > 90% for all treatments. Within in vitro cultures, both S. yirgalemense and S. jeffreyense were able to produce a new cohort of infective juveniles from L. vanillana larvae. Pupae, however, were found to be considerably less susceptible to EPN infection. This is the first study on the use of EPNs to control L. vanillana. The egg parasitoid Trichogramma evanescens Westwood is considered as an efficient biological control agent for managing several lepidopteran pests and it is widely distributed throughout the world. Mass rearing protocols of parasitoids that are currently in use in biocontrol programs require a meticulous quality control plan, in order to optimize their efficacy, but also their progeny production capacity. In this paper, the effect of different factors on the quality control in mass rearing of T. evenescens, using Plodia interpunctella (Hübner) and Galleria mellonella L. as host species, were investigated. The impact of egg agewas significant in the rates of parasitism, for both host species tested. Significantly highest percent of parasitoid emergence was noticed in two day-old eggs for both host species, while one day-old eggs day exhibited the maximum emergence when both species were used togetherin the same trials. Age-dependent storage egg preservation at either 4 or 9°C significantly influenced the parasitism percentages on both species. The highest parasitism percentage was recorded in two day-old G. mellonella eggs that are kept for 15 days at 9°C while the lower in one day-old P. interpunctella eggs for 60 d storage. Moreover, the highest parasitoid mortality was recorded in T. evanescens reared either on P. interpunctella or G. mellonella at 20°C. Rearing of the parasitoid on a mixture of eggs of both host species resulted in higher parasitism, but not always in higher rates of parasitoid emergence. The exopolysaccharide galactosaminogalactan (GAG) has been well characterized in Aspergilli, especially the human pathogen Aspergillus fumigatus. It has been found that a five-gene cluster is responsible for GAG biosynthesis in Aspergilli to mediate fungal adherence, biofilm formation, immunosuppression or induction of host immune defences. Herein, we report the presence of the conserved GAG biosynthetic gene cluster in the insect pathogenic fungus Metarhizium robertsii to mediate either similar or unique biological functions. Deletion of the gene cluster disabled fungal ability to produce GAG on germ tubes, mycelia and appressoria. Relative to the wild type strain, null mutant was impaired in topical infection but not injection of insect hosts. We found that GAG production by Metarhizium is partially acetylated and could mediate fungal adherence to hydrophobic insect cuticles, biofilm formation, and penetration of insect cuticles. In particular, it was first confirmed that this exopolymer is responsible for the formation of appressorium mucilage, the essential extracellular matrix formed along with the infection structure differentiation to mediate cell attachment and expression of cuticle degrading enzymes. In contrast to its production during A. fumigatus invasive growth, GAG is not produced on the Metarhizium cells harvested from insect hemocoels; however, the polymer can glue germ tubes into aggregates to form mycelium pellets in liquid culture. Therophilus javanus (Bhat & Gupta) is an exotic larval endoparasitoid newly imported from Asia into Africa as a classical biological control agent against the pod borer Maruca vitrata (Fabricius). The parasitoid preference for the five larval instars of M. vitrata and their influence on progeny sex ratio were assessed together with the impact of larval host age at the time of oviposition on development time, mother longevity and offspring production. In a choice situation, female parasitoids preferred to oviposit in the first three larval instars. The development of immature stages of the parasitoid was observed inside three-day-old hosts, whereby the first two larval instars of T. javanus completed their development as endoparasites and the third larval instar as ectoparasite. The development time was faster when first larval instars (two- and three-day-old) of the host caterpillars were parasitized compared to second larval instar (four-day-old). The highest proportion of daughters (0.51) was observed when females were provided with four-day-old hosts. The lowest intrinsic rate of increase (r) (0.21 ± 0.01), the lowest rate of increase (λ) (1.23 ± 0.01), and the lowest net reproductive rate (Ro) (35.93 ± 6.51) were recorded on four-day-old hosts. These results are discussed in the light of optimizing mass rearing and release strategies.

Background: Steinernema feltiae is an entomopathogenic nematode used in biological control programs with a global distribution. Populations of this species show phenotypic plasticity derived from local adaptation and vary in different traits, such as location and host penetration. The aim of this work was to describe a Chilean isolate of this nematode species, using integrative approaches., Methods: Nematode morphological and morphometric studies were conducted along with molecular analysis of nuclear genes. The symbiotic bacterium was also identified by sequencing the 16S rRNA gene. Some ecological characteristics were described, including the temperature requirements for the nematode life cycle and the effect of soil water content for optimal reproduction., Results: Morphometric characterization revealed a large intra-specific variability. The isolate identity was also corroborated with the analysis of nuclear genes. Based on the 16S gene, its symbiont bacteria, Xenorhabdus bovienii, was identified. The lowest, optimal and highest temperatures found to limit the infestation and reproduction on Galleria mellonella were 10, 20 and 30 °C, respectively; the emergence from the host larvae occurred approximately 10 days after inoculation. Differences were observed in offspring, and 120 infective juveniles (IJ)/larva was the most prolific dose at 20 °C. The soil water content did not affect the number of IJ invaders, penetration efficacy and IJ emergence time or offspring per larva, but it caused a delay in achieving full mortality at the permanent wilting point with respect to saturation and field capacity., Conclusions: For the first time, a Chilean isolate of S. feltiae is described in detail considering morphological, molecular and ecological aspects. The isolate was shown to be efficient in soil containing water, with optimal temperatures ranging from 15 to 25 °C for host infestation and production of an abundant offspring; these characteristics would allow its potential use as control agents in a wide geographical area of the country.

Busseola fusca (Fuller) (Lepidoptera: Noctuidae), the maize stalk borer, is a widespread crop pest in sub-Saharan Africa that has been the focus of biological research and intensive management strategies. Here, we present a comprehensive annotated transcriptome of B. fusca (originally collected in the Western Province of Kenya) based on ten pooled libraries including a wide array of developmental stages, tissue types, and exposures to parasitoid wasps. Parasitoid wasps have been used as a form of biocontrol to try and reduce crop losses with variable success, in part due to differential infectivities and immune responses among wasps and hosts. We identified a number of loci of interest for pest management, including genes potentially involved in chemoreception, immunity, and response to insecticides. The comprehensive sampling design used expands our current understanding of the transcriptome of this species and deepens the list of potential target genes for future crop loss mitigation, in addition to highlighting candidate loci for differential expression and functional genetic analyses in this important pest species., (© The Author(s) 2020. In this study, a microsporidian pathogen of the date moth (Apomyelois (Ectomyelois) ceratoniae, Zeller, 1839) also known as the carob moth, is described based on light microscopy, ultrastructural characteristics and comparative molecular analysis. The pathogen infects the gut and hemolymph of A. ceratoniae. All development stages are in direct contact with the host cell cytoplasm. Fresh spores with nuclei arranged in a diplokaryon are oval and measured 3.29 ± 0.23 μm (4.18-3.03 μm, n = 200) in length and 1.91 ± 0.23 μm (2.98-1.66 μm, n = 200) in width. Spores stained with Giemsa's stain measured 3.11 ± 0.31 μm (3.72-2.41 μm, n = 150) in length and 1.76 ± 0.23 μm (2.16-1.25 μm, n = 150) in width. Spores have an isofilar polar filament with 10-12 coils. An 1110 bp long alignment of the current microsporidium showed an SSU rRNA gene difference of only 0.0009, corresponding to >99.91% sequence similarity with Nosema fumiferanae, while RPB1 gene sequences were 98.03% similar within an alignment of 969 bp. All morphological, ultrastructural and molecular features indicate that the microsporidian pathogen of A. ceratoniae is the new isolate of the N. fumiferanae and is named here as Nosema fumiferanae TY61.

Bacillus thuringiensis Berliner subsp. kurstaki (Btk) and Habrobracon hebetor Say are both biological control agents of Helicoverpa armigera Hubner. The present study evaluated their compatibility for combined application against this pest by examining the acceptability of Btk-inoculated hosts for H. hebetor females and testing for negative life-history impacts on developing progeny. Second-instar H. armigera larvae fed for 72 h on potted chickpea plants treated with three concentrations of Btk (LC15, LC35, and LC70) and were then used in bioassays of parasitoid development and parasitism behavior. Survival of parasitoids was significantly reduced, and immature development prolonged, on hosts fed chickpea plants treated with LC35 and LC70  Btk, but not on plants treated with LC15  Btk. Parasitoids failed to discriminate against hosts treated with LC15 or LC35  Btk in choice tests, but attacked fewer hosts treated with LC70  Btk, paralyzing and parasitizing more healthy hosts, and laying more eggs on them. In contrast, a no-choice test revealed that more hosts treated with LC35 and LC70  Btk were paralyzed compared with control or LC15-treated hosts, but the numbers of hosts parasitized and eggs laid did not vary among Btk treatments. Thus, females required an experience with healthy hosts, as they had in the choice test, to discriminate against diseased ones. A parasitoid's decision to reject or accept a potential host is fundamental to its fitness. Superparasitism, in which more than one egg of a given parasitoid species can deposit in a single host, is usually considered sub-optimal in systems where the host is able to support the development of only a single parasitoid. It follows that selection pressure may drive the capacity for parasitoids to recognize parasitized hosts, especially if there is a fitness cost of superparasitism. Here, we used microsatellite studies of two distinct populations of Cotesia vestalis to demonstrate that an egg laid into a diamondback moth (Plutella xylostella) larva that was parasitized by a conspecific parasitoid 10 min, 2 or 6 h previously was as likely to develop and emerge successfully as was the first-laid egg. Consistent with this, a naive parasitoid encountering its first host was equally likely to accept a healthy larva as one parasitized 10 min prior, though handling time of parasitized hosts was extended. For second and third host encounters, parasitized hosts were less readily accepted than healthy larvae. If 12 h elapsed between parasitism events, the second-laid egg was much less likely to develop. Discrimination between parasitized and healthy hosts was evident when females were allowed physical contact with hosts, and healthy hosts were rendered less acceptable by manual injection of parasitoid venom into their hemolymph. Collectively, these results show a limited capacity to discriminate parasitized from healthy larvae despite a viability cost associated with failing to avoid superparasitism.

Tritrophic interactions allow plants to recruit natural enemies for protection against herbivory. Here we investigated genetic variability in induced responses to stemborer egg-laying in maize Zea mays (L.) (Poaceae). We conducted a genome wide association study (GWAS) of 146 maize genotypes comprising of landraces, inbred lines and commercial hybrids. Plants were phenotyped in bioassays measuring parasitic wasp Cotesia sesamiae (Cameron) (Hymenoptera: Braconidae) attraction to volatiles collected from plants exposed to stemborer Chilo partellus (Swinhoe) (Lepidoptera: Crambidae) eggs. Genotyping-by-sequencing was used to generate maize germplasm SNP data for GWAS. The egg-induced parasitoid attraction trait was more common in landraces than in improved inbred lines and hybrids. GWAS identified 101 marker-trait associations (MTAs), some of which were adjacent to genes involved in the JA-defence pathway (opr7, aos1, 2, 3), terpene biosynthesis (fps3, tps2, 3, 4, 5, 7, 9, 10), benzoxazinone synthesis (bx7, 9) and known resistance genes (e.g. maize insect resistance 1, mir1). Intriguingly, there was also association with a transmembrane protein kinase that may function as a receptor for the egg elicitor and other genes implicated in early plant defence signalling. We report maize genomic regions associated with indirect defence and provide a valuable resource for future studies of tritrophic interactions in maize. The markers identified may facilitate selection of indirect defence by maize breeders.

Female parasitoid wasps normally inject virulence factors together with eggs into their host to counter host immunity defenses. A newly identified RhoGAP protein in the venom of Microplitis mediator compromises the cellular immunity of its host, Helicoverpa armigera. RhoGAP1 proteins entered H. armigera hemocytes, and the host cellular cytoskeleton was disrupted. Depletion of MmGAP1 by injection of dsRNA or antibody increased the wasp egg encapsulation rate. An immunoprecipitation assay of overexpressed MmGAP1 protein in a Helicoverpa cell line showed that MmGAP1 interacts with many cellular cytoskeleton associated proteins as well as Rho GTPases. A yeast two-hybrid and a pull-down assay demonstrated that MmGAP1 interacts with H. armigera RhoA and Cdc42. Transgenic crops express Cry proteins exhibit high resistant to target insect pests. When we evaluate the effects of Cry proteins on the parasitoid of target insect pest via tritrophic experiments (transgenic plant-target insect pest-parasitoid) host quality of parasitoids might decrease because of insecticidal protein ingestion, this would cause host-quality mediated effects and influence the accuracy of biosafety assessment. In the current study, high dose of Cry2Aa protein was injected into the hemolymph of Plodia interpunctella by microinjection, and the hemolymph was used as the carrier to deliver Cry protein to Habrobracon hebetor, which has been previously reported as an ectoparasitoid of P. interpunctella larval, in order to avoid the "host-quality mediated effects". Results showed that injected Cry2Aa remained at high concentration and bioactive in the hemolymph of P. interpunctella parasitized by H. hebetor, the hemolymph of P. interpunctella could be used as carriers of Cry protein to H. hebetor, and high dose of Cry2Aa have no negative impacts on the development time, weight of pupa, sex ratio, adults weight (male and female), adult longevity and fecundity, and the activity of stress-related enzymes of H. hebetor. However, the hemolymph of P. interpunctella injected into Galanthus nivalis agglutinin (the positive control) showed significant negative impact on these parameters measured in the present study of H. hebetor. This indicated that Cry2Aa protein had no detrimental effects on the biological parameters of H. hebetor measured in the current study. The use of nectar-providing plants to nourish natural enemies of pest species has become a widely-used approach in conservation biological control to reduce pest damage without the indiscriminate use of insecticides. Choice of plant species is crucial to maximize benefits, but suitable species are yet to be identified for many important crop-pest systems. Here we explored the suitability of three candidate nectar plants for use in brassica vegetables to suppress the globally significant pest, Plutella xylostella L. (Lepidoptera: Plutellidae), using the widely-distributed parasitoid, Cotesia vestalis (Haliday) (Hymenoptera: Braconidae). Volatiles of alyssum (Lobularia maritima (L.) Desv) (Brassicaceae) were attractive to the parasitoid and access to flowering shoots increased adult longevity and realized fecundity of C. vestalis. Moreover, adult diamondback moth derived no benefit from this flower. In contrast, buckwheat (Fagopyrum esculentum Moench) (Polygonaceae), a species widely used in conservation biological control in other systems, increased the longevity and fecundity of both pest and parasitoid, rendering it less suitable. A third plant, heronsbill (Portulaca grandiflora Hook.) (Portulacaceae) denied benefit to the pest and promoted longevity of the parasitoid under no-choice conditions but did not improve fecundity and was repellent to female parasitoids under choice conditions. The contrasting effects of this set of plants illustrate the need to test multiple response variables and effects on both pest and natural enemy when seeking optimal nectar plants for use in a novel conservation biological control system.

Insect parasitic nematodes have acquired mechanisms to evade their host immune response for successful parasitism. Despite the importance of understanding of the evolution of evasion mechanisms from host immunity, insect immune response against non-parasitic nematodes has not been well studied. In our previous study, we demonstrated that a non-insect parasitic nematode Caenorhabditis elegans was not encapsulated by haemocytes in the larvae of the greater wax moth Galleria mellonella. To understand how nematodes influence insect haemocytes to escape encapsulation, we examined the effect of C. elegans on haemocytes in the haemocoel of G. mellonella larvae. Injection of nematodes resulted in the decrease of haemocyte density while mortality and spreading ability of haemocytes, the haematopoietic organs were not affected. In vitro co-incubation of haemocytes with nematodes resulted in a decrease of haemocyte density and we observed feeding on haemocytes by nematodes. Injection of C. elegans feeding-delay mutants into insects did not cause the decrease of haemocyte density. The decrease of haemocyte density was due to the nematode's ingestion of haemocytes. Furthermore, an entomopathogenic nematode and other bacterial feeding nematodes also showed similar feeding behaviour. The nematode's ability to feed on haemocytes may have played an important role in the evolution of nematode parasitism in bacterial-feeding nematodes.

Background: The holistic view of bacterial symbiosis, incorporating both host and microbial environment, constitutes a major conceptual shift in studies deciphering host-microbe interactions. Interactions between Steinernema entomopathogenic nematodes and their bacterial symbionts, Xenorhabdus, have long been considered monoxenic two partner associations responsible for the killing of the insects and therefore widely used in insect pest biocontrol. We investigated this "monoxenic paradigm" by profiling the microbiota of infective juveniles (IJs), the soil-dwelling form responsible for transmitting Steinernema-Xenorhabdus between insect hosts in the parasitic lifecycle., Results: Multigenic metabarcoding (16S and rpoB markers) showed that the bacterial community associated with laboratory-reared IJs from Steinernema carpocapsae, S. feltiae, S. glaseri and S. weiseri species consisted of several Proteobacteria. The association with Xenorhabdus was never monoxenic. We showed that the laboratory-reared IJs of S. carpocapsae bore a bacterial community composed of the core symbiont (Xenorhabdus nematophila) together with a frequently associated microbiota (FAM) consisting of about a dozen of Proteobacteria (Pseudomonas, Stenotrophomonas, Alcaligenes, Achromobacter, Pseudochrobactrum, Ochrobactrum, Brevundimonas, Deftia, etc.). We validated this set of bacteria by metabarcoding analysis on freshly sampled IJs from natural conditions. We isolated diverse bacterial taxa, validating the profile of the Steinernema FAM. We explored the functions of the FAM members potentially involved in the parasitic lifecycle of Steinernema. Two species, Pseudomonas protegens and P. chlororaphis, displayed entomopathogenic properties suggestive of a role in Steinernema virulence and membership of the Steinernema pathobiome., Conclusions: Our study validates a shift from monoxenic paradigm to pathobiome view in the case of the Steinernema ecology. The microbial communities of low complexity associated with EPNs will permit future microbiota manipulation experiments to decipher overall microbiota functioning in the infectious process triggered by EPN in insects and, more generally, in EPN ecology.

Koinobiont parasitoids regulate the physiology of their hosts, possibly interfering with the host gut microbiota and ultimately impacting parasitoid development. We used the parasitoid Cotesia flavipes to investigate if the regulation of the host would also affect the host gut microbiota. We also wondered if the effects of parasitization on the gut microbiota would depend on the host-parasitoid association by testing the permissive Diatraea saccharalis and the non-permissive Spodoptera frugiperda hosts. We determined the structure and potential functional contribution of the gut microbiota of the fore-midgut and hindgut of the hosts at different stages of development of the immature parasitoid. The abundance and diversity of operational taxonomic units of the anteromedial (fore-midgut) gut and posterior (hindgut) region from larvae of the analyzed hosts were affected by parasitization. Changes in the gut microbiota induced by parasitization altered the potential functional contribution of the gut microbiota associated with both hosts. Our data also indicated that the mechanism by which C. flavipes interferes with the gut microbiota of the host does not require a host-parasitoid coevolutionary history. Tobacco is one of the major industrial crops cultivated worldwide. Chemical control is the main method employed to reduce damage by insect pests. The use of entomopathogenic fungi represents an alternative to replace insecticides. The search for effective strains in the field constitutes a first step when developing a formulation. The objective of this work was to study genetic differences among isolates of entomopathogenic fungi obtained from tobacco grown soils using ISSR markers. The pathogenicity of the strains towards Helicoverpa gelotopoeon and Diabrotica speciosa was also assessed in order to search for a relationship between virulence and genetic diversity. Nineteen isolates were identified according to morphological features and molecular techniques as Beauveria bassiana (11) and Purpureocillium lilacinum (8). The diversity tree generated by ISSR analysis showed a high diversity among the strains. The pathogenicity towards H. gelotopoeon and D. speciosa was assessed and the logistic models generated showed that B. bassiana isolates LPSc1215 and LPSc1364 were the most pathogenic against both insect pests tested. In the diversity tree, these strains were grouped in a same cluster with a similarity level of approximately 85%, indicating a possible relationship between virulence and the band pattern generated.

A new species of bethylid parasitoid wasp, Goniozus omanensis Polaszek sp. n., is described based on morphology and DNA sequence data. The species is currently known only from the lesser date moth Batrachedra amydraula, a pest of economic importance, but can be reared on two factitious host species. G. omanensis is compared with G. swirskiana, known from the same host in Israel. Background: Parasitic insects are well-known biological control agents for arthropod pests worldwide. They are capable of regulating their host's physiology, development and behaviour. However, many of the molecular mechanisms involved in host-parasitoid interaction remain unknown., Results: We sequenced the genomes of two parasitic wasps (Cotesia vestalis, and Diadromus collaris) that parasitize the diamondback moth Plutella xylostella using Illumina and Pacbio sequencing platforms. Genome assembly using SOAPdenovo produced a 178 Mb draft genome for C. vestalis and a 399 Mb draft genome for D. collaris. A total set that contained 11,278 and 15,328 protein-coding genes for C. vestalis and D. collaris, respectively, were predicted using evidence (homology-based and transcriptome-based) and de novo prediction methodology. Phylogenetic analysis showed that the braconid C. vestalis and the ichneumonid D. collaris diverged approximately 124 million years ago. These two wasps exhibit gene gains and losses that in some cases reflect their shared life history as parasitic wasps and in other cases are unique to particular species. Gene families with functions in development, nutrient acquisition from hosts, and metabolism have expanded in each wasp species, while genes required for biosynthesis of some amino acids and steroids have been lost, since these nutrients can be directly obtained from the host. Both wasp species encode a relative higher number of neprilysins (NEPs) thus far reported in arthropod genomes while several genes encoding immune-related proteins and detoxification enzymes were lost in both wasp genomes., Conclusions: We present the annotated genome sequence of two parasitic wasps C. vestalis and D. collaris, which parasitize a common host, the diamondback moth, P. xylostella. These data will provide a fundamental source for studying the mechanism of host control and will be used in parasitoid comparative genomics to study the origin and diversification of the parasitic lifestyle.

The sugarcane borer, Diatraea saccharalis is one of the hosts more used for parasitoid pupal multiplication in Brazil. The parasitoids pupal of Trichospilus diatraeae and Palmistichus elaeisis are generalist natural enemies with potential to suppress populations of diverse families of lepidopteran pests. The success in the utilization of these natural enemies in the field is directly related to the capacity of search of the host, this capacity might be affected by the presence of the pathogens. In this context, the aim of this essay was to detect the presence of intracellular parasites of Phylum Microsporidia. These pathogens may cause morphological and behavioral alterations. The presence of infection was verified by microscopy and was confirmed by amplification of region small subunit (SSU) of ribosomal RNA using universal primers for microsporidia of Nosema sp. The purified PCR products were submitted to sequencing, and the sequences that had been obtained were edited and aligned with the sequences in a Genbank database. In this way, it was possible to verify the presence of intracellular parasites in T. diatraeae, P. elaeisis and D. saccharalis pertaining to Clade Nosema/Vairimorpha. However, this is the first one report about detection of the microsporidia in the parasitoids T. diatraeae and P. elaeisis.

Trichogramma spp., among the most common parasitoids used for augmentation biological control, often are mass-reared on eggs of the Mediterranean flour moth, Ephestia kuehniella (Zeller). To evaluate removal of nutritional components from the E. kuehniella larval diet and reduce production costs, colonies were maintained using one of three diets: a standard diet consisting of eight ingredients, a reduced diet containing whole wheat flour, glycerol, and Brewer's yeast, or a third minimal diet of only whole wheat flour. The standard diet sustained the fastest larval development, female pupae with the greatest mass, the highest level of adult emergence, and production of the most eggs per female. Eggs from moths reared as larvae on the standard or reduced diet had equivalent mass, length, and percent hatch. Females from larvae fed the minimal diet produced eggs with the least mass that were shorter and had the lowest percent hatch. Eggs from the three E. kuehniella colonies were exposed separately to Trichogramma brassicae Bezdenko females to determine their acceptance for oviposition. More of the eggs from the standard diet were parasitized by the females, eggs from the reduced and minimal diets being less acceptable. The percent emergence of the parasitoids was the same regardless of diet; however, the largest wasps emerged from the standard diet eggs and a greater proportion of them were females. The tomato leafminer, Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae), is a destructive pest of tomato that can cause up to 100% yield loss. The predatory bug Nabis pseudoferus (Remane) (Hemiptera: Nabidae) and the parasitoid Trichogramma brassicae (Bezdenko) (Hymenoptera: Trichogrammatidae) are natural enemies of this pest. Since the interaction between predators and parasitoids in different trophic levels including intraguild predation (IGP) can decrease or increase the efficiency of natural enemies, the effects of age-dependent parasitism of host eggs on IGP between these two species were investigated under laboratory conditions. In no-choice and choice preference tests, the predatory bug was exposed to 40 parasitized and nonparasitized eggs of different ages (24, 48, and 72 h old). Investigation of switching behavior was conducted using various combinations of tomato leafminer eggs (30:90, 45:75, 60:60, 75:45, and 90:30 nonparasitized:parasitized eggs) using eggs of different ages (24, 48, and 72 h old). In no-choice tests, the highest feeding rate of the predatory bug was 39.21 ± 0.36 eggs on 24-h-old nonparasitized eggs and the lowest feeding rate was 1.4 ± 0.80 eggs on 72-h-old parasitized eggs. In choice tests, comparison of the Manly's β indices indicated that the predatory bug preferred to feed on nonparasitized eggs with 48- and 72-h-old eggs, but there was no significant preference for the 24-h-old eggs. Results of switching test showed that the linear regression between Manly's β index and different ratios of nonparasitized eggs to parasitized and nonparasitized eggs was not significant in 72-h-old eggs. However, this regression was significant with 24- and 48-h-old eggs and the predator's preference was dependent upon the ratio of nonparasitized and parasitized tomato leafminer eggs. The rice moth, Corcyra cephalonica Stainton, an alternate host for the production of the parasitoid, Habrobracon hebetor Say, was reared on different diets, including pearl millet [Pennisetum glaucum (L.) R. Br.] (Poales: Poaceae) flour only, and in combinations of flours of sorghum [Sorghum bicolor (L.) Moench] (Poales: Poaceae), peanut (Arachis hypogea L.) (Fabales: Fabaceae), and cowpea [Vigna unguiculata (L.) Walp.] (Fabales: Fabaceae) to identify the optimal and economical proportion to be used under the conditions of Niger. The addition of cowpea or peanut to the pearl millet diet slightly increased C. cephalonica larval development time. Likewise, the addition of cowpea or peanut to cereal diets yielded a higher C. cephalonica larval survival. Female moths emerging from larvae fed on cereal and legume mixed diets produced higher eggs compared to the ones fed on sole and mixed cereals. Among legumes, cowpea addition is most interesting in terms of cost/production of C. cephalonica larvae. However, female moths emerging from larvae fed on different millet cowpea mix (5, 25, and 50%) laid significantly more eggs than those fed on sole pearl millet. Further, individual C. cephalonica larvae fed on 75% pearl millet + 25% cowpea produced significantly more H. hebetor. With an initial 25 C. cephalonica larvae kept for a 3-mo rearing period, the number of H. hebetor parasitoids produced will reach 2.68-10.07 million. Exorista larvarum (L.), a polyphagous gregarious larval parasitoid of lepidopterans, can be mass produced both in vivo, using the greater wax moth Galleria mellonella (L.) (Lepidoptera: Pyralidae) as a factitious host, and in vitro, on artificial media composed of crude components. The present study was focused on another aspect of E. larvarum rearing, namely the influence of adult food on parasitoid performance. The standard food, consisting of lump sucrose and cotton balls soaked in a honey and water solution (1), was compared with other foods or food combinations, namely lump sucrose alone (2), honey and water solution (3), sucrose and water solution either alone (4) or combined with bee-collected pollen (5), and, finally, pollen alone (6). All foods were provided together with distilled water supplied in drinking troughs. Based on the parameters considered (i.e., female longevity, number of eggs laid on host larvae, puparia obtained from eggs, and adults emerged from puparia), pollen alone was deemed to be the most suitable food for adult females of E. larvarum. In particular, the pollen showed a longevity-promoting effect, increasing the number of eggs laid on host larvae throughout the female lifespan. Background: Mermithid nematodes, such as Ovomermis sinensis, display a broad host range including some lepidopteran pests. Infective juveniles penetrate their host through the cuticle, complete their growth within the hemocoel and eventually kill the host upon their emergence. Hence, mermithid nematodes are considered potential biological control agents of insect pests. Our previous data indicate that the infection rate of O. sinensis on cotton bollworm (Helicoverpa armigera) is low, which may be largely due to the strong immune system of the host. However, current knowledge on the interactions of mermithid nematodes with their hosts and the mechanisms employed by hosts to defend themselves against mermithid nematodes is limited., Results: Here, we investigated the response of H. armigera to O. sinensis infection. Parasitism by O. sinensis caused a sharp decline in the survival rate of H. armigera. The hemocytic phagocytosis ability, antibacterial activity, and phenoloxidase (PO) activity in plasma of H. armigera increased at 1 d post parasitism (dpp) but decreased at 3 dpp. Further, we investigated gene expression in the fat body of parasitized and non-parasitized H. armigera larvae at 1, 3, and 5 dpp using a digital gene expression system. In total, 41, 60 and 68 immune-related differentially expressed genes were identified at 1, 3, and 5 dpp, respectively. These genes encoded pattern recognition receptors (PRRs), antimicrobial peptides (AMPs), serine proteases (SPs), SP inhibitors, mucins and other immune-related proteins. The expression of most PRRs, AMPs, SPs, and mucins was upregulated in the fat body of larvae at 1 dpp, downregulated at 3 dpp, and then again upregulated at 5 dpp by O. sinensis. The increased expression of SP inhibitors may contribute to the inhibited PO activity at 5 dpp., Conclusions: This study demonstrates that parasitism by O. sinensis modulates the immune reaction of the host H. armigera by altering the expression of immune-related genes. Our data provide a basis for future investigation of the molecular mechanisms employed by the mermithid nematode O. sinensis to modulate the immunity of the host H. armigera. These data will also likely facilitate the improvement of success in parasitism of H. armigera by O. sinensis.

Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) is a polyphagous pest with a wide geographic distribution. This pest first arrived in Brazil in 2013, and since then studies on possible control methods for it have been necessary. A possible method for the control of H. armigera is using the egg parasitoid Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae). Therefore, the objective of this study was to evaluate the performance of T. pretiosum on H. armigera eggs, which are known to represent suitable hosts for the development of this parasitoid species in the laboratory. Parasitism and emergence rates and the duration of the egg-to-adult period of T. pretiosum were investigated following 24- and 48-h exposures of this parasitoid to H. armigera and Corcyra cephalonica (Stainton) (Lepidoptera: Pyralidae) eggs. The longevity of offspring after the 24-h exposure was studied, as well as the frequency of parasitism and emergence, host preference, and the emergence of offspring from eggs of different ages or oviposited by lepidopterans on different days. Parasitism was 14.4 and 34.9% more frequent on C. cephalonica than on H. armigera after 24 and 48 h of exposure, respectively. In C. cephalonica, parasitism was 27.2% higher after 48 h. Parasitism was more frequent on C. cephalonica eggs collected on the second day of oviposition (76.2%), and on H. armigera on the third day of oviposition (71.1%). Parasitism frequency was lower on 2-day-old C. cephalonica eggs (63.3%) and on 3-day-old H. armigera eggs (41.3%). When tested with a chance of choice between hosts, T. pretiosum preferred H. armigera, while in the test with no chance of choice there was no difference in preference. Thus, T. pretiosum may be considered a tool for the Integrated Pest Management (IPM) of H. armigera.

The parasitoid Trichogramma species are indispensable natural enemies of many lepidopterans and it plays an important role in integrated pest management (IPM) programs throughout the world. Laboratory studies were conducted to compare the susceptibility of three Trichogramma egg parasitoid species to ten common insecticides and three herbicides. The adults of Trichogramma dendrolimi, T. chilonis, and T. ostriniae were exposed to the above-mentioned pesticides by a glass-vial residue method. Among the four neonicotinoids, dinotefuran and thiamethoxam exhibited extremely toxic effects on the Trichogramma dendrolimi and T. chilonis, with Risk Quotient (RQ) values ranging from 1471.2 to 5492.5. However, these two neonicotinoids have a relatively low toxicity to T. ostriniae, with RQ values 433.6 and 915.4, respectively. In addition, Imidacloprid and acetamiprid were slightly to moderately toxic to all the tested parasitic wasps and their RQ values are less than 500. For pyrethroids, all the selected compounds were slightly to moderately toxic to three Trichogramma species except that cyhalothrin was dangerously toxic to T. dendrolimi and T. chilonis, with RQ values 2567.6 and 3950.4. Among the three herbicides tested, pendimethalin, butralin and napropamid were slightly to moderately toxic to egg parasitoids, with all RQ values below 1000. For two avermectins, abamectin were slightly to moderately toxic to all three wasps with RQ values 635.6, 148.3 and 254.2, respectively. However, emamectin benzoate was found to be safe for the parasitoids. Furthermore, T. dendrolimi showed higher sensitivity than T. chilonis and T. ostriniae to the pesticides based on the comparison of LR 50 (application rate causing 50% mortality) values. Nematode parasites rely on successful host infection to perpetuate their species. Infection by individual nematode parasites can be risky, however; any one individual could be killed by the host's immune response. Here we use a model system to show that environmental cues and parasite past experience can be used by entomopathogenic nematodes to reduce individual risk of infection. Past parasite experience can more than double the infective virulence (number of host invaders) of a given cohort of entomopathogenic nematode parasites. This plasticity in individual parasite risk-taking and associated infection can be used to manage infection of parasitic nematodes: enhancing biological control with entomopathogenic nematodes and developing behavioral and chemical strategies to reduce infection by vertebrate and plant parasitic nematodes., Competing Interests: The authors have declared that no competing interests exist.

Parasitic wasps produce several factors including venom, polydnaviruses (PDVs) and specialized wasp cells named teratocytes that benefit the survival of offspring by altering the physiology of hosts. However, the underlying molecular mechanisms for the alterations remain unclear. Here we find that the teratocytes of Cotesia vestalis, an endoparasitoid of the diamondback moth Plutella xylostella, and its associated bracovirus (CvBV) can produce miRNAs and deliver the products into the host via different ways. Certain miRNAs in the parasitized host are mainly produced by teratocytes, while the expression level of miRNAs encoded by CvBV can be 100-fold greater in parasitized hosts than non-parasitized ones. We further show that one teratocyte-produced miRNA (Cve-miR-281-3p) and one CvBV-produced miRNA (Cve-miR-novel22-5p-1) arrest host growth by modulating expression of the host ecdysone receptor (EcR). Altogether, our results show the first evidence of cross-species regulation by miRNAs in animal parasitism and their possible function in the alteration of host physiology during parasitism.

Background: Trichogrammatids are minute parasitoid wasps that develop within other insect eggs. They are less than half a millimeter long, smaller than some protozoans. The Trichogrammatidae are one of the earliest branching families of Chalcidoidea: a diverse superfamily of approximately half a million species of parasitoid wasps, proposed to have evolved from a miniaturized ancestor. Trichogramma are frequently used in agriculture, released as biological control agents against major moth and butterfly pests. Additionally, Trichogramma are well known for their symbiotic bacteria that induce asexual reproduction in infected females. Knowledge of the genome sequence of Trichogramma is a major step towards further understanding its biology and potential applications in pest control., Results: We report the 195-Mb genome sequence of Trichogramma pretiosum and uncover signatures of miniaturization and adaptation in Trichogramma and related parasitoids. Comparative analyses reveal relatively rapid evolution of proteins involved in ribosome biogenesis and function, transcriptional regulation, and ploidy regulation. Chalcids also show loss or especially rapid evolution of 285 gene clusters conserved in other Hymenoptera, including many that are involved in signal transduction and embryonic development. Comparisons between sexual and asexual lineages of Trichogramma pretiosum reveal that there is no strong evidence for genome degradation (e.g., gene loss) in the asexual lineage, although it does contain a lower repeat content than the sexual lineage. Trichogramma shows particularly rapid genome evolution compared to other hymenopterans. We speculate these changes reflect adaptations to miniaturization, and to life as a specialized egg parasitoid., Conclusions: The genomes of Trichogramma and related parasitoids are a valuable resource for future studies of these diverse and economically important insects, including explorations of parasitoid biology, symbiosis, asexuality, biological control, and the evolution of miniaturization. Understanding the molecular determinants of parasitism can also inform mass rearing of Trichogramma and other parasitoids for biological control.

The family Ascoviridae is a recently described virus family whose members are transmitted by parasitoids and cause chronic and lethal infections in lepidopteran insects. Little is known about the biology and ecology of ascoviruses, and few isolates have been found outside the United States. We report here the isolation of a new ascovirus variant from Spodoptera litura in Japan. Full genome sequence and phylogenetic analyses showed that this virus was closely related to variants in Heliothis virescens ascovirus-3a, and it was named HvAV-3j. HvAV-3j has a DNA genome of 191 718 bp, with 189 putative ORFs and a GC content of 45.6 %, and is highly similar to HvAV-3h, which was isolated in China. In a field survey, the endoparasitoid Meteorus pulchricornis caused a high percentage of parasitization in populations of S. litura larvae, and under laboratory conditions M. pulchricornis was able to transmit HvAV-3j from infected to uninfected larvae by oviposition. Meteorus pulchricornis is thus likely to be a major vector for HvAV-3j transmission in Japan. This species is recognized here for the first time as a vector of ascoviruses that parasitizes a range of host species that extends across families.

Biological control requires specific tools for the accurate detection and identification of natural enemies in order to estimate variations in their abundance and their impact according to changes in environmental conditions or agricultural practices. Here, we developed two molecular methods of detection based on PCR-RFLP with universal primers and on PCR with specific primers to identify commonly occurring larval parasitoids of the tortricid fruit pests and to estimate parasitism in the codling moth. Both methods were designed based on DNA sequences of the COI mitochondrial gene for a range of parasitoids that emerged from Cydia pomonella and Grapholita molesta caterpillars (102 parasitoids; nine species) and a range of potential tortricid hosts (40 moths; five species) damaging fruits. The PCR-RFLP method (digestion by AluI of a 482 bp COI fragment) was very powerful to identify parasitoid adults and their hosts, but failed to detect parasitoid larvae within eggs or within young C. pomonella caterpillars. The PCR method based on specific primers amplified COI fragments of different lengths (131 to 463 bp) for Ascogaster quadridentata (Braconidae); Pristomerus vulnerator (Ichneumonidae); Trichomma enecator (Ichneumonidae); and Perilampus tristis (Perilampidae), and demonstrated a higher level of sensibility than the PCR-RFLP method. Molecular estimations of parasitism levels in a natural C. pomonella population with the specific primers did not differ from traditional estimations based on caterpillar rearing (about 60% parasitism in a non-treated apple orchard). These PCR-based techniques provide information about within-host parasitoid assemblage in the codling moth and preliminary results on the larval parasitism of major tortricid fruit pests., Competing Interests: The authors declare no conflict of interest.

Under predicted global climate change, species will be gradually exposed to warmer temperatures, and to a more variable climate including more intense and more frequent heatwaves. Increased climatic variability is expected to have different effects on species and ecosystems than gradual warming. A key challenge to predict the impact of climate change is to understand how temperature changes will affect species interactions. Herbivorous insects and their natural enemies belong to some of the largest groups of terrestrial animals, and thus they have a great impact on the functioning of ecosystems and on the services these ecosystems provide. Here we studied the life history traits of the plant-feeding insect Plutella xylostella and its specialist endoparasitoid Diadegma semiclausum, when exposed to a daily heat pulse of 5 or 10°C temperature increase during their entire immature phase. Growth and developmental responses differed with the amplitude of the heat pulse and they were different between host and parasitoid, indicating different thermal sensitivity of the two trophic levels. With a +5°C heat pulse, the adult parasitoids were larger which may result in a higher fitness, whereas a +10°C heat pulse retarded parasitoid development. These results show that the parasitoid is more sensitive than its host to brief intervals of temperature change, and this results in either positive or negative effects on life history traits, depending on the amplitude of the heat pulse. These findings suggest that more extreme fluctuations may disrupt host-parasitoid synchrony, whereas moderate fluctuations may improve parasitoid fitness.

We evaluated the selectivity of three plant extracts with potential insecticidal effects for the parasitoid Trichogramma pretiosum Riley, which is commonly used in biological pest control. The plant extracts assayed were an acetone extract of Toona ciliata M. Roem., commercial neem oil, and a nanoencapsulated formulation of neem oil (NC40). The toxicity of the plant extracts to T. pretiosum was evaluated according to the recommendations of the International Organization for Biological Control- IOBC Working Group. We assessed the susceptibility of adults of the maternal and F1 generations and immature stages of T. pretiosum to the extracts. Females exposed to egg cards treated with commercial neem oil parasitized almost 70% fewer eggs than control eggs treated with water; and this extract was therefore classified as slightly harmful. When the eggs were offered to females 24h after treatment with neem oil and aqueous NC40, the parasitism rate also decreased, and the two extracts were classified as slightly harmful. Adult emergence was lower for parasitoids that fed on host eggs offered 24h after the treatment with the T. ciliata extract, which was considered slightly harmful. The emergence of T. pretiosum from eggs, larvae and pupae treated with the different plant extracts, did not decrease compared to development stages treated with the water control. The use of T. pretiosum, combined with the application of an ethanol extract of T. ciliata and a nanoencapsulated formulation of neem, appears to be feasible in view of these low toxicity indices., (Copyright © 2016 Elsevier Inc. Intra-guild predation (IGP) is an important phenomenon structuring ecological communities and affects the success of biological control. Here we show that parasitism by the koinobiont wasp Cotesia vestalis is associated with behavioural changes in its larval host (diamondback moth, Plutella xylostella) that reduce risk of IGP. Compared with unparasitised caterpillars, parasitised P. xylostella moved less frequently to new feeding patches on plants and were less likely to fall from the plant. Wolf spiders killed significantly fewer parasitised larvae. Reflecting their reduced movement and capacity to select plant tissue of optimal quality, parasitised caterpillars fed at a lower rate and exhibited delayed development suggesting a trade-off between IGP avoidance and nutrient intake by the host. This change in behaviour to reduce risk may cascade to the first trophic level and help explain the stability of IGP systems.

Metabolomics studies of low-biomass organisms, such as small insects, have previously relied on the pooling of biological samples to overcome detection limits, particularly using NMR. We show that the differentiation of metabolite profiles of individual 1 mg parasitoid wasps of different ages is possible when using a modified sample preparation and a combination of untargeted NMR and LC-MS based metabolomics. Changes were observed between newly emerged and older wasps in glycerolipids, amino acids and circulatory sugars. This advance in chemical profiling has important implications for the study of the behaviour and ecology of parasitoids and many other species of small organisms because predictions and observations are typically made at the level of the individual. Thus, the metabolomic state of low-biomass individuals can now be related to their behaviour and ecological performance. We discuss specifically the utility of age-related metabolomic profiling but our new approach can be applied to a wide range of biological research.
