Abstract: Monochromatic gamma-ray signals constitute a potential smoking gun signature for annihilating or decaying dark matter particles that could relatively easily be distinguished from astrophysical or instrumental backgrounds. We provide an updated assessment of the sensitivity of the Cherenkov Telescope Array (CTA) to such signals, based on observations of the Galactic centre region as well as of selected dwarf spheroidal galaxies. We find that current limits and detection prospects for dark matter masses above 300 GeV will be significantly improved, by up to an order of magnitude in the multi-TeV range. This demonstrates that CTA will set a new standard for gamma-ray astronomy also in this respect, as the world's largest and most sensitive high-energy gamma-ray observatory, in particular due to its exquisite energy resolution at TeV energies and the adopted observational strategy focussing on regions with large dark matter densities. Throughout our analysis, we use up-to-date instrument response functions, and we thoroughly model the effect of instrumental systematic uncertainties in our statistical treatment. Abstract: Galaxy clusters are expected to be dark matter (DM) reservoirs and storage rooms for the cosmic-ray protons (CRp) that accumulate along the cluster's formation history. Accordingly, they are excellent targets to search for signals of DM annihilation and decay at gamma-ray energies and are predicted to be sources of large-scale gamma-ray emission due to hadronic interactions in the intracluster medium. We estimate the sensitivity of the Cherenkov Telescope Array (CTA) to detect diffuse gamma-ray emission from the Perseus galaxy cluster. We perform a detailed spatial and spectral modelling of the expected signal for the DM and the CRp components. For each, we compute the expected CTA sensitivity. The observing strategy of Perseus is also discussed. In the absence of a diffuse signal (non-detection), CTA should constrain the CRp to thermal energy ratio within the radius $R_{500}$ down to about $X_{500}<3\times 10^{-3}$, for a spatial CRp distribution that follows the thermal gas and a CRp spectral index $\alpha_{\rm CRp}=2.3$. Under the optimistic assumption of a pure hadronic origin of the Perseus radio mini-halo and depending on the assumed magnetic field profile, CTA should measure $\alpha_{\rm CRp}$ down to about $\Delta\alpha_{\rm CRp}\simeq 0.1$ and the CRp spatial distribution with 10% precision. Regarding DM, CTA should improve the current ground-based gamma-ray DM limits from clusters observations on the velocity-averaged annihilation cross-section by a factor of up to $\sim 5$, depending on the modelling of DM halo substructure. In the case of decay of DM particles, CTA will explore a new region of the parameter space, reaching models with $\tau_{\chi}>10^{27}$s for DM masses above 1 TeV. These constraints will provide unprecedented sensitivity to the physics of both CRp acceleration and transport at cluster scale and to TeV DM particle models, especially in the decay scenario., Comment: 93 pages (including author list, appendix and references), 143 figures. Abstract: NectarCAM is a camera for the medium-sized telescopes of the Cherenkov Telescope Array (CTA), which covers the energy range of 100 GeV to 30 TeV. The camera is equipped with 265 focal plane modules (FPMs). Each FPM comprises 7 pixels, each consisting of a photo-multiplier tube, a preamplifier, an independently controlled power supply, and a common control system. We developed a dedicated test bench to validate and qualify the industrial FPM production and to measure the performance of each FPM in a dark room before its integration in the camera. We report the measured performance of 61 FPM prototypes obtained with our experimental setup. We demonstrate that the gains of the photo multiplier tubes are stable and that pulse widths, transit time spreads, afterpulse rates and charge resolutions are within the specifications for NectarCAM.

Abstract: The Polarized Instrument for Long-wavelength Observation of the Tenuous interstellar medium (PILOT) is a balloon-borne experiment aiming at measuring the polarized emission of thermal dust at a wavelength of 240 mm (1.2 THz). A first PILOT flight (flight#1) of the experiment took place from Timmins, Ontario, Canada, in September 2015 and a second flight (flight#2) took place from Alice Springs, Australia in april 2017. In this paper, we present the inflight performance of the instrument during these two flights. We concentrate on performances during flight#2, but allude to flight#1 performances if significantly different. We first present a short description of the instrument and the flights. We determine the time constants of our detectors combining inflight information from the signal decay following high energy particle impacts (glitches) and of our internal calibration source. We use these time constants to deconvolve the data timelines and analyse the optical quality of the instrument as measured on planets. We then analyse the structure and polarization of the instrumental background. We measure the detector response flat field and its time variations using the signal from the residual atmosphere and of our internal calibration source. Finally, we analyze the detector noise spectral and temporal properties. The in-flight performances are found to be satisfactory and globally in line with expectations from ground calibrations. We conclude by assessing the expected in-flight sensitivity of the instrument in light of the above in-flight performances.

Abstract: Objective: Unusual clinical course. Background: The finding of pneumoperitoneum frequently leads to operative management for diagnosis and treatment. The etiology of pneumoperitoneum includes perforated viscus, such as perforated peptic ulcers, small or large intestinal perforations, appendicitis, and complicated sigmoid diverticulitis. We describe the preoperative, intraoperative, and postoperative course of a patient with perforated Meckel's diverticulitis presenting with pneumoperitoneum. This unusual presenting finding highlights that Meckel's diverticulum should be included in the differential diagnosis in adolescents and young adults presenting with pneumoperitoneum. Case Report: We describe a case of an 18-year-old male who presented with 1 day of abdominal pain, found to have pneumoperitoneum during workup, attributed to perforated Meckel's diverticulum. CT scans of the abdomen and pelvis were performed, confirming pneumoperitoneum, an inflamed segment of distal ileum, and a non-visualized appendix, which made the diagnosis difficult. Perforated Meckel's diverticulum, likely due to infection, was confirmed by diagnostic laparoscopy. The Meckel's diverticulum was then exteriorized and removed by segmental small bowel resection with primary anastomosis. The final pathology report confirmed perforated Meckel's diverticulum with gastric oxyntic-type mucosa. Conclusions: This case illustrates an uncommon presentation of Meckel's diverticulum in an adolescent with pneumoperitoneum. Pneumoperitoneum requires broadening the diagnosis to include other causes, including Meckel's diverticulum, especially in the setting of an acute abdomen. This case highlights that a high index of suspicion should be kept for Meckel's diverticulum, even in adolescents and young adults with pneumoperitoneum. [ABSTRACT FROM AUTHOR]

Abstract: Future cosmology space missions will concentrate on measuring the polarization of the Cosmic Microwave Background, which potentially carries invaluable information about the earliest phases of the evolution of our universe. 13. An Educological Perspective on Higher Education's Responsibility to Vocational Education.

Abstract: A study was conducted to improve understanding of higher education's impact on the growth, sustenance, and improvement of vocational education programs in higher and secondary education. The research focused on how higher education can improve higher and secondary vocational education during the next 10 years. Research was conducted using the Delphi technique (a series of sequential questionnaires) to gather expert opinion. The sample consisted of 30 leading higher vocational educators and 30 leading secondary vocational educators selected by nomination by the current president and 5 former presidents of the American Vocational Association. The study respondents (75 percent of the sample) identified four techniques as the most useful for improving vocational education programs: (1) improve the image of vocational education to all publics; (2) provide more opportunities for upgrading faculty and staff skills; (3) collaborate more with secondary program leaders; and (4) implement research that documents examples from work settings that illustrate the needs of literacy skills, teamwork, negotiation, problem solving, and critical thinking. The study concluded that higher education must be responsible for taking the initiative to improve vocational education programs. (KC)

Abstract: The MITOMAP () data system for the human mitochondrial genome has been greatly enhanced by the addition of a navigable mutational mitochondrial DNA (mtDNA) phylogenetic tree of ∼3000 mtDNA coding region sequences plus expanded pathogenic mutation tables and a nuclear-mtDNA pseudogene (NUMT) data base. The phylogeny reconstructs the entire mutational history of the human mtDNA, thus defining the mtDNA haplogroups and differentiating ancient from recent mtDNA mutations. Pathogenic mutations are classified by both genotype and phenotype, and the NUMT sequences permits detection of spurious inclusion of pseudogene variants during mutation analysis. These additions position MITOMAP for the implementation of our automated mtDNA sequence analysis system, Mitomaster.

Abstract: Surface snow cover is a highly interacting component of global and regional climate systems. In alpine countries it represents a vital natural resource for ecology, electricity production, water management, public services, tourism etc. and can be associated with major natural hazards. At the same time, snow cover is a key climate change indicator and is subject to an important retreat as a consequence of global warming.The monitoring of the evolution of surface snow cover and the understanding of its spatio-temporal variability require the availability of consistent long-term snow data. This can partly be provided by homogenized station series, but for many applications gridded snow cover products provide an added value. Such a gridded climatology is currently missing for Switzerland. To fill this gap, a new methodology to produce a high-quality long-term gridded snow water equivalent (SWE) climatology for Switzerland has been developed. The approach consists of (1) driving a surface snow cover model by gridded temperature and precipitation data, and (2) statistically correcting the resulting SWE fields employing a variant of the quantile mapping method. The resulting SWE climatology covers the whole of Switzerland at a spatial resolution of 1 km and provides consistent daily SWE fields for the period 1961-2022.This contribution presents the methodology applied and a first overview of the spatio-temporal SWE variability in Switzerland since 1961. Abstract: The European Alps are a major mountain range in Europe with a long history of dense in-situ observations of snow cover, whose analysis is hampered by the fragmentation of observer networks and the varying quality and availability of their data. Here we present a collation of in-situ observations of snow depth and depth of snowfall across the whole Alps. We employed novel quality checks and gap-filling procedures based on spatial correlation, which work in areas with a moderate to high density of stations. We show how the climatology of in-situ snow depth and remotely sensed snow cover duration matches the expected climatic forcing zones. These large-scale climatic zones not only influenced day-to-day variability but also long-term trends. A multivariate attribution study highlights the impact of temperature and precipitation variability on snowfall patterns in a smaller subregion. Finally, future snow cover information is derived from an ensemble of regional climate models.

Abstract: Mutant calreticulin (CALR) proteins resulting from a −1/+2 frameshifting mutation of the CALR exon 9 carry a novel C-terminal amino acid sequence and drive the development of myeloproliferative neoplasms (MPNs). Mutant CALRs were shown to interact with and activate the thrombopoietin receptor (TpoR/MPL) in the same cell. We report that mutant CALR proteins are secreted and can be found in patient plasma at levels up to 160 ng/mL, with a mean of 25.64 ng/mL. Plasma mutant CALR is found in complex with soluble transferrin receptor 1 (sTFR1) that acts as a carrier protein and increases mutant CALR half-life. Recombinant mutant CALR proteins bound and activated the TpoR in cell lines and primary megakaryocytic progenitors from patients with mutated CALR in which they drive thrombopoietin-independent colony formation. Importantly, the CALR-sTFR1 complex remains functional for TpoR activation. By bioluminescence resonance energy transfer assay, we show that mutant CALR proteins produced in 1 cell can specifically interact in trans with the TpoR on a target cell. In comparison with cells that only carry TpoR, cells that carry both TpoR and mutant CALR are hypersensitive to exogenous mutant CALR proteins and respond to levels of mutant CALR proteins similar to those in patient plasma. This is consistent with CALR-mutated cells that expose TpoR carrying immature N-linked sugars at the cell surface. Thus, secreted mutant CALR proteins will act more specifically on the MPN clone. In conclusion, a chaperone, CALR, can turn into a rogue cytokine through somatic mutation of its encoding gene.

Any further distribution of this work must maintain attribution to the author(s) and the title of the work, journal citation and DOI., We provide an updated assessment of the power of the Cherenkov Telescope Array (CTA) to search for thermally produced dark matter at the TeV scale, via the associated gamma-ray signal from pair-annihilating dark matter particles in the region around the Galactic centre. We find that CTA will open a new window of discovery potential, significantly extending the range of robustly testable models given a standard cuspy profile of the dark matter density distribution. Importantly, even for a cored profile, the projected sensitivity of CTA will be sufficient to probe various well-motivated models of thermally produced dark matter at the TeV scale. This is due to CTA's unprecedented sensitivity, angular and energy resolutions, and the planned observational strategy. The survey of the inner Galaxy will cover a much larger region than corresponding previous observational campaigns with imaging atmospheric Cherenkov telescopes. CTA will map with unprecedented precision the large-scale diffuse emission in high-energy gamma rays, constituting a background for dark matter searches for which we adopt state-of-the-art models based on current data. Abstract: Eye drops and ointments are the most prescribed methods for ocular drug delivery. However, due to low drug bioavailability, rapid drug elimination, and low patient compliance there is a need for improved ophthalmic drug delivery systems. This study provides insights into the design of a new drug delivery device that consists of an ocular coil filled with ketorolac loaded PMMA microspheres. Nine different ocular coils were created, ranging in wire diameter and coiled outer diameter. Based on its microsphere holding capacity and flexibility, one type of ocular coil was selected and used for further experiments. No escape of microspheres was observed after bending the ocular coil at curvature which reflect the in vivo situation in human upon positioning in the lower conjunctival sac. Shape behavior and tissue contact were investigated by computed tomography imaging after inserting the ocular coil in the lower conjunctival fornix of a human cadaver. Thanks to its high flexibility, the ocular coil bends along the circumference of the eye. Because of its location deep in the fornix, it appears unlikely that in vivo, the ocular coil will interfere with eye movements. In vitro drug release experiments demonstrate the potential of the ocular coil as sustained drug delivery device for the eye. We developed PMMA microspheres with a 26.5 ± 0.3 wt% ketorolac encapsulation efficiency. After 28 days, 69.9% ± 5.6% of the loaded ketorolac was released from the ocular coil when tested in an in vitro lacrimal system. In the first three days high released dose (48.7% ± 5.4%) was observed, followed by a more gradually release of ketorolac. Hence, the ocular coil seems a promising carrier for ophthalmic drugs delivery in the early postoperative time period.

Abstract: Aim To assess the safety and tolerability of adjuvant subcutaneous trastuzumab (Herceptin® SC, H SC), delivered from an H SC Vial via hand-held syringe (Cohort A) or single-use injection device (Cohort B), with or without chemotherapy, for human epidermal growth factor receptor 2 (HER2)-positive stage I to IIIC early breast cancer (EBC) in the phase III SafeHer study (NCT01566721). Methods Patients received 600 mg fixed-dose H SC every 3 weeks for 18 cycles. The chemotherapy partner was at the investigators' discretion (H SC monotherapy was limited to ≤10% of the population). Data from the first H SC dose until 28 days (plus a 5-day window) after the last dose are presented. Results are descriptive. Results In the overall population, 2282/2573 patients (88.7%) experienced adverse events (AEs). Of the above, 128 (5.0%) patients experienced AEs leading to study drug discontinuation; 596 (23.2%) experienced grade ≥ 3 AEs and 326 (12.7%) experienced serious AEs. Grade ≥ 3 cardiac disorders were reported in 24 patients (0.9%), including congestive heart failure in eight (0.3%). As expected, the AE rates varied according to the timing of chemotherapy in both cohorts, with higher rates in concurrent versus sequential chemotherapy subgroups. In the concurrent chemotherapy subgroup, AEs were more common during the actual period of concurrent chemotherapy compared with the period when patients did not receive concurrent chemotherapy. Conclusion SafeHer confirms the safety and tolerability of the H SC 600 mg fixed dose for 1 year (every 3 weeks for 18 cycles) as adjuvant therapy with concurrent or sequential chemotherapy for HER2-positive EBC. These primary analysis results are consistent with the known safety profile for intravenous H and H SC.

Abstract: A liquid, biocompatible polyester based polymer, which could facilitate injectable formulations by simple mixing with the active substance (drug) is much needed by the pharmaceutical companies. A favourite candidate is polylactic acid (PLA) which is biocompatible and biodegradable. However PLA is solid with high crystallinity. Thus, in this research, hydroxylauric acid (HOLA) was copolymerized with lactic acid (LA) in different ratios by polycondensation technique at 180 °C, without a metal catalyst and avoiding the formation of interfering lactides, to provide a liquid polyester. The copolymers molecular weights were determined by Gel Permeation Chromatography (GPC) and their physical states indicated as solid or liquid were noted. The structures as polyesters were confirmed by FT-IR and 1H NMR spectroscopy. Poly(HOLA:LA) products from reactant ratios 0:100 is solid, while ratios of 20:80, 40:60 are mixed (paste) and 60:40, 80:20 and 100:0 are liquids. Thus, the liquid polyesters from the polycondensation of HOLA and LA without catalyst were picked as potential candidates for dissolving hydrophobic drugs that could be used as injectables in controlled drug delivery experiments.

Abstract: “Lagikway” is an underutilized vegetable in the Philippines. One of the reasons for its low utilization is the lack of information in terms of production and nutritional quality under Philippine conditions. Hence, yield trial and nutritional quality evaluation were conducted from June 2017 to February 2018 involving three lagikway entries – including RXII 13-003, RXII 13- 019, and Lag 152649. The effect of blanching on the phytochemicals of leaves for consumption was also determined. Lagikway rooted cuttings were transplanted in 5 m x 1 m plots with 50 cm x 50 cm distance, and the experiment was carried out in a randomized complete block design (RCBD) with four replications. Young leaves and shoots considered for consumption were harvested 1 mo after transplanting (MAT) and continued at 2-wk intervals. Data revealed that potential yield is at 24.5 tons/ha for RXII 13-003, 21.36 tons/ha for RXII 13-019, and 21.94 tons/ha for Lag 152649. For proximate analysis, the moisture content is highest in RXII 13-019, the fat content is highest in RXII 13-003, and the protein content and the nitrogen-free extract are highest in Lag 152649. Ash and fiber content were the same from all the three lagikway accessions. Significant differences were also observed on the different lagikway accessions in terms of phytochemicals and antioxidant activity, and blanching had greatly affected these parameters. Lag 152649 had the highest phenolics, flavonoids, tannins, and radical scavenging activity (RSA). Hence, Lag 152649 will be recommended for mass propagation and distribution, which will eventually lead to further promotion and utilization of this underutilized vegetable. [ABSTRACT FROM AUTHOR]

Abstract: Eye drops and ointments are the most prescribed methods for ocular drug delivery. However, due to low drug bioavailability, rapid drug elimination, and low patient compliance there is a need for improved ophthalmic drug delivery systems. This study provides insights into the design of a new drug delivery device that consists of an ocular coil filled with ketorolac loaded PMMA microspheres.Nine different ocular coils were created, ranging in wire diameter and coiled outer diameter. Based on its microsphere holding capacity and flexibility, one type of ocular coil was selected and used for further experiments. No escape of microspheres was observed after bending the ocular coil at curvature which reflect the in vivo situation in human upon positioning in the lower conjunctival sac.Shape behavior and tissue contact were investigated by computed tomography imaging after inserting the ocular coil in the lower conjunctival fornix of a human cadaver. Thanks to its high flexibility, the ocular coil bends along the circumference of the eye. Because of its location deep in the fornix, it appears unlikely that in vivo, the ocular coil will interfere with eye movements.In vitro drug release experiments demonstrate the potential of the ocular coil as sustained drug delivery device for the eye. We developed PMMA microspheres with a 26.5 +/- 0.3 wt% ketorolac encapsulation efficiency. After 28 days, 69.9% +/- 5.6% of the loaded ketorolac was released from the ocular coil when tested in an in vitro lacrimal system. In the first three days high released dose (48.7% +/- 5.4%) was observed, followed by a more gradually release of ketorolac. Hence, the ocular coil seems a promising carrier for ophthalmic drugs delivery in the early postoperative time period.
