BACKGROUND AND OBJECTIVES: Environmental deoxyribonucleic acid metabarcoding has become a powerful tool for assessing invertebrate biodiversity in stream sediments, providing a non-invasive approach for ecological monitoring. However, the accuracy of this method heavily depends on the nucleic acid extraction protocol used, which can influence taxonomic recovery and the resulting biotic indices. Previous studies have focused on nucleic acid yield and taxonomic composition, but limited research has evaluated the impact of extraction methods on ecological assessments. This study aims to compare four nucleic acid extraction protocols to determine their effectiveness in maximizing metazoan recovery and improving the reliability of biotic indices derived from metabarcoding data.METHODS: Four nucleic acid extraction protocols were tested for invertebrate nucleic acid metabarcoding from stream sediment: a manual extraction and three Qiagen commercial kits (Powersoil, Blood and Tissue, and Plant Mini kit). Extracted nucleic acid was amplified using cytochrome c oxidase I and 18 Svedberg ribosomal ribonucleic acid hypervariable 4 and 9 markers and sequenced using Illumina sequencing. The relative read abundance was analyzed to assess metazoan recovery and biotic index scores were calculated based metazoan community of respective extraction protocol.FINDINGS: Among the tested methods, PowerSoil kit consistently yielded the lowest metazoan relative read abundance (36.07 to 86.31 percent) from stream sediment and exhibited higher non-target amplification, including bacterial nucleic acid, due to its bead-beating step optimized for prokaryotic nucleic acid extraction. In contrast, other protocols showed superior metazoan recovery (63.83 to 98.58 percent) and minimized bacterial co-amplification, indicating high specificity for eukaryotic nucleic acid. Plant Mini kit performed unexpectedly well, aligning closely with the manual extraction in biotic indices, likely due to its effective inhibitor removal of amplification. Conversely, blood and tissue kit had the highest dissimilarity in biotic indices, potentially attributed to the inhibition, resulting in lower detection of sensitive taxa. Biotic indices derived from cytochrome c oxidase I and 18 Svedberg ribosomal ribonucleic acid hypervariable 9 markers were comparable, with cytochrome c oxidase I showing slightly higher scores due to its greater specificity in targeting key taxa used for index calculation. The 18 Svedberg ribosomal ribonucleic acid hypervariable 4 marker, however, yielded lower biotic index scores and was less effective in metazoan recovery, particularly for invertebrates sensitive to ecological stressors.CONCLUSION: Overall, the choice of extraction method substantially influenced the detection of target metazoan taxa and the reliability of biotic indices, with the manual extraction and Plant Mini kit emerging as the most effective protocols. These findings underscore the importance of protocol selection in metabarcoding of stream sediments, suggesting that optimized methods tailored for metazoan nucleic acid recovery are crucial for accurate ecological assessments.

ABSTRACT The objective was to test the response of Urochloa brizantha cv. Marandu in monoculture and in silvopastoral system (SPS), at two distances from the trees, and define the best defoliation height for SPS. Four intermittent defoliation heights (25, 35, 45 and 55cm) and two distances from tree lines (2.5 and 5.0m) were evaluated in the SPS with a control defoliated with 25cm in full sun. The experiment was performed in a randomized block design with 3 replicates in a 4 × 2 + 1 split plot scheme. The control had higher forage accumulation (46.9kg/ha. day) than the SPS (31.1kg/ha. day). The bulk density was also higher in the control (0.89mg/cm³) than in SPS (0.48mg/cm³). The percentage of leaves (78.06%) and leaf/stem ratio (6.04) did not differ among the treatments. In the SPS, there was an increase of 31.07% in forage accumulation from 25 to 55cm. The forage accumulation and bulk density of Urochloa brizantha cv. Marandu in monoculture is greater than in the SPS regardless of the management goal and the distance from trees. The goal of 55cm in the SPS presented greater forage accumulation.

Starphenes are polycyclic aromatic hydrocarbons and build by variatiTheoremons of three different acene-arms. Starphenes are the basic building blocks for miniaturization of different especially organic electronic devices. It also played an important role in different logical gates. In network topology, each electric circuit, structure or network can be derived in graphs where principal nodes (or simply nodes) alternated to vertices and line segment (branches) become edges. The resolvability parameters of a graph are a relatively new specialized subject in which the whole network is formed to obtain each principal node’s unique location. The metric, edge metric dimension, and generalizations are investigated in this article as resolvability parameters of starphene structure. We proved that all the parameters studied for starphene graph have constant cardinalities. Understanding and dealing with structures is made easier by transforming the whole structure into a novel shape delivered by resolvability parameters.

ABSTRACT The aim of this study was to evaluate Leishmania vectors and investigate the environmental and climatic characteristics related to their transmission. This was a prospective eco-epidemiological study involving the systematic collection of sandflies and a canine serological survey in areas of municipality in Brazil. Blood samples from 1,752 dogs were examined to test for canine visceral leishmaniasis (VL). An entomological evaluation was carried out using 24 light traps in the peridomicile and intradomicile areas of 12 households for 12 months. Climate variables were analyzed using a simple Spearman correlation and a spatial analysis via the fuzzy logic method and K function. The prevalence rate of canine VL was 4.1% and 7.1% in the two areas that were studied, respectively. The entomological study resulted in the capture of 431 sandflies. Most (74%) of the specimens were captured in the peridomiciliary area, and 5.6% of the samples that were analyzed using molecular biology were positive for Leishmania spp. In particular, Leishmania infantum was found in 100% of the positive samples. The spatial analysis showed that one particular area presented a higher concentration of high-density overlapping points of Lutzomyia longipalpis and seropositive dog populations, indicating a higher risk of the concomitant occurrence of both events. The results show that a parasite-reservoir-vector interface is active in the studied areas.

O objetivo deste trabalho foi estudar os efeitos da utilização de farelo de crambe em substituição ao farelo de soja sobre o comportamento ingestivo de bovinos leiteiros. Foram utilizados quatro machos castrados Holandês x Zebu, fistulados no rúmen, com peso vivo médio de 664kg, distribuídos em delineamento quadrado latino 4 x 4. Os tratamentos consistiram em quatro dietas isoproteicas e isoenergéticas, formuladas com relação volumoso:concentrado 60:40 com base na matéria seca (MS). O volumoso foi composto de silagem de milho (51% MS) e feno de Tifton (49% MS), e o concentrado formulado com níveis crescentes de substituição do farelo de soja pelo farelo de crambe em 0%, 2,8%, 6,4% e 11,0% na MS da dieta. O comportamento ingestivo foi avaliado por meio do método direto de avaliação visual, em intervalos de 10 minutos, durante períodos de 24 horas. Registraram-se a frequência de alimentação, a ruminação e o ócio, bem como a posição do animal (em pé ou em decúbito). As variáveis em pé e em decúbito não diferiram entre os tratamentos, assim como os tempos gastos em alimentação, ruminação e ócio. O consumo de matéria seca e de FDN expressos em g/dia e gFDN/dia, respectivamente, a eficiência de ruminação expressa em gMS/min, a eficiência de ruminação expressa em gFDN/dia e o tempo de mastigação total não diferiram significativamente. No entanto, a eficiência de alimentação (gMS/min) variou de forma linear decrescente com a inclusão do farelo de crambe. Os períodos do dia influenciaram todas as atividades. O maior tempo de alimentação foi observado nos períodos após o fornecimento da dieta, e a maior atividade de ruminação foi verificada no período noturno. A substituição de farelo de soja por farelo de crambe não afetou o comportamento ingestivo, exceto para o parâmetro eficiência de alimentação. Nesse sentido, considerando o comportamento ingestivo, recomenda-se a substituição do farelo de soja por farelo de crambe para alimentação de bovinos leiteiros.

O experimento foi realizado na Embrapa Gado de Leite, em delineamento quadrado latino (QL) 4x4, com o objetivo de avaliar o perfil de ácidos graxos (AG) e os índices de qualidade nutricional da gordura do leite de vacas Holandês x Gir (n = 16) sob pastejo em Brachiaria brizantha cv. Avaliou-se o efeito da adição de teores crescentes de ureia (0,0; 0,5 e 1,0%) na matéria natural da cana-de-açúcar sobre a produção e composição do leite, eficiência alimentar e viabilidade econômica. Foram utilizadas 18 vacas em lactação, com produção média inicial de 21,3±0,8kg/dia de leite e com 83±7 dias em lactação, distribuídas em delineamento ensaio de reversão do tipo switch-back, 3x3. As dietas experimentais foram constituídas de cana-de-açúcar como volumoso único e concentrado, na relação de 50:50 (base MS). Não houve efeito da suplementação dos diferentes teores de ureia sobre a produção e composição do leite. Contudo, a eficiência alimentar (produção de leite/consumo de matéria seca) foi melhor nas dietas em que se utilizou ureia, sendo 1,14; 1,17 e 1,17, respectivamente, para as dietas com 0,0; 0,5 e 1,0% de ureia (P

Avaliou-se o desempenho produtivo e econômico de três grupos genéticos de bovinos em sistema com recria a pasto e terminação em confinamento, com ciclo de produção de um ano. Foram utilizados 36 bovinos não castrados, com média de 270±50 dias de idade e peso médio inicial de 241,3±34kg, sendo 12 Guzolandos (1/2 Guzerá x 1/2 Holandês), 12 Guzonel (1/2 Guzerá x 1/2 Nelore) e 12 Tricross (1/2 Simental x 1/2 Guzonel). O delineamento estatístico foi inteiramente ao acaso. Os animais permaneceram em pastejo rotacionado de Brachiaria brizantha cv. Marandu, com suplementação comercial; na terminação, receberam dieta composta por silagem de sorgo e concentrado (50:50) durante 61 dias. Os índices de desempenho avaliados foram o ganho médio de peso diário (GMD), ganho médio de peso no período, peso corporal final e, como indicadores econômicos, a receita total, custo operacional total por arroba (COT/@), margem bruta e margem líquida. O GMD na recria foi semelhante (P>0,05) entre os grupos que apresentaram média de 746 e 456 gramas, respectivamente, para os períodos águas/transição águas-seca e seca. O COT/@ da recria do grupo Tricross (R$69,44) foi superior aos dos grupos Guzonel (R$66,75) e Guzolando (R$ 64,86). No período de terminação, o GMD foi semelhante entre os grupamentos, apresentanto o valor médio de 1,55kg. O COT/@ da terminação em confinamento foi R$ 91,48, R$114,15 e R$122,78, para Tricross, Guzonel e Guzolando, respectivamente. Considerando-se o sistema de ciclo curto completo, obteve-se COT/@ menor para o grupamento Tricross (R$77,92), seguido pelo grupamento Guzonel (R$ 74,89) e Guzolando (R$ 78,43). Nas condições de produção avaliadas, o grupo Tricross foi o mais eficiente economicamente.

The role of the pharmacist in healthcare has evolved greatly over the last half-century, from dispensing to providing direct patient-oriented activities not associated with dispensing. However, pharmacist-led healthcare services in Sri Lanka must undergo reform to fully take advantage of their expertise and training in medicine management and related outcomes in Sri Lankan patients. As befits a profession’s role development and value, professional and educational standards for pharmacists need ongoing development and growth. Currently, university curricula and continuing professional education in Sri Lanka require further development and optimisation to provide the theoretical and practical knowledge and skills regarding quality use of medicines and patient-oriented care. Furthermore, pharmacists’ roles in Sri Lankan hospital and community pharmacist settings need to be recognised and should include the pharmacist as an integral part of the multidisciplinary healthcare team in Sri Lanka. Studies from developed countries and some developing countries have demonstrated that expanded pharmacists’ roles have had a significant positive cost-effective impact on the population’s health. Therefore, the availability of qualified Sri Lankan pharmacists trained to deliver expanded professional services accompanied by greater pharmacist integration into healthcare delivery is crucially important to ensure quality use of medicines within the Sri Lankan healthcare system and optimise the medication-related needs of Sri Lankans.

Avaliaram-se o consumo, metabólitos sanguíneos e a produção e composição do leite de 16 vacas Holandês x Gir, manejadas em pastagem de Brachiaria brizantha cv. Marandu, suplementadas com 6kg/vaca/dia (base matéria natural) de concentrado contendo 0; 1,3; 2,6 e 3,9kg/vaca/dia de grão de soja tostado (GST). Foi utilizado o delineamento com quatro quadrados latinos (QL) 4 x 4, sendo cada fase do QL constituída de 10 dias de período de adaptação à dieta e de cinco para coleta de amostras. A suplementação da dieta com GST resultou em redução linear (P0,05), porém a de ácidos graxos não esterificados aumentou com a inclusão do GST na dieta (P0,05) da adição do GST sobre a produção e composição do leite, exceto para o teor (P=0,10) e produção (P=0,08) de gordura no leite.

Os objetivos desta pesquisa foram caracterizar as recusas por parte da população em permitir a borrifação de seus imóveis, em quatro regiões de Belo Horizonte, no período de 2006 a 2008, e relacionar estas recusas com o surgimento de casos de leishmaniose visceral, no mesmo local e período. Utilizaram-se as localizações onde se encontraram 27 casos humanos de leishmaniose visceral obtidos do SINAN, todos referentes a quatro áreas de abrangência da região noroeste de Belo Horizonte, no período de 2006 a 2008. De um total de 33.579 residências visitadas pelo controle químico vetorial, 9636 (28,70%) aceitaram a borrifação interna e a externa do domicílio, 20741 (61,77%) concordaram com a borrifação apenas do peridomicílio, 141 (0,42%) aceitaram apenas a borrifação do intradomicílio, 909 (2,70%) se encontraram fechadas no momento da borrifação e 2152 (6,41%) recusaram qualquer tipo de borrifação no imóvel. Na maioria das residências, o controle químico foi permitido apenas no peridomicílio. A diferença entre o perfil de recusas dos quarteirões contidos em uma área de 200 metros ao redor de um caso humano de leishmaniose visceral e o mesmo perfil dos quarteirões fora desta área foi significativa (P

Assuming that the IS6110-restriction fragment length polymorphism (RFLP) changes at a constant rate of 3.2 years, this methodology was applied to demonstrate, for the first time, variant patterns of Mycobacterium tuberculosis (MTB) in multiple isolates obtained at short time intervals from sputum and blood of an HIV+ patient with multiple admissions to the Emergency Room and to the multidrug-resistant tuberculosis (MDR-TB) Reference Center of a secondary-care hospital in Rio de Janeiro, Brazil. In sputum, the IS6110-RFLP appeared in isolates with two variant patterns with 10 and 13 IS6110 copies. However, blood presented only the pattern corresponding to 10 copies, suggesting compartmentalization. With regard to the exact match of 10 of 13 bands, this may be a subpopulation with the same clonal origin and this may be related to the IS6110 transposition. A susceptibility test demonstrated an MDR profile (INH R, RIF R, SM R, and EMB R), with the sputum isolate also exhibiting EMB S (R = resistant; S = sensitive). A gene mutation confirmed resistance only to streptomycin. There was agreement between the results of the phenotypic test and the clinical response to MDR-TB treatment, suggesting serious implications with regard to treatment administration based exclusively on molecular methods, and calling attention to the fact that more effective control strategies against the emergence of MDR strains must be implemented by the TB control program to prevent transmission of MDR-MTB strains at health facilities in areas highly endemic for TB.

O trabalho teve como objetivo estudar a transmissão horizontal de Mycobacterium avium-intracellulare (MAI) entre suínos. Doze suínos com 35 a 42 dias de idade, divididos em três grupos de quatro e alojados em salas independentes, foram inoculados por via oral com as cepas VPS1, SC1 e ATCC-13950 de Mycobacterium avium-intracellulare. A seguir, quatro suínos não inoculados foram colocados em cada grupo e mantidos na mesma baia até o abate aos 150 dias da idade. Outro grupo de quatro suínos não inoculados foi utilizado como controle. Pelo exame de fezes verificou-se que houve excreção ativa de MAI entre 14 e 42 dias pós-inoculação. A transmissão da infecção foi avaliada pelo teste de tuberculina aos 140 dias de idade e pelos exames de patologia macro e microscópica e de isolamento do MAI de órgãos. Os quatro suínos em contato com aqueles inoculados com a cepa SC1 foram contaminados, enquanto que com a cepa VPS1 apenas um e com a ATCC-13950 somente dois. Concluiu-se que houve transmissão de MAI entre os suínos mantidos na mesma baia, com as três cepas utilizadas, mas foi mais evidente com a cepa SC1, indicando que o suíno pode ser importante fonte de infecção na disseminação da doença.
