Introduction: Endometriosis is a prevalent and recurrent medical condition associated with symptoms such as pelvic discomfort, dysmenorrhea, and reproductive challenges. Furthermore, it has the potential to progress into a malignant state, significantly impacting the quality of life for affected individuals. Despite its significance, there is currently a lack of precise and non-invasive diagnostic techniques for this condition.Methods: In this study, we leveraged microarray datasets and employed a multifaceted approach. We conducted differential gene analysis, implemented weighted gene co-expression network analysis (WGCNA), and utilized machine learning algorithms, including random forest, support vector machine, and LASSO analysis, to comprehensively explore senescence-related genes (SRGs) associated with endometriosis.Discussion: Our comprehensive analysis, which also encompassed profiling of immune cell infiltration and single-cell analysis, highlights the therapeutic potential of this gene assemblage as promising targets for alleviating endometriosis. Furthermore, the integration of these biomarkers into diagnostic protocols promises to enhance diagnostic precision, offering a more effective diagnostic journey for future endometriosis patients in clinical settings.Results: Our meticulous investigation led to the identification of a cluster of genes, namely BAK1, LMNA, and FLT1, which emerged as potential discerning biomarkers for endometriosis. These biomarkers were subsequently utilized to construct an artificial neural network classifier model and were graphically represented in the form of a Nomogram.

Abstract The aim of this study was to investigate the hepatoprotective effect of Matrine salvianolic acid B salt on carbon tetrachloride (CCl4)-induced hepatic fibrosis in rats. Salvianolic acid B and Matrine has long been used to treat liver fibrosis. Matrine salvianolic acid B salt is a new compound containing Salvianolic acid B and Matrine. Hepatic fibrosis induced by CCl4 was studied in animal models using Wistar rats. Organ coefficient, serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), hexadecenoic acid (HA), laminin (LN), hydroxyproline (Hyp), and glutathione (GSH), malondialdehyde (MDA), superoxide dismutase (SOD) in liver tissues were measured, respectively. Histopathological changes in the livers were studied by hematoxylin-eosin (H&E) staining and Masson Trichrome (MT) examination. The expression of transforming growth factor-β1 (TGF-β1) and α-smooth muscle actin (α-SMA) was observed by immunohistochemical analysis. A significant reduction in serum levels of AST, ALT, HA, LN and Hyp was observed in the Matrine salvianolic acid B salt treated groups, suggesting that the salt had hepatoprotective effects. The depletion of GSH and SOD, as well as MDA accumulation in liver tissues was suppressed by Matrine salvianolic acid B salt too. The expression of TGF-β1 and α-SMA measured by immunohistology was significantly reduced by Matrine salvianolic acid B salt in a dose-dependent manner. Matrine salvianolic acid B salt treatment attenuated the necro-inflammation and fibrogenesis induced by CCl4 injection, and thus it is promising as a therapeutic anti-fibrotic agent against hepatic fibrosis.

A defining unique characteristic of the gut immune system is its ability to respond effectively to foreign pathogens while mitigating unnecessary inflammation. Intestinal macrophages serve as the cornerstone of this balancing act, acting uniquely as both the sword and shield in the gut microenvironment. The GI tract is densely innervated by the enteric nervous system (ENS), the intrinsic nervous system of the gut. Recent advances in sequencing technology have increasingly suggested neuroimmune crosstalk as a critical component for homeostasis both within the gut and in other tissues. Here, we systematically review the ENS–macrophage axis. We focus on the pertinent molecules produced by the ENS, spotlight the mechanistic contributions of intestinal macrophages to gut homeostasis and inflammation, and discuss both existing and potential strategies that intestinal macrophages use to integrate signals from the ENS. This review aims to elucidate the complex molecular basis governing ENS–macrophage signaling, highlighting their cooperative roles in sustaining intestinal health and immune equilibrium. This paper first combines the traditional Adaboost iterative algorithm and logistic regression algorithm to construct an improved Adaboost based regression algorithm. In order to solve the problem of the redundant amount or insufficient amount of output of industrial carbon emissions, the SBM model is divided into two stages, and by merging this method, the industrial carbon emission efficiency measuring model is created, While the Global Moran’s I index is used to assess the geographical impact of industrial carbon emission efficiency. Additionally, a model of the influence of emission efficiency based on the geographical effect is built through the selection of the explanatory variables of the influencing factors. According to the study, the industrial carbon emission efficiency is growing at an annual rate of 1.8% during the period of fast expansion, 0.4% in the steady growth stage, and the Z value of STI is 0.38 is significant in spatial autocorrelation.

Na+/H+ exchanger isoform 1 (NHE1), which is a regulator of intracellular and extracellular pH via ion exchange, has been demonstrated to serve an important role in cell differentiation, migration and invasion in solid tumors and hematological malignancies. However, the potential role of NHE1 in epithelial ovarian cancer (EOC) remains unclear. In the present study, the expression pattern and the prognostic value of NHE1 were investigated in EOC. EOC tissues, non-cancerous tumors and normal ovarian tissues were collected, and the expression levels of NHE1 were determined using the reverse transcription-quantitative polymerase chain reaction, western blotting and immunohistochemistry. The expression pattern of NHE1 was also evaluated in ovarian cancer cell lines using western blotting and immunofluorescence. In addition, the association between the NHE1 expression pattern and the clinicopathological features and the clinical prognosis of patients with EOC was also analyzed. The expression levels of NHE1 were identified to be significantly increased in EOC tissues compared with non-cancerous tumors and normal ovarian tissues (P<0.05). Furthermore, the increased expression of NHE1 was associated with an advanced International Federation of Gynecology and Obstetrics stage (FIGO III–IV; P<0.001) and the presence of high-grade carcinoma (grades 2–3, P<0.001). Overexpressed NHE1 was identified as a risk factor of shorter PFS (P<0.001) and OS (P<0.001). A multivariate Cox's regression analysis revealed that NHE1 was an independent prognostic factor for the prediction of the outcome of patients with EOC. Objectives Predicting heave motion is helpful for improving the performance of the heave compensator and reducing the disturbance of waves on operating equipment. To improve the accuracy and stability of the heave prediction model, a real-time prediction method for ship heave motion is proposed in this paper.Methods Based on the Nonlinear Autoregressive with eXogeneous inputs (NARX) neural network, a single sea-state prediction model is established. The simulated heave motion of the vessel is obtained using the Marine Systems Simulator software tool to verify the model. The prediction model based on NARX is compared with prediction models based on Kalman and BP. On this basis, a multi sea-state prediction model is established by improving the single sea-state model.Results The prediction accuracy requirements of the multi sea-state prediction model are satisfied, and its stability is better than the single sea-state model, with a maximum prediction error of less than 10-4 magnitude in the range of sea state from 2 to 5.Conclusions The simulation results verify the good adaptability of the NARX neural network to the complex wave environment. Its prediction speed and accuracy are higher than the common back-propagation neural network and the traditional filtering method. It still maintains high prediction accuracy under high sea state.

Epigenetic inactivation of GADD45A is a common occurrence in different types of cancer. However, little is known regarding its association with radiosensitivity in cervical cancer (CC). Thus, the present study aimed to investigate the association between aberrant GADD45A methylation and radiosensitivity in CC. SiHa, HeLa and CaSki CC cells were treated with 5-azacytidine (5-azaC), with or without irradiation. The expression levels of GADD45A and AKT related molecules were detected via reverse transcription-quantitative PCR and western blot analyses. The methylation status of GADD45A was assessed via methylation-specific PCR and cell proliferation assays, while clonogenic assays and flow cytometric analysis were performed to assess the function of the genes (GADD45A and AKT) in the CC cell lines. The results demonstrated that methylation of GADD45A was significantly higher in the radioresistant tissues (63.16%) compared with the radiosensitive samples (33.33%). In addition, the surviving fraction of SiHa cells following irradiation with 2 Gy was demonstrated to be highest amongst the three CC cells (CaSki, 57±9.5%; HeLa, 70±4% and SiHa, 75±10%). The survival rate of SiHa cells following treatment with 5-azaC and ionizing radiation (IR) significantly decreased as the radiation dose increased, compared with treatment with IR alone. Following overexpression of GADD45A or treatment with 5-azaC, the radiosensitivity of SiHa cells significantly increased compared with both the control vector and PBS treated groups. In addition, the AKT inhibitor, MK-2206, increased the radiosensitivity of SiHa cells. Notably, aberrant methylation of GADD45A was associated with decreased radiosensitivity in CC, and the PI3K/AKT signaling pathway was essential for radioresistance, which was mediated through downregulation of GADD45A. First, it is indicated that the intrinsic mode functions (IMF) obtained by the empirical mode decomposition (EMD) are not orthogonal. Then an orthogonal technique based on Gram-Schmidt method is proposed to obtain the complete orthogonal intrinsic mode functions. Three ways of the orthogonal processes are suggested and compared in the paper. Finally, the suggested method is validated through the decomposition of a typical time history. The numerical results show that the orthogonal IMF can be obtained easily by the technique proposed in the paper.

To elucidate the expression profiles, methylation states, and clinicopathological significance of the PRDM gene family, focusing on the MECOM gene's role in uterine endometrial cancer (UCEC) and its molecular interactions with the TGF-beta signaling pathway. Our methodology combined detailed bioinformatics analyses using UALCAN and GEPIA with in vitro assessments in HEC-1-A cells. Techniques included CRISPR-Cas9 for gene editing and various cellular assays (CCK-8, flow cytometry, Transwell) to evaluate the effects of MECOM on cell proliferation, migration, and apoptosis, alongside Western blot analysis for protein regulation in the TGF-beta pathway. MECOM was upregulated in UCEC tissues, influencing tumor cell behavior significantly. Knockout studies demonstrated reduced proliferation and migration and increased apoptosis, while overexpression showed reverse effects. Mechanistically, MECOM modulated critical proteins within the TGF-beta pathway, impacting cell cycle dynamics and apoptotic processes. The PRDM gene family, particularly MECOM, plays a crucial role in the pathogenesis and progression of UCEC, suggesting its utility as a target for novel therapeutic interventions. Ovarian carcinoma is one of the major causes of gynecological cancer. This study aimed to evaluate the association of CYP1 family polymorphism with the risk of ovarian carcinoma and chemotherapy resistance. Positive selection was detected among human CYP1A1, CYP1A2, and CYP1B1, and other species. Several positive sites were detected by site models and brach-site models. Meta-analysis was conducted for the sites rs1056836 (MAF 0.39) and rs1056827 (MAF 0.36) of CYP1B1 to clarify the association between gene polymorphisms and ovarian carcinoma risk. Subgroup analysis showed the association of rs1056836 polymorphism with ovarian cancer risk among Caucasians and Asians, while all the six genetic models showed no association among African-Americans. All the six genetic models showed no association of rs1056827 polymorphism with ovarian cancer risk. The polymorphisms of rs1056836 associated with ovarian cancer risk were detected in chemotherapy-sensitive and drug-resistant ovarian cancer patients. DNA was extracted from 62 chemotherapy resistance Ovarian carcinoma tissue samples and 137 chemotherapy-sensitive ovarian carcinoma tissue samples as controls. Gene polymorphisms were genotyped using the Sequenom MassARRAY SNP approach. There was no significant association between the CYP1B1 rs1056836 polymorphism and chemotherapy resistance of ovarian cancer in all genetic models. The results suggest that rs1056836 polymorphism of gene CYP1B1 under obvious selection pressure had a significantly increased risk for ovarian carcinoma.
