Statistical bias correction (BC) is a widely used tool to post-process climate model biases in heat-stress impact studies, which are often based on the indices calculated from multiple dependent variables. This study compares four BC methods (three univariate and one multivariate) with two correction strategies (direct and indirect) for adjusting two heat-stress indices with different dependencies on temperature and relative humidity using multiple regional climate model simulations over South Korea. It would be helpful for reducing the ambiguity involved in the practical application of BC for climate modeling and end-user communities. Our results demonstrate that the multivariate approach can improve the corrected inter-variable dependence, which benefits the indirect correction of heat-stress indices depending on the adjustment of individual components, especially those indices relying equally on multiple drivers. On the other hand, the direct correction of multivariate indices using the quantile delta mapping univariate approach can also produce a comparable performance in the corrected heat-stress indices. However, our results also indicate that attention should be paid to the non-stationarity of bias brought by climate sensitivity in the modeled data, which may affect the bias-corrected results unsystematically. Careful interpretation of the correction process is required for an accurate heat-stress impact assessment.

How the added value of dynamically downscaled climate variables can be transferable to the hydrological impact assessment has been a long standing issue. This study investigates the potential benefit of highresolution climate data locally tailored over South Korea in terms of the reduction of uncertainties in hydrological projections. For this purpose, a large ensemble consisting of three Global Climate Model (GCM) projections and their downscaling products with different resolutions (i.e., 20 and 5 km), and four bias correction (BC) methods is fed into a semi-distributed hydrological model (HM) customized over Korean river basins. The in-depth comparison among the 45-members hydrological simulations proves that the added value of RCM would not be erased by the application of BC. While this study acknowledges the necessity of BC to remove the systematic bias in climate simulations, it is found that the high-resolution dynamical downscaling can significantly narrow the spread brought with different BC methods, thus reducing the uncertainty in the projected hydrological change. The projected streamflow changes for both the mean of wet season and the high flows indicate that there will be an intensified runoff, especially for the extremes, over South Korea under the warming. Altogether, this study provides a valuable exploration of uncertainty reduction in hydrological projections from the perspective of resolution effect of dynamical downscaling, which is meaningful for hydroclimate studies and climate change impact assessment.

Chemotherapic cancer treatment has suffered from side effects and poor target specificity. pH-triggered drug delivery systems offer the opportunity to better deliver anticancer drugs to cancer cells in a more controllable manner. Herein, we reported a facile method to synthesize Mg-doped Ca2SiO4 (CaMgSiO4) nanospheres with highly porous structures as pH-responsive drug delivery. Material characterizations confirmed the uniform Mg doping within calcium silicates. After the Mg doping, the surface area of calcium silicates increased from 185.68 m2/g to 212.71 m2/g. When incorporated with PEG surface coating, the porous CaMgSiO4 nanoparticles displayed over 50% increased doxorubicin (DOX) loading capacity compared with Ca2SiO4 nanoparticles. The pH-triggered DOX release behaviors and cytotoxicity of porous CaMgSiO4 nanoparticles were investigated. As a result, porous CaMgSiO4 nanoparticles were highly sensitive to pH stimulus and exhibited high biocompatibility and strong cancer cell killing effect (over 75%).

In this work, a reasonable strategy for the construction of Li2ZnTi3O8@Na2WO4 composite was employed to promote the Li storage performances of Li2ZnTi3O8. The Li2ZnTi3O8@Na2WO4 composites (5, 10, and 15 wt%) were then prepared by a solution dispersion method. The introduction of Na2WO4 does not change the structures of the samples and they show similar morphologies with particle sizes from 100 to 200 nm. Suitable amount of Na2WO4 modification effectively improves the electrochemical performance of Li2ZnTi3O8. Li2ZnTi3O8@Na2WO4 composites (0, 5, 10, and 15 wt%) deliver the discharge/charge capacities of 137.4/136.4, 164.2/162.3, 189.2/188.1, and 154.5/153.3 mAh g−1 at 0.5 A g−1 after 100 cycles, respectively. Li2ZnTi3O8@Na2WO4 composites (10 wt%) has the highest reversible capacities among all samples. The Na2WO4 shell with an excellent electronic conductivity can reduce electrode polarization, decrease the charge transfer resistance, enhance the Li-ion diffusion coefficient of Li2ZnTi3O8, and then improve the electrochemical kinetics of composites. In addition, the formation of Ti–O bonds at the interface can be helpful for the stabilization of the composite, being beneficial for the improvement of their cycling stabilities. These results reveal that Na2WO4 coating is a facile and effective strategy to promote the Li storage performance of Li2ZnTi3O8.

The information of when and where region-specific patterns in both mean and extreme temperatures leading to heat stress will emerge from the present-day climate variability is important to plan adaptation options, but to date studies on this issue still remain limited and fragmented. Here, we estimate the time of emergence (TOE) of temperature and wet-bulb temperature (Tw), a better indication of heat stress, using fine-scale, long-term regional climate model projections under the RCP2.6 and RCP8.5 scenarios across six different domains. Differently from previous studies, the TOE is determined using three methods applied on impact-relevant variables: two different signal-to-noise frameworks based on summer mean temperature and Tw and a statistical test to identify significant differences in daily extreme distributions. The TOE response to RCP2.6 and RCP8.5 with respect to the end of 20th century variability differs significantly regardless of which TOE metric is applied. For summer mean temperature, the land fraction reaching TOE is expected to exceed 90% by the 2050s under the RCP8.5, whereas the increase rate of land exposure to TOE tends to stagnate over time under the RCP2.6 so that more than 40% of land will not experience TOE by the end of the 21st century. Compared to temperature, the TOE of Tw is reached earlier in most of the wet tropics but is delayed in hot and dry regions because of the nonlinear response of Tw to humidity. For both temperature and Tw, the TOE appears earlier in regions with low baseline variability, such as in the tropics. Despite the uncertainties arising from the choice of TOE metrics, the vast majority of regions in Africa and southeast Asia experience TOE in the early 21st century under both the RCP2.6 and RCP8.5 scenarios, which stresses the urgent need for developing adequate adaptation strategies in these regions.

Lung cancer always ranks first in the number of cancer deaths every year, accounting for 18.4% of total cancer deaths in 2018. Metastasis is the main cause of death in lung cancer patients. The identification of bioactive components of traditional Chinese medicine is very important for the development of novel reagents against non-small cell lung cancer (NSCLC). Rosthorin A has originated from Rabdosia rosthornii (Diels) Hara which excerpts from 'Chinese materia medica', and is known to have 'clear heat phlegm' properties in the folk. Little is known about the biological functions and mechanisms of Rosthorin A in cancer cells at present. The role of EMT in metastasis of a tumor cell is self-evident. Slug is an important EMT inducer, which is related to the development of lung cancer. Cell growth, clone assay, cell migration, cell invasion, and protein expression, and NSCLC transplanted tumor growth were performed in A549, H1299, and H1975 cells. Rosthorin A significantly inhibited the growth of NSCLC cells, it could prolong the survival of nude mice. Rosthorin A inhibited the migration and invasion of A549, H1299, and H1975 cells. Rosthorin A up-regulated E-cadherin expression level and down-regulated the expression of β-catenin, N-cadherin, vimentin, Slug, and Twist. Rosthorin A could promote the expression of E-cadherin and inhibit the development of EMT by downregulating Slug, to inhibit the development and metastasis of NSCLC cells. In summary, Rosthorin A could be used as a promising candidate for the treatment of NSCLC patients with recurrence and metastasis.

Chronic wound repair is a common complication in patients with diabetes mellitus, which causes a heavy burden on social medical resources and the economy. Hypaphorine (HYP) has good anti-inflammatory effect, and chitosan (CS) is used in the treatment of wounds because of its good antibacterial effect. The purpose of this research was to investigate the role and mechanism of HYP-nano-microspheres in the treatment of wounds for diabetic rats. The morphology of HYP-NPS was observed by transmission electron microscopy (TEM). RAW 264.7 macrophages were used to assess the bio-compatibility of HYP-NPS. A full-thickness dermal wound in a diabetic rat model was performed to evaluate the wound healing function of HYP-NPS. The results revealed that HYP-NPS nanoparticles were spherical with an average diameter of approximately 50 nm. The cell experiments hinted that HYP-NPS had the potential as a trauma material. The wound test in diabetic rats indicated that HYP-NPS fostered the healing of chronic wounds. The mechanism was through down-regulating the expression of pro-inflammatory cytokines IL-1β and TNF-α in the skin of the wound, and accelerating the transition of chronic wound from inflammation to tissue regeneration. These results indicate that HYP-NPS has a good application prospect in the treatment of chronic wounds.

To investigate the efficacy of the extract from Yiyuan Yiliu Tang (, YYYLT) on human lung adenocarcinoma cells A549 and human hepatoma cells Bel7402.The cancer cell lines were treated with various concentrations (0, 100, 200, 300, and 400 μg/mL) of the crude water extract of YYYLT and then cell viability, toxicity, cytokine secretion, and cell cycle/apoptosis were determined by MTT assay, LDH assay, and flow cytometry, respectively.The extract from YYYLT significantly suppressed the proliferation of the cancer cell lines and the release of interleukin-2 and tumor necrosis factor-α in a dose-dependent manner. The extract also promoted apoptosis, caused cell cycle arrest at G0/G1 phase, and increased the expression of caspase-3, B-cell lymphoma-2 (Bcl-2), and Bcl-2-associated X proteins.The extract from YYYLT might be a potential treatment for human lung and liver cancers.

Schistosomiasis is an immunopathogenic disease in which a T helper (Th) cell type 2-like response plays vital roles. Hepatic fibrosis is its main pathologic manifestations, which is the leading cause of hepatic cirrhosis. Co-infections of Schistosoma japonicum (Sj) with other pathogens are frequently encountered but are easily ignored in clinical studies, and effective therapeutic interventions are lacking. In this study, we explored the effect of Toxoplasma gondii (Tg) prior infection on Th1/Th2 response, community shifts in gut microbiome (GM), and the pathogenesis of schistosomiasis in murine hosts. Mice were prior infected with Tg before Sj infection. The effects of co-infection on Th1/Th2 response and hepatic fibrosis were analyzed. Furthermore, we investigated this issue by sequencing 16S rRNA from fecal specimens to define the GM profiles during co-infection. Tg prior infection markedly reduced the granuloma size and collagen deposit in livers against Sj infection. Prior infection promoted a shift toward Th1 immune response instead of Th2. Furthermore, Tg infection promoted the expansion of preponderant flora and Clostridiaceae was identified as a feature marker in the GM of the co-infection group. Redundancy analysis (RDA)/canonical correspondence analysis (CCA) results showed that liver fibrosis, Th1/Th2 cytokines were significantly correlated (P &lt, 0.05) with the GM compositions. Tg infection inhibits hepatic fibrosis by downregulating Th2 immune response against Sj infection, and further promotes the GM shifts through &ldquo, gut&ndash, liver axis&rdquo, in the murine hosts. Our study may provide insights into potential anti-fibrosis strategies in co-infection individuals.

Dietary copper supplementation reverses pressure overload-induced cardiac hypertrophy by copper replenishment in the heart. A copper-selective chelator, trientine (triethylenetetramine [TETA]), reverses left ventricular hypertrophy associated with diabetes also by copper replenishment in the heart. The present study was undertaken to address the critical issue how TETA delivers copper to the heart. Adult male Sprague-Dawley rats were subjected to transverse aortic constriction (TAC) to induce cardiac hypertrophy. Eight weeks after the TAC surgery, cardiac hypertrophy was developed and copper content in the heart was reduced. TETA was then administrated by gavage in two different dosages (21.9 or 87.6 mg/kg day) for six weeks. The results showed that in the lower dosage, TETA replenished copper contents in the heart, along with a decrease in the copper concentration in the blood and kidney, and an increase in the urine. In the higher dosage, TETA did not replenish copper contents in the heart, but markedly increased copper concentrations in the urine and decreased those in the blood and kidney. Neither lower nor higher TETA dosage altered copper concentrations in other organs. Corresponding to myocardial copper replenishment, the lower dose TETA suppresses cardiac hypertrophy, as judged by a reduction in the left ventricle wall thickness and a decrease in the heart size, and diminished cardiac fibrosis, as reflected by a decrease in collagen I content. TETA in the higher dose not only did not suppress cardiac hypertrophy, but also caused cardiac hypertrophy in sham-operated rats. TETA-mediated myocardial copper restoration is independent of copper transporter-1 or -2 but related to an energy-dependent transportation. This study demonstrates that low-dose TETA functions as a copper chaperone, selectively delivering copper to the copper-deprived heart through an active transportation; in higher doses, TETA simply retains its chelator function, removing copper from the body by urinary excretion. Impact statement Our study reveals that TETA, traditionally regarded as a copper chelator, in lower doses delivers copper selectively to the heart through a mechanism independent of copper transporter-1 or -2. Copper supplementation by a lower dose of TETA suppresses pressure overload-induced cardiac hypertrophy. Since ischemic heart disease and hypertrophic cardiomyopathy are accompanied by myocardial copper loss, this approach of using a lower dose of TETA to supplement copper to the heart would help treat the disease condition of patients with such cardiac events.

Angiogenesis is a complex physiological process involving the growth of new capillaries. The impaired angiogenesis plays important roles in chronic wounds and ischaemic heart disease. Fibroblast growth factor 2 (FGF-2) exerts pro-angiogenic actions via activation of fibroblast growth factor receptor 1 (FGFR-1). We have identified that vaccarin increased the angiogenic activity of endothelial cells. In this study, we investigated whether FGF-2-mediated FGFR1 signaling pathway participated in vaccarin-mediated neovascularization formation. Human microvascular endothelial cells (HMEC)-1 were incubated with various doses of vaccarin. Our results showed that vaccarin dose-dependently up-regulated FGF-2 levels and phosphorylation of FGFR-1. Neutralization of FGF-2 with anti-FGF-2 antibody also abolished the proliferation, migration and tube formation of HMEC-1 cells induced by vaccarin. Both FGFR-1 inhibitor SU5402 and FGFR-1 siRNA blocked vaccarin-induced cell cycle progression and angiogenesis. The mouse Matrigel model study further unveiled that vaccarin stimulated the neovascularization and microvessel density in vivo, which was prevented by FGFR-1 inhibitor SU5402. Taken together, our results demonstrated for the first time that vaccarin was a novel inducer for FGF-2 expression, followed by phosphorylation of FGFR-1 and subsequent angiogenic behaviors in endothelial cells. Vaccarin may be a promising candidate of angiogenesis activator for neurovascular repair or therapy.

Background: Diabetes mellitus is a growing global health issue nearly across the world. Diabetic patients who are prone to develop diabetes-related complications often exhibit progressive neuropathy (painless and sensory loss). It is usual for small wounds to progress to ulceration, which especially worsens with peripheral arterial disease and in the presence of anaerobic bacteria, culminating into gangrene. In our study, vaccarin (VAC), the main active monomer extracted from Chinese herb vaccariae semen, is proven to have a role in promoting diabetic chronic wound healing through a cytoprotective role under high glucose conditions. Materials and methods: We constructed a pressure ulcer on both VAC-treated and control mice based on a type 1 diabetes (T1DM) model. The wound healing index was evaluated by an experimental wound assessment tool (EWAT). We also determined the effect of VAC on the proliferation and cell migration of human microvascular endothelial cells (HMEC-1) by a cell counting kit (CCK-8), a scratch and transwell assay. Results: The results demonstrated that VAC could promote the proliferation and migration of high glucose-stimulated HMEC-1 cells, which depend on the activation of FOXP2/AGGF1. Activation of the angiogenic factor with G patch and FHA domains 1 (AGGF1) caused enhanced phosphorylation of serine/threonine kinase (Akt) and extracellular regulated protein kinases (Erk1/2). By silencing the expression of forkhead box p2 (FOXP2) protein by siRNA, both mRNA and protein expression of AGGF1 were downregulated, leading to a decreased proliferation and migration of HMEC-1 cells. In addition, a diabetic chronic wound model in vivo unveiled that VAC had a positive effect on chronic wound healing, which involved the activation of the above-mentioned pathways. Conclusions: In summary, our study found that VAC promoted chronic wound healing in T1DM mice by activating the FOXP2/AGGF1 pathway, indicating that VAC may be a promising candidate for the treatment of the chronic wounds of diabetic patients.

This study investigates the performance of the latest version of RegCM4 in simulating summer precipitation over South Korea, comparing nine sensitivity experiments with different combinations of convective parameterization schemes (CPSs) between land and ocean. In addition to the gross pattern of seasonal and monthly mean precipitation, the northward propagation of the intense precipitation band and statistics from extreme daily precipitation are thoroughly evaluated against gridded and in situ station observations. The comparative analysis of 10-year simulations demonstrates that no CPS shows superiority in both quantitative and qualitative aspects. Furthermore, a nontrivial discrepancy among the different observation datasets makes a robust assessment of model performance difficult. Regardless of the CPS over the ocean, the simulations with the Kain–Fritsch scheme over land show a severe dry bias, whereas the simulations with the Tiedtke scheme over land suffer from a limited accuracy in reproducing spatial distributions due to the excessive orographic precipitation. In general, the simulations with the Emanuel scheme over land are better at capturing the major characteristics of summer precipitation over South Korea, despite not all statistical metrics showing the best performance. When applying the Emanuel scheme to both land and the ocean, precipitation tends to be slightly overestimated. This deficiency can be alleviated by using either the Tiedtke or Kain–Fritsch schemes over the ocean instead. As few studies have applied and evaluated the Tiedtke and Kain–Fritsch schemes to the Korean region within the RegCM framework, and this study introduces the potential of these new CPSs compared with the more frequently selected Emanuel scheme, which is particularly beneficial to RegCM users.

This study describes the chemical constituents of Albiziae Cortex and their ability to ameliorate steatosis and promote proliferation and anti-oxidation in vitro. Together, five known lignan glycosides, (7S,8R)-erythro-syringylglycerol-&beta, &ndash, O-4&prime, sinapyl ether 9-O-&beta, D-glucopyranoside (1), (+)-lyoniresinol-9&prime, O-gluco-side (2), (&minus, )-lyoniresinol-9&prime, O-glucoside (3), picraquassioside C (4), and icariside E5 (5), were isolated from the Albiziae Cortex. Their structures were elucidated by extensive NMR and high-resolution mass spectrometry analysis and compared with reported data. Oil Red O staining results revealed that compounds 1, 2, and 3 attenuated lipid accumulation and lipid metabolic disorders in FFAs (oleate/palmitate, 2:1 ratio, 0.3 mM)-exposed HepG2 cells. The Cell Counting Kit 8 (CCK-8) assay results revealed that compounds 1 and 5 can significantly promote human umbilical vein endothelial cell (HUVEC) proliferation, meanwhile, these compounds did not exhibit significant cytotoxicity against HUVECs. In addition, 2&prime, 7&prime, dichlorofluorescein diacetate (DCFH-DA) staining results revealed that high glucose (HG)-induced reactive oxygen species (ROS) production was abolished by compounds 1, 2, and 3. This is the first report of the isolation of lignan skeletons from the genus Albizzia julibrissin with the ability to ameliorate steatosis and promote proliferation and anti-oxidation activities.

Cardiovascular complications are a major leading cause of mortality in patients suffering from type 2 diabetes mellitus (T2DM). Vascular endothelial dysfunction is a core pathophysiological event in the early stage of T2DM and eventually leads to cardiovascular disease. Vaccarin (VAC), an active flavonoid glycoside extracted from vaccariae semen, exhibits extensive biological activities including vascular endothelial cell protection effects. However, little is known about whether VAC is involved in endothelial dysfunction regulation under high glucose (HG) or hyperglycemia conditions. Here, in an in vivo study, we found that VAC attenuated increased blood glucose, increased glucose and insulin tolerance, relieved the disorder of lipid metabolism and oxidative stress, and improved endothelium-dependent vasorelaxation in STZ/HFD-induced T2DM mice. Furthermore, in cultured human microvascular endothelial cell-1 (HMEC-1) cells, we showed that pretreatment with VAC dose-dependently increased nitric oxide (NO) generation and the phosphorylation of eNOS under HG conditions. Mechanistically, VAC-treated HMEC-1 cells exhibited higher AMPK phosphorylation, which was attenuated by HG stimulation. Moreover, HG-triggered miRNA-34a upregulation was inhibited by VAC pretreatment, which is in accordance with pretreatment with AMPK inhibitor compound C (CC). In addition, both reactive oxygen species (ROS) scavenger N-acetyl-L-cysteine (NAC) and VAC abolished HG-evoked dephosphorylation of AMPK and eNOS, increased miRNA-34a expression, and decreased NO production. These results suggest that VAC impedes HG-induced endothelial dysfunction via inhibition of the ROS/AMPK/miRNA-34a/eNOS signaling cascade.

Purpose To explore the potential role and unclear molecular mechanisms of vaccarin in wound healing. Methods Rats’ skin excision model to study the effects of vaccarin on wound healing in vivo . Hematoxylin and eosin staining was performed to evaluate Histopathologic characteristics. Immunohistochemistry was employed to assess the effects of vaccarin in accelerating angiogenesis. Western blot was used to evaluate relative protein expressed levels. Results Vaccarin could significantly promote wound healing and endothelial cells and fibroblasts proliferation in the wound site. Immunohistochemistry and Western blot studies showed that the nodal proteins and receptor (bFGFR) related to angiogenesis signaling pathway were activated, and the microvascular density in the wound site was markedly higher than that in the control group. Conclusions The present study was the first to demonstrate that vaccarin is able to induce angiogenesis and accelerate wound healing in vivo by increasing expressions of p-Akt, p-Erk and p-bFGFR. This process is mediated by MAPK/ERK and PI3K/AKT signaling pathways.

The bamboo shoot of Pleioblastus amarus (Keng) Keng f. is a medicinal and edible plant product in China. In this study, the chemical composition of the total alkaloids from bamboo shoots and bamboo shoot shells of P. amarus (Keng) Keng f. (ABSP and ABSSP, respectively) were separated and investigated by UHPLC/QTOF-MS/MS. The results showed that a total of 32 alkaloids were extracted, with 15 common to both ABSP and ABSSP and 10 and 7 alkaloids distinct to ABSP and ABSSP, respectively. ABSP and ABSSP both decreased the lipopolysaccharide (LPS, 0.5 &mu, g/mL)-induced nitric oxide (NO) production in RAW264.7 murine macrophages with half maximal inhibitory concentration (IC50) values of 78 and 55 &mu, g/mL, respectively. We also found that ABSP and ABSSP (100 &mu, g/mL) could decrease the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at both mRNA and protein levels in LPS-exposed RAW264.7 cells. Moreover, 100 &mu, g/mL of ABSP and ABSSP also significantly inhibited LPS-induced mRNA expression of interleukin 1&beta, (IL-1&beta, ) and tumor necrosis factor &alpha, (TNF-&alpha, ). Additionally, ABSP and ABSSP (100 &mu, g/mL) decreased the phosphorylation of extracellular regulated protein kinase (ERK) in LPS-stimulated RAW264.7 cells. Collectively, the total alkaloids from the bamboo shoots and shells of P. amarus exhibit anti-inflammatory effects in LPS-activated RAW264.7 cells through the inhibition of ERK signaling. This result can provide support for the medicinal use and further study of P. amarus.

Objective Vasculogenic mimicry (VM) is a novel mechanism of tumor blood supply distinct from endothelial vessel (EV). VM is associated with malignancy, invasion, metastasis, and poor prognosis. Hitherto a noninvasive method for the assessment of VM in vivo has been lacking. Methods Contrast-enhanced ultrasound (CEUS) was performed to evaluate the quantitative parameters of tumors in mice. CD31 immunohistochemistry-Periodic Acid-Schiff double staining was conducted to identify the VM or EV in tumor tissues. Correlations between perfusion parameters and VM density was analyzed by Pearson correlation test. Results By the 15th day after tumor inoculation, the EV and VM density was 31.15 ± 7.14 and 14.11 ± 2.99 per 200× field. The maximal intensity (IMAX) was 301.19 ± 191.56%, and the rise time (RT), time to peak (TTP) and mean transit time (mTT) were 17.38 ± 7.82 s, 20.27 ± 9.61 s and 58.09 ± 26.44 s, respectively. Cancer-related fatigue (CRF) is one of the most common and serious complications in cancer patients, which greatly reduces the quality of life. The mechanism induced fatigue may be diverse. In this study, we tried to investigate the effect of 1,25(OH)2D3, the active metabolite of vitamin D on CRF in Lewis lung cancer-bearing mice. Network pharmacological analysis, behavioral testing, western blotting, ELISA and flow cytometry were used. We found that there was an interaction between proteins related to the role of 1,25(OH)2D3 and CRF-related proteins. The results of animal model experiments showed that 1,25(OH)2D3 could mitigate the CRF behavior of tumor-bearing mice, and the treatment of 1,25(OH)2D3 reduced the levels of inflammatory factors, changed the tryptophan metabolism direction, and caused changes in immune cells. Collectively, 1,25(OH)2D3 might improve CRF in tumor-bearing mice by changing the direction of tryptophan metabolism and inflammatory factor levels. This study provided a possible solution for patients with clinical CRF.

A drug vaccarin loaded polymer poly (vinyl alcohol) (PVA)-stilbazole quaternized (SbQ)/Zein was prepared in this study, using co-electrospun method. Then the morphologies and structures of PVA-SbQ/Zein composite nanofibers were observed by scanning electron microscope (SEM) and Fourier transform infrared spectrum (FTIR), respectively. Finally, biocompatibility of PVA-SbQ/Zein nanofibers with drug and without drug was evaluated. Results showed that vaccarin-loaded PVA-SbQ/Zein nanofibers had smooth surface and showed non-toxic to L929 cells. Drug vaccarin could promote cells attachment on nanofibers. The wound healing performance was examined本文采用静电纺丝制备了负载王不留行黄酮苷的聚乙烯醇苯乙烯吡啶盐（PVA-SbQ）/玉米醇溶蛋白（Zein）纳米纤维，对纳米纤维的表面形貌和结构进行了观察，并对其进行了生物相容性评价。将负载王不留行黄酮苷的 PVA-SbQ/Zein 纳米纤维作为实验组，以凡士林纱布为对照组，建立小鼠皮肤损伤模型，对两种敷料在小鼠皮肤上的修复作用进行组织切片观察。实验结果表明，成功制备了负载王不留行黄酮苷的纳米纤维并呈现良好的形貌；负载黄酮苷的 PVA-SbQ/Zein 纳米纤维对 L929 细胞无毒害作用，且黄酮苷的存在可以促进细胞黏附生长。基于 PVA-SbQ/Zein/黄酮苷纳米纤维膜的创伤愈合材料对小鼠伤口愈合的效果要优于凡士林纱布。.

Bacterial cellulose (BC) and bacterial cellulose-vaccarin (BC-Vac) membranes were successfully produced in large scale. BC was synthesized by Gluconacetobacter xylinum. BC-Vac membranes were prepared by immersing BC in vaccarin solution. The surface morphologies of BC and BC-Vac membranes were examined by a scanning electron microscope (SEM) and an atomic force microscopy (AFM). The images showed that BC-Vac exhibited the characteristic 3D nanofibrillar network of BC matrix but there was adhesion between fibers. The mechanical properties of BC and BC-Vac membranes were evaluated and the results indicated that the adding of drug vaccarin into the BC membranes increased the malleability indicated by the increment in elongation at break compared with BC. Fourier transform infrared spectroscopy (FTIR) analysis was conducted to confirm the incorporation of vaccarin in BC-Vac and investigate the hydroxyl interactions between BC and drug vaccarin. Cell viability and cell attachment studies demonstrated that BC and BC-Vac membranes had no cytotoxicity and could be a good carrier for cell growth. The wound healing performance was examined in vivo by rat skin models. Histological observations revealed that wounds treated with BC-Vac epithelialized and regenerated faster than treated with BC. Therefore, BC-Vac was considered as a potential candidate for wound dressing materials.

Endothelial cell injury is a critical component of atherosclerosis and hypertension. Vaccarin is considered to be of potential benefit in the treatment of vascular diseases. The aim of the present study was to evaluate the possible effects of vaccarin in human EA·hy926 cells induced by high glucose, and to investigate its underlying mechanism in the prevention and treatment of high glucose‑induced injury. In the present study, EA·hy926 cells were exposed to 90, 180 and 270 mM high glucose for 24 h, and the induced cell injury was examined using a sulforhodamine B assay. Following treatment with high glucose, it was found that high glucose stimulated cell injury, resulting in reduced cell viability and migratory ability, increased lactate dehydrogenase (LDH) leakage and malondialdehyde (MDA) levels, and decreased superoxide dismutase (SOD) activity. High glucose further accelerated cell apoptosis via activating Notch1 and Hairy and enhancer of split 1. It was found that preincubation with vaccarin protected the EA·hy926 cells from high glucose‑induced cell injury, which promoted cell viability and migratory ability, inhibited the expression levels of LDH and MDA, and enhanced the activity of SOD. Cell migratory ability, LDH leakage, MDA levels and decreasing SOD activity were evaluated using a wound healing assay and corresponding assay kits. Cell apoptosis was detected by flow cytometry with an Annexin V‑fluorescein isothiocyanate/propidium iodide apoptosis detection kit and Hoechst staining. Furthermore, western blotting was used to detect the protein expression levels of Notch1, Hes1 and caspase‑3. In particular, in addition to inducing the downregulation of Notch signaling, vaccarin treatment downregulated the cell apoptotic pathway‑associated protein caspase 3. These findings suggested that vaccarin may be able to selectively protect the vascular endothelium from dysfunction induced by high glucose.

Human-altered flow regime is often claimed to be one of the most serious and continuing threats to aquatic biodiversity in streams and rivers. It is commonly found that both seasonal and subdaily flow patterns are changed below hydropeaking dams due to the multifunctions of the reservoirs such as flood control and water supply. The aim of this paper is to comprehensively quantify the degree of ecologically relevant hydrologic alteration downstream of Gangkouwan Reservoir, focusing particularly on the subdaily flow variation induced by hydropeaking operations. The method is based on the assessment framework set by the indicators of hydrologic alteration (IHA). Hydrologic parameters in groups 1–3 of the IHA were adopted to assess the degree of hydrologic alteration at daily to monthly time scales and based on a daily mean flow data series for both the pre- and post-impact periods. A set of biologically relevant hydrologic parameters based on post-dam hourly flow data, including magnitude and duration of baseflow, magnitude and duration of peaking flows, frequency of peaking flows and ramping rates, were proposed to depict subdaily flow pulses and water condition changes induced by hydropeaking. The results show that Gangkouwan Reservoir made the intra-annual distribution of runoff more consistent, reduced annual peak discharges, and changed the timing of the yearly maximum. The monthly distribution of these hydrologic parameters at the hourly temporal scale provides detailed information on ecologically significant features of the subdaily flow variation induced by hydropeaking operations. This paper provides useful guidance for further investigations on responses of river biota to hydropeaking and provides guidance for suggesting mitigation measures against hydropeaking effects.

The existing incentive mechanisms of crowdsourcing construct the expected utility function based on the assumption of rational people in traditional economics. A large number of studies in behavioral economics have demonstrated the defects of the traditional utility function and introduced a new parameter called loss aversion coefficient to calculate individual utility when it suffers a loss. In this paper, combination of behavioral economics and a payment algorithm based on the loss aversion is proposed. Compared with usual incentive mechanisms, the node utility function is redefined by the loss aversion characteristic of the node. Experimental results show that the proposed algorithm can get a higher rate of cooperation with a lower payment price and has good scalability compared with the traditional incentive mechanism.

Dried stem bark from Albizia julibrissin (AJ) is a highly valued Traditional Chinese Medicine, which has been shown to suppress tumor growth and angiogenesis. Total saponins from AJ (TSAJ) are one of the most bioactive components of AJ extract. The present study evaluated the anti‑tumor and anti‑angiogenic effects of TSAJ in vitro and in vivo. The anti‑angiogenic activity of TSAJ was investigated by measuring the effects on vascular endothelial growth factor (VEGF)‑induced proliferation, migration and tube formation of Ea.hy926 endothelial cells in vitro. The expression levels of proteins associated with VEGF‑induced angiogenesis were determined by western blotting. Furthermore, in vivo Matrigel™ plug and H22 hepatoma tumor models were used to verify the anti‑angiogenic effects of TSAJ. The present study demonstrated that TSAJ significantly inhibited VEGF‑mediated endothelial cell proliferation, migration and tube formation of Ea.hy926 cells in vitro. The anti‑angiogenic effects of TSAJ were modulated by suppression of phosphorylated‑(p‑) focal adhesion kinase, p‑Akt, and p‑extracellular signal‑regulated kinase in the VEGF/VEGF receptor 2 (R2) signaling pathway. Furthermore, oral administration of TSAJ significantly inhibited tumor growth and tumor‑induced angiogenesis, as well as the formation of functional vessels, in the Matrigel™ plug model. These results suggest that TSAJ may be a potential anti‑angiogenic agent that targets the VEGF/VEGFR2 signaling pathway, and inhibits tumor‑induced angiogenesis.

The kidney is an important organ in the regulation of blood pressure, and it is also one of the primary target organs of hypertension. Kidney damage in response to hypertension eventually leads to renal insufficiency. The authors previously demonstrated that vaccarin exhibits a protective role in endothelial injury. However, the effects of vaccarin on the two-kidney, one clip (2K1C) renovascular hypertension model and subsequent kidney injury have yet to be fully elucidated. The present study was designed to investigate the roles and mechanisms of vaccarin in attenuating hypertension and whether vaccarin had beneficial effects on kidney injury. The 2K1C rats had greater fibrosis, apoptosis, reactive oxygen species production, inflammation, angiotensin II (Ang II) and angiotensin type 1 (AT1) receptors in the right kidney compared with normotensive rats, which were alleviated by a high dose of vaccarin and captopril. Vaccarin treatment attenuated hypertension, reduced fibrosis markers, NADPH oxidase (NOX)-2, NOX-4, 3-nitrotyrosine, tumor necrosis factor-α, interleukin 1β (IL-1β), and IL-6 protein levels and altered pro-apoptotic protein levels including caspase-3, anti-apoptosis protein B cell lymphoma (Bcl)-2 and Bcl-2 associated X, apoptosis regulator in the right kidney of 2K1C rats. These findings suggest that the protective effects of vaccarin on the right kidney in renovascular hypertension are possibly due to downregulation of fibrosis, inflammatory molecules, oxidative stress, Ang II, and AT1 receptor levels.

Oxidized low-density lipoprotein (ox-LDL) accumulation is one of the critical determinants in endothelial dysfunction in many cardiovascular diseases such as atherosclerosis. C1q/TNF-related protein 9 (CTRP9) is identified to be an adipocytokine with cardioprotective properties. However, the potential roles of CTRP9 in endothelial function remain largely elusive. In the present study, the effects of CTRP9 on the proliferation, apoptosis, migration, angiogenesis, nitric oxide (NO) production and oxidative stress in human umbilical vein endothelial cells (HUVECs) exposed to ox-LDL were investigated. We observed that treatment with ox-LDL inhibited the proliferation, migration, angiogenesis and the generation of NO, while stimulated the apoptosis and reactive oxygen species (ROS) production in HUVECs. Incubation of HUVECs with CTRP9 rescued ox-LDL-induced endothelial injury. CTRP9 treatment reversed ox-LDL-evoked decreases in antioxidant enzymes including heme oxygenase-1 (HO-1), nicotinamide adenine dinucleotide phosphate (NAD(P)H) dehydrogenase quinone 1, and glutamate-cysteine ligase (GCL), as well as endothelial nitric oxide synthase (eNOS). Furthermore, CTRP9 induced activation of peroxisome proliferator-activated receptor γ co-activator 1α (PGC1-α) and phosphorylation of adenosine monophosphate-activated protein kinase (AMPK). Of interest, AMPK inhibition or PGC1-α silencing abolished CTRP9-mediated antioxidant enzymes levels, eNOS expressions, and endothelial protective effects. Collectively, we provided the first evidence that CTRP9 attenuated ox-LDL-induced endothelial injury by antioxidant enzyme inductions dependent on PGC-1α/AMPK activation.

Endothelial lesion response to injurious stimuli is a necessary step for initiating inflammatory cascades in blood vessels. Hypaphorine (Hy) from different marine sources is shown to exhibit anti-inflammatory properties. However, the potential roles and possible molecular mechanisms of Hy in endothelial inflammation have yet to be fully clarified. We showed that Hy significantly inhibited the positive effects of lipopolysaccharide (LPS) on pro-inflammatory cytokines expressions, including tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), monocyte chemoattractant protein 1 (MCP-1) and vascular cellular adhesion molecule-1 (VCAM-1), as well as induction of the phosphorylation of Akt and mTOR in HMEC-1 cells. The downregulated peroxisome proliferator-activated receptor γ (PPAR-γ) and upregulated toll-like receptor 4 (TLR4) expressions in LPS-challenged endothelial cells were prevented by Hy. Inhibition of both PI3K and mTOR reversed LPS-stimulated increases in TLR4 expressions and decreases in PPAR-γ levels. Genetic silencing of TLR4 or PPAR-γ agonist pioglitazone obviously abrogated the levels of pro-inflammatory cytokines in LPS-treated HMEC-1 cells. These results suggest that Hy may exert anti-inflammatory actions through the regulation of TLR4 and PPAR-γ dependent on PI3K/Akt/mTOR signal pathways. Hy may be considered as a therapeutic agent that can potentially relieve or ameliorate endothelial inflammation-associated diseases.
