Objective To investigate the effects of controlled hypotension induced by sodium nitroprusside on regional cerebral blood flow and oxygenation in pigs. Methods Six healthy female Landrace pigs (aged 4~6 months, weighing 22~25 kg) were included in this study. Sodium nitroprusside was used for controlled hypotension under sevoflurane anesthesia (20%, 30% and 50% lower than the baseline of mean arterial pressure or mean arterial pressure (MAP), corresponding to mild, moderate and severe hypotension, respectively). Oxygen to see (O2C), a monitoring instrument, was used to measure regional cerebral blood flow (rCBF) and regional cerebral oxygen saturation (rSO2). rCBF, rSO2, regional cerebral haemoglobin (rHb), heart rate (HR), peripheral oxygen saturation (SpO2) and incidence of cerebral hypoxia events were recorded under different MAP. Results rCBF was significantly correlated with MAP (r=0.793) and also obviously correlated with rSO2 (r=0.843) in the whole hypotension process. rSO2 had a linear correlation with rHb, MAP and rCBF (R2=0.853, P < 0.05), respectively. And the order of their linear correlation with rSO2 is rHb>MAP>rCBF. During mild hypotension, rCBF and rSO2 were increased by 7.4%(P < 0.05) and 2.5%(P < 0.05), respectively compared with the baseline, and the increase of rCBF was more remarkable. During moderate hypotension, compared with the baseline, rCBF was decreased by 0.9% (P>0.05), while rSO2 was decreased by 5.8% (P < 0.05). During severe hypotension, rCBF and rSO2 were decreased by 7.6% (P < 0.05) and 12.1% (P < 0.05) respectively, and there was no cerebral hypoxia event during mild to severe hypotension. From 5 to 15 min after drug withdrawal, both rCBF and rSO2 were increased by 8.4% and 3.7%, respectively when compared with the baseline (both P < 0.05). Conclusion Controlled hypotension with sodium nitroprusside is beneficial to maintain cerebral perfusion and cerebral oxygenation, but mild hypotension can cause an increase in cerebral blood flow, which is disadvantageous to patients with increased intracranial pressure.

Objective To investigate the accuracy of surgical pleth index (SPI) in monitoring patients with nociceptive stimulation and its predictability for cardiovascular response to nociceptive stimulation in total intravenous anesthesia (TIVA). Methods A total of 102 patients (aged 18 to 65 years, ASAⅠtoⅡ, and BMI of 18.5 to 30 kg/㎡) who underwent elective thyroid surgery in our hospital from February to June 2022 were enrolled in this study. Each patient was induced by intravenous anesthesia, maintained by target-controlled infusion of propofol and remifentanil, and monitored by SPI for analgesia depth. The absolute values of SPI, heart rate (HR) and mean arterical pressure (MAP) of patients 2 min before and during nociceptive stimulation and the maximum values of SPI, HR and MAP within 2 min after nociceptive stimulation were recorded. Receiver operating characteristic (ROC) curve was used to analyze the accuracy of SPI, HR and MAP in monitoring nociceptive stimulation and the predictability of SPI in cardiovascular response to nociceptive stimulation. Results ROC curve analysis indicated that SPI (AUC=0.931, Cutoff value=30) was superior to HR (AUC=0.804, Cutoff value=68) and MAP (AUC=0.759, Cutoff value=80) in monitoring accuracy of tracheal intubation stimulation, ΔSPI (AUC=0.990, Cutoff value=4) was better than ΔHR (AUC=0.972, Cutoff value=3) and ΔMAP (AUC=0.854, Cutoff value=3), ΔSPI was better than SPI, ΔHR was better than HR, and ΔMAP was better than MAP, with their AUC values statistically different (P < 0.05). In terms of monitoring the accuracy of skin incisions, SPI (AUC=0.925, Cutoff value=43) was superior to HR (AUC=0.587, Cutoff value=65) and MAP (AUC=0.804, Cutoff value=76), ΔSPI (AUC=0.998, Cutoff value=7) was better than ΔHR (AUC=0.833, Cutoff value=1) and ΔMAP (AUC=0.943, Cutoff value=3), ΔSPI was better than SPI, ΔHR was better than HR, and ΔMAP was better than MAP, and the above AUC differences were statistically significant (P < 0.05); SPI had predictive value for the cardiovascular response stimulated by tracheal intubation (AUC=0.662, Cutoff value=17, P < 0.05), but had no predictive significance for the cardiovascular response stimulated by skin incisions (P>0.05). Conclusion The absolute value and change of SPI are better than the traditional hemodynamic indexes HR and MAP in monitoring the accuracy of nociceptive stimulation, which can accurately monitor nociceptive stimulation during operation. SPI has predictive value for the cardiovascular response stimulated by intubation stimulation, and has important clinical significance for judging the appropriate timing of intubation.

Isocitrate dehydrogenase (IDH) and O6-methylguanine-DNA methyltransferase (MGMT) genes are critical molecular markers in determining treatment options and predicting the prognosis of adult-type diffuse gliomas. Objectives: this study aimed to investigate whether multimodal MRI enables the differentiation of genotypes in adult-type diffuse gliomas. Methods: a total of 116 adult-type diffuse glioma patients (61 males, 51.5 (37, 62) years old) who underwent multimodal MRI before surgery were retrospectively analysed. Multimodal MRI included conventional MRI, proton magnetic resonance spectroscopy (1H-MRS), and diffusion tensor imaging (DTI). Conventional visual features, N-acetyl-aspartate (NAA)/Creatine (Cr), Choline (Cho)/Cr, Cho/NAA, fractional anisotropy (FA), mean diffusivity (MD), and diffusion histogram parameters were extracted on the whole tumour. Multimodal MRI parameters of IDH-mutant and IDH-wildtype gliomas were compared using the Mann–Whitney U test, Student's t-test, or Pearson chi-square tests. Logistic regression was used to select the MRI parameters to predict IDH-mutant gliomas. Furthermore, multimodal MRI parameters were selected to establish models for predicting MGMT methylation in the IDH-wildtype gliomas. The performance of models was evaluated by the receiver operating characteristics curve. Results: a total of 56 patients with IDH-mutant gliomas and 60 patients with IDH-wildtype glioblastomas (GBM) (37 with methylated MGMT and 17 with unmethylated MGMT) were diagnosed by 2021 WHO classification criteria. The enhancement degree (OR = 4.298, p < 0.001), necrosis/cyst (OR = 5.381, p = 0.011), NAA/Cr (OR = 0.497, p = 0.037), FA-Skewness (OR = 0.497, p = 0.033), MD-Skewness (OR = 1.849, p = 0.035), FAmean (OR = 1.924, p = 0.049) were independent factors for the multimodal combined prediction model in predicting IDH-mutant gliomas. The combined modal based on conventional MRI, 1H-MRS, DTI parameters, and histogram performed best in predicting IDH-wildtype status (AUC = 0.890). However, only NAA/Cr (OR = 0.17, p = 0.043) and FA (OR = 0.38, p = 0.015) were associated with MGMT methylated in IDH-wildtype GBM. The combination of NAA/Cr and FA-Median is more accurate for predicting MGMT methylation levels than using these elements alone (AUC, 0.847 vs. 0.695/0.684). Conclusions: multimodal MRI based on conventional MRI, 1H-MRS, and DTI can provide compound imaging markers for stratified individual diagnosis of IDH mutant and MGMT promoter methylation in adult-type diffuse gliomas. Simple Summary: The gastrointestinal epithelium is important for maintaining intestinal homeostasis. During piglet production, excessive reactive oxygen species (ROS) can lead to intestinal mucosal oxidative injury, endangering the health of piglets. Moreover, oxidative stress has also been reported to be associated with apoptosis and ferroptosis. Probiotics are beneficial to intestinal health. In this study, piglets were fed a diet containing Lactiplantibacillus plantarum P8 (P8) for 29 days and injected with diquat (DQ) to induce oxidative stress on the 22nd day of the experiment in order to investigate the effects and mechanism of P8 on the intestinal oxidative injury. The results suggest that dietary supplementation of P8 contributed to an improvement in jejunal mucosal antioxidant capacity, microstructure and barrier function, and a decrease in apoptosis and ferroptosis. Jejunal transcriptome analysis further reveals that the beneficial effect of P8 may be related to the regulation of PI3K/AKT and NF-κB signaling pathways. Oxidative stress induces apoptosis and ferroptosis, leading to intestinal injury of piglets. Lactiplantibacillus plantarum P8 (P8) has antioxidant capacity, but its roles in intestinal apoptosis and ferroptosis remain unclear. Here, 24 weaned piglets were assigned to three treatments: control (Con), diquat injection (DQ), and P8 supplementation + DQ injection (DQ + P8). The results showed that the increased jejunal oxidative stress, jejunal morphology impairment, and barrier dysfunction in the DQ-treated piglets were decreased by P8 supplementation. TUNEL and apoptosis-related gene expressions showed increased jejunal apoptosis of DQ-treated piglets; however, reduced apoptosis was observed in the DQ + P8 group. In addition, the mitochondrial morphology and ferroptosis-related gene expressions indicated elevated jejunal ferroptosis in the DQ-treated piglets, and the DQ + P8 treatment attenuated the ferroptosis. Transcriptome identified various differentially expressed genes (DEGs) between different treatments. KEGG analysis indicated that the DEGs were enriched in the PI3K-AKT, NF-κB, and apoptosis pathways. The expressions of key DEGs and key proteins in the PI3K-AKT and NF-κB pathways were further verified. In summary, our results indicate that P8 supplementation ameliorated jejunal oxidative stress, morphological damage, barrier dysfunction, apoptosis, and ferroptosis in the DQ-treated piglets. Summary: Cis‐regulatory elements (CREs) are integral to the spatiotemporal and quantitative expression dynamics of target genes, thus directly influencing phenotypic variation and evolution. However, many of these CREs become highly susceptible to transcriptional silencing when in a transgenic state, particularly when organised as tandem repeats. We investigated the mechanism of this phenomenon and found that three of the six selected flower‐specific CREs were prone to transcriptional silencing when in a transgenic context. We determined that this silencing was caused by the ectopic expression of non‐coding RNAs (ncRNAs), which were processed into 24‐nt small interfering RNAs (siRNAs) that drove RNA‐directed DNA methylation (RdDM). Detailed analyses revealed that aberrant ncRNA transcription within the AGAMOUS enhancer (AGe) in a transgenic context was significantly enhanced by an adjacent CaMV35S enhancer (35Se). This particular enhancer is known to mis‐activate the regulatory activities of various CREs, including the AGe. Furthermore, an insertion of 35Se approximately 3.5 kb upstream of the AGe in its genomic locus also resulted in the ectopic induction of ncRNA/siRNA production and de novo methylation specifically in the AGe, but not other regions, as well as the production of mutant flowers. This confirmed that interactions between the 35Se and AGe can induce RdDM activity in both genomic and transgenic states. These findings highlight a novel epigenetic role for CRE–CRE interactions in plants, shedding light on the underlying forces driving hypermethylation in transgenes, duplicate genes/enhancers, and repetitive transposons, in which interactions between CREs are inevitable. Insect silks possess excellent biodegradability, biocompatibility and mechanical properties, and have numerous applications in biomedicine and tissue engineering. However, the application of silk fiber is hindered by its limited supply, especially from non‐domesticated insects. In the present study, the silk yield and organ size of Bombyx mori were significantly improved through genetic manipulation of the target of rapamycin complex 1 (TORC1) pathway components. Silk protein synthesis and silk gland size were decreased following rapamycin treatment, inhibiting the TORC1 signaling pathway both in vivo and ex vivo. The overexpression of posterior silk gland‐specific Rheb and BmSLC7A5 improved silk protein synthesis and silk gland size by activating the TORC1 signaling pathway. Silk yield in BmSLC7A5‐overexpression silkworms was significantly increased by approximately 25%. We have demonstrated that the TORC1 signaling pathway is involved in the transcription and translation of silk genes and transcriptional activators via phosphorylation of p70 S6 kinase 1 and 4E‐binding protein 1. Simple Summary: Bacterial enteritis caused by Salmonella enteritidis infection can reduce the growth performance and increase the mortality of broilers. Antibiotics were the mainstay of treatment for intestinal injury caused by Salmonella enteritidis, but the overuse of antibiotics has resulted in severe drug residues and the emergence of antibiotic-resistant bacteria. Chinese herbs contain natural compounds with high safety and medicinal value, making them good substitutes for antibiotics. Compared with use of a single Chinese herb, the rational combination of multiple herbs could produce synergistic effects to enhance therapeutic outcomes. In this study, the specific compound Chinese herbal medicine (CCHM) consisted of Pulsatilla, Coptis chinensis, Fraxini cortex, Cortex phellodendri, Folium isatidis, Raddix paeoniae alba, Schisandra chinensis, and Fructus mume in a ratio of 15:6:6:6:6:6:6:5. Our results showed that CCHM improved the growth performance, intestinal barrier function, immune response, and cecal microflora of broilers infected with Salmonella enteritidis. This study investigated the effects of CCHM in drinking water on broilers infected with Salmonella enteritidis. One-day-old male Cobb 500 broilers (n = 300) were randomly assigned to five groups: a control (NC) group, a Salmonella enteritidis challenge (SE) group, an antibiotic (AB) group, a low dose of CCHM (CL) group, and a high dose of CCHM (CH) group. Each group had six replicate cages with ten broilers per cage. The broilers in the NC and SE groups were given normal drinking water. From days 12 to 18, the AB group received water treated with ciprofloxacin lactate injection (1 mL/L), while the CL and CH groups received water containing CCHM at doses of 5 mL/L and 10 mL/L, respectively. Broilers in all groups except the NC group were orally given Salmonella enteritidis daily from days 9 to 11. The experimental period was 28 days. The results showed that, compared with the SE group, the CL and CH groups showed improved growth performance; increased immune organ indices, expressions of ileal occludin and ZO-1 proteins, jejunal and ileal villus heights (except at day 19), and cecal Lactobacillus counts on days 19 and 28 (p < 0.05); and decreased jejunal and ileal lesion scores, ileal interleukin 1β (IL-1β) (except at day 19), interferon-γ (IFN-γ), interleukin 6 (IL-6) (except at day 19), secretory immunoglobulin A (slgA) and tumor necrosis factor α (TNF-α) (except at day 19) levels, serum D-lactic acid and diamine oxidase (DAO) (except at day 19) contents, jejunal and ileal crypt depths (except at day 19), and cecal Salmonella and Escherichia coli counts on days 19 and 28 (p < 0.05). On day 28, except for the levels of ileal interleukin 10 (IL-10), TNF-α, slgA, and serum D-lactic acid content, there were no differences among the NC, AB, and CL groups (p > 0.05). In conclusion, drinking water supplemented with CCHM alleviated the intestinal damage caused by Salmonella enteritidis infection and improved growth performance and cecal microbiota in broilers. [Purpose/significance] Through the analysis of the conference information in the field of library and information science, this research analyses the current situation and distribution characteristics of academic conferences in the field of library and information science, and reveal the development of LIS. [Method/process] This research discovers the development of the conference by collecting the information of the “meetings of LIS” from August 31, 2019 to August 31, 2020, the basic information and the communication of the conference. Used quantitative research methods to analyze the meeting time, location, organizer, etc.; explored the difference in attention between domestic and foreign and different platform meetings; conducted text mining on the theme and background of the meeting to reveal features of academic conferences in the new era. [Result/conclusion] The research found that offline academic conferences for LIS are mainly held on weekends, and there is no fixed time for online conferences. The venues are mainly concentrated on universities or cities with rich resources in the subject of LIS, such as Wuhan, Beijing and Shanghai. Scholars pay more attention on foreign conferences than domestic conferences. The conference was held around the innovation, wisdom, integration and openness, and the content of meetings has become increasingly rich.

Previous studies on the early interference of gut microbiota by Bacillus siamensis (B. siamensis) in weaned piglets are rarely reported, and the present trial is a preliminary study. This experiment was conducted to investigate the effects of B. siamensis supplementation on the growth performance, serum biochemistry, immune response, fecal short-chain fatty acids and microbiota of weaned piglets. Sixty weaned piglets were randomly divided into a control group (CON) and a B. siamensis group (BS), which were fed a basal diet and the basal diet supplemented with 5 × 1010 CFU B. siamensis per kg, respectively. Each group had 3 replicates and 10 piglets per replicate. The trial lasted for 28 days. The results showed that B. siamensis significantly increased the serum growth hormone (GH) and insulin-like growth factor (IGF) in piglets. Compared with the CON group, the levels of serum immunoglobulin and inflammatory factors in the BS group were significantly improved. In addition, the serum concentrations of zonulin and endotoxin (ET) in the BS group were lower. The dietary addition of B. siamensis significantly increased fecal short-chain fatty acid (SCFA) levels in piglets. Notably, B. siamensis improved the microbial composition by increasing beneficial genera, including Weissella, Lachnospiraceae_NK4A136_group and Bifidobacterium, and decreasing pathogenic genera, including Pantoea, Fusobacterium and Gemella, in piglet feces. Correlation analysis showed that the benefits of dietary B. siamensis supplementation were closely related to its improved microbial composition. In summary, the addition of B. siamensis can improve the immunity function, inflammatory response, gut permeability and SCFA levels of weaned piglets, which may be achieved through the improvement of their microbiota.
