The hematophagous mite, Dermanyssus gallinae, also known as the Poultry Red Mite (PRM) is the major pest for the egg-laying industry worldwide. It is associated with severe economic loses and means a serious threat for the hen health and welfare. In addition, the mite can feed on humans causing a skin condition known as gamasoidosis. Traditional control methods against the PRM are based on spraying with synthetic acaricides. The lack of effectivity and the development of resistances imply the need for alternative control methods. The present thesis is framed in the One Health approach, focused in the assessment of the zoonotic risk of D. gallinae and in the identification, through omics tools, of vaccine candidates for the alternative control of PRM infestations. Chapter I: General Introduction Chapter I. This chapter provides a general overview of the current research on control methods against PRM infestations. It is also a compilation of the most recent advances in the biology of the mite, the harms to the laying-egg industry and the impact to the environment and human health. A special attention has been paid to the novelties in the vaccine development that has been enhanced with the recent characterization of the mite genome and proteome. D. gallinae was identified by SEM microscopy as the responsible for the dermatitis. In this work the onset of the signs is also associated with a status of severe infestation in the hen house and a close contact with large number of mites while an absence of a clear immune response was observed. In this chapter a vaccinology approach was followed to identify potential vaccines candidates for the control of D. gallinae. Firstly, we characterized the proteome for fed and unfed nymphs and adults. Amongst those proteins that showed different level of expression we selected Calumenin (Deg-CALU) to be evaluated as vaccine candidate. By an on-hen feeding trial where we tested Deg-CALU and Rhm- SUB, as a positive control. The vaccination with Deg-CALU and Rhm-SUB reduced the mite oviposition by a 35 and 44%, respectively. These results supports the consideration of Deg-CALU and Rhm-SUBO as suitable vaccine candidates for the control of PRM infestation [Lima-Barbero, J. Based on the protective effects of the akirin/subolesin vaccines for the control of ectoparasites infestations, the akirin gene for D. gallinae (Deg-akr) was identified and the effectivity of recombinant Deg-AKR vaccines evaluated. An on-hen feeding assay was performed to evaluate the effects on the vaccination with Deg-AKR against PRM infestations. The vaccination induced an effective immune response in the hens that reduced the mite oviposition by a 42% supporting the use of Deg-AKR as a candidate protective antigen aga for the control of PRM population growth. Resumen del trabajo presentado al XIII Congreso de la Sociedad Española para la Conservación y Estudio de los Mamíferos, celebrado en Guadalajara del 6 al 9 de diciembre de 2017., Para poder proceder a la reintroducción de una especie en territorios en los que se ha extinguido, es necesario corregir las causas que provocaron esa extinción. Existe poca información sobre las causas de la extinción del lince ibérico (Lynx pardinus) en Castilla-La Mancha (CLM). En otras áreas de su distribución, el lince se ha visto afectado por procesos infecciosos. El objetivo de este estudio es identificar aquellos agentes infecciosos que circulan en la comunidad de carnívoros simpátricos en CLM. Para ello, se muestrearon las dos áreas destinadas a la reintroducción del lince: Sierra Morena Oriental (SMO) y Montes de Toledo (MT). La monitorización sanitaria consistió en (i) una vigilancia pasiva mediante el muestreo de cadáveres procedentes de atropellos o del control autorizado de depredadores y (ii) una vigilancia activa mediante la captura, muestreo y suelta de individuos vivos. En total, se muestrearon 220 individuos de las especies: zorro (Vulpes vulpes), garduña (Martes foina), gineta (Genetta genetta), turón (Mustela putorius), meloncillo (Herpestes ichneumon), nutria (Lutra lutra), tejón (Meles meles), gato doméstico (Felis silvestris catus) y gato montés (Felis silvestris silvestris). El suero se analizó mediante ELISA para la detección de anticuerpos. Las tasas de prevalencia obtenidas fueron: 70% de positivos para el virus de la leucemia felina (FeLV), 41% coronavirus felino (FCoV), 16% moquillo canino (CDV), 13% Complejo Mycobacterium tuberculosis (CMT), 0% virus de inmunodeficiencia felina (FIV) y 0% parvovirus canino. El gato doméstico y sus híbridos mostraron una elevada seroprevalencia frente a FeLV y FCoV (100%), lo que puede suponer un riesgo para el lince y el gato montés. Respecto a CDV, la especie con mayor seroprevalencia fue el zorro (20%), mientras que en el caso del CMT, causante de la tuberculosis animal, la especie con mayor prevalencia fue el tejón (33%), seguido por el zorro (14%). Este trabajo demuestra la circulación de agentes patógenos, como el FeLV y CMT, entre carnívoros simpátricos del lince ibérico que pueden suponer un riesgo para la especie, por lo que consideramos necesaria una vigilancia sanitaria continuada.

Resumen del trabajo presentado al XXIII Congreso Español de Ornitología, celebrado del 2 al 5 de noviembre 2017 en Badajoz., La “crisis de las vacas locas” causó un escenario muy desfavorable para las aves necrófagas, dando lugar a la recogida del ganado muerto en el campo para reducir el riesgo de transmisión de enfermedades. Al mismo tiempo, debido al colapso de las poblaciones de buitres en Asia y África, Europa y especialmente España tiene el deber de conservar estas especies. Una medida de gran éxito es la creación y el mantenimiento de puntos de alimentación con cadáveres y restos de animales domésticos y cinegéticos. Ante la presencia de tuberculosis bovina (TB) y otras enfermedades en grandes poblaciones de ungulados silvestres, se le concede importancia al movimiento de cadáveres que puedan estar contaminados. Por ello, este trabajo va dirigido a estudiar movimiento de carroñas por las especies necrófagas mediante el uso de cámaras de foto-trampeo. Los datos preliminares indican que en exclusión de mamíferos ambas especies de buitre pueden desplazar restos de carroña (buitre negro) o cadáveres enteros (buitre leonado) a más de 50 metros a favor de pendiente durante el proceso de consumo. Esta información revela la importancia del vallado de los muladares fijos no solo para la exclusión de mamíferos carroñeros.

Resumen del trabajo presentado a la 12th Conference of the European Wildlife Disease Association (EWDA), celebrada en Berlin (Alemania) del 26 al 31 de agosto de 2016., Extensive livestock farming is an important component of the landscape and a primary sector in south-central Spain. ln this type of farms, water and food resources are scarce for a long period along the year, which conduct to wildlife (especially wild boar and red deer) and domestic livestock to share resources. Proper management of these interactions can help to reduce direct and indirect transmission of infectious pathogens at the interface. We visited 53 cattle farms in south-central Spain to assess in situ all the risk points for wildlife-livestock interaction, for which we developed a standardise field protocol to assess biosecurity. We proposed specific biosecurity plans for each farm to reduce interspecific contacts, which are mainly indirect. With the information obtained so far we have been able to conclude that: i) Complex situations where different hosts are present, in addition to the wild, are common. Extensive pig may act as epidemiological driver among wildlife and cattle. ii) ln a not inconsiderable proportion of farms (almost one third), supplementary feeding is provided to wildlife for hunting purposes. iii) Among the measures proposed, apart from wildlife population control, the most frequent recommended practices included ensuring that animals drink together safely in each of the fences and the intelligent management of grazing. iv) The most expensive measures refer to the need for perimeter game fences or livestock fences. Regarding the cost per farm to implement a biosecurity program, provisional data indicate that in 70 % of the costs does not amount to more 100 per cattle. In the near future, the efficacy and practical applicability of these programmes is going to be evaluated by means of different disease models, so as the design of biosecurity protocols for extensive pig and goat.

Resumen del trabajo presentado a la 12th Conference of the European Wildlife Disease Association (EWDA), celebrada en Berlin (Alemania) del 26 al 31 de agosto de 2016., The bouse sparrow (Passer domesticus) is one of the most widely distributed urban birds and has successfully expanded its range to all continents. Recent population declines of this and other garden bird species in central and Northern Europe have raised concern and are considered due to several factors, including pathogens such as the protozoan parasite Trichomonas gallinae. To study the presence of this potential pathogen in Spanish sparrow populations we sampled 76 house sparrows in three areas with different degrees of urbanisation and during both wintering and the reproductive season. Swab samples from the oropharynx and oesopbagus were cultured in trichomonas culture medium enriched with 10% foetal calf serum and tested by direct microscopic observation and PCR for presence of T. gallinae. None of the sparrows was colonised by T. gallinae but two yeasts were detected, Macrorhabdus ornitogaster and Candida sp. in 29% and 23.7% of the cultures, respectively. Candida albicans was identified in most cases by culture in Sabouraud dextrose agar and biochemical tests while Macrorhabdus ornitogaster has characteristic microscopic features. Macrorhabdus ornitogaster was found more frequently in juvenile sparrows (38.89 %, 7 of 18) than adults (25.7 %, 15 of 58; GzLM, p = 0.049, β = -1,304, SE = 0.663). Macrorhabdus prevalence also increases as degree of urbanisation raises (GzLM, p = 0.018, β = 0.906, SE = 0.383). A co-infection effect with Candida sp.has been observed (GzLM, p = 0.034, β = 0.043, SE = 0.383). M. ornithogaster is an ascomycetes yeast tbat is present at tbe junction of the proventriculos and gizzard of wild and domestic birds. It has been described in numerous captive and wild birds, but to our knowledge not previously in house sparrows. Although the sparrows were apparently healthy and M ornithogaster is potentially a commensal, in some passerine, gallinaceous and psittacine species it has been identified as a significant pathogen. The degree of M ornithogaster carriage in sparrows may be affected by environmental conditions and they may act as reservoirs for other more susceptible species. Urban birds have to cope with many stressors that could affect the presence of this commensal yeast. Finally, under stressful conditions fitness of infected individuals could be comprornised and lead to clinical infections.

Resumen del trabajo presentado a la 12th Conference of the European Wildlife Disease Association (EWDA), celebrada en Berlin (Alemania) del 26 al 31 de agosto de 2016., The decline of red kite (Milvus milvus) populations across Southem and Central Europe has been attributed mostly to pesticide and rodenticide exposure, changes in landuse, carrion, rubbish availability, direct persecution and pathogens. In this context, feeding si tes for resident or wintering carrion eaters are of high value but do also create conflict, as the aggregation of individuals at these feeding sites and the exposure to dead livestock could promote pathogen transmission. Capture events of wild birds for tagging purposes are unique opportunities to carry out targeted disease and bealth surveillance. We used samples from wintering red kites captured at the Binaced feeding station in three different migration stages (auturnn, wintering and spring migration) during three wintering seasons: 2013- 2014 (n =52), 2014-2015 (n =59) and 2015-2016 (n = 69) to explore exposure of these birds to different avian and shared pathogens. No antibodies against West Ni1e Virus were found until the 2015/2016 wintering season (89.6 %, 60 of 67). Meanwbile a decreasing prevalence of antibodies against Avian Influenza Virus (AIV) pattem has been observed in the last wintering season (x2 = 6.904, p = 0.032, df = 2; 2013 - 2014 = 10.4 %, 5 of 48; 2014 -2015 = 8.8%, 5 of 57 and 2015-2016 =0%, 0O of 67). [Objectives]: The objective of this study was to analyse the prevalence of tracheal carriage of Staphylococcus aureus/MRSA in storks and to study the resistance and virulence genes in the obtained isolates. [Methods]: Tracheal samples from 92 stork nestlings of two landfill-associated and two natural-habitat colonies were inoculated in specific media for S. aureus and MRSA recovery. Antimicrobial susceptibility was tested, and the presence of resistance, virulence and immune evasion cluster (IEC) genes was analysed by PCR. S. aureus isolates were characterized by spa and agr typing. Staphylococcal cassette chromosome (SCC) mec type was determined for mecC-positive isolates, and MLST was performed for 17 selected S. aureus isolates. [Results]: S. aureus isolateswere identified in 32/92 samples (34.8%), and 38 isolateswere recovered. The prevalence of S. aureus was higher in nestlings from landfills (24/43, 55.8%) than in those fromnatural habitats (8/49, 16.3%). Three birds from landfill-associated colonies carried MRSA, two with mecA-positive strains [clonal complex (CC) 5-spa-t002 and CC398-spa-t011] and one with a mecC-positive strain [sequence type (ST) 3061-CC130-spat843- agr-III-SCCmecXI). None of the MRSA isolates presented IEC genes. Thirty-five MSSA isolates, which showed 18 different spa types (ascribed to CC5, CC7, CC22, CC30, CC45, CC59, CC133 and CC398), were obtained. The agr types detected were I (63%), II (29%) and III (8%). Resistance and virulence genes identified in MSSA were blaZ (n=25), erm(T) (n=9), erm(A) (n=1), tet(M) (n=2), fexA (n=3), str (n=2), tst (n=2), eta (n=1) and cna (n=15). The IEC types B, C, D and G were found in MSSA isolates, and two new STs were identified (ST3060 and ST3061). [Conclusions]: White storks are frequently tracheal carriers of S. aureus, including ST398 isolates. Resumen del trabajo presentado a las V Jornadas Doctorales de la Universidad de Castilla-La Mancha, celebradas en Ciudad Real (España) el 6 de octubre de 2015., Enfermedades infecciosas en aves silvestres ganan cada vez más en importancia. Tanto aquellas compartidas entre fauna silvestre, animales domésticos y/o el hombre para los que las aves pueden actuar como el reservorio clave (p.ej. Influenza aviar IA o fiebre del Nilo FNO) como las de importancia para la conservación. El milano real es una rapaz amenazada con reducciones preocupantes de sus poblaciones en el pasado reciente sin que se haya podido determinar una causa evidente para estas pérdidas. En España existe tanto una población reproductora una parte de la cual migra en invierno a África, mientras los ejemplares que se reproducen en el centro y Norte de Europa vienen a pasar el invierno a España. Los pasos de migración y sobre todo las áreas de parada y descanso son lugares de agregación y potencial intercambio de patógenos. Durante capturas para el anillamiento científico y anillamiento con anillas de lectura a distancia, realizados durante el paso migratorio y la invernada en 2013-2015, se obtuvieron muestras de 111 milanos reales para determinar su exposición a patógenos aviares de interés sanitario. La ausencia de genoma de Flavivirus y anticuerpos frente a Flavivirus permite deducir que los milanos reales no son de importancia en la epidemiología de los Flaviviridae. Sin embargo un 9,9% y un 9% de los milanos habían estado expuestos a virus de influenza aviar y paramixovirus aviar-1 (virus de la enfermedad de Newcastle). Solo uno de los milanos capturados en 2013 y dos de los milanos capturados en 2015 excretaban en el momento de la captura Salmonella spp. Entre los Escherichia coli que suelen formar parte de la flora intestinal de estas rapaces y se aislarón en el 84,7% un 10,8% mostraban un patrón fenotípico de multiresistencia a los antimicrobiales. Dado que los milanos se alimentan en un muladar cadáveres de pollos de granja, será de interés determinar si los E. coli multiresistentes coinciden con los que se puedan hallar en los cadáveres proporcionados o si forman parte de la flora que estas aves traen de sus lugares de procedencia.

Resumen del póster presentado a la 10th European Vertebrate Pest Management Conference, celebrada en Sevilla (España) del 21 al 25 de septiembre de 2015., Some passerine and columbiform species are considered nuisance on livestock farms including poultry, and can carry pathogenic viruses and bacteria. Newcastle Disease (ND) due to pathogenic strains of aPMV-1 is a worldwide distributed viral disease that causes serious economic losses in poultry. High biosecurity and vaccination are preventive measures in industrial poultry production. These measures are not applied in gamebird farms where contact between wild and farmed birds is frequent. To study the risk of aPMV-1 transmission by sympatric species to farmed red-legged partridges in central Spain, we used a commercial competition ELISA kit to detect antibodies against aPMV-1 in sera from wild birds (n=177), including rock pigeon (Columba livia), wood pigeon (Columba palumbus), eurasian collared dove (Streptopelia decaocto), eurasian magpie (Pica pica) and song thrush (Turdus philomelos). To test for NDV exposure and the effect of aggregation on farms and release estates on NDV transmission we additionally tested sera from farm-raised partridges (n=385), partridges harvested in hunting estates where partridges are released regularly (n=73) and from partridges hunted or captured in “release-free” hunting estates (n=93). High prevalence was found in sympatric birds, especially in rock pigeons (64.5 %, n= 30) and passerine birds (56.2 %, n= 73). Resumen del trabajo presentado al XII Congreso de la Sociedad Española para la Conservación y Estudio de los Mamíferos, celebrado en Burgos del 4 al 7 de diciembre de 2015., La ganadería extensiva es un importante componente del paisaje y el sector primario del centro- sur español. En este tipo de explotaciones ganaderas donde los recursos hídricos y el alimento escasean durante gran parte del año, es habitual encontrar a la fauna silvestre (especialmente el jabalí y el ciervo) y el ganado doméstico compartiendo los mismos recursos. Una adecuada gestión de estas interacciones puede contribuir a reducir la transmisión (directa o indirecta) de enfermedades infecciosas en sus zonas de contacto, y así mejorar la relación entre la fauna silvestre y el ser humano. Por ello, se han visitado 38 explotaciones de ganadería bovina en las provincias de Ciudad Real, Córdoba, Sevilla, Huelva, Cádiz y Extremadura. En las cuales se ha realizado una encuesta pormenorizada con los ganaderos para conocer todos los factores de riesgo asociados a su gestión e historia, así como un análisis in situ de los puntos de riesgo y las instalaciones de la explotación, para diseñar protocolos de bioseguridad que ayuden a controlar la transmisión de la tuberculosis animal entre el ganado doméstico y la fauna silvestre. Las ganaderías de estudio se encuentran situadas en paisajes parcheados, donde las fincas cinegéticas y ganaderas se intercalan, siendo la caza mayor un aprovechamiento secundario común. Poseen de media 617 Has y 205 cabezas de ganado bovino, y el 70% albergan cerdo en extensivo, un potencial reservorio de la tuberculosis bovina. La unidad de gestión habitual es la parcela de pasto vallada para el ganado pero no para la fauna silvestre. En estos ambientes semi-áridos el agua se suministra a través de bebederos, charcas, manantiales o arroyos temporales. Es en estos puntos de agua donde la bacteria (complejo Mycobacterium tuberculosis) sobrevive, y donde se pueden producir la mayor parte de las interacciones indirectas que conlleven una potencial transmisión de la enfermedad. Existen grandes diferencias en el tipo y nivel de riesgo de cada ganadería, evidenciando la necesidad de diseñar programas específicos de bioseguridad, con la colaboración y feedback por parte de los ganaderos y una adecuada monitorización de su efectividad.

Resumen del póster presentado al XIX Congreso de la Sociedad Española de Enfermedades Infecciosas y Microbiología Clínica, celebrado en Sevilla del 28 al 30 de mayo de 2015., [Introducción y objetivos]: Los enterococos resistentes a vancomicina (ERV) pueden presentar mecanismos de resistencia adquirida (ERV‑a), o bien mecanismos intrínsecos de resistencia (ERV‑i) asociados a las especies E. gallinarum y E. casseliflavus (portadores de los genes vanC‑1 y vanC‑2, respectivamente). El objetivo de este estudio fue evaluar la presencia de ERV en muestras cloacales de aves silvestres en su medio natural (cigüeña blanca‑Ciconia ciconia) y en producción semi‑extensiva (perdiz roja‑Alectoris rufa) y analizar los fenotipos y genotipos de resistencia de las cepas obtenidas. [Material y métodos]: Muestras cloacales fueron obtenidas en junio‑2014 de 62 pollos de cigüeña blanca que vivían en 4 colonias distintas durante manejos de anillamiento, y en julio/agosto‑2014 de 127 perdices rojas en 5 granjas cinegéticas, tanto reproductores como pollos nacidos en el año. Estas muestras fueron inoculadas en solución salina y sembradas en placas de Slanetz–Bartley con 4 μg/mL de vancomicina (SB‑van). De cada placa se aislaron dos colonias sugestivas de enterococo/muestra y se mantuvieron las dos cuando presentaron distinta especie o diferente fenotipo de resistencia. Para su identificación se realizaron pruebas bioquímicas y moleculares (PCRs específicas). Se determinó la CMI a vancomicina (VAN) y teicoplanina (TEI) por dilución en agar y la sensibilidad a 10 antibióticos por antibiograma. Se estudió, mediante PCR, el genotipo de resistencia a vancomicina (vanA, vanB, vanC‑1, vanC‑2/3, vanD, vanE o vanG) y la presencia de los genes de resistencia erm(A), erm(B), erm(C), tet(M), tet(K), tet(L), aph(3′)‑IIIa, aac(6′)‑aph(2″), ant(6)‑Ia, drfF, dfrG y dfrK. [Resultados]: Se aislaron enterococos en placas SB‑van (potenciales ERV) en 38 de las 62 muestras de cigüeñas blancas analizadas (61,3%). La prevalencia en tres de las colonias fue similar (entre 34,5 y 38,4%) mientras en una colonia fue del 55,1%. En 2 muestras las 2 cepas pertenecieron a distinta especie estudiándose un total de 40 cepas diferentes, 37 de ellas fueron E. gallinarum y 3 E. casseliflavus (todos ERV‑i). no se detectó ningún aislado ERV‑a. Se detectó ERV‑i en 10 de las 127 muestras de perdices (7,9%) procedentes de 4 de las 5 granjas muestreadas. La prevalencia en reproductores fue del 6% y en pollos del 21%. Se estudiaron un total de 10 cepas, todas ellas E. gallinarum. Los rangos de CMIs para VAN y TEI en las 50 cepas fueron 4‑16 μg/mL y 0,25‑2 μg/mL respectivamente. Se detectó resistencia a los siguientes antibióticos: tetraciclina (48%), eritromicina (31%), gentamicina (2%), estreptomicina (12%), kanamicina (12%), trimetoprim‑sulfametoxazol (2%), ampicilina (2%) y ciprofloxacina (13%). Se identificaron los siguientes genes de resistencia: erm(A) (8%), erm(B) (21%), tet(M)(46%), tet(K) (4%), tet(L) (8%), aph(3′)‑IIIa (12%), aac(6′)‑aph(2″) (2%), ant(6)‑Ia (12%), drfF(2%), y dfrG(2%). [Conclusiones]: Se detectó una alta prevalencia de ERV‑i (especialmente E. gallinarum) en las muestras fecales de cigüeñas blancas (61,3%), siendo mucho menor en perdices (7,9%). Sin embargo, no se detectó ERV‑a en ninguna de estas especies animales. Los aislados de E. gallinarum presentan una gran diversidad de genes de resistencia a antibióticos.

Resumen del trabajo presentado al XIX Congreso de la Sociedad Española de Enfermedades Infecciosas y Microbiología Clínica, celebrado en Sevilla del 28 al 30 de mayo de 2015., [Introducción]: La resistencia a meticilina en Staphylococcus aureus (SARM) mediada por los genes mecA y mecC ha adquirido gran interés en los últimos años no sólo por su repercusión en salud humana sino también por la detección en ciertas líneas genéticas asociadas a animales. [Objetivos]: Determinar la prevalencia de S. aureus (SA) en muestras traqueales de cigüeña sana y realizar la caracterización genética de los aislados obtenidos. [Material y métodos]: Se procesaron torundas traqueales de 92 cigüeñas blancas pertenecientes a cuatro colonias ubicadas en diferentes zonas de la provincia de Ciudad Real en junio-2014. Las muestras fueron pre-enriquecidas en BHI-caldo con 6,5% NaCl, y sembradas posteriormente en placas de Manitol-Salt-Agar y Oxacillin-Resistance-Screening-Agar-Base. Los aislados se identificaron con pruebas microbiológicas y moleculares (PCR gen nuc). El fenotipo de resistencia a 16 antimicrobianos se determinó por difusión-disco. La presencia de los genes de resistencia: mecA, mecC, blaZ, blaZ-SCCmecXI, lnu(A), lnu(B), lnu(C), lnu(D), vga(A), vga(C), vga(E), lsa(B), erm(A), erm(B), erm(C), erm(T), erm(F), msr(A), msr(B), mph(C), tet(K), tet(L), tet(M), tet(O), mup(A), str, ant(6)-Ia, ant(3>)(9), fex(A), cfr, catpC194, catpC221, catpC223, fusB, fusC se estudió por PCR. Los aislados SA fueron tipados mediante SCCmec, spa y agr, y, en aquellas cepas con características especiales o con tipos spa más prevalentes, se realizó también MLST para determinar la secuencia tipo (ST). Las cepas fueron asociadas a un complejo clonal (CC) según los resultados de ST y/o tipo-spa detectados. Los genes de virulencia (lukF/lukS, tst, eta, etb, etd, etd2, cna) fueron analizados por PCR. [Resultados]: Se detectó SA en 33/92 muestras (36%) y se caracterizó de 1-3 aislados diferentes/muestra. De los 39 SA estudiados, tres fueron SARM (8%), uno mecC positivo y dos mecA positivos, y pertenecían a las líneas genéticas CC130-spa-t843-agrIII (SCCmecXI), CC398-spa-t011-agrI y CC5-spa-t002-agrII. Respecto a las cepas sensibles a meticilina (SASM), las líneas detectadas fueron (número de cepas/spa-tipo): CC5 (11/t045, t126, t209, t688, t774, t1818), CC398 (9/t571, t3625, t6606), CC133 (6/t1166, t6384), CC30 (3/t012, t1945), CC7 (2/ t091), CC45 (2/t015), CC22 (1/t005), CC59 (1/t216), y CC no determinado (1/t4445). Los agr-tipo detectados en las cepas SASM fueron: agrI (64%), agrII (28%), y agrIII (8%). Un nuevo spa-tipo y 3 nuevas ST fueron descritos en este estudio. Se observó resistencia a [número de cepas/genes]: penicilina [26/blaZ, blaZ-SCCmecXI, mecA], eritromicina-clindamicina inducible [10/erm(T), erm(A)], tetraciclina [3/tet(M), tet(K)], cloranfenicol [3/fex(A)], estreptomicina [2/str], ácido fusídico [2], ciprofloxacina [1], y mupirocina [1]. Los genes de virulencia detectados fueron los siguientes (número de cepas): tst (2), eta (1), etd2 (1), cna (16). [Conclusiones]: Se detecta una elevada prevalencia de S. aureus en cigüeñas, así como una gran diversidad de líneas genéticas. Destaca la detección del gen mecC por primera vez en cigüeña y la presencia de cepas SARM y SASM de la línea genética CC398. En un futuro deberá determinarse la adquisición de estas cepas y el papel de estas aves migratorias en la transferencia de estos microorganismos en entornos naturales.

Trabajo Fin de Máster: Máster universitario en Investigación Básica y Aplicada en Recursos Cinegéticos. Tema: Enfermedades de las aves silvestres., En este estudio se pretende describir y entender la epidemiología de Paramyxovirus aviar (aPMV-1) en diferentes especies de aves a nivel nacional en la última década. Se analizaron 1194 sueros recogidos entre 2005 y 2014 en toda España pero principalmente en Castilla – La Mancha, de nueve especies diferentes: perdiz roja (Alectoris rufa), faisán común (Phaisanus colchicus), gaviota patiamarilla (Larus cachinans), paloma bravía (Columba livia), paloma torcaz (Columba palumbus), tórtola turca (Streptopelia decaocto), urraca (Pica pica), zorzal común (Turdus philomelos) y cigüeña blanca (Ciconia ciconia) mediante ELISA para la detección de anticuerpos frente al aPMV-1. La seroprevalencia media de aPMV-1 era de 15.7%, identificándose algunas especies como la paloma bravía y los Paseriformes con prevalencias significativamente más altas que el resto. Se observó una tendencia temporal a reducirse la seroprevalencia a lo largo de los años y un marcado efecto estacional, siendo invierno y otoño las estaciones con mayores seroprevalencias, posiblemente ligado a una mayor presencia y agregación de aves invernantes, por ejemplo columbiformes migratorias. Dado el aprovechamiento cinegético y manejo intensivo de la perdiz roja se evaluaron factores relacionados con la exposición de la misma a aPMV-. La seroprevalencia de aPMV-1 en perdices provenientes de cotos en los que solo existen poblaciones naturales es significativamente menor que en perdices muestreadas en granjas y en cotos en los que se realiza suelta de animales. Por otra parte las seroprevalencias de aPMV-1 muestran la misma variación estacional que las seroprevalencias en aves silvestres en general. Mediante la Inhibición de Hemaglutinación (IHA) se observaron títulos medios y altos de anticuerpos hemaglutinantes en perdices rojas seropositivas. Sin embargo no se detectó genoma viral de aPMV-1 mediante RT-PCR a tiempo real en los hisopos orales y cloacales tomados de las perdices. Estos resultados reflejan la circulación continua de aPMV-1 en España y la exposición de la perdiz roja al virus, además de un posible efecto de su manejo sobre esta exposición.

The poultry red mite (PRM), Dermanyssus gallinae, is a hematophagous ectoparasite considered as the major pest in the egg-laying industry. Its pesticide-based control is only partially successful and requires the development of new control interventions such as vaccines. In this study, we follow a vaccinology approach to identify PRM candidate protective antigens. Based on proteomic data from fed and unfed nymph and adult mites, we selected a novel PRM protein, calumenin (Deg-CALU), which is tested as a vaccine candidate on an on-hen trial. Rhipicephalus microplus Subolesin (Rhm-SUB) was chosen as a positive control. Deg-CALU and Rhm-SUB reduced the mite oviposition by 35 and 44%, respectively. The poultry red mite (PRM), Dermanyssus gallinae (De Geer, 1778), is a worldwide distributed ectoparasite and considered a major pest affecting the laying hen industry in Europe. Based on available information in other ectoparasites, the mite microbiome might participate in several biological processes and the acquisition, maintenance and transmission of pathogens. However, little is known about the role of PRM as a mechanical carrier or a biological vector in the transmission of pathogenic bacteria. Herein, we used a metaproteomics approach to characterize the alphaproteobacteria in the microbiota of PRM, and variations in its profile with ectoparasite development (nymphs vs. adults) and feeding (unfed vs. fed). The results showed that the bacterial community associated with D. gallinae was mainly composed of environmental and commensal bacteria. Putative symbiotic bacteria of the genera Wolbachia, C. Tokpelaia and Sphingomonas were identified, together with potential pathogenic bacteria of the genera Inquilinus, Neorickettsia and Roseomonas. Significant differences in the composition of alphaproteobacterial microbiota were associated with mite development and feeding, suggesting that bacteria have functional implications in metabolic pathways associated with blood feeding. The poultry red mite (PRM), Dermanyssus gallinae, is a hematophagous ectoparasite of birds with worldwide distribution that causes economic losses in the egg-production sector of the poultry industry. Traditional control methods, mainly based on acaricides, have been only partially successful, and new vaccine-based interventions are required for the control of PRM. Vaccination with insect Akirin (AKR) and its homolog in ticks, Subolesin (SUB), have shown protective efficacy for the control of ectoparasite infestations and pathogen infection/transmission. The aim of this study was the identification of the akr gene from D. gallinae (Deg-akr), the production of the recombinant Deg-AKR protein, and evaluation of its efficacy as a vaccine candidate for the control of PRM. The anti-Deg-AKR serum IgY antibodies in hen sera and egg yolk were higher in vaccinated than control animals throughout the experiment. The results demonstrated the efficacy of the vaccination with Deg-AKR for the control of PRM by reducing mite oviposition by 42% following feeding on vaccinated hens. A negative correlation between the levels of serum anti-Deg-AKR IgY and mite oviposition was obtained.
