Abstract

The Large Synoptic Survey Telescope (LSST) is a large-aperture, wide-field, ground-based survey system that will image the sky in six optical bands from 320 to 1050 nm, uniformly covering approximately $18,000$deg$^2$ of the sky over 800 times. The LSST is currently under construction on Cerro Pach\'on in Chile, and expected to enter operations in 2022. Once operational, the LSST will explore a wide range of astrophysical questions, from discovering "killer" asteroids to examining the nature of Dark Energy. The LSST will generate on average 15 TB of data per night, and will require a comprehensive Data Management system to reduce the raw data to scientifically useful catalogs and images with minimum human intervention. These reductions will result in a real-time alert stream, and eleven data releases over the 10-year duration of LSST operations. To enable this processing, the LSST project is developing a new, general-purpose, high-performance, scalable, well documented, open source data processing software stack for O/IR surveys. Abstract

Background: Real-world data on non-vitamin K oral anticoagulants (NOACs) are essential in determining whether evidence from randomised controlled clinical trials translate into meaningful clinical benefits for patients in everyday practice. RIVER (RIVaroxaban Evaluation in Real life setting) is an ongoing international, prospective registry of patients with newly diagnosed non-valvular atrial fibrillation (NVAF) and at least one investigator-determined risk factor for stroke who received rivaroxaban as an initial treatment for the prevention of thromboembolic stroke. The aim of this paper is to describe the design of the RIVER registry and baseline characteristics of patients with newly diagnosed NVAF who received rivaroxaban as an initial treatment. Methods and results: Between January 2014 and June 2017, RIVER investigators recruited 5072 patients at 309 centres in 17 countries. The aim was to enroll consecutive patients at sites where rivaroxaban was already routinely prescribed for stroke prevention. Each patient is being followed up prospectively for a minimum of 2-years. The registry will capture data on the rate and nature of all thromboembolic events (stroke / systemic embolism), bleeding complications, all-cause mortality and other major cardiovascular events as they occur. Data quality is assured through a combination of remote electronic monitoring and onsite monitoring (including source data verification in 10% of cases). Patients were mostly enrolled by cardiologists (n = 3776, 74.6%), by internal medicine specialists 14.2% (n = 718) and by primary care/general practice physicians 8.2% (n = 417). The mean (SD) age of the population was 69.5 (11.0) years, 44.3% were women. Mean (SD) CHADS2 score was 1.9 (1.2) and CHA2DS2-VASc scores was 3.2 (1.6). Almost all patients (98.5%) were prescribed with once daily dose of rivaroxaban, most commonly 20 mg (76.5%) and 15 mg (20.0%) as their initial treatment; 17.9% of patients received concomitant antiplatelet therapy. Most patients enrolled in RIVER met the recommended threshold for AC therapy (86.6% for 2012 ESC Guidelines, and 79.8% of patients according to 2016 ESC Guidelines). Conclusions: The RIVER prospective registry will expand our knowledge of how rivaroxaban is prescribed in everyday practice and whether evidence from clinical trials can be translated to the broader cross-section of patients in the real world. Abstract

Background: Real-world data on non-vitamin K oral anticoagulants (NOACs) are essential in determining whether evidence from randomised controlled clinical trials translate into meaningful clinical benefits for patients in everyday practice. RIVER (RIVaroxaban Evaluation in Real life setting) is an ongoing international, prospective registry of patients with newly diagnosed non-valvular atrial fibrillation (NVAF) and at least one investigator-determined risk factor for stroke who received rivaroxaban as an initial treatment for the prevention of thromboembolic stroke. The aim of this paper is to describe the design of the RIVER registry and baseline characteristics of patients with newly diagnosed NVAF who received rivaroxaban as an initial treatment. Methods and results: Between January 2014 and June 2017, RIVER investigators recruited 5072 patients at 309 centres in 17 countries. The aim was to enroll consecutive patients at sites where rivaroxaban was already routinely prescribed for stroke prevention. Each patient is being followed up prospectively for a minimum of 2-years. The registry will capture data on the rate and nature of all thromboembolic events (stroke / systemic embolism), bleeding complications, all-cause mortality and other major cardiovascular events as they occur. Data quality is assured through a combination of remote electronic monitoring and onsite monitoring (including source data verification in 10% of cases). Patients were mostly enrolled by cardiologists (n = 3776, 74.6%), by internal medicine specialists 14.2% (n = 718) and by primary care/general practice physicians 8.2% (n = 417). Abstract

The Large Synoptic Survey Telescope (LSST) will be an 8.4m optical survey telescope sited in Chile and capable of imaging the entire sky twice a week. The data rate of approximately 15TB per night and the requirements to both issue alerts on transient sources within 60 seconds of observing and create annual data releases means that automated data management systems and data processing pipelines are a key deliverable of the LSST construction project. The LSST data management software has been in development since 2004 and is based on a C++ core with a Python control layer. The software consists of nearly a quarter of a million lines of code covering the system from fundamental WCS and table libraries to pipeline environments and distributed process execution. The Astropy project began in 2011 as an attempt to bring together disparate open source Python projects and build a core standard infrastructure that can be used and built upon by the astronomy community. This project has been phenomenally successful in the years since it has begun and has grown to be the de facto standard for Python software in astronomy. Astropy brings with it considerable expectations from the community on how astronomy Python software should be developed and it is clear that by the time LSST is fully operational in the 2020s many of the prospective users of the LSST software stack will expect it to be fully interoperable with Astropy. In this paper we describe the overlap between the LSST science pipeline software and Astropy software and investigate areas where the LSST software provides new functionality. We also discuss the possibilities of re-engineering the LSST science pipeline software to build upon Astropy, including the option of contributing affliated packages.

Abstract

Arsenic is a toxic metalloid which is widely distributed in nature. It is normally present as arsenate under oxic conditions while arsenite is predominant under reducing condition. The major discharges of arsenic in the environment are mainly due to natural sources such as aquifers and anthropogenic sources. It is known that arsenite salts are more toxic than arsenate as it binds with vicinal thiols in pyruvate dehydrogenase while arsenate inhibits the oxidative phosphorylation process. The common mechanisms for arsenic detoxification are uptaken by phosphate transporters, aquaglyceroporins, and active extrusion system and reduced by arsenate reductases via dissimilatory reduction mechanism. Some species of autotrophic and heterotrophic microorganisms use arsenic oxyanions for their regeneration of energy. Certain species of microorganisms are able to use arsenate as their nutrient in respiratory process. Detoxification operons are a common form of arsenic resistance in microorganisms. Hence, the use of bioremediation could be an effective and economic way to reduce this pollutant from the environment.

Abstract

The Large Synoptic Survey Telescope (LSST; http://lsst.org) is a planned, large-aperture, wide-field, ground-based telescope that will survey half the sky every few nights in six optical bands from 320 to 1050 nm. It will explore a wide range of astrophysical questions, from studies of the Solar System, to examining the nature of dark energy. LSST is an integrated survey system. The observatory, telescope, camera and data management systems will be built to conduct the LSST survey and will not support 'PI mode' in the classical sense. Instead, the ultimate, science-enabling, deliverable of LSST will be the fully reduced data. This poster presents the baseline design and contents of LSST data products. There will be three main categories. "Level 1" data products will be generated continuously every observing night and include measurements such as alerts to objects that have changed brightness or position. They will be broadcast world-wide using VO protocols. "Level 2" data products will be made available as annual Data Releases and will include images and measurements of quantities such as positions, fluxes, and shapes, as well as variability information such as orbital parameters for moving objects and an appropriate compact description of light curves. The exact contents of Level 2 products will be set by the desire to minimize the necessity to independently reprocess the image data. Finally, approximately 10% of LSST's computing capability will be made available to the community for generation of "Level 3" data products. These will be used to perform custom analyses not fully enabled by Level 1/2, while taking advantage of co-location of computation with the entire LSST data set.

Abstract

A new water-soluble ionic polyacetylene was synthesized by the acid-catalyzed polymerization of 2-ethynylpyridine using acrylic acid. The polymerization proceeded well to give a high yield of polymer. The acetylenic C = C bond stretching (2110 cm-1) and acetylenic = C-H bond stretching (3293 cm-1) peaks of 2-ethynylpyridine monomer were not seen in the FT-IR spectrum of polymer. The electro-optical and electrical properties of the ionic polyacetylene were measured and discussed. The resulting polymer showed strong maximum at 513 nm in the UV-Visible absorption, which is corresponding to photon energy of 2.42 eV. The energy gap width of polymer was estimated to be of 2.08 eV. It was observed that the electrochemical process of polymer is reproducible in the potential range of -1.80 to 1.60 V vs Ag/AgNO3. Abstract

The plasma spray technique was employed to produce an anode layer for solid oxide fuel cells (SOFCs) using calcined micro-sized nickel oxide-8 mol% yttria-stabilized zirconia (NiO-8YSZ) cermet particles as stock powder. The nanostructure of calcined micro-sized NiO-8YSZ cermet particles (CMCEP) generated through the sintering process is closely related to SOFC performance and varied with sintering conditions. Thus, the present study was conducted to analyze the effect of the sintering temperature on the diffusion and nanostructure of particles during the production of CMCEP for SOFC anode material. The distributions of Ni and Zr in grain boundary caused by molecular diffusion were quantified at different sintering temperatures in the range 1,100 °C-1,300 °C. The effects of sintering on nanosurface and structural properties of CMCEP were analyzed through scanning electron and back-scattered electron microscopies and specific surface area. The crystallinity of CMCEP was found to increase with increasing sintering temperature. Smoothing, neck formation, shrinkage, and densification of nano-sized NiO-8YSZ composite granules occurred due to the acceleration of molecular diffusion at sintering temperatures above 1,200 °C, resulting in distinct grain boundary formation. The width of interfacial transition gap between Ni and Zr measured with respect to the grain boundary increased by 69.1% as molecular diffusion accelerated with increasing sintering temperature from 1,100 to 1,300 °C. The results of the nanostructural analysis of the CMCEP surface showed a decrease in the specific surface area (measured by the Brunauer Emmett Teller (BET) method) with increasing calcination temperature. The results of this study are expected to contribute to performance optimization in the future via the control of the porosity, electrical conductivity, and triple phase boundary generation during SOFC anode layer production using plasma sprays. Abstract

Background: Treatment with sorafenib, although associated with inhibition of tumour growth and angiogenesis in in vivo studies, leads to up-regulation of pERK. The addition of MEK inhibition could potentially abrogate this effect and potentiate anti-tumour activity. This phase I study investigated the maximum tolerated dose (MTD), safety, tolerability, pharmacokinetics (PK) and biomarker correlates of selumetinib combined with sorafenib in patients with advanced hepatocellular carcinoma (HCC). Methods: Patients with Child-Pugh (CP) score ≤7 were treated with 400 mg twice daily of sorafenib with escalating doses of selumetinib in a 3 + 3 study design. The dose-limiting toxicity (DLT) evaluation period was 28 days. PK of selumetinib was determined. Angiogenic effect was evaluated with dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI). Results: Twenty-seven patients of Asian ethnicity were enrolled. The MTD was selumetinib 75 mg daily with sorafenib 400 mg twice daily. DLT included grade 3 transaminitis, diarrhoea and fatigue. Most common treatment-related adverse events at MTD (all grades) were diarrhoea (85%), rash (59%), hypertension (44%), fatigue (30%), anorexia (22%) and hand-foot syndrome (22%). Four patients (15%) had PR and 13 (48%) had SD. PR or SD was observed for ≤6 months in seven patients. The median overall survival was 14.4 months. Selumetinib exposures in combination with sorafenib were comparable to other monotherapy studies. A reduction in permeability-surface area product noted in DCE-MRI with treatment correlated with worse survival outcomes. Conclusion: The MTD of selumetinib was 75 mg daily when combined with sorafenib 400 mg twice a day in CP ≥7 HCC. Abstract

The objective of this study was to determine the expression and transferability of tetracycline and erythromycin resistance among 188 MRSA strains from a Malaysian tertiary hospital. The minimum inhibitory concentrations (MICs) for oxacillin, erythromycin, tetracycline and ciprofloxacin ranged from 4 to 512 μg/ml, 0.25 to 256 μg/ml, 0.5 to 256 μg/ml and 0.5 to 512 μg/ml, respectively. Tetracycline-resistant strains showed co-resistance towards ciprofloxacin and erythromycin. There was a significant increase (P<0.05) of high-level tetracycline (≥MIC 256 μg/ml) and erythromycin (≥MIC 128 μg/ml) resistant strains in between the years 2003 and 2008. All erythromycin-resistant strains harboured ermA or ermC gene and all tetracycline-resistant strains harboured tetM or tetK gene. The blaZ was detected in all MRSA strains, whereas ermA, tetM, ermC, tetK and msrA genes were detected in 157 (84%), 92 (49%), 40 (21%), 39 (21%) and 4 (2%) MRSA strains, respectively. The blaZ, tetM, ermC and tetK genes were plasmid-encoded, with ermC gene being easily transmissible. Tn5801-like transposon was present in 78 tetM-positive strains. ermA and tetM genes were the most prevalent erythromycin and tetracycline resistance determinants, respectively, in MRSA strains. The association of resistance genes with mobile genetic elements possibly enhances the spread of resistant traits in MRSA.

Abstract

Griseofulvin(GF) has become the drug of choice as an antifungal agent for patients who suffer from many kinds of fungal infection. In order to clarify hepatic injury by griseofulvin(GF) overload and the effect of UDCA on GF-induced hepatic injury, the authors carried out biochemical, histologic, and ultrastructural studies of liver following treatment with griseofulvin and ursodeoxycholic acid(UDCA) in mice. Urine porphobilinogen excretion in the group treated with GF alone was significantly increased and reached the highest level in the 4th week and declined thereafter. Biochemical studies of the liver function showed no remarkable changes of serum bilirubin levels throughout the experimental period in all groups, except for SGPT and alkaline phosphatase activities which were significantly elevated and reached the highest level in the second week. Then they slightly decreased in GF treated groups(GF alone and GF plus UDCA) in comparison with the control group. Pathologic findings in the group treated with GF alone include focal liver cell necrosis(esp, zone 3), Mallory bodies in hepatocytes(esp, zone 1), Kupffer cell activation, and brown protoporphyrin pigments in the hepatocytes, bile canaliculi and interlobular bile ducts with a marked inflammatory cell infiltration in the portal tracts. Under the polarizing light microscope, bile ductular and canalicular thrombi showed a "Maltese cross" birefringence in mice treated with GF alone. There is no definite finding of fatty change in hepatocyte. Under the microscope, the liver appeared normal with an intact lobular architecture in the GF plus UDCA treated group. Electron microscopically, GF-induced changes include swelling of mitochondria, globular protoporphyrin crystals in the hepatocyte cytoplasm, markedly dilated bile cannaliculi and bile ducts and the formation of a Mallory hyaline bodies in the hepatocytes. There were no noticeable structural changes in the GF plus UDCA-treated group. Therefore the results suggest that GF causes hepatic injury, namely porphyria and cholestasis, and the treatment of UDCA may have cytoprotective and choleretic effects on GF-induced hepatic injuries.
