Adolescents tend to neglect food and their eating pattern is influenced by several factors. Adolescents’ health literacy substantiates their ability to respond to the growing demands of health, being linked to health promotion in several areas, including food. The goal of this work was to analyze the relationship between sociodemographic and contextual variables with secondary school students’ food literacy. A quantitative, cross-sectional, descriptive and analytical study was carried out with a sample of 181 nonprobabilistic secondary school students, aged between 16 and 21 years (mean = 18.09 years ± 1.45), mostly female (58.0%), from 11th grade (35.4%), who are part of a secondary school in central Portugal. As a tool for data collection, the questionnaire of the project “Your PEL – Promote and Empower for Health Literacy in the young population” was used, aggregating three areas: eating behaviors, harmful consumptions and sexuality. The results showed that 58.6% of young people eat 4–5 meals a day, consume fast food weekly (79%) and eat soup (82.3%). Additionally, they eat fruit and vegetables on a daily basis (94.5 and 83.4%, respectively). The results further showed that 10th grade students have higher levels of health and food literacy. It was observed that both girls and boys are interested in receiving information regarding food, transmitted through social networks and by a communication application (84.5 and 73.5%, respectively), with significant differences (χ 2 = 4.768; p = 0.028). Adolescents face unique health challenges and a critical level of health literacy compromises their understanding of information about diet and future health. The results indicate that educational plans to empower adolescents in these areas, which integrate gender and age differences and sources of information as important variables to be considered, are pivotal for increasing levels of health and food literacy.

During the first years of Covid, 2020–2023, a group of seven colleagues across three continents—working outside of institutional contexts—prepared a Practice Note on naturecultures. The Practice Note draws together the long-time work, experience, and thinking of the authors, all of whom work in the field of heritage conservation. It gives focus to the improved integration of nature and culture, and cultural heritage and natural heritage, in the work of caring for and safeguarding important places. By promoting awareness of diversity and mutual respect for multiple views and understandings, the Practice Note is concerned with working together, fostering dialogue, and creating long-lasting and equitable approaches to conservation. In this introduction, we outline the purpose, origins, and the making of the Practice Note.

Around 60% of individuals with neurodevelopmental disorders (NDD) remain undiagnosed after comprehensive genetic testing, primarily of protein-coding genes1. Large genome-sequenced cohorts are improving our ability to discover new diagnoses in the non-coding genome. Here we identify the non-coding RNA RNU4-2 as a syndromic NDD gene. RNU4-2 encodes the U4 small nuclear RNA (snRNA), which is a critical component of the U4/U6.U5 tri-snRNP complex of the major spliceosome2. We identify an 18 base pair region of RNU4-2 mapping to two structural elements in the U4/U6 snRNA duplex (the T-loop and stem III) that is severely depleted of variation in the general population, but in which we identify heterozygous variants in 115 individuals with NDD. Most individuals (77.4%) have the same highly recurrent single base insertion (n.64_65insT). In 54 individuals in whom it could be determined, the de novo variants were all on the maternal allele. We demonstrate that RNU4-2 is highly expressed in the developing human brain, in contrast to RNU4-1 and other U4 homologues. Using RNA sequencing, we show how 5 splice-site use is systematically disrupted in individuals with RNU4-2 variants, consistent with the known role of this region during spliceosome activation. Finally, we estimate that variants in this 18 base pair region explain 0.4% of individuals with NDD. This work underscores the importance of non-coding genes in rare disorders and will provide a diagnosis to thousands of individuals with NDD worldwide.

Considering the lack of consensus related to the impact of selective IL-6 receptor inhibition on bone remodeling and the scarcity of reports, especially on large bone defects, this study proposed to evaluate the biological impact of the selective inhibitor of interleukin-6 receptor (tocilizumab) in an experimental model of critical calvarial defect in rats. In this preclinical and in vivo study, 24 male Wistar rats were randomly divided into two groups (n=12/group): defect treated with collagen sponge (CG) and defect treated with collagen sponge associated with 2 mg/kg tocilizumab (TCZ). The defect in the parietal bone was created using an 8-mm diameter trephine drill. After 90 days, the animals were euthanized, and tissue samples (skull caps) were evaluated through micro-CT, histological, immunohistochemistry, cytokines, and RT-qPCR analyses. Tocilizumab reduced mononuclear inflammatory infiltration (P0.05). The bone cells (osteoblasts, osteoclasts, and osteocytes) in the defect area were similar in both groups (P>0.05). Tocilizumab reduced inflammatory cytokines, decreased osteogenic protein, and increased proteases in a critical bone defect in rats. Ninety days after the local application of tocilizumab in the cranial defect, we did not find a significant formation of bone tissue compared with a collagen sponge.

Introduction: Renal cell carcinoma (RCC) often develops a tumor thrombus extending into the inferior vena cava (IVC). Radical nephrectomy with IVC thrombectomy is the standard treatment, although prognostic factors are yet to be properly established. Objectives: The objectives of this study were to review the clinicopathological features of surgically treated patients with RCC and IVC thrombus and to investigate potential prognostic factors. Materials and Methods: This retrospective analysis covered patients with RCC and IVC thrombus who underwent surgical treatment at a tertiary center over 12 years. Results: Of the 32 patients included, 56% and 41% had nodal (N1) and metastatic (M1) diseases, respectively. Thrombus level was 1 in 25% and 4 in 21.9% of cases, according to the Mayo classification. The median follow-up was 17.0 months. The median overall survival (OS) was 20.0 months, with a median OS of 36.0 months in M0 patients and 10.0 months in M1 patients (log-rank P = 0.029). Stage IV disease (T4 and/or M1 status) (hazard ratio [HR]: 2.85, P = 0.021), fat invasion (HR: 2.52, P = 0.044), positive margins (HR: 2.54, P = 0.037), American Society of Anesthesiologists score (HR: 2.59, P = 0.033), tumor size >100 mm (HR: 2.538, P = 0.033), and higher neutrophil-to-lymphocyte ratio (r2 = 0.304, P = 0.001) were significantly associated with worse OS in univariate analysis. Thrombus level did not impact prognosis. Conclusions: Certain clinicopathological factors, but not thrombus level, appear to influence prognosis. Prospective multicentric randomized studies are needed to better stratify patient risk, improve prognostic prediction, and evaluate systemic therapy responses.

OBJECTIVE: To conduct an integrative literature review on epidemiology of falls in older adults in Brazil, seeking to identify occurrence rate, recurrence, and potentially modifiable factors associated with these events. METHOD: This literature review consisted of searching the Medical Literature Analysis and Retrieval System Online (MEDLINE), Scientific Electronic Library Online (SciELO), and Latin American and Caribbean Center on Health Sciences Information (LILACS) databases for scientific articles on November 14, 2017. The following keywords were used for finding articles: “accidental falls,” AND/OR “elderly,” AND/OR “Brazil.” Epidemiological data were collected from primary studies and then compared. RESULTS: Thirty-five studies were selected. Most of them were performed in the Brazilian Southeast (15) and South (11) regions. The occurrence rate of falls ranged from 10.7 to 59.3%. The most common setting for falls was the home during daytime. The most commonly described circumstances were tripping, slipping, dizziness, and uneven flooring, resulting in falling from one’s own height. The factors most frequently associated with falls were female sex, age greater than 80 years, cognitive impairment, and depressive symptoms. The reported consequences were fractures and fear of falling again. CONCLUSION: As some factors associated with falls in older adults in Brazil are modifiable, fall prevention may reduce morbidity and mortality in this population.

Abstract Gonadal hormone activities mediated by androgen and estrogen receptors, along with cell-autonomous mechanisms arising from the absence of sex-chromosome dosage compensation, are key factors in avian sexual development. In this study, we generate androgen receptor (AR) knockout chickens (AR−/−) to explore the role of androgen signaling in avian sexual development. Despite developing sex-typical gonads and gonadal hormone production, AR−/− males and females are infertile. While few somatic sex-specific traits persist (body size, spurs, and tail feathers), crucial sexual attributes such as comb, wattles and sexual behaviors remain underdeveloped in both sexes. Testosterone treatment of young AR−/− males fails to induce crow behavior, comb development, or regression of the bursa of Fabricius, which are testosterone-dependent phenotypes. These findings highlight the significance of androgen receptor mechanisms in fertility and sex-specific traits in chickens, challenging the concept of a default sex in birds and emphasizing the dominance of androgen signaling in avian sexual development.

Abstract Studies for the preservation of seagrass beds biotopes have met difficulties in establishing appropriate methods assessing their health. We tested the efficiency of space occupation by seagrasses scattered worldwide (dgrass index), which proved to be dependent on clonal growth form and morphometric plasticity. dgrass correlated with the above-ground to below-ground biomass ratio. However, the latter was misleading when high ratios resulted from low below-ground biomass. Nutrient Posphate-limitations were revealed in situations of theoretical Nitrogen-limitation. Enhanced nutrient supply benefitted seagrasses only up to a threshold after which it became detrimental. Better nurtured, healthier meadows with denser canopies increased the organic matter in the sediment and had associated greater abundances of benthic macrofauna. Hence, seagrass biotopes could benefit from moderate anthropogenic nutrient additions. However, organic matter above ≈6% and/or reduced riverine discharges (dams upstream and climate-change-related droughts) were detrimental to healthy meadows, jeopardizing ecosystem services such as macrofauna abundances and carbon sinks.

Abstract A genomic database of all Earth’s eukaryotic species could contribute to many scientific discoveries; however, only a tiny fraction of species have genomic information available. In 2018, scientists across the world united under the Earth BioGenome Project (EBP), aiming to produce a database of high-quality reference genomes containing all ~1.5 million recognized eukaryotic species. As the European node of the EBP, the European Reference Genome Atlas (ERGA) sought to implement a new decentralised, equitable and inclusive model for producing reference genomes. For this, ERGA launched a Pilot Project establishing the first distributed reference genome production infrastructure and testing it on 98 eukaryotic species from 33 European countries. Here we outline the infrastructure and explore its effectiveness for scaling high-quality reference genome production, whilst considering equity and inclusion. The outcomes and lessons learned provide a solid foundation for ERGA while offering key learnings to other transnational, national genomic resource projects and the EBP.

The diverse physiography of the Portuguese land and marine territory, spanning from continental Europe to the Atlantic archipelagos, has made it an important repository of biodiversity throughout the Pleistocene glacial cycles, leading to a remarkable diversity of species and ecosystems. This rich biodiversity is under threat from anthropogenic drivers, such as climate change, invasive species, land use changes, overexploitation, or pathogen (re)emergence. The inventory, characterisation, and study of biodiversity at inter- and intra-specific levels using genomics is crucial to promote its preservation and recovery by informing biodiversity conservation policies, management measures, and research. The participation of researchers from Portuguese institutions in the European Reference Genome Atlas (ERGA) initiative and its pilot effort to generate reference genomes for European biodiversity has reinforced the establishment of Biogenome Portugal. This nascent institutional network will connect the national community of researchers in genomics. Here, we describe the Portuguese contribution to ERGA’s pilot effort, which will generate high-quality reference genomes of six species from Portugal that are endemic, iconic, and/or endangered and include plants, insects, and vertebrates (fish, birds, and mammals) from mainland Portugal or the Azores islands. In addition, we outline the objectives of Biogenome Portugal, which aims to (i) promote scientific collaboration, (ii) contribute to advanced training, (iii) stimulate the participation of institutions and researchers based in Portugal in international biodiversity genomics initiatives, and (iv) contribute to the transfer of knowledge to stakeholders and engaging the public to preserve biodiversity. This initiative will strengthen biodiversity genomics research in Portugal and fuel the genomic inventory of Portuguese eukaryotic species. Such efforts will be critical to the conservation of the country’s rich biodiversity and will contribute to ERGA’s goal of generating reference genomes for European species.

Background: The long-term success of dental implants largely depends on achieving primary stability, previously described as crucial to obtaining osseointegration and immediate loading protocol requirements. Implant thread depths seem to be one of the key factors influencing primary stability, particularly in low-density bone. Insertion torque (IT) and resonance frequency analysis (RFA) are considered the most reliable tests to assess primary stability. The aim of this work was to evaluate how different thread depths of commercially available dental implants affect primary stability in low-density D3 bone. Materials and Methods: An in vitro study was carried out between February 2024 and March 2024. Twenty-four dental implants were divided into four groups (six implants each) according to their thread depths (Group A: 4 mm, Group B: 4.5 mm, Group C: 5 mm, Group D: 5.5 mm) and were inserted in D3-type artificial bone blocks. The main outcome variables were the IT and the Implant Stability Quotient (ISQ) measured in four different areas of the implant (buccal, lingual, mesial, and distal) with an Osstel® ISQ reader. Descriptive and inferential analyses of the data were performed, and the significance value was set at 5%. Results: A total of 24 implants were analyzed. The highest IT values were obtained in Group D, with a mean of 54.03 Ncm (standard deviation (SD) = 8.99), while the lowest measurements were observed in Group A (mean = 25.12; SD: 2.96 N.cm). The mean ISQ values were consistently higher in Group D for each analyzed area, with a mean of 70.13 N.cm (SD = 1.12). Conclusions: Taking into consideration the limitations of this in vitro study, greater thread depths seem to increase the primary stability of dental implants placed in soft bone. Furthermore, a positive correlation was observed between all IT and ISQ values, regardless of the thread depth.

Abstract Repeat expansions in FGF14 cause autosomal dominant late-onset cerebellar ataxia (SCA27B) with estimated pathogenic thresholds of 250 (incomplete penetrance) and 300 AAG repeats (full penetrance), but the sequence of pathogenic and non-pathogenic expansions remains unexplored. Here, we demonstrate that STRling and ExpansionHunter accurately detect FGF14 expansions from short-read genome data using outlier approaches. By combining long-range PCR and nanopore sequencing in 169 patients with cerebellar ataxia and 802 controls, we compare FGF14 expansion alleles, including interruptions and flanking regions. Uninterrupted AAG expansions are significantly enriched in patients with ataxia from a lower threshold (180–200 repeats) than previously reported based on expansion size alone. Conversely, AAGGAG hexameric expansions are equally frequent in patients and controls. Distinct 5’ flanking regions, interruptions and pre-repeat sequences correlate with repeat size. Furthermore, pure AAG (pathogenic) and AAGGAG (non-pathogenic) repeats form different secondary structures. Regardless of expansion size, SCA27B is a recognizable clinical entity characterized by frequent episodic ataxia and downbeat nystagmus, similar to the presentation observed in a family with a previously unreported nonsense variant (SCA27A). Overall, this study suggests that SCA27B is a major overlooked cause of adult-onset ataxia, accounting for 23–31% of unsolved patients. We strongly recommend re-evaluating pathogenic thresholds and integrating expansion sequencing into the molecular diagnostic process.

Human Hsp70-escort protein 1 (hHep1) is a cochaperone that assists in the function and stability of mitochondrial HSPA9. Similar to HSPA9, hHep1 is located outside the mitochondria and can interact with liposomes. In this study, we further investigated the structural and thermodynamic behavior of interactions between hHep1 and negatively charged liposomes, as well as interactions with cellular membranes. Our results showed that hHep1 interacts peripherally with liposomes formed by phosphatidylserine and cardiolipin and remains partially structured, exhibiting similar affinities for both. In addition, after being added to the cell membrane, recombinant hHep1 was incorporated by cells in a dose-dependent manner. Interestingly, the association of HSPA9 with hHep1 improved the incorporation of these proteins into the lipid bilayer. These results demonstrated that hHep1 can interact with lipids also present in the plasma membrane, indicating roles for this cochaperone outside of mitochondria.

The environmental conditions to which dams are exposed play a major role in dictating the progression and manifestation of the alkali–aggregate reaction (AAR). However, in the numerical thermal-mechanical simulation of AAR-affected dams, the solar radiation and its associated shadow effects have received little attention. The spatiotemporal distribution of the solar radiation incidence on the dam surfaces has often been addressed in a simplified way or has just been neglected. Yet, far less attention has been given to shadows cast by the dam’s own geometry or the slopes. The main reasons for these simplifications derive from the fact that contrary to other thermal loads, environmental actions vary in daily and annual cycles, with the added complication that solar radiation also depends on the orientation of the surface with respect to the Sun’s rays. In this way, a conventional thermal finite element code should be modified in order to deal with these two particular issues. Therefore, this article starts with the estimation of the solar radiation distribution by recourse to concepts of astronomy and computer graphics. Then, to illustrate the influence of the nonuniform temperature distribution on dam surfaces due to solar radiation and shading, the analysis of an AAR-affected arch dam is presented in this paper. A comparison of the AAR expansions computed on the dam with or without considering the solar radiation and shading is presented.

Abstract The article aims to assess the connection between sustainable leadership and sustainable entrepreneurship, by providing a framework based on the main findings of the clusters relating the effectiveness of that connection. Using the Web of Science databases and applying inclusion and exclusion criteria, 149 articles were retrieved and analysed. The bibliometric analysis was conducted using R-Studio software. The main findings unveil that sustainable leadership and sustainable entrepreneurship are interrelated concepts that play a key role in promoting responsible business practices and formulating solutions to contemporary socio-environmental challenges. The study’s main contribution lies in highlighting the importance of knowledge about sustainable leadership, sustainable entrepreneurship and their links. In practical terms, analysis of the connection between sustainable leadership and sustainable entrepreneurship reveals how the former plays an important role in promoting the latter, inspiring and supporting entrepreneurs in the adoption of sustainable practices, generating a positive socio-environmental impact as well as economic value for the firm. There is a need for more research on sustainable leadership, especially correlating its effectiveness in promoting sustainable entrepreneurship.

The manufacturing and characterization of nanographite films on substrates form the foundation for advances in materials science. Conductive graphite films are challenging products, as isolating graphite oxide is often necessary. In this study, nanographite suspensions containing non-oxidized graphite flakes were used to fabricate novel thin and ultrathin films via blade coating on industry-standard substrates. Films as thin as 346 nm were successfully fabricated. Moreover, it was possible to induce the orientation of the graphite nanoflakes via blade coating. This orientation led to electrical anisotropy; thus, the electrical behavior of the films in each orthogonal direction differed. After adjusting the coating parameters and the concentration of the nanographite flakes, the electrical conductivity ranged from 0.04 S/cm to 0.33 S/cm. In addition, with such adjustments, the transparency of the films in the visible range varied from 20% to 75%. By establishing a methodology for the tuning of both electrical and optical properties via adjustments in the nanographite suspension and coating parameters, we can fabricate resistant, conductive, and transparent films satisfying certain requirements. The results presented here can be extrapolated to enhance applications, especially for photonics and solar cells, in fields that require electrical conductive materials with high levels of transparency.

Abstract Excess sugar is considered one of the primary factors contributing to overweight status. In Brazil, sugar-sweetened beverages (SSBs) contain a significant amount of this nutrient and are consumed excessively. These beverages are associated with adverse health outcomes and impose costs on the healthcare system. The literature currently lacks studies that aim to attribute specific nutrients or foods as causes of diseases and also evaluate their economic impact, especially in middle- and low-income countries. This study aims to estimate the direct and indirect costs of obesity, stratified by sex and age group, resulting from the excessive consumption of sugar-sweetened beverages in Brazil from 2008 to 2020, and to project these costs for the year 2036. The estimation of obesity costs attributable to excessive consumption of SSBs was based on relative risks and the population prevalence of obesity, considering expenditures on hospitalizations and outpatient procedures in the Unified Health System (SUS). Cost information was obtained from the health information systems available at SUS. The highest burden attributable to the consumption of SSBs was observed among younger individuals and progressively decreased with advancing age. The total direct costs in the period between 2008 and 2020 amounted to approximately US$ 6.33 million, 87% of which was related to expenses for females. Additionally, deaths resulting from the consumption of SSBs cost the economy US$ 40 million due to the premature loss of productivity. The total costs of obesity attributable to the consumption of SSBs are substantial, impacting public spending and generating social and productivity losses that burden the economy. It is crucial to develop and implement cost-effective fiscal and regulatory policies aimed at preventing and combating obesity.
