Abstract: The BL Lac object 3C 371 is one of the targets that are regularly monitored by the Whole Earth Blazar Telescope (WEBT) Collaboration to study blazar variability on both short and long timescales. We aim to evaluate the long-term multiwavelength (MWL) behaviour of 3C 371, comparing it with the results derived for its optical emission in our previous study. For this, we make use of the multi-band campaigns organized by the WEBT Collaboration in optical and radio between January 2018 and December 2020, and of public data from Swift and Fermi satellites and the MOJAVE Very Large Interferometry programme. We evaluate the variability shown by the source in each band with the amplitude variability quantification, as well as possible interband correlation using the z-Discrete Correlation Function. We also present a deep analysis of the optical-UV, X-ray and $\gamma$-ray spectral variability. With the MOJAVE data we perform a kinematics analysis, looking for components propagating along the jet, calculating its kinematics parameters. This set of parameters is later used for the interpretation of the source MWL behaviour, modelling the broadband spectral energy distribution (SED) of the source with theoretical blazar emission scenarios., Comment: Accepted in A&A, 23 pages, 15 figures

Abstract: Due to its peculiar and highly variable nature, the blazar 3C 454.3 has been extensively monitored by the WEBT team. Here, we present for the first time these long-term optical flux and color variability results using data acquired in B, V, R, and I bands over a time span of $\sim$ 2 decades. We include data from WEBT collaborators and public archives such as SMARTS, Steward Observatory, and ZTF. The data are binned and segmented to study the source over this long term when more regular sampling was available. During our study, the long-term spectral variability reveals a redder when brighter (RWB) trend, which, however, stabilizes at a particular brightness cutoff $\sim$ 14.5 mag in the I-band, after which it saturates and evolves into a complex state. This trend indicates increasing jet emission dominance over accretion disk emission until jet emission completely dominates. Plots of the spectral index variation (following $F_{\nu} \propto \nu^{-\alpha}$) reveal a bimodal distribution using a one-day binning. These correlate with two extreme phases of 3C 454.3, an outburst or high flux state and quiescent or low flux state, which are respectively jet and accretion disk dominated. We have also conducted intra-day variability studies of nine light curves and found that six of them are variable. Abstract: The X-ray polarization observations made possible with the Imaging X-ray Polarimetry Explorer (IXPE) offer new ways of probing high-energy emission processes in astrophysical jets from blazars. Here we report on the first X-ray polarization observation of the blazar S4 0954+65 in a high optical and X-ray state. During our multi-wavelength campaign on the source, we detected an optical flare whose peak coincided with the peak of an X-ray flare. This optical-X-ray flare most likely took place in a feature moving along the parsec-scale jet, imaged at 43 GHz by the Very Long Baseline Array. The 43 GHz polarization angle of the moving component underwent a rotation near the time of the flare. In the optical band, prior to the IXPE observation, we measured the polarization angle to be aligned with the jet axis. In contrast, during the optical flare the optical polarization angle was perpendicular to the jet axis; after the flare, it reverted to being parallel to the jet axis. Due to the smooth behavior of the optical polarization angle during the flare, we favor shocks as the main acceleration mechanism. We also infer that the ambient magnetic field lines in the jet were parallel to the jet position angle. The average degree of optical polarization during the IXPE observation was (14.3$\pm$4.1)%. Despite the flare, we only detected an upper limit of 14% (at 3$\sigma$ level) on the X-ray polarization degree; although a reasonable assumption on the X-ray polarization angle results in an upper limit of 8.8% ($3\sigma$). We model the spectral energy distribution (SED) and spectral polarization distribution (SPD) of S4 0954+65 with leptonic (synchrotron self-Compton) and hadronic (proton and pair synchrotron) models. The constraints we obtain with our combined multi-wavelength polarization observations and SED modeling tentatively disfavor hadronic models for the X-ray emission in S4 0954+65., Comment: Submitted to A&A, 16 pages, 5 figures, and 7 tables

Abstract: OT 081 is a well-known, luminous blazar that is remarkably variable in many energy bands. We present the first broadband study of the source which includes very-high-energy (VHE, $E>$100\,GeV) $\gamma$-ray data taken by the MAGIC and H.E.S.S. imaging Cherenkov telescopes. The discovery of VHE $\gamma$-ray emission happened during a high state of $\gamma$-ray activity in July 2016, observed by many instruments from radio to VHE $\gamma$-rays. We identify four states of activity of the source, one of which includes VHE $\gamma$-ray emission. Variability in the VHE domain is found on daily timescales. The intrinsic VHE spectrum can be described by a power-law with index $3.27\pm0.44_{\rm stat}\pm0.15_{\rm sys}$ (MAGIC) and $3.39\pm0.58_{\rm stat}\pm0.64_{\rm sys}$ (H.E.S.S.) in the energy range of 55--300\,GeV and 120--500\,GeV, respectively. The broadband emission cannot be sucessfully reproduced by a simple one-zone synchrotron self-Compton model. Instead, an additional external Compton component is required. We test a lepto-hadronic model that reproduces the dataset well and a proton-synchrotron dominated model that requires an extreme proton luminosity. Emission models that are able to successfully represent the data place the emitting region well outside of the Broad Line Region (BLR) to a location at which the radiative environment is dominated by the infrared thermal radiation field of the dusty torus. In the scenario described by this flaring activity, the source appears to be an FSRQ, in contrast with past categorizations. Abstract: Blazars are beamed active galactic nuclei known for their strong multi-wavelength variability on timescales from years down to minutes. We aim to investigate the suitability of the twisting jet model presented in previous works to explain the multi-wavelength behaviour of BL Lacertae, the prototype of one of the blazar classes. According to this model, the jet is inhomogeneous, curved, and twisting, and the long-term variability is due to changes in the Doppler factor due to variations in the orientation of the jet-emitting regions. We analysed optical data of the source obtained during monitoring campaigns organised by the Whole Earth Blazar Telescope (WEBT) in 2019-2022, together with radio data from the WEBT and other teams, and gamma-ray data from the Fermi satellite. In this period, BL Lacertae underwent an extraordinary activity phase, reaching its historical optical and gamma-ray brightness maxima. The application of the twisting jet model to the source light curves allows us to infer the wiggling motion of the optical, radio, and gamma-ray jet-emitting regions. The optical-radio correlation shows that the changes in the radio viewing angle follow those in the optical viewing angle by about 120 days, and it suggests that the jet is composed of plasma filaments, which is in agreement with some radio high-resolution observations of other sources. The gamma-ray emitting region is found to be co-spatial with the optical one, and the analysis of the gamma-optical correlation is consistent with both the geometric interpretation and a synchrotron self-Compton (SSC) origin of the high-energy photons. We propose a geometric scenario where the jet is made up of a pair of emitting plasma filaments in a sort of double-helix curved rotating structure, whose wiggling motion produces changes in the Doppler beaming and can thus explain the observed multi-wavelength long-term variability., Comment: In press for A&A

Abstract: X-ray polarization is a unique new probe of the particle acceleration in astrophysical jets made possible through the Imaging X-ray Polarimetry Explorer. Here we report on the first dense X-ray polarization monitoring campaign on the blazar Mrk 421. Our observations were accompanied by an even denser radio and optical polarization campaign. We find significant short-timescale variability in both X-ray polarization degree and angle, including a $\sim90^\circ$ angle rotation about the jet axis. We attribute this to random variations of the magnetic field, consistent with the presence of turbulence but also unlikely to be explained by turbulence alone. At the same time, the degree of lower-energy polarization is significantly lower and shows no more than mild variability. Our campaign provides further evidence for a scenario in which energy-stratified shock-acceleration of relativistic electrons, combined with a turbulent magnetic field, is responsible for optical to X-ray synchrotron emission in blazar jets., Comment: 23 pages, including 8 pages of appendices. 12 figures, 3 tables. Abstract: Observations of quasars show that the polarization position angle of the emission coming from them varies greatly over time, including periods called rotations during which the angle changes in an orderly manner. The study proposes a method for identifying such events and assessing their statistical significance. The operation of the method is demonstrated using the example of long-term polarimetric observations of the blazars CTA 102, 3C 454.3, and OT 081. During the analysis of light curves, 51 rotations of the polarization position angle were found and it was shown that for CTA 102 and 3C 454.3 the rotations are predominantly oriented in one direction., Comment: 21 pages, 12 figures

Abstract: The BL Lac object 3C 371 has been observed by the Transiting Exoplanet Survey Satellite (\textit{TESS}) for approximately a year, between July 2019 and July 2020, with an unmatched 2-minute observing cadence. In parallel, the Whole Earth Blazar Telescope (WEBT) Collaboration organized an extensive observing campaign, providing three years of continuous optical monitoring between 2018 and 2020. These datasets allow for a thorough investigation of the variability of the source. The goal of this study is evaluating the optical variability of 3C 371. Taking advantage of the remarkable cadence of \textit{TESS} data, we aim to characterize the intra-day variability (IDV) displayed by the source and identify its shortest variability timescale. With this estimate, constraints on the size of the emitting region and black hole mass can be calculated. Moreover, WEBT data is used to investigate long-term variability (LTV), including understanding spectral behaviour of the source and the polarization variability. Based on the derived characteristics, information on the origin of the variability on different timescales is extracted. We evaluated the variability applying the variability amplitude tool that quantifies how variable the emission is. Moreover, we employed common tools like ANOVA (ANalysis Of VAariance) tests, wavelet and power spectral density (PSD) analyses to characterize the shortest variability timescales present in the emission and the underlying noise affecting the data. Short- and long-term colour behaviours have been evaluated to understand the spectral behaviour of the source. The polarized emission was analyzed, studying its variability and possible rotation patterns of the electric vector position angle (EVPA). Abstract: PG 1553+113 is one of the few blazars with a convincing quasi-periodic emission in the gamma-ray band. The source is also a very high-energy (VHE; >100 GeV) gamma-ray emitter. To better understand its properties and identify the underlying physical processes driving its variability, the MAGIC Collaboration initiated a multiyear, multiwavelength monitoring campaign in 2015 involving the OVRO 40-m and Medicina radio telescopes, REM, KVA, and the MAGIC telescopes, Swift and Fermi satellites, and the WEBT network. The analysis presented in this paper uses data until 2017 and focuses on the characterization of the variability. The gamma-ray data show a (hint of a) periodic signal compatible with literature, but the X-ray and VHE gamma-ray data do not show statistical evidence for a periodic signal. In other bands, the data are compatible with the gamma-ray period, but with a relatively high p-value. The complex connection between the low and high-energy emission and the non-monochromatic modulation and changes in flux suggests that a simple one-zone model is unable to explain all the variability. Abstract: We present multiwavelength polarization measurements of the luminous blazar Mrk~501 over a 14-month period. The 2--8 keV X-ray polarization was measured with the Imaging X-ray Polarimetry Explorer (IXPE) with six 100-ks observations spanning from 2022 March to 2023 April. Each IXPE observation was accompanied by simultaneous X-ray data from NuSTAR, Swift/XRT, and/or XMM-Newton. Complementary optical-infrared polarization measurements were also available in the B, V, R, I, and J bands, as were radio polarization measurements from 4.85 GHz to 225.5 GHz. Among the first five IXPE observations, we did not find significant variability in the X-ray polarization degree and angle with IXPE. However, the most recent sixth observation found an elevated polarization degree at $>3\sigma$ above the average of the other five observations. The optical and radio measurements show no apparent correlations with the X-ray polarization properties. Throughout the six IXPE observations, the X-ray polarization degree remained higher than, or similar to, the R-band optical polarization degree, which remained higher than the radio value. This is consistent with the energy-stratified shock scenario proposed to explain the first two IXPE observations, in which the polarized X-ray, optical, and radio emission arises from different regions., Comment: Accepted for publication in ApJ

Abstract: Variability can be the pathway to understanding the physical processes in astrophysical jets, however, the high-cadence observations required to test particle acceleration models are still missing. Here we report on the first attempt to produce continuous, >24 hour polarization light curves of blazars using telescopes distributed across the globe and the rotation of the Earth to avoid the rising Sun. Our campaign involved 16 telescopes in Asia, Europe, and North America. We observed BL Lacertae and CGRaBS J0211+1051 for a combined 685 telescope hours. We find large variations in the polarization degree and angle for both sources in sub-hour timescales as well as a ~180 degree rotation of the polarization angle in CGRaBS J0211+1051 in less than two days. We compared our high-cadence observations to Particle-In-Cell magnetic reconnection and turbulent plasma simulations. We find that although the state of the art simulation frameworks can produce a large fraction of the polarization properties, they do not account for the entirety of the observed polarization behavior in blazar jets., Comment: 20 pages, 15 figures, 2 tables, accepted for publication in A&A. Abstract: We study the optical flux and polarization variability of the binary black hole blazar OJ 287 using quasi-simultaneous observations from 2015 to 2023 carried out using telescopes in the USA, Japan, Russia, Crimea, and Bulgaria. This is one of the most extensive quasi-simultaneous optical flux and polarization variability studies of OJ 287. OJ 287 showed large amplitude, ~3.0 mag flux variability, large changes of ~37% in degree of polarization, and a large swing of ~215 degrees in the angle of the electric vector of polarization. During the period of observation, several flares in flux were detected. Those flares are correlated with a rapid increase in the degree of polarization and swings in electric vector of polarization angle. A peculiar behavior of anticorrelation between flux and polarization degree, accompanied by a nearly constant polarization angle, was detected from JD 2,458,156 to JD 2,458,292. Abstract: Context. Blazar AO 0235+164, located at redshift z = 0.94, has undergone several sharp multi-spectral-range flaring episodes during the last decades. In particular, the episodes peaking in 2008 and 2015, that received extensive multi-wavelength coverage, exhibited interesting behavior. Aims. We study the actual origin of these two observed flares by constraining the properties of the observed photo-polarimetric variability, those of the broad-band spectral energy-distribution and the observed time-evolution behavior of the source as seen by ultra-high resolution total-flux and polarimetric Very-long-baseline interferometry (VLBI) imaging. Methods. The analysis of VLBI images allows us to constrain kinematic and geometrical parameters of the 7 mm jet. We use the Discrete Correlation Function to compute the statistical correlation and the delays between emission at different spectral ranges. Multi-epoch modeling of the spectral energy distributions allows us to propose specific models of emission; in particular for the unusual spectral features observed in this source in the X-ray region of the spectrum during strong multi spectral-range flares. Results. We find that these X-ray spectral features can be explained by an emission component originating in a separate particle distribution than the one responsible for the two standard blazar bumps. This is in agreement with the results of our correlation analysis that do not find a strong correlation between the X-rays and the remaining spectral ranges. We find that both external Compton dominated and synchrotron self-Compton dominated models can explain the observed spectral energy distributions. However, synchrotron self-Compton models are strongly favored by the delays and geometrical parameters inferred from the observations.

Abstract: The Spin Physics Detector collaboration proposes to install a universal detector in the second interaction point of the NICA collider under construction (JINR, Dubna) to study the spin structure of the proton and deuteron and other spin-related phenomena using a unique possibility to operate with polarized proton and deuteron beams at a collision energy up to 27 GeV and a luminosity up to $10^{32}$ cm$^{-2}$ s$^{-1}$. As the main goal, the experiment aims to provide access to the gluon TMD PDFs in the proton and deuteron, as well as the gluon transversity distribution and tensor PDFs in the deuteron, via the measurement of specific single and double spin asymmetries using different complementary probes such as charmonia, open charm, and prompt photon production processes. Other polarized and unpolarized physics is possible, especially at the first stage of NICA operation with reduced luminosity and collision energy of the proton and ion beams. This document is dedicated exclusively to technical issues of the SPD setup construction.

Abstract: The PHENIX experiment has performed a systematic study of identified charged-hadron ($\pi^\pm$, $K^\pm$, $p$, $\bar{p}$) production at midrapidity in $p$$+$Al, $^3$He$+$Au, Cu$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV and U$+$U collisions at $\sqrt{s_{_{NN}}}=193$ GeV. Identified charged-hadron invariant transverse-momentum ($p_T$) and transverse-mass ($m_T$) spectra are presented and interpreted in terms of radially expanding thermalized systems. The particle ratios of $K/\pi$ and $p/\pi$ have been measured in different centrality ranges of large (Cu$+$Au, U$+$U) and small ($p$$+$Al, $^3$He$+$Au) collision systems. The values of $K/\pi$ ratios measured in all considered collision systems were found to be consistent with those measured in $p$$+$$p$ collisions. However the values of $p/\pi$ ratios measured in large collision systems reach the values of $\approx0.6$, which is $\approx2$ times larger than in $p$$+$$p$ collisions. These results can be qualitatively understood in terms of the baryon enhancement expected from hadronization by recombination. Identified charged-hadron nuclear-modification factors ($R_{AB}$) are also presented. Enhancement of proton $R_{AB}$ values over meson $R_{AB}$ values was observed in central $^3$He$+$Au, Cu$+$Au, and U$+$U collisions. The proton $R_{AB}$ values measured in $p$$+$Al collision system were found to be consistent with $R_{AB}$ values of $\phi$, $\pi^\pm$, $K^\pm$, and $\pi^0$ mesons, which may indicate that the size of the system produced in $p$$+$Al collisions is too small for recombination to cause a noticeable increase in proton production., Comment: 480 authors from 78 institutions, 18 pages, 6 tables, 16 figures. v2 is version accepted for publication in Physical Review C. Abstract: Optical circular polarization observations can directly test the particle composition in black holes jets. Here we report on the first observations of the BL Lac type object S4 0954+65 in high linear polarized states. While no circular polarization was detected, we were able to place upper limits of <0.5% at the 99.7% confidence. Using a simple model and our novel optical circular polarization observations we can constrain the allowed parameter space for the magnetic field strength and composition of the emitting particles. Our results favor models that require magnetic field strengths of only a few Gauss and models where the jet composition is dominated by electron-positron pairs. Abstract: The relevance of the study is explained by the need to consider the functioning of the mechanisms of discursive sensemaking and an impact on the recipients of information, taking into account the new linguosemiotic opportunities which modern political communication provides and which are manifested not only in the real life, but also in the virtual digital environment. The multimedia technologies are gaining an increasing role, allowing to encode discursive messages, using semiotically polycode format, combining the traditional verbal way of presenting information along with iconic, color, graphic, metatext and other formats. The aim of the study is to prove that semiotic parameters of visual metaphor as a polycode sign functioning in the discursive environment within the framework of the pragmatic situation “Elections”, determine its interpretative and semantic potential and interacting with other sign codes result in new meanings and interpretations. The materials for the study of visual metaphors are the official logos used during the 2024 election campaign in Mexico. The research methodology, which allows for a systematic study of visual metaphors semiotics, is based on the interdisciplinary and polyparadigmatic model of discourse studies and implies an integral approach that combines the general scientific methods of analysis and synthesis, comparison and analogy with the method of sociolinguistic interpretation, semantic, interpretive, linguocognitive and discourse analysis. It has been found out that the linguosemiotic paradigm of Mexican electoral discourse integrates verbal and non-verbal semiotic codes to express meanings, conceptualize and categorize reality. By using of various semiotic codes, the metaphor-logo condenses the semantic content and, as a cognitive model based on analogy, acts as a means of sensemaking, modelling the perception of the world in the interests of the addresser. The use of polysemantic iconic signs-images, understandable to a wide audience of voters, translates connotative, ideological and social meanings. Thus, a visual metaphor as a way of logical comprehension and verbalization of reality functions as a significant component of the cognitive space within electoral discourse.

Abstract: We present X-ray polarimetry observations from the Imaging X-ray Polarimetry Explorer (IXPE) of three low spectral peak and one intermediate spectral peak blazars, namely 3C 273, 3C 279, 3C 454.3, and S5 0716+714. For none of these objects was IXPE able to detect X-ray polarization at the 3$\sigma$ level. However, we placed upper limits on the polarization degree at $\sim$10-30\%. The undetected polarizations favor models where the X-ray band is dominated by unpolarized photons upscattered by relativistic electrons in the jets of blazars, although hadronic models are not completely eliminated. Abstract: In 2022 the BL Lac object S4 0954+65 underwent a major variability phase, reaching its historical maximum brightness in the optical and $\gamma$-ray bands. We present optical photometric and polarimetric data acquired by the Whole Earth Blazar Telescope (WEBT) Collaboration from 2022 April 6 to July 6. Many episodes of unprecedented fast variability were detected, implying an upper limit to the size of the emitting region as low as $10^{-4}$ parsec. The WEBT data show rapid variability in both the degree and angle of polarization. We analyse different models to explain the polarization behaviour in the framework of a twisting jet model, which assumes that the long-term trend of the flux is produced by variations in the emitting region viewing angle. All the models can reproduce the average trend of the polarization degree, and can account for its general anticorrelation with the flux, but the dispersion of the data requires the presence of intrinsic mechanisms, such as turbulence, shocks, or magnetic reconnection. The WEBT optical data are compared to $\gamma$-ray data from the Fermi satellite. These are analysed with both fixed and adaptive binning procedures. We show that the strong correlation between optical and $\gamma$-ray data without measurable delay assumes different slopes in faint and high brightness states, and this is compatible with a scenario where in faint states we mainly see the imprint of the geometrical effects, while in bright states the synchrotron self-Compton process dominates., Comment: 12 pages, 13 figures, in press for MNRAS

Abstract: The BL Lac 1ES 2344+514 is known for temporary extreme properties (e.g., a shift of the synchrotron SED peak energy $\nu_{synch,p}$ above 1keV). While those extreme states were so far observed only during high flux levels, additional multi-year observing campaigns are required to achieve a coherent picture. Here, we report the longest investigation of the source from radio to VHE performed so far, focusing on a systematic characterisation of the intermittent extreme states. While our results confirm that 1ES 2344+514 typically exhibits $\nu_{synch,p}>$1keV during elevated flux periods, we also find periods where the extreme state coincides with low flux activity. A strong spectral variability thus happens in the quiescent state, and is likely caused by an increase of the electron acceleration efficiency without a change in the electron injection luminosity. We also report a strong X-ray flare (among the brightest for 1ES 2344+514) without a significant shift of $\nu_{synch,p}$. During this particular flare, the X-ray spectrum is among the softest of the campaign. It unveils complexity in the spectral evolution, where the common harder-when-brighter trend observed in BL Lacs is violated. During a low and hard X-ray state, we find an excess of the UV flux with respect to an extrapolation of the X-ray spectrum to lower energies. This UV excess implies that at least two regions contribute significantly to the infrared/optical/ultraviolet/X-ray emission. Using the simultaneous MAGIC, XMM-Newton, NuSTAR, and AstroSat observations, we argue that a region possibly associated with the 10 GHz radio core may explain such an excess. Finally, we investigate a VHE flare, showing an absence of simultaneous variability in the 0.3-2keV band. Using a time-dependent leptonic modelling, we show that this behaviour, in contradiction to single-zone scenarios, can instead be explained by a two-component model., Comment: Accepted for publication in Astronomy & Astrophysics

Abstract: Chlamydia is a group of zoonotic diseases caused by Chlamydiaceae spp., which carries high potential risks for animal welfare, public health and animal husbandry. The application of effective vaccines for livestock is still one of the major important tools of managing Chlamydia infection. In this study, the main characteristics of the currently available Russian commercial inactivated whole-cell mono-, polyvalent, and combination vaccines for the prevention of either a single Chlamydia infection (monoinfection) or multi-pathogen infection in livestock, cattle, small ruminants, pigs, farm birds, and fur animals are briefly reviewed. All of the vaccines are inactivated, absolutely safe for the immunization of target livestock and naive untreated contacting animals in the same herd and produce no documented serious vaccine-related complications. Since chlamydia has not yet been eradicated in the world, the infection persists in nature and can be transmitted from infected livestock, wild, domestic animals, and migratory birds to unprotected farm animals. Thus, the availability of effective and safe chlamydia vaccines remains an important aspect for the prevention and control over Chlamydia infection in Russian Federation and worldwide.

Abstract: Introduction. Tumor-associated macrophages (TAMs) are essential innate immune cells in the tumor microenvironment. TAMs can stimulate cancer cell proliferation and primary tumor growth, angiogenesis, lymphangiogenesis, cancer cell invasiveness in vessels and metastatic niche formation as well as support chemotherapy resistance. TAMs are phenotypically diverse both in various cancer localizations and in intratumoral heterogeneous compartments. Tumor-specific modeling of TAMs is necessary to understand the fundamental mechanism of pro- and anti-tumor activity, to test their interaction with existing therapies, and to develop TAM- targeted immunotherapy. Aim of study: To investigate cancer-specific transcriptomic features of ex vivo human TAM models. Material and Methods. Here we compared transcriptomic profiles of TAMs for breast, colorectal, ovarian, lung, and prostate cancers ex vivo. Human monocytes were isolated from buffy coats, and then stimulated by the tumor cell conditioned medium ex vivo. Using real-time PCR, we quantified the expression of key TAM biomarkers including inflammatory cytokines, scavenger-receptors, angiogenesis-regulating genes, and matrix remodeling factors. Results. PCR analysis allowed revealing cancer-specific expression profiles of modeled TAMs. By comparing the existing knowledge about TAM phenotypes in human tumors in vivo with the collected data, we discuss the advantages and limitation of ex vivo TAM models derived from human blood monocytes. Conclusion. Monocytes-derived macrophages stimulated with cancer cell-conditioned medium can, to a certain extent, allow modeling of cancer-specific programming of TAMs. Our model system is valuable to examine agents reprogramming key TAM pro-tumoral activities, and for the reproducible analysis of mechanistic events that program tolerogenic status of TAMs towards cancer cells.

Abstract: Cognitive studies describe explicit and implicit means of revealing cognitive and linguistic skills. Allusions play a particular role in distributed cognition in language and speech. This research featured the cognitive potential of linguistic allusion in line with the theory of distributed cognition. The authors used the methods of conceptual and contextual analyses to clarify the main research concepts, describe allusion as a cognitive phenomenon, and define its functions. The article covers such issues as the interaction between the speaker and the language within the linguistic eco-system, the concepts of linguistic ecology and ecological linguistics, and the biological approach in linguistics. Allusion appeared to have constitutive and verifying functions. The constitutive function arranged discourse by correlating the information given by the respondent with the additional meanings that occur in the process of communication. The verifying function provided the accuracy of perception. These functions made it possible to use allusion to describe distributed cognition in speech as consisting of three stages: evaluation, correlation, and interaction.

Abstract: We study the broadband emission of Mrk 501 using multi-wavelength observations from 2017 to 2020 performed with a multitude of instruments, involving, among others, MAGIC, Fermi-LAT, NuSTAR, Swift, GASP-WEBT, and OVRO. Mrk 501 showed an extremely low broadband activity, which may help to unravel its baseline emission. Nonetheless, significant flux variations are detected at all wavebands, with the highest occurring at X-rays and very-high-energy (VHE) $\gamma$-rays. A significant correlation ($>$3$\sigma$) between X-rays and VHE $\gamma$-rays is measured, supporting leptonic scenarios to explain the variable parts of the emission, also during low activity. This is further supported when we extend our data from 2008 to 2020, and identify, for the first time, significant correlations between Swift-XRT and Fermi-LAT. We additionally find correlations between high-energy $\gamma$-rays and radio, with the radio lagging by more than 100 days, placing the $\gamma$-ray emission zone upstream of the radio-bright regions in the jet. Furthermore, Mrk 501 showed a historically low activity in X-rays and VHE $\gamma$-rays from mid-2017 to mid-2019 with a stable VHE flux ($>$0.2 TeV) of 5% the emission of the Crab Nebula. The broadband spectral energy distribution (SED) of this 2-year-long low-state, the potential baseline emission of Mrk 501, can be characterized with one-zone leptonic models, and with (lepto)-hadronic models fulfilling neutrino flux constraints from IceCube. We explore the time evolution of the SED towards the low-state, revealing that the stable baseline emission may be ascribed to a standing shock, and the variable emission to an additional expanding or traveling shock., Comment: 55 pages, 30 figures, 14 tables, accepted by APJS. Abstract: PHENIX presents a simultaneous measurement of the production of direct $\gamma$ and $\pi^0$ in $d$$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV over a $p_T$ range of 7.5 to 18 GeV/$c$ for different event samples selected by event activity, i.e. charged-particle multiplicity detected at forward rapidity. Direct-photon yields are used to empirically estimate the contribution of hard-scattering processes in the different event samples. Using this estimate, the average nuclear-modification factor $R_{d\rm Au,EXP}^{\gamma^{\rm dir}}$ is $0.925{\pm}0.023({\rm stat}){\pm}0.15^{\rm (scale)}$, consistent with unity for minimum-bias (MB) $d$$+$Au events. For event classes with moderate event activity, $R_{d\rm Au,EXP}^{\gamma^{\rm dir}}$ is consistent with the MB value within 5\% uncertainty. These results confirm that the previously observed enhancement of high-$p_T$ $\pi^0$ production found in small-system collisions with low event activity is a result of a bias in interpreting event activity within the Glauber framework. In contrast, for the top 5\% of events with the highest event activity, $R_{d\rm Au,EXP}^{\gamma^{\rm dir}}$ is suppressed by 20\% relative to the MB value with a significance of $4.5\sigma$, which may be due to final-state effects., Comment: 279 authors from 69 institutions, 8 pages, 3 figures, v2 is version accepted for publication in Physical Review Letters. Abstract: Reported here are transverse single-spin asymmetries ($A_{N}$) in the production of charged hadrons as a function of transverse momentum ($p_T$) and Feynman-$x$ ($x_F$) in polarized $p^{\uparrow}$+$p$, $p^{\uparrow}$+Al, and $p^{\uparrow}$+Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV. The measurements have been performed at forward and backward rapidity ($1.4<|\eta|<2.4$) over the range of $1.50$) in $p^{\uparrow}$+$p$ collisions, whereas the $p^{\uparrow}$+Al and $p^{\uparrow}$+Au results show smaller asymmetries. Abstract: Presented are the first measurements of the transverse single-spin asymmetries ($A_N$) for neutral pions and eta mesons in $p$+Au and $p$+Al collisions at $\sqrt{s_{_{NN}}}=200$ GeV in the pseudorapidity range $|\eta|<$0.35 with the PHENIX detector at the Relativistic Heavy Ion Collider. Abstract: Immunotherapy represents a groundbreaking therapeutic approach, based on the immune system’s intrinsic capacity to interfere with tumor progression, that opens the horizons in the treatment of endometrial cancer. However, the clinical efficacy of immunotherapy is hampered by the development of resistance in patients. The resistance to immunotherapy is multifactorial mechanism, encompassed genetic and epigenetic alterations in tumor cells modulating immune checkpoint molecules, resulted in escaping immune surveillance. The tumor microenvironment can orchestrate an immunosuppressive milieu, attenuating the immune response and facilitating tumor progression. To overcome immunotherapeutic resistance in endometrial cancer we must bring to light the mechanisms of intricate interplay between neoplastic cells, the host immune system, and the tumor microenvironment. The identification of predictive biomarkers for immunotherapeutic response and the innovative agents capable of reversing resistance pathways must be developed. Our review summarizes accumulated data on the role of cells of the tumor microenvironment and their regulatory molecules in the mechanisms underlying therapeutic effects of immune checkpoint inhibitors, including resistance to therapy. Major question we raise here – which group of patients is the most favorable to achieve durable immunotherapy response in endometrial cancer?

Abstract: Brain-derived neurotrophic factor (BDNF) is a secreted molecule that plays an important role in the survival and growth of nerve cells. BDNF undergoes complex post-translational processing with cellular proteases. Pro- and mature BDNF forms bind to different receptor types in the brain. BDNF is prominent in the neonatal cerebral cortex. The neonatal period is critical for the proper development of the brain and nervous system. Disruptions in these critical periods can have long-lasting effects on behavior and mental health. Individuals who experience adverse effects in the neonatal period have demonstrated a predisposition to depression and other neurobehavioral disorders. In this work we studied the influence of transient expression (P3–P8) elevation of pro-, mature and mutant forms of BDNF that could not be processed with cellular convertases in the neonatal medial prefrontal cortex (mPFC) on anxiety and depressive-like behavior in adolescents. Elevated expression of mature BDNF (LV-BDNF) increased anxiety and depressive-like behavior at P30. Only immobility in the tail suspension test was increased after expression of mutant BDNF (LV-pBDNF mut). Using our RNA-seq data and available online sn-RNAseq results, we investigated transcriptomic changes in the neonatal mPFC at P8 that underlie subsequent behavioral changes. Mature BDNF expression caused an increased transcriptional response in perivascular stromal cells (PSC) with such genes: Ptgds, Slc6a13, Slc22a6, Bnc2, Slc13a4, Aldh1a2. Based on GWAS data, Ptgds is a candidate gene associated with ADHD and bipolar disorder Pujol-Gualdo et al. (2021); Marín-Méndez et al. (2012); Munkholm et al. (2015). LV-pBDNF mut caused a complete opposite set of transcriptional changes in the PSC compared to LV-BDNF. The observed similar behavioral phenotype after expression of mature and mutant forms of BDNF together with the detected genes related to bipolar disorder underpinned that Bdnf could play a substantial role in the pathogenesis of this neurobehavioral disorder.

Abstract: Blazars are a class of jet-dominated active galactic nuclei with a typical double-humped spectral energy distribution. It is of common consensus the Synchrotron emission to be responsible for the low frequency peak, while the origin of the high frequency hump is still debated. The analysis of X-rays and their polarization can provide a valuable tool to understand the physical mechanisms responsible for the origin of high-energy emission of blazars. We report the first observations of BL Lacertae performed with the Imaging X-ray Polarimetry Explorer ({IXPE}), from which an upper limit to the polarization degree $\Pi_X<$12.6\% was found in the 2-8 keV band. We contemporaneously measured the polarization in radio, infrared, and optical wavelengths. Our multiwavelength polarization analysis disfavors a significant contribution of proton synchrotron radiation to the X-ray emission at these epochs. Abstract: The etiology of cerebral palsy in children with intrauterine hypotrophy at birth and developmental delay is often explained by chronic intrauterine hypoxia. However, children with muscle hypotonia and developmental delay require genetic examination. The aim of this study is to report a case of mitochondrial disease caused by FBXL4 gene mutations and to identify main diagnostic criteria for mitochondrial DNA (mtDNA) depletion syndromes (MDS) in early childhood. Mitochondrial DNA depletion syndrome-13 is associated with FBXL4 gene mutations located in the 6q16.1–q16.27 locus. This disorder was first described in 2013 by P.E. Bonnen and X. Gai independently. MDS are a clinically and genetically heterogeneous group of diseases inherited by an autosomal recessive type and caused by mutations in genes that support the biogenesis and integrity of mtDNA. Encephalomyopathic mtDNA depletion syndrome-13 (MTDPS13) (OMIM: 615471) is an exceedingly rare autosomal recessive disease caused by biallelic mutations in the FBXL4 gene (MIM: 605654) with an estimated prevalence of 1 case per 100,000– 400,000 newborns. The disease onset is usually observed in the neonatal period; 75 % of patients develop symptoms by the age of 3 months. In the majority of cases, mtDNA depletion syndrome-13 manifests itself in the early neonatal period; however, in some patients, the disease onset was registered by the age of 24 months. The disease is characterized by encephalopathy, hypotension, lactic acidosis, severe developmental delay, and changes in the area of basal ganglia revealed by magnetic resonance imaging of the brain. FBXL4-related encephalomyopathy is a multisystem disease primarily affecting the central nervous system, heart, and liver. It is characterized by different clinical manifestations such as lactic acidosis, developmental delay, generalized hypotension, nutritional disorders, and growth retardation. Some patients demonstrate specific facial features, including prominent forehead, sinus-shaped folds, thick eyebrows, long eyelashes, epicanthus, short eye slits, hypertelorism, wide and depressed nose bridge, long and smooth labial groove, thin upper lip, and low-set ears. The disease prognosis is extremely poor; most children die before the age of 4 years. Approximately half of the patients suffer from microcephaly and hyperammonemia. The outcome varies; death was reported in 30 % of cases. Mean time to death was 3 years (median – 2 years). The diagnosis is crucial for medical and genetic counseling and possible prenatal diagnosis.

Abstract: Background and Objectives: The development of resistance of microorganisms to existing antibacterial agents requires constant updating of existing drugs and research in the search for alternative sources of active substances. In recent years, the problem of the emergence of microorganisms resistant to all existing antimicrobial drugs has become systematic and requires significant attention from researchers to search for alternative sources of active substances. The main problem in the development of drugs based on antimicrobial peptides is the search for optimal solutions in the preparation of these substances. Therefore, optimization and search for methods of isolation, analysis and control of protein fractions of water-soluble peptides used for the subsequent development of antibacterial drugs based on them is an urgent task. Materials and Methods: Optimal conditions and methods have been selected for the preparative production of water-soluble peptides isolated from the biomass of Hermetia illucens larvae. Optimization and search of methods for isolation, analysis and control of protein fractions of these water-soluble peptides will ensure the accuracy of the results and obtain optimal amounts of protein fractions. Results: It has been found that the use of molecular sieves makes it possible to obtain a mixture of three peptides with a difference in chromatographic retention time of less than 1 minute, which has been confirmed by three parallel experiments on the isolation and purification of peptides. During HPLC it has been noted that protein fractions 1 and 2 have similar values and the first and third protein fractions have a smaller difference in retention time, which is why there is no complete separation of these chromatographic peaks. Comparison of the percentage of the area of the peptides obtained allows us to talk about the possibility of obtaining peptides of the same size from H. illucens larvae by HPLC, and in combination with DLS to obtain protein fractions with very similar physicochemical and physical characteristics, since this type of chromatography separates substances according to their size. Conclusion: The use of high-performance liquid chromatography makes it possible to establish the reproducibility of the method of isolation of antimicrobial peptides by cold extraction with water and further stages of protein purification, salting and molecular sieve chromatography, which, in correlation with DLS analysis, makes it possible to reliably identify the peptides obtained, and the developed technology of isolation and purification makes it possible to produce these proteins on an industrial scale at low cost.

Pityriasis amiantacea is a reaction of the skin to various inflammatory diseases (seborrheic dermatitis, psoriasis, atopic dermatitis, chronic lichen simplex and others) that manifests by thick gray-yellow scaly crusts surrounding the hair. Сrusts are often attached to hair quite firmly and an effort to remove them leads to hair pulling, that in combination with the inflammatory process and secondary infection can sometimes lead to the formation of secondary traumatic cicatricial alopecia or non-scarring alopecia with subsequent hair growth.Objective. The aim is to study and analyze modern scientific data regarding the clinical features and treatment of pityriasis amiantacea, and demonstrate our own clinical observation.Materials and methods. The article discusses current information about pityriasis amiantacea, a rare condition that can be a manifestation of various inflammatory diseases of the scalp. The article discusses in detail the clinical manifestations and variants of the process, and gives a description of the features of primary conditions that can lead to its development. The authors presented their own clinical case of a case of pityriasis amiantacea in a 21-year-old man, which developed as a complication of seborrheic dermatitis and accompanied by alopecia.Conclusion. There is no single approach to the treatment of pityriasis amiantacea; the choice of therapy depends on the underlying disease. Determining the primary disease that caused the development of pityriasis amiantacea is a difficult diagnostic task, since regardless of the etiology, the manifestations of pityriasis amiantacea on the scalp are almost the same. The persistence of the process and the lack of generally accepted approaches to therapy require further research. It seems especially significant due to the fact that long-term existence and the absence of treatment, it can lead to the formation of secondary traumatic cicatricial alopecia.

The requirements to the second foreign language Olympiad tasks implied: possibility of mass coverage of students with Olympiad tasks; compliance of the tasks with the curriculum; time limit; relying on the students’ experience, including their native language skills and abilities; increasing tasks complexity; variety of tasks forms and content, the tasks contents and novelty; problem nature tasks to stimulate students thinking, language guess; using authentic material for the Olympiad tasks; the use of local history and extralinguistic material. The general complexity of the tasks, and difficulties associated with the nature of the linguistic material were also taken into account.Conclusion. According to the results of the experimental work, the author comes to the following conclusion: the leading principle of organizing distance Olympiads assumes that the university faculty will be focused on the timely identification of the future teachers’ interest to the Korean language, their involvement in preparation for the Olympiad, thus forming their interest in Korean culture and language. Olympiad assignments should be creative so that students learn to find their own ways of solving problems and develop self-education skills. Participation in the Olympiad increases students’ learning motivation, broadens their horizons, and develops their ability to express their opinions. The variety of requirements to the work of participants makes the assessment of each member of the jury quite subjective, but the collective nature of the evaluation procedure ensures correctness and objectivity of the final decision, allows participants to familiarize with the results of their work and consider appeals after each round of the Olympiad participants.

44. Structural Changes in the Dynamics of China’s Exports and Imports

Abstract: The economic policy pursued has turned China into a major trading power. In terms of GDP, the Chinese economy has been the first economy in the world for a number of recent years; China’s rapidly growing trade flows have made it the largest trading partner for many countries. Over the past twenty years there has been rapid growth in exports and imports. The article puts forward a hypothesis about the presence of structural instability in the dynamics of Chinese exports for the period from 2000 to 2020. The hypothesis was confirmed and two periods were identified in the dynamics of exports and imports with different natures of the main development trend: from 2000 to 2008 and from 2009 to 2020. For each stage, trend equations were selected that describe the dynamics of exports and imports and an interval forecast of indexes for 2021 was made. Materials and methods. During the work, dynamic, structural analysis of analytical and statistical information was used; methods of analytical, logical, systemic, correlation and regression analysis were used, as well as analysis of structural changes. The analysis was carried out using the Statistica 10.0 program.Results. A change in the value of the import coverage ratio by exports was revealed before and after structural changes in the dynamics of the main foreign economic indexes and an assessment of their statistical significance was given. An analysis of the influence of export dynamics on GDP dynamics was carried out and it was found that before structural changes, the influence of export dynamics on GDP dynamics was statistically insignificant, and after a structural change, export dynamics began to have a direct impact on changes in China’s GDP. Conclusion. Analysis of official statistical data on the main indexes of China’s foreign economic activity for the period from 2000 to 2020 allowed us to note that the volume of Chinese exports increased over the period 2010-2019 from $1.58 trillion in 2010 (10.34% of global exports), to $2.50 trillion in 2019 (12.81% of global exports). In 2019, China continued to rank first in terms of export value among all countries in the world. Despite the challenging year of 2020, China’s exports reached almost 2.6 trillion US dollars and increased by 4% compared to 2019.China also occupies a leading position in the export of high-tech goods - their volume is 731.9 billion US dollars, the share of exports in the global volume is 25%. At the same time, the dynamics of Chinese exports for the period from 2000 to 2020 was heterogeneous and characterized by structural instability. Two stages can be distinguished with different characteristics of the main development trend: from 2000 to 2008 and from 2009 to 2020. Based on this, the authors came to the conclusion that to describe the main trend in the development of export and import dynamics for the period from 2000 to 2008, the exponential model is best suited. To describe the main trend in the development of export and import dynamics in the period from 2009 to 2020 - logarithmic model. A study of the consistency of changes in export and import volumes showed that in the period from 2009 to 2020 was characterized by greater consistency in China’s main foreign trade flows than the period from 2000 to 2008. In the period before the structural change in the dynamics of exports and imports, the coverage ratio varied from 105.7% to 127.6%; in the period after the structural change, the coverage ratio changed from 108.9% to 135.4%.Checking the statistical significance of the differences in the coefficient of coverage of imports by exports before and after the structural change showed that the differences are not statistically significant. In the period from 2000 to 2008 the dynamics of China’s exports did not have a statistically significant impact on the dynamics of the country’s GDP. From 2009 to 2020 after structural changes in the nature of the dynamics of the main indexes of China’s foreign trade, changes in exports began to have a statistically significant impact on the dynamics of GDP. A 1% increase in China’s exports now results in a 0.25% increase in GDP.The practical significance of the paper is determined by the developed methodology for analyzing structural changes in the dynamics of exports and imports, examined using the example of China.

Abstract: Most of the light from blazars, active galactic nuclei with jets of magnetized plasma that point nearly along the line of sight, is produced by high-energy particles, up to $\sim 1$ TeV. Although the jets are known to be ultimately powered by a supermassive black hole, how the particles are accelerated to such high energies has been an unanswered question. The process must be related to the magnetic field, which can be probed by observations of the polarization of light from the jets. Measurements of the radio to optical polarization - the only range available until now - probe extended regions of the jet containing particles that left the acceleration site days to years earlier (Jorstad et al., 2005; Marin et al., 2018; Blinov et al., 2021), and hence do not directly explore the acceleration mechanism, as could X-ray measurements. Here we report the detection of X-ray polarization from the blazar Markarian~501 (Mrk~501). We measure an X-ray linear polarization degree $\Pi_X \sim10\%$, a factor of $\sim2$ higher than the value at optical wavelengths, with a polarization angle parallel to the radio jet. Abstract: We present multi-epoch optical spectra of the $\gamma$-ray bright blazar 1156+295 (4C +29.45, Ton 599) obtained with the 4.3~m Lowell Discovery Telescope. During a multi-wavelength outburst in late 2017, when the $\gamma$-ray flux increased to $2.5\times 10^{-6} \; \rm phot\; cm^{-2}\; s^{-1}$ and the quasar was first detected at energies $\geq100$ GeV, the flux of the Mg II $\lambda 2798$ emission line changed, as did that of the Fe emission complex at shorter wavelengths. These emission line fluxes increased along with the highly polarized optical continuum flux, which is presumably synchrotron radiation from the relativistic jet, with a relative time delay of $\lesssim2$ weeks. This implies that the line-emitting clouds lie near the jet, which points almost directly toward the line of sight. The emission-line radiation from such clouds, which are located outside the canonical accretion-disk related broad-line region, may be a primary source of seed photons that are up-scattered to $\gamma$-ray energies by relativistic electrons in the jet., Comment: 25 pages, 12 figures

Abstract: Ovarian cancer (OC) is the most lethal gynecologic malignancy worldwide. The major clinical challenge includes the asymptomatic state of the disease, making diagnosis possible only at advanced stages. Another OC complication is the high relapse rate and poor prognosis following the standard first-line treatment with platinum-based chemotherapy. At present, numerous clinical trials are being conducted focusing on immunotherapy in OC; nevertheless, there are still no FDA-approved indications. Personalized decision regarding the immunotherapy, including immune checkpoint blockade and immune cell–based immunotherapies, can depend on the effective antigen presentation required for the cytotoxic immune response. The major aim of our study was to uncover tumor-specific transcriptional and epigenetic changes in peripheral blood monocytes in patients with high-grade serous ovarian cancer (HGSOC). Another key point was to elucidate how chemotherapy can reprogram monocytes and how that relates to changes in other immune subpopulations in the blood. To this end, we performed single-cell RNA sequencing of peripheral blood mononuclear cells (PBMCs) from patients with HGSOC who underwent neoadjuvant chemotherapeutic treatment (NACT) and in treatment-naïve patients. Monocyte cluster was significantly affected by tumor-derived factors as well as by chemotherapeutic treatment. Bioinformatical analysis revealed three distinct monocyte subpopulations within PBMCs based on feature gene expression – CD14.Mn.S100A8.9hi, CD14.Mn.MHC2hi and CD16.Mn subsets. The intriguing result was that NACT induced antigen presentation in monocytes by the transcriptional upregulation of MHC class II molecules, but not by epigenetic changes. Increased MHC class II gene expression was a feature observed across all three monocyte subpopulations after chemotherapy. Our data also demonstrated that chemotherapy inhibited interferon-dependent signaling pathways, but activated some TGFb-related genes. Our results can enable personalized decision regarding the necessity to systemically re-educate immune cells to prime ovarian cancer to respond to anti-cancer therapy or to improve personalized prescription of existing immunotherapy in either combination with chemotherapy or a monotherapy regimen.

Abstract: The PHENIX experiment reports systematic measurements at the Relativistic Heavy Ion Collider of $\phi$-meson production in asymmetric Cu$+$Au collisions at $\sqrt{s_{_{NN}}}$=200 GeV and in U$+$U collisions at $\sqrt{s_{_{NN}}}$=193 GeV. Measurements were performed via the $\phi\rightarrow K^{+}K^{-}$ decay channel at midrapidity $|\eta|<0.35$. Features of $\phi$-meson production measured in Cu$+$Cu, Cu$+$Au, Au$+$Au, and U$+$U collisions were found to not depend on the collision geometry, which was expected because the yields are averaged over the azimuthal angle and follow the expected scaling with nuclear-overlap size. The elliptic flow of the $\phi$ meson in Cu$+$Au, Au$+$Au, and U$+$U collisions scales with second-order-participant eccentricity and the length scale of the nuclear-overlap region (estimated with the number of participating nucleons). At moderate $p_T$, $\phi$-meson production measured in Cu$+$Au and U$+$U collisions is consistent with coalescence-model predictions, whereas at high $p_T$ the production is in agreement with expectations for in-medium energy loss of parent partons prior to their fragmentation. Abstract: Introduction. Chemotherapy is a common treatment for breast cancer. Chemotherapeutic drugs effect blood monocytes, which are major contributors to cancer pathogenesis. However, to date, pro-tumor or anti-tumor programming by chemotherapy of monocytes is controversial.Aim. To characterize changes in phenotypic and transcriptomic profiles of monocytes of breast cancer patients before and after chemotherapeutic treatment.Materials and methods. In a cohort of 50 breast cancer patients, monocyte populations were identified based on their expression of CD14, CD16, CD163, and HLA-DR evaluated by flow cytometry before and after neoadjuvant chemotherapy. Bulk RNA sequencing was adopted to explore the transcriptomic profile of CD14+ monocytes before and after treatment. After treatment, we observed an increase in the activity of signaling pathways related to lipid metabolism and intracellular transport of vesicles from the endoplasmic reticulum, against the background of a decreased response to exposure to interferon γ and interferon α, and foreign molecules (exogenous nucleic acids, viruses and bacteria).Results. In breast cancer patients, neoadjuvant chemotherapy decreased in CD14+16+HLA-DR+ monocytes. Under cytostatic treatment, increased gene expression of MGLL, NR4A2, UCK1, YOD1, ABCA2, PAPSS2, ATP10 (log2FoldChange ≥0.8; false discovery rate (FDR) ≤0.01) and decreased gene expression of KPNA2, ERCC4, JAGN1, RUBCNL, SMYD4, B3GALT4 (log2FoldChange ≥0.8; FDR ≤0.01) were observed in monocytes of patients. Using discriminant analysis, the relative numbers of CD14+16–, CD14+16+, CD14-16+, CD14+16-HLA-DR+, CD14+16+HLA-DR+ and CD14–16+HLA-DR+ monocytes in the blood were found to be valuable in predicting response to neoadjuvant chemotherapy.Conclusion. Thus, association of blood monocytes with chemotherapeutic treatment in breast cancer was revealed.
