Abstract Background Sunflower (Helianthus annuus) is one of the most important economic crops in oilseed production worldwide. The different cultivars exhibit variability in their resistance genes. The NAC transcription factor (TF) family plays diverse roles in plant development and stress responses. With the completion of the H. annuus genome sequence, the entire complement of genes coding for NACs has been identified. However, the reference genome of a single individual cannot cover all the genetic information of the species. Results Considering only a single reference genome to study gene families will miss many meaningful genes. A pangenome-wide survey and characterization of the NAC genes in sunflower species were conducted. In total, 139 HaNAC genes are identified, of which 114 are core and 25 are variable. Phylogenetic analysis of sunflower NAC proteins categorizes these proteins into 16 subgroups. 138 HaNACs are randomly distributed on 17 chromosomes. SNP-based haplotype analysis shows haplotype diversity of the HaNAC genes in wild accessions is richer than in landraces and modern cultivars. Ten HaNAC genes in the basal stalk rot (BSR) resistance quantitative trait loci (QTL) are found. A total of 26 HaNAC genes are differentially expressed in response to Sclerotinia head rot (SHR). A total of 137 HaNAC genes are annotated in Gene Ontology (GO) and are classified into 24 functional groups. GO functional enrichment analysis reveals that HaNAC genes are involved in various functions of the biological process. Conclusions We identified NAC genes in H. annuus (HaNAC) on a pangenome-wide scale and analyzed S. sclerotiorum resistance-related NACs. This study provided a theoretical basis for further genomic improvement targeting resistance-related NAC genes in sunflowers.

As education advances with the times, traditional English EFL teaching can no longer fully adapt to the current educational situation. This paper studies the mining of the Civics element in English teaching integrates it into EFL classroom teaching, and constructs the morality cultivation EFL critical thinking ability teaching strategy based on the critical thinking ability model. The effects of applying this teaching strategy are analyzed and compared to the six dimensions of critical thinking ability: interpretation, analysis, evaluation, reasoning, explanation, and self-regulation ability. The exploration of critical thinking ability development and individual teacher factors is conducted. After the teaching experiment, the experimental group improved by 12.08 points, while the control group only improved by 0.46 points, indicating a significant difference between the two groups’ scores. The Sig. The values of the experimental group in the six dimensions are less than 0.05, and the Sig. The values of the control group are greater than 0.05. The teaching strategy of moral cultivation in EFL critical thinking ability has an obvious effect on improving students’ critical thinking abilities. There is a correlation between the development of critical thinking ability and teachers’ education, and teachers with master’s degrees are better than teachers with bachelor’s degrees.

CircRNAs have been proven to play a pivotal role in cancer progression. The present study aims to explore the roles and related mechanisms of circSETD2 in breast cancer proliferation, migration and invasion. The expression of circSETD2 in BC was assessed by the GEO database and qRT‒PCR. The biological function and underlying molecular mechanism of circSETD2 in BC were explored using in vitro and in vivo experiments, including CCK8, transwell, RIP, western blot, and xenograft mouse models. The expression of circSETD2 was downregulated in BC tumors, in accordance with the GEO database. Overexpression of circSETD2 significantly suppressed cell growth, cell migration and invasion. Mechanistically, circSETD2 reduced the stabilization of YAP1 by competitively binding with HuR, resulting in inactivation of downstream targets such as CTGF, myc and Slug. Our work suggests that the novel signaling axis circSETD2/HuR/YAP1 plays an important role in BC progression. The molecular mechanism underlying this signaling axis may provide a potential therapeutic target for BC treatment.

We establish the formulation for quantum current. Given a symmetry group G, let $\mathcal{C}$:=Rep G be its representation category. Physically, symmetry charges are objects of $\mathcal{C}$ and symmetric operators are morphisms in $\mathcal{C}$. The addition of charges is given by the tensor product of representations. For any symmetric operator O crossing two subsystems, the exact symmetry charge transported by O can be extracted. The quantum current is defined as symmetric operators that can transport symmetry charges over an arbitrary long distance. A quantum current exactly corresponds to an object in the Drinfeld center $Z_1(\mathcal{C})$. The condition for quantum currents to be \TL{superconducting} is also specified, which corresponds to condensation of anyons in one higher dimension. To express the local conservation, the internal hom must be used to compute the charge difference, and the framework of enriched category is inevitable. To illustrate these ideas, we develop a rigorous scheme of renormalization in one-dimensional lattice systems and analyse the fixed-point models. It is proved that in the fixed-point models, superconducting quantum currents form a Lagrangian algebra in $Z_1(\mathcal{C})$ and the boundary-bulk correspondence is verified in the enriched setting. Overall, the quantum current provides a natural physical interpretation to the holographic categorical symmetry.

Objective: The feasibility of the determination of glutamate purity was studied by differential scanning calorimetry. Methods: Indium and Zinc were used to correct the instrument parameters. Under the conditions of nitrogen atmosphere and flow rate of 50 mL/min, the effects of sample particle size, experimental atmosphere, heating rate and sample quality on the DSC measurement results were investigated, and the glutamic acid purity was determined under the optimal conditions. The determination results were compared between DSC and China Pharmacopeia 2015 titration. Results: Under the condition of weighing 3 mg and heating rate at 40 ℃/min, the purity of glutamic acid was 99.74% and the precision was 0.07%. The purity of glutamic acid measured by titration 99.72%. Conclusion: The DSC method has high accuracy and simple operation, and can be used for the determination of glutamate purity.

The competitive removal of Pb2+ and malachite green (MG) from water by three magnetic phosphate nanocomposites (Fe3O4/Ba3(PO4)2, Fe3O4/Sr5(PO4)3(OH), and Fe3O4/Sr5xBa3x(PO4)3(OH), namely "FBP", "FSP", and "FSBP", respectively) was systematically investigated compared with Fe3O4 ("F") nanoparticle. Temperature and adsorbent dosage for competitive removal were optimized to be 20 °C and 0.05 g in 50 mL. The kinetic and isothermal adsorption results were fitted well with the pseudo-second-order model and Langmuir model, respectively. In the competitive removal process, FSP showed a high affinity to Pb2+ (202.8 mg/g) while FBP possessed high selectivity for MG (175.4 mg/g), and FSBP was effective at simultaneous removal of Pb2+ and MG, with a capacity of 143.7 and 90.9 mg/g, respectively. The magnetic contents in nanocomposites allow magnetic separation of materials from the water after treatment. We proposed that the simultaneous removal mechanism by FSBP was due to ion exchange between Pb2+ and Sr2+ in the lattice and then the formation of hydrogen bonds between PO43- outside the material's surface and positively charged hydrogen in MG. This study indicates the potential of these phosphate nanocomposites to be used as effective materials for selective or simultaneous removal of Pb2+ and MG from water.

Abstract Background The phenomenon of codon usage bias is known to exist in many genomes and is mainly determined by mutation and selection. Codon usage bias analysis is a suitable strategy for identifying the principal evolutionary driving forces in different organisms. Sunflower (Helianthus annuus L.) is an annual crop that is cultivated worldwide as ornamentals, food plants and for their valuable oil. The WRKY family genes in plants play a central role in diverse regulation and multiple stress responses. Evolutionary analysis of WRKY family genes of H. annuus can provide rich genetic information for developing hybridization resources of the genus Helianthus. Results Bases composition analysis showed the average GC content of WRKY genes of H. annuus was 43.42%, and the average GC3 content was 39.60%, suggesting that WRKY gene family prefers A/T(U) ending codons. There were 29 codons with relative synonymous codon usage (RSCU) greater than 1 and 22 codons ending with A and U base. The effective number of codons (ENC) and codon adaptation index (CAI) in WRKY genes ranged from 43.47–61.00 and 0.14–0.26, suggesting that the codon bias was weak and WRKY genes expression level was low. Neutrality analysis found a significant correlation between GC12 and GC3. ENC-plot showed most genes on or close to the expected curve, suggesting that mutational bias played a major role in shaping codon usage. The Parity Rule 2 plot (PR2) analysis showed that the usage of AT and GC was disproportionate. A total of three codons were identified as the optimal codons. Conclusion Apart from natural selection effects, most of the genetic evolution in the H. annuus WRKY genome might be driven by mutation pressure. Our results provide a theoretical foundation for elaborating the genetic architecture and mechanisms of H. annuus and contributing to enrich H. annuus genetic resources.

In response to climate change, the proportion of renewable energy penetration is increasing daily. However, there is a lack of flexible energy transfer mechanisms. The optimization effect of low-carbon economic dispatch in a single park is limited. In the context of the sharing economy, this study proposes a research method for multi-park electricity sharing and benefit allocation based on carbon credit trading. Firstly, a framework for multi-park system operation is constructed, and an energy hub model is established for the electrical, cooling, and heating interconnections with various energy conversions. Secondly, a park carbon emission reduction trading model is established based on the carbon credit mechanism, further forming an optimal economic and environmental dispatch strategy for multi-park electricity sharing. Matlab+Gurobi is used for solving. Then, based on asymmetric Nash bargaining, the comprehensive contribution rate of each park is calculated by considering their energy contribution and carbon emission reduction contribution, thereby achieving a fair distribution of cooperation benefits among multiple parks. The results show that the proposed method can effectively reduce the overall operational cost of multiple parks and decrease carbon emissions, and the benefit allocation strategy used is fair and reasonable, effectively motivating the construction of new energy in parks and encouraging active participation in cooperative operations by all parks. Cadmium is one of the most common heavy metal contaminants found in agricultural fields. MutSα, MutSβ, and MutSγ are three different MutS-associated protein heterodimer complexes consisting of MSH2/MSH6, MSH2/MSH3, and MSH2/MSH7, respectively. These complexes have different mismatch recognition properties and abilities to support MMR. However, changes in mismatch repair genes (OsMSH2, OsMSH3, OsMSH6, and OsMSH7) of the MutS system in rice, one of the most important food crops, under cadmium stress and their association with E2Fs, the key transcription factors affecting cell cycles, are poorly evaluated. In this study, we systematically categorized six rice E2Fs and confirmed that OsMSHs were the downstream target genes of E2F using dual-luciferase reporter assays. In addition, we constructed four msh mutant rice varieties (msh2, msh3, msh6, and msh7) using the CRISPR-Cas9 technology, exposed these mutant rice seedlings to different concentrations of cadmium (0, 2, and 4 mg/L) and observed changes in their phenotype and transcriptomic profiles using RNA-Seq and qRT-PCR. We found that the difference in plant height before and after cadmium stress was more significant in mutant rice seedlings than in wild-type rice seedlings. Transcriptomic profiling and qRT-PCR quantification showed that cadmium stress specifically mobilized cell cycle-related genes ATR, CDKB2;1, MAD2, CycD5;2, CDKA;1, and OsRBR1. Furthermore, we expressed OsE2Fs in yeasts and found that heterologous E2F expression in yeast strains regulated cadmium tolerance by regulating MSHs expression. Further exploration of the underlying mechanisms revealed that cadmium stress may activate the CDKA/CYCD complex, which phosphorylates RBR proteins to release E2F, to regulate downstream MSHs expression and subsequent DNA damage repairment, thereby enhancing the response to cadmium stress.

AIM: To evaluate the global trends in and explore hotspots of high myopia (HM) research. METHODS: This bibliometric analysis was used to reveal the publication trends in HM research field based on the Web of Science Core Collection (WoSCC). VOSviewer version 1.6.13 software was used to analyze the data and construct a knowledge map including the yearly publication number, journals, countries, international collaborations, authors, research hotspots, and intellectual base in HM. RESULTS: The search engine found 3544 peer-reviewed publications on HM between 2010 and 2019, and the yearly research output substantially elevated over the past decade. China is the top publishing country, and Sun Yat-sen University was the most active academic institution. Jonas JB is the top publishing scientist, and Investigative Ophthalmology and Visual Science (IOVS) was the most productive journal. The highest cited references mainly focused on epidemiology and management. The keywords formed 6 clusters: 1) refractive surgery; 2) etiology and clinical characteristics; 3) the mechanism of eye growth; 4) management for myopic maculopathy; 5) vitrectomy surgical treatment; 6) myopia-associated glaucoma-like optic neuropathy. CONCLUSION: The evaluation of development trends based on the data extracted from WoSCC can provide valuable information and guidance for ophthalmologists and public health researchers to improve management procedures in HM field.

African swine fever (ASF) is an acute, hemorrhagic, highly contagious disease in pigs caused by African swine fever virus (ASFV). Our previous study identified that the ASFV MGF300-2R protein functions as a virulence factor and found that MGF300-2R degrades IKKβ via selective autophagy. However, the E3 ubiquitin ligase responsible for IKKβ ubiquitination during autophagic degradation still remains unknown. In order to solve this problem, we first pulled down 328 proteins interacting with MGF300-2R through immunoprecipitation-mass spectrometry. Next, we analyzed and confirmed the interaction between the E3 ubiquitin ligase TRIM21 and MGF300-2R and demonstrated the catalytic role of TRIM21 in IKKβ ubiquitination. Finally, we indicated that the degradation of IKKβ by MGF300-2R was dependent on TRIM21. In summary, our results indicate TRIM21 is the E3 ubiquitin ligase involved in the degradation of IKKβ by MGF300-2R, thereby augmenting our understanding of the functions of MGF300-2R and offering insights into the rational design of live attenuated vaccines and antiviral strategies against ASF. Ischemic stroke is a devastating disease leading to neurologic impairment. Compounding the issue is the very limited array of available interventions. The activation of a γ-aminobutyric acid (GABA) type A receptor (GABAAR) has been reported to produce neuroprotective properties during cerebral ischemia, but its mechanism of action is not yet fully understood. Here, in a rat model of photochemically induced cerebral ischemia, we found that muscimol, a GABAAR agonist, modulated GABAergic signaling, ameliorated anxiety-like behaviors, and attenuated neuronal damage in rats suffering cerebral ischemia. Moreover, GABAAR activation improved brain antioxidant levels, reducing the accumulation of oxidative products, which was closely associated with the NO/NOS pathway. Notably, the inhibition of autophagy markedly relieved the neuronal insult caused by cerebral ischemia. We further established an oxygen–glucose deprivation (OGD)-induced PC12 cell injury model. Both in vivo and in vitro experiments demonstrated that GABAAR activation obviously suppressed autophagy by regulating the AMPK-mTOR pathway. Additionally, GABAAR activation inhibited apoptosis through inhibiting the Bax/Bcl-2 pathway. These data suggest that GABAAR activation exerts neuroprotective effects during cerebral ischemia through improving oxidative stress and inhibiting autophagy and apoptosis. Our findings indicate that GABAAR serves as a target for treating cerebral ischemia and highlight the GABAAR-mediated autophagy signaling pathway. [ABSTRACT FROM AUTHOR]
