The Arctic is seasonally exposed to long periods of low temperatures and complete darkness. Consequently, perennial primary producers have to apply strategies to maximize energy efficiency. Global warming is occurring in the Arctic faster than the rest of the globe. The highest amplitude of temperature rise occurs during Polar Night. To determine the stress resistance of the ecosystem-engineering kelp Laminaria digitata against Arctic winter warming, non-meristematic discs of adult sporophytes from Porsangerfjorden (Finnmark, Norway) were kept in total darkness at 0°C and 5°C over a period of three months. Physiological variables, namely maximum quantum yield of photosynthesis (Fv/Fm) and dry weight, as well as underlying biochemical variables including pigments, storage carbohydrates, total carbon and total nitrogen were monitored throughout the experiment. Although all samples remained in generally good condition with Fv/Fm values above 0.6, L. digitata performed better at 0°C than at 5°C. Depletion of metabolic products resulted in a constant decrease of dry weight over time. A strong decrease in mannitol and laminarin was observed, with greater reductions at 5°C than at 0°C. However, the total carbon content did not change, indicating that the sporophytes were not suffering from "starvation stress" during the long period of darkness. A decline was also observed in the accessory pigments and the pool of xanthophyll cycle pigments, particularly at 5°C. Our results indicate that L. digitata has a more active metabolism, but a lower physiological and biochemical performance at higher temperatures in the Arctic winter. Obviously, L. digitata is well adapted to Arctic Polar Night conditions, regardless of having its distributional center at lower latitudes. This study introduces a novel approach to analyze glycosidic linkages in unfractionated polysaccharides from alcohol-insoluble residues (AIRs) of five brown seaweed species. GC-MS analysis of partially methylated alditol acetates (PMAAs) enables monitoring and comparison of structural variations across different species, harvest years, and tissues with and without blanching treatments. The method detects a wide array of fucose linkages, highlighting the structural diversity in glycosidic linkages and sulfation position in fucose-containing sulfated polysaccharides. Additionally, this technique enhances cellulose quantitation, overcoming the limitations of traditional monosaccharide composition analysis that typically underestimates cellulose abundance due to incomplete hydrolysis of crystalline cellulose. Methods: Experimental colonization of Ross 308 broilers with two different strains of C. jejuni was conducted, with groups fed either a basal diet or the same basal diet supplemented with 725 ppm algal extract from Saccharina latissima to provide 290 ppm laminarin. Fecal samples were collected for bacterial enumeration, and caecal samples were obtained before and after the C. jejuni challenge for the determination of microbiota development. Results and discussion: No significant differences in fecal C. jejuni concentrations between the groups fed different diets or exposed to different C. jejuni strains were observed. This suggests that both strains colonized the birds equally well and that the laminarin rich algal extract did not have any inhibitory effect on C. jejuni colonization. Notably, 16S rRNA amplicon sequencing revealed detailed data on the caecal microbiota development, likely influenced by both bird age and C. jejuni colonization, which can be valuable for further development of broiler feed formulations aimed at promoting gut health. Brown seaweed polysaccharides (BSPs) are potential alternative sources of bioactive compounds. The food and pharmaceutical industries have a great interest in BSPs because of their various bioactivities, such as anticancer, antioxidant, and immunomodulatory activities. Studies have demonstrated that the bioactivities of BSPs are significantly influenced by their structural characteristics and chemical constitution; but, their specific conformational relationships are complex and required further study. However, systematic reviews of the biological activities and mechanisms of action of BSPs are currently inadequate. Thus, this paper summarizes recent studies on the physicochemical and structural features, characterization methods, biological activities, and related action mechanisms of BSPs from different sources. Based on these findings, future prospects include the following three aspects. More advanced research techniques need to be developed to identify the exact main and branch structure of BSPs; in-depth studies of the detailed structure of BSPs are needed to elucidate the conformational relationships between polysaccharides and their activities, and clinical trials should be conducted to verify the effectiveness of BSPs in the prevention and treatment of various diseases. The information provided here aims to offer a theoretical foundation for further research on the structural properties, mechanism of action, and potential applications of BSPs in food and pharmaceutical industries as well as, to increase the economic value of BSPs and extend their application range.

Laminaria digitata is a brown seaweed rich in prebiotic polysaccharides, mainly laminarin, but its alginate-rich cell wall could compromise nutrient access. Carbohydrase supplementation, such as individual alginate lyase and carbohydrases mixture (Rovabio® Excel AP), could enhance nutrient digestibility and prebiotic potential. This study aimed to evaluate the effect of these enzymes on nutrient digestibility and gut health of weaned piglets fed with 10% L. digitata. Diets did not affect growth performance (P > 0.05). The majority of the feed fractions had similar digestibility across all diets, but the supplementation of alginate lyase increased hemicellulose digestibility by 3.3% compared to the control group (P = 0.047). Multi-enzyme-assisted laminarin extraction parameters were optimized, and then the prepared laminarin was assessed for inhibition of α-glucosidase activity. The relative proportions of preparative enzymes were investigated to maximize laminarin yield using an orthogonal test design. The effect of enzymatic hydrolysis time, pH, ratio of liquid to material and temperature on laminarin yield were determined using response surface methodology (RSM). Finally, laminarin inhibition of α-glucosidase activity was assessed using an in vitro enzyme inhibition test. Results revealed that the optimal laminarin yield was obtained using a ratio by mass of 10:9:5.5 of cellulase, pectinase and papain, respectively, and an enzymatic hydrolysis time, temperature, pH and ratio of liquid to material of 1.8 h, 49.4 ℃, 6.1 and 59:1 mL/g, respectively. The predicted yield of laminarin under optimal conditions was 18.183%, which resembled the actual laminarin yield of 18.19%±1.04%. Its neutral sugar, acid sugar, protein and sulphate contents were 52.72%, 11.76%, 2.66% and 19.49%, respectively. As laminarin concentration increased from 1 mg/mL to 5 mg/mL, its α-glucosidase inhibition activity increased, with highest inhibition of 79.04%±3.17% and IC50 value of 1.443 mg/mL. This multi-enzyme-assisted extraction procedure generated a high yield of laminarin that exerted clearly demonstrable inhibition of α-glucosidase activity.

Plant resistance inducers (PRIs) are potential alternatives for controlling grapevine downy (DM) and powdery (PM) mildews in vineyards. In a 3-year field study, we evaluated the field efficacy of six commercial PRIs of chemical and natural origin against DM and PM diseases when applied at designated vine growth stages in a mixture with low doses of copper and sulfur, and only when advised by weather-driven disease models. The disease severity and incidence were evaluated for each season at key growth stages (i.e., the end of flowering, berries pea-sized, veraison, and pre-harvest), and areas under the disease progress curves (AUDPC) were calculated and compared with those of nontreated vines. These risk-based applications resulted in a 41% and 61% reduction of interventions against DM and PM, respectively, compared to the official advice for integrated pest management in the growing area. These applications provided a disease control efficacy of 88% for DM and 93% for PM; the disease severity on bunches never exceeded 5%. Overall, when the disease severity was expressed as AUDPC, we observed higher efficacy of all the PRIs for PM, and of laminarin and cerevisane for DM. We also found that potassium phosphonate and fosetyl-Al (commonly used against DM) were effective against PM, and cos-oga (used against PM) was effective against DM. Stroke is the second leading cause of death and a major cause of disability worldwide. Ischemic stroke accounts for approximately >60% of all stroke, and >80% of strokes can be prevented. Middle cerebral artery occlusion (MCAO) is a common cause of stroke in humans. MCAO is associated with blood vessel-related problems, such as reduced vascular plasticity and hypertension. These problems can be attenuated by the use of nutraceuticals such as laminarin. Laminarin is a storage glucan commonly found in brown algae. Accumulating evidence and studies have revealed that laminarin is a promising candidate drug for treating ischaemic stroke. However, details on pharmaceutical targets and molecular mechanisms underlying laminarin's beneficial effects for treating ischaemic stroke remains largely unknown. Herein, we applied network pharmacology, bioinformatic analysis, and middle cerebral artery occlusion model to delineate the protective role of laminarin. By comparing the laminarin- and MCAO-associated genes, we identified 23 potential targets including tumour necrosis factor (TNF), vascular endothelial growth factor A, selectin P, presenilin 1, fibroblast growth factor 2, microtubule-associated protein tau, caspase 3, matrix metallopeptidase 1, 5-hydroxytryptamine receptor 2A, telomerase reverse transcriptase, interleukin 2, signal transducer and activator of transcription 3 (STAT3), ATP binding cassette subfamily B member 1, catalase, superoxide dismutase 2, adenosine A1 receptor, adenosine A2a receptor, 5-hydroxytryptamine receptor 1B, heat shock protein 90 alpha family class A member 1, matrix metallopeptidase 8, BCL2-like 1, galectin 3, and epoxide hydrolase 2 of laminarin against ischaemic stroke. The gene ontology enrichment and Kyoto Encyclopedia of Genes and Genomes enrichment analyses highlighted the importance of this gene cluster in the development of blood vessels, neuronal cell death, brain functions, and neuroinflammation. Furthermore, molecular docking analysis suggested a direct binding of laminarin to its target proteins STAT3 and TNF. Our results provide the pharmaceutical targets and delineate the details regarding the molecular mechanisms underlying the beneficial effects of laminarin against ischaemic stroke. Aberrantly high dietary cholesterol intake and intestinal cholesterol uptake lead to dyslipidemia, one of the risk factors for cardiovascular diseases (CVDs). Based on previous studies, laminarin, a polysaccharide found in brown algae, has hypolipidemic activity, but its underlying mechanism has not been elucidated. In this study, we investigated the effect of laminarin on intestinal cholesterol uptake in vitro, as well as the lipid and morphological parameters in an in vivo model of high-fat diet (HFD)-fed mice, and addressed the question of whether Niemann–Pick C1-like 1 protein (NPC1L1), a key transporter mediating dietary cholesterol uptake, is involved in the mechanistic action of laminarin. In in vitro studies, BODIPY-cholesterol-labeled Caco-2 cells were examined using confocal microscopy and a fluorescence reader. The results demonstrated that laminarin inhibited cholesterol uptake into Caco-2 cells in a concentration-dependent manner (EC50 = 20.69 μM). In HFD-fed C57BL/6J mice, laminarin significantly reduced the serum levels of total cholesterol (TC), total triglycerides (TG), and low-density lipoprotein cholesterol (LDL-C). It also decreased hepatic levels of TC, TG, and total bile acids (TBA) while promoting the excretion of fecal cholesterol. Furthermore, laminarin significantly reduced local villous damage in the jejunum of HFD mice. Mechanistic studies revealed that laminarin significantly downregulated NPC1L1 protein expression in the jejunum of HFD-fed mice. The siRNA-mediated knockdown of NPC1L1 attenuated the laminarin-mediated inhibition of cholesterol uptake in Caco-2 cells. This study suggests that laminarin significantly improves dyslipidemia in HFD-fed mice, likely by reducing cholesterol uptake through a mechanism that involves the downregulation of NPC1L1 expression. Breeder nutrition is an important factor for chick quality since the chick embryo relies on nutrients available in the egg for growth and development. In addition, the egg is providing the chick with important antibodies that are vital during the first weeks of life. Brown algae contains several bioactive compounds, and dietary supplementation with algal extracts have shown improved gut health and immune responses in both pigs and poultry. The aim of this study was to investigate if feeding the brown algae Saccharina latissima, intact or as an extract, to broiler breeders can affect breeder hens’ antibody responses to vaccination, egg quality and transfer of antibodies and nutrients to the egg and thereby improve the quality of newly hatched chicks. Forty-five hens and nine roosters of the parent lines of the fast-growing broiler Ross 308 were included in the experiment where hens were 31 weeks at the start. The hens were housed individually and fed one of three dietary treatments for seven weeks; (a) control, (b) addition of 0.6% algal meal or (c) addition of 0.08% algal extract. The hens were given a booster vaccination against infectious bronchitis virus (IBV) 21 days after the start of experiment. During experimental days 32–42, hens were naturally mated every 5th day and hatching eggs were collected. A total of 255 chicks were hatched, and chick quality was assessed. Moreover, on chick day three, blood was collected from 48 focal chickens and total immunoglobulin Y levels and specific titres to IBV in serum were determined. The results showed that feeding the brown algae Saccharina latissima, intact or as an extract to broiler breeders did not affect egg production, egg quality, antibody responses to vaccination or transfer of antibodies from hen to chick. However, feeding intact algae significantly increased the levels of iodine and decreased the level of selenium in the eggs and resulted in a lower proportion of chicks with maximum quality score. Interestingly, algal feeding, both intact and as an extract, increased the abdominal fat pad in broiler breeders by about 17% without affecting BW. In conclusion, supplementation of broiler breeder diets with algal extract from Saccharina latissima, but not intact algal meal is a promising dietary strategy to increase the abdominal fat pad without causing any adverse effects on nutrient level in eggs or chick quality.

The fungus Fusarium graminearum causes Fusarium head blight (FHB), a serious disease of wheat and barley. FHB results in crop yield loss and contaminates grain with mycotoxins. In a prior study, we discovered that chitin induces tissue‐specific ROS burst in wheat. However, it is unknown whether other fungal cell wall components could induce defense response in wheat. Therefore, we evaluated ROS and defense gene responses in different wheat tissues that had been treated with chitin, laminarin, or both. Different ROS patterns were induced in wheat treated with laminarin or chitin. Furthermore, we found that ROS were enhanced in wheat tissues treated with both chitin and laminarin. The fecal microbiota profile of sows supported the improvement of reproductive performance in sows and the growth performance in suckling offspring. Conclusions: Dietary supplementation of laminarin during late pregnancy and lactation could significantly improve reproductive performance of sows and growth performance of piglets. Plant resistance inducers (PRIs) harbor promising potential for use in downy mildew (DM) control in viticulture. Here, the effects of six commercial PRIs on some epidemiological components of Plasmopara viticola (Pv) on grapevine leaves were studied over 3 years. Disease severity, mycelial colonization of leaf tissue, sporulation severity, production of sporangia on affected leaves, and per unit of DM lesion were evaluated by inoculating the leaves of PRI-treated plants at 1, 3, 6, 12, and 19 days after treatment (DAT). Laminarin, potassium phosphonate (PHO), and fosetyl-aluminium (FOS) were the most effective in reducing disease severity as well as the Pv DNA concentration of DM lesions on leaves treated and inoculated at 1 and 3 DAT; PHO and FOS also showed long-lasting effects on leaves established after treatment (inoculations at 6 to 19 DAT). PRIs also prevented the sporulation of Pv on lesions; all the PRI-treated leaves produced fewer sporangia than the nontreated control, especially in PHO-, FOS-, and cerevisane-treated leaves (>75% reduction). These results illustrate the broader and longer effect of PRIs on DM epidemics. The present study aims to formulate and evaluate the physicochemical characteristics of the resveratrol-laminarin encapsulated liposomal formulation. Resveratrol is a phytoalexin with numerous biological effects like antidiabetic, anti-oxidant, anti-cancer, and anti-inflammatory properties. A polysaccharide extracted from brown algae called laminarin possesses anti-tumor, anti-oxidant, anti-coagulant, anti-inflammatory, and antiobesity properties. Resveratrol, a hydrophobic drug, is entrapped in the liposomal bilayer, and laminarin, a hydrophilic drug, is enclosed in the hydrophilic core. The preparation and characteristics of resveratrol-laminarin-loaded liposomes, including their particle size, zeta potential, entrapment efficiency, and in-vitro release kinetics, were investigated in this study. The thin film lipid hydration method was used to prepare the liposome using soy phospholipids and cholesterol in a 6:1 ratio. Downsizing of the prepared liposomes was carried out by homogenization and sonication techniques. The liposome had a particle size of 120.7 nm after sonication, and a zeta potential of -33.4 mV was observed. The highest entrapment efficiency recorded for resveratrol is 90.69%, and 83.64% for laminarin. Biophysical characterisation of the prepared liposomes was carried out by scanning electron microscope and UV-visible spectrophotometry. The release study of resveratrol and laminarin was observed for six hours. A stable and steady release of the compounds was observed. The stability of the liposomes stored at 4°C was analyzed after three months of preparation. In conclusion, the prepared liposomal formulations exhibited good physicochemical characteristics and are of great therapeutic value in the future. [ABSTRACT FROM AUTHOR]

Brown macroalgae represent one of the most proliferative groups of living organisms in aquatic environments. Due to their abundance, they often cause problems in aquatic and terrestrial ecosystems, resulting in health problems in humans and the death of various aquatic species. To resolve this, the application of Sargassum has been sought in different research areas, such as food, pharmaceuticals, and cosmetics, since Sargassum is an easy target for study and simple to obtain. In addition, its high content of biocompounds, such as polysaccharides, phenols, and amino acids, among others, has attracted attention. One of the valuable components of brown macroalgae is their polysaccharides, which present interesting bioactivities, such as antiviral, antimicrobial, and antitumoral, among others. There is a wide variety of methods of extraction currently used to obtain these polysaccharides, such as supercritical fluid extraction (SFE), pressurized liquid extraction (PLE), subcritical water extraction (SCWE), ultrasound-assisted extraction (UAE), enzyme-assisted extraction (EAE), and microwave-assisted extraction (MAE). Therefore, this work covers the most current information on the methods of extraction, as well as the purification used to obtain a polysaccharide from Sargassum that is able to be utilized as alginates, fucoidans, and laminarins. In addition, a compilation of bioactivities involving brown algae polysaccharides in in vivo and in vitro studies is also presented, along with challenges in the research and marketing of Sargassum-based products that are commercially available. To achieve sustainable development, alternative resources should replace conventional resources such as fossil fuels. In marine ecosystems, many macroalgae grow faster than terrestrial plants. Macroalgae are roughly classified as green, red, or brown algae based on their photosynthetic pigments. Brown algae are considered to be a source of physiologically active substances such as polyphenols. Furthermore, some macroalgae can capture approximately 10 times more carbon dioxide from the atmosphere than terrestrial plants. Therefore, they have immense potential for use in the environment. Recently, macroalgae have emerged as a biomass feedstock for bioethanol production owing to their low lignin content and applicability to biorefinery processes. Herein, we provided an overview of the bioconversion of macroalgae into bioactive substances and biofuels using microbial biotechnology, including engineered yeast designed using molecular display technology. Natural plant populations are polymorphic and show intraspecific variation in resistance properties against pathogens. The activation of the underlying defence responses can depend on variation in perception of pathogen-associated molecular patterns or elicitors. To dissect such variation, we evaluated the responses induced by laminarin (a glucan, representing an elicitor from oomycetes) in the wild tomato species Solanum chilense and correlated this to observed infection frequencies of Phytophthora infestans. We measured reactive oxygen species burst and levels of diverse phytohormones upon elicitation in 83 plants originating from nine populations. We found high diversity in basal and elicitor-induced levels of each component. Further we generated linear models to explain the observed infection frequency of P. infestans. The effect of individual components differed dependent on the geographical origin of the plants. We found that the resistance in the southern coastal region, but not in the other regions, was directly correlated to ethylene responses and confirmed this positive correlation using ethylene inhibition assays. Our findings reveal high diversity in the strength of defence responses within a species and the involvement of different components with a quantitatively different contribution of individual components to resistance in geographically separated populations of a wild plant species. Background: Blooms of marine microalgae play a pivotal role in global carbon cycling. Such blooms entail successive blooms of specialized clades of planktonic bacteria that collectively remineralize gigatons of algal biomass on a global scale. This biomass is largely composed of distinct polysaccharides, and the microbial decomposition of these polysaccharides is therefore a process of prime importance. Results: In 2020, we sampled a complete biphasic spring bloom in the German Bight over a 90-day period. Bacterioplankton metagenomes from 30 time points allowed reconstruction of 251 metagenome-assembled genomes (MAGs). Corresponding metatranscriptomes highlighted 50 particularly active MAGs of the most abundant clades, including many polysaccharide degraders. Saccharide measurements together with bacterial polysaccharide utilization loci (PUL) expression data identified β-glucans (diatom laminarin) and α-glucans as the most prominent and actively metabolized dissolved polysaccharide substrates. Both substrates were consumed throughout the bloom, with α-glucan PUL expression peaking at the beginning of the second bloom phase shortly after a peak in flagellate and the nadir in bacterial total cell counts. Conclusions: We show that the amounts and composition of dissolved polysaccharides, in particular abundant storage polysaccharides, have a pronounced influence on the composition of abundant bacterioplankton members during phytoplankton blooms, some of which compete for similar polysaccharide niches. We hypothesize that besides the release of algal glycans, also recycling of bacterial glycans as a result of increased bacterial cell mortality can have a significant influence on bacterioplankton composition during phytoplankton blooms. Laminarin has antioxidant and immunomodulatory properties and favorably impacts gut microbial composition, providing a potential solution for the treatment of intestinal diseases in fish. The aim of this study was to investigate the effects of laminarin on the growth and intestinal health of juvenile spotted seabass, Lateolabrax maculatus. A total of 450 juveniles (initial body weight: 7.14 ± 0.10 g) were randomly divided into 6 groups with 3 replicates per group and 25 fish per replicate. Six diets were prepared with laminarin supplementation at doses of 0% (Control), 0.4% (P0.4), 0.8% (P0.8), 1.2% (P1.2), 1.6% (P1.6), and 2% (P2). Each group was fed the corresponding diet for 8 weeks. The basic region/leucine zipper motif (bZIP) transcription factor (TF) family is one of the most important gene families in plants, regulating the plant growth, development, reproduction, as well as stress responses. It was well known that some bZIP TFs in higher plants can regulate polysaccharide metabolism. However, bZIP TFs in microalgae and their function on the polysaccharide metabolism are still unknown. In this study, bioinformatic and molecular methods were carried out to analyze bZIP TFs in the diatom Phaeodactylum tricornutum (Pt). As a result, a total of twenty-two PtbZIP genes were identified and subsequently classified into seven distinct clades based on phylogenetic relationships, conserved motifs, and gene structures. The number of introns varied from zero to three among the PtbZIP genes. All PtbZIP genes were randomly distributed to 16 chromosomes, and each containing one or two PtbZIP domains. Among the 22 identified PtbZIP genes, PtbZIP12 and PtbZIP13 genes were identified as a putative pair of duplicated genes. Furthermore, it was observed that laminarin content increased during the day and decreased during the night. It was proposed that the 12 PtbZIPs showing the same circadian rhythms could positively affect the accumulation of laminarin, while the 10 PtbZIP genes with the opposite expression pattern might have a negative effect on the laminarin metabolism. Based on these findings, a regulatory mechanism for laminarin biosynthesis was proposed in P. tricornutum. The results of this study provide valuable information for further analysis of bZIP TFs functions and laminarin metabolism in microalgae, which may be useful for optimizing the mass production of laminarin.

Fucoidan and laminarin are polymolecular compounds that extracted from seaweed. As functional polysaccharides, they possess various biological functions such as immunomodulatory, anti-inflammatory, antiviral and antioxidant bioactivities. Fucoidan and laminarin modulate host immune responses by activating immune cells (macrophages, neutrophils, T lymphocytes) and inducing the expression of cytokines (TNFα, IL-1β, IL-6, IL-8, NF-κB). Intelectin is a novel lectin that plays important roles in bacterial agglutination and bactericidal effects by recognizing the polysaccharide components on the cell surface of pathogens, which can also enhance the phagocytosis of macrophages. The present study intends to explore the immunomodulatory effects of fucoidan, laminarin and intelectin on the macrophages of Megalobrama amblycephala. The results showed that the incubation of both fucoidan and laminarin at the concentration of 100 μg/mL for 4 h could enhance the phagocytic capacity of M. amblycephala macrophages, which also showed remodeled cytoskeleton with extended pseudopodia. In addition, fucoidan and laminarin could induce the expression of M. amblycephala intelectin (MaINTL) in the primary hepatocytes and macrophages, and the recombinant MaINTL (rMaINTL) could bind with fucoidan and laminarin. Furthermore, fucoidan and laminarin induced the expression of cytoskeletal remodeling related genes in the primary M. amblycephala macrophages, such as Rac1 and WASP, which should contribute to the enhancement of seaweed polysaccharides (fucoidan and laminarin) and MaINTL on the phagocytosis of M. amblycephala macrophages. However, even though fucoidan and laminarin could induced the expression of CXCL8, while the chemotaxis of M. amblycephala macrophages on neutrophils was not affected by them. These findings suggested that fucoidan and laminarin enhanced the phagocytic capacity of M. amblycephala macrophages via activation of MaINTL and up-regulation of the expression of downstream cytoskeletal remodeling related genes Rac1 and WASP. Therefore, fucoidan and laminarin could be used as immune enhancers to improve host immunity and disease resistance in M. amblycephala, which provided novel insights into the prevention and protection of fish bacterial diseases.

Simple Summary: Due to consumer demand for safe aquatic foods and environmental concerns in aquaculture, aquaculture production can benefit from investigating the use of immunostimulant b-glucans as feed additives. In this study, we selected laminarin as a feed additive for juvenile largemouth bass (Micropterus salmoides), and investigated the appropriate dose of supplemented laminarin in the diet based on the analysis of the growth performance, antioxidant capacity, immune response and intestinal microbiota of fish. The results indicated that supplemented laminarin in diet at a low level is suggested as a promising immunopotentiator without negative effects on growth performance for juvenile largemouth bass. A 28 day feeding trial was conducted to investigate the growth performance, immune response and intestinal microbiota of laminarin (LAM) supplemented diets in juvenile largemouth bass (Micropterus salmoides). Four hundred and eighty fish (initial average weight: 0.72 ± 0.04 g) were randomly divided into four groups (40 fish per tank with three replicates in each group) Four diets were prepared with LAM supplementation at the doses of 0 (control), 5 g Kg−1 (LL), 10 g Kg−1 (ML) and 15 g Kg−1 (HL), respectively. No significant difference in the specific growth rate (SGR) and hepatosomatic index (HSI) was observed in fish among the four groups, or in the lipid and ash content of fish flesh. In addition, fish in the LL group exhibited much higher antioxidant capacity (p < 0.05), while the diets with the inclusion of 5 and 10 g Kg−1 LAM remarkably decreased the antioxidant capacity of fish (p > 0.05). Dietary LAM at the dose of 5 g Kg−1 inhibited the transcription of interleukin-1β (il-1β) and tumor necrosis factor-α (tnf-α), while promoting the expression of transforming growth factor-β (tgf-β) in fish intestine. Moreover, the beneficial intestinal bacteria Bacteroide, Comamonas and Mycoplasma abundance significantly increased in fish from the LL group, while the content of opportunistic pathogens Plesiomonas, Aeromonas and Brevinema in fish of the HL group was substantially higher than the control group. Overall, the appropriate dose of supplemented LAM in the diet was 5 g Kg−1, while an excessive supplementation of LAM in the diet led to microbial community instability in largemouth bass. Marine-origin polysaccharides, in particular cationic and anionic ones, have been widely explored as building blocks in fully natural or hybrid electrostatic-driven Layer-by-Layer (LbL) assemblies for bioapplications. However, the low chemical versatility imparted by neutral polysaccharides has been limiting their assembly into LbL biodevices, despite their wide availability in sources such as the marine environment, easy functionality, and very appealing features for addressing multiple biomedical and biotechnological applications. In this work, we report the chemical functionalization of laminarin (LAM) and pullulan (PUL) marine polysaccharides with peptides bearing either six lysine (K6) or aspartic acid (D6) amino acids via Cu(I)-catalyzed azide-alkyne cycloaddition to synthesize positively and negatively charged polysaccharide-peptide conjugates. The successful conjugation of the peptides into the polysaccharide's backbone was confirmed by proton nuclear magnetic resonance and attenuated total reflectance Fourier-transform infrared spectroscopy, and the positive and negative charges of the LAM-K6/PUL-K6 and LAM-D6/PUL-D6 conjugates, respectively, were assessed by zeta-potential measurements. The electrostatic-driven LbL build-up of either the LAM-D6/LAM-K6 or PUL-D6/PUL-K6 multilayered thin film was monitored in situ by quartz crystal microbalance with dissipation monitoring, revealing the successful multilayered film growth and the enhanced stability of the PUL-based film. The construction of the PUL-peptide multilayered thin film was also assessed by scanning electron microscopy and its biocompatibility was demonstrated in vitro towards L929 mouse fibroblasts. The herein proposed approach could enable the inclusion of virtually any kind of small molecules in the multilayered assemblies, including bioactive moieties, and be translated into more convoluted structures of any size and geometry, thus extending the usefulness of neutral polysaccharides and opening new avenues in the biomedical field, including in controlled drug/therapeutics delivery, tissue engineering, and regenerative medicine strategies. Seaweeds (macroalgae) are gaining attention as potential sustainable feedstock for the production of fuels and chemicals. This comparative study focuses on the characterization of the microbial production of alcohols from fermentable carbohydrates in the hydrolysate of the macroalgae Laminaria digitata as raw material. The potential of a hydrolysate as a carbon source for the production of selected alcohols was tested, using three physiologically different fermentative microbes, in two main types of processes. For the production of ethanol, Saccharomyces cerevisiae was used as a benchmark microorganism and compared with the strictly anaerobic thermophile Thermoanaerobacterium strain AK17. For mixed production of acetone/isopropanol, butanol, and ethanol (A/IBE), three strictly anaerobic Clostridium strains were compared. All strains grew well on the hydrolysate, and toxicity constraints were not observed, but fermentation performance and product profiles were shown to be both condition- and strain-specific. S. cerevisiae utilized only glucose for ethanol formation, while strain AK17 utilized glucose, mannitol, and parts of the glucan oligosaccharides. The clostridia strains tested showed different nutrient requirements, and were able to utilize glucan, mannitol, and organic acids in the hydrolysate. The novelty of this study embodies the application of different inoculates for fermenting a common brown seaweed found in the northern Atlantic Ocean. It provides important information on the fermentation properties of different microorganisms and pinpoints the value of carbon source utilization when selecting microbes for efficient bioconversion into biofuel and chemical products of interest. Marine algal species comprise of a large portion of polysaccharides which have shown multifunctional properties and health benefits for treating and preventing human diseases. Laminarin, or β-glucan, a storage polysaccharide from brown algae, has been reported to have potential pharmacological properties such as antioxidant, anti-tumor, anti-coagulant, anticancer, immunomodulatory, anti-obesity, anti-diabetic, anti-inflammatory, wound healing, and neuroprotective potential. It has been widely investigated as a functional material in biomedical applications as it is biodegradable, biocompatible, and is low toxic substances. The reported preclinical and clinical studies demonstrate the potential of laminarin as natural alternative agents in biomedical and industrial applications such as nutraceuticals, pharmaceuticals, functional food, drug development/delivery, and cosmeceuticals. This review summarizes the biological activities of laminarin, including mechanisms of action, impacts on human health, and reported health benefits. Additionally, this review also provides an overview of recent advances and identifies gaps and opportunities for further research in this field. It further emphasizes the molecular characteristics and biological activities of laminarin in both preclinical and clinical settings for the prevention of the diseases and as potential therapeutic interventions. Plant resistance inducers (PRIs) are potential alternatives for controlling grapevine downy (DM) and powdery (PM) mildews in vineyards. In a 3-year field study, we evaluated the field efficacy of six commercial PRIs of chemical and natural origin against DM and PM diseases when applied at designated vine growth stages in a mixture with low doses of copper and sulfur, and only when advised by weather-driven disease models. The disease severity and incidence were evaluated for each season at key growth stages (i.e., the end of flowering, berries pea-sized, veraison, and pre-harvest), and areas under the disease progress curves (AUDPC) were calculated and compared with those of nontreated vines. These risk-based applications resulted in a 41% and 61% reduction of interventions against DM and PM, respectively, compared to the official advice for integrated pest management in the growing area. These applications provided a disease control efficacy of 88% for DM and 93% for PM; the disease severity on bunches never exceeded 5%. Overall, when the disease severity was expressed as AUDPC, we observed higher efficacy of all the PRIs for PM, and of laminarin and cerevisane for DM. We also found that potassium phosphonate and fosetyl-Al (commonly used against DM) were effective against PM, and cos-oga (used against PM) was effective against DM. These results broaden the application and integration of PRIs in viticulture.

Aberrantly high dietary cholesterol intake and intestinal cholesterol uptake lead to dyslipidemia, one of the risk factors for cardiovascular diseases (CVDs). Based on previous studies, laminarin, a polysaccharide found in brown algae, has hypolipidemic activity, but its underlying mechanism has not been elucidated. In this study, we investigated the effect of laminarin on intestinal cholesterol uptake in vitro, as well as the lipid and morphological parameters in an in vivo model of high-fat diet (HFD)-fed mice, and addressed the question of whether Niemann–Pick C1-like 1 protein (NPC1L1), a key transporter mediating dietary cholesterol uptake, is involved in the mechanistic action of laminarin. In in vitro studies, BODIPY-cholesterol-labeled Caco-2 cells were examined using confocal microscopy and a fluorescence reader. The results demonstrated that laminarin inhibited cholesterol uptake into Caco-2 cells in a concentration-dependent manner (EC50 = 20.69 μM). In HFD-fed C57BL/6J mice, laminarin significantly reduced the serum levels of total cholesterol (TC), total triglycerides (TG), and low-density lipoprotein cholesterol (LDL-C). It also decreased hepatic levels of TC, TG, and total bile acids (TBA) while promoting the excretion of fecal cholesterol. Furthermore, laminarin significantly reduced local villous damage in the jejunum of HFD mice. Mechanistic studies revealed that laminarin significantly downregulated NPC1L1 protein expression in the jejunum of HFD-fed mice. The siRNA-mediated knockdown of NPC1L1 attenuated the laminarin-mediated inhibition of cholesterol uptake in Caco-2 cells. This study suggests that laminarin significantly improves dyslipidemia in HFD-fed mice, likely by reducing cholesterol uptake through a mechanism that involves the downregulation of NPC1L1 expression.

Laminarin is a polysaccharide isolated from brown marine algae and has a wide range of bioactivities, including immunoregulatory and anti-inflammatory properties. However, the effects of laminarin on atopic dermatitis have not been demonstrated. This study investigated the potential effects of topical administration of laminarin using a Balb/c mouse model of oxazolone-induced atopic dermatitis-like skin lesions. Our results showed that topical administration of laminarin to the ear of the mice improved the severity of the dermatitis, including swelling. Histological analysis revealed that topical laminarin significantly decreased the thickening of the epidermis and dermis and the infiltration of mast cells in the skin lesion. Serum immunoglobulin E levels were also significantly decreased by topical laminarin. Additionally, topical laminarin significantly suppressed protein levels of oxazolone-induced proinflammatory cytokines, such as interleukin-1β, tumor necrosis factor-α, monocyte chemoattractant protein-1, and macrophage inflammatory protein-1α in the skin lesion. These results indicate that topical administration of laminarin can alleviate oxazolone-induced atopic dermatitis by inhibiting hyperproduction of IgE, mast cell infiltration, and expressions of proinflammatory cytokines. Pancreatic ductal adenocarcinoma (PDAC) is a notoriously aggressive type of cancer with a high metastasis rate. It is conventionally treated by surgical resection and neoadjuvant chemotherapy. However, continuous chemotherapy leads to relapse in most PDAC patients due to chemical resistance. Therefore, novel anticancer agents need to be identified and developed. The antitumor activities of laminarin extracted from brown algae against hepatocarcinoma, lung, and colon cancer have been established. However, its effects on pancreatic cancer have remained obscure. Our study identified the anticancer effects of laminarin on pancreatic cancer cells and tried to explain its intracellular mechanisms. We assessed the cell viability of PANC-1 and MIA PaCa-2 cells using MTT assay. Hanging drop method was used for the spheroid formation. Flow cytometry was conducted to evaluate the several intracellular alterations including apoptosis, ROS production, mitochondrial membrane potential (MMP), and calcium concentration induced by laminarin. An invasion test was performed to assess the inhibitory effect of laminarin on cell migration and the invasive genes were evaluated by RT-qPCR. Signaling pathway related with anticancer effects of laminarin was analyzed by western blot. We report that inhibiting laminarin increased the proliferation and viability of the representative pancreatic cancer cell lines, MIA PaCa-2 and PANC-1. Laminarin triggered apoptosis and mitochondrial impairment as evidenced by depolarized mitochondrial membranes, disrupted calcium, and suppressed cell migration caused by reactive oxygen species production and related intracellular signaling pathways. Moreover, laminarin showed synergistic effects when combined with 5-FU, a standard anticancer agent for PDAC. The present study is the first to report that laminarin exerts anticancer effect through ROS production in pancreatic cancer cells. Plant resistance inducers (PRIs) harbor promising potential for use in downy mildew (DM) control in viticulture. Here, the effects of six commercial PRIs on some epidemiological components of Plasmopara viticola (Pv) on grapevine leaves were studied over 3 years. Disease severity, mycelial colonization of leaf tissue, sporulation severity, production of sporangia on affected leaves, and per unit of DM lesion were evaluated by inoculating the leaves of PRI-treated plants at 1, 3, 6, 12, and 19 days after treatment (DAT). Laminarin, potassium phosphonate (PHO), and fosetyl-aluminium (FOS) were the most effective in reducing disease severity as well as the Pv DNA concentration of DM lesions on leaves treated and inoculated at 1 and 3 DAT; PHO and FOS also showed long-lasting effects on leaves established after treatment (inoculations at 6 to 19 DAT). PRIs also prevented the sporulation of Pv on lesions; all the PRI-treated leaves produced fewer sporangia than the nontreated control, especially in PHO-, FOS-, and cerevisane-treated leaves (>75% reduction). These results illustrate the broader and longer effect of PRIs on DM epidemics. The findings open up new perspectives for using PRIs in a defense program based on single, timely, and preventative field interventions.

Brown macroalgae represent one of the most proliferative groups of living organisms in aquatic environments. Due to their abundance, they often cause problems in aquatic and terrestrial ecosystems, resulting in health problems in humans and the death of various aquatic species. To resolve this, the application of Sargassum has been sought in different research areas, such as food, pharmaceuticals, and cosmetics, since Sargassum is an easy target for study and simple to obtain. In addition, its high content of biocompounds, such as polysaccharides, phenols, and amino acids, among others, has attracted attention. One of the valuable components of brown macroalgae is their polysaccharides, which present interesting bioactivities, such as antiviral, antimicrobial, and antitumoral, among others. There is a wide variety of methods of extraction currently used to obtain these polysaccharides, such as supercritical fluid extraction (SFE), pressurized liquid extraction (PLE), subcritical water extraction (SCWE), ultrasound-assisted extraction (UAE), enzyme-assisted extraction (EAE), and microwave-assisted extraction (MAE). Therefore, this work covers the most current information on the methods of extraction, as well as the purification used to obtain a polysaccharide from Sargassum that is able to be utilized as alginates, fucoidans, and laminarins. In addition, a compilation of bioactivities involving brown algae polysaccharides in in vivo and in vitro studies is also presented, along with challenges in the research and marketing of Sargassum-based products that are commercially available.

Until today, the coronavirus disease 2019 (COVID-19) pandemic has caused 6,043,094 deaths worldwide, and most of the mortality cases have been related to patients with long-term diseases, especially cancer. Autophagy is a cellular process for material degradation. Recently, studies demonstrated the association of autophagy with cancer development and immune disorder, suggesting autophagy as a possible target for cancer and immune therapy. Laminarin is a polysaccharide commonly found in brown algae and has been reported to have pharmaceutic roles in treating human diseases, including cancers. In the present report, we applied network pharmacology with systematic bioinformatic analysis, including gene ontology (GO) enrichment, Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis, reactome pathway analysis, and molecular docking to determine the pharmaceutic targets of laminarin against COVID-19 and cervical cancer via the autophagic process. Our results showed that the laminarin would target ten genes: CASP8, CFTR, DNMT1, HPSE, KCNH2, PIK3CA, PIK3R1, SERPINE1, TLR4, and VEGFA. The enrichment analysis suggested their involvement in cell death, immune responses, apoptosis, and viral infection. In addition, molecular docking further demonstrated the direct binding of laminarin to its target proteins, VEGFA, TLR4, CASP8, and PIK3R1. Antibody immunity has not been studied in organ transplant recipients (OTRs) with cryptococcosis. We determined serum antibody levels in OTRs: 23 cryptococcosis cases and 21 controls. Glucuronoxylomannan immunoglobulin M (IgM) and laminarin IgM were lower in cases than controls, were inversely associated with cryptococcosis status, and may hold promise as markers of cryptococcosis. Transcription factors (TFs) have an important role in the regulation of the gene expression network. The role of TFs in the immune response of freshwater crayfish is poorly understood, but leveraging the regulatory mechanisms of immune response could augment the resistance against the invasive oomycete pathogen, Aphanomyces astaci. Previous studies indicated that the TFs CCAAT/enhancer-binding protein (C/EBP) and putative Krüppel homolog-1 protein (Kr-h1) might play a role in immune and stress response of the noble crayfish (Astacus astacus). Here, we aimed to further characterise these two gene products to gain a better understanding of their evolutionary origin, domain organisation and expression patterns across different crayfish tissues. Furthermore, we conducted an immune stimulation experiment to observe the potential changes in the gene expression of C/EBP and Kr-h1 under immune challenge in different crayfish tissues. Our results showed that both C/EBP and Kr-h1 are closely related to other C/EBP s and Kr-h1 s in Malacostraca. Gene expression analysis revealed that both TFs are present in all analysed tissues, with higher expression of C/EBP in the gills and Kr-h1 in the abdominal muscle. Immune stimulation with laminarin (mimicking β-1–3-glucan in the oomycete cell wall) showed an activation of the crayfish immune system, with an overall increase in the total haemocyte count (THC) compared to untreated control and crayfish buffered saline (CBS) treatment. On the gene expression level, an up-regulation of the C/EBP gene was detected in the laminarin treated group in hepatopancreas and heart, while no changes were observed for the Kr-h1 gene. Our results indicate an early change in C/EBP expression in multiple tissues during immune stimulation and suggest its involvement in the immune response of the noble crayfish. Stroke is the second leading cause of death and a major cause of disability worldwide. Ischemic stroke caused by atherosclerosis accounts for approximately 87% of all stroke cases. Ischemic stroke is a preventable disease; therefore, a better understanding of the molecular mechanisms underlying its pathogenesis and recovery processes could provide therapeutic targets for drug development and reduce the associated mortality rate. Laminarin, a polysaccharide, is a nutraceutical that can be found in brown algae. Accumulating evidence suggests that laminarin could reduce the detrimental effects of neuroinflammation on brain damage after stroke. However, the molecular mechanism underlying its beneficial effects remains largely unknown. In the present study, we used a middle cerebral artery occlusion (MCAO) rat model and applied comparative transcriptomics to investigate the molecular targets and pathways involved in the beneficial effects of laminarin on ischemic stroke. Our results show the involvement of laminarin targets in biological processes related to blood circulation, oxygen supply, and anti-inflammatory responses in the normal brain. More importantly, laminarin treatment attenuated brain damage and neurodeficits caused by ischemic stroke. These beneficial effects are controlled by biological processes related to blood vessel development and brain cell death through the regulation of canonical pathways. Our study, for the first time, delineated the molecular mechanisms underlying the beneficial effects of laminarin on ischemic stroke prevention and recovery and provides novel therapeutic targets for drug development against ischemic stroke.

Brown seaweed polysaccharides (BSPs) are potential alternative sources of bioactive compounds. The food and pharmaceutical industries have a great interest in BSPs because of their various bioactivities, such as anticancer, antioxidant, and immunomodulatory activities. Studies have demonstrated that the bioactivities of BSPs are significantly influenced by their structural characteristics and chemical constitution; but, their specific conformational relationships are complex and required further study. However, systematic reviews of the biological activities and mechanisms of action of BSPs are currently inadequate. Thus, this paper summarizes recent studies on the physicochemical and structural features, characterization methods, biological activities, and related action mechanisms of BSPs from different sources. Based on these findings, future prospects include the following three aspects. More advanced research techniques need to be developed to identify the exact main and branch structure of BSPs; in-depth studies of the detailed structure of BSPs are needed to elucidate the conformational relationships between polysaccharides and their activities, and clinical trials should be conducted to verify the effectiveness of BSPs in the prevention and treatment of various diseases. The information provided here aims to offer a theoretical foundation for further research on the structural properties, mechanism of action, and potential applications of BSPs in food and pharmaceutical industries as well as, to increase the economic value of BSPs and extend their application range. Marine algae-based drug discovery has recently received a lot of attention. This study was conducted to extract laminarin-enriched solvent extracts from Padina tetrastromatica and Sargassum cinereum and to evaluate their anticancer activity against the HeLa cell line in vitro (MTT assay). Furthermore, their toxicity was determined through a zebra fish model study. P. tetrastromatica and S. cinereum biomasses have a higher concentration of essential biomolecules such as carbohydrates, protein, and crude fiber, as well as essential minerals (Na, Mg, K, Ca, and Fe) and secondary metabolites. Methanol extracts, in particular, contain a higher concentration of vital phytochemicals than other solvent extracts. The laminarin quantification assay states that methanol extracts of P. tetrastromatica and S. cinereum are rich in laminarin, which is primarily confirmed by FTIR analysis. In an anticancer study, laminarin-MeE from P. tetrastromatica and S. cinereum at concentrations of 750 and 1000 μg mL−1 demonstrated 100% activity against HeLa cells. The Zebra fish model-based toxicity study revealed that the laminarin-enriched MeE of P. tetrastromatica and S. cinereum is non-toxic. These findings revealed that the laminarin-enriched MeE of P. tetrastromatica and S. cinereum has significant anticancer activity without causing toxicity. • P. tetrastromatica and S. cinereum biomass contain more biomolecules. • Methanol extract of P. tetrastromatica & S. cinereum have more metabolites. • P. tetrastromatica & S. cinereum methanol extract enriched with laminarin. • FT-IR analysis revealed that these brown algae enriched with laminarin. • 750–1000 μg mL−1 dosage of laminarin-MeE of these algae showed anticancer activity. • Weaned piglets have immature digestive systems and require high−quality feedstuffs. • Seaweeds such as Laminaria digitata are a tool to deal with postweaning stress. • Enzymatic supplementation can improve digestive accessibility of nutrients. • Seaweed improved hemicellulose digestibility and gut microbiome composition. • Alginate lyase supplementation improved the digestibility of ADF. Laminaria digitata is a brown seaweed rich in prebiotic polysaccharides, mainly laminarin, but its alginate-rich cell wall could compromise nutrient access. Carbohydrase supplementation, such as individual alginate lyase and carbohydrases mixture (Rovabio® Excel AP), could enhance nutrient digestibility and prebiotic potential. This study aimed to evaluate the effect of these enzymes on nutrient digestibility and gut health of weaned piglets fed with 10% L. digitata. Diets did not affect growth performance (P > 0.05). The majority of the feed fractions had similar digestibility across all diets, but the supplementation of alginate lyase increased hemicellulose digestibility by 3.3% compared to the control group (P = 0.047). Additionally, we observed that algal zinc was more readily available compared to the control group, even without enzymatic supplementation (P < 0.001). However, the increased digestibility of some minerals, such as potassium, raises concerns about potential mineral imbalance. Seaweed groups had a higher abundance of beneficial bacteria in colon contents, such as Prevotella , Oscillospira and Catenisphaera. Furthermore, the addition of alginate lyase led to a lower pH in the colon (P < 0.001) and caecum (P < 0.001) of piglets, which is possibly a result of released fermentable laminarin, and is consistent with the higher proportion of butyric acid found in these intestinal compartments. L. digitata is a putative supplement to enhance piglet gut health due to its prebiotic polysaccharides. Alginate lyase supplementation further improves nutrient digestibility and prebiotic potential. Enriching foods with long‐chain n‐3 polyunsaturated fatty acids requires a delivery emulsion system, which is both thermodynamically and oxidatively stable. The antioxidant and stabilizing properties of three types of polysaccharide extracts from brown algae Saccharina latissima with a mixed composition of polysaccharides (SA: 98% sodium alginate, SF: 90% alginate and 9% fucoidan, SL: 14.5% fucoidan, 9.5% laminarin, and co‐extracted nonpolysaccharides) were evaluated. SA, SF, SL, and REF (added commercial sodium alginate) showed in vitro ferrous ion chelating ability in the order: SA (99%) > SL (78%) > REF (31%) > SF (16%). The difference in antioxidant activity between SA, REF, and SF appeared related to structural differences of alginate (M/G ratio). A storage trial was conducted using 70% (w/w) fish oil‐in‐water delivery emulsions added sodium caseinate (NaCas) (0.23 wt%) as emulsifier in combination with SA, SF, SL, or commercial NaAlg (REF) in different concentrations (C1 = 0.1, C2 = 0.2, C3 = 0.3, and C4 = 0.4 wt%). A control with only NaCas was included (CON). The physical (e.g., creaming and droplet‐size distribution) and oxidative (peroxide value and volatiles) stability of the emulsions, were evaluated (12 days, dark at 20˚C). Acceptable physical stability (creaming index) was found for REF (all concentrations), SF = 0.2 wt% (C2), SL and SA = 0.3 wt% (C3) and 0.4 wt% (C4). In general, the oxidative stability decreased by adding REF, SA, and SF (except for REF at C1), as prooxidant activity was observed. However, SA showed antioxidant activity against the formation of 2‐ethylfuran. A diverse range of molecules with anticancer, antiviral, and antibiotic activities has been identified and isolated from algal species. Such metabolites could address unmet clinical needs and be used to fight human diseases. Further, the natural compounds derived from algae present several advantages in terms of sustainable supply, low cytotoxicity and low cost. Seaweeds represent one of the richest known sources of natural antivirals, as well as a significant source of other bioactive compounds. Increasing evidence has demonstrated that algae comprise a vast untapped reservoir of pharmaceutical potential with many underexplored species and underexploited drug candidates, which may be derived from these ‘plants of the sea’.In recent decades, new therapeutic lead compounds including polysaccharides, lectins, terpenes, alkaloids, carotenoids, and sterols, with a broad spectrum of antiviral activities, have been isolated from marine macro- and micro-algae. The current high volume of research studies exploring the antiviral potency of seaweeds epitomises the increased interest in this field. This review focuses on bioactive marine macromolecules with antiviral activities which may serve as a starting point for enhanced knowledge and a stimulus for wider research in this field. The review also seeks to improve the understanding of the antiviral mechanisms and complex structures of algal-derived macromolecules. Since the purification of such structurally complex and frequently heterogeneous extracts and molecular classes remain fraught with difficulty, efforts to decipher the structure and composition of these macromolecules would offer enhanced possibilities for future scientific and pharmaceutical advances.

Hatching concepts such as on-farm hatching provide an opportunity to supply newly hatched chickens with optimal nutrition that support growth and development of a healthy gut. Brown algae contain bioactive compounds, especially laminarin and fucoidan that may improve intestinal health and immune responses. This study aimed to examine the effects of early access to feed and water posthatch and feed supplementation with algal extract rich in laminarin from Laminaria digitata, on growth performance, organ and microbiota development and antibody production. A total of 432 Ross 308 chicks were allotted to 36 rearing pens in a 2 × 3 factorial design with two hatching treatments and three dietary treatments. During chick placement, half of the pens were directly provided access to feed and water (Early) while half of the pens were deprived of feed and water for 38 h (Late). The chicks were fed three different starter diets until day 6; a wheat-soybean meal-based control diet, a diet with low inclusion of algal extract (0.057%) and a diet with high inclusion of algal extract (0.114%). Feed intake and BW were registered on pen basis at placement, days 1, 6, 12, 19, 26, 33 and 40. To induce antibody responses, all chicks were vaccinated against avian pneumovirus on day 10. Three chicks per pen were selected as focal animals and used for blood sampling on days 10 and 39. On days 6, 19, and 40, two birds per pen were killed and used for organ measurement and caecal digesta sampling for gut microbiota analysis using the Illumina Miseq PE 250 sequencing platform. Results showed that algal extract did not influence gut microbiota, gut development or vaccine-induced antibody responses. However, during the first 38 h, early-fed chicks consumed on average 19.6 g of feed and gained 27% in BW, while late-fed chicks lost 9.1% in BW which lowered BW and feed intake throughout the study (P

The aim of the study was to investigate the effects of laminarin on natural killer (NK) cell cytotoxicity of immunosuppressive mice and its mechanism. Cyclophosphamide (cy) was used to make an immunosuppressive model of mice. The mice of two groups were given interventions by gavage with laminarin 500 mg/kg and 1000 mg/kg every day for 10 days. MACS was adopted to isolate spleen NK cells, and cytotoxicity of NK cells and IL-12, IFN- level in serum were detected in vivo . Cytotoxicity of NK92-MI cells, activating receptors (NKp30, NKp44, NKp46 and NKG2D) and perforin and granzyme B expression were detected in vitro . Compared to the normal control group, the cytotoxicity of NK cells, IL-12 and IFN- level in serum in the cy model group were all reduced significantly (p < 0.01). Compared to the cy model group, laminarin increased the cytotoxicity of NK cells, IL-12 and IFN- levels in serum significantly (p < 0.05). In vitro, laminarin increased the cytotoxicity, NKp30 and NKG2D, perforin and granzyme B expressions of NK92-MI cells (p < 0.01). This research showed that laminarin can promote NK cell cytotoxicity in immunosuppressive mice by increasing the levels of IL-12 and IFN- in serum and expressions of NKp30 and NKG2D, perforin and granzyme B.

Marine-origin polysaccharides, in particular cationic and anionic ones, have been widely explored as building blocks in fully natural or hybrid electrostatic-driven Layer-by-Layer (LbL) assemblies for bioapplications. However, the low chemical versatility imparted by neutral polysaccharides has been limiting their assembly into LbL biodevices, despite their wide availability in sources such as the marine environment, easy functionality, and very appealing features for addressing multiple biomedical and biotechnological applications. In this work, we report the chemical functionalization of laminarin (LAM) and pullulan (PUL) marine polysaccharides with peptides bearing either six lysine (K6) or aspartic acid (D6) amino acids via Cu(I)-catalyzed azide-alkyne cycloaddition to synthesize positively and negatively charged polysaccharide-peptide conjugates. The successful conjugation of the peptides into the polysaccharide’s backbone was confirmed by proton nuclear magnetic resonance and attenuated total reflectance Fourier-transform infrared spectroscopy, and the positive and negative charges of the LAM-K6/PUL-K6 and LAM-D6/PUL-D6 conjugates, respectively, were assessed by zeta-potential measurements. The electrostatic-driven LbL build-up of either the LAM-D6/LAM-K6 or PUL-D6/PUL-K6 multilayered thin film was monitored in situ by quartz crystal microbalance with dissipation monitoring, revealing the successful multilayered film growth and the enhanced stability of the PUL-based film. The construction of the PUL-peptide multilayered thin film was also assessed by scanning electron microscopy and its biocompatibility was demonstrated in vitro towards L929 mouse fibroblasts. The herein proposed approach could enable the inclusion of virtually any kind of small molecules in the multilayered assemblies, including bioactive moieties, and be translated into more convoluted structures of any size and geometry, thus extending the usefulness of neutral polysaccharides and opening new avenues in the biomedical field, including in controlled drug/therapeutics delivery, tissue engineering, and regenerative medicine strategies.
