Background. Fresh pasta filata cheese is considered as one of the most important foods in the Venezuelan diet. It is typically produced by small-scale producers using raw milk. The objective of this research was to molecularly characterize the pathogenic potential of Escherichia coli strains isolated from pasta filata cheese manufactured and marketed in Venezuela. Methodology. In the period between January and March of 2019, a total of 36 strains of E. coli were isolated from a variety of pasta filata cheeses including 17 samples of mozzarella, 16 of telita, and 3 of guayanés. These strains were isolated according to the Venezuelan Commission of Industrial Standards (COVENIN) and identified by conventional methods (biochemical and phenotypic tests). Antimicrobial susceptibility was determined using the disk diffusion technique. Phylogenetic grouping and detection of virulence genes were performed by Polymerase Chain Reaction amplification. Diversity and genetic relationships were determined by Rep-PCR. Results: All strains were susceptible to the tested antibiotics. Phylogroup A (n=19) was the most frequent (52.8%), followed by groups D (n=11; 30.6%), and B1 (n=2; 5.6%). The majority of isolates carried at least two virulence genes, one coding for adhesion mechanisms (fimH) and the other for iron uptake (fyuA). Only one strain of phylogroup A presented a profile consisting of four virulence genes (fimH, fyuA, kpsMT II, and papAH). Four strains that could not be classified according to Clermont's scheme carried resistance genes as well. A heterogeneous population structure was observed by Rep-PCR of the strains. Conclusion: Results support the hypothesis that the E. coli strains isolated from the three types of pasta filata cheeses manufactured and marketed in Venezuela have identical characteristics and virulence factors to Extraintestinal Pathogenic E. coli strains observed in animals and humans, posing a potential health risk. Therefore, it is essential to improve hygienic and sanitary controls at all stages of cheese production and to implement measures for epidemiological surveillance of potentially pathogenic bacterial strains present in Venezuelan, artisanal pasta filata cheeses. An experimental demonstration for generating harmonics in the optical Vernier effect (OVE) by cascading tapered single-mode fibres (SMF28) is presented. The tapered optical fibres conformed to a pair of in-series Mach-Zehnder fibre interferometers (MZFI), in which one MZFI was a reference while the other was a sensor. The first-order harmonic was achieved by increasing the distance between tapers of the sensor MZFI by a multiple of (i + 1) times the distance between tapers of the reference MZFI plus a mismatch factor. A series of experiments using an individual MZFI and two MZFIs with traditional and harmonic OVE as displacement sensors were performed to compare their sensitivities and magnification factors and demonstrate the improving the sensitivity. The displacement sensor with the first-order harmonic exhibited a maximum sensitivity of −33.3 nm/mm with an M-factor of 8.53 in the mm range.

En este trabajo se hace un análisis de las diferentes fallas que se pueden presentar en los instrumentos de un proceso químico (válvula y transmisor). El método propuesto detecta y diagnostica el instrumento con problemas en el proceso. Un modelo de un proceso químico es usado para ver cómo trabaja el método propuesto, para diferenciar una falla de válvula de una falla de transmisor, se utiliza un análisis en el dominio wavelet; luego se mide la diferencia entre las fallas utilizando técnicas de reconocimiento de patrones, en este caso Coeficiente de Correlación y Distancia Euclidiana.

En este trabajo se presentan los aspectos referentes al modelado de flujo en tuberías para detectar y localizar fugas, con un enfoque basado en observadores estado. El modelo de tubería está basado en las ecuaciones del Golpe de Ariete, discretizadas por diferencias finitas. Este requiere dos condiciones de frontera: presión o flujo a la entrada y salida de la tubería. Para mejorar el modelo, se modificó la forma de considerar el efecto de la fricción y la forma de introducir las condiciones de frontera. Se muestra, con simulaciones comparadas con datos reales, la importancia de considerar la variación del coeficiente de fricción, y además que la correcta selección de las condiciones de frontera, incluyendo las ecuaciones de la bomba y restricciones de flujo, permiten una mejor representación del comportamiento dinámico del flujo. Igualmente, se muestra que el modelo puede utilizarse para estimar el flujo de fuga y su posición utilizando observadores de estado, y se toma como ejemplo el filtro tipo Kalman Extendido. Se observó adicionalmente que, con el modelo obtenido, se puede trabajar con solo dos variables medidas: presión de salida y flujo de entrada, en lugar de las cuatro consideradas en trabajos anteriores.

Three-dimensional (3D) data from airborne laser scanning (ALS) and, more recently, digital aerial photogrammetry (DAP) have been successfully used to model forest attributes. While multi-temporal, wall-to-wall ALS data is not usually available, aerial imagery is regularly acquired in many regions. Thus, the combination of ALS and DAP data provide a sufficient temporal resolution to properly monitor forests. However, field data is needed to fit new forest attribute models for each 3D data acquisition, which is not always affordable. In this study, we examined whether transferability of growing stock volume (GSV) models may provide an improvement in the efficiency of forest inventories updating. We used two available ALS datasets acquired with different characteristics in 2009 and 2010, respectively, generated two DAP point clouds from imagery collected in 2010 and 2017, and utilized field data from two ground surveys conducted in 2009 and 2016-2017. We first analyzed the stability of point cloud derived metrics. Then, Support Vector Regression models based on the most stable metrics were fitted to assess model transferability by applying them to other datasets in four different cases: (1) ALS-ALS, (2) DAP-DAP temporal, (3) ALS-DAP and (4) ALS-DAP temporal. Some metrics were found to be enough stable in each case, so they could be used interchangeably between datasets. The application of models to other datasets resulted in unbiased predictions with relative root mean square error differences ranging from -8.27% to 14.59%. Results demonstrated that 3D-based GSV models may be transferable between point clouds of the same type as well as point clouds acquired using different technologies such as ALS and DAP, suggesting that DAP data may be used as a cost-efficient source of information for updating ALS-assisted forest inventories.

Se presenta un Trabajo de Investigación, dentro del contexto de “Profesiones Asistenciales”, donde estudiamos el Estrés Laboral en su fase más avanzada y más concretamente los síndromes de Burnout (profesional quemado) y Mobbing (acoso moral en trabajo). El objetivo general es determinar el nivel del constructo Estrés Laboral, a través del estudio de estos síndromes, detectándose una alta incidencia del Síndrome de Burnout entre los docentes de secundaria. El instrumento utilizado, para realizar esta investigación, ha consistido en un protocolo de recogida de información autoadministrado que consta de: Registro de variables personales y de condiciones de trabajo; Inventario de Burnout de Maslach (MBI) e Inventario de terror psicológico de Leymann (LIPT). Finalmente se obtiene una alta incidencia de burnout, no se detecta un alto porcentaje de mobbing, no podemos descartar una posible relación entre burnout y mobbing y se establece el perfil de profesorado burnout.

Anastrepha ludens is a pestiferous tephritid fly species exhibiting extreme polyphagy. It develops optimally in hosts rich in sugar but low nitrogen content. We studied the geographical influence on the composition of A. ludens 's larval and newly emerged adult gut microbiota in altitudinal (0-2,000 masl) and latitudinal (ca. 800 km from 17° to 22°N latitude) transects along the coastline of the state of Veracruz, Mexico. In the 16 collection sites, we only collected Citrus x aurantium fruit (238 samples of A. ludens larvae and adults, plus 73 samples of pulp) to control for host effect, hypothesizing that there exists a conserved core microbiota that would be dominated by nitrogen-fixing bacteria. We found that latitude triggered more significant changes in the gut microbiota than altitude. Northern and southernmost samples differed the most in microbiota composition, with a trade-off between Acetobacteraceae and Rhizobiaceae driving these differences. As hypothesized, the core microbiota in each sampling site, contained the functional group of nitrogen-fixing bacteria. We conclude that A. ludens larvae can acquire multiple diazotrophic symbionts along its wide distribution range where it infests fruit with a high C:N ratio in the pulp., Competing Interests: The authors declare that they have no competing interests., (© 2024 Aluja et al.)

Introduction: Artisanal raw milk cheese can be an important source of bacteria potentially harmful to humans, such as pathogenic Escherichia coli. The objective of this study was to determine the genetic diversity, pathogenicity, and antimicrobial resistance of 45 E. coli strains isolated from artisanal raw milk cheese in Venezuela., Methods: These strains were isolated according to the procedures established by the Venezuelan Commission of Industrial Standards (COVENIN) and identified by conventional methods. Antimicrobial resistance was determined by the disk diffusion method, while phylogenetic grouping and detection of 6 virulence genes (fimH, kpsMT II, papAH, PAI, fyuA and usp) were performed by PCR amplification. Strain typing was performed by Rep-PCR., Results: Of the 45 isolates, 73.3% were susceptible, while 26.7% were resistant to at least one of the tested antibiotics. Phylogenetic grouping revealed a relatively homogeneous distribution. Phylogenetic group A dominated in 82.2% of the strains followed by B1 and D (8.9% each). Three major virulence factors, fimH, fyuA, and kpsMTII, were genetically encoded in most strains. Rep-PCR typing of E. coli strains revealed a heterogeneous population structure., Conclusions: E. coli isolated from artisanal dairy products share characteristics and virulence genes with extraintestinal pathogenic E. coli (ExPEC) strains from animals and humans, which represents a public health risk. Anastrepha ludens (Mexican fruit-fly) is a highly polyphagous fruit fly species (Tephritidae) attacking wild and commercial fruit from Mexico to Panama. Here we report on a recent habitat and host range expansion as A. ludens lately started to attack apples (Malus domestica) in Mexico, a phenomenon likely influenced by global warming. We document natural infestations in apple-growing regions in the States of Nuevo León and Hidalgo, Mexico where A. ludens has started to attack the cultivars 'Golden Delicious', 'Rayada' and 'Criolla'. No infestations were found in the apple-growing region of Zacatlán, Puebla. To determine apple cultivar susceptibility to the attack of this emerging pest, we ran forced infestation assays in enclosed fruit-bearing branches in all three apple-growing regions and studied the metabolome of all fruit. A clear pattern emerged indicating that the cultivar 'Golden Delicious' was the most susceptible, with 'Criolla' exhibiting complete resistance in one location (Puebla). Although A. ludens can develop in this new host, development rates (egg-adult) and adult emergence were affected when compared with the performance in the natural host 'Marsh' grapefruit. Warmer temperatures and specific secondary metabolites of some apple cultivars are likely contributing to the territorial and host expansion of A. ludens., Competing Interests: Competing interests The authors declare no competing interests. Background: In the Americas, deaths by diseases avoidable with vaccines are a significant contributor to child mortality. An essential means of reducing this is through broad vaccine coverage. The COVID-19 pandemic has posed a potential disruption to vaccine coverage due to its effects on the healthcare system., Objectives: this study aims to evaluate the impact of the COVID-19 pandemic on DTP3 vaccination coverage in the Americas, investigating trends from 2012 to 2022 to identify significant changes, regional disparities, and the overall effect of the pandemic on progress towards global immunization targets., Methods: This study used the coverage data for the third dose of the diphtheria, tetanus, and pertussis vaccine (DTP3) pulled from UNICEF databases spanning 2012 to 2022. We conducted a Joinpoint regression to identify points of significant trend changes. The annual percentage change (APC) and 95% confidence intervals (95% CIs) were calculated for America and its regions. We also used segmented regression analysis. Using the Chi-square test, we compared DTP3 vaccination coverage for each country between 2019 and 2022., Results: Overall, America saw a decrease in vaccine coverage during this period, with an APC of -1.4 (95% CI -1.8; -1.0). This trend varied across regions. In North America, the decrease was negligible (-0.1% APC). South America showed the steepest decrease, with an APC of -2.5%. Central America also declined, with an APC of -1.3%. Our findings suggest a concerning trend of declining DTP-vaccination rates in the Americas, exacerbated in certain regions, in the wake of the COVID-19 pandemic. The absolute decrease in vaccine coverage in the Americas was -4% between 2019 and 2022, with the most important drop being in Central America (-7%). However, six countries reported increased vaccination rates post-COVID-19, led by Brazil, with a 7% increase. Conversely, twenty-two countries registered a decline in DTP3 vaccine coverage, with the average decrease being -7.37%. This decline poses an important challenge to achieving the WHO's target of 90% coverage for the third dose of DTP by 2030, as evidenced by the reduction in the number of countries meeting this target from 2019 to 2022., Conclusions: The COVID-19 pandemic has impacted vaccine coverage in America, leading to a decrease, especially across Central America.

Research on larval rearing and nutrition of tephritid flies on artificial diets is key for the sterile insect technique. Here, we examined the effects of the type of gel (calcium alginate, agar, or carrageenan), at varying percentages in artificial diets for the polyphagous pest Anastrepha ludens , on the physicochemical and nutritional traits of the diets, and the effects of the type of gel, the gel content and the larval density (larvae/g of diet) used in production, quality parameters for mass-reared tephritids, diet removal (an indirect estimation of diet consumption), and nutritional traits of flies. Regardless of the gel content, calcium alginate diets were firmer and more resistant to penetration than the agar and carrageenan diets. The larval recovery, pupation, pupal weight, and flight ability of A. ludens were lower in calcium alginate diets than in agar and carrageenan diets. Diet removal was higher in calcium alginate diets; however, low levels of ammonium and high levels of uric acid in excretions from larvae on these diets suggest an alteration in protein metabolism. The firmness and penetration resistance characteristics of calcium alginate diets may have limited movement and feeding of larvae, but this could be overcome by the collective feeding of large groups of larvae. Our findings provide insights into the mechanism governing gel-diet rearing systems for A. ludens .

Using light, transmission, scanning electron, and confocal microscopy, we carried out a morphological study of antennal sensilla and their ultrastructures of the Mexican Fruit Fly Anastrepha ludens (Loew), an economically important species that is a pest of mangos and citrus in Mexico and Central America. Our goal was to update the known information on the various sensilla in the antennae of A. ludens , involved in the perception of odors, temperature, humidity, and movement. Based on their external shape, size, cuticle-thickness, and presence of pores, we identified six types of sensilla with 16 subtypes (one chaetica in the pedicel, four clavate, two trichoid, four basiconic, one styloconic, and one campaniform-like in the flagellum, and three additional ones in the two chambers of the sensory pit (pit-basiconic I and II, and pit-styloconic)), some of them described for the first time in A. ludens . We also report, for the first time, two types of pores in the sensilla (hourglass and wedge shapes) that helped classify the sensilla. Additionally, we report a campaniform-like sensillum only observed by transmission electronic microscopy on the flagellum, styloconic and basiconic variants inside the sensory pit, and an "hourglass-shaped" pore in six sensilla types. We discuss and suggest the possible function of each sensillum according to their characteristics and unify previously used criteria in the only previous study on the topic.

Anastrepha ludens is a polyphagous frugivorous tephritid that infests citrus and mango. Here, we report the establishment of a laboratory colony of A. ludens reared on a larval medium that is a waste for the citrus industry, specifically, orange ( Citrus × sinensis ) fruit bagasse. After 24 generations of rearing on a nutritionally poor orange bagasse diet, pupae weighed 41.1% less than pupae from a colony reared on a nutritionally rich artificial diet. Larvae from the orange bagasse diet had 6.94% less protein content than larvae from the artificial diet, although their pupation rate was similar. Males from the orange bagasse diet produced a scent bouquet with 21 chemical compounds and were sexually competitive, but they had significantly shorter copulations when compared to males from the artificial diet and from the wild host, Casimiroa edulis , which had relatively simple scent bouquets. The chemical complexity in the odors of males from the orange bagasse diet might initially have attracted females to novel scent combinations, but, once in the copula, they may have been able to sense negative characteristics in males, leading them to terminate copulations soon after they began. We conclude that A. ludens can adjust morphological, life history, nutritional, and chemical traits when adapted to a larval environment consisting of fruit bagasse.

Climate change, particularly global warming, is disturbing biological processes in unexpected ways and forcing us to re-study/reanalyze the effects of varying temperatures, among them extreme ones, on insect functional traits such as lifespan and fecundity/fertility. Here we experimentally tested, under both laboratory and field conditions, the effects of an extreme range of temperatures (5, 10, 15, 20, 30, 40, and 45 °C, and the naturally varying conditions experienced in the field), on survivorship/lifespan, fecundity, and fertility of four pestiferous fruit fly species exhibiting contrasting life histories and belonging to two phylogenetic groups within the genus Anastrepha : A. ludens, A. obliqua, A. striata , and A. serpentina . In the field, we also measured the length of the entire life cycle (egg to adult), and in one species ( A. ludens ), the effect on the latter of the host plant (mango and grapefruit). Under laboratory conditions, none of the adults, independent of species, could survive a single day when exposed to a constant temperature of 45 °C, but A. striata and A. serpentina females/males survived at the highly contrasting temperatures of 5 and 40 °C at least 7 days. Maximum longevity was achieved in all species at 15 °C (375, 225, 175 and 160 days in A. ludens, A. serpentina, A. striata and A. obliqua females, respectively). Anastrepha ludens layed many eggs until late in life (368 days) at 15 °C, but none eclosed. Eclosion was only observed in all species at 20 and 30 °C. Under natural conditions, flies lived ca. 100 days less than in the laboratory at 15 °C, likely due to the physiological cost of dealing with the highly varying environmental patterns over 24 h (minimum and maximum temperatures and relative humidity of ca. 10-40 °C, and 22-100%, respectively). In the case of A. ludens , the immature's developmental time was shorter in mango, but adult survival was longer than in grapefruit. Chemical ecology studies on ambrosia beetles are typically conducted with either wild or laboratory-reared specimens. Unlike laboratory-reared insects, important aspects that potentially influence behavioral responses, such as age, physiological state, and prior experience are unknown in wild specimens. In this study, we compared the electroantennographic (EAG) responses of laboratory-reared and wild X. affinis and X. ferrugineus to 70% ethanol and bark odors (host kairomones) of Bursera simaruba , Mangifera indica , and Persea schiedeana aged for 2, 24, and 48 h. Chemical analyses of each odor treatment (bark species x length of aging) were performed to determine their volatilome composition. EAG responses were different between laboratory-reared and wild X. ferrugineus when exposed to ethanol, whereas wild X. affinis exhibited similar EAG responses to the laboratory-reared insects. Ethanol elicited the strongest olfactory responses in both species. Among the bark-odors, the highest responses were triggered by B. simaruba at 48 h in X. affinis , and P. schiedeana at 24 and 48 h in X. ferrugineus . Volatile profiles varied among aged bark samples; 3-carene and limonene were predominant in B. simaruba , whereas α-copaene and α-cubebene were abundant in P. schiedeana. Further studies are needed to determine the biological function of B. simaruba and P. schiedeana terpenes on X. affinis and X. ferrugineus, and their potential application for the development of effective lures.

With the aim of identifying key factors that determine oviposition decisions by Anastrepha obliqua for management purposes, we conducted a behavioral study under natural/semi-natural field conditions to identify where exactly in the fruit (upper, middle, or lower sections) females preferred to lay eggs in a highly susceptible mango cultivar ("Criollo"), and whether sunlight incidence and fruit chemical compounds influenced oviposition site selection by this pestiferous fly. Females oviposited in shaded, upper fruit sections where pulp had higher total carbohydrate concentrations but similar total protein, lipid, and polyphenol concentrations than non-oviposited sections. Peel had higher overall nutrient and mangiferin/quercetin-3-D-galactoside (polyphenols) concentrations. An untargeted metabolomic analysis of oviposited and non-oviposited fruit sections identified abscisic acid (ABA) and dihydrophaseic acid glucoside, a by-product of ABA catabolism, as potential chemical markers that could play a role in fruit acceptance behaviors by female flies. We conclude that females preferentially oviposit in fruit sections with optimal chemical and environmental conditions for larval development: more carbohydrates and antioxidants such as mangiferin and ferulic acid and lesser sunlight exposure to avoid lethal egg/larval desiccation/overheating. We make specific recommendations for A . obliqua management based on female host selection behavior, a tree pruning scheme exposing fruit to direct sunlight, application of a host marking pheromone, and the use of egg sinks in the orchard.

Background: Baricitinib is a Janus kinase (JAK) inhibitor with a broader anti-inflammatory activity than tocilizumab and an antiviral potential although no head-to-head trials are available. The benefits of adding baricitinib to patients with COVID-19 experiencing clinical progression despite the standard of care (SOC), including corticosteroids and tocilizumab, are also unknown. Methods: A cohort study included microbiologically confirmed COVID-19 hospitalizations. The primary outcome was 28-day mortality. Secondary outcomes were 60- and 90-day mortality, the composite outcome "28-day invasive mechanical ventilation (IMV) or death" and the safety of the combination. Propensity score (PS) matching was used to identify the association between baricitinib use and the outcomes of interest. Results: Of 1,709 admissions, 994 patients received corticosteroids and tocilizumab and 110 of them received baricitinib after tocilizumab. PS matched 190 (95:95) patients with baricitinib + SOC vs. SOC, of whom 69.5% received remdesivir. No significant effect of baricitinib was observed on 28-day [39 events; adjusted hazard ratio (aHR), 0.76; 95% CI, 0.31-1.86], 60-day (49 events, aHR, 1.17; 95% CI, 0.55-2.52), or 90-day mortality (49 events; aHR, 1.14; 95% CI, 0.53-2.47), or on the composite outcome 28-day IMV/death (aHR, 0.88; 95% CI, 0.45-1.72). Secondary infections during hospitalization were not different between groups (17.9 vs. 10.5%, respectively; p = 0.212) and thromboembolic events were higher with baricitinib (11.6% vs. 3.2%; p = 0.048), but differences vanished after the adjustment [aHR 1.89 (0.31-11.57), p = 0.490]. Conclusion: The addition of baricitinib did not substantially reduce mortality in hospitalized patients with COVID-19 having clinical progression despite the therapy with tocilizumab and corticosteroids. We studied the microbiota of a highly polyphagous insect, Anastrepha ludens (Diptera: Tephritidae), developing in six of its hosts, including two ancestral ( Casimiroa edulis and C. greggii ), three exotic ( Mangifera indica cv. Ataulfo, Prunus persica cv. Criollo, and Citrus x aurantium ) and one occasional host ( Capsicum pubescens cv. Manzano), that is only used when extreme drought conditions limit fruiting by the common hosts. One of the exotic hosts ("criollo" peach) is rife with polyphenols and the occasional host with capsaicinoids exerting high fitness costs on the larvae. We pursued the following questions: (1) How is the microbial composition of the larval food related to the composition of the larval and adult microbiota, and what does this tell us about transience and stability of this species' gut microbiota? (2) How does metamorphosis affect the adult microbiota? We surveyed the microbiota of the pulp of each host fruit, as well as the gut microbiota of larvae and adult flies and found that the gut of A. ludens larvae lacks a stable microbiota, since it was invariably associated with the composition of the pulp microbiota of the host plant species studied and was also different from the microbiota of adult flies indicating that metamorphosis filters out much of the microbiota present in larvae. The microbiota of adult males and females was similar between them, independent of host plant and was dominated by bacteria within the Enterobacteriaceae. We found that in the case of the "toxic" occasional host C. pubescens the microbiota is enriched in potentially deleterious genera that were much less abundant in the other hosts. In contrast, the pulp of the ancestral host C. edulis is enriched in several bacterial groups that can be beneficial for larval development. We also report for the first time the presence of bacteria within the Arcobacteraceae family in the gut microbiota of A. ludens stemming from C. edulis . Adequate pupation substrates and substrate volume are critical factors in the mass-rearing of insects for Sterile Insect Technique (SIT) applications. To identify an ideal pupation substrate for a reproductive colony of Ceratitis capitata (Wiedemann) VIENNA 8 genetic sexing strain, we first examined pupation in cellulose from recycled paper (cellulose I), sawdust, fine wheat bran, vermiculite and coconut fiber using a volume of 2.5-12.5 mL of substrate for each 5 mL volume of fly larvae. We found a positive relationship between substrate volume and pupation, with cellulose I generating the highest proportions of pupation and coconut fiber the lowest. Higher proportions of female flies (white pupae) pupated in sawdust. The proportion of female fliers increased as substrate volume rose in sawdust and coconut fiber, whereas it decreased in vermiculite and cellulose. In a second experiment, we tested three types of cellulose differing in physicochemical characteristics (celluloses I, II and III), sawdust, and fine wheat bran using a substrate:larvae ratio of 1:1. The three types of cellulose produced the highest pupation levels. The highest proportions of female fliers were observed in sawdust, and cellulose types III and II. Cellulose III and sawdust at relatively low volumes were more cost-effective to produce one million pupae than other substrates, including fine wheat bran used in a mass-rearing facility in Mexico.

Background: The interdependencies of viral replication and the host immune response in patients with coronavirus disease 2019 (COVID-19) remain to be defined. We investigated the viral determinants of antibody response, the predictors of nonseroconversion, and the role of antibodies on viral dynamics., Methods: This was a prospective study in patients hospitalized with COVID-19 that was microbiologically confirmed by real-time polymerase chain reaction (RT-PCR). Serial nasopharyngeal and oropharyngeal swabs and plasma samples were obtained for measuring severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) RNA and antibodies (total and S-IgG/N-IgG), respectively., Results: Of 132 patients included, 99 (75%) showed positive antibody titers after a median (Q1-Q3) of 11 (8-14) days. The median (Q1-Q3) follow-up was 74.5 (63.0-87.0) days. In an adjusted linear regression model, time to seropositivity was inversely associated with peak log SARS-CoV-2 viral load ( P = .009) and positively with time to viral clearance ( P = .004). Adjusted predictors of S-IgG levels were time to viral clearance ( P < .001), bilateral lung infiltrates on admission ( P = .011), and the time-dependent SARS-CoV-2 RNA ( P < .001) and SARS-CoV-2 RNA area under the curve ( P = .001). Thirty-three (25%) patients showed undetectable antibody titers. Patients who did not seroconvert had higher cycle threshold values of RT-PCR (38.0 vs 28.0; P < .001), had shorter time to viral clearance (3.0 vs 41.0; P < .001), and were more likely to have SARS-CoV-2 only detected on fecal samples ( P < .001). Nonseroconvertors had also lower levels of blood inflammatory biomarkers on admission and lower disease severity., Conclusions: Viral replication determines the magnitude of antibody response to SARS-CoV-2, which, in turn, contributes to viral clearance. Anastrepha ludens is a key pest of mangoes and citrus from Texas to Costa Rica but the mechanisms of odorant perception in this species are poorly understood. Detection of volatiles in insects occurs mainly in the antenna, where molecules penetrate sensillum pores and link to soluble proteins in the hemolymph until reaching specific odor receptors that trigger signal transduction and lead to behavioral responses. Scrutinizing the molecular foundation of odorant perception in A. ludens is necessary to improve biorational management strategies against this pest. After exposing adults of three maturity stages to a proteinaceous attractant, we studied antennal morphology and comparative proteomic profiles using nano-LC-MS/MS with tandem mass tags combined with synchronous precursor selection (SPS)-MS3. Antennas from newly emerged flies exhibited dense agglomerations of olfactory sensory neurons. We discovered 4618 unique proteins in the antennas of A. ludens and identified some associated with odor signaling, including odorant-binding and calcium signaling related proteins, the odorant receptor co-receptor (Orco), and putative odorant-degrading enzymes. Antennas of sexually immature flies exhibited the most upregulation of odor perception proteins compared to mature flies exposed to the attractant. This is the first report where critical molecular players are linked to the odor perception mechanism of A. ludens .

Excessive interleukin-6 signaling is a key factor contributing to the cytokine release syndrome implicated in clinical manifestations of COVID-19. Preliminary results suggest that tocilizumab, a humanized monoclonal anti-interleukin-6 receptor antibody, may be beneficial in severely ill patients, but no data are available on earlier stages of disease. An anticipated blockade of interleukin-6 might hypothetically prevent the catastrophic consequences of the overt cytokine storm. We evaluated early-given tocilizumab in patients hospitalized with COVID-19, and identified outcome predictors. Consecutive patients with initial Sequential-Organ-Failure-Assessment (SOFA) score < 3 fulfilling pre-defined criteria were treated with tocilizumab. Serial plasma biomarkers and nasopharyngeal swabs were collected. Of 193 patients admitted with COVID-19, 64 met the inclusion criteria. After tocilizumab, 49 (76.6%) had an early favorable response. Adjusted predictors of response were gender, SOFA score, neutrophil/lymphocyte ratio, Charlson comorbidity index and systolic blood pressure. At week-4, 56.1% of responders and 30% of non-responders had cleared the SARS-CoV-2 from nasopharynx. Temporal profiles of interleukin-6, C-reactive protein, neutrophil/lymphocyte ratio, NT-ProBNP, D-dimer, and cardiac-troponin-I differed according to tocilizumab response and discriminated final in-hospital outcome. No deaths or disease recurrences were observed. Preemptive therapy with tocilizumab was safe and associated with favorable outcomes in most patients. Biological and clinical markers predicted outcomes.

Background: The virological and immunological effects of the immunomodulatory drugs used for COVID-19 remain unknown. We evaluated the impact of interleukin (IL)-6 blockade with tocilizumab on SARS-CoV-2 viral kinetics and the antibody response in patients with COVID-19., Methods: Prospective cohort study in patients admitted with COVID-19. Serial nasopharyngeal and plasma samples were measured for SARS-CoV-2 RNA and S-IgG/N-IgG titers, respectively., Findings: 138 patients with confirmed infection were included; 76 (55%) underwent IL-6 blockade. Median initial SOFA (p = 0•016) and SARS-CoV-2 viral load (p<0•001, Mann-Whitney-Wilcoxon test) were significantly higher among anti-IL-6 users. Patients under IL-6 blockade showed delayed viral clearance in the Kaplan-Meier curves (HR 0•35 [95%CI] [0•15-0•81], log-rank p = 0•014), but an adjusted propensity score matching model did not demonstrate a significant relationship of IL-6 blockade with viral clearance (HR 1•63 [0•35-7•7]). Cox regression showed an inverse association between SARS-CoV-2 RNA clearance and the initial viral load (HR 0•35 [0•11-0•89]). Patients under the IL-6 blocker showed shorter median time to seropositivity, higher peak antibody titers, and higher cumulative proportion of seropositivity in the Kaplan Meier curves (HR 3•1 [1•9-5] for S-IgG; and HR 3•0 [1•9-4•9] for N-IgG; log-rank p<0•001 for both). However, no significant differences between groups were found in either S-IgG (HR 1•56 [0•41-6•0]) nor N-IgG (HR 0•96 [0•26-3•5]) responses in an adjusted propensity score analysis., Interpretation: Our results suggest that in patients infected with SARS-CoV-2, IL-6 blockade does not impair the viral specific antibody responses. Although a delayed viral clearance was observed, it was driven by a higher initial viral load. Background: Acute encephalitis can occur in different viral diseases due to infection of the brain or by an immune mechanism. Severe novel coronavirus disease 2019 (COVID-19) is associated with a major immune inflammatory response with cytokine upregulation including interleukin 6 (IL-6). We report a case presenting with acute encephalitis that was diagnosed as having severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) infection with hyperinflammatory systemic response and recovered after therapy with immunoglobulins and cytokine blockade., Case Report: A 39-year-old-man was brought to the Emergency Department with drowsiness, mental disorientation, intermittent fever and headache. A brain magnetic resonance imaging showed extensive involvement of the brain including cortical and subcortical right frontal regions, right thalamus, bilateral temporal lobes and cerebral peduncles, with no leptomeningeal enhancement. Cerebrospinal fluid (CSF) showed a leukocyte count of 20/µL (90% lymphocytes), protein level of 198 mg/dL, and glucose of 48 mg/dL. SARS-CoV-2 was detected in nasopharyngeal swabs by reverse-transcriptase-PCR (RT-PCR) but it was negative in the CSF. Remarkable laboratory findings in blood tests included low lymphocyte count and elevated ferritin, IL-6 and D-dimer. He had a complicated clinical course requiring mechanical ventilation. Intravenous immunoglobulins and cytokine blockade with tocilizumab, an IL-6 receptor antagonist, were added considering acute demyelinating encephalomyelitis. In insects, the quality of sex pheromones plays a critical role in mating success and can be determined by the ability of larvae/adults to accrue chemical precursors. We tested the host-quality-effect hypothesis by analyzing the chemical composition of scent bouquets emitted by calling males of two polyphagous tephritid species ( Anastrepha ludens and A . obliqua ) that originated from 13 fruit species representing diverse plant families. In A . ludens , we worked with an ancestral host (Rutaceae), nine exotic ones (Rutaceae, Anacardiaceae, Rosaceae, Solanaceae, Lythraceae), and two species never attacked in nature but that represent candidates for host-range expansion (Solanaceae, Myrtaceae). In A . obliqua , we tested an ancestral, a native, and an exotic host (Anacardiaceae), one occasional (Myrtaceae), and one fruit never attacked in nature (Solanaceae). We identified a core scent bouquet and significant variation in the bouquet's composition depending on the fruit the larvae developed in. We also tested the possible microbial role on the scent bouquet by treating adults with antibiotics, finding a significant effect on quantity but not composition. We dwell on plasticity to partially explain our results and discuss the influence hosts could have on male competitiveness driven by variations in scent bouquet composition and how this could impact insect sterile technique programs., Competing Interests: The authors declare no conflict of interest.

The development of cost-effective diets for mass-rearing fruit flies (Diptera: Tephritidae) and their parasitoids in pest control programs based on the Sterile Insect Technique is a high priority worldwide. To this end, we tested carrageenan, agar, gelatin and two types of pregelatinized corn starches as gelling agents at varying percentages in a yeast-reduced liquid larval diet for rearing the Mexfly, Anastrepha ludens . Only diets with 0.234% ( w / w ) agar or 0.424% carrageenan were identified as diets with potential for mass-rearing A. ludens in terms of the number of pupae recovered from the diet, pupal weight, adult emergence, flight ability and diet cost. Comparative experiments showed that yeast-reduced agar and carrageenan gel diets produced heavier pupae and higher proportions of flying adults than the standard mass-rearing diet. The gel-agar and mass-rearing diets produced more pupae than the gel-carrageenan diet, but the latter produced more suitable larvae as hosts for rearing of Diachasmimorpha longicaudata (Hymenoptera: Braconidae) females, a widely used fruit fly biocontrol agent. Yeast-reduced agar and carrageenan gel diets could represent cost-effective fruit fly mass-rearing diets if a practical system for gel diet preparation and dispensation at fruit fly mass-rearing facilities is developed.

Scientifically-based, tephritid fly host status determination lies at the heart of strategic regulatory decisions impinging on international fruit trade. Here we conducted intensive field and laboratory studies with peaches as controls, to determine the host status of Physalis peruviana for the Medfly- Ceratitis capitata , as this fruit is experiencing a consumption boom worldwide. A total of 98,132 Uchuvas (local name), collected in Colombia from the plant or the ground over a three-year period (2016-2018) did not yield a single C . capitata larva or pupa, thus reaching a Probit 9 level with 99.9968% efficacy and 96% confidence level. Field-cage studies with enclosed fruit-bearing Uchuva plants, exposing fruit with an intact, damaged or totally removed husk to the attack of C . capitata , also failed to yield infestations. Highly artificial choice experiments, exposing gravid females to unripe and fully ripe fruit, resulted in an absence of infestations, even when overripe Uchuvas were artificially damaged. The husk and surface resins/waxes inhibit fly landings on fruit and oviposition activity. Considering our results and the fact that the foliage, husk and fruit of P . peruviana are repellent/toxic to insects, we conclude that this plant should be treated as a non-natural and non-conditional host of C . capitata .

With the aim of understanding the mechanisms involved in the regurgitation behavior of tephritid flies, we performed a structural study of the digestive system of the economically important fruit-fly pest, Anastrepha ludens (Loew) using optical, scanning electronic microscopy (SEM) and transmission electron microscopy (TEM), plus a feeding assay. Most structures studied are similar to those previously reported in other adult dipterans, but, importantly, we found sexual differences in some structures that apparently affect regurgitation. We report for the first time sexual differences in the crop duct nerve and large numbers of dense core vesicles within the nerve bundle. Male nerve bundles are bigger and have more secretory vesicles than female ones. The close proximity to the muscles of both the crop lobes and duct suggest that these vesicles (i.e., possibly neurosecretions) might help modulate the muscles regulating regurgitation. The salivary glands are connected to the crop via tracheae, however, SEM/TEM studies failed to find any direct structural connection. Results of the feeding assay indicate that, independently of food type (sucrose or protein) and age, males regurgitate significantly more than females. Regurgitation behavior may also play an important role in capturing bacteria in the environment, and possibly help adults eliminate ingested toxicants such as insecticides. The walnut husk fly Rhagoletis completa (Cresson), native to the Midwestern United States and Mexico, is invasive in California and Europe. It is one of the most important pests of walnuts in areas gathering 30% of the world production. Knowledge of life-history regulation is important for the design of management strategies. Research on dormancy has been performed on invasive populations, and not on populations at the southern extreme of its native range. Here, we examined the effect of winter length on fly and parasitoid emergence, survival, and duration of dormancy. Percent emergence was higher for chill periods at 5°C ranging from 8 to 20 wk. No or insufficient chill resulted in low emergence and a significant proportion of individuals in prolonged dormancy (>1 yr). Duration of dormancy was longer for pupae at constant temperatures and a 4-wk chill period than longer winter durations. Dormancy was longer for Mexican than that reported for U.S. populations, suggesting the existence of a latitudinal cline where populations at southern latitudes have evolved slower metabolic rates. Three parasitoid species were found associated with R. completa (Aganaspis alujai (Wharton and Ovruski) (Hymenoptera: Figitidae), Diachasmimorpha juglandis Muesebeck, and Diachasmimorpha mellea Gahan) (Hymenoptera: Braconidae). Results suggest that rearing of R. completa is possible by subjecting pupae to chill periods between 8 and 20 wk. Overwintering mortality of flies and A. alujai could be further reduced above 5°C. Introduction: the triathlon is an endurance sport and individual that consists of three different disciplines: swimming, cycling and running. The aim of the study was to describe and analyze the anthropometric characteristics, body composition and somatotype in male college triathletes., Methodology: observational and descriptive study of anthropometric characteristics, body composition and somatotype of 39 male college athletes from 24 ± 4,5 years, participants in the championship of Spain university triathlon sprint mode (Alicante 2010), from different universities Spanish. According to anthropometric measurement techniques adopted by the International Society for the Advancement of Kinanthropometry (ISAK) and the Spanish Group Cineantropometría (GREC) by an accredited assessor ISAK Level II., Results: we find athletes of stunting, where you destacanvalores below normal in the subscapularis, supraspinatus, triceps and biceps skinfold, percentage of muscle mass (45.27 ± 3.29%) and fat mass (10.22 ± 2.92%) and bone (16.65 ± 1.34%) and where mesomorphy somatotipo predominates., Discussion: the triathletes and runners have lower size that cyclists and swimmers. Triathletes and cyclists show a similar weight, less than swimmers line, and more than 10km runners. Iliac crest skinfold, abdominal and thigh front cyclists are less than triathletes. The percentage of fat mass of runners triathletes and swimmers are similar, however the muscle mass of athletes usually less than cyclists but similar to other forms. The semicontinuous heterophase polymerization of styrene in the presence of cross-linking and porogen agents was carried out. Latexes with close to 20% solid content, which contained mesoporous nanoparticles with 28 nm in average diameters, up to 0.5 cm3/g in porosity and 6-8 nm in pore diameters were obtained. By varying the monomer dosing rate over the micellar solution, an unexpected direct dependence of instantaneous conversion on the monomer dosing rate was found. This was ascribed to the higher average number of radicals per particle attained in the polymerization at the higher dosing rate, which in turn would arise from the higher gel percentage in the polymer. It is believed that the cross-linked chains prevent encounters between radicals, delaying the bimolecular termination reactions and allowing the existence of more than one radical inside the particles, which in turn increases the propagation rate.

Aim: To evaluate the ability of interleukin (IL)-15 to control T cell functions through its influence on CD30 and OX40 expressing cells in Celiac Disease (CD). In peripheral blood (PB), by examining the expression of OX40 in conventional effectors cells and T cells with a phenotypic specialization of regulatory cells [CD4+CD25high forkhead box protein 3 (Foxp3)+], and the co stimulation of IFN-γ and IL-4 production within CD30 and OX40 positive subsets of T cells. At the duodenal mucosa, by assessing the expression of CD30 and OX40 in intraepithelial (IE) and lamina propria (LP) lymphocytes (IEL, LPL)., Patients and Methods: PB and duodenal mucosal biopsies were obtained from 38 patients with classic CD (Cel) and 38 healthy controls (HC). Analysis of cell surface and/or intracellular antigens was performed in anti-CD3-treated PB mononuclear cells (PBMC) before and after treatment with recombinant IL-15 (rIL-15), and in IE and LP cellular suspensions prepared from duodenal biopsies pre-treated with/without rIL-15., Results: A subpopulation of CD3+OX40+ T blasts was induced in Cel and HC by a 3days treatment of PBMC with anti-CD3 and decreased its size thereafter, regardless of the presence of rIL-15. However, the addition of rIL-15 to T blasts distinctively induced the survival of T cells with a regulatory phenotype that expresses OX40 antigen in Cel (p<0.05). Celiac patients showed higher frequencies of IFN-γ-producing CD3+CD30+ blasts before and after treatment with rIL-15 (p<0.05, vs. HC). IL-15 increased the frequencies of CD3+CD30+ LPL (HC: p<0.05, Cel: p<0.05) but not of CD3+OX40+ LPL, and CD30 or OX40 positive IEL., Conclusions: The distinctive control of OX40+ cells with a T regulatory phenotype mediated by the influence of IL-15 comes out as new function of this cytokine in the context of CD. Background: Rapid and reliable identification of quarantine pests is essential for plant inspection services to prevent introduction of invasive species. For insects, this may be a serious problem when dealing with morphologically similar cryptic species complexes and early developmental stages that lack distinctive characters useful for taxonomic identification. DNA based barcoding could solve many of these problems. The standard barcode fragment, an approx. 650 base pairs long sequence of the 5'end of the mitochondrial cytochrome oxidase I (COI), enables differentiation of a very wide range of arthropods. However, problems remain in some taxa, such as Tephritidae, where recent genetic differentiation among some of the described species hinders accurate molecular discrimination., Results: In order to explore the full species discrimination potential of COI, we sequenced the barcoding region of the COI gene of a range of economically important Tephritid species and complemented these data with all GenBank and BOLD entries for the systematic group available as of January 2012. We explored the limits of species delimitation of this barcode fragment among 193 putative Tephritid species and established operational taxonomic units (OTUs), between which discrimination is reliably possible. Furthermore, to enable future development of rapid diagnostic assays based on this sequence information, we characterized all single nucleotide polymorphisms (SNPs) and established "near-minimal" sets of SNPs that differentiate among all included OTUs with at least three and four SNPs, respectively., Conclusions: We found that although several species cannot be differentiated based on the genetic diversity observed in COI and hence form composite OTUs, 85% of all OTUs correspond to described species. Because our SNP panels are developed based on all currently available sequence information and rely on a minimal pairwise difference of three SNPs, they are highly reliable and hence represent an important resource for developing taxon-specific diagnostic assays. For selected cases, possible explanations that may cause composite OTUs are discussed.
