1. Bosonic Quantum Error Correction with Neutral Atoms in Optical Dipole Traps

Abstract

Bosonic quantum error correction codes encode logical qubits in the Hilbert space of one or multiple harmonic oscillators. A prominent class of bosonic codes are Gottesman-Kitaev-Preskill (GKP) codes of which implementations have been demonstrated with trapped ions and microwave cavities. In this work, we investigate theoretically the preparation and error correction of a GKP qubit in a vibrational mode of a neutral atom stored in an optical dipole trap. This platform has recently shown remarkable progress in simultaneously controlling the motional and electronic degrees of freedom of trapped atoms. The protocols we develop make use of motional states and, additionally, internal electronic states of the trapped atom to serve as an ancilla qubit. We compare optical tweezer arrays and optical lattices and find that the latter provide more flexible control over the confinement in the out-of-plane direction, which can be utilized to optimize the conditions for the implementation of GKP codes. Concretely, the different frequency scales that the harmonic oscillators in the axial and radial lattice directions exhibit and a small oscillator anharmonicity prove to be beneficial for robust encodings of GKP states. Finally, we underpin the experimental feasibility of the proposed protocols by numerically simulating the preparation of GKP qubits in optical lattices with realistic parameters., Comment: 18 pages, 7 figures

2. Solution approaches for evaporation-driven density instabilities in a slab of saturated porous media

Abstract

This work considers the gravitational instability of a saline boundary layer formed by an evaporation-induced flow through a fully-saturated porous slab. Evaporation of saline waters can eventually result in the formation of salt lakes as salt accumulates. As natural convection can impede the accumulation of salt, establishing a relation between its occurrence and the value of physical parameters such as evaporation rate, height of the slab or porosity is crucial. One step towards determining when gravitational instabilities can arise is to compute the ground-state salinity, that evolves due to the uniform upwards flow caused by evaporation. The resulting salt concentration profile exhibits a sharply increasing salt concentration near the surface, which can lead to a gravitationally unstable setting. In this work, this ground state is analytically derived within the framework of Sturm-Liouville theory. Then, the method of linear stability in conjunction with the quasi-steady state approach is employed to investigate the occurrence of instabilities. These instabilities can develop and grow over time depending on the Rayleigh number and the dimensionless height of the porous medium. To calculate the critical Rayleigh number, which can determine the stability of a particular system, the eigenvalues of the linear perturbation equations have to be computed. Here, a novel fundamental matrix method is proposed to solve this eigenvalue problem and shown to coincide with an established Chebyshev-Galerkin method in their shared range of applicability. Finally, a 2-dimensional direct numerical simulation of the full equation system via the finite volume method is employed to validate the time of onset of convective instabilities predicted by the linear theory. Moreover, the fully nonlinear convection patterns are analyzed.

Abstract

Abstract The increasing availability of biobanks is changing the way individuals are identified for genomic research. This study assesses the validity of a self-reported clinical diagnosis of schizophrenia. The study included 1744 clinically-ascertained participants with schizophrenia or schizoaffective disorder depressed-type (SA-D) diagnosed by self-report and/or research interview and 1453 UK Biobank participants with self-reported and/or medical record diagnosis of schizophrenia or SA-D. Unaffected controls included a total of 501,837 participants. We assessed the positive predictive values (PPV) of self-reported clinical diagnoses against research interview and medical record diagnoses. Polygenic risk scores (PRS) and phenotypes relating to demographics, education and employment were compared across diagnostic groups. The variance explained (r2) in schizophrenia PRS for each diagnostic group was compared to samples in the Psychiatric Genomics Consortium (PGC). In the clinically-ascertained participants, the PPV of self-reported schizophrenia for a research diagnosis of schizophrenia was 0.70, which increased to 0.81 after expanding the research diagnosis to schizophrenia or SA-D. In UK Biobank, the PPV of self-reported schizophrenia for a medical record diagnosis was 0.74. Compared to participants who self-reported, participants with a clinically-ascertained research diagnosis were younger and more likely to have a high school qualification. Participants with a medical record diagnosis in UK Biobank were less likely to be employed or have a high school qualification than those who self-reported. Schizophrenia PRS did not differ between participants that had a diagnosis from self-report, research diagnosis or medical records. Polygenic liability r2, for all diagnosis definitions, fell within the distribution of PGC schizophrenia cohorts. Self-reported measures of schizophrenia are justified in genomic research to maximise sample size and reduce the burden of in-depth interviews on participants, although within sample validation of diagnoses is recommended.

Abstract

Sand nourishment is a common and often applied coastal protection measure along the beaches of the German Baltic Sea coast. At the tourist beach of Ahrenshoop, large-scale sand nourishments are completed every five years along a 4-km stretch. The nourishment in question was executed between the end of October 2021 and mid-February 2022. Meiofaunal communities at the beach-water interface were monitored at six stations before (September 2021) and for one year after the impact (March and September 2022, March 2023). To assess the effects of sand nourishment on meiofauna communities, two complementary approaches were employed: whole community metabarcoding and morphological determination and counting of the animals. Both methods produced consistent results regarding community response to disturbance and subsequent recovery. During the period from autumn 2021 to spring 2023, drastic changes in the composition of communities were observed. Directly after the impact, the taxa Acari and Annelida nearly disappeared, abundances of Copepoda decreased substantially, and Platyhelminthes increased notably. The fact that the studied beach held a low diversity may have facilitated that metabarcoding and morphological determination followed the same trend. After initial studies comprising both methods, efficient assessments using only metabarcoding may be considered for comparable sites and situations.

Abstract

Abstract The combination of lineage tracing and immunohistochemistry has helped to identify subpopulations and fate of hepatic stellate cells (HSC) in murine liver. HSC are sinusoidal pericytes that act as myofibroblast precursors after liver injury. Single cell RNA sequencing approaches have recently helped to differentiate central and portal HSC. A specific Cre line to lineage trace portal HSC has not yet been described. We used three Cre lines (Lrat-Cre, PDGFRβ-CreERT2 and SMMHC-CreERT2) known to label mesenchymal cells including HSC in combination with a tdTomato-expressing reporter. All three Cre lines labeled populations of HSC as well as smooth muscle cells (SMC). Using the SMMHC-CreERT2, we identified a subtype of HSC in the periportal area of the hepatic lobule (termed zone 1-HSC). We lineage traced tdTomato-expressing zone 1-HSC over 1 year, described fibrotic behavior in two fibrosis models and investigated their possible role during fibrosis. This HSC subtype resides in zone 1 under healthy conditions; however, zonation is disrupted in preclinical models of liver fibrosis (CCl4 and MASH). Zone 1-HSC do not transform into αSMA-expressing myofibroblasts. Rather, they participate in sinusoidal capillarization. We describe a novel subtype of HSC restricted to zone 1 under physiological conditions and its possible function after liver injury. In contrast to the accepted notion, this HSC subtype does not transform into αSMA-positive myofibroblasts; rather, zone 1-HSC adopt properties of capillary pericytes, thereby participating in sinusoidal capillarization.

Abstract There are 90 independent genome-wide significant genetic risk variants for Parkinson’s disease (PD) but currently only five nominated loci for PD progression. The biology of PD progression is likely to be of central importance in defining mechanisms that can be used to develop new treatments. We studied 6766 PD patients, over 15,340 visits with a mean follow-up of between 4.2 and 15.7 years and carried out genome-wide survival studies for time to a motor progression endpoint, defined by reaching Hoehn and Yahr stage 3 or greater, and death (mortality). There was a robust effect of the APOE ε4 allele on mortality in PD. We also identified a locus within the TBXAS1 gene encoding thromboxane A synthase 1 associated with mortality in PD. We also report 4 independent loci associated with motor progression in or near MORN1, ASNS, PDE5A, and XPO1. Only the non-Gaucher disease causing GBA1 PD risk variant E326K, of the known PD risk variants, was associated with mortality in PD. Further work is needed to understand the links between these genomic variants and the underlying disease biology. However, these may represent new candidates for disease modification in PD.

Abstract

Background Physical health conditions are more common in individuals with autism. Some, like epilepsy, have considerable evidence supporting their increased prevalence, but many diseases lack literature to make strong conclusions. Aims To investigate the prevalence of physical health comorbidities in autism. Method We undertook a nested cross-sectional study, using a sample from the National Centre for Mental Health database. It included participants from England and Wales who reported a clinician-made diagnosis of autism (n = 813), and a control sample without autism or mental illness (n = 2781). Participants had provided a medical history at enrolment. Analysis was carried out by binomial logistic regressions controlling for age, gender, smoking status, and antipsychotic and mood stabiliser use. A subanalysis of individuals with concurrent intellectual disability (n = 86) used binomial logistic regression with the same control variables. Results Many physical health conditions were significantly more common in autism. Sixteen out of 28 conditions showed increased odds, with the highest odds ratios observed for liver disease, chronic obstructive pulmonary disease, kidney disease, osteoporosis and rheumatoid arthritis. A subanalysis demonstrated a similar pattern of physical health in individuals with autism with and without concurrent intellectual disability. Some conditions, including osteoporosis, hyperthyroidism, head injury and liver disease, had larger odds ratios in individuals with concurrent intellectual disability. Conclusions Physical health conditions occur more commonly in individuals with autism, and certain conditions are further increased in those with concurrent intellectual disability. Our findings contribute to prior evidence, including novel associations, and suggest that people with autism are at greater risk of physical health problems throughout adulthood.

Abstract

Forms of algorithmic management (AM) play an essential role in organizing food-delivery work by deploying artificial intelligence-based systems to coordinate driver routes. Given the risks of precarity and threats posed by AM, which are typically related to (migrant) platform work, the question arises to what extent structures of co-determination can positively shape this type of work and the technologies involved. Based on an in-depth case study within a large food-delivery company, this article is guided by two questions: (1) How do companies use algorithm-based management and performance control, and how do the couriers perceive them? (2) What priorities, strategies, resources, and achievements do works councils and trade unions have with regard to co-determination practices? Our analyses indicate that algorithmic management poses problems of non-transparency and information asymmetry, which in turn call for new forms of and procedures for co-determination. Our study does not find evidence that AM practices aim to individually profile and discipline couriers. The main challenges for the works council and trade unions arise from the couriers’ generally precarious working and employment conditions; data- and AM-related issues do not represent the central area of conflict. However, our study identifies new demands regarding the co-determination of AM and underlines the importance of institutional regulation at the legal and sectoral level.

Abstract Introduction A three-pronged approach to acne treatment combining an antibiotic, antimicrobial, and retinoid may be more efficacious than single/double treatments while potentially reducing antibiotic resistance. This study evaluated the efficacy and safety of the first fixed-dose, triple-combination topical acne product, clindamycin 1.2%/adapalene 0.15%/benzoyl peroxide (BPO) 3.1% gel (CAB) using pooled phase 3 data. Methods In two identical phase 3 (N = 183; N = 180), double-blind, 12-week studies, participants aged ≥ 9 years with moderate-to-severe acne were randomized 2:1 to receive once-daily CAB or vehicle gel. Endpoints included ≥ 2-grade reduction from baseline in Evaluator’s Global Severity Score and clear/almost clear skin (treatment success) and least-squares mean percent change from baseline in acne lesion counts. Treatment-emergent adverse events (TEAEs) and cutaneous safety/tolerability were evaluated. Results At week 12, 50.0% of participants achieved treatment success with CAB versus 22.6% with vehicle gel (P 70% reductions in inflammatory and noninflammatory lesions at week 12 (77.9% and 73.0%, respectively), which were significantly greater than vehicle (57.9% and 48.2%; P

Abstract

Abstract Organoboron compounds have a wide range of applications in numerous research fields, and methods to incorporate them in biomolecules are much sought after. Here, on‐resin chemical syntheses of aliphatic and vinylogous peptide boronic acids are presented by transition metal‐catalyzed late‐stage hydroboration of alkene and alkyne groups in peptides and peptoids, for example on allyl‐ and propargylglycine residues, using readily available chemicals. These methods yield peptide boronic acids with much shorter linkers than previously reported on‐resin methods. Furthermore, the methods are regio‐ and stereoselective, compatible with all canonical amino acid residues and can be applied to short, long, and in part even “difficult” peptide sequences. In a feasibility study, the protected peptide vinylboronic acids are further derivatized by the Petasis reaction using salicylaldehyde derivatives. The ability of the obtained peptide boronic acids to reversibly bind to carbohydrates is demonstrated in a catch‐release model experiment using a fluorescently labeled peptide boronic acid on cross‐linked dextran beads. In summary, this highlights the potential of the target compounds for drug discovery, glycan‐specific target recognition, controlled release, and diagnostics.
