Abstract

Six of Japanese tree frogs(Hyla japonica)were sent to the Russian space station MIR (FRIS experiment group, 1990 and Izumi-Kurotani, et al., 1991). Their postures, movements, behaviors were observed under microgravity. Floating frogs showed a typical posture in space. They stretched four legs out and bent their body backward. Response behaviors to the external stimuli were similar to those on ground. When frogs stayed on the surface of something, many frogs bent their neck backward. Adaptation to microgravity was expressed as improvement of landing after jumping in later experiment. After 8 days on orbit, all the tree frogs were recovered alive to ground. Readaptation to normal gravity was also investigated.

Abstract

The rodent granular retrosplenial cortex (GRS) is reciprocally connected with the hippocampus. It is part of several networks implicated in spatial learning and memory, and is known to contain head-direction cells. There are, however, few specifics concerning the mechanisms and microcircuitry underlying its involvement in spatial and mnemonic functions. In this report, we set out to characterize intrinsic properties of a distinctive population of small pyramidal neurons in layer 2 of rat GRS. These neurons, as well as those in adjoining layer 3, were found to exhibit a late-spiking (LS) firing property. We established by multiple criteria that the LS property is a consequence of delayed rectifier and A-type potassium channels. These were identified as Kv1.1, Kv1.4 and Kv4.3 by Genechip analysis, in situ hybridization, single-cell reverse transcriptase-polymerase chain reaction, and pharmacological blockade. 13. Functional Thalamocortical Synapse Reorganization from Subplate to Layer IV during Postnatal Development in the Reeler-Like Mutant Rat (Shaking Rat Kawasaki).

Abstract

Transient synapse formation between thalamic axons and subplate neurons is thought to be important in thalamocortical targeting. Shaking rat Kawasaki (SRK), having reversed cortical layering similarly observed in reeler mouse, provides an interesting model system to test this idea. The spatial and temporal pattern of excitation was investigated using optical recording with voltage-sensitive dyes in thalamocortical slice preparations from SRK. At postnatal day 0 (P0), a strong optical response was elicited within the superplate of the SRK in the cell layer corresponding to subplate in wild-type (WT) rats. By P3, this response rapidly descended into deep cortical layers comprised of layer IV cells, as identified with 5-bromo-2'-deoxynridine birthdating at embryonic day 17. During the first 3 postnatal days, both the subplate and cortical plate responses were present, but by P7, the subplate response was abolished. Tracing individual axons in SRK revealed that at P0-P3, a large number of thalamocortical axons reach the superplate, and by PT-P10, the ascending axons develop side branches into the lower or middle cortical layers. Synaptic currents were also demonstrated in WT subplate cells and in SRK superficial cortical cells using whole-cell recording. These currents were elicited monosynaptically, because partial AMPA current blockade did not modify the latencies. These results suggest that the general developmental pattern of synapse formation between thalamic axons and subplate (superplate) neurons in WT and SRK is very similar, and individual thalamic arbors in cortex are considerably remodeled during early postnatal development to find layer IV equivalent neurons. 14. Ca2+-Dependent Photoreceptor Potential in Paramecium Bursaria

Abstract

Photostimulation of deciliated specimens of chlorella-free P. bursaria elicited a transient depolarization of the membrane potential. The amplitude of this receptor potential became larger as light intensity was increased and the relationship showed a Michaelis equation. The action spectrum of the receptor potential showed two peaks at around 420 and 560 nm. When constant current was injected to depolarize the membrane gradually, the receptor potential showed a decrease in amplitude. The potential at which the response disappeared depended on the external concentration of Ca2+ (27 mV/log [Ca2+]o), and the amplitude of the receptor potential was also dependent upon external Ca2+ concentration. Therefore, the receptor potential is primarily caused by a transient increase in the membrane conductance to Ca2+.

15. Ionic Mechanism of Thermoreception in Paramecium

Abstract

The localization of thermoreceptors in Paramecium, and the ionic basis of thermoreception, was investigated in posterior and anterior fragments of cells. Transverse section of the animals was used to obtain these fragments, which sealed up and swam actively. In the anterior fragment, an increase in the frequency of directional changes in swimming and depolarization of the membrane was produced by cooling below the temperature of the culture. In the posterior fragment, these effects were produced by wanning above culture temperature. Reversal potentials of these effects were found by injection of constant current to change membrane potential. In the anterior fragment, the reversal potential of the response to cooling was more negative than the resting potential and was potassium-dependent (52 mV/ log[K+]0). In the posterior fragment, the reversal potential of the warming response was above resting potential and was primarily calcium-dependent (28 mV/ log[Ca2+]0). It is concluded that cooling results in changes in the frequency of directional changes in swimming of Paramecium by causing a transient change in the membrane conductance for potassium, whereas warming produces its effects by a transient change in calcium conductance.
