14. Model Predictive Control of Spatially Distributed Systems with Spatio-Temporal Logic Specifications.

Abstract

In this paper, for spatially distributed systems, we propose a new method of model predictive control with spatio-temporal logic specifications. We formulate the finite-time control problem with specifications described by SSTLf (signal spatio-temporal logic over finite traces) formulas. In the problem formulation, the feasibility is guaranteed by representing control specifications as a penalty in the cost function. Time-varying weights in the cost function are introduced to satisfy control specifications as well as possible. The finite-time control problem can be written as a mixed integer programming (MIP) problem. According to the policy of model predictive control (MPC), the control input can be generated by solving the finite-time control problem at each discrete time. 16. Multi-Agent Surveillance Based on Travel Cost Minimization

Abstract

The multi-agent surveillance problem is to find optimal trajectories of multiple agents that patrol a given area as evenly as possible. In this paper, we consider the multi-agent surveillance problem based on travel cost minimization. The surveillance area is given by an undirected graph. The penalty for each agent is introduced to evaluate the surveillance performance. Through a mixed logical dynamical system model, the multiagent surveillance problem is reduced to a mixed integer linear programming (MILP) problem. In model predictive control, trajectories of agents are generated by solving the MILP problem at each discrete time. Furthermore, a condition that the MILP problem is always feasible is derived based on the Chinese postman problem. Finally, the proposed method is demonstrated by a numerical example.

Abstract

In a multi-agent system, it is important to consider a design method of cooperative actions in order to achieve a common goal. In this paper, we propose two novel multi-agent reinforcement learning methods, where the control specification is described by linear temporal logic formulas, which represent a common goal. First, we propose a simple solution method, which is directly extended from the single-agent case. In this method, there are some technical issues caused by the increase in the number of agents. Next, to overcome these technical issues, we propose a new method in which an aggregator is introduced. Finally, these two methods are compared by numerical simulations, with a surveillance problem as an example.

Abstract

The differentiation of αβT cells from thymic precursors is a complex process essential for adaptive immunity. Here we exploited the breadth of expression data sets from the Immunological Genome Project to analyze how the differentiation of thymic precursors gives rise to mature T cell transcriptomes. We found that early T cell commitment was driven by unexpectedly gradual changes. In contrast, transit through the CD4(+)CD8(+) stage involved a global shutdown of housekeeping genes that is rare among cells of the immune system and correlated tightly with expression of the transcription factor c-Myc. Selection driven by major histocompatibility complex (MHC) molecules promoted a large-scale transcriptional reactivation. We identified distinct signatures that marked cells destined for positive selection versus apoptotic deletion. Differences in the expression of unexpectedly few genes accompanied commitment to the CD4(+) or CD8(+) lineage, a similarity that carried through to peripheral T cells and their activation, demonstrated by mass cytometry phosphoproteomics. The transcripts newly identified as encoding candidate mediators of key transitions help define the 'known unknowns' of thymocyte differentiation.

Abstract

Helicobacter pylori (H. pylori) infects the gastric mucosa and persists for the life of the host. Bacterial persistence may be due to the induction of regulatory T cells (Tregs) whichmay have protective effects against other diseases such as asthma. It has been shown that H. pylori modulates the T cell response through dendritic cell reprogramming but the molecular pathways involved are relatively unknown. The goal of this study was to identify critical elements of dendritic cell (DC) activation and evaluate potential influence on immune activation. Microarray analysis was used to demonstrate limited gene expression changes in H. pylori stimulated bone marrow derived DCs (BMDCs) compared to the BMDCs stimulated with E. coli. IRAK-M, a negative regulator of TLR signaling, was upregulated and we selectedit for investigation of its role in modulating the DC and T cell responses. IRAK-M(-/-) and wild type BMDC were compared for their response to H. pylori. Cells lacking IRAK-M produced significantly greater amounts of proinflammatory MIP-2 and reduced amounts of immunomodulatory IL-10 than wild type BMDC. IRAK-M(-/-) cells also demonstrated increased MHC II expression upon activation. However, IRAK-M(-/-) BMDCs were comparable to wild type BMDCs in inducing T-helper 17 (TH17) and Treg responses as demonstrated in vitro using BMDC CD4+ T cells co-culture assays,and in vivo though the adoptive transfer of CD4(+) FoxP3-GFP T cells into H. pylori infected IRAK-M(-/-) mice. These results suggest that H. pylori infection leads to the upregulation of anti-inflammatory molecules like IRAK-M and that IRAK-M has a direct impact on innate functions in DCs such as cytokine and costimulation molecule upregulation but may not affect T cell skewing.

Abstract

This study proposes a novel stabilizing controller for nonlinear systems using group-wise sparse inputs. The input variables are divided into several groups. In the situations when the input constraints can be ignored, one input becomes active for each group at each moment. Our method improves energy efficiency, as sparse input vectors often reduce the standby power of inactive actuators. Large-scale systems, such as those consisting of multiple subsystems, often require the manipulation of multiple inputs simultaneously to be controlled. Our method can be applied to such systems due to the group-wise sparsity of the inputs. The proposed controller is based on the control Lyapunov function approach and includes Sontag's universal formula as a special case. The controllers designed in our method have best-effort property, which means even when a restriction for the decreasing rate of the Lyapunov function cannot be fulfilled, the controller minimizes the time derivative of the Lyapunov function within the input constraint. The effectiveness of the proposed method can be confirmed through simulations.

Abstract

This study proposes a controller design method based on block sparse optimization for dynamical network systems. The objective of the controller is to stabilize dynamical network systems with a given convergence rate. The block sparse optimization minimizes the number of controlled nodes. This study is unique in that the structure of the controller is constrained by the network topology of the system. Additionally, the proposed design problem is separable in terms of the distributed optimization over networks. The proposed method is applicable to controller design for the pinning control of consensus systems and the optimal vaccine allocation for epidemic spreading processes.
