2. Process analytical technology for the manufacture of biotherapeutic protein products

Abstract

The emergence of new bioprocessing technologies for integrated, continuous biotherapeutics manufacturing has instigated a need to establish new process analytical technologies (PAT) that are able to monitor product attributes that ultimately define the safety and efficacy of the drug. The aim of this Engineering Doctorate has been to develop new analytical technologies that address a problematic facet of biologics manufacturing: the propensity for proteins to unfold and aggregate. A high-throughput system for circular dichroism (CD) and fluorescence spectroscopy has been developed for automated capillary-based structural analysis of biotherapeutics loaded in 96-well plate format. The system was used to examine the conformational stability of monoclonal antibody (mAb) using chemical denaturation (urea) and low pH elution buffers. Subsequently, an in-line methodology was developed for monitoring structural changes of mAb immediately following affinity chromatography (Protein A/Protein G/Byzen Pro) through measured changes in CD\(_{240 nm}\) g-factor and fluorescence \(\lambda_{max}\), establishing a new technique for real time release testing in bioprocessing. Finally, a fused silica microcapillary (220 μm i.d.) flow cell was developed for high-throughput/in-line CD analysis of concentrated protein solutions and further demonstrated as being suitable for the alignment of macromolecules for rapid linear dichroism (LD) detection.

Abstract

The BabA adhesin mediates high-affinity binding of Helicobacter pylori to the ABO blood group antigen-glycosylated gastric mucosa. Here we show that BabA is acid responsive-binding is reduced at low pH and restored by acid neutralization. Acid responsiveness differs among strains; often correlates with different intragastric regions and evolves during chronic infection and disease progression; and depends on pH sensor sequences in BabA and on pH reversible formation of high-affinity binding BabA multimers. We propose that BabA's extraordinary reversible acid responsiveness enables tight mucosal bacterial adherence while also allowing an effective escape from epithelial cells and mucus that are shed into the acidic bactericidal lumen and that bio-selection and changes in BabA binding properties through mutation and recombination with babA-related genes are selected by differences among individuals and by changes in gastric acidity over time. These processes generate diverse H. pylori subpopulations, in which BabA's adaptive evolution contributes to H. pylori persistence and overt gastric disease.

5. English-speaking war correspondents of the Spanish Civil War : why was objectivity impossible?

Abstract

Clear Blue Waters of the Danube was planned and drafted from October 2007 to December 2012. It is written from the perspective of Daniel Rourke, a young man whose life is changed forever by the arrival into the family home of Marija Kovač, a Croatian refugee. The wars leading to the break-up of Yugoslavia, notably the Croatian War of Independence from 1990-5 and the Bosnian Civil War from 1992-5, provide the novel's historical background. Preparation included interviews with conflict survivors, witnesses, soldiers who fought in the war, and those who were children during the fighting. Research visits to Serbia, Croatia and Bosnia and Herzegovina took place during the summers of 2008 and 2009. I also drew upon conversations with former Yugoslav refugees from my time working in London during the 1990s and early 2000s. Other information was selected from biographies, historical records, documentary films, diaries and reports by the International Criminal Tribunal for the Former Yugoslavia. Although the novel notes the key moments of Yugoslavia's violent break-up, Clear Blue Waters of the Danube is not a political thriller. It follows a young man on a journey of self-discovery that takes him away from the family home, first to London, then across the Balkans. By establishing the truth about terrible incidents from the past, he comes to a greater understanding about himself and his previous behaviour. More importantly he is able to re-evaluate the relationship with his father that lies at the heart of everything he does, and in whose shadow he has always lived. The question of whether a writer is truly able to separate himself from his/her subject matter is investigated in greater depth throughout my critical project. Planned between October 2007 and June 2008 then written over the following two years, the perspectives of English-speaking war correspondents during the Spanish Civil War from 1936-1939 are examined. Newspaper articles, memoirs, biographies and films are scrutinised. Although the allegiances of British newspapers were split more or less evenly, the majority of writers and reporters supported the Republican effort and invested huge amounts of personal feeling into their work. For a war fought over such contrasting values, a degree of bias was perhaps inevitable. As I began my research, my aim was to investigate to what extent objectivity in such circumstances was even possible. If news reports bore the hallmarks of fiction, what then of the Spanish Civil War novel? The final part of the project deals with Ernest Hemingway and For Whom the Bell Tolls. As a journalist, Hemingway had engaged in propaganda on behalf of the Republic and readily accepted the weak evidence behind the denunciation of Republican dissidents. Following the war‟s conclusion, he returned to Cuba to write his novel of the Spanish Civil War, For Whom the Bell Tolls. Ironically having written newspaper reports to spread misinformation, he elected to use the form of a novel to reveal his version of what had actually happened. Can fiction reveal the 'truth' about events when supposedly non-fiction texts cannot? My thesis asks fundamental questions about why we write and what we choose to write about. Can any writer truly separate him/herself from the subject matter? Can our understanding ever be full and free from bias and prejudice? Or do a writer's values permeate the work to the extent that, whether a newspaper article or a novel is written, genuine objectivity becomes impossible? Is the quest for objectivity a desirable or realistic aspiration?

17. Overcoming Obstacles to DNA Group Engagement

Abstract

Discipleship is the primary command given by Jesus to his disciples. The elements of discipleship growth are best carried out in smaller cell groups of ten to twenty people. In the model used by the researcher, these cell groups (called missional communities) are subdivided into groups of three comprised of the same gender. These subdivided groups are called DNA groups. It is within DNA groups that a more intimate environment is created for transparency, biblical learning, and accountability. The researcher found three primary obstacles to engagement in a previous iteration of DNA groups. Those three obstacles are overscheduling / busyness, an unwillingness to have difficult conversations, and a general incompetency in using the Bible alone for personal spiritual formation due to an over-dependence on premade curricula. The researcher did both a presurvey and a postsurvey. The intervention was a series of presentations on how to overcome each of the three obstacles. Comparison of the data from the surveys demonstrates that teaching alone is not enough to create measurable change in personal behavior. However, the missional community model contains within it the components necessary to create measurable change over a longer period of time.

Abstract

Introduction Radiotherapy provides excellent outcome in early stage glottic cancer; however, the optimal radiotherapy dose fractionation remains unknown. Objective To investigate the outcome of patients with T2N0M0 treated with either hypofractionated (HypoFxn) or conventionally fractionated radical (ConFxn) radiotherapy. Methods According to our institutional protocol, patients with T2N0M0 glottic cancer can be treated either with ConfFxn or HypoFxn radiotherapy, as per clinician's and patient's choice, following shared decision making discussing the advantages and disadvantages of both modalities. A total of 77 patients with T2N0M0 squamous cell carcinoma of glottis treated with either HypoFxn 55Gy in 20 fractions (n = 19) or ConFxn 63 to 65Gy in 30 fractions (n = 58) were included. Results With median follow-up of 3.4 years, there was no significant difference in disease-free survival (median: HypoFxn = 65.2 months, and ConFxn = 75.3 months; p = 0.874), local recurrence free survival rates (median: HypoFxn = 78.8 months vs. ConFxn = 81.2 months; p = 0.274), and overall survival (median: HypoFxn = 65.9 months vs. ConFxn = 67.7 months; p = 0.532). Elective neck irradiation was given to 43 patients, all in the ConFxn group, and this was associated with poorer local control (p = 0.027). The use of radiotherapy modality, three-dimensional conformal radiotherapy (3DRT) versus intensity modulated radiotherapy (IMRT), was not a prognostic factor (p = 0.36). In the HypoFxn group, grade III acute dysphagia requiring nasogastric tube was 16%, compared with 25% in the ConFxn group (p = 0.446). Conclusion HypoFxn radiotherapy provides a comparable treatment outcome with acceptable toxicity. The addition of prophylactic irradiation of the neck lymph nodes has no impact on regional control. Abstract

Here we characterise four dyes and assess the complementarity of linear dichroism and FRET in biomimetic light-harvesting antennae optimisation., It is a challenge within the field of biomimetics to recreate the properties of light-harvesting antennae found in plants and photosynthetic bacteria. Attempts to recreate these biological structures typically rely on the alignment of fluorescent moieties via attachment to an inert linear scaffold, e.g. DNA, RNA or amyloid fibrils, to enable Förster resonance energy transfer (FRET) between attached chromophores. While there has been some success in this approach, refinement of the alignment of the chromophores is often limited, which may limit the efficiency of energy transfer achieved. Here we demonstrate how linear dichroism spectroscopy may be used to ascertain the overall alignment of chromophores bound to the M13 bacteriophage, a model linear scaffold, and demonstrate how this may be used to distinguish between lack of FRET efficiency due to chromophore separation, and chromophore misalignment. This approach will allow the refinement of artificial light-harvesting antennae in a directed fashion.

Abstract

Previous xenogeneic immunization experiments in rhesus macaques with simian immunodeficiency virus (SIV) grown in human CD[4.sup.+] T cells consistently elicited protection from challenge with live SIV. However, the mechanism of protection has not been established. We present evidence that xenogeneic immunization induced significant CD8 suppressor factor, RANTES (regulated upon activation, normal T cell expressed and secreted), macrophage inflammatory protein (MIP) 1[Alpha], and MIP-1[Beta] (P < 0.001 - P < 0.02). The concentrations of these increased significantly in protected as compared with infected macaques (P < 0.001). Xenogeneic stimulation in vitro also up-regulated CD8 suppressor factors (SF; P < 0.001) and the [Beta] chemokines which were neutralized by antibodies to the 3 [Beta] chemokines. Recombinant human RANTES, MIP-1[Alpha] and MIP-1[Beta] which bind to simian CCR5, suppressed SIV replication in a dose-dependent manner, with RANTES being more effective than the other two chemokines. The results suggest that immunization with SIV grown in human CD[4.sup.+] T cells induces CD8-suppressor factor, RANTES, MIP-1[Alpha] and MIP-1[Beta] which may block CCR5 receptors and prevent the virus from binding and fusion to CD[4.sup.+] cells.
