5. High-throughput isolation of gentiopicroside from Gentiana lutea and the study on its anticancer properties in vitro

Abstract

The study aims at the isolation of gentiopicroside (GPC) from Gentiana lutea L. root extracts. GPC is a bioactive glycosylated secoiridoid from Gentiana spp. (Gentianaceae) proved to exhibit broad pharmaceutical activities, including anti-inflammatory, hepatoprotective and spasmolytic action. Due to its marked potential, an efficient recovery method of high purity GPC from G. lutea roots was elaborated using the combination of Centrifugal Partition Extraction (CPE) that offers the advantage of industrial scalability, and Preparative High Performance Liquid Chromatography (prep-HPLC) techniques. For this purpose the root hydroalcoholic extract of G. lutea was initially fractionated in a 0.3 L volume CPE column using the biphasic solvent system that was composed of heptane: ethyl acetate: ethanol: water 1:8:3:5 (v/v/v/v) in elution-extrusion mode. As a result, 2.2 g of GPC (purity > 90%) and 0.77 g of GPC-enriched fractions (purity > 80%) were obtained from 31.5 g of the extract within 6 h (5 consecutive CPE fractionations). The following prep-HPLC based purification step increased the quantity and purity of the finally isolated secoiridoid to 2.21 g of > 96% pure GPC. The successful recovery of large amounts of pure GPC enabled the study on the anti-proliferative activity against the following cancer and normal cell lines: lung (A549, NCI-H1299), breast (MCF7, MDA-MB-468), glioblastoma (T98G), rhabdomyosarcoma (TE671), colon (HT-29, DLD-1) cancer cells and normal colon epithelial cells (CCD 841 CoTr). The MTT assay enabled the evaluation of the dose-dependent growth inhibition effect that was evident in all analyzed cancer cell lines, however, TE671 cells were the most sensitive (IC50 = 0.078 mg/mL) and MDA-MB-468 cells were the least sensitive (IC50 = 0.208 mg/mL) to the GPC treatment. The tested compound showed only a weak cytotoxic effect against normal cells (CCD 841 CoTr). © 2023 Elsevier B.V.

Abstract

A novel two-phase aerosolisation mode utilizing porous materials is investigated, aiming to improve aerosol delivery for medical inhalation. Sintered stainless steel filters with varied pore sizes (PS) from 0.2 μm to 7 μm were used to generate aerosols from a 0.9 wt.% sodium chloride solution. Droplet sizes and delivery rates were measured using laser diffraction spectroscopy. Further measurements included shadow imaging. Results indicate that aerosolisation occurs within a specific range of PS with droplet sizes increasing with increasing PS. The droplets generated are suitable for inhalation therapies. A hypothesis is established about the process of droplet formation which states that different PS within the porous material serve distinct functions that contribute to the breakup of liquid films into aerosol particles. Droplet formation is the result of film breakup in pores filled with fluid. This low-energy aerosolisation method has the potential to be used in handheld devices for sensitive drug formulations, overcoming the limitations of current technologies. Further research is needed to optimize the pore size distribution and enhance aerosol generation efficiency.

20. Identification of mushroom and murine tyrosinase inhibitors from Achillea biebersteinii Afan. Extract

Abstract

Growing scientific evidence indicates that Achillea biebersteinii is a valuable source of active ingredients with potential cosmetic applications. However, the data on its composition and pharmacological properties are still insufficient. This study aims to optimize the extraction procedure of the plant material, evaluate its phytochemical composition, and compare anti-tyrosinase potential of A. biebersteinii extracts obtained by various methods. In order to identify compounds responsible for the tyrosinase inhibitory activity of A. biebersteinii, the most active anti-tyrosinase extract was fractionated by column chromatography. The fractions were examined for their skin lightening potential by mushroom and murine tyrosinase inhibitory assays and melanin release assay. HPLC-ESI-Q-TOF-MS/MS analysis of the total extract revealed the presence of several phenolic acids, flavonoids, flavonoid glucosides, and carboxylic acid. Among them, fraxetin-8-O-glucoside, quercetin-O-glucopyranose, schaftoside/isoschaftoside, gmelinin B, 1,3-dicaffeoylquinic acid (1,3- DCQA), and ferulic acid were found in the fractions with the highest skin lightening potential. Based on obtained qualitative and quantitative analysis of the fractions, it was assumed that the caffeoylquinic acid derivatives and dicaffeoylquinic acid derivatives are more likely responsible for mushroom tyrosinase inhibitory activity of A. biebersteinii extracts and fractions. Ferulic acid was proposed as the most active murine tyrosinase inhibitor, responsible also for the reduced melanin release from B16F10 murine melanoma cells. © 2021 by the authors. Abstract

Palmatine (PLT) is a natural isoquinoline alkaloid that belongs to the class of protoberber-ines and exhibits a wide spectrum of pharmacological and biological properties, including anti-cancer activity. The aim of our study was to isolate PLT from the roots of Berberis cretica and investigate its cytotoxic and anti-proliferative effects in vitro alone and in combination with doxorubicine (DOX) using human ER+ /HER2− breast cancer cell lines. The alkaloid was purified by column chromatog-raphy filled with silica gel NP and Sephadex LH-20 resin developed in the mixture of methanol: water (50:50 v/v) that provided high-purity alkaloid for bioactivity studies. The purity of the alkaloid was confirmed by high resolution mass measurement and MS/MS fragmentation analysis in the HPLC-ESI-QTOF-MS/MS-based analysis. It was found that PLT treatment inhibited the viability and proliferation of breast cancer cells in a dose-dependent manner as demonstrated by MTT and BrdU assays. PLT showed a quite similar growth inhibition on breast cancer cells with IC50 values ranging from 5.126 to 5.805 µg/mL. In contrast, growth of normal human breast epithelial cells was not affected by PLT. The growth inhibitory activity of PLT was related to the induction of apoptosis, as determined by Annexin V/PI staining. Moreover, PLT sensitized breast cancer cells to DOX. Isobolographic analysis revealed synergistic and additive interactions between studied agents. Abstract

Acoustic resonances are computed numerically in rectangular two-dimensional deep and shallow open cavities. Unconfined cavities as well as cavities confined in a two-dimensional infinitely long duct are considered. Because of radiation losses these resonances are generally complex valued. All computations are made for zero mean flow. Perfectly matched layer absorbing boundary conditions are employed to avoid unphysical reflections at the necessarily finite grid boundaries. In confined cavities a few resonant modes, so-called trapped modes, exist with zero radiation losses, and significant differences are observed between confined and unconfined cavity resonances. A comparison with the frequencies of the modes observed by Rossiter suggests that the dominant Rossiter modes lock in at resonant cavity frequencies.

Abstract

Magnoflorine (MGN) is a quaternary aporphine alkaloid that exhibits numerous therapeutic properties, including neuropsychopharmacological, anti-anxiety, immunomodulatory, anti-inflammatory, antioxidant, or antifungal activities. The aim of the present study was an investigation of the influence of MGN on viability, proliferation, induction of apoptosis, and cell cycle arrest in NCI-H1299 lung, MDA-MB-468 breast, T98G glioma, and TE671 rhabdomyosarcoma cancer cells. MGN was isolated from the roots of Berberis cretica L. by counter-current partition chromatography (CPC). Cell viability and proliferation assessments were performed by means of MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) and 5-bromo-2′-deoxyuridine (BrDU) assays, respectively. The induction of apoptosis and cell cycle progression was measured using fluorescence-activated cell sorting analysis. MGN in high doses inhibits proliferation, induces apoptosis, and inhibits cell cycle in S/G2 phases in a dose-dependent manner. MGN seems to be a promising anti-cancer compound in therapy of some types of lung, breast, glioma, and rhabdomyosarcoma cancers, for which current standard therapies are limited or have severe strong side effects. © 2020 by the authors. Abstract

In surveillance problems dense clutter/dense target situations call for refined data association and tracking techniques. In addition, closely spaced targets may exist which are not resolved. This phenomenon has to be considered explicitly in the tracking algorithm. We concentrate on two targets which temporarily move in close formation and derive a generalization of MHT methods on the basis of a simple resolution model.

Abstract

The vision of NFDI4Chem is the digitalisation of all key steps in chemical research to support scientists in their efforts to collect, store, process, analyse, disclose and re-use research data. Measures to promote Open Science and Research Data Management (RDM) in agreement with the FAIR data principles are fundamental aims of NFDI4Chem to serve the chemistry community with a holistic concept for access to research data. To this end, the overarching objective is the development and maintenance of a national research data infrastructure for the research domain of chemistry in Germany, and to enable innovative and easy to use services and novel scientific approaches based on re-use of research data. NFDI4Chem intends to represent all disciplines of chemistry in academia. We aim to collaborate closely with thematically related consortia. In the initial phase, NFDI4Chem focuses on data related to molecules and reactions including data for their experimental and theoretical characterisation. This overarching goal is achieved by working towards a number of key objectives: Key Objective 1: Establish a virtual environment of federated repositories for storing, disclosing, searching and re-using research data across distributed data sources. Connect existing data repositories and, based on a requirements analysis, establish domain-specific research data repositories for the national research community, and link them to international repositories. Key Objective 2: Initiate international community processes to establish minimum information (MI) standards for data and machine-readable metadata as well as open data standards in key areas of chemistry. Identify and recommend open data standards in key areas of chemistry, in order to support the FAIR principles for research data. Finally, develop standards, if there is a lack. Abstract

Objectives/Hypothesis: TNM‐classification inadequately estimates patient‐specific overall survival (OS). We aimed to improve this by developing a risk‐prediction model for patients with advanced larynx cancer. Study Design: Cohort study. Methods: We developed a risk prediction model to estimate the 5‐year OS rate based on a cohort of 3,442 patients with T3T4N0N+M0 larynx cancer. The model was internally validated using bootstrapping samples and externally validated on patient data from five external centers (n = 770). The main outcome was performance of the model as tested by discrimination, calibration, and the ability to distinguish risk groups based on tertiles from the derivation dataset. The model performance was compared to a model based on T and N classification only. Results: We included age, gender, T and N classification, and subsite as prognostic variables in the standard model. After external validation, the standard model had a significantly better fit than a model based on T and N classification alone (C statistic, 0.59 vs. 0.55, P < .001). The model was able to distinguish well among three risk groups based on tertiles of the risk score. Adding treatment modality to the model did not decrease the predictive power. As a post hoc analysis, we tested the added value of comorbidity as scored by American Society of Anesthesiologists score in a subsample, which increased the C statistic to 0.68. Conclusions: A risk prediction model for patients with advanced larynx cancer, consisting of readily available clinical variables, gives more accurate estimations of the estimated 5‐year survival rate when compared to a model based on T and N classification alone.
