ObjectiveTo investigate the contamination status of Burkholderia gladioli in commercially available mushroom products in Chengdu， analyze its pathogenic characteristics， and provide support for food safety risk monitoring.MethodsAccording to GB 4789.29—2020， the identification of suspected colony by microbial mass spectrometry was added. One hundred and twenty-one mushroom foods were detected， and the whole genome of the isolated strains was sequenced to analyze their genetic characteristics and the carrying status of bon genes related to the biosynthesis of Bongkrekic acid.ResultsThe positive rate of Burkholderia gladioli in 121 samples was 50.41% （61/121）， with a positive rate of 67.14% （47/70） for Tremella； 10 strains of Burkholderia gladioli isolated from 4 food samples carried bon genes clusters； There was a dominant clone group that could mainly contaminated Tremella， without carrying the bon gene clusters.ConclusionTremella could be easily contaminated by Burkholderia gladioli， so it is necessary to strengthen the risk monitoring for the detection of this pathogen in key foods， especially for the strains carrying the bon gene clusters.

Warfarin is an anticoagulant commonly used as an oral drug in preventing and treating thromboembolic diseases. The international normalized ratio (INR) is a clinical monitoring anticoagulation intensity index that adjusts the dose based on important references. In particular, INR value must be strictly monitored when warfarin is used for anticoagulation therapy in infected patients. Herein, we report a 54-year-old female patient diagnosed with pulmonary embolism and venous thrombosis of the lower limbs. After the warfarin administration, the INR was always substandard. The patient did not take other warfarin-interacting drugs or foods during the hospital stay. Metagenome next-generation sequencing suggested a bloodstream infection caused by Escherichia coli, which was further confirmed by blood culture. After meropenem administration for anti-infective treatment, the INR value rose rapidly to a standard level. Considering the lack of relevant reports, this case is the first report of potential interaction between E. coli and warfarin. Further, in patients with thromboembolic diseases complicated by infection, antibiotics should be chosen reasonably with close monitoring of the INR to avoid the interaction of warfarin and antibiotics and to ensure the effectiveness and safety of warfarin treatment.

ObjectiveIn order to discover the pulsed field gel electrophoresis (PFGE) serotypes and drug resistance of Salmonella agona (S. agona) in Sichuan Province during 2008 to 2018, so as to understand the contamination status of S. agona and provide reference data for outbreak warning, traceability investigation and antibiotic use strategy.MethodsA total of 61 S. agona strains after the serological and biochemical identification were analyzed by PFGE and tested for minimal inhibitory concentration of 14 antibiotics.ResultsThe isolates of S. agona were divided into 41 PFGE types, some strains isolated from clinical cases in different years and regions had the same PFGE type, one pork isolate had the same PFGE type as some clinical isolates. The antibiotic resistance test indicated 28 of the isolates were sensitive to 14 kinds of antibiotics, the other 33 strains showed different degrees of drug resistance, nearly half of the strains were insensitive to tetracycline; and there were 16 multidrug-resistant strains. All of the isolates were sensitive to imipenem.ConclusionThe PFGE types of S. agona isolated in Sichuan from 2008 to 2018 showed diversity, and they were sporadic prevalent in Sichuan Province. The drug resistance was serious with an increasing trend.

Introduction: Pulmonary embolism (PE), the most serious presentation of venous thromboembolism (VTE), is associated with a high rate of mortality and expense. Clinical studies on pregnant women with PE are scarce. The aim of this study was to analyze the clinical impact of fibrinolytic enzyme activation inhibitor-1 (PAI-1) 4G/5G genetic polymorphisms, methylenetetrahydrofolate reductase (MTHFR) rs1801131 (A1298C) and rs1801133 (C677T) genetic polymorphisms, and establish a predictive model for pregnant women. Material and Methods: Between September 2022 and August 2023, 53 pregnant women with PE were enrolled. Using the propensity score matching method, 106 consecutive pregnant women without VTE were 1:2 matched. The relevant patient data were collected, and the susceptibility genes for PE were detected to determine genetic polymorphisms, and PE susceptibility in pregnant women, as well as to develop predictive models. Results: Our study showed that 4G/4G homozygous mutations increased the risk of pregnant PE fourfold (OR = 4.46, 95% CI = 1.59– 12.50, P = 0.004), whereas the 4G allele mutation increased the risk twofold (OR = 2.33, 95% CI = 1.35– 4.04, P = 0.002). A nomogram was established to predict the risk of pregnant women with PE by four predictive features including PAI-1 genetic polymorphisms, international normalized ratio (INR), antithrombin-III (AT-III) activity, and platelet count (PLT). The area under the curve (AUC) of the nomogram was 0.821 (0.744– 0.898). The AUC of the internal validation group was 0.822 (0.674– 0.971). Decision curve analysis revealed that the nomogram has a higher net benefit in the following threshold: probability interval of ≥ 15%. Conclusion: The PAI-1 4G/4G genotype is an independent risk factor for pregnant women with PE; furthermore, the presence of the 4G allele can increase the risk of PE. The ongoing COVID-19 pandemic still threatens human health around the world. The methyltransferases (MTases) of SARS-CoV-2, specifically nsp14 and nsp16, play crucial roles in the methylation of the N7 and 2′-O positions of viral RNA, making them promising targets for the development of antiviral drugs. In this work, we performed structure-based virtual screening for nsp14 and nsp16 using the screening workflow (HTVS, SP, XP) of Schrödinger 2019 software, and we carried out biochemical assays and molecular dynamics simulation for the identification of potential MTase inhibitors. For nsp14, we screened 239,000 molecules, leading to the identification of three hits A1–A3 showing N7-MTase inhibition rates greater than 60% under a concentration of 50 µM. For the SAM binding and nsp10-16 interface sites of nsp16, the screening of 210,000 and 237,000 molecules, respectively, from ZINC15 led to the discovery of three hit compounds B1–B3 exhibiting more than 45% of 2′-O-MTase inhibition under 50 µM. These six compounds with moderate MTase inhibitory activities could be used as novel candidates for the further development of anti-SARS-CoV-2 drugs. [ABSTRACT FROM AUTHOR]

The CDPK family genes play crucial roles in signal transduction pathways during plant development and stress response. In this study, we comprehensively analyzed the CDPK family genes in mustard (Brassica juncea L.), resulting in the identification of 101 genes (BjuCDPK1–101) located on chromosomes AA_Chr01 to BB_Chr08. RNA-seq analysis showed that most BjuCDPK genes were predominantly expressed in the root and flower bud, suggesting their organ-specific expression patterns. Furthermore, the expression levels of most BjuCDPK genes were significantly altered by cold stress. The promoter regions of BjuCDPK genes had abundant cis-acting elements related to hormones and stress. The interaction network analysis predicted that BjuCDPK proteins respond to abiotic stress probably via interaction with ABF1, ABF4, and/or DI19. Our findings provide valuable information for further the investigation of the cold stress adaption of mustard via the CDPK signaling pathway. [ABSTRACT FROM AUTHOR]
