The purpose of this study was to investigate the effect of extraction and application of okra gum as an aqueous film coating agent. Powdered okra pods dispersed in demineralized water was heated at 80 ± 2°C for 30 minutes in the presence of sodium chloride. The filtrate was successively centrifuged at 4000 rpm for 30, 60, or 120 minutes and freeze dried. The samples were used as film former at different concentrations in aqueous film coating operations. Near infrared (nIR) absorption spectra, photomicrographs, and some physicochemical properties of the coated tablets were evaluated. The okra gum samples had different nIR spectra and possessed good processing and application quality due to relatively low viscosity. A six-fold concentration of this gum from the novel extraction yielded glossy theophylline tablets within a short time. A t (18) = 2.895, P < 0.005, t critical = 1.734 were obtained for the independent analysis of the hardness of core and coated theophylline tablets. A 3.0% concentration of the okra samples at a flow rate of 3 ml/min for 100 minutes showed that F = 3.798, DF = 29, P < 0.035, F critical = 3.354 in tablet hardness among samples and F = 15.632, DF = 29, P < 0.0001, F critical = 2.152 were obtained on film thickness among tablet samples during the coating and drying operation. Novel extraction process enhanced the film coating potential of okra gum by delivering more solids on the substrate at a shorter time with improved operation efficiency. [ABSTRACT FROM AUTHOR]

Background: Grewia gum has received attention as a polymeric pharmaceutical excipient in the recent times, being employed as a suspending, film coating, mucoadhesive, and binding agent. The low aqueous solubility, however, has limited its characterization and application. Objective: The purpose of this study was to fractionate and evaluate some physicochemical properties of the gum. Materials and Methods: Aqueous dispersion of the gum was treated at 80°C for 30 min in the presence of sodium chloride and was subsequently fractionated by successively centrifuging it at 3445 rpm for 30 min. Skeletal density, solubility, particle size, and rheological as well as thermal characteristics of the fractions were evaluated. The 1H nuclear magnetic resonance (NMR) and near infrared (NIR) profiles of the fractions were also investigated. The solubility of the gum increased up to fourfold while the viscosity decreased from 244 to as low as70 cP at 40 rpm with some fractions. Results: Grewia gum and the fractions showed good thermal stability exhibiting no thermal events, but charred irreversibly at 297°C irrespective of the fraction. The molecular weight averages by weight and by number of the fractions were between 233,100 and 235,000. The 1H nuclear magnetic resonance (NMR) spectra showed broad peaks. The NMR and NIR spectra suggested the presence of -OH and -OCH3 functional groups in this gum. Conclusion: The fractionation improved solubility and facilitated further investigations on its characteristics that may have implication on its processing, application, and optimization as a potential pharmaceutical excipient. The plant Irvingia gabonensis contains lipids and polymer extractives that can be a good source of excipients for oral pharmaceutical formulations. These constituents are usually extracted from the seeds with the aid of either organic solvents or enzymes. The purpose of this paper was to describe a novel, simpler, cheaper, and safer method for the simultaneous extraction of lipids and gum from I. gabonensis. A 100.0 g of seeds of I. gabonensis in 200 mL deionized water was heated at 78°C in the presence of 2.0 g sodium chloride for 1 h, and the mixture was allowed to stand for the lipids to be separated and removed by filtration. The samples were air dried at25-30°C. The liquid fraction was centrifuged at 3445 rpm for 30 min, and the supernatant portion containing the gum was freeze dried. The samples were investigated for their true, bulk, and tapped densities as well as particle size and particle size distributions. The rheological, and near infrared absorption spectra as well as thermal behavior of the samples were also studied. The lipid and the polymeric components of I. gabonensis seeds were successfully extracted simultaneously. The true densities of the fatty and gum components were, respectively, 1.000 and 1.544 g/cm3. The melting point of the fat was 40°C and Tg of the gum was 232°C. These values were similar to those described in the literature for the components using the traditional method, demonstrating the effectiveness and efficiency of this simple novel method. Oral candidiasis (OC), caused by the fungal pathogen Candida albicans, is the most common opportunistic infection in HIV(+) individuals and other immunocompromised populations. The dramatic increase in resistance to common antifungals has emphasized the importance of identifying unconventional therapeutic options. Antimicrobial peptides have emerged as promising candidates for therapeutic intervention due to their broad antimicrobial properties and lack of toxicity. Histatin-5 (Hst-5) specifically has exhibited potent anticandidal activity indicating its potential as an antifungal agent. To that end, the goal of this study was to design a biocompatible hydrogel delivery system for Hst-5 application. The bioadhesive hydroxypropyl methylcellulose (HPMC) hydrogel formulation was developed for topical oral application against OC. The new formulation was evaluated in vitro for gel viscosity, Hst-5 release rate from the gel, and killing potency and, more importantly, was tested in vivo in our mouse model of OC. The findings demonstrated a controlled sustained release of Hst-5 from the polymer and rapid killing ability. Based on viable C. albicans counts recovered from tongues of treated and untreated mice, three daily applications of the formulation beginning 1 day postinfection with C. albicans were effective in protection against development of OC. Interestingly, in some cases, Hst-5 was able to clear existing lesions as well as associated tissue inflammation. These findings were confirmed by histopathology analysis of tongue tissue. Coupled with the lack of toxicity as well as anti-inflammatory and wound-healing properties of Hst-5, the findings from this study support the progression and commercial feasibility of using this compound as a novel therapeutic agent., (Copyright © 2016, American Society for Microbiology.
