Abstract

Pulmonary drug delivery system is one of the most intriguing and beneficial route for drug delivery as it offers several advantages over the conventional oral route of administration. Many devices are being studied to achieve optimum drug delivery through pulmonary route. Nebulizers, dry powder inhalers, pressurized metered dose inhalers are developed and marketed to target delivery through the pulmonary system. Nebulizers are devices which convert liquid into aerosols of appropriate size for inhalation which can efficiently reach the pulmonary region. The different types of nebulizers include jet nebulizer, ultrasonic nebulizer, vibrating mesh nebulizer. They are further subdivided according to the modification in their structure. The advancement in science has led to the development of more ‘intelligent nebulizers’ which include I-neb nebulizer and AKITA nebulizer, they monitor the breathing pattern of the patient and release the drug accordingly resulting in minimal wastage of the drug. Nebulizers sales have shown growth in the market owing to an increase in the incidences of chronic obstructive pulmonary disorder, asthma in the young and elderly, thus, a combination of smart nebulizer and microparticle formulation will help in achieving the desired results. 7. Formulation development, characterization and assessment of In-Vitro antifungal efficacy against Candida albicans of diallyl disulphide liposomal gel using 32 factorial design

Abstract

Objective: The aim of this study was to develop the diallyl disulphide (DADS) liposomal dispersion and further the dispersion was to formulate into gel. Methods: The DADS liposomal system was developed using thin film hydration method. 32 factorial design was employed to study the effect of various factors to obtain the optimized liposomal system. Further, it was converted to gel using suitable gelling agent. The optimized liposomal system and gel formulations were evaluated for various parameters. The optimized gel was evaluated for its in-vitro antifungal efficacy. The stability studies for gel were carried as per International conference on harmonisation (ICH) guidelines. Results: The optimized liposomal system was having vesicle size of 208.6nm, % entrapment was 91.7±1.16% and loading was 11.12±0.23%. % drug permeation was 61.10±0.85%. The zeta potential showed moderate stability. The optimized DADS liposomal gel showed satisfactory results. Ex-vivo permeation studies showed that DADS liposomal gel possesses sustained release and drug retention study proved better retention of DADS from liposomal gel than conventional gel in skin. The stability studies showed DADS liposomal gel was stable for 30 days at 25℃/60%RH, 40℃/75%RH and refrigeration (5±3℃). Conclusion:  The results from the present study show that the sustained release profile of optimized DADS liposomal gel. The retention of DADS from liposomal gel was more than from conventional gel which helps in better pharmacological activity with least side effects. 9. PREPARATION AND EVALUATION OF IRBESARTAN-CYCLODEXTRIN INCLUSION COMPLEXES

Abstract

In the present work effect of methyl-β-cyclodextrin (MβCD) on physical properties and dissolution rate of Irbesartan (IRB) was studied.Based on AL-type ofphase solubility diagram obtained, solid binary systems of the drug with MβCD were prepared in 1:1 molar ratio by various methods. Complexes were characterized using Differential Scanning Calorimetry and powder X-Ray Diffractometry. It could be concluded that IRB can form inclusion complex with MβCD. The dissolution profiles of inclusion complexes were compared with those of IRB alone and the physical mixture. The dissolution rate of IRB was increased remarkably by MβCD inclusion complexation.

Abstract

The aim of the present investigation was to develop a modified release effervescent floating drug delivery system of famotidine for 12 h dosage regimen to improve its bioavailability. Effervescent floating tablets were prepared by direct compression method taking into account its advantages over wet granulation by using directly compressible excipients like Carbopol® 71G and Cellactose® 80. The incorporation of sodium bicarbonate aided in the buoyancy with effervescent approach. The prepared tablets were evaluated for floating lag time (FLT), total floating time (TFT), in vitro drug release along with general parameters. 23 factorial design was used for optimization. The tablets showed desired release of more than 98 % over the period of 12 h which may increase bioavailability of selected candidate. The release of famotidine was found to be influenced by the polymer concentration.
