Abstract Background With increasing global limate warm, high temperature (HT) is one of limiting factors for soybean yield and quality. Exploring HT resistance-related functional genes and their corresponding molecular mechanisms is of great value. In our previous report, compared with HD14 (HT sensitive), JD21 is an HT-resistant variety, and further analysis of the transcriptome and proteome has revealed the HT tolerance mechanism of JD21 anthers. We found that compared with those of HD14 (28.72%), the leaves of JD21 also exhibited HT resistance, and the degree of leaf wilting in JD21 plants after HT stress treatment was 11.02%; however, the regulatory mechanism of the response of JD21 to HT stress is still unclear. Results In this study, comparative transcriptome analysis of JD21 and HD14 soybean leaves after HT stress and field control plants was performed by RNA-seq analysis. The results showed that the number of upregulated differentially expressed genes (DEGs) in JD21 and HD14 was greater than the number of downregulated DEGs after HT stress, and the number of up- or down-regulated DEGs in JD21 was higher than those of HD14. Bioinformatics analysis revealed that many DEGs were involved in various molecular functions and metabolic pathways. QRT‒PCR analysis verified that the gene expression pattern results determined via RNA–seq was reliable. In addition, through analysis of gene expression level and conserved domain, 18 key candidate genes related to the response of soybean leaves to HT stress were screened. Conclusions This study systematically revealed the regulation mechanism of soybean leaves molecular transcription level by RNA-seq, and several key candidate DEGs (transcription factor, HSPs, HSFs, GmCYP78A6, etc.) involved in the response to HT stress were identified based on the bioinformatics analysis. The results provided a theoretical basis for studying the response mechanism of soybean leaves to HT stress.

Abstract Sand gradation is an important reference factor for concrete proportion design, which has a significant impact on the strength and workability of concrete. The efficiency of using the traditional vibrating sieving method to measure the gradation is low, and the machine vision-based sand gradation inspection method can realize the automation of sand gradation inspection, which can significantly improve the inspection efficiency and reduce the inspection cost. The particle size of construction sand is usually between 0.075 and 4.75 mm, and to capture images of sand below 0.15 mm requires a high-resolution camera that can only maintain a small object distance and a small shooting field of view, so it is difficult for the image acquisition system to capture images of all the sand particles, and this is a challenging problem in sand grading inspection. In order to solve this problem, this study proposes a sand gradation detection method based on localized sampling, and develops a hardware system for localized sampling and a software system for gradation detection. The hardware system uses a flexible vibrating disk to uniformly disperse the sand particles, realizing random local sampling of sand particles, and calculating the grain size gradation of the test samples based on the local data of multiple local sampling. The experimental results show that the local sampling gradation and the sample gradation have a similar distribution, and the local gradation detection error of multiple sampling is smaller. This study provides an effective method for the automated detection of sand grain size gradation between 0.075 and 4.75 mm, which can significantly increase the frequency of construction sand gradation detection, improve the quality control level of construction sand, and improve the quality of construction.

Abstract Background Under global warming, high temperature (HT) has become a major meteorological factor affecting soybean production. To explore the candidate genes and regulatory mechanism of the soybean bud response to HT stress, previously identified as HT-tolerant (‘Handou14’; HD14) and HT-sensitive (‘Jiadou36’; JD36) were treated for 5 days in an artificial climate incubator either with HT (43 °C (day)/ 33 °C (night), 16 h light/8 h darkness) or the non-stress growth condition (25 °C, 16 h light/8 h darkness) as the control at the bud stage were used as experimental materials in this study. After HT treatment, changes in physiological indicators including hypocotyl length, enzyme activity and hormone content were detected; at the same time, the cotyledons, hypocotyls, and main roots were collected for transcriptome sequencing analysis. Analyzing the mechanisms of HT stress response in the bud stage of HD14 and JD36 at physiological and transcription levels. Results Analysis of physiological indicator showed that the activities of superoxide dismutase (SOD) were significantly increased 47.4% and 41.2% in the cotyledon of HD14 and the main root of JD36, and the contents of peroxidase (POD) were significantly increased 61.5% and 125% in the hypocotyl of HD14 and JD36; the contents of malonaldehyde (MDA) were significantly increased 44.8% and 22.2% in the main root of HD14 and JD36 after HT treatment. Abstract To elucidate the specific and shared genetic background of schizophrenia (SCZ) and bipolar disorder (BPD), this study explored the association of polygenic liabilities for SCZ and BPD with educational attainment and cognitive aging. Among 106,806 unrelated community participants from the Taiwan Biobank, we calculated the polygenic risk score (PRS) for SCZ (PRSSCZ) and BPD (PRSBPD), shared PRS between SCZ and BPD (PRSSCZ+BPD), and SCZ-specific PRS (PRSSCZvsBPD). Based on the sign-concordance of the susceptibility variants with SCZ/BPD, PRSSCZ was split into PRSSCZ_concordant/PRSSCZ_discordant, and PRSBPD was split into PRSBPD_concordant/PRSBPD_discordant. Ordinal logistic regression models were used to estimate the association with educational attainment. Linear regression models were used to estimate the associations with cognitive aging (n = 27,005), measured by the Mini-Mental State Examination (MMSE), and with MMSE change (n = 6194 with mean follow-up duration of 3.9 y) in individuals aged≥ 60 years. PRSSCZ, PRSBPD, and PRSSCZ+BPD were positively associated with educational attainment, whereas PRSSCZvsBPD was negatively associated with educational attainment. PRSSCZ was negatively associated with MMSE, while PRSBPD was positively associated with MMSE. The concordant and discordant parts of polygenic liabilities have contrasting association, PRSSCZ_concordant and PRSBPD_concordant mainly determined these effects mentioned above. PRSSCZvsBPD predicted decreases in the MMSE scores. Using a large collection of community samples, this study provided evidence for the contrasting effects of polygenic architecture in SCZ and BPD on educational attainment and cognitive aging and suggested that SCZ and BPD were not genetically homogeneous.

Abstract Enhancing and strengthening food production capacity has always been a top priority in agricultural research, serving as a cornerstone for ensuring national food security and stable economic development. This study, based on panel data spanning from 2011 to 2021 across 30 provinces in China, delves into the mechanism through which the digital economy impacts food production capacity. Employing a double fixed effect model, a mediation effect model, and a panel threshold model, we uncover several key findings: The digital economy significantly boosts food production capacity, with robustness tests affirming the reliability of our results. Mechanism analysis reveals that the digital economy enhances food production capacity by elevating total factor productivity and bolstering agricultural resilience. The threshold effect underscores that urbanization levels exhibit a single-threshold impact, wherein the influence of the digital economy on food production capacity intensifies upon crossing this threshold. Heterogeneity analysis reveals that the digital economy significantly boosts food production capacity in central and primary grain-producing regions, while its impact is comparatively weaker in the eastern and western regions, as well as in non-primary grain-producing areas. In summary, this research sheds light on the pivotal role of the digital economy in augmenting food production capacity, offering valuable insights into regional variations and thresholds in its impact across China.

Abstract Background Both serum uric acid (SUA) levels and body mass index (BMI) are recognized as important risk factors for hypertension. The current study aimed to investigate the interaction effects between SUA levels and overweight (defined as BMI ≥ 24 kg/m2 in Chinese) on the incidence of hypertension among Chinese adults. Methods 1124 hypertensive participants and 7283 non-hypertensive participants, extracted from the China Health and Nutrition Survey (CHNS), were analyzed. Participants were categorized based on their SUA levels and BMI, to investigate the interaction effects between SUA levels and overweight on hypertension. Results In comparison with the reference group (BMI 6.39 mg/dL) of SUA among Chinese adults. Therefore, strategies to lower SUA levels could be considered as a potential approach to mitigate hypertension risk in overweight individuals within this specific subgroup.

Abstract To investigate the effect of waste glass material particle sizes on the mechanical and durability properties of concrete, a two-phase experimental approach was conducted. First, comprehensive tests were performed to examine the effects of glass sand and glass powder of different particle sizes on concrete performance. Subsequently, based on the experimental results, an orthogonal test was designed to optimize the replacement amounts of composite particle sizes. The results indicated that an appropriate amount of glass sand can enhance the mechanical properties and durability of concrete, while excessive amounts or larger particle sizes may have adverse effects. The pozzolanic effect of glass powder also contributes to performance improvement, but the replacement rate should be limited to 20%. Under composite particle size conditions, the compressive, tensile, and shear strengths of concrete increased by 35.56%, 21.74%, and 13.79%, respectively, while durability significantly improved, with water absorption reduced by 20.73% and chloride ion permeability decreased by 63.10%. At a total replacement rate of 20%, the optimal proportions were determined to be 2.86% for 0.6 mm glass sand, 1.43% for 1.18 mm glass sand, 8.57% for 50–60 μm glass powder, and 7.14% for 60–70 μm glass powder. The incorporation of composite particle sizes improved the microstructure of concrete, thereby enhancing its mechanical and durability properties.

Abstract Despite a rapid expansion of Porcine reproductive and respiratory syndrome virus (PRRSV) sublineage 8.7 over recent years, very little is known about the patterns of virus evolution, dispersal, and the factors influencing this dispersal. Relying on a national PRRSV surveillance project established over 20 years ago, we expand the available genomic data of sublineage 8.7 from China. We perform independent interlineage and intralineage recombination analyses for the entire study period, which showed a heterogeneous recombination pattern. A series of Bayesian phylogeographic analyses uncover the role of Guangdong as an important infection hub within Asia. The spatial spread of PRRSV is highly linked with a composite of human activities and the heterogeneous provincial distribution of the swine industry, largely propelled by the smaller-scale Chinese rural farming systems in the past years. We sequence all four available modified live vaccines (MLVs) and perform genomic analyses with publicly available data, of which our results suggest a key “leaky” period spanning 2011–2017 with two concurrent amino acid mutations in ORF1a 957 and ORF2 250. Overall, our study provides an in-depth overview of the evolution, transmission dynamics, and potential leaky status of HP-PRRS MLVs, providing critical insights into new MLV development.

Abstract The tight sandstone reservoris are important exploration and production target in the Xujiahe Formation in Xinchang area of Western Sichuan. Due to the various lithofacies and diagenetic alteration in distributary channel belt sandstones, the reservoirs exhibit strong heterogeneity. The strong heterogeneity makes the identification of petrologically superior sandbodies a great challenge. In order to better understand reservoir quality, this study constructs a model between diagenetic alterations and reservoir quality within distributary channel belts. 73 plug samples were taken from 17 wells located in the channel belt. An integration of thin sections, X-ray diffraction, carbon and oxygen isotope ratios, cathodoluminescence, and micro-CT were analysed for the genesis and distribution of reservoir quality. The results show that subaqueous distributary channels are formed under various hydrodynamics, resulting in different lithofacies. The thicknesses of channels can be divided into less than 2 m, 2–6 m, and more than 6 m. The diagenetic alteration in lithofacies were identified. Coarse-grained sandstones with low content of rock fragments are relatively weakly compacted with less calcite and chlorite cementation. Fine-grained sandstones with high content of rock fragments are strongly compacted with calcite cementation during early diagenesis. The dissolution pores are related to the soluble mineral content and the distribution of mudstones. The subaqueous distributary channels formed in high-energy, moderate-energy, and low-energy hydrodynamic settings have experienced different diagenetic intensities. Additionally, fractures can effectively improve the reservoir properties. The distribution of good quality reservoirs is controlled by the combined influence of diagenetic heterogeneity and fractures. This would help to promote better understanding of reservoir quality within the subaqueous distributary channel belts in braided-river delta front.

Abstract Left ventricular (LV) hypertrophy (LVH) is frequently observed in patients with hypertension (HTN). LV myocardial work (MW) has recently emerged as a non‐invasive method to assess systolic myocardial deformation relative to afterload conditions. The authors investigated the characteristics of myocardial work with different degrees of LVH in HTN patients. From December 2020 to February 2024, 255 HTN patients and 26 healthy controls undergoing transthoracic echocardiography were included in the current study. Hypertension patients were divided into quintile groups based on left ventricular mass index (LVMI), for the first to fourth LVMI quantiles, global work index (GWI) and global constructive work (GCW) were higher compared to the control group, but the difference was not statistically significant. In the sixth LVMI quantile, GWI and GCW showed a significant decrease. The restricted cubic splines showed that both GWI and GCW exhibited an inverted U‐shaped relationship with LVMI. In view of the energy occurrence structure of “rich coal, poor oil and little gas” in China, and the situation that coal will occupy the dominant position of energy consumption for a long time,The development history of frozen shaft mechanized drilling and blasting is reviewed in the field of coal mining in China. As the main construction method of shaft construction, the application of dynamite greatly improves the drilling efficiency of frozen shaft. The application of supporting large mechanized equipment such as umbrella drilling, rock grabbing machine loading, large derrick and equipment hoisting, and large formwork building, greatly improve the wellbore completion speed; Shaft drilling rig, reverse drilling rig and shaft boring machine as special shaft sinking methods under special formation conditions, also have their own application range. Aiming at the characteristics of water-rich weakly consolidated formation and the development trend of smart mine in western China, there are shortcomings in using freezing & borehole-blasting method and large drilling equipment to develop the shaft of weakly consolidated formation at kilometer level, while using freezing & shaft boring machine is one of the trends of ultra-deep shaft construction. Combining with the construction cases of freezing & shaft boring machine abroad, reasonable suggestions are put forward for shaft excavation of water-rich weak consolidated rock formation in western China. The freezing wall thickness and strength are calculated according to the current design theory, the permanent shaft support adopts double-layer reinforced concrete wall structure, the rock-breaking form of shaft boring machine under low temperature environment, and the method of hierarchical slag discharge to realize the parallel operation of digging - supporting - transporting. Finally, the next research focus of this construction method is put forward. The synergistic mechanism between shaft wall structure and weakly consolidated surrounding rock should be clarified to realize optimization of shaft wall structure, and the support - operation cooperation mode of shaft boring machine to improve shaft excavation and support efficiency and improve informatization level. Realize shaft driving towards mechanization and intelligence.

Abstract Background It has been proposed that having a psychiatric disorder could increase the risk of developing a gastrointestinal disorder, and vice versa. The role of familial coaggregation and shared genetic loading between psychiatric and gastrointestinal disorders remains unclear. Methods This study used the Taiwan National Health Insurance Research Database; 4,504,612 individuals born 1970–1999 with parental information, 51,664 same-sex twins, and 3,322,959 persons with full-sibling(s) were enrolled. Genotyping was available for 106,796 unrelated participants from the Taiwan Biobank. A logistic regression model was used to examine the associations of individual history, affected relatives, and polygenic risk scores (PRS) for schizophrenia (SCZ), bipolar disorder (BPD), major depressive disorder (MDD), and obsessive-compulsive disorder (OCD), with the risk of peptic ulcer disease (PUD), gastroesophageal reflux disease (GERD), irritable bowel syndrome (IBS), and inflammatory bowel disease (IBD), and vice versa. Results Here we show that parental psychiatric disorders are associated with gastrointestinal disorders. Full-siblings of psychiatric cases have an increased risk of gastrointestinal disorders except for SCZ/BPD and IBD; the magnitude of coaggregation is higher in same-sex twins than in full-siblings. The results of bidirectional analyses mostly remain unchanged. PRS for SCZ, MDD, and OCD are associated with IBS, PUD/GERD/IBS/IBD, and PUD/GERD/IBS, respectively. PRS for PUD, GERD, IBS, and IBD are associated with MDD, BPD/MDD, SCZ/BPD/MDD, and BPD, respectively. Conclusions There is familial coaggregation and shared genetic etiology between psychiatric and gastrointestinal comorbidity. Individuals with psychiatric disorder-affected relatives or with higher genetic risk for psychiatric disorders should be monitored for gastrointestinal disorders, and vice versa.

Abstract Background The syncytiotrophoblast (SCT) layer in the placenta serves as a crucial physical barrier separating maternal-fetal circulation, facilitating essential signal and substance exchange between the mother and fetus. Any abnormalities in its formation or function can result in various maternal syndromes, such as preeclampsia. The transition of proliferative villous cytotrophoblasts (VCT) from the mitotic cell cycle to the G0 phase is a prerequisite for VCT differentiation and their fusion into SCT. The imprinting gene P57Kip2, specifically expressed in intermediate VCT capable of fusion, plays a pivotal role in driving this key event. Moreover, aberrant expression of P57Kip2 has been linked to pathological placental conditions and adverse fetal outcomes. Methods Validation of STK40 interaction with P57Kip2 using rigid molecular simulation docking and co-immunoprecipitation. STK40 expression was modulated by lentivirus in BeWo cells, and the effect of STK40 on trophoblast fusion was assessed by real-time quantitative PCR, western blot, immunofluorescence, and cell viability and proliferation assays. Co-immunoprecipitation, transcriptome sequencing, and western blot were used to determine the potential mechanisms by which STK40 regulates P57Kip2. Results In this study, STK40 has been identified as a novel interacting protein with P57Kip2, and its expression is down-regulated during the fusion process of trophoblast cells. Overexpressing STK40 inhibited cell fusion in BeWo cells while stimulating mitotic cell cycle activity. Further experiments indicated that this effect is attributed to its specific binding to the CDK-binding and the Cyclin-binding domains of P57Kip2, mediating the E3 ubiquitin ligase COP1-mediated ubiquitination and degradation of P57Kip2. Moreover, abnormally high expression of STK40 might significantly contribute to the occurrence of preeclampsia. Conclusions This study offers new insights into the role of STK40 in regulating the protein-level homeostasis of P57Kip2 during placental development.

Abstract To study the resistance of rice husk ash-rubber-fiber reinforced concrete (RRFC) to dry–wet cycle/chloride erosion under a hygrothermal environment, the optimal combination was selected by an orthogonal test. The peak strain, residual strain, and fatigue damage strength of the optimal group of RRFC samples under cyclic loading and unloading after dry–wet cycle/chloride erosion under different environments and temperatures were compared and analyzed. After that, microscopic analysis and anti-erosion mechanism analysis were carried out. The results show that the axial peak and residual strain of RRFC specimens increase continuously during the repeated loading–unloading process, and the increase of axial peak and residual strain in the first five cycles is the most obvious. Among them, RRFC has the most significant increase in axial peak strain after 14 dry–wet cycles, which is 11.73%. The rice husk ash reacted with Ca(OH)2 in the specimen to precipitate C—S—H gel, which improved the specimen’s corrosion resistance and fatigue resistance. The rubber in the specimen has high elasticity, which reduces the fatigue damage of the specimen during cyclic loading and unloading, thus showing higher fatigue failure strength.

Abstract Intravesical therapy (IT) is widely used to tackle various urological diseases. However, its clinical efficacy is decreased by the impermeability of various barriers presented on the bladder luminal surface, including the urinary mucus layer and the densely packed tissue barrier. In this study, we report a mucoadhesive-to-penetrating nanomotors-in-hydrogel system for urothelium-oriented intravesical drug delivery. Upon intravesical instillation, its poloxamer 407 (PLX) hydrogel gelated and adhered to the urothelium to prolong its intravesical retention. The urea afterwards diffused into the hydrogel, thus generating a concentration gradient. Urease-powered membrane nanomotors (UMN) without asymmetric surface engineering could catalyze the urea and migrate down this concentration gradient to deeply and unidirectionally penetrate the urothelial barrier. Moreover, the intravesical hybrid system-delivered gemcitabine could effectively inhibit the bladder tumor growth without inducing any side effect. Therefore, our mucoadhesive-to-penetrating nanomotors-in-hydrogel system could serve as an alternative to IT to meet the clinical need for more efficacious therapeutics for urological diseases. Abstract Abdominal aortic aneurysms (AAA) are a significant health concern in developed countries due to their considerable mortality rate. The crucial factor of the progression of AAA is the release of neutrophils and neutrophil extracellular traps (NETs). Magnetic particle imaging (MPI) is a new imaging technique that offers the capability to detect superparamagnetic iron oxide nanoparticles (SPION) with exceptional sensitivity. We aimed to investigate the functional imaging of MPI for the detection and monitoring of neutrophil infiltration within AAA. A novel multimodal imaging agent targeting neutrophils, PEG-Fe3O4-Ly6G–Cy7 nanoparticles (Ly6G NPs), were designed by coupling Fe3O4 nanoparticles with Ly6G antibodies and Cy7. The targeting and sensitivity of Ly6G NPs were assessed using MPI and fluorescence imaging (FLI) in the AAA mouse model. After the inhibition of NETosis, the degree of neutrophil infiltration and AAA severity were assessed using MPI with Ly6G NPs. Ly6G NPs accurately localized and quantitatively analyzed AAA lesion sites in mice using MPI/FLI/CT. Compared to the control group, elevated MPI and FLI signal intensities were detected at the abdominal aortic lesion site, and neutrophil infiltration and NETs accumulation were detected by histological analysis in the AAA models. After the inhibition of NETs accumulation in vivo, pathological damage in the abdominal aorta was significantly reduced, along with a decrease in the accumulation of Ly6G NPs and MPI signals. This multimodal MPI strategy revealed that nanoparticles targeting Ly6G can be used to detect neutrophil infiltration within AAA and monitor AAA severity.

Since low-light images are of poor quality and carry noise, resulting in insufficient contrast and brightness, unclear details in the image, and the problem of high cost of obtaining paired low-light image datasets, a no-reference low-light image enhancement method for feature map enhancement is proposed under the influence of the Mach band effect of biological vision. Firstly, the model uses the enhanced filter block (EFB) to enhance the feature of images and feature maps, which can suppress noise and enhance the feature details to improve the ability of the network to learn features. Then, skip connection is combined with efficient spatial attention (ESA) module to extract global context information and local regional features by fusing enhanced shallow features and deep features, which effectively retains image color information, avoids detail loss, and improves the generalization ability of the network. Finally, the pixel estimation curve is used to adjust the dynamic range of the pixels in the low-light image and enhance the brightness of the low-light image. Experimental results show that the PSNR, SSIM, LPIPS and NIQE of the image processed by the algorithm reach 17.709 dB, 0.657, 0.239 and 3.486 respectively. Compared with the existing mainstream algorithms, it can better achieve the purpose of image enhancement, and effectively improves the image brightness and detail information, while maintaining the naturalness of the image.

In this work, a novel graphite-based electrochemical filter was developed for the efficient and environmentally friendly treatment of uranium tailings leachate. The filter utilizes graphite particles as filler and carbon felt as a conductor, powered by a DC power supply and driven by a peristaltic pump. In a non-circulating continuous operation filter experiment, uranium tailings leachate was tested for uranium removal efficiency under different operating parameters such as voltage and residence time, using both synthetic wastewater and actual uranium mine wastewater. This study is based on the fundamental principles of metal electrochemical reduction. The efficacy of this device for the continuous flow treatment of uranium tailings leachate was systematically investigated. The mechanism of uranium tailings leachate treatment by the electrochemical filter was elucidated through electrochemical analysis and product characterization. The results highlight that voltage and hydraulic retention time are critical factors influencing the efficiency of electrochemical filter treatment. Specifically, 4.0 V and 10 hours were identified as the optimized voltage and suitable hydraulic residence time, respectively, for the treatment of uranium tailings leachate using the electrochemical filter, resulting in a uranium removal rate exceeding 97%. In actual uranium tailings leachate applications, the maximum extraction rate can reach 5275 μmol/(m3∙h), confirming the feasibility of electrochemical filter in removing uranium from actual uranium tailings leachate and its good selectivity in complex water quality applications, the device can be quickly applied to uranium tailings leachate collection facilities and achieve in-situ uranium extraction. The voltage increase can significantly enhance the uranium extraction performance and can be portablely deployed in uranium mine water. In-situ uranium processing and recovery is achieved in processing and collection facilities. Characterization techniques such as field emission scanning electron microscope-energy spectrum analyzer(FESEM-EDS) and X-ray photoelectron spectrometer(XPS) were employed to analyze the reaction products on the graphite particles, revealing that uranium was evenly distributed on the surface of graphite particles and exhibits a stable crystal structure, the main mechanism responsible for uranium removal was adsorption-reduction, with uranium recovery products deposited on the electrode surface in the form of a mixture of U(Ⅳ) and U(Ⅵ). A cost estimation shows that the operational cost of extracting uranium from uranium tailings leachate using this device falls within the range of 176.6 ￥/kgU to 587.5 ￥/kgU. This cost is significantly lower than that of conventional land-based uranium resource development, making it economically viable. Abstract Background An elevated endogenous cortisol level due to the peripartum stress is one of the risk factors of postpartum bovine uterine infections. Selenium is a trace element that elicits anti-inflammation and antioxidation properties. This study aimed to reveal the modulatory effect of selenium on the inflammatory response of primary bovine endometrial stromal cells in the presence of high-level cortisol. The cells were subjected to lipopolysaccharide to establish cellular inflammation. The mRNA expression of toll-like receptor 4 (TLR4), proinflammatory factors, and selenoproteins was measured with qPCR. The activation of NF-κB and MAPK signalling pathways was detected with Western blot and immunofluorescence. Results The pretreatment with sodium selenite (2 and 4 µΜ) resulted in a down-regulation of TLR4 and genes encoding proinflammatory factors, including interleukin (IL)-1β, IL-6, IL-8, tumour necrosis factor α, cyclooxygenase 2, and inducible nitric oxide synthase. Selenium inhibited the activation of NF-κB and the phosphorylation of mitogen-activated protein kinase kinase, extracellular signal-regulated kinase, p38MAPK and c-Jun N-terminal kinase/stress-activated protein kinase. The suppression of those genes and pathways by selenium was more significant in the presence of high cortisol level (30 ng/mL). Meanwhile the gene expression of glutathione peroxidase 1 and 4 was promoted by selenium, and was even higher in the presence of cortisol and selenium. Conclusions The anti-inflammatory action of selenium is probably mediated through NF-κB and MAPK, and is augmented by cortisol in primary bovine endometrial stromal cells.

Abstract Fluctuations in gas emission or concentration at working face represent primary indicators of impending coal and gas outbursts, making them essential for monitoring processes. However, the direct use of original signals for predictive purposes may cause false warnings due to the inability to recognize valuable potential information. This can result in decreased prediction accuracy. Additionally, gas emission or concentration is affected by potential hazards such as sensor calibration, borehole spraying, and air duct damage, leading to complexity of their behavior and recognition process. This paper proposes an intelligent recognition method leveraging YOLOv8 neural network to discern coal and gas outburst precursors and potential hazards. By employing continuous wavelet transform (CWT) on the gas concentration signal processed by critical slowing down (CSD) method, a two-dimensional time–frequency representation is generated. This representation is then fed into YOLOv8 model to recognize the outburst precursor characteristics and potential hazards. The research results show that YOLOv8-based intelligent recognition adeptly identifies the relevant precursor characteristics and potential hazards, enhancing both coal mine safety protocols and the accuracy of early outburst warning mechanisms.

Objectives: This study aims to examine the change in financial strain, health behaviors, and psychological well-being of family caregivers of older adults during the COVID-19 pandemic and explore the differences in mental health outcomes by gender, race, and relationship status. Methods: Using the 2020 National Health and Aging Trends Study COVID-19 supplement, our sample included 2026 family caregivers of older adults. Structural equation modeling was conducted. Results: Caregivers with financial strain showed worse mental health than those with no financial strain. Female or adult children caregivers reported significantly less time walking, more financial strain, and a higher level of negative mental health outcomes compared to male or spouse caregivers; non-White caregivers reported greater positive mental health outcomes compared to White caregivers during the pandemic. Discussion: Health professionals should consider the financial and mental health impact of COVID-19 among family caregivers when designing and delivering caregiver support programs. Innovation: This study provides nationally representative estimates of several important health behaviors and health outcomes for caregivers of older adults during and after the COVID-19 pandemic, helping to fill the knowledge gap about the characteristics of caregivers whose health and well-being were most affected by the pandemic.

Paediatric Mycoplasma pneumoniae pneumonia (MPP) is a heterogeneous disease with a diverse spectrum of clinical phenotypes. No studies have demonstrated the relationship between underlying endotypes and clinical phenotypes as well as prognosis about this disease. Thus, we conducted a multicentre prospective longitudinal study on children hospitalized for MPP between June 2021 and March 2023, with the end of follow-up in August 2023. Blood samples were collected and processed at multiple time points. Multiplex cytokine assay was performed to characterize serum cytokine profiles and their dynamic changes after admission. Cluster analysis based on different clinical phenotypes was conducted. Among the included 196 patients, the levels of serum IL-17A and IL-6 showed remarkable variabilities. Four cytokine clusters based on the two cytokines and four clinical groups were identified. Significant elevation of IL-17A mainly correlated with diffuse bronchiolitis and lobar lesion by airway mucus hypersecretions, while that of IL-6 was largely associated with lobar lesion which later developed into lung necrosis. Besides, glucocorticoid therapy failed to inhibit IL-17A, and markedly elevated IL-17A and IL-6 levels may correlate with lower airway obliterans. Our study provides critical relationship between molecular signatures (endotypes) and clustered clinical phenotypes in paediatric patients with MPP.

ABSTRACTStaphylococcus pseudintermedius (S. pseudintermedius) is a common pathogen that causes canine corneal ulcers. However, the pathogenesis remained unclear. In this study, it has been demonstrated that S. pseudintermedius invaded canine corneal epithelial cells (CCECs) intracellularly, mediating oxidative damage and pyroptosis by promoting the accumulation of intracellular reactive oxygen species (ROS) and activating the NLRP3 inflammasome. The canine corneal stroma was infected with S. pseudintermedius to establish the canine corneal ulcer model in vivo. The intracellular infectious model in CCECs was established in vitro to explore the mechanism of the ROS – NLRP3 signalling pathway during the S. pseudintermedius infection by adding NAC or MCC950. Results showed that the expression of NLRP3 and gasdermin D (GSDMD) proteins increased significantly in the infected corneas (p < 0.01). The intracellular infection of S. pseudintermedius was confirmed by transmission electron microscopy and immunofluorescent 3D imaging. The composite charge structure may have the potential to enhance the overall damage capability of the weapon. In this regard, three-dimensional LS-DYNA simulation software (dynamics calculation software from LSTC) was used to simulate the explosion process of charges in a partially confined space. The effects of different aluminized charge structures on the propagation and parameters of blast waves in a cubic cabin structure with a pressure relief port were obtained. The results showed that the reflected waves generated local pressure convergence at the junction and corner areas of adjacent walls in the cabin structure. The numbers of significant reflections of the blast waves at the center and corners of the wall were 3 and 2, respectively. The overpressure of the reflected wave at the corner was more than 3.2 times that at the center of the sidewall. Moreover, the number of reflections, overpressure ratio, and charge structure were not directly related. The advantages of composite structure charges in enhancing the overpressure and wave velocity of incident waves and the overpressure and impulse of reflected waves were not significant. However, the internal and external composite charge structure had more advantages than did the upper and lower structures, and its impulse TNT equivalent reached more than 70% of that of a single-structure explosive. The results can provide theoretical support and technical guidance for the design of charge structures and cabin structures.

The curing process regulates metabolite transformations of leaves and significantly influences the formation of tobacco quality. This study investigated the major physicochemical compositions and metabolic profiles under normal curing (NC), excessive curing (EC), and insufficient curing (IC) treatments. The results indicated that the contents of nicotine, nitrogen, potassium, and chlorine remained stable among treatments, while the sugar content in EC was significantly lower than in IC. LC-MS/MS identified 845 metabolites, with flavonoids as the most abundant class. Comparative analyses identified a series of differentially expressed metabolites (DEMs) among fresh leaf, NC, EC, and IC leaves at the end of 42°C, 54°C, and 68°C, respectively. At the end of 68°C, 256 up-regulated and 241 down-regulated common DEMs across treatments were isolated in comparison to fresh leaf, underscoring the consistency of metabolic changes during curing. Notably, nonivamide varied markedly across treatments, suggesting its potential as a key curing indicator. NC_68°C displayed 11 up-regulated and 17 down-regulated unique DEMs, differing from EC_68°C and IC_68°C, suggesting their potential availability in evaluating tobacco leaf quality. KEGG pathway analysis revealed temporal shifts in metabolic pathways, particularly those involved in secondary metabolite biosynthesis (such as flavonoids, flavones, flavonols) and amino acid metabolism, during the transition from yellowing to color-fixing. Correlation analysis isolated the top 25 DEMs correlated with curing degree and stage, which might play pivotal roles in the curing process and could serve as potential biomarkers for assessing curing degree and stage. Specifically, D-(+)-cellobiose displayed the strongest negative correlation with curing degree, while 5,7-dihydroxychromone exhibited the highest positive correlation coefficient. Furthermore, curcurbitacin IIa showed the highest positive correlation with curing stage, followed by hesperetin and 8-shogaol. Additionally, random forest analysis emphasized morellic acid as a core molecular metabolite across curing degrees, suggesting its potential as a biomarker. Debiased sparse partial correlation (DSPC) network analysis further pinpointed hispidulin as a key metabolite, underscoring its significance in elucidating flavonoid metabolism during the curing process. Collectively, this study enhances the understanding of metabolite transformations underlying tobacco curing processes and provides a valuable reference for optimizing curing strategies to achieve desired outcomes.

African swine fever virus (ASFV) is a nuclear cytoplasmic large DNA virus (NCLDV) that causes devastating hemorrhagic diseases in domestic pigs and wild boars, seriously threatening the development of the global pig industry. IFN-I plays an important role in the body's antiviral response. Similar to other DNA viruses, ASFV has evolved a variety of immune escape strategies to antagonize IFN-I signaling and maintain its proliferation. In this study, we showed that the ASFV early protein pK205R strongly inhibited interferon-stimulated genes (ISGs) as well as the promoter activity of IFN-stimulated regulatory elements (ISREs). Mechanistically, pK205R interacted with the intracellular domains of IFNAR1 and IFNAR2, thereby inhibiting the interaction of IFNAR1/2 with JAK1 and TYK2 and hindering the phosphorylation and nuclear translocation of STATs. Subsequently, we generated a recombinant strain of the ASFV-pK205R point mutation, ASFV-pK205R7PM. Notably, we detected higher levels of ISGs in porcine alveolar macrophages (PAMs) than in the parental strain during the early stages of ASFV-pK205R7PM infection. Moreover, ASFV-pK205R7PM attenuated the inhibitory effect on IFN-I signaling. In conclusion, we identified a new ASFV immunosuppressive protein that increases our understanding of ASFV immune escape mechanisms.

Abstract The essential role of the inwardly rectifying potassium channel Kir5.1 (KCNJ16) in controlling electrolyte homeostasis and blood pressure has been demonstrated in human and animal studies. Previous studies have identified several bi‐allelic mutations of KCNJ16 in humans, causing severe hypokalemia, renal salt wasting, and disturbed acid–base homeostasis. Here, we identified a novel homozygous variant of KCNJ16, I26T, in an Amish patient affected with polydipsia, developmental delay, and chronic metabolic acidosis with low serum bicarbonate concentration. Subsequently, we generated the rat model with I26T mutation using Dahl salt‐sensitive rat (I26T rat) to characterize this variant. The male mutant rats displayed similar blood pressure and electrolyte homeostasis under baseline and with a high salt (4% NaCl) challenge. Blood pH, HCO3− and renal damage also remained similar between WT and I26T rats after high salt challenge. Additionally, single‐channel patch clamp analysis revealed similar channel activity in CHO cells overexpressed with WT and I26T mutant Kir4.1/5.1 channels. In summary, this study reported a novel variant in KCNJ16, namely I26T, which is likely a benign variant and not associated with pathologic phenotype in either human or Dahl salt‐sensitive rats, indicating that the type/location of variant should be considered when diagnosing and treating patients with KCNJ16 mutations.

As the world’s largest carbon emitter and the second-largest economy, China’s soaring carbon emissions are intricately linked to its historical reliance on GDP-based performance appraisal for local officials. In response, the Chinese government initiated reforms in 2014 to phase out the GDP-centric evaluation system (POGE) in underdeveloped regions, aiming to reduce carbon emissions and foster sustainable development. Notably, China met its Nationally Appropriate Mitigation Actions target ahead of schedule in 2020. However, research on the impact of the POGE policy on carbon emissions reduction remains scarce. This study addresses this critical gap by using county-level panel data from 2011 to 2020 and applying staggered Difference-in-Differences (DID) and Propensity Score Matching-Difference in Differences (PSM-DID) models to assess the POGE policy’s effects on per capita carbon emissions. Our findings reveal that the POGE policy has significantly lowered per capita carbon emissions by 13.1% to 14.1% in underdeveloped regions, with a marked regional gradient from east to west. The policy has also facilitated a shift in local industrial structure, favoring the primary industry over the secondary, further contributing to emission reduction. Overall, the POGE policy has not only improved the ecological environment but also supported local economic growth in these regions. These insights offer valuable policy implications for policymakers in China and other developing nations in crafting effective emission reduction strategies to achieve UN Sustainable Development Goal 13 (Climate Action) by 2030.

Abstract Purpose This study aimed to investigate the feasibility of using computed tomography (CT) attenuation values to differentiate hypodense brain lesions, specifically acute ischemic stroke (AIS) from asymmetric leukoaraiosis (LA) and old cerebral infarction (OCI). Materials and methods This retrospective study included patients with indeterminate hypodense lesions identified via brain CT scans conducted between June 2019 and June 2021. All lesions were confirmed through head MRI/diffusion-weighted imaging within 48 h after CT. CT attenuation values of hypodense lesions and symmetrical control regions were measured. Additionally, CT attenuation value difference (ΔHU) and ratio (RatioHU) were calculated. One-way analysis of variance (ANOVA) was used to compare age and CT parameters (CT attenuation values, ΔHU and RatioHU) across the groups. Finally, receiver operating characteristic (ROC) analysis was performed to determine the cutoff values for distinguishing hypodense lesions. Results A total of 167 lesions from 146 patients were examined. The CT attenuation values for AIS(n = 39), LA(n = 53), and OCI(n = 75) were 18.90 ± 6.40 HU, 17.53 ± 4.67 HU, and 11.90 ± 5.92 HU, respectively. Abstract Lycibarbarspermidines are unusual phenolamide glycosides characterized by a dicaffeoylspermidine core with multiple glycosyl substitutions, and serve as a major class of bioactive ingredients in the wolfberry. So far, little is known about the enzymatic basis of the glycosylation of phenolamides including dicaffeoylspermidine. Here, we identify five lycibarbarspermidine glycosyltransferases, LbUGT1-5, which are the first phenolamide-type glycosyltransferases and catalyze regioselective glycosylation of dicaffeoylspermidines to form structurally diverse lycibarbarspermidines in wolfberry. Notably, LbUGT3 acts as a distinctive enzyme that catalyzes a tandem sugar transfer to the ortho-dihydroxy group on the caffeoyl moiety to form the unusual ortho-diglucosylated product, while LbUGT1 accurately discriminates caffeoyl and dihydrocaffeoyl groups to catalyze a site-selective sugar transfer. Crystal structure analysis of the complexes of LbUGT1 and LbUGT3 with UDP, combined with molecular dynamics simulations, revealed the structural basis of the difference in glycosylation selectivity between LbUGT1 and LbUGT3. Site-directed mutagenesis illuminates a conserved tyrosine residue (Y389 in LbUGT1 and Y390 in LbUGT3) in PSPG box that plays a crucial role in regulating the regioselectivity of LbUGT1 and LbUGT3. Our study thus sheds light on the enzymatic underpinnings of the chemical diversity of lycibarbarspermidines in wolfberry, and expands the repertoire of glycosyltransferases in nature.

Abstract Peanut (Arachis hypogaea L.) is an important oilseed crop worldwide. However, soil salinization becomes one of the main limiting factors of peanut production. Therefore, developing salt-tolerant varieties and understanding the molecular mechanisms of salt tolerance is important to protect peanut yield in saline areas. In this study, we selected four peanut varieties with contrasting response to salt challenges with T1 and T2 being tolerance and S1 and S2 being susceptible. High-throughput RNA sequencing resulted in more than 314.63 Gb of clean data from 48 samples. We identified 12,057 new genes, 7,971of which have functional annotations. KEGG pathway enrichment analysis of uniquely expressed genes in salt-tolerant peanut revealed that upregulated genes in the root are involved in the MAPK signaling pathway, fatty acid degradation, glycolysis/gluconeogenesis, and upregulated genes in the shoot were involved in plant hormone signal transduction and the MAPK signaling pathway. Na+ content, K+ content, K+/ Na+, and dry mass were measured in root and shoot tissues, and two gene co-expression networks were constructed based on weighted gene co-expression network analysis (WGCNA) in root and shoot. In this study, four key modules that are highly related to peanut salt tolerance in root and shoot were identified, plant hormone signal transduction, phenylpropanoid biosynthesis, starch and sucrose metabolism, flavonoid biosynthesis, carbon metabolism were identified as the key biological processes and metabolic pathways for improving peanut salt tolerance. The hub genes include genes encoding ion transport (such as HAK8, CNGCs, NHX, NCL1) protein, aquaporin protein, CIPK11 (CBL-interacting serine/threonine-protein kinase 11), LEA5 (late embryogenesis abundant protein), POD3 (peroxidase 3), transcription factor, and MAPKKK3. There were some new salt-tolerant genes identified in peanut, including cytochrome P450, vinorine synthase, sugar transport protein 13, NPF 4.5, IAA14, zinc finger CCCH domain-containing protein 62, beta-amylase, fatty acyl-CoA reductase 3, MLO-like protein 6, G-type lectin S-receptor-like serine/threonine-protein kinase, and kinesin-like protein KIN-7B. The identification of key modules, biological pathways, and hub genes in this study enhances our understanding of the molecular mechanisms underlying salt tolerance in peanuts. This knowledge lays a theoretical foundation for improving and innovating salt-tolerant peanut germplasm.

Abstract Catalytic cascade transformations of simple starting materials into highly functionalized molecules bearing a stereochemically defined multisubstituted alkene, which are important in medicinal chemistry, natural product synthesis, and material science, are in high demand for organic synthesis. The development of multiple reaction pathways accurately controlled by catalysts derived from different ligands is a critical goal in the field of catalysis. Here we report a cobalt-catalyzed strategy for the direct coupling of inexpensive 1,3-enynes with two molecules of acrylates to construct a high diversity of functionalized 1,3-dienes containing a trisubstituted or tetrasubstituted olefin. Such cascade reactions can proceed through three different pathways initiated by oxidative cyclization to achieve multiple bond formation in high chemo-, regio- and stereoselectivity precisely controlled by ligands, providing a platform for the development of tandem carbon-carbon bond-forming reactions.

Abstract Background Exosomes assume a pivotal role as essential mediators of intercellular communication within tumor microenvironments. Within this context, long noncoding RNAs (LncRNAs) have been observed to be preferentially sorted into exosomes, thus exerting regulatory control over the initiation and progression of cancer through diverse mechanisms. Results Exosomes were successfully isolated from cholangiocarcinoma (CCA) CTCs organoid and healthy human serum. Notably, the LncRNA titin-antisense RNA1 (TTN-AS1) exhibited a conspicuous up-regulation within CCA CTCs organoid derived exosomes. Furthermore, a significant elevation of TTN-AS1 expression was observed in tumor tissues, as well as in blood and serum exosomes from patients afflicted with CCA. Importantly, this hightened TTN-AS1 expression in serum exosomes of CCA patients manifested a strong correlation with both lymph node metastasis and TNM staging. Remarkably, both CCA CTCs organoid-derived exosomes and CCA cells-derived exosomes featuring pronounced TTN-AS1 expression demonstrated the capability to the proliferation and migratory potential of CCA cells. Validation of these outcomes was conducted in vivo experiments. Conclusions In conclusion, our study elucidating that CCA CTCs-derived exosomes possess the capacity to bolster the metastasis tendencies of CCA cells by transporting TTN-AS1. These observations underscore the potential of TTN-AS1 within CTCs-derived exosomes to serve as a promising biomarker for the diagnosis and therapeutic management of CCA.

Abstract Strain-specific probiotics can present antioxidant activity and reduce damage caused by oxidation. Streptococcus alactolyticus strain FGM (S. alactolyticus strain FGM) isolated from the chicken cecum shows potential probiotic properties which have been previously demonstrated. However, the antioxidant properties of S. alactolyticus strain FGM remain unknown. In this view, cell-free supernatant (CFS), intact cells (IC) and intracellular extracts (CFE) of strain FGM and 3 strains of Lactobacillus (LAB) were prepared, and their scavenging capacities against DPPH, hydroxyl radicals and linoleic acid peroxidation inhibitory were compared in this study. The effects of strain FGM cell-free supernatant (FCFS) on NO production, activity of SOD and GSH-Px in RAW264.7 cells and LPS-induced RAW264.7 cells were analyzed. The metabolites in the supernatant were quantitated by N300 Quantitative Metabolome. Abstract Background This study aimed to investigate the impact of intraoperative hypothermia on the recovery period of anesthesia in elderly patients undergoing abdominal surgery. Methods A prospective observational study was conducted based on inclusion and exclusion criteria. A total of 384 elderly patients undergoing abdominal surgery under general anesthesia were enrolled in a grade A tertiary hospital in Chengdu, Sichuan Province from October 2021 and October 2022. After anesthesia induction, inflatable warming blankets were routinely used for active heat preservation, and nasopharyngeal temperature was monitored to observe the occurrence of intraoperative hypothermia. Patients were divided into hypothermia group and nonhypothermia group according to whether hypothermia occurred during the operation. Anesthesia recovery time and the incidence of adverse events or unwanted events during anesthesia recovery between the two groups were compared. Results The numbers (percentage) of 384 patients who underwent abdominal surgery developed intraoperative hypothermia occurred in 240 (62.5%) patients, all of whom had mild hypothermia. There were statistically significant differences between mild hypothermia after active warming and nonhypothermia in the occurrence of shivering (χ2 = 5.197, P = 0.023) and anesthesia recovery time (Z = -2.269, P = 0.02) in elderly patients undergoing abdominal surgery during anesthesia recovery, and there were no statistically significant differences in hypoxemia, nausea or vomiting, hypertension, hypokalemia, hypocalcemia, analgesic drug use,postoperative wound infection or postoperative hospitalization days. Conclusions The incidence of intraoperative mild hypothermia after active warming was high in elderly patients who underwent abdominal surgery. Mild hypothermia increased the incidence of shivering and prolonged anesthesia recovery time in elderly patients undergoing abdominal surgery.

Abstract Background Breast cancer is the most prevalent cancer among women globally, and surgical procedures continue to be the primary treatment. However, over 50% of patients experience preoperative anxiety due to the unknown and fear associated with surgery. Although drug therapy is commonly used to address this anxiety, its side effects have led to a heated debate regarding its effectiveness. Consequently, non-pharmacological therapies, such as preoperative education, have emerged as an alternative approach to alleviate anxiety. WeChat, a widely popular social media platform, offers a public platform that can potentially be utilized for effective preoperative education. This study aims to evaluate the use of WeChat public platform as a tool for preoperative education in patients undergoing breast surgery. Methods This is a prospective, randomized, and controlled trial will involve 392 adult women scheduled for breast cancer resection. Participants will be randomly assigned to either the WeChat education group or the regular group. In addition to regular preoperative visits, the WeChat education group will also watch science videos through the WeChat public platform. The regular group will only receive education from ward nurses during preoperative visits. The primary outcome measure will be the incidence of preoperative anxiety, defined by scores of the State Anxiety Inventory (SAI) exceeding 40 points. Secondary outcome measures include the incidence of severe anxiety (SAI > 44) on the day before surgery, incidence of anxiety 72 h after surgery, incidence of severe anxiety 72 h after surgery, NRS scores for pain at rest and during activity 24, 48, and 72 h after surgery, incidence of nausea and vomiting within 24 h after surgery, subjective sleep score at 1 week postoperatively, quality of life QoR-15 scores at 1 and 3 months postoperatively, incidence of chronic pain at 3 months postoperatively, bowel function recovery, length of hospital stay, and hospitalization expenses. Discussion This is the first clinical trial to investigate the use of WeChat public platform for delivering preoperative education on perioperative anxiety in breast cancer patients. By utilizing the renowned WeChat public platform, our study aims to improve patient outcomes by providing video education that explains the disease, surgery, and anesthesia in a more accessible manner, thereby reducing the incidence of perioperative anxiety. If our hypothesis is confirmed, this non-pharmacological approach can be universally acknowledged as a cost-effective and practical method in clinical care. Its application can also be extended to other medical fields beyond breast cancer. Trial registration ClinicalTrials.gov, NCT05291494. Registered on 29 December 2021.

AIM: To evaluate the effectiveness and safety of intravitreal ranibizumab (IVR) for diabetic macular edema (DME) in vitrectomized versus non-vitrectomized eyes. METHODS: The PubMed, EMBASE, Web of Science, Cochrane, EBSCO were comprehensively searched for studies comparing vitrectomized and non-vitrectomized eyes with DME. Clinical outcomes of best-corrected visual acuity (BCVA), central macular thickness (CMT), the mean number of intravitreal injection and adverse events were extracted and analyzed. Lower extremity peripheral artery disease (PAD) is a growing global health problem. New methods to diagnose PAD have been explored in recent years. At present, the majority of imaging methods for PAD focus on the macrovascular blood flow, and the exploration of microcirculation and tissue perfusion of PAD remains largely insufficient. In this report, we applied three new imaging technologies, i.e., second near-infrared region (NIR-II, 900–1,880 nm wavelengths) imaging, optical coherence tomography angiography (OCTA), and laser speckle flowgraphy (LSFG), in a PAD patient with a healthy human subject as control. Our results showed that the PAD patient had poorer tissue perfusion than the control without observed adverse effects. Moreover, compared with the first near-infrared region (NIR-I, 700–900 nm wavelengths) imaging results, NIR-II imaging had a higher signal-to-background ratio and resolution than NIR-I imaging and detected microvessels that were not detected by NIR-I imaging. These observations suggested that NIR-II imaging, OCTA, and LSFG are potentially safe and effective methods for diagnosing PAD.

BackgroundHulatang is a traditional specialty snack in Henan, China, and is well known for its unique flavor.MethodsIn this study, the volatile organic compounds (VOCs) in four kinds of Hulatang from two representative regions in Henan Province (Xiaoyaozhen and Beiwudu) were evaluated using headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS).ResultsThe results showed that Xiaoyaozhen Hulatang exhibited more ethers, fewer terpenes and ketones than Beiwudu Hulatang. Additionally, Hulatang from different regions were classified using the orthogonal partial least squares-discriminant analysis (OPLS-DA) based on GC-IMS data. Twenty aroma substances were selected as the potential markers using the variable importance in the projection (VIP) variable selection method. Additionally, fifteen aroma components significantly contributing to the aroma of Hulatang were screened using the relative odor activity value (ROAV) (ROAV > 1). Combined with the sensory score results, twelve key substances with significant correlation with odor perception were selected. The flavor characteristics of the key substances revealed that the flavor of Hulatang was mainly composed of volatile components with camphor, green, almond, fatty, spicy, herbal, vegetable, fruity, floral, musty, and solvent aromas.ConclusionOverall, the experimental results provide a theoretical basis for evaluating the flavor characteristics of Hulatang from different regions using GC-IMS.

Abstract Facile evaluation of formation kinetics of key intermediate is crucial for a comprehensive understanding of electrochemical ammonia oxidation reaction (AOR) mechanisms and the design of efficient electrocatalysts. Currently, elucidating the formation kinetics of key intermediate associated with rate‐determining step is still challenging. Herein, 4‐phtalamide‐N‐(4′‐methylcoumarin) naphthalimide (CF) is developed as a molecular probe to detect N2H4 intermediate during AOR via electrochemiluminescence (ECL) and further investigated the formation kinetics of N2H4 on Pt catalysts with different crystal planes. CF probe can selectively react with N2H4 to release ECL substance luminol. Thus, N2H4 intermediate as a key intermediate can be sensitively and selectively detected by ECL during AOR. For the first time, Pt(100) facet is discovered to exhibit faster N2H4 formation kinetics than Pt(111) facet, which is further confirmed by Density functional theory calculation and the finite element simulation. The AOR mechanism under the framework of Gerischer and Mauerer is further validated by examining N2H4 formation kinetics during the dimerization process (NH2 coupling). The developed ECL active probe and the discovered facet‐dependent formation kinetics of key intermediates provide a promising new tool and strategy for the understanding of electrochemical AOR mechanisms and the design of efficient electrocatalysts.

Attention deficit hyperactivity disorder (ADHD) is one of the most common neurodevelopmental disorders in childhood. Numerous resting-state functional magnetic resonance imaging (rs-fMRI) studies in ADHD have been performed using traditional low-frequency bands (0.01–0.08 Hz). However, the neural activity patterns of frequency subbands in ADHD still require further investigation. The purpose of this study is to explore the frequency-dependent characteristics and neural activity patterns of ADHD subtypes. We selected the ADHD combined type (ADHD-C, N = 25), ADHD inattentive type (ADHD-I, N = 26) and typically developing (TD, N = 28) children from the ADHD-200 Consortium. Based on the slow-5 band (0.01–0.027 Hz) and slow-4 band (0.027–0.073 Hz), we generated static and dynamic fractional amplitude of low-frequency fluctuation (fALFF) and regional homogeneity (ReHo) maps for each participant. A flexible-factorial analysis of variance model was performed on static and temporal dynamic rs-fMRI measurements within two subbands. Results revealed that the orbital-frontal gyrus, precuneus, superior temporal gyrus and angular gyrus were found to have obvious frequency band and group interaction effects. The intrinsic neural activity differences among three groups were more prominent in the slow-5 frequency band compared to the slow-4 band. In addition, the indices of significant interaction regions showed correlations with the progression of the disease and the features in slow-5 showed an advantageous diagnostic performance compared with those in slow-4. The results suggested the intrinsic neural activities of ADHD subtypes were frequency-dependent. The frequency-specific analysis of static and dynamic brain activity may provide a deeper understanding of neurophysiological dysfunction patterns in ADHD subtypes and provide supplementary information for assessing ADHD subtypes.

Humic electron mediators can facilitate the reductive dehalogenation of organohalogenated compounds by accelerating electron transfer. To investigate the effect of humic electron mediators on the microbial anaerobic reductive dechlorination of Polychlorinated biphenyls (PCBs), three types of humic electron mediators, humin (HM), humic acid (HA), and anthraquinone-2,6-disulfonic acid (AQDS, HA analogs), were added to PCB dechlorination cultures enriched from different sources in terrestrial and marine environments (T and M cultures). The results showed that meta- and para-site dechlorination occurred in the M culture, while only meta-site dechlorination occurred in the T culture. The dechlorination process N and the dechlorination process H or H′ are presented in both cultures. HM enhanced PCB dechlorination metabolic activity in both cultures mainly by promoting meta-site dechlorination. HA showed a weak promoting effect on the M culture by promoting para-chlorine removal but inhibited the dechlorination metabolism of the terrestrial-origin culture, inhibiting meta-chlorine removal. AQDS showed inhibitory effects on both cultures by inhibiting the microbial removal of meta-chlorine. High-throughput sequencing and qPCR results suggest that HM is not a carbon source for the potential dechlorinating metabolism of Dehalococcoides but may promote reductive dechlorination by changing the community structure, and AQDS may inhibit anaerobic reductive dechlorination of PCBs by inhibiting the growth of Dehalococcoides. This study provides insights into the mechanism of enhancing PCB microbial dechlorination mediated by humic substances and plays a significant role in extending the application prospects of PCBs bioremediation technology.

BackgroundAtherosclerosis (AS) is a chronic arterial pathology and a leading cause of vascular disease-related mortality. Fatty streaks in the arterial wall develop into atherosclerosis and characteristic plaques. Clinical interventions typically involve lipid-lowering medications and drugs for stabilizing vulnerable plaques, but no direct therapeutic agent specifically targets atherosclerosis. Garlic, also locally known as DASUAN, is recognized as a widely sold herbal dietary supplement esteemed for its cardiovascular benefits. However, the specific mechanisms of garlic’s anti-atherosclerotic effects remain unclear.AimsThis study aims to elucidate the pharmacological mechanisms through which garlic ameliorates atherosclerosis.MethodsThe study identified the major active components and targets of garlic by screening the TCMSP, TCM-ID, and, ETCM databases. Atherosclerosis-associated targets were obtained from the DisGeNET, GeneCards, and DiGSeE databases, and garlic intervention targets were determined through intersection. Utilizing the intersected genes, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were conducted using R software. A garlic component-disease target network was constructed using Cytoscape. RNA-seq datasets from the GEO database were utilized to identify differentially expressed genes (DEGs) associated with atherosclerosis. The target genes were intersected with DEGs and the FerrDb (ferroptosis database). Molecular docking predicted the binding interactions between active components and the core targets. In vitro and in vivo experiments validated the identified core targets.ResultsThe integration of garlic drug targets with atherosclerotic disease targets identified 230 target genes. Intersection with RNA-seq DEGs revealed 15 upregulated genes, including 8 target genes related to ferroptosis. Molecular docking indicated favorable affinities between garlic active components [Sobrol A, (+)-L-Alliin, Benzaldoxime, Allicin] and target genes (DPP4, ALOX5, GPX4). Experimental validation showed that GARLIC reduces the expression of ferroptosis-related genes in AS, suggesting its therapeutic potential through the regulation of ferroptosis.ConclusionGarlic ameliorates atherosclerosis by targeting intra-plaque ferroptosis and reducing lipid peroxidation. These findings provide novel insights into the pharmacological mechanisms underlying the efficacy of garlic in treating AS.

BackgroundBile acids (BAs), products of gut microbiota metabolism, have long been implicated in atherosclerotic disease pathogenesis. Characterizing the serum bile acid profile and exploring its potential role in carotid atherosclerosis (CAS) development are crucial tasks.MethodsIn this study, we recruited 73 patients with CAS as the disease group and 77 healthy individuals as the control group. We systematically measured the serum concentrations of 15 bile acids using ultrahigh-performance liquid chromatography-mass spectrometry (UPLC-MS/MS). Multivariate logistic regression and least absolute shrinkage and selection operator (LASSO) regression were applied to analyze the impact of bile acids on the disease and select the key BAs. The possible molecular mechanism was elucidated by network pharmacology.Results(1) The BA profile of patients with CAS significantly differed. (2) Multifactorial logistic regression analysis identified elevated levels of GCDCA (OR: 1.01, P < 0.001), DCA (OR: 1.01, P = 0.005), and TDCA (OR: 1.05, P = 0.002) as independent risk factors for CAS development. Conversely, GCA (OR: 0.99, P = 0.020), LCA (OR: 0.83, P = 0.002), and GUDCA (OR: 0.99, P = 0.003) were associated with protective effects against the disease. GCA, DCA, LCA, and TDCA were identified as the four key BAs. (3) TNF, FXR, GPBAR1, ESR1 and ACE were predicted to be targets of BAs against AS. These four BAs potentially impact AS progression by triggering signaling pathways, including cAMP, PPAR, and PI3K-AKT pathways, via their targets.ConclusionThis study offers valuable insights into potential therapeutic strategies for atherosclerosis that target bile acids.

High sugar content in peanut seeds is one of the major breeding objectives for peanut flavor improvement. In order to explore the genetic control of sugar accumulation in peanut kernels, we constructed a recombinant inbred line population of 256 F2:6-7 lines derived from the Luhua11 × 06B16 cross. A high-resolution genetic map was constructed with 3692 bin markers through whole genome re-sequencing. The total map distance was 981.65 cM and the average bin marker distance was 0.27cM. A major stable QTL region (qSCB09/qSSCB09) was identified on linkage group (LG) B09 associated with both sucrose content (SC) and soluble sugar content (SSC) explaining 21.51-33.58% phenotypic variations. This major QTL region was consistently detected in three environments and mapped within a physical interval of 1.56 Mb on chromosome B09, and six candidate genes were identified. These results provide valuable information for further map-based cloning of favorable allele for sugar content in peanut.

Aiming at the frost heave characteristics of long-distance and large-section underground freezing projects in cities, the double-line tunnel of Shanghai Metro Line 18 undercrossing an operation station was select as an example,a three-dimensional numerical model was established by using finite element software based on the thermal-mechanical coupling equation. Combined with the physical parameters obtained from laboratory tests, the evolution law of frost heave displacement field of the project was studied, and the influence of measures such as the staggered peak freezing and the adjustment of brine temperature on the frost heave displacement field was explored. Under the experimental conditions, the results show that: ① the deformation of the station floor caused by frost heave which mainly occurs in the active freezing period. When the double-line tunnel is frozen at the same time, the vertical displacement of the station floor at 45days of freezing reaches 77.72% of the displacement at 90 days of freezing. ② When the freezing time reaches 90 days, compared with simultaneous freezing, the displacement of the station floor reduced by 7.7% under the condtion of staggered peak freezing. The superposition effect of frost heave in the same time period is effectively avoided, and the frost heave effect is reduced to a certain extent. ③ When the freezing time reaches 90 days, compared with simultaneous freezing, the displacement of the station floor reduced by 34.2% under the condtion of adjusting the brine temperature, it shown that the expansion rate and frost heave effect of frozen soil are effectively controlled. In engineering practice, measures such as staggered freezing and adjusting brine temperature are used together to control frost heave, and the maximum vertical deformation of the station floor is 25.41 mm. The deformation law of the station floor obtained by numerical simulation is basically consistent with the measured data, which effectively guided the construction of the project.

Abstract Background Endometritis is a common bovine postpartum disease. Rapid endometrial repair is beneficial for forming natural defense barriers and lets cows enter the next breeding cycle as soon as possible. Selenium (Se) is an essential trace element closely related to growth and development in animals. This study aims to observe the effect of Se on the proliferation of bovine endometrial epithelial cells (BEECs) induced by lipopolysaccharide (LPS) and to elucidate the possible underlying mechanism. Results In this study, we developed a BEECs damage model using LPS. Flow cytometry, cell scratch test and EdU proliferation assay were used to evaluate the cell cycle, migration and proliferation. The mRNA transcriptions of growth factors were detected by quantitative reverse transcription-polymerase chain reaction. The activation of the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (AKT) and Wnt/β-catenin pathways were detected by Western blotting and immunofluorescence. The results showed that the cell viability and BCL-2/BAX protein ratio were significantly decreased, and the cell apoptosis rate was significantly increased in the LPS group. Compared with the LPS group, Se promoted cell cycle progression, increased cell migration and proliferation, and significantly increased the gene expressions of TGFB1, TGFB3 and VEGFA. Se decreased the BCL-2/BAX protein ratio, promoted β-catenin translocation from the cytoplasm to the nucleus and activated the Wnt/β-catenin and PI3K/AKT signaling pathways inhibited by LPS. Conclusions In conclusion, Se can attenuate LPS-induced damage to BEECs and promote cell proliferation and migration in vitro by enhancing growth factors gene expression and activating the PI3K/AKT and Wnt/β-catenin signaling pathways.

Abstract Background The neural cells in the brains of patients with Parkinson’s disease (PWP) display aberrant synchronized oscillatory activity within the beta frequency range. Additionally, enhanced gamma oscillations may serve as a compensatory mechanism for motor inhibition mediated by beta activity and also reinstate plasticity in the primary motor cortex affected by Parkinson’s disease. Transcranial alternating current stimulation (tACS) can synchronize endogenous oscillations with exogenous rhythms, thereby modulating cortical activity. The objective of this study is to investigate whether the addition of tACS to multidisciplinary intensive rehabilitation treatment (MIRT) can improve symptoms of PWP so as to enhance the quality of life in individuals with Parkinson’s disease based on the central-peripheral-central theory. Methods The present study was a randomized, double-blind trial that enrolled 60 individuals with Parkinson’s disease aged between 45 and 70 years, who had Hoehn-Yahr scale scores ranging from 1 to 3. Participants were randomly assigned in a 1:1 ratio to either the tACS + MIRT group or the sham-tACS + MIRT group. The trial consisted of a two-week double-blind treatment period followed by a 24-week follow-up period, resulting in a total duration of twenty-six weeks. The primary outcome measured the change in PDQ-39 scores from baseline (T0) to 4 weeks (T2), 12 weeks (T3), and 24 weeks (T4) after completion of the intervention. The secondary outcome assessed changes in MDS-UPDRS III scores at T0, the end of intervention (T1), T2, T3, and T4. Additional clinical assessments and mechanistic studies were conducted as tertiary outcomes. Discussion The objective of this study is to demonstrate that tACS can enhance overall functionality and improve quality of life in PWP, based on the framework of MIRT. Additionally, it seeks to establish a potential correlation between these therapeutic effects and neuroplasticity alterations in relevant brain regions. The efficacy of tACS will be assessed during the follow-up period in order to optimize neuroplasticity and enhance its potential impact on rehabilitation efficiency for PWP.
