Aspergillus niger strain LCP521 harbours a highly processive epoxide hydrolase (EH) that is of particular interest for the enantioselective bio-organic synthesis of fine chemicals. In the present work, we report the isolation of the gene and cDNA for this EH by use of inverse PCR. The gene is composed of nine exons, the first of which is apparently non-coding. The deduced protein of the A. niger EH shares significant sequence similarity with the mammalian microsomal EHs (mEH). In contrast to these, however, the protein from A. niger lacks the common N-terminal membrane anchor, in line with the fact that this enzyme is, indeed, soluble in its native environment. Recombinant expression of the isolated cDNA in Escherichia coli yielded a fully active EH with similar characteristics to the fungal enzyme. Sequence comparison with mammalian EHs suggested that Asp192, Asp348 and His374 constituted the catalytic triad of the fungal EH. This was subsequently substantiated by the analysis of respective mutants constructed by site-directed mutagenesis. The presence of an aspartic acid residue in the charge-relay system of the A. niger enzyme, in contrast to a glutamic acid residue in the respective position of all mEHs analysed to date, may be one important contributor to the exceptionally high turnover number of the fungal enzyme when compared with its mammalian relatives. Recombinant expression of the enzyme in E. coli offers a versatile tool for the bio-organic chemist for the chiral synthesis of a variety of fine chemicals.

KWIN-Veehouderij is een jaarlijkse uitgave van het Praktijkonderzoek Veehouderij (PV). De KWIN bevat statistische prijsinformatie over onderdelen van veehouderijbedrijven en normen voor het maken van berekeningen, bedrijfsevaluaties en begrotingen. KWIN-Veehouderij heeft betrekking op de sectoren melkvee, vleesvee, geiten, schapen, paarden, varkens, pluimvee, nertsen, konijnen en palingen. : :Bij de normen staat een toelichting over de inhoud, de wijze van berekening en de bron van de gegevens. Generaliseren is bij het opstellen van normen onvermijdelijk. De normen moet u dan ook zien als een richtlijn, die echter wel een breed draagvlak hebben omdat ze met afgevaardigden uit de sectoren zijn afgesproken. Afhankelijk van de ideeën van de ondernemer en het bedrijf waarvoor u begroot, kunt u de gegeven normen aanpassen. : :Saldoberekeningen : :Voor alle sectoren is er een saldoberekening. Voor het maken van een saldoberekening voor de melkveehouderij is bij het PV het computerprogramma BBPR beschikbaar. Deze berekent, afhankelijk van de specifieke bedrijfssituatie, saldo- of netto-bedrijfsresultaten. BBPR werkt met de normen uit KWIN-Veehouderij. Alle in KWIN-Veehouderij gegeven saldoberekeningen zijn voorbeeldberekeningen. Voor specifieke bedrijfsbegrotingen moet u ze aanpassen aan de bedrijfssituatie. : :Wat kunt u ermee? : :KWIN-Veehouderij leent zich voor verschillende doeleinden. De normen geven inzicht in de hoogte van de prijzen op middellange termijn. Hierdoor zijn ze geschikt als basis voor planning- en evaluatiesystemen. Het voordeel van KWIN-Veehouderij is dat u alle informatie van de betreffende diersoorten bij elkaar hebt.

Het rapport geeft een goed beeld van de bedrijfsvoering, de bedrijfsresultaten en de resultaten van het onderzoek. Belangrijke doelstellingen zijn gehaald of heel dicht benaderd.

In de praktijk blijkt de veevervanging op het Lagekostenbedrijf de nodige knelpunten op te leveren.

Ook in 2000 was het Lagekostenbedrijf weer zelfvoorzienend voor ruwvoer bij een laag bemestingsniveau op het grasland.

Het doel om zoveel en zo lang mogelijk te beweiden, heeft het Lagekostenbedrijf in 1999 ruimschoots kunnen waarmaken. Witte klaver heeft een belangrijke bijdrage geleverd aan de voerproductie.

Met BBPR kunnen veehouders hun bedrijf analyseren en alternatieve bedrijfssituaties begroten. Doordat BBPR nu ook standaardoverzichten kan maken sluit het programma veel beter aan bij managementsoftware.

Wild water buffaloes (Bubalus arnee) are categorized as endangered on the IUCN Red List. With a global population of less than 4,000, the species has a very limited distribution spanning over less than 20,000 km2 in Nepal, India, Sri Lanka, Cambodia, Myanmar, Thailand and Bhutan. In Nepal, Koshi Tappu Wildlife Reserve was designated for wild Asian buffalo; this reserve contains approximately 219 individuals, although there is no precise information on the number of pure-bred wild buffalo. The conservation of wild buffalo is of global concern; there are numerous threats due to habitat fragmentation, as well as competition, hybridization, habitat overlap, disease and parasites transmitted by domestic livestock. The reserve provides an area of only 0.80 km2 per buffalo, which is insufficient maintaining a viable population of wild buffalo. It is therefore important to identify further potential habitat and to begin translocation of some of the individuals. A Wild Buffalo Conservation Alliance can play a key role in influencing conservation strategies by conducting detailed research including feasibility studies on translocation, developing awareness, holding workshops and preparing a conservation action plan. In this way the alliance can help to increase the number of wild buffaloes and to maintain a viable population of the species.
