ABSTRACT Chicken abdominal fat (AF) is an economically important trait, and many studies have been conducted on genetic selection for AF. However, previous studies have focused on detecting functional chromosome mutations or regions using gene chips. The present study used the specific-locus amplified fragment sequencing (SLAF-seq) technology to perform a genome-wide association study (GWAS) on purebred Wengshang Barred chicken. A total of 1,286,715 single-nucleotide polymorphisms (SNPs) were detected, and 175,211 SNPs were selected as candidate SNPs for genome-wide association analysis using TASSEL general linear models. Two SNPs markers reached genome-wide significance. Of these, rs7943847, rs127627362 were significantly associated with AF at 120 days. These SNPs are close to eight genes (SLC16A6, ARSG, WIPI1, PRKAR1A, FAM20A, ABCA8, ABCA9, CPQ,). These results would enrich the studies on AF and promote the use of Chinese chicken, especially the Wenshang Barred chicken.

The degree of steam overlay and descriptions of the migration rules for vapor front is considered one of the greatest challenges to research the seepage laws of steam assisted gravity drainage. In order to surmount these challenges, the theory of percolation mechanics are introduced, combined with the well group structure of steam assisted gravity drainage, we do workings such as establishing the mathematical model of the pressure field, applying the steam overlap theory and simplifying the steam overlap coefficient, adopting the technology of database and mapping to display the trends of vapor front intuitively. At last, combined with production factor and reservoir thickness, we analyze the swept volume of steam and the influence rules of the vapor front migration. Understanding the achievements will provide a scientific basis to the improvement of steam’s sweep volume in symmetry elements, displaying the working degree of reservoirs intuitively as well as the efficient development of steam assisted gravity drainage.

Sea cucumbers have been emerged as important models to study organ regeneration and development owing to the capacity to regenerate its organs quickly after evisceration. Evisceration is a special defense mechanism for sea cucumber to eject all of internal organs when they encounter predators or adverse environmental conditions. However, little was known about the dynamics of bacterial community in coelomic fluid after evisceration. In the present study, evisceration was induced by intracelomic injection of 0.35 M KCl, and the significantly alternation of bacterial community in coelomic fluid of sea cucumber Apostichopus japonicus was observed with lower diversity and total bacterial load at 7 dpe (days post evisceration) and 14 dpe. The bacterial community was tended to restore at 28 dpe. In particular, relative abundances of Bacteroidetes and Rubritaleaceae, which involved in degradation of polysaccharides and lipid, increased significantly at 7 dpe (p < 0.05), and returned to the original level at 28 dpe. In addition, the predicted functions of bacterial community indicated that the bacteria associated with metabolism pathways of amino acid, lipid and carbohydrate also increased significantly at 7 dpe. These results suggested that the bacterial community in coelomic fluid of A. japonicus was highly dynamic and could rebuild a stable community structure after evisceration. It was suggested that the enriched metabolic related beneficial bacteria at early stage played a role after evisceration in terms of decomposing polysaccharides and lipid to provide energy.

ABSTRACT The effects of plant essential oil (PEO) on the production performance and immune function of laying hens were evaluated to provide theoretical basis for promoting the natural plant extracts. Eight thousand 1-day-old healthy laying hens were randomly divided into a control group or PEO group, with four replicates per treatment and 1000 hens per replicate. The PEO diet was supplemented with 3g/kg plant extract. Diets were fed for 56 days. The tibia length and keel length were detected on an empty stomach at the end of the trial. Blood samples were collected on the 28th and 56th days to detect the level of C3, C4, IL-1, IL-2, IL-17 and immunoglobulin in the serum. ABSTRACT To investigate the hypothesis that APS can attenuate E. coli-induced microvascular dysfunction in chicken intestine, 60 0-day old male chickens were divided into three groups with 5 replications of 4 chicks. Chicken in the APS group were treated with 15 mg APS daily while the others were given 0 mg APS for 6 days. Then all 7-day old chicken were injected intraperitoneally by E. coli, except for the chicken in the control group. After 4 h, all chicken’s intestinal samples were collected to detect gene expressions involved in inflammatory factors and adhesion molecules. Results showed that APS attenuated the signs of edema and hemorrhage in 7-day old chicken intestinal mucosa which were induced by E. coli injection. The Pacific oyster Crassostrea gigas is an important species living in the intertidal zones. It is of great significance to study the mechanism of oysters to adapt air exposure. In the present study, weighted gene co-expression network analysis (WGCNA) with the transcriptome data of gills and adductor muscle was conducted to investigate the metabolic transformation of C. gigas under air exposure. GO enrichment of modules specifically expressed in adductor muscle of oysters exposed to air for five, seven and nine days revealed the phased expression of respiratory chain, protein turnover and lipid metabolism, indicating the conversion of energy metabolism. During air exposure, “respiratory chain” and “ribosome biogenesis” were enriched in the muscle on the fifth day, suggesting that glycogen metabolism was dominant in the early stages of air exposure. On the seventh day, many terms about the regulation of proteolysis were enriched, indicating that carbohydrates were not be able to meet the metabolic needs in the oyster adductor muscle, and proteins began to be degraded for energy supply. The processes related to lipid metabolism were enriched on the ninth day. The extremely high glycogen content of C. gigas allowed it to maintain a basic metabolic activity for a long time with a conservative compensation strategy. GO and KEGG enrichments of the modules sensitive to air exposure in gills were mainly involved in “response to endoplasmic reticulum stress”, “Endoplasmic Reticulum (ER) to Golgi transport vesicle membrane” and “protein processing in endoplasmic reticulum”. It revealed that the mechanism of oyster adapting to air exposure was a complex regulatory network depending on the ER. Hub gene network and PPI network analyses found that some transcription factors containing zinc finger domains regulated the biochemical reactions for stress adaptation, indicating that the ER, as a regulatory element sensitive to external stress, could regulate apoptosis, autophagy and protein degradation in gills of C. gigas under air exposure. These results would provide new insights into the adaptation of C. gigas to air exposure in terms of energy metabolism and homeostasis.

A need for multi-functional assessment tools evaluating trade-offs and co-benefits for various types of Nature-Based Solutions (NBS) has been increasingly identified in recent years. Methodologically, concepts for a tool are presented which include quantifying the demand and potential for NBS to enhance ecosystem service (ES) provision, and linking ESs to readily quantifiable and legislatively-relevant environmental quality indicators (EQIs). The objective of tool application is to identify optimal NBS placement across a diverse set of socio-environmental indicators, whilst also incorporating issues of relative location of areas of implementation and benefit accrual. Embedded within the tool is the importance of evaluating outcomes in terms of economic benefits and of sustainable development goals. The concepts are illustrated with simplified examples, relating to the case of implementing urban forestry as an exemplar NBS. By summarising the knowledge base it is demonstrated that benefits of NBS are substantially scale-dependent in two main respects; those of extent and proximity to receptors. Evaluation tools should be capable of quantifying scale-dependence. The substantive importance of these considerations and how their dynamics vary between indicators and services is illustrated graphically through schematic functions. When developed, the tool should be used as a focus for consultation and co-design to pinpoint the size of NBS necessary to achieve a sufficient level of benefit for a particular receptor. This could be measured against target levels of benefit for each indicator, distinguishing between primary intended outcomes and those co-benefits or trade-offs that are secondary or unintended.

The Chinese horseshoe crab Tachypleus tridentatus is an endangered marine benthic species of great biomedical value and scientific significance. In this study, T. tridentatus hemocyte transcriptome was sequenced to rapidly develop molecular resources using the Illumina paired-end sequencing platform. Deep sequencing generated a total of 38402764 reads that consisted of 7.7 Gbp raw data, which were assembled into 38759 unigenes with an average length of 668.6 bp. A total of 608 microsatellite loci and 71959 high-confidence single nucleotide polymorphisms were discovered. All the assembled unigenes were annotated by running BLASTx similarity searches on Nr, Swiss-Prot, COG, and KEGG databases; of which 17446 (45.0%) unigenes showed significant matches (E-value

High-throughput sequencing (HTS) is rapidly becoming a popular and robust tool to characterize biodiversity of complex communities, especially for those dominated by microscopic species such as zooplankton. The popular use of HTS-based methods has prompted a possible method of inferring relative species abundance from sequencing data. However, these methods remain largely untested in many communities as to whether sequence data can reliably quantify relative species abundance. Here we tested the relationship between species abundance and sequence abundance in zooplankton using 2 methods: (1) spiking known amounts of indicator species into existing zooplankton communities, and (2) comparing results obtained from parallel replicates for the same natural zooplankton communities. Although we detected a general trend that low-abundance species usually corresponded to low-abundance sequence reads, further statistical analyses revealed that sequencing data could not reliably quantify relative species abundance, even for the same indicator species spiked into different zooplankton communities. The distribution of sequence reads statistically varied even between parallel replicates of the same natural zooplankton communities. Our study reveals that sequence abundance may generally qualitatively reflect species abundance as the general trend between these 2 variables exists; however, extra caution is required when using HTS-based approaches to make quantitative inferences regarding zooplankton communities.

Xanthomonas oryzae pv. Oryzae (Xoo) are plant pathogenic bacteria that can cause serious blight of rice. We have demonstrated that Xoo can infect the model organism C. elegans and p38 MAPK pathway plays specific roles in defense against the pathogen in our previous paper. Based on that p38 MAPK pathway can be activated in a range of tissues, it is intriguing to compare the tissue-specific activities of this pathway in host innate immunity. Here, transgenic worms that sek-1 expressed specifically in neurons system, ciliated sensory neurons, and intestine respectively are used to determine the nematode survival and transcriptional levels of immune-related genes. We report that SEK-1 and TOL-1 are not involved in C. elegans avoidance behavior, and ingestion of nematodes is related to the aversion and also the characteristics of bacteria. In addition, tol-1 and sek-1 participate the immune response to the infection by Xoo; sek-1 also exhibits tissue-specific activities in host innate immunity. Our findings suggest that overlapping immune effect may exist between the tol-1 and sek-1.

Heat shock proteins (HSPs) are molecular chaperones that help organisms cope with stressful conditions. In this study, a novel small HSP (sHSP) gene was identified from the hard clam Meretrix meretrix (designated as Mm-HSP20). The full length of the cDNA is 1222 bp, consisting of a 5’-terminal untranslated region (5’UTR) of 103 bp, a 3’UTR of 565 bp, and a 522 bp open reading frame encoding a polypeptide of 173 amino acids. Sequence comparison showed that Mm-HSP20 had a moderate degree of homology to the sHSP of other organisms. A sHSP feature domain, an alpha-crystallin domain, and a V/IXI/V motif in the C-terminal extension were identified in the Mm-HSP20 amino acid sequence, indicating that Mm-HSP20 is a new member of the sHSP family. The Mm-HSP20 transcript was constitutively expressed in 6 different test tissues, with the highest expression level detected in the digestive gland by fluorescent quantitative real-time PCR. The temporal expression of Mm-HSP20 mRNA in the digestive gland and hemocytes was evaluated after exposure to cadmium (Cd) (40 µg l-1 CdCl2) and benzo[a]pyrene (50 µg l-1 B[a]P) individually, and in combination (40 µg l-1 CdCl2 +5 0 µg l-1 B[a]P). Mm-HSP20 expression level increased significantly after Cd and B[a]P exposure in both tissue types. Conversely, significant Mm-HSP20 transcript repression was detected in the 24 h B[a]P-treated digestive gland samples. Considerable up-regulation of Mm-HSP20 mRNA level was observed after multiple exposures. These results indicate that Mm-HSP20 plays a role in mechanisms involved in coping with environmental stress in hard clams. Mm-HSP20 may be suitable for use as a biomarker for heavy metal and B[a]P contamination.

Background: To determine the prevalence and characteristics of birth defects in perinatal infants in Hubei Province during 200l-2008.Methods: The prevalence of birth defects in perinatal infants delivered after 28 weeks or more was analyzed in Hubei surveillance hospitals during 200l-2008.Results: The incidence of birth defects in perinatal infants from 200l to 2008 was 120.0 per 10,000 births, and was increased by about 41% from 81. 1 in 2001 to 138.5 per 10,000 births in 2008. The incidence in the first 4 years (2005-2008) was much higher than the latter four (2001-2004) (χ2=77.64 , P 0.05), but that between male and female was significant (χ2=5.24 , P 0.05). The two leading birth defects were cleft lip and/or palate and polydactyly, followed by congenital heart disease, hydrocephaly, external ear malformation and neural tube defects. The prevalence of congenital heart disease was rising.Conclusions: Eight years birth defects data indicate that the birth defect rate was on the rise and the birth defects prevalence in Hubei province should be valued.

Antimicrobial peptides (AMPs) represent the most universal immune effectors. Molluscs constitute the second largest animal phylum, after Arthropods, in term of number of species. Only a negligible number has been investigated regarding AMPs. The choice of the species to be studied relied on their economical importance and availability. First studies on molluscan AMPs dated from 1996 and were based on biological activities of biochemical-purified fractions. Such approach released all the original structures we know, with biological activity sometimes different from one isoform to another. Then, molecular biology techniques were applied to molluscan AMPs starting in 1999. Complete screening of genome expression in various situations became available, as well as some exotic Molluscs, the ones collected from deep-sea hydrothermal vent, for instance. Full sequences of active peptides and precursors, and gene organizations were established. A breakthrough consisted in the discovery of numerous AMP variants, even within the same animal. In addition, computer homology revealed the existence of already known AMPs in new studied molluscan species.

Enterococcus faecalis infection can cause serious diseases including cancer development. Recently it has been reported that mitochondrial reactive oxygen species (mROS) are required for host immune defenses against bacteria and many mutations in mitochondrial electron transport chain (mETC) genes have an effect on mROS production. To identify the exact role of mROS during E. faecalis infection, we thus decide to knockdown the expression of mev-1 and isp-1 in Caenorhabditis elegans using RNAi. The knockdown of mev-1 and isp-1 causes increased susceptibility and increased resistance to E. faecalis infection, respectively. The mev-1(RNAi) can also down-regulate antimicrobial genes (C17H12.8, mtl-1 and bli-3), whereas these antimicrobial genes are up-regulated in isp-1(RNAi) animals after bacterial infection. Further, significant increase of mitochondrial superoxide and mitochondrial sod expressions have been observed in isp-1(RNAi) animals. Conversely, the mev-1(RNAi) worms show a decrease of mitochondrial superoxide and mitochondrial sod expressions. Prooxidant paraquat, which is a mitochondrial superoxide generator, can increase survival rate of mev-1(RNAi) animals after E. faecalis infection. All together, the enhancement of mitochondrial superoxide contributes to anti-bacterial immunity and a better knowledge of them should open new avenues for preventive strategies against bacterial infection and also limiting the development of infection-associated cancer.

Bone sections including either titanium or porous tantalum implant devices used for interbody spinal fusion were investigated with position-resolved small angle X-ray scattering (sSAXS). The samples were obtained from six-month-old pigs that had undergone surgery three months prior to sacrifice. The aim of the study was to explore the possibility of using sSAXS to obtain information about thickness, orientation and shape/arrangement of the mineral crystals in bone near the implant surfaces. Detailed sSAXS scans were carried out in two different regions of bone adjacent to the implant in each of the implant samples. In the implant vicinity the mineral crystals tended to be aligned with the surface of the implants. The mean crystal thickness was between 2.1 and 3.0 nm. The mineral crystal thickness increased linearly with distance from the implant in both regions of the porous tantalum implant and in one of the regions in the titanium sample. In the second region of the titanium sample the thickest mineral crystals were found close to the implant surface. The observed differences in mineral thickness with distance from the implant surfaces might be explained by differences in mechanical load induced by the implant material and the geometrical design of the implant. The study shows that sSAXS is a powerful tool to characterize the nanostructure of bone near implant surfaces.

Plasma cholesteryl ester (CE) synthesis by lecithin cholesterol acyltransferase (LCAT) is activated by apolipoprotein (apo)A-I. We studied the effect of plasma apoA-I concentration on LCAT activation, using normal, heterozygous or homozygous apoA-I-deficient mice made by gene targeting. Plasma esterified cholesterol concentrations of mice fed chow diets were ordered (mean +/- SEM): 105 +/- 7 (normal) > 70 +/- 5 (heterozygotes) > 26 +/- 2 (homozygotes) mg/dl. Plasma free cholesterol concentrations were similar among the three genotypes. Endogenous LCAT activity, measured as the decrease in plasma free cholesterol after a 1 h incubation at 37 degrees C, was ordered: 44 +/- 3 (normal) > 21 +/- 2 (heterozygotes) > 5 +/- 1 (homozygotes) nmol CE formed/h per ml plasma. Using a recombinant exogenous substrate consisting of egg yolk phospholipid, [14C]cholesterol, and apoA-I, CE formation of normals and heterozygotes was similar (27.4 +/- 0.6 and 28.8 +/- 1.3 nmol/h per ml plasma, respectively), but was significantly less for homozygotes (19.2 +/- 1.7 nmol/h per ml plasma). However, using a small unilamellar vesicle substrate particle containing phospholipid and [14C]cholesterol, CE formation was ordered: 1.6 +/- 0.1 (normal) = 1.6 +/- 0.1 (heterozygotes) > 0.6 +/- 0.1 (homozygotes) nmol/h per ml plasma; addition of apoA-I to the plasma of homozygous animals restored CE formation to normal levels (1.6 +/- 0.1). CE fatty acid analysis demonstrated that plasma from homozygous mice contained significantly more saturated and monounsaturated and fewer polyunsaturated fatty acids compared to normal and heterozygous mice.(ABSTRACT TRUNCATED AT 250 WORDS)

Estrogen treatment raises plasma high density lipoprotein (HDL) levels, which may reduce cardiovascular risk. To identify the responsible mechanisms as well as the importance of the route of administration, we treated eight healthy postmenopausal women in a double-blind crossover study with three treatments for 6 weeks each: oral estradiol, 2 mg daily; transdermal estradiol, 0.1 mg twice weekly; and placebo. At the end of each treatment, apoA-I of HDL2 (d 1.063-1.125 g/ml) and HDL3 (d 1.125-1.210 g/ml) was endogenously labeled by a constant intravenous infusion of trideuterated leucine. HDL2 and HDL3 were separated by preparative ultracentrifugation. The pool sizes and enrichment curves of HDL apoA-I were used to calculate production rates and fractional catabolic rates (FCR). Oral estradiol increased the levels of HDL2 apoA-I by 37% (P < 0.005) and of HDL3 apoA-I by 11% (P < 0.05). These increased apoA-I levels resulted entirely from increased production, by 36% for HDL2 (P < 0.01), and by 19% for HDL3, (P < 0.05) as their FCRs were unchanged (0.20 pool/d with placebo and 0.21 with estradiol for HDL2, and 0.19 with placebo and 0.21 with estradiol for HDL3). The isotopic enrichment curves of HDL2 apoA-I and HDL3 apoA-I were identical, implying that apoA-I rapidly cycles between HDL particles, or that rapid interconversion of these subfractions occurs. The changes in HDL apoA-I metabolic rates were positively correlated with changes in VLDL-apoB metabolic rates measured previously. Transdermal estradiol, with systemic potency similar to that of oral estradiol, had no significant effect on HDL levels or metabolic rates. Thus, the “first pass” effect of oral estradiol on the liver and/or intestine appears to increase HDL apoA-I levels (particularly in HDL2) by increasing HDL apoA-I production, and not by reducing HDL apoA-I catabolism.

To help address the built environmental issues of both heat island and stormwater runoff, strategies that make pavements cooler and permeable have been investigated through measurements and modeling of a set of pavement test sections. The investigation included the hydraulic and thermal performance of the pavements. The permeability results showed that permeable interlocking concrete pavers have the highest permeability (or infiltration rate, ∼0.5 cm s ^−1 ). The two permeable asphalt pavements showed the lowest permeability, but still had an infiltration rate of ∼0.1 cm s ^−1 , which is adequate to drain rainwater without generating surface runoff during most typical rain events in central California. An increase in albedo can significantly reduce the daytime high surface temperature in summer. Permeable pavements under wet conditions could give lower surface temperatures than impermeable pavements. The cooling effect highly depends on the availability of moisture near the surface layer and the evaporation rate. The peak cooling effect of watering for the test sections was approximately 15–35 °C on the pavement surface temperature in the early afternoon during summer in central California. The evaporative cooling effect on the pavement surface temperature at 4:00 pm on the third day (25 h after watering) was still 2–7 °C lower compared to that on the second day, without considering the higher air temperature on the third day. A separate and related simulation study performed by UCPRC showed that full depth permeable pavements, if designed properly, can carry both light-duty traffic and certain heavy-duty vehicles while retaining the runoff volume captured from an average California storm event. These preliminarily results indicated the technical feasibility of combined reflective and permeable pavements for addressing the built environment issues related to both heat island mitigation and stormwater runoff management.
