Research on the development of environmentally friendly plant protection systems against fungal pathogens using symbiotic bacteria Xenorhabdus sp. – symbionts of entomopathogenic nematodes (EPN) has been a new direction in agricultural practice in recent years and undoubtedly represent relevance and scientific significance. The studies used suspensions of live and autoclaved cultures of symbiotic bacteria of symbionts of various types of EPN (Steinernema carpocapsae, S. feltiae and S. feltiae protense) with a bacterial cell titer of 107 CFU/ml in comparison with the biological preparation Phytosporin-M (dough) and water as a control. In laboratory conditions at a temperature of 25 oC, there were established differences in the antibiotic activity of the primary forms of producers of Xenorhabdus sp., isolated from different types of EPN. The greatest inhibition of the fungal growth zone on the 4th day was observed for metabolites of the S. carpocapsae strain against Alternaria solani. Biological effectiveness in suppressing this pathogen was 51 %. Field research conducted in 2022-2023 in the conditions of the Republic of Karelia on the mid-early potato variety ʻRed Scarlettʼ, showed that under epiphytotic conditions (low air temperatures and excessive waterlogging) double spraying of vegetative plants with a suspension of live and autoclaved cultures of symbiotic bacteria (EPN-1-1, EPN-2 and EPN-2 -1) reduced the development of rhizoctoniosis compared to the control variant by 50, 64 and 60 %, respectively. It was found that double treatment with a live and autoclaved aqueous suspension of bacteria, symbionts of the subspecies S. feltiae was more effective and ensured a reduction in the degree of development of scab symptoms by 1.3–2.8 times and the spread of rhizoctonia by 1.5–2.0 times. It has been also established that 2-fold spraying of plants with a live and autoclaved suspension of symbiotic bacteria S. feltiae during the growing season significantly increases the yield of tubers by 35–22 %, respectively. Thus, the use of biologically active secondary metabolites of Xenorhabdus sp. has significant potential as biological plant protection agents against potato pathogens.

The combined use of drugs with different mechanisms of action is the main principle of musculoskeletal pain control in rheumatic diseases. However, there are few studies evaluating the efficacy of this approach in real practice.Objective: to determine the efficacy and safety of the combined use of celecoxib, diacerein, and the combination of glucosamine + chondroitin in osteoarthritis (OA) and chronic nonspecific low back pain (NSLBP). Material and methods. Statistical analysis of data obtained during a 3-month open observational study was performed. We included 1569 patients (63.6 % women and 36.4 % men, mean age 58.7 ± 11.0 years) with knee OA (kOA), hip OA (hOA), generalized OA (gOA), and chronic NSLBP with moderate/severe pain (≥ 4 on a numeric rating scale, NRS 0–10) who required nonsteroidal anti-inflammatory drugs. Celecoxib 200 mg twice daily was prescribed, with the dose reduced to 200 mg per day or taken “as needed" after significant pain relief; diacerein 50 mg twice daily; and a medication of glucosamine 250 mg and chondroitin 200 mg, 2 capsules 2–3 times daily. Outcomes were assessed after 3 months using the dynamics of pain, fatigue, dysfunction (according to NRS), and the “Patient Acceptable Symptom State” (PASS) indicator. Results and discussion. 80.2 % of patients completed the 3 month course of treatment, 4.4 % discontinued treatment due to adverse events (AEs), and for 15.4 % of patients there was no follow-up. After 3 months of treatment ≥ 50 % decrease (from baseline) in the severity of symptoms was noted in 83.4 % of patients for pain on movement, in 83.7 % for pain at rest, in 78.6 % for pain at night, in 80.8 % for dysfunction, and in 83.4 % for fatigue. 87.7 % of patients reported PASS. There were no significant differences in treatment outcomes for different localizations of OA and NSLBP: a ≥ 50 % pain reduction in kOA was achieved in 81.6 % of patients, in hOA – in 82.2 %, in gOA – in 85.0 %, in NSLBP – in 88.1 %. AEs were registered in 350 (22.4 %) patients, the most frequent was dyspepsia (n = 280, 17.8 %), diarrhea was recorded in 37 (2.4 %) cases. No serious AEs requiring hospitalization were registered. Conclusion. Combination therapy with celecoxib, diacerein, and a combination of glucosamine and chondroitin significantly reduces the severity of symptoms of OA and NSLBS.

8. Heating Structure of the Second Half of the XVIII Century (based on the archaeological materials of Tobolsk)

This study deals with description of the furnace structure, studied during archaeological work on the territory of the upper part of one of the oldest towns in Siberia – Tobolsk (founded in 1587). Stratigraphic surveys together with a numismatic collection allowed the authors to date the revealed heating structure to the second half of the XVIII century. It was established that the furnace was built at the bottom of the cellar; its base was a frame of scaffolds, on which the brickwork was made (the reuse of bricks from pulled down furnaces was recorded). The studied part of the furnace base has been preserved to a height of 1 m; the maximum studied dimensions are 170×160 cm. Based on the analysis of the masonry, the furnace found to be rectangular in shape. It was decorated with tiles. The authors believe that the studied furnace design is not typical for Tobolsk of the second half of the XVIII century. It was in the house of a wealthy citizen.

In yeast Saccharomyces cerevisiae, there are two translation termination factors, eRF1 (Sup45) and eRF3 (Sup35), which are essential for viability. Previous studies have revealed that presence of nonsense mutations in these genes leads to amplification of mutant alleles (sup35-n and sup45-n), which appears to be necessary for the viability of such cells. However, the mechanism of this phenomenon remained unclear. In this study, we used RNA-Seq and proteome analysis to reveal the complete set of gene expression changes that occur during cellular adaptation to the introduction of the sup35-218 nonsense allele. Our analysis demonstrated significant changes in the transcription of genes that control the cell cycle: decreases in the expression of genes of the anaphase promoting complex APC/C (APC9, CDC23) and their activator CDC20, and increases in the expression of the transcription factor FKH1, the main cell cycle kinase CDC28, and cyclins that induce DNA biosynthesis. We propose a model according to which yeast adaptation to nonsense mutations in the translation termination factor genes occurs as a result of a delayed cell cycle progression beyond the G2-M stage, which leads to an extension of the S and G2 phases and an increase in the number of copies of the mutant sup35-n allele.

Many retinal degenerative diseases result in vision impairment or permanent blindness due to photoreceptor loss or dysfunction. It has been observed that Pde6brd1 mice (rd1), which carry a spontaneous nonsense mutation in the pde6b gene, have a strong phenotypic similarity to patients suffering from autosomal recessive retinitis pigmentosa. In this study, we present a novel mouse model of retinitis pigmentosa generated through pde6b gene knockout using CRISPR/Cas9 technology. We compare this Pde6b-KO mouse model to the rd1 mouse model to gain insights into the progression of retinal degeneration. The functional assessment of the mouse retina and the tracking of degeneration dynamics were performed using electrophysiological methods, while retinal morphology was analyzed through histology techniques. Interestingly, the Pde6b-KO mouse model demonstrated a higher amplitude of photoresponse than the rd1 model of the same age. At postnatal day 12, the thickness of the photoreceptor layer in both mouse models did not significantly differ from that of control animals; however, by day 15, a substantial reduction was observed. Notably, the decline in the number of photoreceptors in the rd1 model occurred at a significantly faster rate. These findings suggest that the C3H background may play a significant role in the early stages of retinal degeneration.

In the evolution of invertebrates, the transition from egg-layers to brooders occurred many times. However, the molecular mechanisms underlying this transition are still not well understood. Recently diverged species genus Littorina (Mollusca, Gastropoda, Caenogastropoda, Littorinimorpha): Littorina saxatilis, L. arcana, L. compressa, L. obtusata and L. fabalis might be a fruitful model for elucidation of these mechanisms. All five species sympatrically inhabit an intertidal zone. Only L. saxatilis is ovoviviparous while the other four species form clutches. Although in L. saxatilis jelly gland of the pallial oviduct function as a brood pouch, it is not deeply modified at the morphological level in comparison to egg-laying relatives. Comparative analysis of transcriptomic profiles of the pallial oviducts of these closely related species might help to uncover the molecular mechanisms of the egg-laying to brooding transition. Unraveling of the mechanisms underlying this transition in L. saxatilis is important not only in aspects of reproduction biology and strategy, but also in a broader view as an example of relatively fast evolutionary transformations. We generated an RNA-seq dataset (224 104 446 clean reads) for oviducts of five species genus Littorina. Libraries of all five species were sequenced using Illumina HiSeq 2500; additional reads for L. arcana were obtained using Illumina NovaSeq 6000. Transcriptomic profiles were analyzed in pooled samples (of three individuals) with two biological replicates for each species (each biological replicate was prepared and sequenced as a separate library). The transcriptome was assembled de novo and annotated with five assembles corresponding to each species. The raw data were uploaded to the SRA database, the BioProject IDs are PRJNA662103 (“obtusata” group) and PRJNA707549 (“saxatilis” group).

12. Burial Practice of Russian Population in Tobolsk Irtysh Region in 18th-19th Centuries: Cases of Deviations from Official Legislation

The article deals with the issue of non-compliance with the legislation in the burial practice of the Russian population of the Tobolsk Irtysh region in the 18th-19th centuries. The archival materials of the Fund of the Tobolsk Spiritual Consistory of the State Archive in Tobolsk, reflecting the cases of deviation from the legally established practice of burial, are introduced into scientific circulation. The materials of records management on the facts of the death of a newborn child of the settler Ivan Minin in 1774, who died unbaptized, are analyzed, as well as the death and burial of the peasant Alexey Vakhrushev made without the consent of the civil authorities in 1815 in the territory of the Abalak Znamensky monastery. As a source for the reconstruction of the funeral rites of the Russian population of Siberia, the materials obtained as a result of archaeological research of two children’s burials from the Russian settlement “Yarkova village” in the Tobolsk Irtysh region are involved. Based on the analysis of the features of the burial rite, the burials are dated by the middle of the 19th - early 20th centuries. In connection with the absence of other burials nearby, it is concluded that these burials are a “home” (family) cemetery of unbaptized babies, located in the garden near the cliff, free from economic activity on the site of the peasant estate.

13. Gut microbiota in chronic liver diseases in children

The impact of gut microbiota on the development of various diseases is of great interest to researchers. However, data on the taxonomic diversity of the intestinal microbiota in chronic liver diseases in children are lacking.Purpose. To study the taxonomic diversity of the fecal microbiota in children with chronic liver diseases in comparison with healthy patients.Material and methods. A metagenomic analysis of the intestinal microbiota of 24 children with chronic liver diseases (mean age 10.3 ± 4.7 years) was carried out with the isolation of the target fragment of the 16S rRNA gene. The group included 18 children with autoimmune liver diseases and 6 children with non-autoimmune liver diseases. The comparison group consisted of fecal samples of 34 apparently healthy children.Results. The conducted study revealed 684 types of microorganisms in the studied samples of patients’ feces. An analysis of the conducted studies showed that fecal samples of healthy children and patients with chronic liver diseases differ in bacterial diversity. The dominant taxa in healthy children were Neisseria flavescens, in patients with chronic liver diseases, the dominant taxa were Bifidobacterium longum, Bifidobacterium adolescentis, Blautia massiliensis. At the same time, Bifidobacterium longum, Bifidobacterium adolescentis, Blautia massiliensis in fecal samples of patients with chronic liver diseases was 8 times as high.Conclusion. Studies have shown differences in the composition of the intestinal microbiota in healthy children and children with chronic liver diseases.

A number of [PSI+]-no-more (PNM) mutations, eliminating [PSI+] prion, were previously described in SUP35. In this study, we designed and analyzed a new PNM mutation based on the parallel in-register β-structure of Sup35 prion fibrils suggested by the known experimental data. In such an arrangement, substitution of non-charged residues by charged ones may destabilize the fibril structure. We introduced Q33K/A34K amino acid substitutions into the Sup35 protein, corresponding allele was called sup35-M0. The mutagenized residues were chosen based on ArchCandy in silico prediction of high inhibitory effect on the amyloidogenic potential of Sup35. The experiments confirmed that Sup35-M0 leads to the elimination of [PSI+] with high efficiency. Our data suggested that the elimination of the [PSI+] prion is associated with the decreased aggregation properties of the protein. The new mutation can induce the prion with very low efficiency and is able to propagate only weak [PSI+] prion variants. We also showed that Sup35-M0 protein co-aggregates with the wild-type Sup35 in vivo. Moreover, our data confirmed the utility of the strategy of substitution of non-charged residues by charged ones to design new mutations to inhibit a prion formation.

16. Identification of New FG-Repeat Nucleoporins with Amyloid Properties

Amyloids are fibrillar protein aggregates with a cross-β structure. More than two hundred different proteins with amyloid or amyloid-like properties are already known. Functional amyloids with conservative amyloidogenic regions were found in different organisms. Protein aggregation appears to be beneficial for the organism in these cases. Therefore, this property might be conservative for orthologous proteins. The amyloid aggregates of the CPEB protein were suggested to play an important role in the long-term memory formation in Aplysia californica, Drosophila melanogaster, and Mus musculus. Moreover, the FXR1 protein demonstrates amyloid properties among the Vertebrates. A few nucleoporins (e.g., yeast Nup49, Nup100, Nup116, and human Nup153 and Nup58), are supposed or proved to form amyloid fibrils. In this study, we performed wide-scale bioinformatic analysis of nucleoporins with FG-repeats (phenylalanine–glycine repeats). We demonstrated that most of the barrier nucleoporins possess potential amyloidogenic properties. Furthermore, the aggregation-prone properties of several Nsp1 and Nup100 orthologs in bacteria and yeast cells were analyzed. Only two new nucleoporins, Drosophila melanogaster Nup98 and Schizosaccharomyces pombe Nup98, aggregated in different experiments. At the same time, Taeniopygia guttata Nup58 only formed amyloids in bacterial cells. These results rather contradict the hypothesis about the functional aggregation of nucleoporins.

17. The easternmost record of the largest anguine lizard that has ever lived – Pseudopus pannonicus (Squamata, Anguidae): new fossils from the late Neogene of Eastern Europe

We here report on new material of Pseudopus pannonicus, the iconic and largest-known representative of the lizard clade Anguinae, from several late Neogene localities across Moldova, Ukraine, and regions of the North Caucasus – the last representing the easternmost known occurrence of this extinct species. Today, Pseudopus apodus, the last extant Pseudopus representative, is found in a variety of habitats ranging from South-East Europe to Central Asia. In the late Cenozoic of Europe, however, several extinct species of Pseudopus existed. Among them, interestingly, P. pannonicus displayed the largest spatiotemporal range of the genus, occurring from Spain to the North Caucasus and known from the Late Miocene to the Early Pleistocene. Although it has been reported in a plethora of European localities, P. pannonicus is a taxon “with several questionings related to its few diagnostic features vs. numerous features shared with P. apodus”. The elements described here exhibit some variability, but their overall morphology undoubtedly resembles that of previously described material of P. pannonicus. The lacrimal from Tatareshty, moreover, represents the first fossil lacrimal reported for P. pannonicus. Besides, the fairly complete maxilla with a length of almost 3.7 cm is the largest maxilla ever reported for this taxon, expanding our knowledge of its gigantism. In addition, several features are described and discussed regarding their diagnostic relevance for P. pannonicus. The relationship between body size and some of these features was tested statistically. Consequently, two cranial characters and one vertebral feature peculiar to P. pannonicus were retained in the diagnosis of the species.

The article presents the result of treatment of patient P. with trauma caused by an attack of a bear during the hunting. Patient had open comminuted displaced fracture of the lower third of right ulnar bone with formation of large infected wound of right forearm and right hand with injury of flexor tendons of IV and V fingers, extensor tendon of V finger, ulnar nerve. At the admission to the Central District Hospital of Mama, primary surgical treatment of wound, plaster immobilization were conducted, wound weren't stitched up. At the treatment in Irkutsk Scientific Center of Surgery and Traumatology, secondary surgical treatment of wound of right forearm, necretomy, ultrasound treatment of wound, laceration repair of wound with local tissues, transosseous osteosynthesis of right forearm - hand were conducted. As a result of conducted treatment we achieved total reduction of necrotic and purulent process, repair of vast infected wound of forearm and hand, full adhesion of fracture of ulnar bone and restoration of a function of right arm. Using ultrasound treatment of wounds and transosseous osteosynthesis at openfractures of forearm bones and chronic purulent infection is a treatment of choice and can cause good clinical outcome.

Yeast is a convenient model for studying protein aggregation as it is known to propagate amyloid prions. [PSI+] is the prion form of the release factor eRF3 (Sup35). Aggregated Sup35 causes defects in termination of translation, which results in nonsense suppression in strains carrying premature stop codons. N-terminal and middle (M) domains of Sup35 are necessary and sufficient for maintaining [PSI+] in cells while preserving the prion strain’s properties. For this reason, Sup35NM fused to fluorescent proteins is often used for [PSI+] detection and investigation. However, we found that in such chimeric constructs, not all fluorescent proteins allow the reliable detection of Sup35 aggregates. Particularly, transient overproduction of Sup35NM-mCherry resulted in a diffuse fluorescent pattern in the [PSI+] cells, while no loss of prions and no effect on the Sup35NM prion properties could be observed. This effect was reproduced in various unrelated strain backgrounds and prion variants. In contrast, Sup35NM fused to another red fluorescent protein, TagRFP-T, allowed the detection of [PSI+] aggregates. Analysis of protein lysates showed that Sup35NM-mCherry is actively degraded in the cell. This degradation was not caused by vacuolar proteases and the ubiquitin-proteasomal system implicated in the Sup35 processing. Even though the intensity of this proteolysis was higher than that of Sup35NM-GFP, it was roughly the same as in the case of Sup35NM-TagRFP-T. Thus, it is possible that, in contrast to TagRFP-T, degradation products of Sup35NM-mCherry still preserve their fluorescent properties while losing the ability to decorate pre-existing Sup35 aggregates. This results in diffuse fluorescence despite the presence of the prion aggregates in the cell. Thus, tagging with fluorescent proteins should be used with caution, as such proteolysis may increase the rate of false-negative results when detecting prion-bearing cells.

Over the last 25 years, researchers, mostly paleontologists, have developed a system of rank-free, phylogenetically defined names for the primary clades of turtles. As these names are not considered established by the PhyloCode, the newly created nomenclatural system that governs the naming of clades, we take the opportunity to convert the vast majority of previously defined clade names for extinct and extant turtles into this new nomenclatural framework. Some previously defined names are converted with minor adjustments. We also define a number of new clade names to close apparent nomenclatural gaps. In total, we establish 113 clade names, of which 79 had already received phylogenetic definitions and 34 are new.

A measurement of the inclusive jet production in proton-proton collisions at the LHC at root s = 13TeV is presented. The double-differential cross sections are measured as a function of the jet transverse momentum p(T) and the absolute jet rapidity vertical bar y vertical bar. The anti- k(T) clustering algorithm is used with distance parameter of 0.4 (0.7) in a phase space region with jet p(T) from 97 GeV up to 3.1TeV and vertical bar y vertical bar < 2.0. Data collected with the CMS detector are used, corresponding to an integrated luminosity of 36.3 fb(-1) (33.5 fb(-1)). The measurement is used in a comprehensive QCD analysis at next-to-next-to-leading order, which results in significant improvement in the accuracy of the parton distributions in the proton. Simultaneously, the value of the strong coupling constant at the Z boson mass is extracted as alpha(S)(m(Z)) = 0.1170 +/- 0.0019. For the first time, these data are used in a standard model effective field theory analysis at next-to-leading order, where parton distributions and the QCD parameters are extracted simultaneously with imposed constraints on the Wilson coefficient c(1) of 4-quark contact interactions.

Differential cross section measurements of W±γ production in proton-proton collisions at s=13 TeV are presented. The data set used in this study was collected with the CMS detector at the CERN LHC in 2016-2018 with an integrated luminosity of 138 fb-1. Candidate events containing an electron or muon, a photon, and missing transverse momentum are selected. The measurements are compared with standard model predictions computed at next-to-leading and next-to-next-to-leading orders in perturbative quantum chromodynamics. Constraints on the presence of TeV-scale new physics affecting the WWγ vertex are determined within an effective field theory framework, focusing on the O3W operator. A simultaneous measurement of the photon transverse momentum and the azimuthal angle of the charged lepton in a special reference frame is performed. This two-dimensional approach provides up to a factor of ten more sensitivity to the interference between the standard model and the O3W contribution than using the transverse momentum alone. © 2022 CERN.

This paper is devoted to the description of a new species of xinjiangchelyid turtle – Annemys variabilis sp. nov. – represented by thousands of isolated bones and several more complete specimens from the Middle Jurassic (Bathonian) Itat Formation of the Berezovsk coal mine, Krasnoyarsk Territory, western Siberia, Russia. The description is based on a sample of the best-preserved specimens. The new species differs from other Annemys spp. by a combination of cranial and shell characters. The basisphenoids (about 200 specimens) of A. variabilis demonstrate variation in 12 characters, some of which are reported in turtles for the first time. About 20 shell characters of A. variabilis are subject to ontogenetic or interindividual variation. Some of these characters are variable in other Annemys spp. and other xinjiangchelyids. The formula of the cervical vertebrae [(2(3()4()5()6()7()8) or (2(3()4()5()6()7()8(] with two opisthocoelous vertebrae (2 and 3), four or five amphicoelous vertebrae (4–7 or 4–8), and one procoelous vertebra is unique for this species among xinjiangchelyids, which were previously known to have only amphicoelous cervicals. Other characters of the non-shell postcranium correspond to those of other xinjiangchelyids. Of the three phylogenetic analyses performed in this study, Analysis 1 does not support the monophyly of either Xinjiangchelyidae or Annemys, Analysis 2 shows paraphyly of Xinjiangchelyidae and monophyly of the Annemys clade (A. latiens, A. levensis, A. variabilis and A. wusu), and Analysis 3 supports monophyly of the Annemys clade with A. latiens, A. levensis and A. variabilis, only in the majority rule consensus tree. The diversity of the turtle assemblage of the Itat Formation is re-assessed as containing 2–3 taxa (A. variabilis [=Testudines indet. 2], Testudines indet. 1 and 3), which is in agreement with similar diversities seen in some other Middle Jurassic Asian turtle assemblages. Darevskia rock lizards include 29 sexual and seven parthenogenetic species of hybrid origin distributed in the Caucasus. All seven parthenogenetic species of the genus Darevskia were formed as a result of interspecific hybridization of only four sexual species. It remains unknown what are the main advantages of interspecific hybridization along with switching on parthenogenetic reproduction in evolution of reptiles. Data on whole transcriptome sequencing of parthenogens and their parental ancestors can provide value impact in solving this problem. Here we have sequenced ovary tissue transcriptomes from unisexual parthenogenetic lizard D. unisexualis and its parental bisexual ancestors to facilitate the subsequent annotation and to obtain the collinear characteristics for comparison with other lizard species. Here we report generated RNAseq data from total mRNA of ovary tissues of D. unisexualis, D. valentini and D. raddei with 58932755, 51634041 and 62788216 reads. Obtained RNA reads were assembled by Trinity assembler and 95141, 62123, 61836 contigs were identified with N50 values of 2409, 2801 and 2827 respectively. For further analysis top Gene Ontology terms were annotated for all species and transcript number was calculated. The raw data were deposited in the NCBI SRA database (BioProject PRJNA773939). Amyloids are fibrillar protein aggregates with a cross-β structure and unusual features, including high resistance to detergent or protease treatment. More than two hundred different proteins with amyloid or amyloid-like properties are already known. Several examples of nucleoporins (e.g., yeast Nup49, Nup100, Nup116, and human NUP153) are supposed to form amyloid fibrils. In this study, we demonstrated an ability of the human NUP58 nucleoporin to form amyloid aggregates in vivo and in vitro. Moreover, we found two forms of NUP58 aggregates: oligomers and polymers stabilized by disulfide bonds. Bioinformatic analysis revealed that all known orthologs of this protein are potential amyloids which possess several regions with conserved ability to aggregation. The biological role of nucleoporin amyloid formation is debatable. We suggest that it is a rather abnormal process, which is characteristic for many proteins implicated in phase separation.

Genus Littorina subgenus Neritrema (Mollusca, Caenogastropoda) includes the “obtusata” group of closely related species (Littorina obtusata and L. fabalis). The anatomy of the adult reproductive system (pallial oviduct) is the only reliable feature used for species identification in females of these species. Reproductive system anatomy and reproduction-associated proteins often diverge between sibling species. Despite being of high evolutionary interest, the molecular basis of this divergence remains poorly understood. We performed proteotranscriptomic comparison of oviducts of L. obtusata and L. fabalis by RNA-seq on Illumina HiSeq 2500 and two-dimensional protein electrophoresis (2D DIGE) with MS/MS identification of the species-specific proteins. The interspecies differences in the oviduct were associated with (1) metabolic proteins reflecting overall physiological differences between L. obtusata and L. fabalis, (2) receptor proteins, and (3) transcripts related to transposable elements (TEs). Various receptors identified may recognize a wide variety of ligands from pathogen-associated molecular patterns to specific carbohydrates on the sperm surface. Therefore, these may participate in immune defense as well as in sperm storage and regulation. Species-specificity of multiple TE sequences (coding for reverse transcriptase and ribonuclease H) may indicate the important role of these genomic elements in the Littorina species divergence, which has not been reported previously.

Simple Summary Genitalia and reproduction-associated proteins are often species-specific and might evolve rapidly. The situation in which the morphology of the reproductive system is the only difference between two or several closely related species has been reported on multiple occasions. Nevertheless, the reasons for such rapid divergence of the reproductive system is still poorly investigated. To shed some light on the issue, we performed a transcriptomic and proteomic comparison of pallial oviducts from the two sibling species of gastropods Littorina obtusata and L. fabalis. The main identified differences were associated with three functional groups of genes: transposable elements, which enhance genome variation and promote the evolution of new genes, receptor proteins potentially involved in friend or foe recognition, and various enzymes. We hypothesize that these functional groups reflect both the mechanism (transposable elements) and the directions (friend or foe recognition and reproductive physiology) of the rapid evolution of the reproductive system. (1) Hypophosphatasia (HPP) is a rare inherited disease caused by mutations (pathogenic variants) in the ALPL gene which encodes tissue-nonspecific alkaline phosphatase (TNSALP). HPP is characterized by impaired bone mineral metabolism due to the low enzymatic activity of TNSALP. Knowledge about the structure of the gene and the features and functions of various ALPL gene variants, taking into account population specificity, gives an understanding of the hereditary nature of the disease, and contributes to the diagnosis, prevention, and treatment of the disease. The purpose of the study was to describe the spectrum and analyze the functional features of the ALPL gene variants, considering various HPP subtypes and clinical symptoms in Russian children. (2) From 2014–2021, the study included the blood samples obtained from 1612 patients with reduced alkaline phosphatase activity. The patients underwent an examination with an assessment of their clinical symptoms and biochemical levels of TNSALP. DNA was isolated from dried blood spots (DBSs) or blood from the patients to search for mutations in the exons of the ALPL gene using Sanger sequencing. The PCR products were sequenced using a reagent BigDye Terminator 3.1 kit (Applied Biosystems). Statistical analysis was performed using the GraphPad Prism 8.01 software. (3) The most common clinical symptoms in Russian patients with HPP and two of its variants (n = 22) were bone disorders (75%), hypomyotonia (50%), and respiratory failure (50%). The heterozygous carriage of the causal variants of the ALPL gene was detected in 225 patients. A total of 2 variants were found in 27 patients. In this group (n = 27), we identified 28 unique variants of the ALPL gene, of which 75.0% were missense, 17.9% were frameshift, 3.6% were splicing variants, and 3.6% were duplications. A total of 39.3% (11/28) of the variants were pathogenic, with two variants being probably pathogenic, and 15 variants had unknown clinical significance (VUS). Among the VUS group, 28.6% of the variants (7/28) were discovered by us for the first time. The most common variants were c.571G > A (p.Glu191Lys) and c.1171del (Arg391Valfs*12), with frequencies of 48.2% (13/28) and 11% (3/28), respectively. It was found that the frequency of nonsense variants of the ALPL gene was higher (p < 0.0001) in patients with the perinatal form compared to the infantile and childhood forms of HPP. Additionally, the number of homozygotes in patients with the perinatal form exceeded (p < 0.01) the frequencies of these genotypes in children with infantile and childhood forms of HPP. On the contrary, the frequencies of the compound-heterozygous and heterozygous genotypes were higher (p < 0.01) in patients with infantile childhood HPP than in perinatal HPP. In the perinatal form, residual TNSALP activity was lower (p < 0.0005) in comparison to the infantile and childhood (p < 0.05) forms of HPP. At the same time, patients with the heterozygous and compound-heterozygous genotypes (mainly missense variants) of the ALPL gene had greater residual activity (of the TNSALP protein) regarding those homozygous patients who were carriers of the nonsense variants (deletions and duplications) of the ALPL gene. Residual TNSALP activity was lower (p < 0.0001) in patients with pathogenic variants encoding the amino acids from the active site and the calcium and crown domains in comparison with the nonspecific region of the protein.

Thousands of yeast genomes have been sequenced with both traditional and long-read technologies, and multiple observations about modes of genome evolution for both wild and laboratory strains have been drawn from these sequences. In our study, we applied Oxford Nanopore and Illumina technologies to assemble complete genomes of two widely used members of a distinct laboratory yeast lineage, the Peterhof Genetic Collection (PGC), and investigate the structural features of these genomes including transposable element content, copy number alterations, and structural rearrangements. We identified numerous notable structural differences between genomes of PGC strains and the reference S288C strain. We discovered a substantial enrichment of mid-length insertions and deletions within repetitive coding sequences, such as in the SCH9 gene or the NUP100 gene, with possible impact of these variants on protein amyloidogenicity. High contiguity of the final assemblies allowed us to trace back the history of reciprocal unbalanced translocations between chromosomes I, VIII, IX, XI, and XVI of the PGC strains. We show that formation of hybrid alleles of the FLO genes during such chromosomal rearrangements is likely responsible for the lack of invasive growth of yeast strains. Taken together, our results highlight important features of laboratory yeast strain evolution using the power of long-read sequencing.

A search for electroweak production of supersymmetric (SUSY) particles in final states with one lepton, a Higgs boson decaying to a pair of bottom quarks, and large missing transverse momentum is presented. The search uses data from proton-proton collisions at a center-of-mass energy of 13 TeV collected using the CMS detector at the LHC, corresponding to an integrated luminosity of 137 fb(-1). The observed yields are consistent with backgrounds expected from the standard model. The results are interpreted in the context of a simplified SUSY model of chargino-neutralino production, with the chargino decaying to a W boson and the lightest SUSY particle (LSP) and the neutralino decaying to a Higgs boson and the LSP. Charginos and neutralinos with masses up to 820 GeV are excluded at 95% confidence level when the LSP mass is small, and LSPs with mass up to 350 GeV are excluded when the masses of the chargino and neutralino are approximately 700 GeV.

The upper Eocene Youganwo Formation in Maoming Province, Guangdong Province, China, produced abundant remains of turtles and crocodiles, but mammalian remains are rare. The previously found mammals from Youganwo Formation include a nimravid carnivore and an amynodontid perissodactyl. Here we report on a new brontotheriid perissodactyl from the Youganwo Formation. Maobrontops paganus gen. et sp. nov. is described based on a maxillary fragment with P4 and M1–2 (SYSU-M-4). Maobrontops paganus gen. et sp. nov. is characterized by the combination of a simple P4 without a hypocone with derived molars having large molar fossae and large anterolingual cingular cusp. A parsimony implied weighting character analysis places Maobrontops gen. nov. in the clade Embolotheriita as a sister taxon to the terminal subclade containing Nasamplus, Protembolotherium, and Embolotherium. Maobrontops paganus gen. et sp. nov. is one of the largest Asian brontotheres. The brontotheriid fauna of South China is endemic and includes at least three valid taxa: Dianotitan from Brontotheriita and Pygmaetitan and Maobrontops gen. nov. from Embolotheriita.

We construct global smooth approximate solution to a scalar conservation law with arbitrary smooth monotonic initial data. Different kinds of singularities interactions which arise during the evolution of the initial data are described as well. In order to solve the problem, we use and further develop the weak asymptotic method, recently introduced technique for investigating nonlinear waves interactions.

During embryogenesis of the sea urchin, a large number of REs are expressed, but the role of these elements in the regulation of biological processes remains unknown. The aim of this study was to identify the RE expression at different stages of embryogenesis. REs occupied 44% of genomic DNA of Strongylocentrotus purpuratus. The most prevalent among these elements were the unknown elements—in total, they contributed 78.5% of REs (35% in total genome occupancy). It was revealed that the transcription pattern of genes and REs changes significantly during gastrulation. Using the de novo transcriptome assembly, we showed that the expression of RE is independent of its copy number in the genome. We also identified copies that are expressed. Only active RE copies were used for mapping and quantification of RE expression in the single-cell RNA sequencing data. REs expression was observed in all cell lineages and they were detected as population markers. Moreover, the primary mesenchyme cell (PMC) line had the greatest diversity of REs among the markers. Our data suggest a role for RE in the organization of developmental domains during the sea urchin embryogenesis at the single-cell resolution level.

Although fossils of Trionychidae are found as far back as the Early Cretaceous, their evolutionary history, including the origin of the crown clade, is poorly understood. Here, we use a microCT scan of the skull of ‘Trionyx’ kyrgyzensis from the late Early Cretaceous (Albian) of Kyrgyzstan as the basis for a thorough redescription of the cranial morphology of this species. New information includes the pattern of cranial circulation, which is comparable to all extant trionychids. A unique combination of cranial and postcranial characters of ‘Trionyx’ kyrgyzensis allows us to establish for it a new genus Petrochelys gen. nov. The data regarding Petrochelys kyrgyzensis are incorporated into a phylogenetic analysis. When Petrochelys kyrgyzensis is analysed without including additional, extinct taxa, it is placed in a highly derived position within the extant Nilssonia. However, the addition of five Cretaceous–Palaeogene taxa breaks up hypothesized long branches and results in more basal positions for Petrochelys kyrgyzensis within recovered trees. All results support the hypothesis that Petrochelys kyrgyzensis is a crown trionychid and a member of Trionychinae. Turtle horny shell has a scute pattern, which is conservative through evolution and across species. The discovery of epidermal placodes as the scute primordia and their strict topographical association to the somites of the turtle embryo suggested a new interpretation of the developmental mechanism of the scute pattern. Here, we tested the hypothesis that horny scutes develop from a mosaic of placodes corresponding exactly to the paths of myoseptae, with vertebral and pleural scutes developing staggered in adjacent segments, and marginal scutes developing in every segment. This scheme predicts little variation in marginals and suggests intercalary supernumerary scutes as potential variations for the vertebral and pleural rows. We examined spatial and numerical variations of the horny shell in 655 newly hatched olive ridley sea turtle, Lepidochelys olivacea, which is known to have a highly variable horny shell. In total, 120 patterns of carapacial scutes and 10 patterns of scutes on plastron, differing in the number and position of scutes were found. The number of vertebral scutes varied from 4 to 10. Variations with five, six and seven vertebrals occurred with the greatest and nearly equally frequency (31.5% on average). Pleural scutes were from 5 to 10 at one or both sides, and the typical symmetric pattern for sea turtles with five pairs of pleurals was only seen in ca. 12% of specimens. In contrast, the majority of the specimens (92.7%) had just 13 pairs of marginals, showing a stable normal pattern. Similarly, on plastron the horny scutes were conservative, too; about 85% of specimens standardly had six pairs of plastral scutes and all specimens had four pairs of inframarginals. Despite a high level of variation of vertebral and pleural scutes in olive ridley turtle, all patterns fall into the theoretical spectrum of possible variants predicted by the segment-dependent model of development of the turtle horny shell. Therefore, the results of our analysis support the existence of direct morphogenetic correlation between the number and distribution of normal and supernumerary scutes and metamere organization of the turtle embryo.

The article analyses the main definitions (concepts) of the notion «intelligentsia». The author justifies the use of term «intelligentsia» (as the tool of the research) in the notion of «knowledge workers». It also reveals the principles of counting the number of intelligentsia in Kubanregion at the turn of the XIX–XX centuries. Basing on the materials of the first Russian population census in 1897, author determines the number and occupational pattern of Kuban intelligentsia, its relative density in society. Kuban intelligentsia counted about 7000 people, that is 0,08 per cent of all regional population. The article also discusses the Kuban intelligentsia peculiarities, presents its activity conditions in Cossack region. The article analyses Kuban intelligentsia limits capabilities to influence the social life and the intellectual development of society.

This study was focused on neuropsychological analysis of the cognitive functions in patients with parasellar meningiomas undergoing radiation therapy. In these patients, the unilateral hippocampus adjacent to the tumor was partially exposed to irradiation. A novel approach was used to assess particular memory functionsincluding pattern separation, in subpopulations of subjects prior to the course of radiotherapy, immediately after the course, and 1–2 years after therapy. Our results show a tendency for pattern separation decline in the long-term follow-up. A possible association of these disorders with the effect of the hippocampal region irradiation and inhibition of adult neurogenesis is discussed. This paper presents the results of mathematical modeling of heat transfer in the field emission process in a conic cathode of small dimensions with its possible melting considered. It is shown that the possibility of melting is determined by the cathode vertex angle. The melting is modeled in the framework of the phase field system using the proposed methods for the formation of the liquid phase zone.

Herein we report morphologically and histologically diagnostic trionychid specimens from the Barremian–Aptian (129.4–113.0 Ma) of Japan. One specimen (FPDM-V9487) is an associated skeleton that consists of a scapula, a humerus, an ischium, and hypoplastra. The limb and girdle bones are similar in morphology to those of modern trionychids. The hypoplastron also resembles that of modern trionychids; however, it is distinctive in that it completely lacks callosities. The other three new specimens are fragmentary costals that show a reduction in the shell bones and an absence of scute sulci. Two of the newly reported costals and FPDM-V0127 were examined histologically. Bone fiber bundles organized in a plywood-like structure, which is unique to trionychids, were identified. Fossil occurrence data indicate that morphologically and histologically typical trionychids already inhabited the coastal region of Asia (e.g., western Japan) as early as the Aptian. Modern turtles exhibit a broad scope of ecological adaptations, including coastal marine and pelagic habitats, and, during their evolutionary history, turtles repeatedly exploited the aquatic environment as well. Although some pleurodiran clades also ventured into the marine realm, it is the cryptodires that did so most extensively. Among those, three major radiation phases are distinguished, with the first phase consisting of basal eucryptodiran taxa inhabiting littoral or near costal environments (Late Jurassic, Europe); the second phase including more open marine chelonioids (starting in the late Early Cretaceous, mainly North America and Eurasia); and the third phase (starting in the Palaeocene/Eocene, global distribution) including the highly-nested chelonioids, such as the modern cheloniid and dermochelyid turtles and closest relatives. A review of previously published as well as unpublished data of shell microstructures of these groups and those of some of the earliest aquatic turtles from the Middle Jurassic, Heckerochelys romani and Eileanchelys waldmani, show that bones are strongly influenced functionally as a result of life spent in an aquatic medium, whereas there are little to no characters of systematic value in the bones. We confirm the general tetrapod pattern that pelagic forms tend to show osteoporotic-like shell structures and neritic forms tend to have more bone ballast, especially by retaining a thickened external compacta. © 2014 The Linnean Society of London, Biological Journal of the Linnean Society, 2014, 112, 701–718.
