Abstract

Central to all machine learning algorithms is data representation. For multi-agent systems, selecting a representation which adequately captures the interactions among agents is challenging due to the latent group structure which tends to vary depending on context. However, in multi-agent systems with strong group structure, we can simultaneously learn this structure and map a set of agents to a consistently ordered representation for further learning. In this paper, we present a dynamic alignment method which provides a robust ordering of structured multi-agent data enabling representation learning to occur in a fraction of the time of previous methods. We demonstrate the value of this approach using a large amount of soccer tracking data from a professional league.

Abstract

Background. There are challenges related to the accurate and efficient measurement of lymphedema in people with breast cancer. The LymphaTech 3D Imaging System (LymphaTech, Atlanta, GA, USA) is a mobile, noninvasive platform that provides limb geometry measurements. Objective. The objective of this study was to estimate the reliability and validity of the LymphaTech for measuring arm volume in the context of women seeking care in a specialty breast cancer rehabilitation clinic. Design. This was a cross-sectional reliability and convergent validity study. Methods. People who had stage I to IV breast cancer with lymphedema or were at risk for it were included. Arm volume was measured in 66 participants using the LymphaTech and perometer methods. Test-retest reliability for a single measure, limb volume difference, and agreement between methods was analyzed for 30 participants. A method-comparison analysis was also used to assess convergent validity between methods. Results. Both LymphaTech and perometer methods displayed intraclass correlation coefficients (ICCs) of [greater than or equal to]0.99. The standard errors of measurement for the LymphaTech and length-matched perometer measurements were nearly identical. Similar intraclass correlation coefficients (0.97) and standard errors of measurement (38.0-40.7 mL) were obtained for the between-limb volume difference for both methods. The convergent validity analyses demonstrated no systematic difference between methods. Limitations. The sample size was not based on a formal sample size calculation. LymphaTech measurements included interrater variance, and perometer measurements contained intrarater variance. Conclusions. The LymphaTech had excellent test-retest reliability, and convergent validity was supported. Abstract

Abstract Background Lymphedema is a common complication of breast cancer treatment that affects one in five breast cancer survivors, yet there is no reliable method to detect lymphedema in the subclinical range. The objective of this study was to determine the feasibility and reliability of using an infrared 3D scanning device (ISD) as a peri-operative limb volume measurement tool. Methods Fifteen patients were analyzed based on inclusion criteria. Peri-operative measurements were obtained using tape measure and an ISD. Volumes were calculated using a standard algorithm for tape measure and a custom algorithm for ISD measurements. Linear regression models were used to assess ISD and tape measurement volume and circumference correlation. One-way ANOVA was used to compare change in percent difference at set time points post-operatively (2–3 weeks, 4–6 weeks, and 7–12 weeks) for both ISD and tape measure. t tests for unequal variances with the Bonferroni correction were performed among these groups. Results There is a positive linear correlation (R2 = 0.8518) between absolute volume measurements by the ISD and tape measure. Analyses over 2–10 weeks post-operatively showed that the ISD was able to detect volume changes in both the unaffected and the affected arm. Furthermore, the affected arm tended to have a greater increase in volume in the majority of patients, indicating these patients could be at risk for lymphedema. Conclusions Technology utilizing infrared 3D scanners can reliably measure limb volume pre- and post-treatment similarly to tape measure in a small sample of patients. Further research using 3D scanning technology with a longer follow up is warranted.

13. Expansion and cellular characterization of primary human adherent cells in the Quantum® Cell Expansion System, a hollow-fiber bioreactor system

Abstract

Primary adherent cell types can be expanded in the Quantum® Cell Expansion System (Quantum system), an automated platform that utilizes a hollow-fiber bioreactor. This system can replace manual cell culture and produce cells that retain their phenotypes and functionality. Bone- marrow-derived and adipose-derived mesenchymal stem/stromal cells have previously been successfully expanded on the Quantum system. We have now successfully used the Quantum system to expand fibroblasts and myoblasts. Hollow-fiber bioreactors were coated with adherence-supporting proteins, and then cells were loaded and expanded in the appropriate growth medium for 7 to 15 d. Cells were harvested from the bioreactors using enzymatic reagents. Harvested cell yields ranged from 100 × 106 to 1 × 109 cells, with viability typically above 90%. The number of doublings obtained from Quantum system harvests ranged from 4 to 9. The Quantum system is a functionally closed expansion system that can reduce contamination due to minimal interventions and can automate the culture process to reduce labor and reagent costs.

Abstract Methadone Maintenance Treatment (MMT) is among the most widely studied treatments for opiate dependence with proven benefits for patients and society. When misused, however, methadone can also be lethal. The issue of methadone diversion is a major concern for all MMT programs. A potential source for such diversion is from those MMT patients who receive daily take home methadone doses. Using a reverse phase high performance liquid chromatography method, seven of the nine patients who were randomly selected to have all of their remaining methadone take home doses (within a 24 hour period) analyzed, returned lower than expected quantities of methadone. This finding suggests the possibility that such patients may have tampered with their daily take home doses. Larger prospective observational studies are clearly needed to test the supposition of this pilot study.

Abstract

Competition models may be used by ecologists as basic guidelines in analyzing issues that contribute to the decline of species population. To be used for that purpose, a mathematical model must be prudently parameterized. Therefore, this paper examines the effects of interference competition by lions with human–related mortality to the population dynamics of African wild dogs. The model was carefully parameterized and validated with estimated data by employing the theory of basic reproduction number and by running the sensitivity analysis. Numerical simulation of the model was executed by using MATLAB to explore the outcome of certain key parameters when changes are applied on them.

20. The Lived Experience of Beginning Teachers Defining their Pedagogical Way of Being

Abstract

This hermeneutic phenomenological study explores the lived experience of beginning teachers defining their pedagogical way of being. Many beginning teachers have found frustration and disillusionment during their first year in the classroom, leading to high levels of attrition. Those beginning teachers who remain in the classroom may develop a way of being that opposes how they are as an individual and that pulls them further from the students in their care. How do the individuals who experience this phenomenon make meaning of that experience? What insights about the preparation and support of beginning teachers can be drawn from these experiences? This research is conducted in the tradition of hermeneutic phenomenology, grounded in the work of philosophers such as Heidegger (1962), Gadamer (2006/1975), and Merleau-Ponty (1962). Drawing from these philosophers, van Manen (1997) provides a detailed process used to conduct this form of research. This methodology serves to uncover the essences of this phenomenon, eliciting lived experience through hermeneutic conversation. To uncover the nature of the lived experience of beginning teachers defining their pedagogical way of being, six participants were recruited from urban, suburban, and rural secondary public schools in a south central county in Pennsylvania. The phenomenological text from this study not only reiterates the often noted "challenges" inherent in the first year experience, but also identifies a language of beginning. I seek to understand this language by connecting it to the three Buddhist ways of being, which guides my questioning of the "basics" of the beginning teaching experience, offering a new way of being in the flowing language of becoming. The experiences shared by the participants in this study uncovered many insights that may assist those charged with the care of beginning teachers during the periods of pre-service "formation" and in-service "orientation." Abstract

Human leukocyte antigen (HLA)-matched cord blood (CB) transplantation is a procedure for the treatment of certain hematological malignancies, hemoglobinopathies, and autoimmune disorders. However, one of the challenges is to provide a sufficient number of T cell-depleted hematopoietic stem and progenitor cells. Currently, only 4%-5% of the CB units stored in CB banks contain enough CD34 + cells for engrafting 70-kg patients. To support this clinical need, we have developed an automated expansion protocol for CB-derived CD34 + cells in the Quantum system's dynamic perfusion bioreactor using a novel cytokine cocktail comprised of stem cell factor (SCF), thrombopoietin (TPO), fms-like tyrosine kinase 3 ligand (Flt-3L), interleukin-3 (IL-3), IL-6, glial cell line-derived neurotrophic factor (GDNF), StemRegenin 1 (SR-1), and a fibronectin-stromal-cell-derived factor-1 (SDF-1)-coated membrane. In an 8-day expansion of a 2 × 10 6 positively selected CD34 + cell inoculum from 3 donor lineages, the mean cell harvest and cell viability were 1.02 × 10 8 cells and 95.5%, respectively, and the mean frequency of the CD45 + CD34 + immunophenotype was 54.3%. The mean differentiated cell frequencies were 0.5% for lymphocytes, 15.8% for neutrophils, and 15.4% for platelets.
