(1) Background: Ocimum basilicum L. is an aromatic medicinal plant of the Lamiaceae family known as sweet basil. It is used in traditional medicine for its beneficial effects on gastrointestinal disorders, inflammation, immune system, pyrexia or cancer among others. Ocimum basilicum (OB) leaf extracts contain many phytochemicals bearing the plant health effects but no reports is available on the potential bioactivity of stem extracts. Our investigation aimed at assessing the differential biological activity between basil leaf and stem to promote this co-product valorization. (2) Method: For this purpose we explored phytochemical composition of both parts of the plant. Antioxidant activity was evaluated through total polyphenol content measure, DPPH and ORAC tests. Anti-inflammatory markers on stimulated macrophages, including NO (nitric oxide), TNFa (tumor necrosis factor alpha), IL-6 (interleukin 6), MCP1 (monocyte attractant protein 1) and PGE-2 (prostaglandin E2), were evaluated. In addition, we investigated OB effects on jejunum smooth muscle contractility. (3) Results: OB extracts from leaves and stems demonstrated a different biological activity profile at the level of both antioxidant, anti-inflammatory and smooth muscle relaxation effects. (4) Conclusion: Taken together our results suggest that Ocimum basilicum extracts from co-product stems, in addition to leaves, may be of interest at the nutrition-health level with specific therapeutic potential.

Besides the wide use of its roots, cassava leaves have been used locally as green vegetables and for medicinal purposes. However, nutritional health data regarding cassava leaves is limited, therefore we investigated its composition and associated potential bioactivity interest for human health. (2) Methods: Cassava leaf bioactivity investigations focused on antioxidant properties (free radical scavenging) in association with immunomodulatory activities on inflammatory murine macrophages to measure the impact of cassava extract on the production of pro-inflammatory cytokines such as Interleukin-6, Tumor Necrosis Factor alpha, Monocyte Chemoattractant Protein-1, Prostaglandin-E2 and mediators such as nitric oxide. (3) Results: Antioxidant and immunomodulatory bioactivities were significant, with a concentration-dependent inhibition of cytokines production by inflammatory macrophages; (4) Conclusions: Taken together, our results tend to suggest that Manihot esculenta leaves might be underrated regarding the potential nutrition-health interest of this vegetal matrix for both human nutrition and prophylaxis of metabolic disease with underlying low grade inflammation status.

Banana bunchy top virus (BBTV) is a six-component ssDNA virus (genus Babuvirus, family Nanoviridae) transmitted by aphids, infecting monocots (mainly species in the family Musaceae) and likely originating from South-East Asia where it is frequently associated with self-replicating alphasatellites. Illumina sequencing analysis of banana aphids and leaf samples from Africa revealed an alphasatellite that should be classified in a new genus, phylogenetically related to alphasatellites of nanoviruses infecting dicots. Alphasatellite DNA was encapsidated by BBTV coat protein and accumulated at high levels in plants and aphids, thereby reducing helper virus loads, altering relative abundance (formula) of viral genome components and interfering with virus transmission by aphids. BBTV and alphasatellite clones infected dicot Nicotiana benthamiana, followed by recovery and symptomless persistence of alphasatellite, and BBTV replication protein (Rep), but not alphasatellite Rep, induced leaf chlorosis. Transcriptome sequencing revealed 21, 22 and 24 nucleotide small interfering (si)RNAs covering both strands of the entire viral genome, monodirectional Pol II transcription units of viral mRNAs and pervasive transcription of each component and alphasatellite in both directions, likely generating double-stranded precursors of viral siRNAs. Consistent with the latter hypothesis, viral DNA formulas with and without alphasatellite resembled viral siRNA formulas but not mRNA formulas. Alphasatellite decreased transcription efficiency of DNA-N encoding a putative aphid transmission factor and increased relative siRNA production rates from Rep- and movement protein-encoding components. Alphasatellite itself spawned the most abundant siRNAs and had the lowest mRNA transcription rate. Collectively, following African invasion, BBTV got associated with an alphasatellite likely originating from a dicot plant and interfering with BBTV replication and transmission. Plant bioactive extracts represent a major resource for identifying drugs and adjuvant therapy for type 2 diabetes. To promote early screening of plants' antidiabetic potential, we designed a four in vitro tests strategy to anticipate in vivo bioactivity. Two antidiabetic plants were studied: Ocimum gratissimum L. (Oc) leaf extract and Musanga cecropoides R. Br. ex Tedlie (Mu) stem bark extract. Chemical compositions were analyzed by LCMS and HPLC. Antidiabetic properties were measured based on (1) INS-1 cells for insulin secretion, (2) L6 myoblast cells for insulin sensitization (Glut-4 translocation), (3) L6 myoblast cells for protection against hydrogen peroxide (H2O2) oxidative stress (cell mortality), and (4) liver microsomial fraction for glucose-6-phosphastase activity (G6P). Oc extract increased insulin secretion and insulin sensitivity, whereas it decreased oxidative stress-induced cell mortality and G6P activity. Mu extract decreased insulin secretion and had no effect on insulin sensitivity or G6P activity, but it increased oxidative stress-induced cell mortality. Results were compared with NCRAE, an antidiabetic plant extract used as reference, previously characterized and reported with increased insulin secretion and insulin sensitivity, protection against oxidative stress, and decreased G6P activity. The proposed set of four in vitro tests combined with chemical analysis provided insight into the interest in rapid early screening of plant extract antidiabetic potential to anticipate pharmaco-toxicological in vivo effects.

Population breeding through recurrent selection is based on the repetition of evaluation and recombination among best-selected individuals. In this type of breeding strategy, early evaluation of selection candidates combined with genomic prediction could substantially shorten the breeding cycle length, thus increasing the rate of genetic gain. The objective of this study was to optimize early genomic prediction in an upland rice (Oryza sativa L.) synthetic population improved through recurrent selection via shuttle breeding in two sites. To this end, we used genomic prediction on 334 S0 genotypes evaluated with early generation progeny testing (S0:2 and S0:3) across two sites. Four traits were measured (plant height, days to flowering, grain yield, and grain zinc concentration) and the predictive ability was assessed for the target site. For days to flowering and plant height, which correlate well among sites (0.51–0.62), an increase of up to 0.4 in predictive ability was observed when the model was trained using the two sites. For grain zinc concentration, adding the phenotype of the predicted lines in the nontarget site to the model improved the predictive ability (0.51 with two-site and 0.31 with single-site model), whereas for grain yield the gain was less (0.42 with two-site and 0.35 with single-site calibration). Through these results, we found a good opportunity to optimize the genomic recurrent selection scheme and maximize the use of resources by performing early progeny testing in two sites for traits with best expression and/or relevance in each specific environment.

Resorption is the active withdrawal of nutrients before leaf abscission. This mechanism represents an important strategy to maintain efficient nutrient cycling; however, resorption is poorly characterized in old-growth tropical forests growing in nutrient-poor soils. We investigated nutrient resorption from leaves in 39 tree species in two tropical forests on the Guiana Shield, French Guiana, to investigate whether resorption efficiencies varied with soil nutrient, seasonality, and species traits. The stocks of P in leaves, litter, and soil were low at both sites, indicating potential P limitation of the forests. Accordingly, mean resorption efficiencies were higher for P (35.9%) and potassium (K; 44.6%) than for nitrogen (N; 10.3%). K resorption was higher in the wet (70.2%) than in the dry (41.7%) season. P resorption increased slightly with decreasing total soil P; and N and P resorptions were positively related to their foliar concentrations. We conclude that nutrient resorption is a key plant nutrition strategy in these old-growth tropical forests, that trees with high foliar nutrient concentration reabsorb more nutrient, and that nutrients resorption in leaves, except P, are quite decoupled from nutrients in the soil. Seasonality and biochemical limitation played a role in the resorption of nutrients in leaves, but species-specific requirements obscured general tendencies at stand and ecosystem level.

The olive tree (Olea europaea L. subsp. europaea var. europaea) is currently one of the most widely cultivated temperate fruit trees in the world. It is characterised by a large genetic diversity, as well as a large variation in phenotypic traits. Across the Mediterranean Basin, the nuclear genetic diversity of olive is organized into three main groups: 'West', 'Central', 'East', with an additional group ('Mosaic' or 'admixed') corresponding to varieties resulting from the hybridization of varieties belonging to at least two of these three groups. The first aim of the present study is to test whether the phenotypic traits of the cultivated varieties differ among these four genetic groups. To do so, nine leaf traits and five pit traits were measured on 35 varieties belonging to these four groups. The second aim of this study is to use this combination of traits to assess whether some trait covariations previously observed across and/or within species hold within olive, a sclerophyllous species characteristic of the Mediterranean Basin. We found that: (1) Trait values span a wide range of variation across varieties, from approximately 7-fold for pit volume, the most variable trait, to 1.3 fold for leaf dry matter content, the least variable one; (2) For most traits, there was no significant difference among the four genetic groups recognized across the Mediterranean Basin; (3) Leaf size was positively related to leaf mass per area, a trait describing the morpho-anatomy of the leaf; this is the consequence of leaf mass increasing more rapidly than leaf area, and agrees with the hypothesis that bigger leaves require to be stiffer to compensate for mechanical damage and gravity; (4) The two underlying components of leaf mass per area, leaf thickness and density, contributed equally to its determination; (5) Leaf and pit sizes were positively related. A structural equation modelling approach allowed us to show that this relationship was mediated through leaf mass per area

The objective of this study was to characterise the nutritional potential of leaves and identify a diversity centre with low cyanide and high nutrient content among 178 Latin American cassava genotypes. This field‐based collection, represents the seven diversity centres, held at The International Center for Tropical Agriculture (CIAT Palmira, Colombia) by the Cassava Program. The cyanide, all‐trans‐β‐carotene and lutein concentrations in cassava leaves ranged from 346 ‐ 7484 ppm dry basis (db), from 174‐547 μg.g‐1 db and 15 ‐ 181 μg · g‐1 db respectively. Cassava leaves also showed significant levels of essential amino acids leucine, lysine, phenylalanine, valine and threonine, and average total protein content of 26.24 g · 100g‐1 db. Among seven diversity centres, South American rainforest group showed low cyanide and high carotene content in leaves. In addition, VEN77 and PAN51 genotypes stood out for having low cyanide in leaves and roots and high carotene in leaves. This genetic diversity can be used to select high potential progenitors for breeding purposes.

Phenolic compounds are involved in plant response to environmental conditions and are highly present in leaves of Coffea arabica L., originally an understory shrub. To increase knowledge of C. arabica leaf phenolic compounds and their patterns in adaptation to light intensity, mature leaves of Ethiopian wild accessions, American pure lines and their relative F1 hybrids were sampled in full sun or under 50% shade field plots in Mexico and at two contrasting elevations in Nicaragua and Colombia. Twenty-one phenolic compounds were identified by LC-DAD-MS2 and sixteen were quantified by HPLC-DAD. Four of them appeared to be involved in C. arabica response to light intensity. They were consistently more accumulated in full sun, presenting a stable ratio of leaf content in the sun vs. shade for all the studied genotypes: 1.6 for 5-CQA, F-dihex and mangiferin and 2.8 for rutin. Moreover, 5-CQA and mangiferin contents, in full sun and shade, allowed for differentiating the two genetic groups of Ethiopian wild accessions (higher contents) vs. cultivated American pure lines. They appear, therefore, to be potential biomarkers of adaptation of C. arabica to light intensity for breeding programs. We hypothesize that low 5-CQA and mangiferin leaf contents should be searched for adaptation to full-sun cropping systems and high contents used for agroforestry systems.

The programmed loss of a plant organ is called abscission, which is an important cell separation process that occurs with different organs throughout the life of a plant. The use of floral organ abscission in Arabidopsis thaliana as a model has allowed greater understanding of the complexities of organ abscission, but whether the regulatory pathways are conserved throughout the plant kingdom and for all organ abscission types is unknown. One important pathway that has attracted much attention involves a peptide ligand-receptor signalling system that consists of the secreted peptide IDA (INFLORESCENCE DEFICIENT IN ABSCISSION) and at least two leucine-rich repeat (LRR) receptor-like kinases (RLK), HAESA (HAE) and HAESA-LIKE2 (HSL2). In the current study we examine the bioactive potential of IDA peptides in two different abscission processes, leaf abscission in Populus and ripe fruit abscission in oil palm, and find in both cases treatment with IDA peptides enhances cell separation and abscission of both organ types. Our results provide evidence to suggest that the IDA–HAE–HSL2 pathway is conserved and functions in these phylogenetically divergent dicot and monocot species during both leaf and fruit abscission, respectively.

In complex coffee-based agroforestry, quantifying interactions within the agrosystem that impact on coffee disease regulation and on coffee yield is a major stake to design sustainable cropping systems. To this end, we analyzed the interaction network between shade trees, coffee trees (Catimor variety), coffee foliar diseases complex (CFDC; majority of Mycena citricolor) and soil characteristics. The system is characterized by 40 variables measured in 60 plots spread on three farms (monitored for 2 years) in Nicaragua. We used Partial Least Square Path Modeling (PLSPM) to study the network interaction. We built 6 blocks with the more significant variables of each component: shade trees (shade percentage, species), soil (Cation Exchange Capacity, P), CFDC (incidence, severity), coffee trees age and size, coffee growth and coffee yield. The second part of the PLSPM was performed between blocks. Shade trees, mostly the shade percentage, had direct positive effects on CFDC and soil quality, and negative effects on coffee growth and yield. Shade had also an indirect negative effect on coffee trees by increasing CFDC, which impedes coffee growth and yield. Soil variables being negatively related to CFDC, shade had an indirect effect on coffee trees. Reducing excessive shade cover seems to be a solution to enhance positive impacts of shade trees on coffee yield. Overall, shade management requires an analysis of trade-offs between soil quality, diseases regulation and yield gains.

Leaf traits are often strongly correlated with yield, which poses a major challenge in rice breeding. In the present study, using a panel of Vietnamese rice landraces genotyped with 21,623 single-nucleotide polymorphism markers, a genome-wide association study (GWAS) was conducted for several leaf traits during the vegetative stage. Vietnamese landraces are often poorly represented in panels used for GWAS, even though they are adapted to contrasting agrosystems and can contain original, valuable genetic determinants. A panel of 180 rice varieties was grown in pots for four weeks with three replicates under nethouse conditions. Different leaf traits were measured on the second fully expanded leaf of the main tiller, which often plays a major role in determining the photosynthetic capacity of the plant. The leaf fresh weight, turgid weight and dry weight were measured; then, from these measurements, the relative tissue weight and leaf dry matter percentage were computed. The leaf dry matter percentage can be considered a proxy for the photosynthetic efficiency per unit leaf area, which contributes to yield. By a GWAS, thirteen QTLs associated with these leaf traits were identified. Eleven QTLs were identified for fresh weight, eleven for turgid weight, one for dry weight, one for relative tissue weight and one for leaf dry matter percentage. Eleven QTLs presented associations with several traits, suggesting that these traits share common genetic determinants, while one QTL was specific to leaf dry matter percentage and one QTL was specific to relative tissue weight. Interestingly, some of these QTLs colocalize with leaf- or yield-related QTLs previously identified using other material. Several genes within these QTLs with a known function in leaf development or physiology are reviewed.

Coffee Leaf Rust (CLR) has been reported in over 50 coffee growing countries causing significant economic losses in Arabica coffee. The aim of this study was to determine the effect of shade on leaf life span in relation to severity of coffee leaf rust. To achieve this objective, three shade levels (shaded, partial shade, unshaded) were applied in three agro-ecological zones (AEZ) i.e. Upper-Midland (UM) I, II and III. Each treatment was replicated 4 times in 7 farms in each AEZ. All the farms had similar agronomic management levels. Infected leaves were counted per tree and disease severity was scored every month, between January and September, 2014 which is the peak period for CLR. Yields per plot were estimated using the method of Cilas and Descroix, 2004. The results showed that shaded trees retained the infected leaves 8 weeks longer than the unshaded in all the agro-ecological zones. Similarly, yield estimate from the shaded coffee (1521kg/ha clean coffee) was significantly higher than the unshaded (1050kg/ha clean coffee). Although leaves remain longer on shaded trees, allowing more time for the disease to develop and progress, severity level of shaded trees remains lower (12.8%) than unshaded (19.0%), suggesting the expression of regulation mechanism under the control of shade. Moreover, full shade generated by dense canopy, such as the one from mango or avocado tree best manages CLR across all the tested three agro-ecological zones.

Plant DNA and RNA extraction methods are well established, with a wide range of protocols, depending on the purposes of each laboratory/research. Nowadays, quick, inexpensive and easy plant DNA and RNA extraction methods are highly sought after. We developed an optimized protocol for plant DNA and RNA extraction that uses an inexpensive bench drill and plastic bags and does not require liquid nitrogen. DNA from leaves and RNA from leaves and roots of banana, pineapple, citrus, papaya, passion fruit and cassava, were extracted using a basic cetyltrimethylammonium bromide method. Both nucleic acids were quantified and evaluated for quality based on agarose gel electrophoresis. The DNA and RNA extractions were successful for all species, and RNA quality in pellets was maintained after storage at room temperature for three weeks. This protocol can reduce costs considerably in laboratories with ongoing routine activities of DNA and RNA extraction for genetic diversity and gene expression analyses, where other conventional methods have not been successful due to explant, condition of samples and quantity and quality of nucleic acids. This is especially relevant for many laboratories in developing countries where the cost and availability of liquid nitrogen may be a constraint.

Variation of plant functional traits may predict ecosystem function1 , yet there is little work linking functional traits of crops and service trees, their interactions, and processes in agrofo­restry systems2, specifically the transmission of pathogens. Focusing on coffee as a model, we explore how multi-species functional traits adjust abiotic processes that affect the dispersal and persistence of coffee leaf rust (CLR)3. We hypothesize that shade tree canopy and leaf traits will mediate CLR transmission via abiotic modifications and that key coffee functional leaf traits will suppress CLR under different agroforestry systems. This study was conducted in CATIE's coffee agroforestry research trial in Costa Rica. Throughfall kinetic energy under diverse shade tree canopies was modified by shade tree composition, canopy characteristics (e.g. crown base height; CBH) and functional leaf traits (e.g. specific leaf area; SLA). Certain shade tree traits such as canopy depth, CBH and SLA also related to trends in plant-level CLR incidence (Figure 1 ). CLR tended to favour certain coffee leaf functional traits, where coffee plants with low mean leaf nitrogen concentration and high mean SLA had higher plant­level CLR incidence. These results suggest that managing the functional leaf trait variation of shade trees and targeting key coffee functional leaf traits can change the persistence of CLR, thus improving our understanding of alternative resistance measures available in agroforestry.

Breeders select for yield, thereby indirectly selecting for traits that contribute to it. We tested if breeding has affected a range of traits involved in plant architecture and light interception, via the analysis of a panel of 60 maize hybrids released from 1950‐2015. This was based on novel traits calculated from reconstructions derived from a phenotyping platform. The contribution of these traits to light interception was assessed in virtual field canopies composed of 3D plant reconstructions, with a model tested in a real field. Two categories of traits had different contributions to genetic progress. (i) The vertical distribution of leaf area had a high heritability and showed a marked trend over generations of selection. Leaf area tended to be located at lower positions in the canopy, thereby improving light penetration and distribution in the canopy. This potentially increased the carbon availability to ears, via the amount of light absorbed by the intermediate canopy layer, (ii) Neither the horizontal distribution of leaves in the relation to plant rows nor the response of light interception to plant density showed appreciable trends with generations. Hence, among many architectural traits, the vertical distribution of leaf area was the main indirect target of selection.

Centella asiaticais aherbal plant generally used for its curative properties. Two foliar morphotypes were recently identified in Madagascar: a reniform tetraploid (2n = 4x = 36) in the Center and the East of the island and a round diploid (2n = 2x = 18) in the West. The objective of this study was to evaluate the nutritional properties of these two morphotypes and to compare them with other green leafy vegetables. Reniform leaves were collected in Antananarivo and Moramanga and round leaves were collected in Tsiroanomandidy and Analavory. Macronutrient content wasdetermined by standard methods, mineral contents were analysed by inductively coupled plasma atomic emissionspectroscopy. Ascorbic acid and carotenoids were quantified byHPLC.Food composition showed significant differences between the morphotypes. Reniform leaves have higher protein content (19–22 % dry weight (DW)) than round leaves (17–21 % DW). Lipid content are from 2.5 to 6.0 % DW. Reniform leaves have higher iron content than round leaves. Iron content of C. asiatica's leaves range from 3.8 to 12.5 mg/100 g fresh weight (FW). Reniform leaves from Moramanga have the highest protein, lipid and iron content.Round leaves from Analavory have the highest calcium and magnesium content, which can cover 41 % and 17 % of the Recommended Dietary Allowances (RDA), respectively. Leaves of C. asiaticahave low vitamin C content (1.3 to 7.7 mg/100 g FW). Consumption of 100 g of C. asiaticaleaves could cover 10 to 21 % of vitamin A daily requirements of women for reniform leaves and 21 to 37 % of vitamin A requirement of women for round leaves. Round leaves from Analavory have the highest β-carotene content. It is hoped that from these findings, the people of Madagascar will be encouraged to include C. asiaticain their diets for its nutritional benefits.

The programmed loss of a plant organ is called abscission, which is an important cell separation process that occurs with different organs throughout the life of a plant. The use of floral organ abscission in Arabidopsis thaliana as a model has allowed greater understanding of the complexities of organ abscission, but whether the regulatory pathways are conserved throughout the plant kingdom and for all organ abscission types is unknown. One important pathway that has attracted much attention involves a peptide ligand-receptor signalling system that consists of the secreted peptide IDA (INFLORESCENCE DEFICIENT IN ABSCISSION) and at least two leucine-rich repeat (LRR) receptor-like kinases (RLK), HAESA (HAE) and HAESA-LIKE2 (HSL2). In the current study we examine the bioactive potential of IDA peptides in two different abscission processes, leaf abscission in Populus and ripe fruit abscission in oil palm, and find in both cases treatment with IDA peptides enhances cell separation and abscission of both organ types. Our results provide evidence to suggest that the IDA–HAE–HSL2 pathway is conserved and functions in these phylogenetically divergent dicot and monocot species during both leaf and fruit abscission, respectively.
