Post-traumatic stress disorder (PTSD) is an anxiety disorder of considerable prevalence in individuals who have experienced a traumatic event. Studies of the neural substrate of this disorder have focused on the role of areas such as the hippocampus, the amygdala and the medial prefrontal cortex. We show that the ventral tegmental area (VTA), which directly modulates these areas, is part of this circuitry. Using a rat model of PTSD, we show that a brief but intense foot shock followed by three brief reminders can cause long-term behavioral changes as shown by anxiety-like, nociception, and touch-sensitivity tests. We show that an intraperitoneal injection of a dopamine (DA) antagonist or a bilateral inactivation of the VTA administered immediately before the traumatic event decrease the occurrence or intensity of these behavioral changes. Furthermore, we show that there is a significant decrease of baseline VTA dopaminergic but not GABAergic cell firing rates 2 weeks after trauma. Our data suggest that VTA DA neurons undergo long-term physiological changes after trauma and that this brain area is a crucial part of the circuits involved in PTSD symptomatology.

The CB(1) cannabinoid receptor is implicated in the rewarding properties of many drugs of abuse, including cocaine. While CB(1) receptor involvement in the acute rewarding properties of cocaine is controversial, CB(1) antagonists such as SR141716 (rimonabant) have clearly been found to prevent cue- and cocaine-elicited reinstatement of cocaine self-administration in rodents. Here we demonstrate the novel involvement of CB(1) receptors in the maintenance of behavioral sensitization to cocaine in C57BL/6 mice. Consistent with previous reports, the induction of locomotor sensitization following repeated daily cocaine was not prevented by systemic pretreatment of either rimonabant, Delta(9)-tetrahydrocannabinol (THC), or a 1:1 mixture of THC and cannabidiol (CBD). In contrast, established cocaine sensitization was markedly disrupted following subchronic treatment with rimonabant alone. This effect was notably context-dependent, in that rimonabant did not diminish established cocaine sensitization if delivered in the home cage, but only if the rimonabant-injected mice were exposed to activity chambers previously paired with cocaine. These findings are consistent with CB(1) receptor involvement in conditioned cocaine-seeking behaviors, and further suggest that endocannabinoid (eCB)-mediated synaptic plasticity may act specifically within drug-paired environments to maintain cocaine-directed behavioral responses.

Recreational abuse of toluene-containing volatile inhalants by adolescents is a significant public health problem. The mechanisms underlying the abuse potential of such substances remain unclear, but could involve increased activity in mesoaccumbal dopamine (DA) afferents innervating the nucleus accumbens (ACB). Here, using in vitro electrophysiology, we show that application of behaviorally relevant concentrations of toluene directly stimulates DA neurons in the ventral tegmental area (VTA), but not surrounding midbrain regions. Toluene stimulation of VTA neurons persists when synaptic transmission is reduced. Moreover, unlike non-DA neurons, the magnitude of VTA DA neuron firing does not decline during longer exposures designed to emulate 'huffing'. Using dual-probe in vivo microdialysis, we show that perfusion of toluene directly into the VTA increases DA concentrations in the VTA (somatodendritic release) and its terminal projection site, the ACB. These results provide the first demonstration that even brief exposure to toluene increases action potential drive onto mesoaccumbal VTA DA neurons, thereby enhancing DA release in the ACB. The finding that toluene stimulates mesoaccumbal neurotransmission by activating VTA DA neurons directly (independently of transynaptic inputs) provide insights into the neural substrates that may contribute to the initiation and pathophysiology of toluene abuse.

Inhalant abuse is a common, potentially lethal, form of drug abuse. Although the putative psychotropic compo- nent of some popularly abused inhalants appears often to be the organic solvent toluene, its effects on midbrain neurones which comprise reward pathways have not been established. Therefore, the present study was designed to assess the response of ventral tegmental dopamine neurones during toluene inhalation. Electrophysiological determinations were made using extracellular single-unit recordings in ketamine anaesthetized rats that were exposed to acute (1-15.3 rnin.) concentrations of toluene vapor (1 1,500 ppm) similar to those consumed by inhalant abusers. Toluene exposure through a tracheal breathing tube elicited two distinctly different patterns of response in dopamine neurons. One pattern consisted of an initial stimulation of neuronal firing (+221%,t72%; 8.5 min.). The other pattern consisted of only an inhibition of firing regardless of the length of exposure. Furthermore, the changes in firing rates were paralleled by changes in number of action potentials contained in bursts. Blood samples taken at the time of the dopamine recordings revealed comparable toluene concen- trations (4-79 pg/ml, n=24) regardless of the patterns of response. These results suggest that mesolimbic dopamine neurotransmission can be changed by an exposure paradigm comparable to that used by human abusers, and that these changes may be integral to the reinforcing effects underlying inhalant abuse. The widespread abuse of inhalants (i.e., spray paints, glues, and lacquers) is a significant and potentially lethal problem among youth. An epidemiological study showed that in 1995, 23% of the adolescents in the United States were ex- perimenting with inhalants (Mathias 1996). Notably, up- wards of 38% of users may die during their first inhalant experience (Flanagan & Ives 1994). Although inhalant use may also lead to the abuse of other substances, a significant percentage of users prefer and continue to abuse inhalants to the point of irreversible neurotoxic consequences (Delva et ul. 1997). Although the overall incidence of inhalant abuse is extensive there is little experimental data implicat- ing the abuse liability of solvents such as toluene with a specific CNS site or mechanism of action. The acute consumption of inhalants produces a variety of psychotropic effects including an initial euphoria, intoxi- cation, slurred speech, ataxia, numbness of the extremities, amnesia, hostile behaviour, and hallucinations. As inhal- ation continues, seizures, tinnitus, diffuse somatic pain, tremor, and death can occur (Flanagan & Ives 1994). Sev- eral reports also link chronic "huffing" to permanent liver, kidney, heart and cranial nerve damage, cerebral, cerebellar, and brainstem atrophy, neuropathies, dependence, and psy- chosis (for a review see Arlien-S~rborg 1992). Many consider the organic solvent toluene the essential psychotropic com- ponent found in the more commonly abused inhalants. Even non-human primates lever press for bursts of toluene

Phencyclidine (PCP) and PCP-like drugs increased firing rates and the amount of burst activity of A10 dopamine neurons recorded extracellularly in anesthetized rats. These effects correlated to their potency as noncompetitive N-methyl-D-aspartate (NMDA) antagonists but not to their affinity for the sigma-receptor. In contrast, the direct acting NMDA antagonists, CGS 19755, (+)CPP and NPC 12626, produced no alterations in either firing rate or burst patterns. However, pretreatment with either CGS 19755 or (+)CPP effectively attenuated the excitatory effects of PCP. In contrast to the findings obtained in the whole animal, PCP in the midbrain slice preparation did not activate dopamine neurons, even though PCP selectively blocked the excitations induced by NMDA but not those of the nonNMDA agonists, kainate and AMPA. In the self-administration test system a progressive-ratio schedule of reinforcement was used to assess the reinforcing strength of PCP and the PCP congeners, TCP and BTCP. In comparison to BTCP, which produced breaking points comparable to those occurring with equivalent doses of cocaine, PCP and TCP had considerably less reinforcing efficacy. These behavioral differences appeared to reflect the affinity of the compounds for the dopamine reuptake site versus the PCP binding site on the NMDA-ion channel complex. Thus, PCP's psychotomimetic effects and abuse liability properties may result from the differential mechanisms by which it affects limbic and cortical dopamine neurotransmission.

Background: Cortical spreading depression (CSD) is a wave of depolarization followed by depression of bioelectrical activity that slowly propagates through the cortex. CSD is believed to be the underlying mechanism of aura in migraine; however, whether CSD can elicit pain associated with migraine headache is unclear. Methods: Awake, freely moving rats were monitored for both CSD events and behavioral responses resulting from dural-cortical pinprick and/or KCl injection to the occipital cortex. Results: We observed tactile allodynia of the face and hindpaws, as well as enhanced Fos expression within the trigeminal nucleus caudalis (TNC) following CSD induced by KCl injection into the cortex, but not by pinprick. Application of KCl onto the dura elicited cutaneous allodynia and increased Fos staining in the TNC but did not elicit CSD events. Conclusions: These data suggest that sustained activation of trigeminal afferents that may be required to establish cutaneous allodynia may not occur following CSD events in normal animals.

The abuse of volatile inhalants remains a prominent, yet poorly understood, form of substance abuse among youth. Nevertheless, the identification of a mechanism underlying the reinforcing properties of inhalants has been hampered by the lack of a clearly identifiable neural substrate upon which these chemicals act. One ingredient that is common to many abused inhalants is toluene, an organic solvent that is self-administered by nonhuman primates and rodents. Most drugs of abuse have been found to elicit forward locomotion in rats, an effect owing to the activation of mesoaccumbal dopamine (DA) pathways. Thus, the present study was undertaken using two different approaches to determine whether toluene-induced locomotor hyperactivity is also ultimately dependent upon DA neurotransmission in the mesolimbic nucleus accumbens (NAC). Here we report on the effects of 6-hydroxydopamine (6-OHDA) lesions of the NAC or pretreatment with the metabotropic mGlu2/3 receptor agonist LY379268 on toluene-induced locomotor activity. Both procedures, which are known to alter neurotransmission within the NAC, significantly attenuated toluene's locomotor stimulatory effects. These results provide strong support for a central mechanism of action of inhalants, which in the past has been more typically attributed to general nonspecific mechanisms throughout the brain. Moreover, as with other drugs of abuse, the NAC may be the final common pathway subserving toluene's abuse liability.

FOOT-SHOCK induced stress promotes a five to sixfold increase in β-endorphin plasma levels. Similar increases were also found for ACTH plasma levels. In the hypothalamus, foot-shock induced stress promotes a decrease of β-endorphin assayed by radioimmunoassays. These data suggest that physiological increases in plasma β-endorphin levels induced by stress do not result in elevated levels of brain β-endorphin.

Basal levels of immunoreactive (ir) beta-endorphin, corticotropin (ACTH), and prolactin (PRL) in plasma of male rats decrease after dexamethasone pretreatment (400 microgram/kg at 24 hr and 200 microgram/kg at 2 hr before). Inescapable electric footshocks increase ir-beta-endorphin, ACTH, and PRL plasma levels and this effect is blocked by dexamethasone pretreatment. Morphine (20 mg/kg) also increases ir-beta-endorphin, ACTH, and PRL levels. Dexamethasone pretreatment blocks the morphine-induced release of ir-beta-endorphin but does not prevent the morphine-induced release of PRL. Naloxone, the opiate antagonist, decreases basal plasma levels of PRL and partially blocks the stress-induced increase of PRL, but it has no effect on the basal or stress-induced release of ir-beta-endorphin. These results are consistent with the proposal that beta-endorphin may interact with an opiate receptor involved in the regulation of PRL secretion.

The atypical excitation by opiates and opioid peptides of hippocampal pyramidal cells can be antagonized by iontophoresis of naloxone, the gamma-aminobutyric acid antagonists bicuculline, or magnesium ion. The recurrent inhibition of these cells evoked by transcallosal stimulation of the contralateral hippocampus is blocked by enkephalin but only shortened by acetylcholine. The results suggest that the opioids excite pyramidal neurons indirectly by inhibition of neighboring inhibitory interneurons (probably containing gamma-aminobutyric acid). This mechanism may be pertinent to the electrographic signs of addictive drugs.
